The aerosol release during the professional application of two different isocyanate based two component spray systems was identified and the physicochemical properties of the released airborne aerosols were characterized. For this purpose, aerosol release fractions were measured using a mass balance method described by Schwarz and Koch. Besides the release of total aerosol mass special emphasis was directed to the content of free monomeric MDI (4,4'- and 2,4'-diphenylmethane diisocyanate) in three particle size fractions relevant for inhalation uptake: inhalable, thoracic, and respirable size fraction. Two products were investigated: a two component PUR (polyurethane) spray foam (Elastopor) and a polyurea spray coating (Elastocoat). The mass fraction of the applied products released with the overspray as inhalable aerosol is 6.3 × 10-4 (Elastopor) and 4.0 × 10-4 (Elastocoat). Of the released total overspray aerosol 75 or 80% were in the thoracic size range, and 26 or 47% in the respirable regime for the PUR spray foam or the polyurea spray coating, respectively. At the time point of release the content of monomeric MDI in the aerosol corresponds to the composition of the bulk product. However, analysis of air samples indicates that <1% of the spray foam aerosol mass release fraction is attributed to free monomeric 4,4'- and 2,4'-MDI. For the Spray Coating the monomeric MDI fraction is <0.1%. Higher oligomers of MDI and prereacted oligomeric reaction products make up a few percent of the aerosol. This results in a total fraction of 0.0023% (spray foam) and 0.00015% (spray coating), respectively, of the sprayed monomeric MDI that is transferred into an inhalable aged aerosol. This data demonstrates, that during professional spraying only a small fraction of the total applied mass is released as airborne aerosol. The potential distribution of the theoretically inhalable aerosol in the respiratory tract and a low residual monomer content is described, significantly contributing to a refined safety assessment of the spray applications at the workplaces., (© The Author(s) 2019. A versatile and simple mass balance method for the measurement of the release fraction of thoracic and respirable particles of non-volatile compounds of spray products is presented. The release fractions are defined as the ratio between the mass of suspended non-volatile particulate matter in the thoracic and respirable particle size range and the total mass of non-volatile material released with the spray action. For its determination, a spray bolus of short duration and of defined mass is sprayed into a well stirred control chamber. The respirable and thoracic aerosol mass associated with the spray bolus is determined by measuring the time averaged mass concentration inside the control volume and the half time of the exponential concentration decrease to be expected in well stirred systems to correct for mass losses during sampling.  The method is used for a wide range of spray products and technologies for which the release fractions vary by orders of magnitude. A set of data is presented elucidating the relationship between spray technology and fine particle release. Furthermore, a simple rule of thumb was derived from the data that allows for estimation of the release fractions based on a characteristic diameter of the spray droplets. The usefulness of the mass balance method for substance classification as well as for generating input data for exposure assessment and indoor air quality modeling is discussed.

A field study was carried out in order to derive a factor for the conversion of historic worker exposure data on airborne beryllium (Be) obtained by sampling according to the 37-mm closed faced filter cassette (CFC) 'total' particulate method into exposure concentration values to be expected when sampling using the 'Gesamtstaubprobenahmesystem' (GSP) inhalable sampling convention. Workplaces selected to represent the different copper Be work processing operations that typically occur in Germany and the EU were monitored revealing a broad spectrum of prevailing Be size distributions. In total, 39 personal samples were taken using a 37-mm CFC and a GSP worn side by side for simultaneous collection of the 'total' dust and the inhalable particulates, respectively. In addition, 20 static general area measurements were carried out using GSP, CFC, and Respicon samplers in parallel, the latter one providing information on the extra-thoracic fraction of the workplace aerosol. The study showed that there is a linear relationship between the concentrations measured with the CFC and those measured with the GSP sampler. The geometric mean value of the ratios of time-weighted average concentrations determined from GSP and CFC samples of all personal samples was 2.88. The individual values covered a range between 1 and 17 related to differences in size distributions of the Be-containing particulates. An experimental system was developed for the rapid measurement of the aspiration/transfer efficiency of aerosol samplers in a wind tunnel. We attempted to measure the aspiration and particle transfer characteristics of two inlets commonly used for sampling airborne Particulate Matter (PM): the 'Total Suspended Particulate' or TSP inlet, and the louvered 'dichotomous sampler inlet' typically used in sampling PM10 or PM2.5. We were able to determine the fraction of the external aerosol that enters the inlet and is transferred through it, and hence is available for collection by a filter, or further size fractionation into PM10 or PM2.5. This 'sampling efficiency' was analysed as a function of dimensionless aerodynamic parameters in order to understand the factors governing inlet performance. We found that for the louvered inlet the sampling efficiency increases as the external wind increases. Under all conditions expected in practical use the louvered inlet aspirates sufficient PM to allow either PM10 or PM2.5 to be selected downstream. The TSP inlet's sampling efficiency decreases with increasing external wind, and the TSP inlet is likely to under-sample the coarse end of the PM10 fraction at moderate and high external winds. As this inlet is generally not used with a downstream size fractionator, changes in sampling efficiency directly affect the measured aerosol concentration. We also investigated whether it is possible to dimensionally scale the PM inlets to operate at either higher or lower flow rates, while preserving the same sampling characteristics as the current full-scale, 16.67 L min(-1) versions. In the case of the louvered inlet, our results indicate that scaling to lower flow rates is possible; scaling to higher flow rates was not tested. For the TSP sampler, the sampling efficiency changes if the sampler is scaled to operate at smaller or larger flow rates, leading to unreliable performance.

The Respicon has been introduced as a sampler for health related measurements of airborne contaminants at workplaces. The instrument is aimed at simultaneous collection of three health related aerosol fractions: (a) the coarser inhalable fraction, defining the aerosol fraction that may enter the nose and mouth during breathing; (b) the intermediate thoracic fraction, defining the fraction that may penetrate beyond the larynx and so reach the lung; and (c) the finer respirable fraction, defining the fraction that may penetrate to gas exchange region of the lung. The instrument has a number of features attractive to occupational hygienists: in addition to providing the three aerosol fractions simultaneously, it is light and compact enough to be used as a personal sampler. yet can be a tripod mounted for area sampling, it can provide samples not only for gravimetric analysis but also microscopic and chemical analyses; and it is also available in a photometric direct-reading version. The instrument has previously been evaluated as an area sampler and, in this mode of operation, has shown reasonable accuracy in collecting respirable, thoracic and inhalable particles, the latter up to particle diameters of ca. 80 microm. Except for some scattered unpublished data there exist no systematic investigations in the Respicon's performance when used as a personal sampler in the industrial environment. In this paper, we will report on a study of side by side comparison of the Respicon with the IOM inhalable sampler, regarded as a reference instrument for the inhalable fraction. The main study was performed at six different workplaces in a nickel refinery. Statistical analysis of the gravimetrically-determined concentration data reveals consistently lower aerosol exposure values for the Respicon as compared to the IOM sampler. The data for the nickel workplaces are compared with findings from other studies. The results are interpreted in the light of the overall results and the possibility of introducing a correction factor is discussed.

The application of biocidal products by foam is considered an alternative to droplet spraying when disinfecting surfaces or fighting infestations. Inhalation exposure to aerosols containing the biocidal substances cannot be ruled out during foaming. In contrast to droplet spraying, very little is known about aerosol source strength during foaming. In this study, the formation of inhalable aerosols was quantified according to the aerosol release fractions of the active substance. The aerosol release fraction is defined as the mass of active substance transferred into inhalable airborne particles during foaming, normalised to the total amount of active substance released through the foam nozzle. Aerosol release fractions were measured in control chamber experiments where common foaming technologies were operated according to their typical conditions of use. These investigations include foams generated mechanically by actively mixing air with a foaming liquid as well as systems that use a blowing agent for foam formation. The values of the aerosol release fraction ranged from 3.4 × 10−6 to 5.7 × 10−3 (average values). For foaming processes based on mixing air and the foaming liquid, the release fractions could be correlated to the process and foam parameters such as foam exit velocity, nozzle dimensions, and foam expansion ratio. Inhalative exposure can occur accidentally when using cosmetic spray products. Usually, a tiered approach is applied for exposure assessment, starting with rather conservative, simplistic calculation models that may be improved with measured data and more refined modelling. Here we report on an advanced methodology to mimic in-use conditions for antiperspirant spray products to provide a more accurate estimate of the amount of aluminium possibly inhaled and taken up systemically, thus contributing to the overall body burden. Four typical products were sprayed onto a skin surrogate in defined rooms. For aluminium, size-related aerosol release fractions, i.e. inhalable, thoracic and respirable, were determined by a mass balance method taking droplet maturation into account. These data were included into a simple two-box exposure model, allowing calculation of the inhaled aluminium dose over 12 min. Systemic exposure doses were calculated for exposure of the deep lung and the upper respiratory tract using the Multiple Path Particle Deposition Model (MPPD) model. The total systemically available dose of aluminium was in all cases found to be less than 0.5 µg per application. With this study it could be demonstrated that refinement of the input data of the two-box exposure model with measured data of released airborne aluminium is a valuable approach to analyse the contribution of antiperspirant spray inhalation to total aluminium exposure as part of the overall risk assessment. A computer-controlled spray-generating and spray-exposure unit called RHINOCON was designed and constructed to offer the possibility of depositing, for example, environmental or pharmaceutical aerosols at the air/liquid interface under realistic exposure conditions. Here, isolated human respiratory cells are chosen to simulate, as far as possible, the in vivo situation. The exposure unit is equipped to hold up to 6 commercially available pump-spray dispensers, which are prepared mechanically for exposure as described in the manufacturer's users' guide. A transwell insert containing the cells is placed opposite each dispenser in a special plate head first. After starting the program, the spray dispensers are shaken and actuated automatically. The spray is released into an air flow passing over the cells, where the spray droplets are deposited due to their inertia. After exposure, the inserts can easily be removed and processed to measure a variety of cell toxicity parameters, such as cell viability or cell metabolism. Validation of the experimental setup was performed in an initial set of experiments: (1) by determination of the deposited amount of nebulized test substance on transwell membranes, and (2) by investigation of the cellular reactions of a human respiratory epithelial cell line (HFBE 21) exposed to nebulized hydrogen peroxide or to standard medium as a reference control. One of the uses of MDI is as an alternative to formaldehyde in the manufacture of furniture, its main route of exposure to humans being by inhalation. There have been no previous studies on the potential prenatal toxic effects of this compound. To close this gap in information, gravid Wistar rats, Crl:(WI)BR, were exposed by whole-body inhalation to clean air (control) and to 1, 3, and 9 mg/m3 MDI, respectively, for 6 hr per day from Days 6 to 15 post conception (p.c). Rats were killed on Day 20 p.c. and the following results were obtained: Treatment caused a dose-dependent decrease in food consumption in all substance-treated groups during exposure, returning to normal values after cessation of treatment. The lung weights in the high-dose group were significantly increased compared to the sham-treated control animals. Treatment did not influence any other maternal and/or fetal parameters investigated (maternal weight gain, number of corpora lutea, implantation sites, pre– and postimplantation loss, fetal and placental weights, gross and visceral anomalies, degree of ossification), although a slight but significant increase in litters with fetuses displaying asymmetric sternebra(e) was observed after treatment with the highest dose of 9 mg/m3. Although the relevance of an increase of this minor anomaly in doses which cause toxic effects in dams (reduced food consumption, increased lung weights) is limited and the number observed is within the limits of biological variability, a substance-induced effect in the high-dose group cannot be excluded with certainty. Consequently, a no embryotoxic effect level of 3 mg/m3 was determined. [ABSTRACT FROM AUTHOR]
