5-Aryl-4-acyl-3-hydroxy-1-(2,2-dimethoxyethyl)-3-pyrrolin-2-ones were synthesized by the reaction of acylpyruvic acid methyl esters with a mixture of an aromatic aldehyde and aminoacetaldehyde dimethylacetal. The proposed structures are confirmed by IR and PMR spectroscopy. The results of an investigation of the antibacterial and analgesic activity of the synthesized compounds are presented. A series of 4-acyl-5-aryl-3-hydroxy-1-(2-hydroxyethylaminoethyl)-3-pyrrolin-2-ones (I) and their hydrochlorides (II) were synthesized via reactions of methyl esters of acylpyruvic acids with a mixture of aromatic aldehyde and N-hydroxyethylethylenediamine. The antibacterial activity of compounds I and the influence of compounds I and II on blood coagulation were studied. All compounds demonstrate pronounced anticoagulant or hemostatic action. 1-(3-Aminopropyl)-5-aryl-4-acyl-3-hydroxy-3-pyrrolin-2-ones ( I–X) were prepared via reaction of methyl acylpyruvates and a mixture of aromatic aldehyde and 1,3-diaminopropane in a 1:1:1 molar ratio. The products were converted to hydrochlorides XI–XV in 48–90% yields. 1,3-Di(4-acetyl-3-hydroxy-2-oxo-5-phenyl-3-pyrrolin-1-yl)propane ( XVI) was produced in 16% yield via reaction of methyl acetylpyruvate with a mixture of benzaldehyde and 1,3-diaminopropane in a 2:2:1 molar ratio. A series of 2,4-dibenzyloxy(diallyloxy)carbonyl-3-aryl-5-hydroxy-5-methylcyclohexanones have been obtained via reactions of benzylacetoacetate and allylacetoacetate with aromatic aldehydes in the presence of a basic catalyst (piperidine). The proposed structures are confirmed by the results of 1H NMR spectroscopy, IR absorption spectroscopy, and high-resolution mass spectrometry measurements. Reaction of acetoacetic esters, aromatic aldehydes, and ammonia formed under Hantzsch conditions 2,6-dimethyl-3,5-dialkoxycarbonyl-1,4-dihydropyridines.
