The vast areas of permafrost peatlands in the West Siberian Lowland (WSL) hold a significant amount of carbon currently at risk due to human-caused global warming. The rapid thawing of about 20–60% of the frozen soils of Western Siberia adds to the destruction of the region's signature palsa landscape and the development of thermokarst lakes or fens. Due to the complex interactions between climate and the ecosystem that shape its formation and degradation, permafrost reacts in different ways to environmental changes. Understanding recent changes in Western Siberia requires knowledge of previous environmental transitions, which needs to be enhanced further. This study, aimed to create a hydrological testate amoebae (TA) transfer function (TF) from the Khanymei area in the WSL to reconstruct past hydrological conditions in permafrost peatlands. In addition, this research intended to examine the influence of TA communities living in lichens (genus Cladonia) on the performance of TF by integrating these data with a calibration data set. In the summer of 2019, 76 modern samples of Sphagnum , Cladonia , and vascular plants from permafrost and non-permafrost sites were collected. Moreover, one peat core was extracted from the same area to reconstruct the depth to water table (DWT) quantitatively. The Shannon's diversity index, a quantitative measure of species richness, was calculated for each sample to estimate the TA species diversity in a community. Non-metric multidimensional scaling ordination was applied to the dataset to explore the similarities in TA communities between samples and determine whether the DWT controls the TA community structure. Results showed that the typical transition of TA species along the microtopographic (wet–dry) gradient is influenced by the presence of lichens on the surface of the peatland. Statistical analysis confirmed that TA communities in Cladonia are less affected by DWT. Therefore, the TF was constructed on the entire dataset (n = 75) and the dataset with Cladonia excluded (n = 55). Both these TFs showed satisfactory results. Even though the TF constructed on the entire dataset showed fewer predictive abilities, it provided a complete picture of the current vegetation conditions of Khanymei peatlands, where lichens are an essential element of the ecosystem. In rich fens, unlike bogs, the key drivers structuring testate amoeba communities are related to nutrient status, suggesting the potential for transfer functions to quantitatively reconstruct changing nutrient status from palaeoecological records. Such records could be useful tools to investigate the long-term impacts of pollution and landscape change. Here, we derive and test transfer functions for pH, water-table depth, conductivity and Ca and Mg concentrations using a data set from Polish fens. Results show that transfer functions for Ca and conductivity have apparent predictive power for surface samples; these models will require further validation and testing with palaeoecological data. Testate amoeba transfer functions for fen nutrient status may be a valuable addition to the peatland palaeoecologist’s tool-kit, although further work will be required to demonstrate their usefulness in practice. Abstract: Among the non-pollen micro-fossils commonly encountered in Quaternary sediment samples prepared for pollen analysis are many shells of testate amoebae. Testate amoebae are eukaryotic micro-organisms which are increasingly used in ecological and palaeoecological studies, particularly as indicators of hydrological change in Sphagnum-dominated peatlands. In this study we address the extent to which testate amoebae are used in palynological research, the key challenges to more widespread use, and the extent to which ecological information is retained in the testate amoeba assemblages of standard palynological slides. To achieve this we review the literature on the use of testate amoebae in palynology, compare testate amoeba records produced by palynological and water-based preparation methods and carry out simulations using previously-derived datasets. Our results show that testate amoebae are widely encountered in Quaternary palynological studies, primarily in peatlands, but the information which they can provide is undermined by limited taxonomic knowledge. Many taxa are destroyed in pollen preparations, but for taxa that are retained patterns of abundance parallel those determined using water-based preparation methods. Although the loss of sensitive taxa limits the ecological information contained in testate amoeba assemblages the information preserved is likely to be useful in a multiproxy approach to palaeoenvironmental reconstruction. To help improve taxonomic awareness and encourage the use of testate amoebae in palynology we present a basic introduction to testate amoeba taxonomy and a guide to the taxonomic literature. In this study we assess the possible consequences of violation of this requirement by simulating taxonomic errors in testate amoeba data. Combinations of easily confused taxa were selected, and data manipulated to reflect confusion of these taxa; transfer functions based on unmodified data were then applied to these modified data sets. Initially these experiments were carried out one error at a time using four modern training sets; subsequently, multiple errors were separately simulated both in four modern training sets and in four palaeoecological data sets. Some plausible taxonomic confusions caused major biases in reconstructed values. In the case of two palaeoecological data sets, a single consistent taxonomic error was capable of changing the pattern of environmental reconstruction beyond all recognition, totally removing any real palaeoenvironmental signal. Tropical peatlands represent globally important carbon sinks with a unique biodiversity and are currently threatened by climate change and human activities. It is now imperative that proxy methods are developed to understand the ecohydrological dynamics of these systems and for testing peatland development models. Testate amoebae have been used as environmental indicators in ecological and palaeoecological studies of peatlands, primarily in ombrotrophic Sphagnum-dominated peatlands in the mid- and high-latitudes. We present the first ecological analysis of testate amoebae in a tropical peatland, a nutrient-poor domed bog in western (Peruvian) Amazonia. Litter samples were collected from different hydrological microforms (hummock to pool) along a transect from the edge to the interior of the peatland. We recorded 47 taxa from 21 genera. The most common taxa are Cryptodifflugia oviformis, Euglypha rotunda type, Phryganella acropodia, Pseudodifflugia fulva type and Trinema lineare. One species found only in the southern hemisphere, Argynnia spicata, is present. Arcella spp., Centropyxis aculeata and Lesqueresia spiralis are indicators of pools containing standing water. Canonical correspondence analysis and non-metric multidimensional scaling illustrate that water table depth is a significant control on the distribution of testate amoebae, similar to the results from mid- and high-latitude peatlands. A transfer function model for water table based on weighted averaging partial least-squares (WAPLS) regression is presented and performs well under cross-validation (r $^{2}_{apparent} \,=\, 0.76, \text {RMSE} \,=\, 4.29; \mathrm {r}^{2}_{jack} \,=\, 0.68, \text {RMSEP} \,=\, 5.18$). The transfer function was applied to a 1-m peat core, and sample-specific reconstruction errors were generated using bootstrapping. In this article, we examined the indicative value of a relationship between two non-pollen palynomorphs (NPPs), fungal HdV-10, related to the presence of Calluna vulgaris, and HdV-31A, which is testate amoeba – Archerella flavum. Both are frequently present on slides designated for pollen analysis, prepared from Sphagnum peat. We analysed three profiles from three extensive ombrotrophic peatlands in northern Poland, in which the content of testate amoebae (TA) was examined and TA-inferred depth to the water table (DWT) was reconstructed. The new analysis of palynological samples regarding NPPs revealed that strong increases in HdV-10 content were mostly simultaneous to HdV-31A declines. However, the relations between both types were not statistically significant. The rapid increases in HdV-10, despite the fact that this type of conidiospores is related to drier habitats on peatlands, were tentatively interpreted as an indicator of rapid rises in the water table level which, in consequence, might have stimulated the production of these spores by fungi. In addition, a negative correlation between HdV-31A and Arcella discoides and positive one, but weaker, in case of HdV-10 (attributed by some authors to species Trichocladium opacum (Corda) S Hughes) and A. discoides show a link between mutual fluctuations of both NPPs and hydrological instabilities on peatland. The Bagno Kusowo bog, the westernmost peatland subjected to the study, displayed an intriguing agreement between the presence of peat sections with strong increases in HdV-10 and cold climate events affecting Europe. Our study reveals that counting even a limited number of NPP types during the standard pollen analysis of Sphagnum peat may support the interpretation of results, especially, in cases when the investigation lacks testate amoeba analysis. The Central European area has been extensively studied using qualitative reconstruction techniques focusing on the development of peatlands in the past; however, research based on quantitative techniques is still lacking, especially in relation to highlands and mountainous areas. In this study, we focused on the hydrological changes and human-induced disturbances that affected two raised bogs from the Orawa-Nowy Targ Basin (Carpathian region): Puścizna Krauszowska and Puścizna Mała. We aimed to reconstruct the development of peatlands and changes in water table under different intensities of human activities. Two peat sequences from two different bogs, both possessing absolute chronologies based on high-resolution 14C dating, were examined in terms of pollen, non-pollen palynomorphs, plant macrofossils and testate amoeba records. We detected an asynchronous decrease in the water table level on the bogs that took place between the 4th and the 7th century ad, which occurred simultaneously with a global cold period ad 300–600 (Migration Period) in case of the Puścizna Mała bog. A concurrent but insignificant human impact on bogs was recorded. A distinct wet shift corresponding to the Wolf solar minimum (ca. ad 1280–1340) in Puścizna Mała was detected during ca. ad 1300–1350. The effect of this climatic event on bog hydrology is difficult to estimate because of the simultaneous human-induced deforestation. Drainage and substantial acceleration of peat extraction in the 19th and the 20th century ad led to the significant disturbances in peatland; however, these bogs were still susceptible to dry climatic events. We present a short note on the species composition of testate amoebae in a poor fen on mineral soil near the Pacific Coast in the hilly area of Central Honshu, Japan. In total 45 species and subspecific taxa belonged to 21 genera and 14 families of testate amoebae were recorded. Eight species and nine subspecies are newly recorded from Japan. However, most species from the list can be considered as distributed worldwide and associated mostly to oligotrophic/acid Sphagnum conditions. [ABSTRACT FROM AUTHOR]

This study explores the history of the development of Sphagnum communities in an ombrotrophic peatland – Bagno Kusowo – over the past 650 years, based on high-resolution plant macrofossil and testate amoebae analysis. Our research provided information related to the length of peatland existence and the characteristics of its natural/pristine state before the most recent human impacts. Changes in the Sphagnum communities before human impact could have resulted from climate cooling during the ‘Little Ice Age’ (LIA). In this cold and unstable hydrological period, among vascular plants, Eriophorum vaginatum and Baeothryon caespitosum dominated in the peatland vegetation. Peat-forming Sphagnum communities survived the drainage conducted during the 20th century at the Bagno Kusowo bog. We provide three important messages through this study: (1) testate amoebae reflect similar hydrological trends in two peat cores despite considerable microhabitat variability, (2) average long-term water level 10 cm below the surface should be a target for active bog conservation and (3) sites like Bagno Kusowo are extremely important to preserve the remains of pristine biodiversity (including genetic diversity of plants and protists) that was completely removed from most of the raised bogs in Europe due to human activities, for example, drainage. In the decade since the first pan-European testate amoeba-based transfer function for peatland palaeohydrological reconstruction was published, a vast amount of additional data collection has been undertaken by the research community. Here, we expand the pan-European dataset from 128 to 1799 samples, spanning 35° of latitude and 55° of longitude. After the development of a new taxonomic scheme to permit compilation of data from a wide range of contributors and the removal of samples with high pH values, we developed ecological transfer functions using a range of model types and a dataset of ∼1300 samples. We rigorously tested the efficacy of these models using both statistical validation and independent test sets with associated instrumental data. Model performance measured by statistical indicators was comparable to other published models. Comparison to test sets showed that taxonomic resolution did not impair model performance and that the new pan-European model can therefore be used as an effective tool for palaeohydrological reconstruction. Our results question the efficacy of relying on statistical validation of transfer functions alone and support a multi-faceted approach to the assessment of new models. We substantiated recent advice that model outputs should be standardised and presented as residual values in order to focus interpretation on secure directional shifts, avoiding potentially inaccurate conclusions relating to specific water-table depths. The extent and diversity of the dataset highlighted that, at the taxonomic resolution applied, a majority of taxa had broad geographic distributions, though some morphotypes appeared to have restricted ranges. Transfer functions are widely used in palaeoecology to infer past environmental conditions from fossil remains of many groups of organisms. In contrast to traditional training-set design with one observation per site, some training-sets, including those for peatland testate amoeba-hydrology transfer functions, have a clustered structure with many observations from each site. Here we show that this clustered design causes standard performance statistics to be overly optimistic. Model performance when applied to independent data sets is considerably weaker than suggested by statistical cross-validation. We discuss the reasons for these problems and describe leave-one-site-out cross-validation and the cluster bootstrap as appropriate methods for clustered training-sets. Using these methods we show that the performance of most testate amoeba-hydrology transfer functions is worse than previously assumed and reconstructions are more uncertain. [ABSTRACT FROM AUTHOR]

We investigated the relationships between testate amoebae (Arcellinida, Euglyphida), vegetation and water chemistry along environmental gradients in minerotrophic peatlands (fens) in western Poland. We hypothesized that: a) hydrochemistry significantly influences structure of testate amoeba communities, and b) testate amoeba communities are more closely correlated with the hydrochemical variables (environment) than with the vegetation data. Testate amoeba communities and vegetation from 71 sample plots were investigated together with the hydro-chemistry and hydrology based on 16 environmental variables and vegetation composition. Testate amoeba communities revealed a distinctive poor-rich gradient in analysed fens. Mineral-rich habitats, which were dominated by brown mosses, were preferred by a higher number of taxa than acidic habitats, which were dominated by Sphagnum. We recorded a total of 107 testate amoebae taxa. The average species richness of testate amoebae for brown mosses was higher (20) than for Sphagnum (13). We found that testate amoebae communities were similarly correlated with vascular plants, mosses and environmental parameters. Results of direct ordination demonstrate that hydrology, pH, Mg2+ and sodium remain the most important environmental control for the entire data set. CCA showed that in case of brown mosses hydrology, sodium and oxygen affect testate amoeba communities significantly whereas in Sphagnum only sodium emerge as most significant determining testate amoeba assemblages. Testate amoebae (TA) are a common and diverse group of protists and are especially abundant in peatlands. The structure of peatland TA communities is well correlated to surface moisture and water table depth (WTD). For that reason, TA are widely used as proxy indicators in ecological and palaeoecological studies. Peatlands are abundant across Asia, but the diversity and ecology of the TA that inhabit these systems are poorly documented. It is therefore unclear whether TA can be used as palaeohydrological indicators in the manner in which they commonly are in Europe and North-America. There is particular uncertainty as to the efficacy of this approach in the lower latitudes. We compiled existing and new data on testate amoebae from 1124 Sphagnum -dominated samples from 42 individual peatlands covering broad latitudinal (25°–66° N) and longitudinal (68°–129° E) ranges. Using a consensus taxonomic framework, we built a checklist of TA and developed TA-based hydrological transfer functions for Asian peatlands. The results showed that three models, weighted averaging (WA), weighted average partial least squares (WA-PLS), and maximum likelihood (ML), predicted similar WTD values for full samples, while the modern analogue technique (MAT) produced the strongest (R2 boot = 0.58) relationship between observed and estimated water-table depths (WTDs). Removing outlier samples improved the R2 values of observed vs. estimated WTDs, with ML then demonstrating the strongest predictive power (R2 boot = 0.68, RMSEP boot = 8.98 cm). The predictive capability of the developed WTD transfer function is comparable to equivalent models for Europe and North America and thus can be used for palaeohydrological reconstructions for boreal to subtropical peatlands in Asia. • A testate amoebae taxonomic checklist was compiled from Asian peatlands. • We developed continental scale testate amoeba-based hydrological transfer functions. • The water-table depth transfer functions can be used for paleohydrological reconstructions.
