Building an optical filtration function into a microfluidic chip is a promising way of simplifying the optical detection system of a microfluidic device. In this paper, a PDMS microfluidic chip filter that is capable of transmitting chlorophyll fluorescence and blocking interfering light in the visible wavelength range was developed for living algae detection with a smartphone. The chip was fabricated by sealing a layer of crystal violet solution in a PDMS layer on the top of the Sudan II-doped PDMS slab, which has a straight microchannel. Optimum dye concentrations and thicknesses for the crystal violet solution layer and Sudan II-doped PDMS slab were investigated and determined by spectrum measurements. It was found that the cut-on range of this integrated microfluidic chip is extended to about 625 nm and the transmittance in the chlorophyll fluorescence range (650 nm to 710 nm) is as high as 95%, when 25 mg L

A conductivity-difference-based method for increasing dielectrophoretic (DEP) force for particle separation in a microfluidic chip is presented in this paper. By applying a direct-current (DC) voltage across two immiscible electrolyte solutions with a conductivity difference, an enhanced electric field gradient is generated at the liquid-liquid interface. Theoretical analysis based on equivalent circuit theory found that the gradient of the electric field squared increases with the decrease in the conductivity ratio of the two liquids (main channel to the side channel). As a result, the particle separation distance (an indicator of DEP force) increases with the decrease in the conductivity ratio, which is both numerically predicted and experimentally verified. Numerical simulations also show that the separation distance increases with the increase in the magnitude of the electric field and the decrease in the width of the orifice. The method presented in this paper is simple and advantageous for increasing DEP force without applying higher DC voltages or fabricating smaller orifices.

Insulator-based dielectrophoretic (iDEP) microdevices have been limited to work with Newtonian fluids. We report an experimental study of the fluid rheological effects on iDEP focusing and trapping of polystyrene particles in polyethylene oxide, xanthan gum, and polyacrylamide solutions through a constricted microchannel. Particle focusing and trapping in the mildly viscoelastic polyethylene oxide solution are slightly weaker than in the Newtonian buffer. They are, however, significantly improved in the strongly viscoelastic and shear thinning polyacrylamide solution. These observed particle focusing behaviors exhibit a similar trend with respect to electric field, consistent with a revised theoretical analysis for iDEP focusing in non-Newtonian fluids. No apparent focusing of particles is achieved in the xanthan gum solution, though the iDEP trapping can take place under a much larger electric field than the other fluids. This is attributed to the strong shear thinning-induced influences on both the electroosmotic flow and electrokinetic/dielectrophoretic motions.

Insulator-based dielectrophoresis (iDEP) exploits the electric field gradients formed around insulating structures to manipulate particles for diverse microfluidic applications. Compared to the traditional electrode-based dielectrophoresis, iDEP microdevices have the advantages of easy fabrication, free of water electrolysis, and robust structure, etc. However, the presence of in-channel insulators may cause thermal effects because of the locally amplified Joule heating of the fluid. The resulting electrothermal flow circulations are exploited in this work to trap and concentrate nanoscale particles (of 100 nm diameter and less) in a ratchet-based iDEP microdevice. Such Joule heating-enabled electrothermal enrichment of nanoparticles are found to grow with the increase of alternating current or direct current electric field. It also becomes more effective for larger particles and in a microchannel with symmetric ratchets. Moreover, a depth-averaged numerical model is developed to understand and simulate the various parametric effects, which is found to predict the experimental observations with a good agreement.

Vortex formation near a two-part cylinder with zeta potentials of different values but the same sign under an external DC electric field is numerically investigated in this paper. The cylinder, inserted in a straight microchannel filled with an aqueous solution, is composed of an upstream part and a downstream part. When a DC electric field is applied in the channel, under certain conditions, the vortex will form near the cylinder due to the different velocities of electroosmotic flow generated on the cylinder surface. The numerical results reveal that the larger the velocity difference of electroosmotic flow generated on the two-part cylinder and the smaller the channel width, the more conducive to vortex formation in the channel. In addition, if the zeta potential ratios of cylinder downstream part to upstream part and channel wall to cylinder upstream part are unchanged, the DC electric field strength and the zeta potential value do not affect the pattern of vortices formed in the channel. This study provides a way for vortex formation in microchannels and has the potential application in microfluidic devices.

Real-time monitoring of large marine structures’ health, including drilling platforms, submarine pipelines, dams, and ship hulls, is greatly needed. Among the various kinds of monitoring methods, optical fiber sensors (OFS) have gained a lot of concerns and showed several distinct advantages, such as small size, high flexibility and durability, anti-electromagnetic interference, and high transmission rate. In this paper, three types of OFS used for marine structural health monitoring (SHM), including point sensing, quasi-distributed sensing, and distributed sensing, are reviewed. Emphases are given to the applicability of each type of the sensors by analyzing the operating principles and characteristics of the OFSs. The merits and demerits of different sensing schemes are discussed, as well as the challenges and future developments in OFSs for the marine SHM field.

Binding behaviors of proteins are important for applications in the field of biochemistry. Though a standard assay has a favorable limit of detection (LOD), it is mainly limited to indirect observation via fluorescence labeling. We reported and demonstrated a novel label-free sensing approach based on a near-infrared (NIR) surface plasmon resonance (SPR) sensing chip modified with a graphene-gold surface architecture in this paper. The NIR excitation wavelength can greatly improve the sensitivity of SPR sensing derived from the wavelength modulation-based methodology. Moreover, benefiting from the excellent electro-optical properties of graphene in NIR range, the graphene-gold surface architecture was built to further improve the sensing sensitivity. A fiber-optic biosensing platform based on ultra-narrowband cladding mode resonances was developed on a high-reflectivity fiber Bragg grating (FBG) for targeting biomolecular detection. The multiple cladding modes with a high sensitivity to the refractive index (RI) were excited in the FBG by coupling between the forward-propagating guided core mode of the multimode fiber and the backward-propagating guided cladding mode of the FBG without any damage to the fiber structure or any change to the standard FBG manufacturing process. The full width at half maximum and the Q-factor of the typical cladding mode resonance operation of the proposed sensor are 80 pm and 19,270, respectively, which are better than those of most fiber-optic biosensors reported to date. In addition, the FBG sensor demonstrated a high sensitivity in protein detection and a high selectivity in serum sample assays. The sensitivity of this sensor was further increased simply by coating it with graphene oxide (GO) sheets on the sensing surface without using a signal amplification strategy. Furthermore, an ultra-low limit of detection (LOD) of 32 pM was obtained by the GO-coated FBG sensor for IgG detection. The proposed FBG sensor provides a competitive fiber-optic platform for biomolecular detection. It has a great potential for applications in label-free biosensing.

Discharge mode is an important parameter for ozone synthesis by dielectric barrier discharge (DBD). Currently, it is still challenging to stably generate glow discharge with oxygen at atmospheric pressure. In this paper, a DBD reactor with a layer of silver placed between the electrode and the dielectric layer (SL-DBD) was developed. Experimental results show that both streamer and glow corona discharge were stably generated under sinusoidal excitation with a 0.5 mm discharge gap in a parallel-plate DBD, due to the increased electric field strength in the discharge gap by the silver layer. It was also found that, in the SL-DBD reactor, glow corona discharge enhances the discharge strength by 50 times. The spectral peak of O at 777 nm in SL-DBD is increased to 28,800, compared with 18,389 in a reactor with a streamer only. The SL-DBD reactor produces ozone with a concentration of as high as 150 g/m3 and shows good stability in an 8 h durability test.

Recent studies have demonstrated the strong influences of fluid rheological properties on insulator-based dielectrophoresis (iDEP) in single-constriction microchannels. However, it is yet to be understood how iDEP in non-Newtonian fluids depends on the geometry of insulating structures. We report in this work an experimental study of fluid rheological effects on streaming DEP in a post-array microchannel that presents multiple contractions and expansions. The iDEP focusing and trapping of particles in a viscoelastic polyethylene oxide solution are comparable to those in a Newtonian buffer, which is consistent with the observations in a single-constriction microchannel. Similarly, the insignificant iDEP effects in a shear thinning xanthan gum solution also agree with those in the single-constriction channel except that gel-like structures are observed to only form in the post-array microchannel under large DC electric fields. In contrast, the iDEP effects in a both viscoelastic and shear thinning polyacrylamide solution are significantly weaker than in the single-constriction channel. Moreover, instabilities occur in the electroosmotic flow and appear to be only dependent on the DC electric field. These phenomena may be associated with the dynamics of polymers as they are electrokinetically advected around and through the posts. This article is protected by copyright. All rights reserved.

This paper develops a numerical simulation model to research the deformable particle-particle interactions caused by dielectrophoresis (DEP) under AC electric fields. The DEP force is calculated by using Maxwell stress tensor method, and the hydrodynamic force is obtained by calculating the hydrodynamic stress tensor. Simulation results show that the DEP interactive motion will facilitate the particles forming particle chains that are parallel to the electric field, and the particles with low shear modulus present a lower x-component velocity. Also, the electric field intensity and particles radius have some effects on the DEP motions, and for different particles, smaller particles with larger electric field intensity easily reach a larger velocity. The numerical research may provide universal guidance for biological cells manipulation and assembly.

This paper presents the detection of nanoparticles and microorganisms using a recently developed side-orifice-based resistive pulse sensor (SO-RPS). By decreasing the channel height of the detection section of the SO-RPS, the detection sensitivity was increased and an average signal to noise ratio (S/N) of about 3 was achieved for 100 nm polystyrene particles. It was also found that spherical particles generate symmetrical signals. Algae with irregular shapes generate signals with more complex patterns. A scatter plot of signal magnitude versus signal width was proven to be reliable for differentiating bacteria from the nanoparticles and two types of algae. The side orifice for detecting heterogeneous nanoparticles and microorganisms is advantageous to avoid orifice clogging and the large flow resistance.

Flow-electrode capacitive deionization (FCDI) has gained lots of attention for the continuous operation, fast and scalable desalination process. However, the flow electrode slurry is poorly conductive and resulting in the high ions transport resistance, which restricts its desalination efficiency. In this work, a novel FCDI system was constructed by using the porous nickel foam (NFM) as the current collector, and its desalination performance was enhanced via the assistance of electro-catalytic water splitting. In detail, the functionalized current collectors (NFM-Co3O4 with enhanced oxygen evolution activity and NFM-Co(OH)2 with enhanced hydrogen evolution activity) were prepared through a two-step procedure of electrodeposition and subsequently anodic reaction/cathodic reaction. A desalination efficiency of 90.22% was obtained by the FCDI system adopted functionalized NFM (F-NFM-FCDI) after 100 min operation at applied voltage of 2.0 V, which was much higher than that of the FCDI system adopted bare NFM (NFM-FCDI, 65.37%). Moreover, F-NFM-FCDI system possessed higher charge efficiency (CE) of 81.24% and lower normalized energy consumption (NEC) of 237.54 J·mmol−1 under this condition. Mechanism analysis demonstrated that the enhancement on the desalination performance of F-NFM-FCDI system was attributed to the improved conductivity in flow-electrode chamber, which was due to the increase in the concentration of H+ and OH– caused by the enhanced electro-catalytic water splitting reaction during charging. In summary, this work provided a novel and facile strategy to improve the desalination performance of FCDI system by coupling with electro-catalytic water splitting.

Electrokinetic movement of spherical polystyrene particles at different liquid-fluid interfaces was experimentally investigated in this paper. A novel method was developed to place the particles rightly at the interfaces formed in a large plastic container. The velocity was measured by an optical microscope. The experimental results show that the particles (3, 5 and 10 μm in diameter) at the water-air interface, water-dodecane interface and NaCl solution-air interface move in the opposite direction of the applied electric field. The magnitude of the particles’ velocity increases linearly with the increase in the applied electric field. Moreover, for particles of the same size, the electrokinetic velocity at the liquid-fluid interfaces is larger than particles’ electrophoretic velocity in the bulk liquid phase. Under the same electric field, however, the electrokinetic velocity of smaller particles at the liquid-fluid interfaces is larger than that of larger particles. Such results are attributed to the surface charges at the liquid-fluid interface and the particle-liquid interface.

Dielectrophoresis (DEP) is a non-destructive, accurate, and label-free cell manipulating technique and DEP applications have been found in various fields. Assessment of cell viability is one of the important applications and many investigations have been reported. In this paper, cell polarization and its modeling, some key parameters employed for living/dead cell separation, as well as electrode configurations are reviewed. Focus is given to the latest development of DEP devices employed for the assessment of cell viability. Experimentally determined factors for separating living/dead cells, such as the conductivity of suspending medium and the frequency of applied electric field, are summarized. The future directions and potential challenges in this field are also outlined.

Controlling zeta potential of PDMS surface coated with a layer of PEG is important for electroosmosis and electrophoresis in PDMS made microfluidic chips. Here, zeta potentials of PDMS surfaces modified by simple physisorption of PEG of different concentrations in phosphate buffer solutions, pure water, and PEG solution were reported. Coating PEG on PDMS surfaces was achieved by immersing a PDMS layer into the PEG solution for 10 min and then taking it out and placing it in an oven at 80℃ for 10 h. To avoid damaging the PEG layer on the PDMS surface, an induction current method was employed for zeta potential measurement. Zeta potentials of PEG modified PDMS in electrolyte solutions were measured. The results show that 2.5% PEG can effectively modify PDMS surface with positive zeta potential value in phosphate buffer solutions, pure water and 10% PEG solution. Further increase in PEG solution beyond 5% for surface modification has no obvious effect on zeta potential change.

The separation of particles and cells in a uniform mixture has been extensively studied as a necessity in many chemical and biomedical engineering and research fields. This work demonstrates a continuous charge-based separation of fluorescent and plain spherical polystyrene particles with comparable sizes in a ψ-shaped microchannel via the wall-induced electrical lift. The effects of both the direct current electric field in the main-branch and the electric field ratio in between the inlet branches for sheath fluid and particle mixture are investigated on this electrokinetic particle separation. A Lagrangian tracking method based theoretical model is also developed to understand the particle transport in the microchannel and simulate the parametric effects on particle separation. Moreover, the demonstrated charge-based separation is applied to a mixture of yeast cells and polystyrene particles with similar sizes. Good separation efficiency and purity are achieved for both the cells and the particles.

A novel method of sheathless particle focusing by induced charge electrokinetic flow in a microchannel is presented in this paper. By placing a pair of metal plates on the opposite walls of the channel and applying an electrical field, particle focusing is achieved due to the two pairs of vortex that constrain the flow of the particle solution. As an example, the trajectories of particles under different electrical fields with only one metal plate on one side channel wall were numerically simulated and experimentally validated. Other flow focusing effects, such as the focused width ratio (focused width/channel width) and length ratio (focused length/half-length of metal plate) of the sample solution, were also numerically studied. The results show that the particle firstly passes through the gaps between the upstream vortices and the channel walls. Afterwards, the particle is focused to pass through the gap between the two downstream vortices that determine the focused particle position. Numerical simulations show that the focused particle stream becomes thin with the increases in the applied electrical field and the length of the metal plates. As regards to the focused length ratio of the focused stream, however, it slightly increases with the increase in the applied electrical field and almost keeps constant with the increase in the length of the metal plate. The size of the focused sample solution, therefore, can be easily adjusted by controlling the applied electrical field and the sizes of the metal plates.

Traditionally, a resistive pulse sensor (also known as Coulter counter) works by letting a particle pass through a small orifice in an electrolyte solution. The detection sensitivity mainly relies on the volume ratio of the particle to the orifice. This paper presents a novel resistive pulse sensor which has a sensing orifice located on the side wall of a microchannel. In this way, the sensor can detect and count particles (or cells) without requiring particles (or cells) passing through the sensing gate. An equation was derived to relate the magnitudes of the detected signals and the electrical resistances. Results show that the magnitudes of the detected signals can be increased by applying voltages from more than one voltage input channels simultaneously. Under the same conditions, the magnitudes of the detected signals become larger when the diameters of particles are larger. Higher detection sensitivity can be obtained simply by increasing either the magnitudes of the applied voltages or the number of the voltage input channels, or reducing the opening of the side sensing gate to a size that is even smaller than the diameter of the particle. Due to the high detection sensitivity, detection of 1 μm particles by a relatively large sensing gate of 5 × 10 × 10 μm (width × length × height) was successfully demonstrated with a signal to noise ratio (S/N) of approximately 3. This sensor was also applied to detect and count human red blood cells and lymphocyte cells. Results show that this method can clearly distinguish the cells with different sizes based on the pre-determined-thresholds. Because this sensor does not require cells to pass through the sensing gate, the channel clogging problem can be avoided. More importantly, the detection sensitivity can be tuned by applying different voltages without fabricating a smaller sensing gate.

Electrokinetic motion of a micro oil droplet beneath a glass slide was experimentally investigated in this paper. The micro oil droplets were released under the glass slide in an aqueous solution and the motion along the glass slide was measured by a microscope. The experimental results indicate that while the electrokinetic mobility increases with the applied electric field, it decreases with the oil droplet size and the ionic concentration of the aqueous solution, respectively. By changing the zeta potential of the glass-liquid interface using polybrene coating from negative to positive, the direction of the electrokinetic mobility is reversed and the absolute value of the electrokinetic mobility increases significantly. Finally, pH effects were also investigated, and it was found that the electrokinetic mobility of the droplets reaches a maximum at pH = 6∼8.

A novel microfluidic method of continually detecting and counting beads-labeled cells from a cell mixture without fluorescence labeling was presented in this paper. The detection system is composed of a microfluidic chip (with a permanent magnet inserted along the channel), a signal amplification circuit, and a LabView® based data acquisition device. The microfluidic chip can be functionally divided into separation zone and detection zone. By flowing the pre-labeled sample solution, the target cells will be sequentially separated at the separation zone by the permanent magnet and detected and counted at the detection zone by a microfluidic resistive pulse sensor. Experiments of positive separation and detection of T-lymphocytes and negative separation and detection of cancer cells from the whole blood samples were carried out to demonstrate the effectiveness of this method. The methodology of utilizing size difference between magnetic beads and cell-magnetic beads complex for beads-labeled cell detection is simple, automatic, and particularly suitable for beads-based immunoassay without using fluorescence labeling.

Previous studies have reported a lateral migration in particle electrophoresis through a straight rectangular microchannel. This phenomenon arises from the inherent wall-induced electrical lift that can be exploited to focus and separate particles for microfluidic applications. Such a dielectrophoretic-like force has been recently found to vary with the buffer concentration. We demonstrate in this work that the particle zeta potential also has a significant effect on the wall-induced electrical lift. We perform an experimental study of the lateral migration of equal-sized polystyrene particles with varying surface charges under identical electrokinetic flow conditions. Surprisingly, an enhanced focusing is observed for particles with a faster electrokinetic motion, which indicates a substantially larger electrical lift for particles with a smaller zeta potential. We speculate this phenomenon may be correlated with the particle surface conduction that is a strong function of particle and fluid properties.

Zeta potential is an important parameter for characterizing the electrokinetic properties of a solid-liquid interface. In this paper, zeta potentials of polydimethylsiloxane surfaces modified by polybrene (PB) solutions of different concentrations in Phosphate buffer solution and pure water were reported. The zeta potentials were measured by an induction current method. The measurements were validated both by a calibration curve based on the data reported in the published papers and by comparing the zeta potential determined by using the Smoluchowski equation and the measured velocity of the electrokinetic motion of particles in a microchannel.

Electrophoretic mobility of oil droplets of micron sizes in PBS and ionic surfactant solutions was measured in this paper. The experimental results show that, in addition to the applied electric field, the speed and the direction of electrophoretic motion of oil droplets depend on the surfactant concentration and on if the droplet is in negatively charged SDS solutions or in positively charged hexadecyltrimethylammonium bromide (CTAB) solutions. The absolute value of the electrophoretic mobility increases with increased surfactant concentration before the surfactant concentration reaches to the CMC. It was also found that there are two vortices around the oil droplet under the applied electric field. The size of the vortices changes with the surfactant and with the electric field. The vortices around the droplet directly affect the drag of the flow field to the droplet motion and should be considered in the studies of electrophoretic mobility of oil droplets. The existence of the vortices will also influence the determination and the interpretation of the zeta potential of the oil droplets based on the measured mobility data.

A novel and simple method of determining both the critical micelle concentration (CMC) and polarities of ionic surfactants at the same time is presented in this paper. This method is based on the dependence of electrical potential at the liquid-air interface on the aggregation of the surfactant. When the measuring electrode contacts the liquid-air interface, the difference between the electrical potential of the electrode surface and the electrical potential of the liquid-air interface generates an electrical current through an electrical circuit. The dependences of the magnitudes of the measured signals on the concentrations of sodium dodecyl sulfate and hexadecyltrimethylammonium bromide were measured. The results show that the magnitude of the measured signal increases with the increasing surfactant concentration before it reaches to the CMC. The signal magnitude remains constant after the CMC. The value of CMC can be determined from the intersection point of the two lines before and after CMC. The CMC values calculated with this method are in good agreement with those reported in the reference papers. The experimental results also show that the directions of the signals generated by anionic and cationic surfactants are in opposite direction, which can be used to determine the polarities of the surfactants. The method described in this paper provides a simple and reliable tool to determine the CMC value and the polarity of ionic surfactants.

High-throughput particle counting by a differential resistive pulse sensing method in a microfluidic chip is presented in this paper. A sensitive differential microfluidic sensor with multiple detecting channels and one common reference channel was devised. To test the particle counting performance of this chip, an experimental system which consists of the microfluidic chip, electric resistors, an amplification circuit, a LabView based data acquisition device was developed. The influence of the common reference channel on the S/N of particle detection was investigated. The relationship between the hydraulic pressure drop applied across the detecting channel and the counting throughput was experimentally obtained. The experimental results show that the reference channel designed in this work can improve the S/N by ten times, thus enabling sensitive high-throughput particle counting. Because of the greatly improved S/N, the sensing gate with a size of 25 × 50 × 10 μm (W × L × H) in our chips can detect and count particles larger than 1.5 μm in diameter. The counting throughput increases with the increase in the flowing velocity of the sample solution. An average throughput of 7140/min under a flow rate of 10 μL/min was achieved. Comparing with other methods, the structure of the chip and particle detecting mechanism reported in this paper is simple and sensitive, and does not have the crosstalking problem. Counting throughput can be adjusted simply by changing the number of the detecting channels.

Preconcentrating samples of dilute particles or cells to a detectable level is required in many chemical, environmental and biomedical applications. A variety of force fields have thus far been demonstrated to capture and accumulate particles and cells in microfluidic devices, which, however, all take place within the region of microchannels and may potentially cause channel clogging. This work presents a new method for the electrokinetic preconcentration of 1 μm-diameter polystyrene particles and E. coli cells in a very-low-conductivity medium inside a microfluidic reservoir. The entire microchannel can hence be saved for a post-concentration analysis. This method exploits the strong recirculating flows of induced-charge electroosmosis to concentrate particles and cells near the corners of the reservoir-microchannel interface. Positive dielectrophoresis is found to also play a role when small microchannels are used at high electric fields. Such an in-reservoir electrokinetic preconcentration method can be easily implemented in a parallel mode to increase the flow throughput, which may potentially be used to preconcentrate bacterial pathogens in water.

The electrokinetic motion of a negatively charged spherical particle at an oil-water interface in a microchannel is numerically investigated and analyzed in this paper. A three-dimensional (3D) transient numerical model is developed to simulate the particle electrokinetic motion. The channel wall, the surface of the particle and the oil-water interface are all considered negatively charged. The effects of the direct current (DC) electric field, the zeta potentials of the particle-water interface and the oil-water interface, and the dynamic viscosity ratio of oil to water on the velocity of the particle are studied in this paper. In addition, the influences of the particle size are also discussed. The simulation results show that the micro-particle with a small value of negative zeta potential moves in the same direction of the external electric field. However, if the zeta potential value of the particle-water interface is large enough, the moving direction of the particle is opposite to that of the electric field. The velocity of the particle at the interface increases with the increase in the electric field strength and the particle size, but decreases with the increase in the dynamic viscosity ratio of oil to water, and the absolute value of the negative zeta potentials of both the particle-water interface and the oil-water interface. This work is the first numerical study of the electrokinetic motion of a charged particle at an oil-water interface in a microchannel.

An electrokinetic microfluidic chip is developed to detect and sort target cells by size from human blood samples. Target-cell detection is achieved by a differential resistive pulse sensor (RPS) based on the size difference between the target cell and other cells. Once a target cell is detected, the detected RPS signal will automatically actuate an electromagnetic pump built in a microchannel to push the target cell into a collecting channel. This method was applied to automatically detect and sort A549 cells and T-lymphocytes from a peripheral fingertip blood sample. The viability of A549 cells sorted in the collecting well was verified by Hoechst33342 and propidium iodide staining. The results show that as many as 100 target cells per minute can be sorted out from the sample solution and thus is particularly suitable for sorting very rare target cells, such as circulating tumor cells. The actuation of the electromagnetic valve has no influence on RPS cell detection and the consequent cell-sorting process. The viability of the collected A549 cell is not impacted by the applied electric field when the cell passes the RPS detection area. The device described in this article is simple, automatic, and label-free and has wide applications in size-based rare target cell sorting for medical diagnostics.

Capacitive detection of living microalgae from the sample suspension in a microfluidic chip is presented in this paper. A three-dimension capacitance sensor was designed by placing two cylindrical copper wires across the microfluidic channel. The frequency-dependent capacitive changes for both living and dead Dunaliella Salina were analyzed with Maxwell mixture theory and measured experimentally in this study. The results show that the amplitude of capacitive change is mainly influenced by algae's size, living status and the applied alternative current frequency. Specifically, capacitive change increases with the increase in algae size and decreases with the increase in frequency. For the same kind of algae, the capacitance change caused by the living algal, however, is larger than that of the dead one under the same frequency, which can be used to differentiate the living algae from the sample suspension. The method of frequency-dependent capacitive detection of live algae described in this paper is simple and sensitive. The experimentally determined frequency window can serve as a guideline to differentiate the living algal from the dead one.

Evaluation of radiation dose is very important for the detection of radiation damage. γ-H2AX is a popular biological dosimeter to evaluate the radiation effect. Typically, bulky and expensive commercial flow cytometers are used to detect γ-H2AX. This paper presents a miniaturized and high sensitive cytometer using a microfluidic chip for evaluating the radiation dose by detecting the mean immunofluorescence intensity of γ-H2AX. A compact optical focusing system and a shift-phase differential amplifier are designed to improve the detection sensitivity. Sample lymphocyte cells are stained by FITC fluorescent dye after being irradiated by UVC. Comparison experiments between the developed miniature cytometer and a commercial flow cytometer were conducted under different radiation doses. The developed microfluidic cytometer also demonstrates a good linear correlation between the measured fluorescence intensity and the irradiation dose with a detection limit similar to that of the commercial flow cytometer. The developed cytometer can evaluate quantitatively the radiation dose by the mean fluorescence intensity of γ-H2AX with a significantly smaller amount of blood samples than a commercial flow cytometer.

This paper reports a novel and very simple method for measuring the zeta potential of electrolyte solution–air interface. When a measuring electrode contacts the electrolyte solution–air interface, an electrical current will be generated due to the potential difference between the electrode–air surface and the electrolyte solution–air interface. The amplitude of the measured electric signal is linearly proportional to this potential difference; and depends only on the zeta potential at the electrolyte solution–air interface, regardless of the types and concentrations of the electrolyte. A correlation between the zeta potential and the measured voltage signal is obtained based on the experimental data. Using this equation, the zeta potential of any electrolyte solution–air interface can be evaluated quickly and easily by inserting an electrode through the electrolyte solution–air interface and measuring the electrical signal amplitude. This method was verified by comparing the obtained results of NaCl, MgCl2 and CaCl2 solutions of different pH values and concentrations with the zeta potential data reported in the published journal papers.

Sweat test based on chloride ion selective electrode is the common method for the diagnosis of Cystic Fibrosis (CF). In our previous study, the possibility of using chloride ion sensitive fluorescent dye to determine the chloride ion concentration with a small volume of sample has been explored. However, we found the conventional Stern–Volmer Equation is applicable only for large systems such as a fluorescent microscope with a large volume of sample and dye solution; and it is not valid for portable devices using a small photo detector and a small volume of sample and dye solution. Therefore, in this paper, extensive experiments were performed by using a miniature fluorescent detection system; the effects of the parameters including the concentration and the volume of fluorescent dye solution, the coefficient of Stern–Volmer plot, and the volume ratio of the fluorescent dye solution and the chloride solution were studied; and a modified Stern–Volmer Equation was obtained. Using the modified Stern–Volmer correlation, the miniature fluorescent detection device can detect the chloride ion concentration correctly and rapidly.

This paper reports a lab-on-a-chip device that can automatically detect and sort particles based on their size differences with a high resolution. The PDMS-glass microfluidic chip is made by soft-lithography technique. A differential resistive pulse sensor is employed to electrically detect the sizes of the particles in EOF generated by applying DC voltages across channels. The detected resistive pulse sensor signals, whose amplitudes are proportional to particles' sizes, will automatically trigger the sorting process that is controlled by applying a voltage pulse (36 V) whenever a target particle is detected. This method was applied to automatically detect and sort polystyrene particles and microalgae in aqueous solutions. Sorting 5 μm polymer particle from a mixture of 4- and 5-μm polystyrene particles in aqueous solution, i.e. 1 μm sorting resolution, was demonstrated. The device described in this paper is simple, automatic, and label-free with high sorting resolution. It has wide applications in sample pretreatment and target particles detection.

A novel and simple method of improving the particle detection sensitivity of a microfluidic resistive pulse sensor was presented in this paper. This novel electrokinetic flow focusing method utilizes a focusing solution (with high resistivity) flowing from the upstream focusing channel to the downstream focusing channel. The focusing solution in the sensing gate works like a virtual insulation wall that greatly narrows the gate and thus improves the detection sensitivity. An equation was derived to relate the magnitude of the output signal to the resistivity and the width of the focusing solution. The width of the focused particle solution under different voltages was numerically predicted. The results show that the magnitude of output signal increases with the decrease in the width of the focused particle stream. More importantly, the detection sensitivity can be improved by decreasing the space occupied by the focusing solution in the upstream and downstream channels as much as possible. Detection of 1 μm particle with a sensing gate of 30 × 40 × 10 μm (width × length × height) was successfully achieved. The proposed method is simple and advantageous in detecting smaller particles without fabricating a small sensing gate.

Electroosmotic flow is the transport method of choice in microfluidic devices over traditional pressure-driven flow. To date, however, studies on electroosmotic flow have been almost entirely limited to inside microchannels. This work presents the first experimental study of Joule heating effects on electroosmotic fluid entry from the inlet reservoir (i.e., the well that supplies fluids and samples) to the microchannel in a polymer-based microfluidic chip. Electrothermal fluid circulations are observed at the reservoir-microchannel junction, which grow in size and strength with the increasing alternating current to direct current voltage ratio. Moreover, a 2D depth-averaged numerical model is developed to understand the effects of Joule heating on fluid temperature and flow fields in electrokinetic microfluidic chips. This model overcomes the problems encountered in previous unrealistic 2D and costly 3D models, and is able to predict the observed electroosmotic entry flow patterns with a good agreement.

This paper reports a microfluidic method of continuous separation of marine algae and particles by DC dielectrophoresis. The locally non-uniform electric field is generated by an insulating PDMS triangle hurdle fabricated within a PDMS microchannel. Both the particles and algae are subject to negative DEP forces at the hurdle where the gradient of local electric-field strength is the strongest. The DEP force acting on the particle or the algae depends on particles’ or algae’s volume, shape and dielectric properties. Thus the moving particles and algae will be repelled to different streamlines when passing the hurdle. In this way, combined with the electroosmotic flow, continuous separation of algae of two different sizes, and continuous separation of polystyrene particles and algae with similar volume but different shape were achieved. This first demonstration of DC DEP separation of polystyrene particles and algae with similar sizes illustrates the great influence of dielectric properties on particle separation and potentials for sample pretreatment.

By incorporating a viscous liquid force model into DEM simulations, the effect of liquid bridging between two tablets during a collision was studied. It was shown that when compared to the viscous force, the capillary force is several orders of magnitude smaller. In order to predict the capture of a falling tablet by the liquid bridge, it was necessary to adjust the limiting separation distance used in the viscous force model. Comparison of experimental results with the simulations, using oils with viscosities in the range of 100–1,000 cP, showed that there was a linear relationship between the limiting separation distance and the viscosity of the silicone oils. This viscosity-dependent separation distance varied in the range of 1–6 mm.

A novel method for measuring the zeta potential of PDMS-electrolyte solution interface is presented in this paper. When an electrolyte solution passes the electrode coated with PDMS film, an electrical current will be induced due to the electrostatic potential difference between the PDMS-air interface and the PDMS-electrolyte solution interface. Based on this principle, the relationship between the measured electrical signal and the zeta potential of PDMS-electrolyte solution interface is experimentally investigated. The results show that the magnitude of the measured signal is linearly proportional to this potential difference. An empirical correlation between the zeta potential and the measured voltage signal was obtained. Good agreement was found when comparing the zeta potential calculated by this empirical equation with that reported in the published journal papers. The zeta potential measurement method presented in this paper is simple and accurate and can be used for measuring zeta potentials of other dielectric-electrolyte interfaces.

Detection of living microalgae cells is very important for ballast water treatment and analysis. Chlorophyll fluorescence is an indicator of photosynthetic activity and hence the living status of plant cells. In this paper, we developed a novel microfluidic biosensor system that can quickly and accurately detect the viability of single microalgae cells based on chlorophyll fluorescence. The system is composed of a laser diode as an excitation light source, a photodiode detector, a signal analysis circuit, and a microfluidic chip as a microalgae cell transportation platform. To demonstrate the utility of this system, six different living and dead algae samples (Karenia mikimotoi Hansen, Chlorella vulgaris, Nitzschia closterium, Platymonas subcordiformis, Pyramidomonas delicatula and Dunaliella salina) were tested. The developed biosensor can distinguish clearly between the living microalgae cells and the dead microalgae cells. The smallest microalgae cells that can be detected by using this biosensor are 3 μm ones. Even smaller microalgae cells could be detected by increasing the excitation light power. The developed microfluidic biosensor has great potential for in situ ballast water analysis.

This paper reports a lab-on-a-chip device that counts the number of bacteria flowing through a microchannel. The bacteria number counting is realized by a microfluidic differential Resistive Pulse Sensor (RPS). By using a single microfluidic channel with two detecting arm channels placed at the two ends of the sensing section, the microfluidic differential RPS can achieve a high signal-to-noise ratio. This method is applied to detect and count bacteria in aqueous solution. The detected RPS signals amplitude for Pseudomonas aeruginosa ranges from 0.05 V to 0.17 V and the signal-to-noise ratio is 5-17. The number rate of the bacteria flowing through the sensing gate per minute is a linear function of the sample concentration. Using this experimentally obtained correlation curve, the concentration of bacteria in the sample solution can be evaluated within several minutes by measuring the number rate of the bacteria flowing through the sensing gate of this microfluidic differential RPS chip. The method described in this paper is simple and automatic, and have wide applications in determining the bacteria and cell concentrations for microbiological and other biological applications.

Trapping and preconcentrating particles and cells for enhanced detection and analysis are often essential in many chemical and biological applications. Existing methods for diamagnetic particle trapping require the placement of one or multiple pairs of magnets nearby the particle flowing channel. The strong attractive or repulsive force between the magnets makes it difficult to align and place them close enough to the channel, which not only complicates the device fabrication but also restricts the particle trapping performance. This work demonstrates for the first time the use of a single permanent magnet to simultaneously trap diamagnetic and magnetic particles in ferrofluid flows through a T-shaped microchannel. The two types of particles are preconcentrated to distinct locations of the T-junction due to the induced negative and positive magnetophoretic motions, respectively. Moreover, they can be sequentially released from their respective trapping spots by simply increasing the ferrofluid flow rate. In addition, a three-dimensional numerical model is developed, which predicts with a reasonable agreement the trajectories of diamagnetic and magnetic particles as well as the buildup of ferrofluid nanoparticles.
