In oocyte cryopreservation programs, vitrification has overthrown conventional slow freezing both in veterinary and human medicine. In animals, its feasibility in field conditions makes it the preferred technique for the safeguard of genetic resources from zoo or wild animals, including threatened felids, for which the domestic cat is an excellent model. However, many cellular injuries, such as cytoskeleton, mitochondria and meiotic spindle alterations, DNA damage, zona pellucida hardening and cumulus cell loss, might occur following vitrification. After warming, although the exact mechanisms are still unclear, degeneration is a frequent outcome for cat vitrified oocytes. For immature (germinal vesicle) gametes, in vitro maturation after warming is a challenge, and cleavage after fertilization barely reaches 15-30%, while for mature (metaphase II) cryopreserved gametes it can get to 30-50%. Anyway, the progression to late embryos stages is often impaired, and improvements are needed. Standard cryopreservation protocol and the use of conventional in vitro culture systems after warming may not be enough for vitrified oocytes to recover and demonstrate their full developmental potential. Physical or chemical factors applied to oocytes undergoing vitrification, as an enrichment to the vitrification step, or to the culture microenvironment, could create more favorable conditions and promote vitrified oocyte survival and development. From the use of three-dimensional culture systems to the regulation of metabolic activities and cellular pathways, this review aims to explore all the possibilities employed so far, including the studies performed by our own lab, and the future perspectives, to present the most effective strategies for cat oocyte vitrification and the best time for their application (i.e., before, during, or after vitrification-warming).

An egg of the Critically Endangered flapper skate Dipturus intermedius was successfully incubated to hatching in captivity, in what is believed to be a first for the species. Water conditions (temperature, salinity, flow rate) were recorded, with mean water temperatures ranging from a monthly mean of 8.3±1.2 to 13.2±0.3oC and salinity from a monthly mean of 30.5±1.2 to 36.6±2.3 ppt. Hatching occurred after 534 days, suggesting that flapper skate eggs take approximately 5700 growing degree-days to incubate to hatching. The egg's prolonged embryonic development raises concerns about flapper skate eggs' vulnerability to anthropogenic disturbance. Publisher PDF

Beta‐microseminoproteins (MSMBs) are small disulfide‐rich proteins that are conserved among vertebrates. These proteins exhibit diverse biological activities and were mainly reported to play a role in male fertility, immunity, and embryogenesis. In this work, we focused on the chicken MSMB3 protein that was previously depicted as an egg antibacterial protein. We report that MSMB3 protein is exclusively expressed in the reproductive tissues of laying hens (in contrast to chicken MSMB1 and MSMB2 paralogs), to be incorporated in the egg white during the process of egg formation. We also showed that chicken MSMB3 possesses highly conserved orthologs in bird species, including Neognathae and Palaeognathae. Chicken MSMB3 was purified from egg white using heparin affinity chromatography and was analyzed by top‐down and bottom‐up proteomics. Several proteoforms could be characterized, and a homodimer was further evidenced by NMR spectroscopy. The X‐ray structure of chicken MSMB3 was solved for the first time, revealing that this protein adopts a novel dimeric arrangement. The highly cationic MSMB3 protein exhibits a distinct electrostatic distribution compared with chicken MSMB1 and MSMB2 structural models, and with published mammalian MSMB structures. The specific incorporation of MSMB3 paralog in the egg, and its phylogenetic conservation in birds together with its peculiar homodimer arrangement and physicochemical properties, suggests that the MSMB3 protein has evolved to play a critical role during the embryonic development of avian species. These new data are likely to stimulate research to elucidate the structure/function relationships of MSMB paralogs and orthologs in the animal kingdom., Beta‐microseminoproteins (MSMBs) are small proteins conserved among vertebrates. The chicken genome contains three MSMB paralogs, which exhibit distinct tissue expression profiles, with chicken beta‐MSMB 3 (MSMB3) protein being egg‐specific. Collectively, results from protein sequence analysis, mass spectrometry analysis, and 3D structure data highlight that chicken MSMB3 possesses specific features that may be related to its presumed role in avian reproduction. ​

Microbes have central roles in the development and health of animals, being the introduction of specific microbial species a potential conservation strategy to protect animals from emerging diseases. Thus, insight into the microbiota of the species and their habitats is essential. In this manuscript, we report for the first time the bacterial composition of all the components (eggshells of hatched and unhatched eggs, internal content of unhatched eggs, intestinal content of hatchling and pipping sea turtles, and sand) of three nesting beaches of Caretta caretta along the Italian coasts of the Mediterranean Sea. The analysis of 26 amplicon samples was carried out using next-generation sequencing analysis, targeting V3–V4 regions of the bacterial 16S rRNA gene. Samples featured mainly Proteobacteria, Actinobacteria, Firmicutes, and Bacteroidetes, whose percentages depended on the sample type. Our results showed that, although from different sampling sites, the internal content of the unhatched eggs, intestinal content of hatchling and pipping sea turtles share the microbiota, which was yet different from that of eggshells and sand of the same nesting beach. This study suggests the maternal and environmental influence alongside a protective role of eggshells in shaping the egg microbiota of Caretta caretta sea turtles.

Introduction: Oral immunotherapy (OIT) is effective at inducing desensitization in food-allergic individuals, and is a valid therapeutic option for those allergic to peanut, cow’s milk and egg. However, there is a high rate of dose-related adverse events, and at least one fatality to OIT has been reported. Areas covered: We provide an update on the broader framework of issues which will impact on the availability and uptake of OIT. Expert opinion: The need for standardized products remains controversial. A licensed product exists for peanut-OIT, but OIT can also be safely achieved using peanut-containing foods at much lower cost. For other allergens, OIT can only be done with non-pharma products – something which has been done safely for over 2 decades. There is a need to develop personalized protocols for OIT, particularly for the 20% of patients unable to tolerate standard OIT. Cost-effectiveness is dependent on improved quality of life, but evidence for this is currently lacking, and is a key evidence gap. OIT is likely to be cost-effective, particularly if noncommercial products are used. There may be a trade-off: in patients with lower reaction thresholds, a commercial product may be needed for initial updosing, until a level of desensitization is achieved when they can be switched to natural food products.

The novel severe acute respiratory syndrome (SARS) coronavirus, SARS-CoV-2, is responsible for the global COVID-19 pandemic. Effective interventions are urgently needed to mitigate the effects of COVID-19 and likely require multiple strategies. Egg-extracted antibody therapies are a low-cost and scalable strategy to protect at-risk individuals from SARS-CoV-2 infection. Commercial laying hens were hyperimmunized against the SARS-CoV-2 S1 protein using three different S1 recombinant proteins and three different doses. Sera and egg yolk were collected at three and six weeks after the second immunization for enzyme-linked immunosorbent assay and plaque-reduction neutralization assay to determine antigen-specific antibody titers and neutralizing antibody titers, respectively. In this study we demonstrate that hens hyperimmunized against the SARS-CoV-2 recombinant S1 and receptor binding domain (RBD) proteins produced neutralizing antibodies against SARS-CoV-2. We further demonstrate that antibody production was dependent on the dose and type of antigen administered. Our data suggests that antibodies purified from the egg yolk of hyperimmunized hens can be used as immunoprophylaxis in humans at risk of exposure to SARS-CoV-2.

Abstract Aedes koreicus is an invasive alien mosquito species native to Asia now introduced in several European countries, including northern Italy. In this temperate region, mosquito populations survive cold winter temperatures thanks to diapausing eggs or adults, depending on the species. In its native area, Ae. koreicus was reported to overwinter in the egg stage, but to the best of our knowledge, it is not confirmed whether overwintering eggs are actually diapausing or only in a quiescence stage, i.e., they might hatch as soon as external conditions are favorable. Based on previous laboratory studies, we established a diapausing Ae. koreicus colony, maintained at 21 °C with a photoperiod of 12L:12D. Females were allowed to lay eggs, which were consequently placed in water at different time intervals after oviposition, from 30 days to 5 months. We found that diapausing eggs younger than 3 months have a poor hatching rate, while after about 100 days we observed that almost all eggs hatched. Our findings highlight that water immersion alone did not lead to the hatching of eggs, as age was found to be a significantly important factor. We thus confirm effective diapause, occurring at the egg stage, for Ae. koreicus in a recently invaded area. Moreover, our quantification of diapause duration and hatching success might help in better designing future experiments and improving modeling efforts. Graphical Abstract

Background Hen's egg is one of the most common allergens causing infantile food allergy. Consuming heated egg yolk slightly contaminated with egg white (EY with scEW) improves diet quality. Most children with egg allergies can safely consume 1/25 of a heated whole egg (low‐dose egg). Although low‐dose egg has similar antigenicity to EY with scEW, clinical reproducibility is unknown. Methionine (Met), an essential amino acid in poultry diets, when overdosed may cause hyperhomocysteinemia, which is mainly a trigger for cardiovascular diseases in humans. Homocysteine is neutralized (remethylated) in the presence of folic acid (FA), which also plays an important role in hematopoiesis and participates in the synthesis of DNA, and its deficiencies may result in the development of neural tube defects. One of the basic tools in studying the impact of both xenobiotics and nutrients on the animal organism is hematological analysis. Thus, the aim of this study was to determine the effect of in ovo supplementation with Met and FA on the hematological parameters of broiler chickens. On the 17th day of incubation, embryonated eggs (Ross 308) were injected with 5 or 25 mg of Met per egg (M5 and M25), 3 and 15 mg of FA per egg (F3 and F15), or a mixture of these 2 compounds (M5/F3 and M25/F15). The broilers were reared in accordance with welfare regulations and fed with commercial diets ad libitum. Blood samples were collected on the first, seventh, and 35th day of rearing (D1, D7, and D35), and complete hematological analysis was performed. The observed changes in red blood cell parameters probably result from physiological changes occurring during bird growth. Mean erythrocyte volume decreased with the age of chickens in the control, M5, and M25 groups, but not in those supplied with FA. Among supplemented groups, the number of white blood cells on D1 was lower only in group M5 than in the sham (C) group. The analysis of leukograms showed no significant differences between the groups. Comparing D1 with D7 in the group injected with a higher dose of Met and FA (MF25/15), a statistically significant increase in the percentage of lymphocytes and a significant decrease in the percentage of heterophils were observed. In addition, in the group injected with a higher FA dose (F15), there was statistically significant reduction in the percentage of eosinophils and a significant increase in the percentage of monocytes at day 7 compared with day 1. It seems that Met supplementation led to temporary immunosuppression in the animals.

Public discourse around “greenhouse gases” (GHG) has led to the application of life-cycle assessments to ascertain the “global warming potential” of human activities. Life-cycle assessments applied to agricultural systems typically do not consider positive contributions (i.e., fixation of atmospheric carbon dioxide [CO2]) or consider complex interrelationships among commodities within the larger agricultural sector. The purpose of this article is to present an argument for a paradigm shift and that poultry production should be considered as a value-adding activity within modern crop production systems for GHG foot-printing purposes. To this end, a case study based on 2018 production data is presented where poultry production (chicken and eggs) was contextualized as a sub-component of wheat and corn production in the Canadian provinces of Alberta and Ontario, respectively. Total GHG footprint was calculated to be 3.05 and 3.29 million tonnes (MT) of CO2 equivalent (eq) for Alberta wheat and Ontario corn production, respectively. The GHG footprint of chicken production was calculated to be 0.39 and 1.38 MT CO2 eq in Alberta and Ontario, respectively. The GHG footprint of egg production calculated to be 0.12 and 0.47 MT of CO2 eq in Alberta and Ontario, respectively. When carbon (C) fixation as crop biomass is included in the scenario, the combined crop–poultry system C balance in 2018 favored net fixation of 40.70 and 35.15 MT of CO2 eq in Alberta and Ontario, respectively. The calculated total GHG footprint of poultry production in Alberta and Ontario corresponded to only 1.2 and 5.5% of the calculated total net CO2 fixation of their respective cropping systems. This case study demonstrates that by failing to acknowledge real world estimates of C fixation by crop biomass, GHG foot-printing exercises largely misrepresent reality and can thus perpetuate faulty assumptions about the environmental footprint of animal agriculture. The authors propose that the calculations presented herein provide grounds to postulate the hypothesis that modern, integrated crop-livestock agricultural systems in Canada (and elsewhere) act as net sinks for atmospheric CO2.

Background Avian eggs have a proteinaceous cuticle. The quantity of cuticle varies and the deposition of a good cuticle in the uterus (Shell-gland) prevents transmission of bacteria to the egg contents. Results To understand cuticle deposition, uterus transcriptomes were compared between hens with i) naturally good and poor cuticle and, ii) where manipulation of the hypothalamo-pituitary-gonadal-oviduct axis produced eggs with or without cuticle. The highest expressed genes encoded eggshell matrix and cuticle proteins, e.g. MEPE (OC-116), BPIFB3 (OVX-36), RARRES1 (OVX-32), WAP (OVX-25), and genes for mitochondrial oxidative phosphorylation, active transport and energy metabolism. Expression of a number of these genes differed between hens laying eggs with or without cuticle. There was also a high expression of clock genes. PER2, CRY2, CRY1, CLOCK and BMAL1 were differentially expressed when cuticle deposition was prevented, and they also changed throughout the egg formation cycle. This suggests an endogenous clock in the uterus may be a component of cuticle deposition control. Cuticle proteins are glycosylated and glycosaminoglycan binding genes had a lower expression when cuticle proteins were deposited on the egg. The immediate early genes, JUN and FOS, were expressed less when the cuticle had not been deposited and changed over the egg formation cycle, suggesting they are important in oviposition and cuticle deposition. The uterus transcriptome of hens with good and poor cuticle deposition did not differ. Conclusions We have gained insights into the factors that can affect the production of the cuticle especially clock genes and immediate early genes. We have demonstrated that these genes change their expression over the period of eggshell formation supporting their importance. The lack of differences in expression between the uterus of hens laying eggs with the best and worse cuticle suggest the genetic basis of the trait may lie outside the oviduct.

Different strains of commercial laying hens have been molded by varying selection pressures, impacting their production, health, and behavior. Therefore, assumptions that all laying hen strains use the given resources within aviary systems similarly and maintain equal health and performance may be false. We investigated interactions among patterns of aviary resource use by 2 strains of white and 2 strains of brown laying hens (4 units per strain, 144 hens per unit) with daily egg production, location of egg laying, keel fractures, and footpad damage across the lay cycle. Hens' distribution among resources (litter, nest, wire floor, ledge, and perch) was recorded during light and dark periods at 28, 54, and 72 wk of age. Daily egg production and location were recorded, and 20% of hens per unit were randomly selected and assessed for keel bone damage, foot health, and plumage quality. Production and health risks associated with hens' resource use were assessed using multivariable regression. During the day, more brown hens occupied wire floors, while larger numbers of white hens were on perches and litter. More brown hens were on lower-tier wire floors in the dark, while more white hens occupied top tiers. Brown hens laid more eggs outside nests, showed lower incidence of keel fractures, and had better plumage quality than white hens. White hens had higher odds of keel fractures (4.2) than brown hens. Odds of keel fractures were 3.7 and 5.7 times higher at 54 and 72 wk than at 28 wk in all strains (P ≤ 0.05). Occupying the upper tier at night increased odds of keel fractures by 5.4 times. Occupying perches was associated with lower odds of foot lesions and poor plumage quality in all strains across the lay cycle (P ≤ 0.05). Finally, white hens were associated with lower odds of non-nest laying (0.76), whereas higher nest use by brown hens resulted in higher odds of non-nest egg laying (1.56) across the lay cycle (P ≤ 0.05). Distinct strain differences in resource use in an aviary were associated with different risks to hens' production, health, and welfare.

Objective Food allergy encompasses a range of food hypersensitivities. Different clinical phenotypes for food allergy likely exist in much the same way as endotype discovery is now a major research theme in asthma. We discuss the emerging evidence for different reaction phenotypes (ie, symptoms experienced after allergen exposure in food allergic individuals) and their relevance for clinical practice. Results Food anaphylaxis may be pathophysiologically different than anaphylaxis caused by nonfood triggers. Currently, there are no robust, clinically useful predictors of severity in food allergy. It is likely that patient-specific reaction phenotypes exist in food allergy, which may affect the risk of severe anaphylaxis. Allergen immunotherapy may modulate these phenotypes. Conclusion Data are emerging to confirm our clinical experience that many food allergic patients experience stereotypical symptoms after allergen exposure, both in the community and at supervised oral food challenge, in a manner that varies among patients. Integrating data sets from different cohorts and applying unbiased machine-based learning analyses may demonstrate specific food allergy endotypes in a similar way to asthma. Whether this results in improvements in patient management (eg, through facilitating risk stratification or affecting the decision to prescribe an epinephrine autoinjector and, perhaps, the number of devices) remains to be determined, but given our current inability to predict which patients are most at risk of severe food allergic reactions, this will clearly be an important area of research in the future.

Per- and poly-fluorinated substances (PFASs) are man-made chemicals that have been used for a variety of applications and can end up in the food chain. New opinions on the risk assessment were recently published by the European Food Safety Authority, emphasising the need for more sensitive methods. From this, minimum required LOQs for the analytical method for analysis of milk and egg have been calculated for perfluorooctanoic acid (PFOA) and GenX (hexafluoropropylene oxide dimer acid, HFPO-DA). A fully validated method is described for analysis of 13 PFASs, including PFOA and HFPO-DA, in milk and egg. All compounds, except perfluorodecane sulphonate (PFDS), can be quantitatively determined in these matrices with a trueness ranging from 87% to 119% and a relative within-laboratory reproducibility between 12% and 41%. Also the method proved suitable for confirmation of the identity of the individual PFASs. The LOQ for HFPO-DA in milk and egg is 0.05 ng g−1, well below the calculated required LOQ. For PFOA in egg the determined LOQ is 0.025 ng g−1, nicely below the required level of 0.03 ng g−1. In milk the required LOQ was not achieved: 0.005 instead of 0.003 ng g−1. However, on six out of eight days an LOQ of 0.0025 ng g−1 was demonstrated. It is concluded that the required LOQs are achievable when instrument performance is optimal. The current method can be expanded with long chain PFASs by using a cellulose filter instead of the PTFE filter vials. The presented method was applied for a small-scale study in The Netherlands.

Imprudent and superfluous use of antibiotics contributes to the selection of resistant bacteria, which is a large threat to human health. Therefore analytical procedures have been implemented in the poultry production sector to check if antibiotic treatments are registered, aiming to achieve more prudent use of antibiotics. These methods rely on the analysis of feathers, a matrix in which antibiotic residues persist. However, other routes besides direct administration, through which poultry feathers could contain antibiotic residues, should also be taken into account. In this research the vertical transmission from parent hen to broiler was investigated through a controlled animal study for the antibiotics enrofloxacin, doxycycline and sulfachlorpyridazine. Vertical transmission was observed for all antibiotics to both egg and egg shell. Also it is demonstrated that the transferred antibiotics from parent hen to chick are subsequently excreted via the chick’s droppings. Through this route, the broilers’ environment is contaminated. If eggs are hatched that were taken during treatment of the parent hen, this indirect route and/or the direct vertical transmission can eventually result in the detection of low concentrations of antibiotic residues in the broilers’ feathers at greater age

International audience; A total of 144 French selected breed (Galor) female guinea fowl (GF) of 42 wk of age were enrolled for a feeding trial of 15, 30, and 45 D duration. The birds were randomly assigned to 18 cages, each containing 8 birds. A total of 3 isonitrogenous and isocaloric dietary treatments were trialed, each diet comprising 6 replications (cages), which meant a total of 48 birds per diet. The GF were fed either a control diet C (commercial diet "FACI ponte 20", SIPRA, Ivory Coast, usually used for all poultry species) or the diet C supplemented with 5% Euphorbia heterophylla seeds (diet E) and the diet C supplemented with 5% Hevea seed meal (Hevea brasiliensis) (diet H). Animal performance were assessed for 3 periods (days 0 to 15, 0 to 30, and 0 to 45), and egg quality and composition were assessed at 15, 30, and 45 D of the trial. The results indicated no mortality during the trial. The laying rate was the highest (43.9%) with diet E and the lowest with diet C (32.5%), the laying rate with diet H being intermediate (38.5%). Diet E containing Euphor-bia seeds led to a reduced cholesterol content of the eggs. Additionally, inclusion of Euphorbia seeds and, to a lesser extent, of the Hevea seed meal in the diet led to n-3 polyunsaturated fatty acid enriched GF eggs, with thereby, improved nutritional value. A sensory test did not find any difference between the 3 diets on trial.

Brominated flame retardants (BFR) are primarily used as flame retardant additives in insulating materials. These lipophilic compounds can bioaccumulate in animal tissues, leading to human exposure via food ingestion. Although their concentration in food is not yet regulated, several of these products are recognised as persistent organic pollutants; they are thought to act as endocrine disruptors. The present study aimed to characterise the occurrence of two families of BFRs (hexabromocyclododecane (HBCDD) and polybrominated diphenyl ethers (PBDE)) in hen eggs and broiler or pig meat in relation to their rearing environments. Epidemiological studies were carried out on 60 hen egg farms (34 without an open-air range, 26 free-range), 57 broiler farms (27 without an open-air range, 30 free-range) and 42 pig farms without an open-air range in France from 2013 to 2015. For each farm, composite samples from either 12 eggs, five broiler pectoral muscles or three pig tenderloins were obtained. Eight PBDE congeners and three HBCDD stereoisomers were quantified in product fat using gas chromatography–high-resolution mass spectrometry, or high-performance liquid chromatography–tandem mass spectrometry, respectively. The frequencies of PBDE detection were 28% for eggs (median concentration 0.278 ng/g fat), 72% for broiler muscle (0.392 ng/g fat) and 49% for pig muscle (0.403 ng/g fat). At least one HBCDD stereoisomer was detected in 17% of eggs (0.526 ng/g fat), 46% of broiler muscle (0.799 ng/g fat) and 36% of pig muscle (0.616 ng/g fat). Results were similar in concentration to those obtained in French surveillance surveys from 2012 to 2016. Nevertheless, the contamination of free-range eggs and broilers was found to be more frequent than that of conventional ones, suggesting that access to an open-air range could be an additional source of exposure to BFRs for animals. However, the concentration of BFRs in all products remained generally very low. No direct relationship could be established between the occurrence of BFRs in eggs and meat and the characteristics of farm buildings (age, building materials). The potential presence of BFRs in insulating materials is not likely to constitute a significant source of animal exposure as long as the animals do not have direct access to these materials.

Anacardic acid is naturally found in various parts of the cashew tree (Anacardium occidentale L.) and marketed as calcium anacardate. This product has antibacterial, antifungal and antioxidant activity, and has been used in humans in the treatment and prevention of cardiovascular and cerebrovascular diseases and tumors. In meat and egg production systems, anacardic acid is used as a substitute for growth-promoting antibiotics. The study objective was to evaluate effects of adding calcium anacardate as source of anacardic acid in laying Japanese quail diet on bird performance and eggs quality. A total of 252 Japanese quail with 22 week-old were studied, using a completely randomized design composed of six treatments with six replicates and seven quails per experimental unit. Treatments applied were: diet without growth promoter; diet with 0,1% growth promoter; and four diets without growth promoter and addition of 0.25; 0.50; 0.75 and 1.0% calcium anacardate (CA), respectively. The data were submitted to analysis of variance and treatment means compared with a SNK test (5%). The data from treatments containing CA were submitted to regression analysis. Treatments did not influence feed intake, egg production, egg weight, egg volume, feed conversion ratio and eggs quality parameters. The addition up to 1% of calcium anacardate in laying Japanese quail diet, does not influence their performance and egg quality.

Using a variety of genetic resources, the aim of this study is to see how genetic background affects egg traits in chickens. Three different chicken genotypes (a commercial Brown layer, BOR; 2 Japanese indigenous breeds, NGY and YKD) were investigated effects on genotype in 10 external and internal egg quality traits along with 20 yolk and albumen free amino acid traits. Significant effects on genotype in 10 external and internal egg quality traits and 18 yolk and 17 albumen amino acid traits were found (P < 0.05). In sizes and weights of egg and eggshell redness, there were significant differences among all combinations of genotype (BOR > NGY > YKD). In 14 yolk (Asn, Ser, Gln, Gly, His, Arg, Ala, Pro, Tyr, Val, Met, Leu, Phe, and Lys) and 8 albumen amino acid traits (Gln, Gly, His, Arg, Val, Ile, Leu, and Lys), BOR was significantly higher than NGY and YKD, while the opposite relations were seen in 2 amino acid traits (Cys and GABA). Moreover, phenotypic correlation analyses revealed that positive correlations among amino acid traits within each yolk and albumen were broadly seen (0.30 < r < 0.98, P < 0.05). However, there are almost no phenotypic correlations in amino acids between yolk and albumen in BOR and NGY, but negative correlations in YKD, which implying a potential use of untapped genetic resources for modifying amino acid balance. These results indicate genetic background affects not only sizes and weights of egg but also amino acid contents and their balance of yolk and albumen.

The aim of this research was to investigate the effects of laying hen genotype and age on eggshell cuticle deposition. A total of 4,320 brown eggs were obtained from 3 modern hen strains (A, B, and C), currently used worldwide for commercial egg production, at different intervals of age (20–30, 40–50, and 60–70 wk). Four samplings of 120 randomly collected eggs were carried out for each genotype/interval of age. Eggs were individually weighed and cuticle blue staining was used to assess quality and degree of cuticle coverage. On each egg, the eggshell color profile was assessed before and after staining using the CIE L*a*b* system and these values were used to calculate ΔE*ab. A 4-point scale visual score (VS) system was also applied to estimate the degree of cuticle coverage after staining (0 = no coverage, 1 = partial coverage, 2 = total coverage - low degree, 3 = total coverage - high degree). The effects of genotype and age and their interaction on eggshell color attributes were assessed by means of factorial ANOVA, while omnibus Chi-Square and Chi-squared Automatic Interaction Detector algorithm were applied for the analysis of VS data. Overall, both genotype and age affected the eggshell color profile as well as the degree of cuticle coverage. Hen strain A showed better cuticle deposition in comparison with B and particularly C one, being ΔE*ab values significantly higher. The VS evaluation revealed that eggs with impaired cuticle coverage degree increased with the hen age (23, 34, and 37%, respectively for 20–30, 40–50, and 60–70 wk; P < 0.05). However, a significant interaction between genotype and age was observed: transition from early to late hen age resulted in a significantly different pattern of ΔE*ab changes in each genotype. The classification tree analysis confirmed that the hen genotype has a greater effect than the age on cuticle deposition. In conclusion, considering the importance of the cuticle in table egg production, these results highlight the crucial role exerted by the genotype on eggshell cuticle coverage.

Cathepsin D (CatD, EC 3.4.23.5) is a member belonging to the subfamily of aspartic endopeptidases, which are classified into the MEROPS clan AA, family A1. Helminth parasites express a large set of different peptidases that play pivotal roles in parasite biology and pathophysiology. However, CatD is less well known than the other classes of peptidases in terms of biochemical properties and biological functions. In this study, we identified 2 novel CatDs (CsCatD1 and CsCatD2) of Clonorchis sinensis and partially characterized their properties. Both CsCatDs represent typical enzymes sharing amino acid residues and motifs that are tightly conserved in the CatD superfamily of proteins. Both CsCatDs showed similar patterns of expression in different developmental stages of C. sinensis, but CsCatD2 was also expressed in metacercariae. CsCatD2 was mainly expressed in the intestines and eggs of C. sinensis. Sera obtained from rats experimentally infected with C. sinensis reacted with recombinant CsCatD2 beginning 2 weeks after infection and the antibody titers were gradually increased by maturation of the parasite. Structural analysis of CsCatD2 revealed a bilobed enzyme structure consisting of 2 antiparallel β-sheet domains packed against each other forming a homodimeric structure. These results suggested a plausible biological role of CsCatD2 in the nutrition and reproduction of parasite and its potential utility as a serodiagnostic antigen in clonorchiasis.

Dysregulation of the immune system is associated with many pathologies, including cardiovascular diseases, diabetes, and cancer. To date, the most commonly used models in biomedical research are rodents, and despite the various advantages they offer, their use also raises numerous drawbacks. Recently, another in vivo model, the chicken embryo and its chorioallantoic membrane, has re-emerged for various applications. This model has many benefits compared to other classical models, as it is cost-effective, time-efficient, and easier to use. In this review, we explain how the chicken embryo can be used as a model for immune-based studies, as it gradually develops an embryonic immune system, yet which is functionally similar to humans’. We mainly aim to describe the avian immune system, highlighting the differences and similarities with the human immune system, including the repertoire of lymphoid tissues, immune cells, and other key features. We also describe the general in ovo immune ontogeny. In conclusion, we expect that this review will help future studies better tailor their use of the chicken embryo model for testing specific experimental hypotheses or performing preclinical testing.

The avian egg perivitelline layer (PL) is a proteinaceous structure that encloses the egg yolk. It consists of the inner and the outer perivitelline layers (IPL and OPL, respectively) that are assumed to play distinct roles in bird reproduction. To gain insight into their respective function, we analyzed the proteome of IPL and OPL in chicken unfertilized eggs after mechanical separation, using a GeLC-MS/MS strategy. Of the 412 proteins identified, 173 proteins were uniquely recovered in IPL and 98 proteins in OPL, while 141 proteins were identified in both sublayers. Genes coding the most abundant proteins were shown to be expressed either in the liver/ovary (IPL formation) or in the oviduct (OPL formation), but rarely in both. The presence of oviduct-specific proteins (including LYZ, VMO1, AvBD11, PTN, OVAL and LOC10175704) in IPL strongly suggests that they participate in the physical association of IPL to OPL, whose tight attachment was further evidenced by analyses of IPL/OPL interfaces (by scanning electron microscopy). Functional annotation of identified proteins revealed functions associated with fertilization and early development for IPL, while OPL would rather participate in egg defense and embryogenesis. Collectively, our data highlight the complementary functions of IPL and OPL that are major determinants of bird reproductive success. SIGNIFICANCE: The present study unveils for the first time the individual proteomes of the two sublayers composing the chicken egg perivitelline layer (PL), which allowed to assign their respective putative biological roles in avian reproduction. The combination of proteomics with gene expression and ultrastructural analyses provides insightful data on the structure and biochemistry of the avian PL. The functional annotation of PL proteins highlights the multifaceted biological functions of this structure in reproduction including fertilization, embryonic development, and antimicrobial protection. This work will stimulate further research to validate predicted functions and to compare the physiology and the functional specificities of PL in egg-laying species.

Methane is a short-lived greenhouse gas (GHG) modelled distinctly from long-lived GHGs such as carbon dioxide and nitrous oxide to establish global emission budgets for climate stabilisation. The Paris Agreement requires a 24–47% reduction in global biogenic methane emissions by 2050. Separate treatment of methane in national climate policies will necessitate consideration of how global emission budgets compatible with climate stabilisation can be downscaled to national targets, but implications of different downscaling rules for national food production and climate neutrality objectives are poorly understood. This study addresses that knowledge gap by examining four methods to determine national methane quotas, and two methods of GHG aggregation (GWP100 and GWP*) across four countries with contrasting agriculture, forestry and other land use (AFOLU) sectors and socio-economic contexts (Brazil, France, India and Ireland). Implications for production of methane-intensive food (milk, meat, eggs and rice) in 2050 and national AFOLU climate neutrality targets are explored. It is assumed that methane quotas are always filled by food production where sufficient land is available. Global methane budgets for 1.5 °C scenarios are downscaled to national quotas based on: grand-parenting (equal percentage reductions across countries); equity (equal per capita emissions); ability (emission reductions proportionate to GDP); animal protein security (emissions proportionate to animal protein production in 2010). The choice of allocation method changes national methane quotas by a factor of between 1.7 (India) and 6.7 (Ireland). Despite projected reductions in emission-intensities, livestock production would need to decrease across all countries except India to comply with quotas under all but the most optimistic sustainable intensification scenarios. The extent of potential afforestation on land spared from livestock production is decisive in achieving climate neutrality. Brazil and Ireland could maintain some degree of milk and beef export whilst achieving territorial climate neutrality, but scenarios that comply with climate neutrality in India produce only circa 30% of national calorie and protein requirements via rice and livestock. The downscaling of global methane budgets into national policy targets in an equitable and internationally acceptable manner will require simultaneous consideration of the interconnected priorities of food security and (land banks available for) carbon offsetting.

Background The dominant Gasterophilus species in the desert steppe (Xinjiang, China) Gasterophilus pecorum poses a serious threat to the reintroduced Przewalski’s horses. We investigated the distribution pattern of G. pecorum eggs in June 2017. Methods Two sampling methods, transect and grid, were used, and the results were analyzed via geostatistics by semivariance. The nest quadrat was used to determine the optimal quadrat size. Results Eggs were found in 99 quadrats (63.1%) and 187 clusters (1.5%) of Stipa caucasica on the steppe. The mean oviposition count of a cluster was 3.8 ± 1.6. Three-eggs is the mode of which females oviposit on each ovigerous S. caucasica (22.0%). Semivariogram analysis revealed that the distance of spatial dependence for eggs was 921 m, 1233 m and 1097 m for transect 1, transect 2 and grid methods, respectively, while spatial continuity was 62%, 77% and 57.0% for transect 1, transect 2 and grid, respectively. The eggs showed a patchy, aggregated distribution pattern. This suggested the spherical model is most applicable. The proportion of ovigerous S. caucasica was significantly correlated with the distance from water resources (r = − 0.382, p = 0). Conclusion Our findings indicated that diversification of G. pecorum oviposition was a new adaptative strategy for its survival in the desert steppe ecological niche. This made it more efficient at infecting hosts in the local environment. Areas surrounding water resources, especially around the drinking paths of equids (500 m radius surrounding the water), were concentrated epidemic areas. It is suggested that more attention to be paid to the ecological characteristics of G. pecorum in order to develop control measures that would reduce the infection risk for Przewalski’s horses.

Eggs are highly nutritious foods, yet intake by children in Ethiopia is low. We hypothesized that a nutrition-sensitive poultry intervention improves nutritional status of children 6–18 months using a 6-month cluster randomized controlled community trial. Intervention group (IG) children received a gift of two egg-laying hens in a ceremony where children’s ownership of the chickens was declared by community leaders. Parents promised to add more hens and feed the owner-child one-egg-a-day. Trained community workers reinforced egg feeding, environmental sanitation and poultry husbandry. Control group (CG) mothers received usual nutrition education on child feeding. At baseline 29.6% of children were stunted, 19.4% underweight and 8.6% wasted. Egg consumption significantly increased only in IG, at 6 months. The intervention increased weight-for-age and weight-for-height z-scores by 0.38 (95% CI = 0.13–0.63) and 0.43 (95% CI = 0.21–0.64), respectively. Binary logit model indicated IG children were 54% (Odds ratio [OR] = 0.46; 95% CI = 0.26–0.84) and 42% (OR = 0.58; 95% CI = 0.37–0.91) less likely to be underweight and stunted, respectively, compared to CG. IG children attained the milestone of running (p = 0.022; AHR = 1.43; 95% CI = 1.05–1.95), kicking a ball (p = 0.027; AHR = 1.39; 95% CI = 1.04–1.87) and throwing a ball (p = 0.045; AHR = 1.37; 95% CI = 1.01–1.86) earlier than CG. This nutrition-sensitive child-owned poultry approach should be implemented where animal-source food intake is low.

Non-destructive testing (NDT) for eggshell faults is highly important for the egg industry, as cracked eggs account for around 3% of total production. The most commonly used method at present, candling, is labor intensive, while computer vision systems are expensive and complicated. In this paper, we present a simple, yet efficient, novel method for eggshell crack detection by acoustic spectroscopy. Altogether, 693 sound recordings were evaluated by different classification methods. The results show a cross-validated 2.1% total classification error, with only 0.87% false positive rate, which is the crucial metric for fresh eggs. Adapting the developed method to an industrial setting may lead to a reliable, fast and cost-effective detection method.

Fertilization is essential for species survival. Although Izumo1 and Juno are critical for initial interaction between gametes, additional molecules necessary for sperm:egg fusion on both the sperm and the oocyte remain to be defined. Here, we show that phosphatidylserine (PtdSer) is exposed on the head region of viable and motile sperm, with PtdSer exposure progressively increasing during sperm transit through the epididymis. Functionally, masking phosphatidylserine on sperm via three different approaches inhibits fertilization. On the oocyte, phosphatidylserine recognition receptors BAI1, CD36, Tim-4, and Mer-TK contribute to fertilization. Further, oocytes lacking the cytoplasmic ELMO1, or functional disruption of RAC1 (both of which signal downstream of BAI1/BAI3), also affect sperm entry into oocytes. Intriguingly, mammalian sperm could fuse with skeletal myoblasts, requiring PtdSer on sperm and BAI1/3, ELMO2, RAC1 in myoblasts. Collectively, these data identify phosphatidylserine on viable sperm and PtdSer recognition receptors on oocytes as key players in sperm:egg fusion., Izumo and Juno are receptors on sperm and eggs respectively required for fusion, but other factors for sperm-egg fusion are poorly studied. Here, the authors report that phosphatidylserine, found mainly on cells marked for death, is also present on motile sperm, recognized by egg receptors, and is required for sperm-egg fusion.

Objectives Salmonella is the most important causes of foodborne illness especially from poultry and poultry products. So the aim of this study was to carryout phenotypic characterization, antimicrobials susceptibility pattern and risk factors of salmonella isolates from farms and markets eggs, cloacae swabs of chickens and stool of egg collectors. A cross-sectional study was conducted from January 2018 to September 2018. Samples were, processed; salmonella was isolated, Phenotypically identified by OmniLog and antimicrobials susceptibility were carried out. Result Over all; 11(2.65%) of salmonella enterica were phenotipically characterized out of 415 samples from farms egg content (n=83), farms eggshell (n=83), cloacae (n=83), market eggshell (n=83) and market egg contents (n=83) with 2.4%, 0%, 2.4%, 4.8% and 3.6% prevalence, respectively. Out of isolates, 8(72.72%) displayed multidrug resistance. All isolates showed susceptibility to Gentamicin, Kanamycin and Streptomycin. Lack of separating cracked eggs, washing hand, eggs stay longer unsold, and mixing excreta with feed were associated risk factors for salmonella presence (p-value

Contaminated liver fluke egg in the environment has led to the high prevalence of human opisthorchiasis associated with cholangiocarcinoma in Southeast Asia. To find the effective lessening methods of Opisthorchis viverrini eggs in the contaminated environment, we investigated the temperature conditions for killing of these trematode eggs in vitro. Numerous O. viverrini eggs were obtained in the proximal part of uteri of adult worms from experimental hamsters. Mature eggs with miracidium were allocated by experimental groups (2 control: positive and negative and 4 treatment: 50, 60, 70, and 80°C) with 0.85% saline, and treated by the experimental plan. Eggs in each experimental groups were observed under the confocal microscope after stain with Propidium Iodide (PI) to evaluate the effect of temperatures. Eggs in 70 and 80°C groups were all killed after over 10 min heated. Majority of eggs in 60°C (10, 15, and 30 min heated), 70 and 80°C (5 min heated) groups were inactivated. However in 50°C group, below half of eggs were to be killed in all time lapse (10, 15 and 30 min). In order to prevent O. viverrini infection and cholangiocarcinoma, direct treatment of sewage by heating at 70 or 80°C at least 10 min is essential. Therefore, treatment of O. viverrini eggs at a high temperature is a potential method for controlling egg contamination in sewage.

Food allergy is a growing health problem worldwide; thus, there is an urgent need for robust, specific, and sensitive analytical methods for detecting allergens. Mass spectrometry is an alternative to the existing methods, and it can overcome their limitations. One of the first steps in the development of any analytical method is the identification of the analytes to be further studied. In the case of allergen detection by mass spectrometry, the analytes are peptides. In this study, a strategy was developed for identifying potential peptide biomarkers in processed food products. This strategy was applied to processed egg matrices, and 16 potential peptide biomarkers were identified for the further detection and quantification of egg by means of mass spectrometry. With an empirical approach based on dedicated sample preparation, including tandem Lys-C/trypsin enzymatic digestion and high-resolution mass spectrometry analysis, hundreds of peptides from egg proteins were identified. This list of peptides was further refined with a series of criteria, obtained from empirical evidence, to identify the ideal biomarkers for the development of a quantitative method. These criteria include the resistance to food processing and the specificity of the peptides for eggs but also the effects of amino acid modifications and enzymatic digestion efficiency.

Background Salmonella enterica serovar Enteritidis (S. Enteritidis) is one of the most common serovars, associated with human salmonellosis. The food-borne outbreak of this bacterium is mainly related to the consumption of contaminated poultry meat and poultry products, including eggs. Therefore, rapid and accurate detection, besides investigation of virulence characteristics and antimicrobial resistance profiles of S. Enteritidis in poultry and poultry egg samples is essential. A total of 3125 samples (2250 poultry and 875 poultry egg samples), sent to the administrative centers of veterinary microbiology laboratories in six provinces of Iran, were examined for Salmonella contamination, according to the ISO 6579 guideline. Next, duplex PCR was conducted on 250 presumptive Salmonella isolates to detect invA gene for identification of the genus Salmonella and sdf gene for identification of S. Enteritidis. Subsequently, the S. Enteritidis isolates were examined for detection of important virulence genes (pagC, cdtB, msgA, spaN, tolC, lpfC, and spvC) and determination of antibiotic resistance patterns against nalidixic acid, trimethoprim-sulfamethoxazole, cephalothin, ceftazidime, colistin sulfate, and kanamycin by the disk diffusion method. Results Overall, 8.7 and 2.3% of poultry samples and 6.3 and 1.3% of eggs were contaminated with Salmonella species and S. Enteritidis, respectively. The invA and msgA genes (100%) and cdtB gene (6.3%) had the highest and the lowest prevalence rates in S. Enteritidis isolates. The spvC gene, which is mainly located on the Salmonella virulence plasmid, was detected in 50.8% of S. Enteritidis isolates. The S. Enteritidis isolates showed the highest and the lowest resistance to nalidixic acid (87.3%) and ceftazidime (11.1%), respectively. Unfortunately, 27.0% of S. Enteritidis isolates were multidrug-resistant (MDR). Conclusion The rate of contamination with Salmonella in the poultry and egg samples, besides the presence of antimicrobial resistant and MDR Salmonella isolates harboring the virulence genes in these samples, could significantly affect food safety and subsequently, human health. Therefore, continuous monitoring of animal-source foods, enhancement of poultry farm control measures, and limiting the use of antibiotics for prophylactic purposes in food producing animals, are essential for reducing the zoonotic risk of this foodborne pathogen for consumers and also choosing effective antibiotics for the treatment of salmonellosis.

The appearance, size, and weight of poultry meat and eggs are essential for production economics and vital in the poultry sector. These external characteristics influence their market price and consumers' preference and choice. With technological developments, there is an increase in the application and importance of vision systems in the agricultural sector. Computer vision has become a promising tool in the real-time automation of poultry weighing and processing systems. Owing to its noninvasive and nonintrusive nature and its capacity to present a wide range of information, computer vision systems can be applied in the size, mass, volume determination, and sorting and grading of poultry products. This review article gives a detailed summary of the current advances in measuring poultry products' external characteristics based on computer vision systems. An overview of computer vision systems is discussed and summarized. A comprehensive presentation of the application of computer vision-based systems for assessing poultry meat and eggs was provided, that is, weight and volume estimation, sorting, and classification. Finally, the challenges and potential future trends in size, weight, and volume estimation of poultry products are reported.

Elevating levels of environmental lead (Pb) results in serious hazards to health of animals and human beings. In this study, daily diet with three different levels of Pb (Pb nitrate at doses of 1, 10, and 100 mg/kg body weight) were fed to ISA Brown layers. It showed that the kidney and liver have the relatively high Pb concentration (2.34 and 0.51 ppm) after culture, while the meat has the Pb concentration as low as 0.07 ppm (lower than the standard of Codex Alimentarius). It was also confirmed that egg laying worked as a potential pathway for hens to excrete Pb as Pb concentrations in eggshell and yolk increased from 0.10 to 3.11 ppm. We studied the correlations between egg geometrical parameters (i.e., egg shape index, sphericity, geometric mean diameter, surface area, and volume) and eggshell qualities, or the organic matrix in eggshell. Eggs were collected from 5 poultry breeds belonging to 3 species (commercial Hy-line Brown Chicken, Shaoxing Duck, Jinding Duck, Taihu Goose, and Zhedong White Goose). The geometrical parameters showed high variation among 3 species of poultry, and even between breeds in the same species. The five geometrical parameters were grouped into 2 sets, one contained shape index and sphericity, the other comprised geometric mean diameter, surface area, and volume. The parameters in the same set can be perfectly fitted to one another. Egg weight, shell membrane weight, and calcified shell weight were significantly correlated with geometric mean diameter, surface area, and volume. In accordance with false discovery rate-adjusted P value, both shell membrane relative weight and calcified shell thickness showed no significant correlations with any of the geometrical parameters. However, the correlations between geometrical parameters and other shell variables (calcified shell weight, shell relative weight, calcified shell thickness uniformity, and eggshell breaking strength) depend on breed. Both constitutive proportions and percentage contents of 3 eggshell matrix components (acid-insoluble, water-insoluble, and both acid and water facultative-soluble matrix) had no effects on egg shape and size. The correlations between the amounts of various shell matrix, egg shape and size depend on breed or species. This study provides a methodology and the correlation between geometrical parameters and eggshell qualities, and between geometrical parameters and organic matrix components in calcified shells.

Scaffoldin, an S100 fused-type protein (SFTP) with high amino acid sequence similarity to the mammalian hair follicle protein trichohyalin, has been identified in reptiles and birds, but its functions are not yet fully understood. Here, we investigated the expression pattern of scaffoldin and cornulin, a related SFTP, in the developing beaks of birds. We determined the mRNA levels of both SFTPs by reverse transcription polymerase chain reaction (RT-PCR) in the beak and other ectodermal tissues of chicken (Gallus gallus) and quail (Coturnix japonica) embryos. Immunohistochemical staining was performed to localize scaffoldin in tissues. Scaffoldin and cornulin were expressed in the beak and, at lower levels, in other embryonic tissues of both chickens and quails. Immunohistochemistry revealed scaffoldin in the peridermal compartment of the egg tooth, a transitory cornified protuberance (caruncle) on the upper beak which breaks the eggshell during hatching. Furthermore, scaffoldin marked a multilayered peridermal structure on the lower beak. The results of this study suggest that scaffoldin plays an evolutionarily conserved role in the development of the avian beak with a particular function in the morphogenesis of the egg tooth.

The invasive mosquito Aedes japonicus japonicus (Theobald, 1901) settled in 2013 in the Alsace region, in the northeast of France. In this temperate area, some mosquito species use diapause to survive cold winter temperatures and thereby foster settlement and dispersal. This study reports diapause and its seasonality in a field population of Ae. japonicus in the northeast of France. For two years, eggs were collected from May to the beginning of November. They were most abundant in summer and became sparse in late October. Diapause eggs were determined by the presence of a fully developed embryo in unhatched eggs after repeated immersions. Our study showed effective diapause of Ae. japonicus in this part of France. At the start of the egg-laying period (week 20), we found up to 10% of eggs under diapause, and this rate reached 100% in October. The 50% cut-off of diapause incidence was determined by the end of summer, leading to an average calculated maternal critical photoperiod of 13 h 23 min. Interestingly, diapause was shown to occur in part of the eggs even at the earliest period of the two seasons, i.e. in May of each year. Even though we observed that the size of eggs was positively correlated with diapause incidence, morphology cannot be used as the unique predictive indicator of diapause status due to overlapping measurements between diapausing and non-diapausing eggs. Dans une telle région tempérée, certaines espèces de moustiques ont recours à la diapause pour survivre aux températures froides de l’hiver, ce qui favorise donc l’établissement et la dispersion de ces espèces. Cette étude montre le phénomène de diapause et la saisonnalité d’une population sauvage d’Ae. japonicus dans le nord-est de la France. Pendant deux ans, les œufs ont été collectés de mai à début novembre. Ils étaient plus abondants durant l’été et plus rarement présents en octobre. Les œufs diapausants ont été déterminés par la présence d’un embryon entièrement développé dans les oeufs non éclos après des immersions répétées. Notre étude a mis en évidence une diapause effective d’Ae. japonicus dans le nord-est de la France. Au début de la période de ponte (semaine 20), jusqu’à 10 % des œufs a été trouvé à l’état de diapause et ce taux atteint 100 % en octobre. Un taux de 50 % de diapause a été atteint à la fin de l’été, amenant à une photopériode critique maternelle calculée de 13 heures 23 minutes en moyenne. De façon intéressante, il a été montré que la diapause se produisait chez une partie des œufs, et ce même dès le début des deux saisons, en mai de chaque année. Bien que nous ayons observé que les mesures des œufs étaient positivement corrélées avec le taux de diapause, la morphologie des œufs seule ne peut pas être utilisée comme unique indicateur de prédiction du statut de diapause, à cause d’un chevauchement des mesures entre les œufs diapausants et non diapausants. Cette étude offre de nouvelles connaissances sur les caractéristiques de la diapause et sur les traits d’invasion d’Ae. japonicus.

In this study, liquid egg, albumen, and egg yolk were artificially inoculated with E. coli. Ultrasound equipment (20/40 kHz, 180/300 W; 30/45/60 min) with a circulation cooling system was used to lower the colony forming units (CFU) of E. coli samples. Frequency, absorbed power, energy dose, and duration of sonication showed a significant impact on E. coli with 0.5 log CFU/mL in albumen, 0.7 log CFU/mL in yolk and 0.5 log CFU/mL decrease at 40 kHz and 6.9 W absorbed power level. Significant linear correlation (p < 0.001) was observed between the energy dose of sonication and the decrease of E. coli. The results showed that sonication can be a useful tool as a supplementary method to reduce the number of microorganism in egg products. With near-infrared (NIR) spectra analysis we were able to detect the structural changes of the egg samples, due to ultrasonic treatment. Principal component analysis (PCA) showed that sonication can alter C–H, C–N, –OH and N–H bonds in egg. The aquagrams showed that sonication can alter the properties of H2O structure in egg products. The observed data showed that the absorbance of free water (1412 nm), water molecules with one (1440 nm), two (1462 nm), three (1472 nm) and four (1488 nm) hydrogen bonds, water solvation shell (1452 nm) and strongly bonded water (1512 nm) of the egg samples have been changed during ultrasonic treatment.
