The present study reveals purification and characterization of a C-type lectin from the serum of pearl spot, Etroplus suratensis (Es-Lec). The Es-Lec was purified by affinity chromatography with mannose coupled sepharose CL-4B column and it exhibits single band with a molecular weight of 75 kDa in SDS-PAGE. The surface morphology of purified Es-Lec displays the homogeneous nature of protein. A distinct peak with a retention time of 2.958 min was appeared in high performance liquid chromatography (HPLC), X-ray diffraction (XRD) analysis expresses a single peak at 31.8372 and MALDI-TOF peaks which shows the purity and crystalline nature of the protein respectively. Functional analysis of purified Es-Lec exhibits yeast agglutination activity against Saccharomyces cerevisiae and has the ability to agglutinate the human erythrocytes, which was observed by light microscopy and haemagglutination inhibition was also done. In addition, purified Es-Lec showed the broad spectrum of antibacterial activity against Gram negative Vibrio parahaemolyticus and Aeromonas hydrophila. Antibiofilm potential of purified Es-Lec against selected Gram-negative bacteria exhibited the disruption of biofilm architecture at the concentration of 50 μg ml−1 and also it exhibited antiviral and anticancer activity.

This paper characterize microplastics (MPs) in the heavily urbanized, brackish water Vembanad Lake (India), focussing on some commercially important bottom-feeding fishes and shellfish (Arius maculatus, Etroplus suratensis, E. maculatus and Villorita sp.). The average abundance of MPs was higher in the water column (872 ± 573 nos./m3) than in finfishes (15 ± 13 particles per fish) and shellfish (23 ± 20 nos./ind.). Fish consumption from contaminated water-bodies is a serious health issue. This study conducted to reveal the presence of heavy metals and bisphenols in Vembanad lake, an exploiting tourist spot in Kerala, receiving untreated agricultural, domestic, municipal, and industrial effluents. We evaluated aquatic contaminant impact on hepatic stress markers in Etroplus suratensis from fragile Vembanad lake. The significant difference in water physiochemical parameters, the concentration of heavy metals, and bisphenols (BPA and BPS) were studied. Hepatic tissue of E. suratensis inhabited in lake featured with high iron (11.29 ± 0.39 ppm) and BPA (0.02412 ± 0.0031 µg/mL) content along with an increased hepatic stress marker and distorted hepatic structure. The study highlights the presence of high iron and BPA in edible fish. The study recommends monitoring of physiochemical characters of freshwater lakes is essential for better survival of freshwater flora and fauna.

Flow cytometry was used for estimating the genome size of five brackishwater finfish and four shrimp species. The genome size for Lutjanus argentimaculatus was 0.95 ± 0.10 and 0.79 ± 0.01 pg for Scatophagus argus. The genome sizes for Chanos chanos (0.72 ± 0.01 pg), Etroplus suratensis (1.71 ± 0.16 pg) and Liza macrolepis (0.87 ± 0.02 pg) which are important aquaculture species are reported for the first time in this study. The phylogenetic tree constructed using sixty-seven sequence accessions of cytochrome c oxidase subunit 1 (COI) gene of Lates calcarifer revealed two separate clades. The Indian Lates calcarifer species with estimated genome size of 0.44 ± 0.02 pg belonged to a clade different than that of South East Asia and Australia reported to have larger genome size. The genome size for the four major species of genus Penaeus (Penaeus monodon, Penaeus indicus, Penaeus vannamei and Penaeus japonicus) were found in similar range. The genome size of female shrimps ranged from 2.91 ± 0.03 pg (P. monodon) to 2.14 ± 0.02 pg (P. japonicus). In male shrimps, the genome size ranged from 2.86 ± 0.06 pg (P. monodon) to 2.19 ± 0.02 pg (P. indicus). Significant difference was observed in the genome size between male and female shrimp of all species except in P. monodon. The highest relative difference of 12.78% was observed in the genome size between the either sex in P. indicus. The interspecific relative difference of 30.59% in genome size was highest between the male shrimps of P. monodon and P. indicus and 35.98% between the female shrimps of P. monodon and P. japonicus. The stored gills and pleopod tissues could be successfully used up to 3 weeks to estimate the genome size in shrimps.

This article discusses bioaccumulation and temporal variation of heavy metals in three edible lagoon fish species with references to gender. Cd and As were undetected in the three fish species (i.e., below the method detection limits of 1 μg Cd/kg wet weight and 2 μg As/kg wet weight) irrespective of the periods, and Cd was undetected in the water too (i.e., below the detection limit of 0.0003 mg/L). The present study exhibits the occurrence of MPs in different matrices (water, sediment and biota) from the Sal estuary, Goa, situated on the central west coast of India. The average numbers of MPs in the water column and sediment were 48 ± 19 MP particles/L (MPs/L) and 3950 ± 930 MP particles/kg (MPs/kg), respectively. In shellfish (whole soft tissue), the average concentrations of MPs were 4 ± 2 (Crassostrea sp.), 3.2 ± 1.8 (Perna viridis) and 0.7 ± 0.3 (Paphia malbarica) MPs/g body weight (bw), respectively. The highest MPs were recorded in finfish (gastro-intestinal tract) (Mugil cephalus)7.8 ± 4, followed by (Gerres filamentosus) 5.3 ± 4.9, (Arius jella) 4.6 ± 2.6, and (Etroplus suratensis)1.4 ± 0.3 MP/g bw. MP fibres were predominant in all matrices. Interestingly, a dominance of small sized (10–300 μm) MPs was recorded in biota. Among the 37 polymer types identified by μ-FTIR, the most prevalent ones were, polyacrylamide (PAM) , polyacetylene, ethylene vinyl alcohol (EVOH), polyvinyl chloride (PVC) and polyamide (nylon). Notably, the polymers dominant in the gut of finfish and in whole shellfish were equally prominent in sediment and the water column. This study highlights the presence of MPs in commercially important shellfish and finfish samples from the Sal estuary. This study clearly shows the presence of MPs in various types of marine organisms in the Sal estuary. As shellfish is locally consumed as a delicacy and plays a major role in the seafood industry, the MPs may pose a hazard for human health. There is also an ecological risk as MPs are also found in water and sediment and in the digestive tract of finfish.

Viral nervous necrosis (VNN) affects more than 120 species mostly belonging to the order Perciformes. However, none of the brackishwater species belonging to the family Cichlidae under the order Perciformes are reported to be susceptible. Hence, the present experiment was undertaken to study the susceptibility of the brackishwater cichlid, pearlspot, Etroplus suratensis to NNV. Thirty-day-old pearlspot larvae were infected with NNV by immersion. Mortality was recorded till 14 days post-infection, and the infected larvae were subjected to nested RT-PCR and histology. The virus was isolated from infected larvae using SSN-1 cells. To study the replication of the virus in vitro, primary cultured brain cells of E. suratensis and IEK cells were infected with NNV. No mortality was observed in any of the control or experimentally infected larvae. However, the experimentally infected larvae were positive for NNV by nested RT-PCR and the virus was isolated using SSN-1 cells. Further, the infected pearlspot brain cells and IEK cells showed cytopathic effect at second and third passage of the virus and they were positive for NNV by nested RT-PCR. Pearlspot is relatively resistant to VNN although the virus could replicate in the larvae and in cell culture.

Innate immune system recognises pathogen-associated molecular patterns (PAMPs) by limited number of germline encoded and non-clonally developed pathogen recognition receptors (PRRs). Retinoic acid-inducible gene I (RIG-I)-like receptors (RLRs) are important cytosolic PRRs for sensing viral RNAs. The receptor encoded by melanoma differentiation associated gene 5 (MDA5), an RLR, recognises viral RNA and enhances antiviral response in host cells. The full-length MDA5 cDNA in Etroplus suratensis was cloned and found to have 3673 nucleotides encoding a polypeptide of 978 amino acids. The deduced amino acid sequence contains four main structural domains: two CARD domains in the N-terminal region, a DExDc (DEAH/DEAD box helicase domain), HELICc (C-terminal helicase) domain and a C-terminal regulatory domain (RD). Phylogenetic analysis revealed a close relationship of E. suratensis MDA5 ( Es MDA5) with MDA5 of Neolamprologus brichardi and Oreochromis niloticus , both belonging to Cichlidae family. Es MDA5 transcripts were ubiquitously expressed in all the 12 tissues tested in healthy fish. Although, transcript level was found to be the highest in muscle, high expression was also detected in the spleen, head kidney and hindgut. In poly I:C-injected fish, Es MDA5 transcripts showed peak expression in the spleen, intestine and heart at 12 h post-injection (hpi). However, in gill and kidney tissues, maximum up-regulation of Es MDA5 was observed at 6 and 48 hpi, respectively. Further, liver tissue showed an increasing trend in expression profile from 6 to 48 hpi. Interferon promoter stimulator-1 (IPS-1) gene, an adaptor triggering RIG-I- and MDA5-mediated type I interferon induction, also showed up-regulated expression at initial time-points in poly I:C-injected E. suratensis . The constitutive expression and up-regulation of Es MDA5 and the IPS-1 genes in different tissues indicate that Es MDA5 may play an important role in sensing viral PAMPs in conjunction with IPS-1.

Copper (Cu) and zinc (Zn) play a vital role in the growth and development, however increased uptake causes deleterious effects in normal functioning of organisms. We have demonstrated in this contribution the tolerance limit of Cu and Zn on Etroplus suratensis (pearl spot) by Continuous Flow Through (CFT) bioassay and the biomarker responses. The accumulation for Cu, Zn and selected trace metals (Cr, Cd, Ni and Pb) from field conditions, as well Geo-accumulation index (I geo) and Contamination factor (C.F) suggested moderate to heavy pollution in the Cochin estuarine system. The 96 h LC50 values for Cu was 1.74 ± 0.04 mg L−1and that for Zn was 24.36 ± 0.58 mg L−1 at 95% confidence interval. No observed effect concentration (NOEC) and low observed effect concentration (LOEC) for Cu and Zn were derived based on the survival rates. Chronic toxicity values for Cu and Zn were 0.23 mg L−1 and 2.005 mg L−1 respectively for 30 days period. The histological, biochemical, hematological and behavioral parameters showed significant variations at sublethal concentrations. Lamellar hyperplasia in gills, vacuolation combined with necrosis in liver, increased occurrence of melanomacrophage centres in spleen were noticed at chronic levels for both Cu and Zn. Tissue specific bioconcentration was observed for zinc and copper in gill and liver respectively, with least rate of bioconcentration observed in muscle tissues. Fishes are always exposed to various environmental stresses and the chances of succumbing to such stresses are of great physiological concern. Any change in temperature from the ambient condition can induce various metabolic and physiological changes in the body. The present study evaluates the effects of temperature induced stress on the antioxidant profile of Etroplus suratensis such as superoxide dismutase, catalase, glutathione peroxidase and lipid peroxidation. Fishes of same size were kept in a thermostatized bath at three different temperature regimes viz 16 °C, 27 °C (ambient temperature) and 38 °C for 72 h. These temperatures were selected based on the CT Max (Critical Thermal Maximum) and CT Min (Critical Thermal Minimum) exhibited by E. suratensis. Superoxide dismutase and catalase activity was found maximum in brain and muscle respectively during the 48th hour of exposure in fishes kept at 38 °C. At 16 °C the antioxidant response of glutathione peroxidase was maximum in muscles, whereas the lipid peroxidation rate was found to be high in gills compared to other tissues. The profound increase in the levels of oxidative stress related biomarkers indicate that the thermal stressors severely affected oxidative state of E. suratensis by the second day of experiment. Such down-regulation of redox state accompanied with the induction of oxidative stress cascade may lead to physiological damage in various tissues in fishes, in vivo. However current data indicate that a transition to low and high temperature environment from ambient condition severely affected the levels and profile of the antioxidant markers overtime in E. suratensis.

The present study was conducted to elucidate the osmoregulatory ability of the fish pearl spot (Etroplus suratensis) to know the scope of this species for aquaculture under various salinities. Juvenile pearl spot were divided into three groups and acclimated to freshwater (FW), brackish water (BW) or seawater (SW) for 15 days. The fish exhibited effective salinity tolerance under osmotic challenges. Although the plasma osmolality and Na(+), K(+) and Cl(-) levels increased with the increasing salinities, the parameters remained within the physiological range. The muscle water contents were constant among FW-, BW- and SW-acclimated fish. Two Na+/K+-ATPase α-isoforms (NKA α) were expressed in gills during acclimation in FW, BW and SW. Abundance of one isoform was up-regulated in response to seawater acclimation, suggesting its role in ion secretion similar to NKA α1b, while expression of another isoform was simultaneously up-regulated in response to both FW and SW acclimation, suggesting the presence of isoforms switching phenomenon during acclimation to different salinities. Nevertheless, NKA enzyme activities in the gills of the SW and FW individuals were higher (p 0.05) than in BW counterparts. Immunohistochemistry revealed that Na(+)/K(+)-ATPase immunoreactive (NKA-IR) cells were mainly distributed in the interlamellar region of the gill filaments in FW groups and in the apical portion of the filaments in BW and SW groups. The number of NKA-IR cells in the gills of the FW-acclimated fish was almost similar to that of SW individuals, which exceeded that of the BW individuals. The NKA-IR cells of BW and SW were bigger in size than their FW counterparts. Besides, the relative abundance of branchial Na(+)/K(+)/2Cl(-) co-transporter showed stronger evidence in favor of involvement of this protein in hypo-osmoregulation, requiring ion secretion by the chloride cells. To the best of our knowledge, this is the first study reporting the wide salinity tolerance of E. suratensis involving differential activation of ion transporters and thereby suggesting its potential as candidate for fish farming under different external salinities.

Cell lines of Etroplus suratensis established in our laboratory were evaluated for their potential use as screening tools for the ecotoxicological assessment of tannery effluent. The cytotoxic effect of tannery effluent in three cell lines derived from eye, kidney and gill tissue of E. suratensis was assessed using multiple endpoints such as Neutral Red (NR) assay, Coomassie Blue (CB) protein assay and Alamar Blue (AB) assay. Acute toxicity tests on fish were conducted by exposing E. suratensis for 96 h to tannery effluent under static conditions. The toxic effect of tannery effluent on the survival of fish was found to be concentration and time dependent. The tannery effluent at the concentration of 15% caused 100% mortality at 96 h whereas the lower concentration (0.5%) caused 13.33% mortality. The cytotoxicity of tannery effluent was found to be similar in the three cell lines tested, independent of the toxic endpoints employed. EC(50) values, the effective concentration of tannery effluent resulting in 50% inhibition of cytotoxicity parameters after 48 h exposure to tannery effluent were calculated for eye, kidney and gill cell lines using NR uptake, AB and cell protein assays. Statistical analysis revealed good correlation with r(2)=0.95-0.99 for all combinations between endpoints employed. Linear correlations between each in vitro EC(50) and the in vivo LC(50) data, were highly significant p0.001 with r(2)=0.977, 0.968 and 0.906 for AB(50), NR(50), and CB(50), respectively.

Four novel cell lines from tissues of eye, gill, kidney and brain of Etroplus suratensis were developed and characterized. The cell lines of eye, gill, kidney and brain were sub-cultured for 245, 185, 170 and 90 passages, respectively, since 2008. These cell lines showed predominantly epithelial-like cells. Effects of temperature and foetal bovine serum concentration on the growth of these cell lines were examined and optimum growth was found at the temperature of 28° C with 20% foetal bovine serum. All the four cell lines were successfully cryopreserved and revived at different passage levels. Cell-cycle analysis of these cell lines was carried out by fluorescence-activated cell sorting. Polymerase chain reaction (PCR) products obtained from the cells and tissues of E. suratensis with primers specific to the conserved region of 16S ribosomal RNA and cytochrome oxidase I genes of E. suratensis revealed the origin of cell lines from E. suratensis. Antibodies raised against the tissues and cells of eye, kidney and gill were highly cross reacted to their specific tissue and cells of E. suratensis. Chromosomal analysis revealed that E. suratensis cells have a normal diploid karyotype with 2n = 48. The cells of these cell lines were successfully transfected with pEGFP vector DNA. The eye (IEE), gill (IEG) and kidney (IEK) cell lines were found to be susceptible to nodavirus but resistant to infectious pancreatic necrosis virus (IPNV). The cells of gill, kidney and eye were applied to test the cytotoxicity of tannery effluents.

Mercury, a global pollutant, has become a real threat to the developing countries like India and China, where high usage of mercury is reported. Mercury and other heavy metals deposited in to the aquatic system can cause health risk to the biota. The common edible fishes such as Mugil cephalus, Arius arius, Lutjanus ehrenbergii, Etroplus suratensis were collected from Cochin backwaters, Southwest India and analysed for mercury and other heavy metals (zinc, cadmium, lead and copper) in various body parts. Kidney and liver showed highest concentration of metals in most fishes. The omnivore and bottom feeder (E. suratensis) showed high concentration of mercury (14.71 mg/kg dry weight) and other metals (1.74 mg/g—total metal concentration). The average mercury concentration obtained in muscle was 1.6 mg/kg dry weight (0.352 mg/kg wet weight), which is higher than the prescribed limits (0.3 mg/kg wet weight). The concentration of other heavy metals in the muscles of fishes were found in a decreasing order Zn>Cu>Cd>Pb and are well below WHO permissible limits that were safe for human consumption. Metal selectivity index (MSI) obtained for all the metals except mercury showed that both carnivores and omnivores have almost same kind of affinity towards the metals especially Zn and Cd, irrespective of their feeding habit. The MSI values also indicate that the fishes have the potential to accumulate metals. High tissue selectivity index (TSI) values were reported for kidney, muscle and brain for all metals suggests that the metal concentration in these tissues can serve as an indication of metal polluted environment. Even if the daily intakes of Zn, Cd, Pb and Cu from these fishes are within the provisional maximum daily intake recommended by WHO/FAO, the quality is questionable due to the high hazard index obtained for mercury (>1). Fishes like E. suratensis being a favourite food of people in this region, the high consumption of it can lead to chronic disorders as this fish has high concentration of metals.

Aims: This study aimed to determine the effect of packaging [air, modified atmosphere (MA)] on microbial growth, sensory and chemical parameters and also on shelf life of fresh pearl spot (Etroplus suratensis Bloch) and on the selection of microbial association. Methods and Results: Fresh pearl spot (whole, gutted) were packaged under both 100% air and MAs (40%CO2/60% O2, 50%CO2/50%O2, 60% CO2/40%O2, 70% CO2/30% O2 and 40% CO2/30% O2/30% N2) and stored at 0°C. Microbial growth (counts of total aerobic bacteria, H2S–producing bacteria, Lactic acid bacteria, Brochothrix thermosphacta, yeast and mould), chemical spoilage indicators (pH, total volatile basic nitrogen) and sensory characteristics were monitored. Microbial changes in Pearl spot packed under 100% air and 40% CO2/30%O2/30% N2 were similar. The total volatile basic nitrogen values increased, but the values never exceeded the acceptability limit of 25 mg 100 g−1. Conclusions: MA 60% CO2 : 40%O2 was found to be better with a shelf life of 21 days whereas air stored samples had a shelf-life of 12–14 days only. Significance and Impact of the Study: Storage of pearl spot under MAs 60% CO2 : 40%O2 is a promising method to extend shelf-life. Longer shelf life expands the market potential of pearl spot and reduces waste during distribution and retail display.

Chemical partitioning of heavy metals (Fe, Mn, Cu, Cr, Pb, Zn, Ni) were determined in surface sediments of three reservoirs at the Delta region of Cauvery River, India. The abundance of metals in sediments varied in the following descending order: Fe, Mn, Cr, Zn, Cu, Ni, and Pb. Higher concentrations of Zn, Pb, Fe, Mn and Cu in exchangeable and carbonate fraction indicated toxicity risk to the biota. Therefore, to understand the extent of bioaccumulation, six commercial fish species were collected from the same sites and analyzed for heavy metals distribution in different organs. Among the metals found in fish samples, iron was observed in the highest concentration, followed by Zn, Pb, Cr, Mn, Cu, and Ni. Concentrations of Pb, Cr and Zn in many fish samples exceeded the permissible limits of Food and Agriculture Organization. The concentrations of Pb (17.7-31.7%), Cr (6.2-15.1%), Cu (15.2-30.5%) and Zn (30-40%) associated with exchangeable and carbonate fractions had significant positive correlation with the respective metal concentrations in fish. Among the fish species, Catla catla and Etroplus suratensis showed the highest accumulation of metals suggesting risk for human consumption.

The genotoxic potential of methyl parathion and phosphamidon, two commercial formulations of organophosphorus pesticides, was evaluated through induction of sister chromatid exchanges (SCE) and chromosome aberrations in fish gill tissues. Fishes exposed to the medium containing 0.05, 0.1 and 0.2 ppm of methyl parathion or 0.5, 1.0, and 2.0 ppm of phosphamidon for a duration of 96 h revealed significant increase in the number of SCE and chromosome aberrations against control values. This demonstrates the feasibility of the fish in vivo system in detecting genotoxic potential of pollutants.

Etroplus suratensis, a southern Asian cichlid, has established populations in Singapore. The fish, which was first collected in 1995, was probably introduced via the aquarium trade or through the Johor River in Malaysia. The growth, feeding and reproductive characteristics were found to follow its ecology in its native range in southern Asia, and its establishment in Singapore could be due to the similarity in environment and availability of food. Changes in intestinal length indicated diet shifts from a predominantly herbivorous to an omnivorous one as it matured. Stomach contents did not provide comparable evidence. Diet similarity to two other introduced cichlids may imply that interspecies resource competition exists. Closer monitoring is needed to determine its effects on the local environment.

A new cell line [pearlspot fin (PSF)] has been developed from caudal fin of Etroplus suratensis, a brackish/freshwater fish cultivated in India. The cell line was maintained in Leibovitz's L-15 supplemented with 10% fetal bovine serum (FBS). The PSF cell line consisted predominantly of epithelial-like cells. The cells were able to grow at temperatures between 25 degrees C and 32 degrees C with optimum temperature of 28 degrees C. The growth rate of PSF cells increased as the FBS proportion increased from 2% to 20% at 28 degrees C with optimum growth at the concentration of 10% FBS. One marine fish virus (fish nodavirus) was tested on this cell line and found not susceptible. After confluency, the cells were subcultured with a split ratio of 1:2. The cells showed epithelial-like morphology and reached confluency on the third d after subculture. Polymerase chain reaction amplification of mitochondrial 16S rRNA and COI indicated identity of this cell line with those reported from this fish species, confirming that the cell line was of pearlspot origin. The cells were successfully cryopreserved and revived at the tenth, 25th, and 35th passages. The bacterial extracellular products from Vibrio cholerae MTCC 3904 were found to be toxic to PSF. Karyotyping analysis indicated that the modal chromosome number was 48.

Effect of packaging atmosphere (air and under different modified atmospheres (MAs), 40% CO2/60% O2, 50%/50% O2, 60% CO2/40% O2, 70% CO2/30% O2 and 40% CO2/30% O2/30% N2) on the microbial and biochemical attributes of fresh pearlspot (Etroplus suratensis Bloch) stored at 0-2 degrees C was investigated. Trimethylamine nitrogen (TMA-N) and thiobarbituric acid (TBA) values remained lower than the proposed acceptability limits throughout the storage period. Results demonstrated that storage of pearlspot under air and MA 40% CO2/30% O(2)/30% N(2) resulted in growth of Enterobacteriaceae, Aeromonas and H(2)S-producing bacteria including Shewanella putrefaciens, while all other packaging atmospheres did not allow multiplication of Enterobacteriaceae and Aeromonas within 3 weeks.
