The frontal sinuses are cavities inside the frontal bone located at the junction between the face and the cranial vault and close to the brain. Despite a long history of study, understanding of their origin and variation through evolution is limited. This work compares most hominin species? holotypes and other key individuals with extant hominids. It provides a unique and valuable perspective of the variation in sinuses position, shape, and dimensions based on a simple and reproducible methodology. We also observed a covariation between the size and shape of the sinuses and the underlying frontal lobes in hominin species from at least the appearance of Homo erectus. Our results additionally undermine hypotheses stating that hominin frontal sinuses were directly affected by biomechanical constraints resulting from either chewing or adaptation to climate. Last, we demonstrate their substantial potential for discussions of the evolutionary relationships between hominin species. Variation in frontal sinus shape and dimensions has high potential for phylogenetic discussion when studying human evolution.

Reconstructing the detailed dietary behaviour of extinct hominins is challenging1—particularly for a species such as Australopithecus africanus, which has a highly variable dental morphology that suggests a broad diet2,3. The dietary responses of extinct hominins to seasonal fluctuations in food availability are poorly understood, and nursing behaviours even less so; most of the direct information currently available has been obtained from high-resolution trace-element geochemical analysis of Homo sapiens (both modern and fossil), Homo neanderthalensis4 and living apes5. Here we apply high-resolution trace-element analysis to two A. africanus specimens from Sterkfontein Member 4 (South Africa), dated to 2.6–2.1 million years ago. Elemental signals indicate that A. africanus infants predominantly consumed breast milk for the first year after birth. A cyclical elemental pattern observed following the nursing sequence—comparable to the seasonal dietary signal that is seen in contemporary wild primates and other mammals—indicates irregular food availability. These results are supported by isotopic evidence for a geographical range that was dominated by nutritionally depauperate areas. Cyclical accumulation of lithium in A. africanus teeth also corroborates the idea that their range was characterized by fluctuating resources, and that they possessed physiological adaptations to this instability. This study provides insights into the dietary cycles and ecological behaviours of A. africanus in response to food availability, including the potential cyclical resurgence of milk intake during times of nutritional challenge (as observed in modern wild orangutans5). The geochemical findings for these teeth reinforce the unique place of A. africanus in the fossil record, and indicate dietary stress in specimens that date to shortly before the extinction of Australopithecus in South Africa about two million years ago. Trace-element analysis of teeth from the hominin Australopithecus africanus, dated to 2.6–2.1 million years ago, sheds light on the weaning sequence of this species and its responses to seasonal food scarcity

Paranthropus robustus is a small-brained extinct hominin from South Africa characterized by derived, robust craniodental morphology. The most complete known skull of this species is DNH 7 from Drimolen Main Quarry, which differs from P. robustus specimens recovered elsewhere in ways attributed to sexual dimorphism. Here, we describe a new fossil specimen from Drimolen Main Quarry, dated from approximately 2.04-1.95 million years ago, that challenges this view. DNH 155 is a well-preserved adult male cranium that shares with DNH 7 a suite of primitive and derived features unlike those seen in adult P. robustus specimens from other chronologically younger deposits. This refutes existing hypotheses linking sexual dimorphism, ontogeny and social behaviour within this taxon, and clarifies hypotheses concerning hominin phylogeny. We document small-scale morphological changes in P. robustus associated with ecological change within a short time frame and restricted geography. This represents the most highly resolved evidence yet of microevolutionary change within an early hominin species.

The Cradle of Humankind (Cradle) in South Africa preserves a rich collection of fossil hominins representing Australopithecus, Paranthropus and Homo

We report here on evidence of early Homo around 1.0 Ma (millions of years ago) in the central plains of southern Africa. The human material, a first upper molar, was discovered during the systematic excavation of a densely-packed bone bed in the basal part of the sedimentary sequence at the Cornelia-Uitzoek fossil vertebrate locality. We dated this sequence by palaeomagnetism and correlated the bone bed to the Jaramillo subchron, between 1.07 and 0.99 Ma. This makes the specimen the oldest southern African hominine remains outside the dolomitic karst landscapes of northern South Africa. Cornelia-Uitzoek is the type locality of the Cornelian Land Mammal Age. The fauna contains an archaic component, reflecting previous biogeographic links with East Africa, and a derived component, suggesting incipient southern endemism. The bone bed is considered to be the result of the bone collecting behaviour of a large predator, possibly spotted hyaenas. Acheulian artefacts are found in small numbers within the bone bed among the fossil vertebrates, reflecting the penecontemporaneous presence of people in the immediate vicinity of the occurrence. The hominine tooth was recovered from the central, deeper part of the bone bed. In size, it clusters with southern African early Homo and it is also morphologically similar. We propose that the early Homo specimen forms part of an archaic component in the fauna, in parallel with the other archaic faunal elements at Uitzoek. This supports an emergent pattern of archaic survivors in the southern landscape at this time, but also demonstrates the presence of early Homo in the central plains of southern Africa, beyond the dolomitic karst areas.

Palaeomagnetic analysis was conducted on speleothems from Members 1–5 at Sterkfontein Cave, South Africa. Palaeomagnetic analysis of siltstone and speleothem from the bulk of Member 4 indicate a reversed magnetic polarity that dates the deposits and its Australopithecus africanus fossils to between 2.58 and ∼2.16 Ma. Further confirmation of this age comes in the form of two short normal polarity events correlated to the Reunion (∼2.16 Ma) and Huckleberry Ridge (∼2.05 Ma) events in speleothem capping the bulk of Member 4 and coeval with deposition of the final phase of Member 4, including A. africanus fossil Sts 5. At ∼2.16–2.05 Ma, Sts 5 is the youngest representative of A. africanus yet discovered. Palaeomagnetic analysis of the Silberberg Grotto deposits identifies a single short geomagnetic field event in flowstone overlying the StW 573 Australopithecus fossil, which is suggested to represent the Reunion event at ∼2.16 Ma. This further supports the uranium lead age estimates of 2.3–2.2 Ma for the StW 573 fossil. Based on a reversed polarity for the deposits below the skeleton it cannot be older than 2.58 Ma. If StW 573 is considered to be a second species of Australopithecus then this indicates that two species of Australopithecus are present at Sterkfontein between 2.6 and 2.0 Ma. All of the Member 5 deposits date to less than 1.8 Ma based on a comparison of palaeomagnetic, faunal, and electron spin resonance age estimates. The StW 53 fossil bearing infill (M5A) is intermediate in age between Member 4 and the rest of Member 5 (B-C) at around 1.78–1.49 Ma. The rest of Member 5 (B-C) containing Oldowan and Acheulian stone tools and Homo and Paranthropus fossils was deposited gradually between 1.40 and 1.07 Ma, much younger than previously suggested.

Speleological, paleomagnetic, mineral magnetic, and biochronological analyses have been undertaken at the Gondolin hominin-bearing paleocave, North West Province, South Africa. Two fossiliferous but stratigraphically separate sequences, GD2 and GD1/3, which were once part of a large cavern system, have been identified. Although some comparative paleomagnetic samples were taken from the GD 1, 3, and 4 localities that are currently under investigation, the research presented here focuses on the fossil-rich, in situ deposits at locality GD 2, excavated by E.S. Vrba in 1979. The GD 2 deposits are dominated by normal-polarity calcified clastic deposits that are sandwiched between clastic-free flowstone speleothems. The lower flowstone has a sharp contact with the red siltstone deposits and is of reversed polarity. The capping flowstone shows a change from normal to reversed polarity, thereby preserving a polarity reversal. While the paleomagnetic work indicates that the GD 2 fossil material was deposited during a normal-polarity period, the shortness of the sequence made matching of the magnetostratigraphy to the geomagnetic polarity time scale (GPTS) impossible without the aid of biochronology. While lacking multiple time-sensitive taxa, the recovery of specimens attributable to Stage III Metridiochoerus andrewsi is consistent with a deposition date between 1.9 and 1.5 Ma. A comparison of the magnetostratigraphy with the GPTS therefore suggests that the fauna-bearing siltstone of GD 2 date to the Olduvai normal-polarity event, which occurred between 1.95 and 1.78 Ma, and that the reversal from normal to reversed polarity identified in the capping flowstone dates to 1.78 Ma. The main faunal layers therefore date to slightly older than 1.78 Ma. Deposits from the GD 1 locality are dominated by reversed directions of magnetization, which show that this deposit is not of the same age as the faunal layers from the GD 2 locality.

Following the discovery of the "Taung Child" (Australopithecus africanus) in 1924 in the Buxton-Norlim Limeworks near Taung, the fossil-bearing deposits associated with the Dart and Hrdlička pinnacles have been interpreted as the mined remnants of cave sediments that formed within the Plio-Pleistocene Thabaseek Tufa: either as a younger cave-fill or as contemporaneous carapace caves. When combined with the Plio-Pleistocene dolomitic cave deposits from the "Cradle of Humankind," a rather restricted view emerges that South African early hominins derived from cave deposits, whereas those of east and central Africa are derived from fluvio-lacustrine and paleosol deposits. We undertook a sedimentological and paleomagnetic analysis of the pink-colored deposit (PCS) from which the "Taung Child" is purported to have derived and demonstrate that it is a calcrete, a carbonate-rich pedogenic sediment, which formed on the paleo-land surface. The deposit extends 100 s of meters laterally beyond the Dart and Hrdlička Pinnacles where it is interbedded with the Thabaseek Tufa, indicating multiple episodes of calcrete development and tufa growth. The presence of in situ rhizoconcretions and insect trace fossils (Celliforma sp. and Coprinisphaera sp.) and the distinctive carbonate microfabric confirm that the pink deposit is a pedogenic calcrete, not a calcified cave sediment. Paleomagnetic and stratigraphic evidence indicates that a second, reversed polarity, fossil-bearing deposit (YRSS) is a younger fissure-fill formed within a solutional cavity of the normal polarity tufa and pink calcrete (PCS). These observations have implications for the dating, environment, and taphonomy of the site, and increase the likelihood of future fossil discoveries within the Buxton-Norlim Limeworks.

We report the current species distribution and population estimate for the chacma baboon (Papio ursinus) in KwaZulu-Natal Province (KZN), South Africa, based on an analysis of estimated area of occupancy and estimated home range size. This estimate suggests a total population size of approximately 11,000 individuals for KZN. Much of the province is uninhabited, with a density in occupied areas of approximately 1.8 animals per km2. The current population size may be more than an order of magnitude smaller than historical population size. Chacma baboons now exhibit a highly fragmented and discontinuous distribution in KZN, with 58% of the population residing within protected areas, and more than half of these troops reside in areas >1,500 m above average sea level. The small population and highly fragmented distribution of chacma baboons in KZN, combined with rapidly increasing human population size and transformation of natural habitat, suggest this species requires greater conservation attention.

The manufacture of stone tools and their use to access animal tissues by Pliocene hominins marks the origin of a key adaptation in human evolutionary history. Here we report an in situ archaeological assemblage from the Koobi Fora Formation in northern Kenya that provides a unique combination of faunal remains, some with direct evidence of butchery, and Oldowan artifacts, which are well dated to 1.95 Ma. This site provides the oldest in situ evidence that hominins, predating Homo erectus , enjoyed access to carcasses of terrestrial and aquatic animals that they butchered in a well-watered habitat. It also provides the earliest definitive evidence of the incorporation into the hominin diet of various aquatic animals including turtles, crocodiles, and fish, which are rich sources of specific nutrients needed in human brain growth. The evidence here shows that these critical brain-growth compounds were part of the diets of hominins before the appearance of Homo ergaster/erectus and could have played an important role in the evolution of larger brains in the early history of our lineage.

Genetic and anatomical evidence suggests that Homo sapiens arose in Africa between 200 and 100 thousand years (kyr) ago, and recent evidence indicates symbolic behaviour may have appeared approximately 135-75 kyr ago. From 195-130 kyr ago, the world was in a fluctuating but predominantly glacial stage (marine isotope stage MIS6); much of Africa was cooler and drier, and dated archaeological sites are rare. Here we show that by approximately 164 kyr ago (+/-12 kyr) at Pinnacle Point (on the south coast of South Africa) humans expanded their diet to include marine resources, perhaps as a response to these harsh environmental conditions. The earliest previous evidence for human use of marine resources and coastal habitats was dated to approximately 125 kyr ago. Coincident with this diet and habitat expansion is an early use and modification of pigment, probably for symbolic behaviour, as well as the production of bladelet stone tool technology, previously dated to post-70 kyr ago. Shellfish may have been crucial to the survival of these early humans as they expanded their home ranges to include coastlines and followed the shifting position of the coast when sea level fluctuated over the length of MIS6.

Reconstructing Plio-Pleistocene African paleoenvironments is important for models of early hominin evolution, but is often hampered by low-resolution or discontinuous climatic data. Here, we present high-resolution stable oxygen and carbon isotope time series data from two flowstones (secondary cave deposits) from the South African hominin-bearing Makapansgat Valley. The age of the older of the two flowstones (Collapsed Cone) is constrained by magnetostratigraphy to approximately 4-5 Ma; the younger flowstone (Buffalo Cave) grew between 2.0-1.5 Ma, as determined by magnetostratigraphy and orbital tuning of the isotopic data. The carbon isotope data is used as a proxy for the proportion of C(4) grasses in the local environment and the oxygen isotope data reflects monsoon rainfall intensity. The carbon isotope evidence indicates that in the late Miocene/early Pliocene, the local environment was dominated by C(3) vegetation, whereas, in the Plio-Pleistocene, it was composed of a mixture of C(3) and C(4) vegetation. This suggests that C(4) grasses became a significant part of the Makapansgat Valley ecosystem at approximately 4-5 Ma, towards the end of the late Neogene global expansion of C(4) grasses. After this initial expansion, South Africa experienced further fluctuations in the proportion of C(3) and C(4) vegetation during the Plio-Pleistocene, in response to regional and global climatic changes. Most notably, the Buffalo Cave flowstone provides evidence for C(4) grass expansion at ca. 1.7 Ma that we suggest was a response to African aridity caused by the onset of the Walker Circulation in the Pacific Ocean at this time.

Reactions catalyzed by human milk N-acetyl lactosamine synthetase in the presence and absence of a-lactalbumins have been investigated by steady-state kinetics. N-Acetyl lactosamine synthesis and lactose synthesis in the absence of a-lactalbumin appear to proceed by an ordered sequential reaction, with substrates attaching in the order : Mn2+, UDP-galactose and monosaccharide. Under the conditions used (pH 7.4, 37 “C) the attachment of Mn2+ is not at thermodynamic equilibrium and it appears that the enzyme can accept either free UDP-galactose or its Mn2+ complex as substrate. Evidence is presented which suggests that the Mn2+ complex of UDP may be the final product released from the enzyme. Reactions in the presence of a-lactalbumin proceed by a similar ordered mechanism. Kinetic effects observed in the presence of human a-lactalbumin with three different monosaccharide acceptors, and in the presence of bovine a-lactalbumin with glucose, can be reasonably explained only by assuming that a-lactalbumin attaches to the enzyme immediately before monosaccharides, contrary to suggestions by other workers. It is proposed that a-lactalbumin attaches to an enzyme . Mn2+ . UDPgalactose complex at thermodynamic equilibrium, producing a new enzyme form with increased affinity for monosaccharides. The inhibitory effects of a-lactalbumin on N-acetyl lactosamine synthesis are attributed to inhibition resulting from attachment of the protein to a central complex in an alternative pathway in the reaction scheme. The kinetic effects of four a-lactalbumins with human galactosyl transferase are characterized and intrinsic differences shown to be independent of the source of galactosyl transferase. The function deduced for a-lactalbumin in the lactose synthetase system is discussed in relation to its structure and a procedure is indicated for quantitatively measuring activity differences in a-lactalbumins.

The sequence complexity of mRNA from seminal vesicles has been examined in rats of different androgen status. Total cellular poly(A)-rich RNA was isolated from normal rats, rats castrated seven days previously and castrated rats induced with testosterone in vivo for three days. Radioactive DNA complementary to each RNA preparation was synthesised with reverse transcriptase from avian myeloblastosis virus and then hybridised to its own RNA in vast RNA excess. Computer analysis of the results suggests that in each case the data are best explained by assuming the RNA to be composed of three classes distinguished by their relative abundance. In normal and induced rats, approximately 40 % of the mRNA is accounted for by about one species represented up to 240000 times per cell. Castration has little effect on the proportion of total mRNA occupied by the highly abundant mRNA but greatly decreases the cellular concentration (3 1 100 copies per cell). In normal rats the remainder of the mRNA of the seminal vesicle comprises about 17 middle abundant sequences (15800 copies per cell) and at least 6000 scarce sequences (10 copies per cell). Heterologous hybridisations are virtually indistinguishable over their entire rot range. This suggests that androgen manipulation has little effect on the total sequence complexity of seminal vesicle mRNA and is accompanied by no gross differences in the relative abundance of the majority of mRNA species. In this respect, testosterone action in the seminal vesicle differs substantially from those of other steroid hormones in other steroid-responsive systems and even from that of testosterone itself in another accessory sexual tissue of the male rat, the ventral prostate.

The role of Mn2+ in the reaction catalysed by the galactosyl transferase of bovine milk and colostrum was studied by steady-state kinetic methods at pH 7.4 and 37°C. The association constants for the binding of Mn2+ by UDP-galactose and by the buffer species used (3-[N-morpho-lino]propanesulphonate) were measured by electron spin resonance spectroscopy and the concentrations of all the species present in the substrate mixture of Mn2+· UDP-galactose and N-acetyl-glucosamine were calculated. Previous evidence indicated that high Mn2+ concentrations activate the reaction beyond what is expected from the Michaelis-Menten equation. At such levels (up to 40 mM), the concentration of free Mn2+ was found to be proportional to total Mn within 3%. Experimental data produced as a function of total Mn concentration and total UDP-galactose (i.e. free UDP-galactose plus MnUDP-galactose) concentration could be interpreted in terms of rate equations written in relatively simple form with free Mn2+ and total UDP-galactose concentrations as variables, corresponding to a large variety of kinetic models involving Mn2+, UDP-galactose and MnUDP-galactose as substrates. A group of related mechanisms could not be eliminated at this stage and experiments were carried out in which the concentrations of two of the substrates were varied at either a constant ratio or a constant product depending on how they were involved in the Mn2+: UDP-galactose association reaction. A choice between the two final mechanisms was made by applying statistical non-linear least-squares methods. The most likely mechanism was one in which, after the addition of Mn2+ to the enzyme, the substrates UDP-galactose and MnUDP-galactose added as alternatives to produce two branches, in each of which was added N-acetylglucosamine followed by the release of N-acetyllactosamine, then the Mn2+ complex of UDP, and finally (in the branch in which MnUDP-galactose had been the substrate) free Mn2+.

RNA complexity analysis of poly(A)-containing RNA isolated from the lactating guinea-pig mammary gland demontrates that the complexity within the nuclear population was three--four-fold greater than that in the equivalent post-nuclear polyribosomal population [see Craig et al., Biochem. J. (1979) 181, 737-756]. Up to 21 000 different sequences distributed in two abundance groups were detected in the nucleus, whereas 5000 sequences distributed in three abundance groups were present in the post-nuclear population. All poly(A)-containing RNA sequences present in the post-nuclear fraction could be detected in the nuclear poly(A)-containing population. Analysis of the relative distribution of the three post-nuclear abundance populations within the nuclear poly(A)-containing RNA population demonstrates that the abundant and moderately abundant post-nuclear sequences were present a similar concentrations within the nucleus, and comprised the abundant nuclear population. The abundant and moderately abundant post-nuclear sequences were present at 62200 and 785 copies of each sequence/cell in the post-nuclear fraction respectively, and 44 and 118 copies of each sequence/cell in the nuclear fraction respectively. The scarce post-nuclear sequences (18.5 copies of each sequence/cell) were also present at low levels in the nuclear fraction (0.1 copy of each sequence/cell). the results are discussed in terms of the post-transcriptional regulation of gene expression in the lactating guinea-pig mammary gland.

K m values of β - N -acetylgucosaminidase ( 2- acetamido -2- deoxy -β- d - glucoside acetamidodeoxyglucohydrolase EC 3.2.1.30), β - N -acetylgalactosaminidase (EC 3.2.1.53), β-galactosidase (β- d -galactoside galactohydrolase EC 3.2.1.23) and α- l -fucosidase (α- l -fucoside fucohydrolase EC 3.2.1.51) of distal colonic tumours, induced in rats by 1,2-dimethylhydrazine, were found to be significantly different compared with the values for the enzymes of the colonic mucosa of the control and tumour-bearing animals and of the proximal colonic tumours. The inhibition kinetics data also showed a significant difference between the enzymes of the distal colon tumours and of other experimental tissues. The data on the effect of pH on enzyme kinetics (p K values) showed no significant difference in the catalytic groups of the active centres of enzymes from tumours and from the control colonic mucosa. Tumour β - N -acetylglucosaminidase and β - N -acetylgalactosaminidase compared with the enzymes from other experimental tissues were found to be different in their thermal inactivation kinetics. K m values of 14 days old foetal intestinal β - N -acetylglucosaminidase and β - N -acetylgalactosaminidase were significantly different from the values obtained for the adult mucosal enzymes but were similar to those of the distal colonic tumour enzymes.

Rat colonic beta-N-acetylhexosaminidase (2-acetamido-2-deoxy-beta-D-glucoside acetamidodeoxyglucohydrolase, EC 3.2.1.30) has been separated into three forms by DEAE-cellulose chromatography with an increasing salt gradient. It was not possible to separate the glucosaminidase activity from the galactosaminidase activity by a variety of chromatographic procedues, but the ratio of the two specific activities varied during purification. The pH optima were however identical, for both activities and all three forms. Kinetic measurements including inhibition by substrate analogues showed differences between the two activities as well as among the three forms. A common active site model was inconsistent with the results. Data from mixed substrate experiments were consistent with a model wherein the two activities reside in seperate active sites, each able to be inhibited by the substrate for the other site. The effect of acetate and SH reagents confirmed the two-site model. Treatment with neuraminidase, thimerosal, p-hydroxymercuribenzoate, HgCl2 and AgNO3 or heating at 50 degrees C did not produce any effect on the A form that could be identified as a conversion to the B form. Measurement of the effects on both activities supported the two-site model. It is concluded that the relationship between the A and B forms in the rat colonic mucosa hexosaminidases must be different from that reported for such enzymes from other sources.

The haemocyanin from the freshwater gastropod Lymnaea stagnalis was digested with proteolytic enzymes under conditions where it existed as whole (native) molecules (mol.wt. approx. 9 × 10(6)), or as one-tenth molecules. Digestion of whole molecules yielded a fragment of mol.wt. approx. 110,000 believed to correspond to the ‘collar’ of the molecule, and an aggregate some 20–30 times the size of the original native molecule formed by end-to-end polymerization of the molecule after removal of the collar. Digestion of one-tenth molecules yielded a mixture of products that could be separated into three fractions by gel filtration. Analysis of these by sodium dodecylsulphate/polyacrylamide-gel electrophoresis revealed that they typically contained two or three components. The collar fragment was present as a component of the intermediate-molecular-weight fraction, and it dissociated on sodium dodecyl sulphate/polyacrylamide gels to give two bands corresponding to apparent mol.wts. 65,000 and 60,000. The c.d. spectra of the separated fractions were recorded and fitted with Gaussian curves by a computer procedure. The fractions each possessed distinct c.d. spectra, by which they could be identified: the collar-fragment c.d. and absorption spectra showed the most striking differences compared with those of the other fragments. The results were interpreted in terms of the postulated existence, within the haemocyanin molecule, of multi-domain structures, each comprising a single polypeptide chain of mol.wt. 200,000–300,000.

Bovine pancreatic ribonuclease is the only enzyme for which the primary structure is known (Hirs, Moore and Stein, 1960; Smyth, Stein and Moore, 1962; Potts et al., 1962; Gross and Witkop, 1962). The nature of the secondary and tertiary structure is, as yet, little understood although models have been proposed (Scheraga, 1960; Parks, 1960; Panar and Westheimer, 1961), but it is wise to postpone discussion of the relationship between catalytic function and structural detail until definite information is available. The chemistry of ribonuclease has been adequately reviewed (Scheraga and Rupley, 1962; Westheimer, 1962) and only data of direct revelance to the catalytic mechanism will be discussed in this paper.

The formation of individual functional mRNA sequences in higher organisms requires many steps in addition to transcription. These include RNA splicing, polyadenylation, base modification, transport from nucleus to cytoplasm and assembly into polyribosomes. Various control mechanisms must also operate. These will function on a quantitative basis to account for the differing frequency of the various classes of cytoplasmic mRNAs, and also on a qualitative basis, because in higher organisms not all the nuclear poly (A)-containing RNA molecules are found in a cytoplasmic poly(A)-containing RNA population from the same tissue1–10. During our studies on the mechanisms controlling the accumulation of the poly (A)-containing RNA sequences which occur with high and moderately high frequency in the cytoplasm of the lactating guinea pig mammary gland, it became apparent that >75% of the 20,000 or so poly (A)-containing nuclear RNA sequences were not found in the cytoplasmic poly(A)-containing RNA fraction4. Here we demonstrate that many of the poly(A)-containing RNA sequences retained in the nucleus of the lactating guinea pig mammary gland are also present in the nucleus and cytoplasm of the liver of the male guinea pig. These observations provide new evidence for a predominant role of post-transcriptional mechanisms in the regulation of structural gene expression in guinea pig tissue.

Male Wistar rats were treated for three weeks with cyproterone (1·7 or 5·1 mg/day) or cyproterone acetate (2 or 6 mg/day). The adrenal weights of animals treated with either dose of cyproterone acetate were significantly less (P < 0·001) than those of untreated animals. In contrast, the adrenal weights of animals treated with cyproterone did not differ from those of the controls. The concentrations of corticosterone in the plasma were significantly less (P < 0·001) in both groups treated with cyproterone acetate compared with those of the controls; only the higher dose of cyproterone reduced the plasma concentration of corticosterone (P < 0·001). Cyproterone acetate inhibited the rat adrenal 3β-hydroxysteroid dehydrogenase–5-ene,4-ene-isomerase complex in vitro, with both pregnenolone and dehydroepiandrosterone as substrates. Analysis of the reaction rates suggested an uncompetitive mode of inhibition. These results suggest that in rats the antiandrogens cyproterone and cyproterone acetate may provoke adrenal insufficiency by inhibition of steroid biosynthesis. Furthermore, indirect evidence from the mass and morphology of the adrenal suggests that cyproterone acetate may also suppress production or secretion of ACTH by the pituitary gland.
