Effects of subchronic administration of antioxidants against pulmonary damage mediated by cigarette smoke were investigated in rats. Rats were continuously received ascorbic acid, N-acetylcysteine and ginseng extract together drinking water from day 25 after birth. After 30 days of antioxidant supplementation, rats were exposed to cigarette smoke generated from six cigarettes (11 mg tar) for 20 min per day throughout 30 days, and then several biochemical markers related to the redox status in vivo were analyzed in the respiratory system. The cigarette smoke induced mild histological changes in trachea and lungs. The activity of superoxide dismutase (SOD) in the lung was significantly increased, and catalase and glutathione peroxidase activities were increased less than SOD, but total sulfhydryl compounds (Total-SH) content was decreased by cigarette smoking. In spite of the increase in activities of antioxidant enzymes, the inhibitory capacity of lung preparations on in vitro lipid peroxidation using ox brain homogenates was decreased and the change in the capacity was not related to the changes of these intracellular enzymes activities, but with the content of Total-SH. On the other hand, the content of thiobarbituric acid reactive substances and the ratio of elastase to anti-protease in the lung homogenates were significantly increased. Supplementation of antioxidants, however, effectively attenuated all of such alterations induced by cigarette smoke. These results indicate that although cigarette smoking induces antioxidant enzymes in the lung as a self defense mechanism, it seems to be not sufficient to protect the pulmonary system, and that chronic antioxidant feeding could be effective to reduce pulmonary damage induced by free radicals.

Mushrooms are popular both as food and as a source of natural compounds of biopharmaceutical interest. Some mushroom-derived compounds such as beta-glucan have been shown to be immunostimulatory; this study explores the anti-inflammatory properties of hispidin analogues derived from the mushroom, Inonotus xeranticus. We sought to identify the molecular mechanism of action of these hispidin analogues by determining their effects on lipopolysaccharide (LPS)-mediated inflammatory responses in a macrophage cell line.The production of inflammatory mediators was determined by Griess assay, reverse transcription-PCR and ELISA. The inhibitory effect of davalliactone on LPS-induced activation of signalling cascades was assessed by western blotting, immunoprecipitation and direct kinase assay.In activated RAW264.7 cells, davallialactone strongly downregulated LPS-mediated inflammatory responses, including NO production, prostaglandin E2 release, expression of proinflammatory cytokine genes and cell surface expression of co-stimulatory molecules. Davallialactone treatment did not alter cell viability or morphology. Davallialactone was found to exert its anti-inflammatory effects by inhibiting a signalling cascade that activates nuclear factor kappa B via PI3K, Akt and IKK, but not mitogen-activated protein kinases. Treatment with davallialactone affected the phosphorylation of these signalling proteins, but not their level of expression. These inhibitory effects were not due to the interruption of toll-like receptor 4 binding to CD14. In particular, davallialactone strongly inhibited the LPS-induced phosphorylation and kinase activity of Src, implying that Src may be a potential pharmacological target of davallialactone.Our data suggest that davallialactone, a small molecule found in edible mushrooms, has anti-inflammatory activity. Davallialactone can be developed as a pharmaceutically valuable anti-Src kinase agent.

The lack of appropriate, clinically relevant, cell-based model systems has limited prostate cancer research and the development of new therapeutic modalities. Here we report the isolation and characterization of a new adherent prostate cancer cell line, derived from the dura mater of a cancer patient.Prostate cancer tissue was harvested at autopsy from a metastatic lesion to the dura mater of a patient with hormone refractory prostate cancer. This tissue was xenografted into SCID mice and later harvested and plated on tissue culture dishes. For characterization, soft agar clonegenic assay, in vivo xenograft growth, in vitro doubling time, karyotype analysis, immunocytochemistry for cytokeratin-18, androgen receptor, and PAP (prostatic acid phosphatase) expression, RT PCR for PAP, PSMA (prostate specific membrane antigen), expression and northern and western blot analysis to determine expression of Rb and p53, were performed.DuCap grows in vitro (passage 55), forms colonies in soft agar, produces tumors in SCID mice (xenograft passage 12), and is androgen sensitive. DNA content was hypertriploid. PSA was detected in mouse serum and media. Cells were AR, PAP and cytokeratin-18 positive by immunocytochemistry. PSMA and PAP were detected by RT-PCR. AR, P53, and Rb were expressed in Northern blot analysis. P53 protein was detected in Western blot analysis but Rb protein was not.This cell line exhibits many phenotypic characteristics of clinical prostate carcinoma, including expression of PSA, PSMA, PAP and AR.

Expression of the stimulatory G protein, G(S)alpha, can vary over a 3-fold range in human tissues and in rodent central nervous system. In fact, the offspring of alcoholics have higher levels of G(S)alpha expression in certain tissues compared with the offspring of nonalcoholics. The aim of this research was to test the hypothesis that a causal relationship exists between the level of expression of G(S)alpha and induction of the adenylyl cyclase (AC) cascade. The methodology employed transient transfection of HEK 293 cells with a cDNA for the 52-kDa form of G(S)alpha under regulation by inducible metallothionein promoters. Transfectants were exposed to varying concentrations (0-125 microM) of zinc sulfate that produced a 3-fold range of membrane G(S)alpha expression. The range of G(S)alpha expression produced was found to mimic a physiologically relevant spectrum of G(S)alpha expression in membranes derived from human tissues and rat brain. It was observed that induction of G(S)alpha expression increased constitutive as well as stimulated cAMP accumulation. Moreover, induction of G(S)alpha expression increased events distal to the accumulation of cAMP including the phosphorylation of the transcription factor, cAMP response element binding protein and transcriptional activation of cAMP-dependent reporter genes. In summary, these studies show that the amount of G(S)alpha expression has a marked impact on the level of activity of the AC cascade from the membrane through to the nucleus. It is hypothesized that individuals who differ in G(S)alpha expression may also differ in the expression of certain cAMP-dependent genes.

Fibroblast growth factor 2 (FGF-2) is not only a potent mitogen, it is a modulator for corneal endothelial cells. To define how the modulation activities of FGF-2 are mediated, we used pharmacologic inhibitors to examine the association of phospholipase C-gamma 1 (PLC-gamma) with FGF receptor or with cytoskeleton.Cell proliferation was determined either by the incorporation of 3H-thymidine into DNA or by counting cell numbers in the absence or presence of the inhibitors. The protein expression was analyzed by immunoprecipitation and immunoblot analysis. Cell shape change was determined by phase-contrast microscopy.FGF-2 stimulated DNA synthesis, whereas genistein inhibited the FGF-2-mediated cell proliferation in a dose-dependent manner, regardless of the concentration of FGF-2. The PLC-gamma 1 specific antisense oligonucleotide primer was able to inhibit cell proliferation by 25% in the absence of FGF-2; however, the antisense primer was not able to override the action of FGF-2. Fibroblast growth factor receptor was phosphorylated on treatment of the cells with FGF-2; however, 24-hour treatment with the growth factor significantly reduced phosphorylation of the receptor. Phospholipase C gamma 1 appears to be abundant in cytoplasm in the absence and presence of FGF-2, and a minor portion of the molecule is translocated to membrane after treatment with FGF-2; genistein inhibited the translocation. When the cytoskeleton fraction of the normal and the modulated corneal endothelial cells was immunoprecipitated with PLC-gamma 1 antibodies, PLC-gamma 1, actin, and vinculin were coprecipitated in both cell cultures. Phospholipase C gamma 1 associated with cytoskeleton was phosphorylated on treatment of the cells with FGF-2. In the presence of FGF-2 of the modulated cells, cytochalasin B, which did not revert the modulated cell morphology, abolished the association of PLC-gamma 1 with actin and vinculin; colchicine, which did revert the modulated cell shape to the polygonal shape, did not block the association of these three molecules. Interestingly, colchicine slightly enhanced the stimulatory effect of FGF-2 on corneal endothelial proliferation in contrast to the effect of cytochalasin B, which slightly decreased the FGF-2 action on cell proliferation.The association of PLC-gamma 1 with cytoskeleton plays a role in cell proliferation, whereas the association of PLC-gamma 1 with actin and vinculin has no effect on cell shape changes. These findings indicate that FGF-2 appears to use distinct signaling pathways for cell proliferation and cell shape changes in corneal endothelial cells.

The influence of food restriction (FR) on the induction of liver microsomal cytochrome P-450 (P-450) was examined in rats, FR by 40% for 6 weeks caused increase in the levels of P-450, cytochrome b5 (b5), NADPH P-450 reductase, NADH b5 reductase, P-450 dependent testosterone 6 beta-, 16 alpha- and 16 beta-hydroxylases (P0.05). Treatment of 3-methylcholanthrene (MC) to food restricted rats (FR rats) induced more P-450, but phenobarbital (PB) induced it less. MC-inducible P-450 from FR rats did not increase its specific ethoxycoumarin O-deethylase activity but it increased testosterone 2 alpha-hydroxylase activity, and the maximum absorption of its CO complex was shifted from 448 nm to 450 nm. PB-inducible P-450 showed less increase in the activities of its specific benzphetamine N-demethylase and it rather significantly decreased testosterone 16 alpha-hydroxylase activity (P0.05). These results indicate that the drugs did not induce their unique types of P-450 in FR rats. Such alterations in P-450 induction in FR rats seem to be closely related to the level of P-450 and its related components enhanced by FR, and it could result in modulation of mixed function oxidase system.

The metabolism of 16 alpha-hydroxytestosterone by cytochrome P-450 system was studied with two different inductions. At least 5 different metabolites with higher polarity were produced from 16 alpha-hydroxytestosterone. MT-B, one of their products, appeared to be specifically increased by 3-methylcholanthrene- and phenobarbital-inducible P-450, respectively. This suggests that monohydroxylated testosterones are further metabolized by the P-450 system and that the reactions are isozyme specific. This can contribute at least in part to the diversity of steroid metabolism by P-450.

Summary A highly conserved acrosomal antigen reactive to a monoclonal antibody (HS-63), generated against human sperm, was purified to homogeneity with a combination of conventional procedures and immunoaffinity chromatography using a soluble extract of mouse and rabbit testes. The molecular weight of the purified antigen was 42–50 kD when analysed by sodium dodecylsulphate polyacrylamide gel electrophoresis. The high specificity of the purified antigen to monoclonal antibody HS-63 was shown by indirect immunofluorescent inhibition assay, enzyme-linked immunosorbent assay, Western blot analysis and radioimmunosorbent assay. The purified antigen was used for isoimmunization of mice and rabbits. Following successive immunizations, antisera of high titres were raised and reacted specifically with antigen on the sperm acrosome and in testes of several mammalian species, but not with somatic tissues. These isoimmune sera exhibited strong inhibition on mouse in-vitro fertilization and human sperm penetration of zona-free hamster eggs. The results of this study suggest that the sperm-specific acrosomal antigen reacting with HS-63 could be a good candidate for the development of immunocontraceptive vaccines in humans and in other animals.

Serum, peripheral and thoracic lymph concentrations of tobramycin, dibekacin and amikacin were determined at various time intervals up to 6 hours following intramuscular administration of 4 mg/kg to dogs. Peak serum concentrations of each antibacterial agents occurred within one hour after administration. The peak lymphatic concentrations occurred in 1.5 approximately 2 hours after administration. Following peak peripheral lymph concentration, the concentration in peripheral lymph exceeded than that of serum. The concentrations of each antibacterial agent in thoracic lymph were nearly equal to serum concentration. Peak peripheral lymph concentrations were always lower than the peak serum concentrations. 2. Serum and pancreatic fluid concentrations of tobramycin, dibekacin and amikacin were determined at various time intervals up to 6 hours following intramuscular administration of 4 mg/kg to dogs. Peak pancreatic fluid concentrations of each antibacterial agents occurred within 1.5 hours after administration. The concentrations rate of tobramycin to the pancreatic fluid was superior to those of dibekacin and amikacin.

This tissue was aseptically xenografted into SCID mice and later harvested and plated on tissue culture dishes. For characterization, soft agar clonegenic assay, in vivo xenograft growth, in vitro doubling time, karyotype analysis, immunocytochemistry for cytokeratin-18 expression immunochemistry for PSA (prostate specific antigen), RT PCR for PAP (prostatic acid phosphatase) and northern blot and western blot analysis to determine expression of Rb and p53, were performed. Androgen receptor expression was measured by transient transfection with a luciferase reporter construct.VCaP cells are immortal in vitro and can be passaged serially in vivo. They express large quantities of prostate specific antigen (PSA). This cell line also expresses prostatic acid phosphatase (PAP), cytokeratin-18 and the androgen receptor, and is androgen sensitive in vitro and in vivo.This cell line was derived from a metastatic tumor to the vertebrae of a prostate cancer patient. It exhibits many of the characteristics of clinical prostate carcinoma, including expression of PSA, PAP, and AR. We believe that VCaP will be a useful addition to the existing models of prostate cancer, and enable more advanced study of the mechanisms of prostate cancer progression and metastasis.

The crossreactivity between bull spermatozoa and monoclonal antibodies initially raised against mouse spermatozoa and human spermatozoa was tested by indirect immunofluorescent assay. The three anti-human spermatozoa monoclonal antibodies examined (HSK-9, HS-11, HS-63) crossreacted with methanol-fixed bull spermatozoa, whereas the anti-mouse spermatozoa monoclonal antibodies (MS-4 and MS-7) did not. A separate experiment was conducted to determine the binding ability of HSK-9, HS-11 and HS-63 with live (fresh) bull spermatozoa incubated (39 degrees C in CO2 incubator) in a capacitation medium (modified Tyrode's supplemented with 10 micrograms heparin ml-1). The binding of the monoclonal antibodies to the intra-acrosomal antigens of live bull spermatozoa was determined at 0, 2, 4, 6 and 8 h of incubation. At the beginning of incubation, binding was minimal (3.2 +/- 1.7%), but a much higher percentage of spermatozoa exhibited fluorescent staining after 2 h. The maximal binding was observed after incubation for 8 h (72.0 +/- 8.2%). The third experiment was performed to determine binding of HS-11 to frozen-thawed spermatozoa and to test whether there was any variation among bulls in HS-11 binding to spermatozoa, and to assess whether such binding is an indication of sperm capacitation. Frozen-thawed semen samples from five bulls were assessed for antibody binding after 0, 2, 4 and 6 h of incubation. Maximal binding was observed at 4 h. Lysophosphatidylcholine (100 micrograms ml-1) induced acrosome reaction assay was performed to assess sperm capacitation at various intervals.(ABSTRACT TRUNCATED AT 250 WORDS)

Lactate dehydrogenase-X (LDH-X), a glycolytic enzyme found only in mammalian testes and spermatozoa, is encoded by a single gene (Ldh-x) in the mouse haploid genome. Several studies have demonstrated that LDH-X is associated with germ cells at specific stages of development. We have examined the expression of the Ldh-x gene during mouse spermatogenesis and testis maturation using in situ mRNA hybridization and immunocytochemistry. The results showed that transcription and translation of the Ldh-x gene are initiated at the pachytene stage of germ cell differentiation. However, although the amount of LDH-X protein increased as the germ cells progressed to maturation, its mRNA level was greatly decreased. These observations were confirmed by Northern analysis of total RNA derived from fractionated spermatogenic cells and developing testes. Furthermore, Northern studies also indicated two sizes of Ldh-x transcripts among different populations of spermatogenic cells in mature mouse testis.
