Anthracnose caused by Colletotrichum species is the most important disease of chayote (Sicyos edulis) in Brazil. The etiology of chayote anthracnose has been assigned to the species C. orbiculare, an important plant pathogenic fungus also reported as the causal agent of anthracnose in other cucurbits worldwide. However, there is no recent survey of the Colletotrichum species causing anthracnose in chayote in Brazil. In this study, Colletotrichum isolates associated with anthracnose on the fruit and leaves of chayote, from various producing regions in Brazil, were collected and identified. Haplotype analysis based on sequences of the β-tubulin genomic region (TUB2) of 44 Colletotrichum isolates was carried out as a first measure of genetic diversity. A subset of 22 isolates were sequenced using the partial sequences of actin (ACT), glyceraldehyde-3-phosphate dehydrogenase (GAPDH), and the rDNA ITS (ITS) region. Maximum likelihood analysis was performed using the concatenated sequences. The multilocus sequence analysis revealed four previously described species, Colletotrichum chrysophilum, C. menezesiae, C. plurivorum, and C. karsti, and two novel species, named C. cucurbitacearum and C. sicyi. All species were able to induce typical symptoms of anthracnose in chayote fruits but varied in their aggressiveness. The species C. menezesiae and C. sicyi were the most aggressive, while C. plurivorum was the least aggressive. The species C. orbiculare was not found to cause chayote anthracnose in Brazil. Anthracnose caused by Colletotrichum species is a serious disease of loquat (Eriobotrya japonica), but the species of Colletotrichum that are associated with the disease in Japan have not been adequately surveyed. In this study, 46 isolates were isolated from diseased leaves and fruits of loquat in Kagawa and Tokushima prefectures in 2017 and 2018. On the basis of morphological features and molecular approaches, seven Colletotrichum species were identified: C. fioriniae, C. nymphaeae, C. karsti, C. aenigma, C. fructicola, C. gloeosporioides s. s., and C. siamense. The most prevalent species was C. karsti (43.5% of total species), followed by C. fioriniae (21.7%) and C. gloeosporioides s. s. (13.0%). All identified species were confirmed to cause loquat anthracnose by fulfilling Koch's postulates. The loquat isolates of C. fioriniae, C. fructicola, and C. siamense also caused symptoms on all tested plants of kiwifruit, satsuma mandarin, persimmon, and peach. Anthracnose, caused by several Colletotrichum species, is a major restricting factor for guava production, but no comprehensive study of the causal agent has been conducted from the plant host centre of diversity. This work characterizes isolates from guava, mainly from the fruit, but also leaves and flowers, representing most Brazilian physiographies according to the partial sequences of the rDNA‐ITS, ApMAT, TUB2, HIS3 and GAPDH gene regions. In addition, the pathogenicity and aggressiveness to fruits of two widely planted guava varieties (SLG and RM) are described. Guava‐derived Colletotrichum isolates were found in five complexes: gloeosporioides, acutatum, boninense, gigasporum and orchidearum. The gloeosporioides complex was the most prevalent (81%), followed by the acutatum complex (14%). A total of 16 Colletotrichum species were naturally associated with guava anthracnose: C. aeschynomenes, C. asianum, C. chrysophilum, C. fructicola, C. gigasporum, C. gloeosporioides, C. karsti, C. melonis, C. musae, C. nymphaeae, C. paranaense, C. siamense, C. sojae, C. syzygicola, C. theobromicola and C. tropicale. Apart from C. nymphaeae and C. gloeosporioides, all the remaining 14 taxa are reported for the first time in P. guajava. The most aggressive species belonged to the C. gloeosporioides complex. C. siamense was the most prevalent, especially in warmer regions, followed by C. chrysophilum, mostly in temperate environments. The most aggressive species were C. siamense, C. chrysophilum, C. fructicola and C. tropicale. Fruits of the variety SLG were consistently more resistant to anthracnose than the fruits of RM. Euonymus japonicus plays important roles in the process of urban landscape construction as an evergreen shrub. Anthracnose is an important limiting factor affecting the healthy growth of E. japonicus, seriously influencing improvement of urban landscape ecosystems. However, the Colletotrichum species associated with anthracnose on E. japonicus are unclear. In this study, diseased leaves of E. japonicus with typical anthracnose symptoms were collected in five nurseries in Beijing, China. Among 45 Colletotrichum isolates obtained, there were two distinct morphotypes, which were identified as C. aenigma or C. siamense by morphological characteristics and multilocus phylogenetic analysis. The growth rates of these two species were determined at different incubation temperatures, and the results showed that both species grew well at 10–35°C, particularly at 25 or 30°C, while C. aenigma isolates were adapted to high growth rates at a wider range of temperatures than C. siamense. Pathogenicity assays indicated that the two species showed varying degrees of pathogenicity on E. japonicus. Wounding was conducive to the pathogenicity of C. aenigma or C. siamense on E. japonicus either by inoculation with mycelial discs or conidial suspensions. This study presents the first report of C. aenigma causing anthracnose on E. japonicus worldwide and of C. siamense in north China causing anthracnose on E. japonicus. The morphological features of Colletotrichum spp. associated with anthracnose on E. japonicus were also compared in detail in this study. Winterberry holly (Ilex verticillata) is an economically valuable landscaping ornamental plant. Serious outbreaks have been reported, in its leaf tips curl upward, irregular black brown spots appear on leaves, and extensive defoliation is commonly observed. The incidence in Hangzhou was estimated at 50% and resulted in large economic losses for growers in 2018. Samples were collected from the main cultivation area in Zhejiang Province. In total, 11 fungal isolates were obtained from diseased leaves through a single-spore purification method, and isolate LVY 9 exhibited strong pathogenicity. Based on morphology and molecular phylogenetic analyses based on multilocus sequence typing of the glyceraldehyde-3-phosphate dehydrogenase (GAPDH), internal transcribed spacer (ITS) regions, actin (ACT), calmodulin (CAL), and chitin synthase (CHS-1) genes, we identified the pathogen as Colletotrichum siamense, causative agent of anthracnose of winterberry holly. Anthracnose caused by Colletotrichum spp. is one of the major problems in mango production worldwide, including Thailand. All mango cultivars are susceptible, but Nam Dok Mai See Thong (NDMST) is the most vulnerable. Through a single spore isolation method, a total of 37 isolates of Colletotrichum spp. were obtained from NDMST showing anthracnose symptoms. Identification was performed using a combination of morphology characteristics, Koch's postulates, and phylogenetic analysis. The pathogenicity assay and Koch's postulates on leaves and fruit confirmed that all Colletotrichum spp. tested were causal agents of mango anthracnose. Multilocus analysis using DNA sequences of internal transcribed spacer (ITS) regions, β-tubulin (TUB2), actin (ACT), and chitin synthase (CHS-1) was performed for molecular identification. Two concatenated phylogenetic trees were constructed using either two-loci of ITS and TUB2, or four-loci of ITS, TUB2, ACT, and CHS-1. Both phylogenetic trees were indistinguishable and showed that these 37 isolates belong to C. acutatum, C. asianum, C. gloeosporioides, and C. siamense. Our results indicated that using at least two loci of ITS and TUB2, were sufficient to infer Colletotrichum species complexes. Of 37 isolates, C. gloeosporioides was the most dominant species (19 isolates), followed by C. asianum (10 isolates), C. acutatum (5 isolates), and C. siamense (3 isolates). In Thailand, C. gloeosporioides and C. acutatum have been reported to cause anthracnose in mango, however, this is the first report of C. asianum and C. siamense associated with mango anthracnose in central Thailand. Colletotrichum is regarded as one of the 10 most important genera of plant pathogens in the world. It causes diseases in a wide range of economically important plants, including peaches. China is the largest producer of peaches in the world but little is known about the Colletotrichum spp. affecting the crop. In 2017 and 2018, a total of 286 Colletotrichum isolates were isolated from symptomatic fruit and leaves in 11 peach production provinces of China. Based on multilocus phylogenetic analyses (ITS, ACT, CAL, CHS-1, GAPDH, TUB2, and HIS3) and morphological characterization, the isolates were identified to be C. nymphaeae, C. fioriniae, and C. godetiae of the C. acutatum species complex, C. fructicola and C. siamense of the C. gloeosporioides species complex, C. karsti of the C. boninense species complex, and one newly identified species, C. folicola sp. nov. This study is the first report of C. karsti and C. godetiae in peaches, and the first report of C. nymphaeae, C. fioriniae, C. fructicola, and C. siamense in peaches in China. C. nymphaeae is the most prevalent species of Colletotrichum in peaches in China, which may be the result of fungicide selection. Pathogenicity tests revealed that all species found in this study were pathogenic on both the leaves and fruit of peaches, except for C. folicola, which only infected the leaves. Persimmon (Diospyros kaki) anthracnose is a major threat in production areas worldwide. Most of the studies are focused on Colletotrichum horii , but other species have been reported as well. The association of distinct Colletotrichum species present in Brazilian persimmon production regions as well as their host ranges are yet elusive. The aims of this work were to identify and characterize Colletotrichum species associated with the persimmon anthracnose. In a survey performed in four production regions of Brazil, 88.7% and 11.3% out of 231 isolates were identified as members of Colletotrichum gloeosporioides species complex (Cgc) or Colletotrichum acutatum species complex (Cac), respectively. A subset of 18 isolates were identified through multilocus phylogenetic analysis, using ITS-rDNA region and two loci, namely GAPDH and TUB2. This study revealed the presence of four species: C. horii (38.8%) and Colletotrichum fructicola (27.7%) from the Cgc and Colletotrichum nymphaeae (27.7%) and Colletotrichum melonis (5.8%) , from the Cac. Additionally, 13 isolates were selected for morphological, physiological, and pathogenic analyses. Contrasting characteristics were observed among species of the Cgc and Cac complexes. The optimal temperature for mycelial growth and germination were higher for Cgc species. The percentage of appressoria melanisation also varied across complexes. All the identified species were able to cause anthracnose-like symptoms on persimmon fruit, leaves, shoots, and sepals. Colletotrichum species from persimmon were also able to infect apple and pear. Anthracnose caused by species of Colletotrichum is considered one of the main postharvest diseases for avocado. In this study, Colletotrichum isolates associated with avocado anthracnose, collected in different states of Brazil, were evaluated through phylogenetic analysis, morphological characterization, and pathogenicity assays. Moreover, the events during pathogen infection of avocados were examined by scanning electron microscopy. To assess the genetic diversity of 54 Colletotrichum isolates, partial sequence analysis of the gene gapdh was performed. According to the generated groupings and the geographical origins of isolates, a subset of 14 strains was selected for performing multilocus phylogeny analysis (using sequences of gapdh, act, tub2, and ApMat). Two species previously described were identified: C. siamense belonging to the C. gloeosporioides species complex and Colletotrichum karstii belonging to the C. boninense species complex. All Colletotrichum strains evaluated caused typical symptoms of anthracnose in avocado fruits. Conidia of the most virulent strain germinated between 6 and 12 hr after inoculation (hai). Penetration through wounds occurred 48 hai, tissue colonization occurred between 144 and 240 hai, and sporulation took place at 240 hai via the production of an acervulus, conidiophores, and conidia. The findings shed light on the aetiology of avocado anthracnose in Brazil and provide a better understanding of the infection process of this pathogen, which may assist in the development of disease management strategies. Pomegranate is a fruit rich in vitamins and secondary metabolites used in traditional medicine and industry. However, production losses have been associated the anthracnose, disease caused by Colletotrichum species. This is an important disease of the pomegranate, as it affects the yield and the quality of the fruits. The present study aimed to investigate which species of Colletotrichum are associated with anthracnose disease in pomegranate in the Northeast region of Brazil, using multi-locus phylogenetic analysis and morpho-cultural characteristics. The total DNA extracted was amplified with GAPDH, TUB2, CAL, ACT genes and the ITS-rDNA region. The sequences obtained were used for the construction of phylogenetic trees of Bayesian inference. The mycelial growth rate, size and shape of the conidia and appressories were evaluated for the morpho-cultural characterization of the species. Six isolates were analysis and three species belonging to the C. gloeosporioides complex were identified in this study. Samples of anthracnose were collected from six main blueberry cultivation areas in Sichuan Province. In total, 74 Colletotrichum isolates were obtained through a single-spore purification method and identified to the species through morphological characteristics and phylogenetic analyses based on partial DNA sequences of glyceraldehyde-3-phosphate dehydrogenase (GAPDH), internal transcribed spacer (ITS) regions, and the β-tubulin (TUB2), actin (ACT) and calmodulin (CAL) genes. Among all species, Colletotrichum fructicola was the most dominant species, with an isolation percentage of up to 66.2% in Sichuan, followed by Colletotrichum siamense (17.6%), C. kahawae (5.4%), C. karstii (5.4%), C. nymphaeae (2.7%) and C. sichuaninese (2.7%). Pathogenicity tests showed all species were able to cause typical anthracnose symptoms on blueberry leaves and stems. Colletotrichum fructicola was the predominant species with strong aggressiveness. Moreover, C. fructicola, C. kahawae, C. sichuaninese and C. nymphaeae are first reported here to cause blueberry anthracnose. This study provides a comprehensive reference for the association of different Colletotrichum spp., which may support the sustainable management of blueberry anthracnose.

Small cardamom ( Elettaria cardamomum Maton) is extensively cultivated in the Western Ghats of South India either as a monocrop under the forest trees or as an intercrop along with arecanut and coffee plantations. Colletotrichum species responsible for severe outbreaks of anthracnose on small cardamom in South India are reported. Small cardamom anthracnose, popularly known as 'Chenthal', manifests itself on the foliage as yellowish lesions, which later coalesce to form large blighted areas. In advanced stages, the affected leaves dry up giving a burnt appearance to the plant. Twenty-five isolates of Colletotrichum were isolated from leaves of small cardamom in Karnataka, Kerala and Tamil Nadu states of India. The isolates were characterized through morphological studies and multilocus phylogenetic analysis ( ITS, ACT, CHS-1, GAPDH, TUB2, CYLH3, GS and ApMat gene regions) to test whether different species are present and identified: C. karstii (2 isolates), C. gloeosporioides (1) , C. siamense (7), C. syzygicola (6), Colletotrichum sp (5), and C. guajavae (4), as the cause of anthracnose on small cardamom for the first time. Although Colletotrichum acutatum was recently investigated and shown to be a species complex comprising about 30 species, the name is still used in its broad sense for anthracnose pathogens of fruits in Brazil. In this study, a multilocus molecular analysis was carried out based on a dataset of ITS, HIS3, GAPDH, CHS-1, TUB2 and ACT sequences of Colletotrichum strains belonging to the C. acutatum species complex from fruits collected in different regions in Brazil combined with sequences of ex-type and other reference strains of species belonging to this complex. The strains were revealed to belong to Colletotrichum nymphaeae , Colletotrichum melonis , Colletotrichum abscissum and one new species, namely Colletotrichum paranaense, from apple and peach. Morphological descriptions of the new species and a strain closely related to but diverging from C. melonis are provided. From the data presently available, the most common species on apple fruits in Brazil is C. nymphaeae . In a pathogenicity test, strains of all four species caused lesions on detached apple, peach and guava fruits, except for strain CBS 134730 that did not infect guava fruits. [ABSTRACT FROM AUTHOR]

Colletotrichum species responsible for severe outbreaks of anthracnose on onion in southwest India are reported. These fungi produced white, oval sunken lesions on the leaves and neck at the onset of infection followed by appearance of necrotic centres with grey-colored concentric rings of acervuli, which eventually covered the whole leaves and subsequently led to collapse of the plant. Thirty seven isolates of Colletotrichum recovered from diseased leaves and neck were characterized based on morphological characteristics and partial sequences of ACT, TUB2, CHS-1, CAL, CYLH3, GAPDH, GS and ITS genes. The combined multilocus sequence analysis coupled with a critical examination of the phenotypic characters revealed the presence of two species, Colletotrichum siamense (27 isolates) and Colletotrichum truncatum (10 isolates). Pathogenicity of both species was ascertained on onion seedlings of cv. Arka kalyan. To our knowledge, this is the first report of C. siamense and C. truncatum causing anthracnose disease on onion. Colletotrichum species are widely known as key anthracnose pathogens of several economic plants. In this study, Colletotrichum species associated with leaf anthracnose isolated from various plants in Thailand were subjected to morphological and molecular analyses. The ITS rDNA regions of these strains were sequenced and aligned with those of type strains in the genus in order to establish if they can be assigned to any known species. Strains that could not be identified were further sequenced for partial actin (ACT), β-tubulin (TUB2) and glyceraldehydes-3-phosphate dehydrogenase (GAPDH) genes and employed in a phylogenetic analysis to reveal their relationships with other closely related taxa. The multilocus sequence analysis, together with a critical examination of the phenotypic characters, revealed three new species. Thirty-eight strains of Colletotrichum were isolated from diseased Citrus leaves in Guizhou and Yunnan provinces in China. Based on morphological characters and multi-locus sequence data, the strains were identified as C. boninense (1 strain), C. brevispora (1), C. fructicola (2), C. gloeosporioides (29), C. karstii (2), C. simmondsii (1) and Colletotrichum murrayae; the latter represents a new species which is described in this paper. Colletotrichum gloeosporioides was originally isolated from Citrus sinensis in southern Italy and was the most common species associated with diseased leaves of Citrus in China. Twenty-two strains of Colletotrichum, representing eight species, were isolated from eight genera of Orchidaceae in Yunnan and Guizhou provinces in China. Fourteen strains were from lesions and are pathogens, four were from fallen/dead flowers or stems and four were isolated as endophytes. The strains are characterized through morphological studies and multilocus phylogenetic analysis (ACT, Tub 2, CAL, CHS I, GPDH and ITS). Colletotrichum orchidearum and the new species Colletotrichum karstii are described and illustrated, based on morphological characters and multilocus sequence data. Colletotrichum species associated with leaf and flower anthracnose of jasmine ( Jasminum sambac) in the Ho Chi Minh region of Vietnam are reported. The disease of jasmine plantations was considered serious as it likely reduced flower yield. Leaves were colonized by Colletotrichum species which formed chlorotic regions with light brown necrotic centres, which eventually covered the whole leaf and subsequently caused defoliation and dieback and whole flowers were blighted. Nine strains of Colletotrichum species were isolated from diseased leaves and flowers and partial ITS rDNA sequences were analysed and morphologies compared across similar species. Based on ITS sequence analysis and morphological characters, three strains were identified as C. truncatum, while one strain was identified as C. siamense. The remaining five strains did not cluster with any known species for which type sequences are available and therefore partial actin (ACT), β-tubulin (TUB2), calmodulin (CAL), glutamine synthetase (GS), glyceraldehyde-3-phosphate dehydrogenase (GPDH) genes of the isolates were sequenced. Based on the reconstructed multiloci molecular phylogeny, two taxa are formally introduced as new species. Another strain was not well resolved in the phylogenetic tree and herein described as Colletotrichum sp. Further studies are needed to prove its distinctiveness. Licania tomentosa (Chrysobalanaceae), also known as "oiti," is a forest tree mainly used for urban afforestation in Brazil. Although anthracnose caused by Colletotrichum gloeosporioides is the main disease that threatens this tree, the identification of the species was based on morphological characteristics only. Owing to the need to use the molecular approach to pinpoint the identity of this pathogen with precision, the aim of this study was to identify endophytic and pathogenic Colletotrichum isolates from L. tomentosa based on both morphological and molecular data. For prior identification, partial sequences of the GAPDH region were obtained of all the 35 isolates (10 endophytic and 25 pathogenic). After analysis, ten isolates, representative of each clade, were selected for multilocus phylogenetic analysis (ACT, CAL, CHS‐1, GAPDH, TUB2, SOD2 and ITS). In addition, a tree based on the ApMat region was obtained for comparison with the multilocus tree. Morphological characterization (colony growth, conidial size and appressoria shape) was also performed for each species. To prove pathogenicity, L. tomentosa leaves were inoculated on the adaxial surface by mycelial plugs and conidial suspension. All isolates obtained belong to the Colletotrichum gloeosporioides complex. The Apmat tree has the same topology as the multilocus tree, allowing for the discrimination of the different species of Colletotrichum on L. tomentosa. Endophytic isolates of C. fructicola, C. queenslandicum, and C. siamense were acquired whereas pathogenic isolates were identified as C. siamense and C. tropicale, although all species were pathogenic on the wounded leaves of L. tomentosa.
