The aim of this study was to investigate the effect of an organophosphorus insecticide omethoate (OM), on certain oxidative stress biomarkers (malondialdehyde (MDA), reduced glutathione (GSH), glutathione-S-transferase (GST), glutathione reductase (GR), catalase (CAT)) in tongue, lung, stomach and muscle tissues of adult frogs (Rana ridibunda Pallas). Animals were exposed to 10 and 20 parts per million dosages of OM for 24, 48, 72 or 96 h. According to the results, MDA level increased significantly in lung and stomach tissues. GSH content fluctuated in lung and muscle while it elevated in tongue and stomach tissues. With regard to antioxidant enzymes (GST, GR and CAT), their activities reduced in tongue, while they increased in lung and fluctuated in stomach and muscle tissues. It can be concluded that exposure of frogs to OM are characterized by increased MDA levels and fluctuated enzyme activities and GSH contents. This may reflect the potential role of these parameters as useful biomarkers for assessment of OM toxicity.

The study was aimed at demonstrating changes in the antioxidant defense systems [Reduced glutathione (GSH), glutathione reductase (GR), superoxide dismutase (SOD), glutathione-S-transferase (GST) and catalase (CAT)] forming an antioxidative barrier and oxidative stress parameter (Malondialdehyde=MDA) in the various tissues of Sprague-Dawley rats which were administrated plant growth regulators (PGRs) [Abcisic acid (ABA) and Gibberellic acid (GA(3))] during 25 days. Seventy five parts per million of ABA and GA(3) as drinking water were administered orally to rats (Sprague-Dawley albino) ad libitum for 25 days continuously. The PGRs treatments caused different effects on antioxidant defense systems and MDA content of experimented rats compared to controls. The lipid peroxidation end product MDA significantly increased in the spleen and lungs of rats treated with ABA and GA(3) without significantly change in the other tissues. The GSH levels were significantly increased in the lungs and stomach of rats treated with ABA without any change in the tissues of rats treated with GA(3). Antioxidant enzyme activities such as SOD significantly increased in the spleen of rats treated with ABA and GA(3). Meanwhile, SOD significantly increased in the kidney of rats treated with GA. CAT significantly decreased in the lungs treated with ABA but did not change significantly in all the rest of rat tissues treated with both the PGRs. On the other hand, the ancillary enzyme GR activity decreased in the spleen and increased in the kidney with GA(3) treatment. The drug metabolizing enzyme GST activity significantly decreased in the heart of rats treated with GA(3) but increased in the spleen and lungs of rats treated with both PGRs. The observations presented led us to conclude that administration of subacute ABA and GA(3) promotes lipid peroxidatin content and alters in the antioxidative systems in the rat's various tissues. These data, along with changes, suggest that the PGRs produced oxidative stress in rats during the period of a 25-day subchronic exposure.

In the present study, the influence of subchronic effects of two plant growth regulators (PGRs) [Abcisic acid (ABA) and Gibberellic acid (GA3)] on antioxidant defense systems [reduced glutathione (GSH), glutathione reductase (GR), superoxide dismutase (SOD), glutathione-S-transferase (GST) and catalase (CAT)] and lipid peroxidation level (malondialdehyde = MDA) in various tissues of the rat were investigated during treatment as a drinking water model. 75 ppm of ABA and GA3 in drinking water were continuously administered orally to rats (Sprague-Dawley albino) ad libitum for 50 days. The PGRs treatments caused different effects on the antioxidant defense systems and MDA content of dosed rats compared to controls. The lipid peroxidation end product MDA significantly increased in the lungs, heart and kidney of rats treated with GA3 without significant change in the spleen. ABA caused also a significant increase in MDA content in the spleen, lungs, heart and kidney. The GSH levels were significantly depleted in the spleen, lungs and stomach of rats treated with ABA without any change in the tissues of rats treated with GA3 except the kidney where it increased. Antioxidant enzyme activities such as SOD significantly increased in the lungs and stomach and decreased in the spleen and heart tissues of rats treated with GA3. Meanwhile, SOD significantly decreased in the spleen, heart and kidney and increased in the lungs of rats treated with ABA. While CAT activity significantly decreased in the lungs of rats treated with GA3, a significant increase occurred in the heart of rats treated with both PGRs. On the other hand, the ancillary enzyme GR activity in the tissues were either significantly depleted or not changed with PGRs treatment. The drug metabolizing enzyme GST activity significantly decreased in the lungs of rats treated with ABA but increased in the stomach of rats treated with both PGRs. As a conclusion, the rats resisted oxidative stress via the antioxidant mechanism. But the antioxidant mechanism could not prevent the increases in lipid peroxidation in rat's tissues. This data, along with changes, suggests that PGRs produced substantial systemic organ toxicity in the spleen, lungs, stomach, heart and kidney during a 50-day period of subchronic exposure.

This study aims to investigate the effects of the plant growth regulators (PGRs) (2,3,5-triiodobenzoic acid (TIBA), Naphthaleneacetic acid (NAA), and 2,4-dichlorofenoxyacetic acid (2,4-D)) on serum marker enzymes (aspartate aminotransferase (AST), alanin aminotransferase (ALT), creatine phosphokinase (CPK), and lactate dehydrogenase (LDH)), antioxidant defense systems (reduced glutathione (GSH), glutathione reductase (GR), superoxide dismutase (SOD), glutathione-S-transferase (GST), and catalase (CAT)), and lipid peroxidation content (malondialdehyde = MDA) in various tissues of rats. 50 and 100 ppm of PGRs as drinking water were administered orally to rats (Sprague-Dawley albino) ad libitum for 25 days continuously. The PGRs treatment caused different effects on the serum marker enzymes, antioxidant defense systems, and the MDA content in experimented rats compared to controls. Results showed that TIBA caused a significant decrease in serum AST activity with both the dosage whereas serum CPK was significantly increased with 100 ppm dosage of TIBA. Meanwhile, serum AST, CPK, and LDH activities were significantly increased with both dosage of NAA and 2,4-D. The lipid peroxidation end-product MDA significantly increased in the all tissues treated with both dosages of PGRs without any change in the brain and erythrocyte of rats treated with both the dosages of 2,4-D. The GSH depletion in the kidney and brain tissues of rats treated with both dosages of PGRs was found to be significant. Furthermore, the GSH depletion in the erythrocyte of rats treated with both dosages of PGRs except 50 ppm dosage of 2,4-D was significant too. Also, the GSH level in the liver was significantly depleted with 50 ppm of 2,4-D and NAA, whereas the GSH depletion in the same tissue did not significantly change with the treatment. The activity of antioxidant enzymes was also seriously affected by PGRs; SOD significantly decreased in the liver, heart, kidney, and brain of rats treated with both dosages of NAA, whereas the SOD activity in the erythrocytes, liver, and heart was either significantly decreased or not changed with two doses of 2,4-D and TIBA. Although the CAT activity significantly increased in the erythrocyte and brain of rats treated with both doses of PGRs, it was not changed in the liver, heart, and kidney. Meanwhile, the ancillary enzyme GR activity significantly increased in the brain, heart, and liver but decreased in the erythrocyte and kidney of rats treated with both doses of PGRs. The drug-metabolizing enzyme GST activity significantly increased in the heart and kidney but decreased in the brain and erythrocytes of rats treated with both dosages of PGRs. As a conclusion, the results indicate that PGRs might affect antioxidant potential enzymes, the activity of hepatic damage enzymes, and lipid peroxidation dose independently. Also, the rats resisted to oxidative stress via antioxidant mechanism but the antioxidant mechanism could not prevent the increases in lipid peroxidation in rat's tissues. Cisplatin (CP) is a widely used cytotoxic agent against cancer, and high doses of CP have been known to cause nephrotoxicity and hepatotoxicity. Some reports claim that antioxidants can reduce CP-induced toxicity. This study investigated the hepatoprotective, nephroprotective, and antioxidant activity of Urtica dioica L methanolic extract (UDME) against CP toxicity in Erhlich ascites tumor (EAT)-bearing mice. Levels of serum hepatic enzymes, renal function markers, and oxidant/antioxidant parameters of liver tissue were measured. Mice were inoculated with EAT on day 0 and treated with nothing else for 24 hours. After a single dose of CP administration on day 1, the extract was given at the doses of 50, 100, 200, and 400 mg/kg body weight daily during 6 days. Almost all doses of UDME performed a significant ( P < 0.05) preventive role against CP toxicity by decreasing aspartate aminotransferase, alanine aminotransferase, lactate dehydrogenase, blood urea nitrogen, creatinine, lipid peroxidation, protein oxidation levels, and myeloperoxidase activity, as well as increasing reduced glutathione content, superoxide dismutase, catalase, glutathione S-transferase, and glutathione peroxidase activities. This suggests that UDME has a protective capacity and antioxidant activity against CP toxicity in EAT-bearing mice, probably by promoting antioxidative defense systems. [ABSTRACT FROM AUTHOR]

The aim of this study was to determine the antiulcer and antioxidant activities of Centaurium erythraea L (small centaury) in aspirin (ASA) induced acute gastric ulcer model. The gastroprotective effect of the 50% aqueous-ethanolic small centaury (SC) extract was investigated in rats at a dose of ASA 200 mg/kg body weight. Twenty-one Sprague-Dawley albino rats were divided into three groups of seven rats each as follows: (1) control group; (2) acute ASA-treated group and (3) ASA plus SC group. At the end of the 4-h drug administration, ulcer index, oxidant and antioxidant levels were measured and compared between the groups. The percentage of lesion area to total gastric surface area (ulcer index) was significantly reduced (77%) in ASA plus SC group as compared with acute ASA-treated group. The oral administration of ASA decreased catalase (CAT), reduced glutathione (GSH), and increased lipid peroxidation (LPO) levels. Although myeloperoxidase (MPO) activity was increased by ASA, it was found to be lower in the ASA plus SC group. GSH and Vitamin A levels were determined higher in the ASA plus SC group compared with ASA group. Coal dust causes lung diseases in occupational exposure. Reactive oxygen species have been implicated in the pathogenesis of its toxicity. In this study, serum enzymes, lipid profile and other biochemical values with oxidant/antioxidant status in whole blood and serum of central heating system workers (CHSW; the persons responsible for heating the apartment with coal) were determined to reflect the cell injury. Blood samples were obtained from CHSW (n = 25) and healthy individuals (n = 25). All values were measured in whole blood and serum. ANOVA was used for the estimation of statistical data. In the group of CHSW, creatinine, ferritin, alanin aminotransferase, aspartate aminotransferase, creatine phosphokinase, gamma glutamyl transferase, lactate dehydrogenase and glutathione reductase activities as well as triglyceride, very low density lipoprotein, protein carbonyl and malondialdehide were significantly higher, while transferrin, high density lipoprotein and catalase (CAT) activities were lower than the group of healthy individuals. This result is consistent with hypothesis that respirable coal dust generates lipid and protein oxidation and induces leakage of serum enzymes by cell damage. The article presents the results of a study which investigated the effects of sublethal concentrations of plant growth regulators such as dichlorophenoxyacetic acid, triiodobenzoic acid and naphthaleneacetic acid on the antioxidant defense systems found in the spleen and lung tissues of rats, such as glutathione reductase, adenosine deaminase and myeloperoxidase. Results indicated that the plant growth regulators in question inhibit the antioxidative defense system and promote malondialdehyde content, causing significant organ toxicity.

Abstract: In the present study, the influence of two different PGRs, indoleacetic acid (IAA) and kinetin (Kn) on immune potential enzymes, adenosine deaminase (ADA) and myeloperoxidase (MPO), and antioxidant defense enzymes such as glutathione peroxidase (GPx), glutathione reductase (GR), and superoxide dismutase (SOD) in various tissues of rats were investigated during the treatment as a drinking water model. 100ppm of IAA and Kn as drinking water were administered orally to rats (Sprague–Dawley albino) ad libitum for 21 days continuously. The PGRs treatments caused different effects on the immune potential and antioxidant defense enzymes of experimented rats compared to controls. Results show that IAA caused a significant decrease in GR activity in the lungs and liver and an increase in the spleen. Also, IAA caused a significant decline in GPx activity in the lungs and an increase in the heart. SOD was significantly reduced in the heart, while increased in the lungs. Furthermore, IAA caused a significant decrease in ADA activity in the heart and blood whereas an increase in the kidney and spleen. MPO activity was also significantly increased in the heart by IAA treatment. The activity of enzymes were also seriously affected by Kn; GR activity decreased in the lungs, brain, and blood while GPx activity decreased in the spleen, brain, and heart. ADA activity was also significantly reduced in the blood whereas MPO activity rose in the spleen. In addition, SOD activity lowered in all tissues except for lungs where a significant increment was determined. As a conclusion, the results indicate that PGRs might affect on antioxidant and immune potential enzymes. These data, along with the determined changes suggest that PGRs produced substantial systemic organ toxicity in the erythrocyte, liver, brain, heart, lungs, spleen, and kidney during the period of a 21-day subacute exposure. Abstract: This study aims to investigate the effects of indoleacetic acid (IAA) and kinetin (Kn), which are plant growth regulators (PGRs), on antioxidant defense systems [reduced glutathione (GSH), glutathione-S-transferase (GST), catalase (CAT)], and lipid peroxidation level (malondialdehyde, MDA) various tissues of rats. Rats (Sprague–Dawley albino) were exposed to 100ppm IAA and Kn. One hundred parts per million of PGRs was administered orally to rats ad libitum for 21days continuously. The PGRs treatments caused different effects on the content of MDA and antioxidant defense system in comparison to those of control rats. According to the results, the subchronic treatments of IAA caused significant decrease in the GSH concentration and CAT activity in erythrocyte. Kn decreased GSH concentration in erythrocyte too. While the MDA concentration in brain was increased significantly by IAA and Kn, Kn decreased significantly brain CAT and GST activity. The liver GST activity was decreased by IAA and Kn. But, liver CAT activity was increased by IAA. On the other hand, while IAA treatment caused a significant decrease kidney GST activity, Kn caused a significant decrease both kidney GST and CAT activity. Also, while heart CAT activity was decreased by IAA, heart GST activity was decreased by both IAA and Kn. Moreover, MDA concentration in heart was increased by Kn treatment. It was concluded that IAA might effect MDA and antioxidant defense on the animals at subchronic treatment. [Copyright &y& Elsevier]
