International audience; Cultivated pearls frequently exhibit morphological irregularities making obvious that mineral deposition was irregularly distributed onto nucleus surface. Taking advantage of experimental cultivations with short durations (from 10 days to few months), these irregular deposits predating occurrence of the nacre were investigated in Polynesian pearls by biochemical characterizations and a series of physical methods. Diversity in the resulting data suggests that various in‐depth alterations of the biomineralization mechanism may have occurred during the grafting process, leading to diversity in the biochemical pathways to nacreous deposition. This allows a precise discussion of current views about pearl formation. The “reversed shell theory” is formally disproved through point to point comparison with development of the shell growing edge. Similarity of pearl formation with “regeneration” or “shell repair” is also discussed, emphasizing the differences between these concepts.

Advancement and diversification of the aquaculture industry is reliant on the development of captive breeding and rearing protocols for novel fish species. Orbicular batfish (Platax orbicularis), a major emerging species in Polynesian aquaculture, live in brackish and marine waters around coral reefs, and are highly prized by Pacific Island communities for their high-quality meat. The present study describes the larval growth of P. orbicularis from hatching until 16 days after hatching (DAH) using meristics and thyroid hormone (T3) quantification. Our study highlighted that metamorphosis of P. orbicularis is critical to their production in aquaculture, as found for other species. Levels of the thyroid hormone T3 in P. orbicularis reached a peak at 12 DAH (i.e. hormonal metamorphosis). This peak occurred in concert with important morphological changes and increased mortality and growth between 9 and 12 DAH, clearly illustrating the sensitivity of this fish during metamorphosis. Overall, our study sheds light on the metamorphosis and larval development of a novel aquaculture species, and the interplay between hormonal and morphological changes throughout a critical period of its rearing. These findings may promote the development of protocols for mass-scale production of orbicular batfish in captivity, which may be particularly beneficial to the aquaculture industries of many Pacific Island countries.

International audience; Trophic conditions and water temperature strongly influence bivalve physiological processes and metabolism. In black-lip pearl oyster Pinctada margaritifera, these parameters have been shown to affect shell biomineralization. The present study investigated the effect of preoperative food level (i.e., microalgal concentration) and temperature on pearl biomineralization. Donor and recipient oysters were conditioned at different levels of food and temperature during the preoperative phase to evaluate the influence of these factors on 1) pearl retention rate (grafting success), 2) expression of genes involved in biomineralization in the mantle and pearl sac and 3) pearl quality traits. Our study confirmed the influence of both microalgal concentration and temperature on shell growth. Food level of donor oysters was decisive for pearl biomineralization, with donors that had been fed at a high microalgal concentration producing pearl sacs with significantly higher biomineralization capabilities and faster nacre establishment during early stages of pearl formation. However, food level showed no effects on quality traits of the pearls harvested 12 months postgrafting, while preoperative temperature only influenced the relative expression of two genes in pearl sacs at 12 months postgrafting. No significant effects of the preoperative conditioning of recipient oysters were detected in either experiment considering gene expression measurements and pearl quality traits. However, mortality was significantly lower in grafted recipient oysters fed at an intermediate trophic level. Finally, pearl weight was shown to be positively correlated with recipient oyster growth.

The purpose of our study is to investigate (1) the influence of phenotype plasticity and local adaptation on pearl-oyster physiology by testing the persistence of growth differentiation of two pearl oyster populations (Arutua and Mangareva) in common garden experiment; (2) to quantify the influence of biofouling development on the growth of each pearl oyster population. According to our observations, the growth rate in terms of total oyster weight suggested better growth performance of the pearl oyster Pinctada margaritifera in Mangareva (0.21–0.24 ± 0.01 g.day−1) than Arutua (0.14–0.15 ± 0.01 g.day−1). However, similar growth performances are observed at the Vairao common garden site for oyster stocks from Mangareva (0.07 ± 0.01 mm.day−1 or 0.15 ± 0.01 g.day−1) and Arutua (0.07 ± 0.01 mm.day−1 or 0.13 ± 0.01 g.day−1). Our results suggest that growth performance variability observed at the pearl farming sites of Arutua and Mangareva is due more to phenotypic plasticity than to local adaptation. This result thus accords a major importance to site selection for the pearl farming optimization process. Biofouling dynamics on Pinctada margaritifera shells differed radically between Arutua and Mangareva sites. In Arutua, biofouling colonization was relatively slow (0.016 g.oyster−1.month−1) and was mainly composed of sponges and bivalves. On Mangareva, the colonization process appeared faster and more continuous over the study period (0.15–0.18 g.oyster−1.month−1) and the biofouling community was dominated by tunicates. On the basis of our results obtained on growth performance between cleaned and uncleaned stock in Arutua and Mangareva, biofouling development after 14–15 months of colonization does not appear to have any negative effect on P. margaritifera growth. Due to the high cost of biofouling management in pearl farming facilities, our results suggest once more that reconsideration of the pearl farming management process is needed.

International audience; The possible consequences of acidification on pearl farming are disruption of oyster metabolism and change in growth. In the laboratory, we studied the impact of pCO2 (3540, 1338 and 541μatm) on the physiology of pearl oysters exposed for 100 days. This experiment was repeated after an interval of one year. Several physiological compartments were examined in pearl oysters: the scope for growth by measuring ingestion, assimilation and oxygen consumption, gametogenesis by means of histological observations, shell growth by measurement and observation by optical and electronic microscopy, and at molecular level by measuring the expression of nine genes of mantle cells implied in the biomineralisation process. Results from both experiments showed that high pCO2 had no effect on scope for growth and gametogenesis. High pCO2 (3540 μatm) significantly slowed down the shell deposit rate at the ventral side and SEM observations of the inside of the shell found signs of chemical dissolution. Of the nine examined genes high pCO2significantly decreased the expression level of one gene (Pmarg-PUSP 6). This study showed that shell growth of the pearl oyster would be slowed down without threatening the species since the management of energy and reproduction functions appeared to be preserved. Further investigations should be conducted on the response of offspring to acidification.

Marine natural products isolated from the sponge Fascaplysinopsis cf reticulata, in French Polynesia, were investigated as an alternative to antibiotics to control pathogens in aquaculture. The overuse of antibiotics in aquaculture is largely considered to be an environmental pollution, because it supports the transfer of antibiotic resistance genes within the aquatic environment. One environmentally friendly alternative to antibiotics is the use of quorum sensing inhibitors (QSIs). Quorum sensing (QS) is a regulatory mechanism in bacteria which control virulence factors through the secretion of autoinducers (AIs), such as acyl-homoserine lactone (AHL) in gram-negative bacteria. Vibrio harveyi QS is controlled through three parallel pathways: HAI-1, AI-2, and CAI-1. Bioassay-guided purification of F. cf reticulata extract was conducted on two bacterial species, i.e., Tenacibaculum maritimum and V. harveyi for antibiotic and QS inhibition bioactivities. Toxicity bioassay of fractions was also evaluated on the freshwater fish Poecilia reticulata and the marine fish Acanthurus triostegus. Cyclohexanic and dichloromethane fractions of F. cf reticulata exhibited QS inhibition on V. harveyi and antibiotic bioactivities on V. harveyi and T. maritimum, respectively. Palauolide (1) and fascaplysin (2) were purified as major molecules from the cyclohexanic and dichloromethane fractions, respectively. Palauolide inhibited QS of V. harveyi through HAI-1 QS pathway at 50 μg ml–1 (26 μM), while fascaplysin affected the bacterial growth of V. harveyi (50 μg ml–1) and T. maritimum (0.25 μg). The toxicity of fascaplysin-enriched fraction (FEF) was evaluated and exhibited a toxic effect against fish at 50 μg ml–1. This study demonstrated for the first time the QSI potential of palauolide (1). Future research may assess the toxicity of both the cyclohexanic fraction of the sponge and palauolide (1) on fish, to confirm their potential as alternative to antibiotics in fish farming.

International audience; The pearl culture is one of the most lucrative aquacultures worldwide. In many South Pacific areas, it depends on the exploitation of the pearl oyster Pinctada margaritifera and relies entirely on the environmental conditions encountered in the lagoon. In this context, assessing the impact of climatic stressors, such as global warming and ocean acidification, on the functionality of the resource in terms of renewal and exploitation is fundamental. In this study, we experimentally addressed the impact of temperature (22, 26, 30 and 34 °C) and partial pressure of carbon dioxide pCO2 (294, 763 and 2485 μatm) on the biomineralization and metabolic capabilities of pearl oysters. While the energy metabolism was strongly dependent on temperature, results showed its independence from pCO2 levels; no interaction between temperature and pCO2 was revealed. The energy metabolism, ingestion, oxygen consumption and, hence, the scope for growth (SFG) were maximised at 30 °C and dramatically fell at 34 °C. Biomineralization was examined through the expression measurement of nine mantle's genes coding for shell matrix proteins involved in the formation of calcitic prisms and/or nacreous shell structures; significant changes were recorded for four of the nine (Pmarg-Nacrein A1, Pmarg-MRNP34, Pmarg-Prismalin 14 and Pmarg-Aspein). These changes showed that the maximum and minimum expression of these genes was at 26 and 34 °C, respectively. Surprisingly, the modelled thermal optimum for biomineralization (ranging between 21.5 and 26.5 °C) and somatic growth and reproduction (28.7 °C) appeared to be significantly different. Finally, the responses to high temperatures were contextualised with the Intergovernmental Panel on Climate Change (IPCC) projections, which highlighted that pearl oyster stocks and cultures would be severely threatened in the next decade

Pearl oyster grafting is a complex surgical operation that should lead to pearl formation after approximately eighteen months. Although this technique has been used for many years in French Polynesia, the grafting process is still not standardised. While studies have been carried out in order to improve graft performance and yield, these remain highly variable due to post-grafting mortality, nucleus rejection and unreliable pearl quality, all of which constrain pearl farm profitability. The present study uses histological analysis to monitor oysters that either rejected or retained their nuclei. Both groups of oysters are compared in terms of evolution of the graft, which could influence retention, and the development of a pearl sac in cases where grafting was successful. Data show that rejection phenomena are linked to a number of causes, notably an inflammatory reaction in the "receiving" oyster, the presence of numerous tissue lesions and the quality of the grafted tissue. These results suggest that study is needed on the different concomitant elements of the grafting process: the graft "donor" oysters, the nucleus and the "receiving" oyster and their interactions.

International audience; The present study investigated the effect of electrolysis on the biomineralization capacities of juveniles of the mollusk Pinctada argaritifera for the first time. Size-selected individuals from two groups, “Medium” and “Large”, from a multi-parental family produced in a hatchery system were subjected to electrolysis under a low voltage current over a nine-week experimental period. The growth of the juveniles was individually monitored and assessed weekly by wet weight and shell height measurements. At the end of the experiment, mantle tissue was sampled for biomineralization-related gene expression analysis. Electrolysis significantly increased pearl oyster growth in terms of shell height and wet weight for Large juveniles from the 5th and the 2nd week, respectively, until the end of the experiment. However, differences were only significant for Medium individuals from the 7th week for shell height and from the 9th week for wet weight. Furthermore, transcriptional analysis of six known biomineralization genes coding for shell matrix proteins of calcitic prisms and/or nacreous shell structures revealed that five were significantly overexpressed in the mantle mineralizing tissue under electrolysis: three in common between the two size class groups and two that were expressed exclusively in one or the other group. Finally, we found no statistical difference of the shell thickness ratio between individuals undergoing electrolysis and control conditions. Taken together, our results indicate, for the first time in a calcifying marine organism, that electrolysis influences molecular mechanisms involved in biomineralization and may stimulate some parameters of pearl oyster growth rate.

The emerging orbicular batfish (Platax orbicularis) aquaculture is the most important fish aquaculture industry in French Polynesia. However, bacterial infections are causing severe mortality episodes. Therefore, there is an urgent need to find an effective management solution. Besides the supplying difficulty and high costs of veterinary drugs in French Polynesia, batfish aquaculture takes place close to the coral reef, where use of synthetic persistent drugs should be restricted. Medicinal plants and bioactive algae are emerging as a cheaper and more sustainable alternative to chemical drugs. We have studied the effect of local Polynesian plants and the local opportunistic algae Asparagopsis taxiformis on batfish when orally administered. Weight gain and expression of two immune-related genes (lysozyme g e Lys G and transforming growth factor beta - TGF-b1) were studied to analyze immunostimulant activity of plants on P. orbicularis. Results showed that several plants increased Lys G and TGF-b1 expression on orbicular batfish after 2 and 3 weeks of oral administration. A. taxiformis was the plant displaying the most promising results, promoting a weight gain of 24% after 3 weeks of oral administration and significantly increasing the relative amount of both Lys G and TGF-b1 transcripts in kidney and spleen of P. orbicularis.

The pathogenicity of Vibrio penaeicida Strains KH-1 and AM101, their culture-free supernatant (CFS), and their protein fraction obtained by 40% of ammonium sulfate precipitation (PFs40) were assessed in experimental challenges against juvenile Litopenaeus vannamei. Live Vibrio cells, CFS, and PFs40 from the AM101 strain produced a significantly higher mortality (p < 0.05) compared to the KH-1 strain. Toxicity and median lethal doses (LD50) of Fast Protein Liquid Chromatography (FPLC) products were evaluated on L. vannamei. The first FPLC fraction sample (A) from PFs40 of the AM101 strain displayed LD50 values of 1.68 and 5.61 mu g protein ind.(-1), respectively. The second FPLC process from Fraction A showed a peak (A1) also with toxic effects to shrimp. PFs40, Fraction A, and Peak At showed a 38.5 kDa molecular band (SDS-PAGE), with activity on a gelatin protease zymogram. The lethal effect of PFs40 and Fraction A was inhibited by Proteinase K, CuCl2, E-64, and heat (60 and 100 degrees C) treatments, but was not inhibited by EDTA-Na-2, aprotinin, and soy trypsin treatments. These results and the zymogram inhibition test suggest the presence of a cysteine protease-like proteinaceous exotoxin as a dominant protease, secreted by V. penaeicida Strain AM101.

Aquaculture production in French Polynesia and New Caledonia relies upon domesticated broodstock of two introduced species (the shrimp Litopenaeus stylirostris , and the fish Lates calcarifer ) and on natural stock of the local pearl oyster Pinctada margaritifera . For L. stylirostris , refrigeration of shrimp sperm lowers the sanitary risk linked to the re-introduction of variability in Tahiti that is proved necessary by microsatellite markers. They also allow paternity controls that will improve future genetic management. Selective breeding programs for shrimp are underway in Tahiti and under assessment at pilot scale in New Caledonia. Conservation strategies are under elaboration for local populations of P. margaritifera that have been proven genetically differentiated. The recent development of pearl oyster hatcheries is the first step towards genetic improvement of this species. Thanks to geographical isolation and sanitary regulations, French Polynesia and New Caledonia are not affected by the viral outbreaks affecting shrimp elsewhere in the world (e.g., white spot and Taura syndrome viruses) and the pearl oyster in Japan (Akoya virus). Furthermore, a zoosanitary surveillance network of pearl oyster stocks is coming up in order to detect the emergence of any pathogen. Recently, alternative methods for antibiotherapy in shrimp larval rearing have been tested using probiotic bacteria, and could find an application in pearl oyster larval rearing.

The aim of this study was to assess the relative contribution of natural productivity and compound food to the growth of the juvenile blue shrimp Litopenaeus stylirostris reared in a biofloc system. Two experiments were carried out based on the same protocol with three treatments: clear water with experimental diet (CW), biofloc with experimental diet (BF) and biofloc unfed (BU). Shrimp survival was significantly higher in biofloc rearing than in CW rearing. The contribution of the biofloc to shrimp diet was estimated through measurement of carbon and nitrogen stable isotope ratios in shrimp and food sources. Different isotopic compositions between feeds were obtained by feeding natural productivity with a mixture rich in fish meal and the shrimps with a pellet containing a high level of soy protein concentrate. Using a two source one-isotope mixing model, we found that the natural productivity of the biofloc system contributed to shrimp growth at a level of 39.8% and 36.9%, for C and N, respectively. The natural food consumed by the shrimps reared in the biofloc system resulted in higher gene expression (mRNA transcript abundance) and activities of two digestive enzymes in their digestive gland: α-amylase and trypsin. The growth of shrimp biomass reared in biofloc was, on average, 4.4 times that of those grown in clear water. Our results confirmed the best survival and promoted growth of shrimps using biofloc technology and highlighted the key role of the biofloc in the nutrition of rearing shrimps. Statement of relevance In this study, we have applied an original protocol to determine the respective contribution of natural productivity and artificial feeds on the alimentation of the juvenile blue shrimp L. stylirostris reared in biofloc system by using C and N natural stable isotope analysis. Moreover, we have compared, in shrimp digestive gland, the α-amylase and trypsin enzyme activities at biochemical and molecular levels for two different shrimp rearing systems, biofloc and clear water. In our knowledge, the use of molecular tool to study the influence of biofloc consumption on digest process of shrimp was never carried out. We think that our research is new and important to increase knowledge on biofloc topic.

International audience; This study compares the antioxidant and antimicrobial transcriptional expression of blue shrimps reared according to two different systems, BioFloc Technology (BFT) and Clear sea Water (CW) and their differential responses when facing an experimental sublethal hydrogen peroxide stress. After 30 days of rearing, juvenile shrimps were exposed to H2O2 stress at a concentration of 30 ppm during 6 hours. The oxidative stress caused by H2O2 was examined in the digestive glands of the shrimp, in which antioxidant enzyme (AOE) and antimicrobial peptide (AMP) gene expression were analysed by quantitative real-time PCR. Results showed that rearing conditions did not affect the expression of genes encoding AOEs or AMPs. However, H2O2 stress induced a differential response in expression between shrimps from the two rearing treatments (BFT and CW). Comparative analysis of the expression profiles indicates that catalase transcripts were significantly upregulated by H2O2 stress for BFT shrimps while no change was observed for CW shrimps. In contrast, H2O2 caused down-regulation of superoxide dismutase and glutathione transferase transcripts and of the three AMP transcripts studied (penaeidin 2 and 3, and crustin) for CW shrimps, while no effect was observed on BFT shrimp transcript levels. These results suggested that BFT shrimps maintained antioxidant and AMP responses after stress and therefore can effectively protect their cells against oxidative stress, while CW shrimp immune competence seems to decrease after stress

Vibrio species have become a major source of concern for shrimp culture because of their close association with low survival rates in hatcheries or growout ponds. New shrimp pathogens belonging to the Vibrio genus have been described although their virulence is not yet fully understood. Indeed, they may act as opportunistic agents in secondary infections or be true pathogens. This review presents the usefulness of infection models with vibriosis pathogens for pathogenicity experiments, testing of curative or prophylactic treatments and the study of host-factors influencing bacterial virulence. Furthermore, some guidelines for experimental trials are given to evaluate the in vivo virulence of Vibrio isolates.

International audience; The Gram-negative bacterium Vibrio harveyi is known to be highly pathogenic for the European abalone Haliotis tuberculata, which is a gastronomically important marine gastropod with a high commercial value. Since 1998, some particular bacterial strains are described as implicated in recurrent mortality outbreaks in French farm and field stocks of abalone. Recently, a 9.6 kb plasmid named pVCR1, was shown to be harbored by one highly V. harveyi virulent ORM4 strain suggesting its involvement in virulence phenotype. Thus, we have developed in the present study two TaqMan real-time PCR assays allowing to (i) rapidly and specifically detect, by a duplex procedure and in less than 2 h, both V. harveyi and the presence of plasmid pVCR1 from unidentified bacterial colony and to (ii) quantify both V. harveyi and the plasmid pVCR1 in the hemolymph of abalone or its surrounding seawater. Quantification curves of V. harveyi or ORM4 strain seeded in hemolymph or artificial sea water samples were equivalent showing excellent qPCR efficacies and detection level as low as 18 V. harveyi cell-equivalent genomic DNA in a PCR reaction well. This qPCR allowed us to monitor V. harveyi ORM4 strain in experimentally infected H. tuberculata. These diagnosis assays could provide powerful and useful tools to better understand the epidemiology of vibriosis caused by V. harveyi in different cultured marine species including H. tuberculata.

Phenoloxidases (POs) are a group of copper proteins including tyrosinase, catecholase and laccase. In several insects and crustaceans, antibacterial substances are produced through the PO cascade, participating in the direct killing of invading microorganisms. However, although POs are widely recognised as an integral part of the invertebrate immune defence system, experimental evidence is lacking that these properties are conserved in molluscs, and more particularly in the Pacific oyster Crassostrea gigas. In the present study, Vibrio splendidus LGP32 and Vibrio aestuarianus 02/041 growths were affected, after being treated with C. gigas haemocyte lysate supernatant (HLS), and either a common substrate of POs, l-3,4-dihydroxyphenylalanine (l-DOPA), to detect catecholase-type PO activity, or a specific substrate of laccase, p-phenylenediamine (PPD), to detect laccase-type PO activity. Interestingly, a higher bacterial growth inhibition was observed in the presence of PPD than in the presence of l-DOPA. These effects were suppressed when the specific PO inhibitor, phenylthiourea (PTU), was added to the medium. Results of the present study suggest, for the first time in a mollusc species, that antibacterial activities of HLS from C. gigas potentially involve POs, and more particularly laccase catalysed reactions. Highlights ► Bacterial growth inhibition was studied in two Vibrio oyster bacterial pathogens. ► Different Crassostrea gigas tissues, haemolymph fractions, and PO substrates, were tested. ► The inhibition occurred only in the presence of C. gigas HLS and PO substrates. ► Bacterial growth inhibition was suppressed with a PO inhibitor. ► Results suggest that HLS antibacterial activities involve laccase-type PO-catalysed reactions.

The understanding of reciprocal interactions between Crassostrea gigas and Vibrio sp., whether these be virulent or avirulent, is vital for the development of methods to improve the health status of cultured oysters. We describe an original non-invasive experimental infection technique using cohabitation, designed to explore these interactions. Using real-time PCR techniques we examined the dynamics of virulent and avirulent Vibrio sp. in oyster hemolymph and tank seawater, and made a parallel study of the expression of four genes involved in oyster immune defense: Cg -BPI, Cg -EcSOD, Cg -IκB, Cg -TIMP. No mortality occurred in control animals, but oysters put in cohabitation for 2–48 h with animals previously infected by two Vibrio pathogens suffered mortalities from 2 to 16 days post-cohabitation. Our results show that virulent Vibrio infect healthy individuals after only 2 h of cohabitation, with values ranging from 4.5×10 2 to 2×10 4 cells ml −1 hemolymph. Simultaneously, an approximate ten-fold increase of the total Vibrio population was observed in control animals, with a 6.6–78.5-fold up-expression of targeted genes. In contrast, oysters exposed to harmful bacteria had mean expression levels strongly down-regulated by a factor of 9.2–29 (depending on the gene) compared with control animals. Although oysters were still found to be infected by virulent Vibrio after 6–48 h of cohabitation, no significant differences were noted when comparing levels of each transcript in control and infected oysters at the same sampling times during this period: the important differences were noted before 6 h cohabitation. Taken together, our data support (1) the hypothesis that virulent Vibrio disturbs the immune response of this invertebrate host both rapidly and significantly, although this occurs specifically during an early and transient period during the first 6 h of cohabitation challenge, and that (2) expression of targeted genes is not correlated with vibriosis resistance.

To assess the impact of pollution induced by pesticides on Pacific oyster, Crassostrea gigas, health in France, in vivo effects of combined pesticide exposure and bacterial challenge on cell activities and gene expression in hemocytes were tested using flow cytometry and real-time PCR. As a first step, an in vivo model of experimental contamination was developed. Pacific oysters were exposed to a mixture of eight pesticides (atrazine, glyphosate, alachlor, metolachlor, fosetyl-alumimium, terbuthylazine, diuron and carbaryl) at environmentally relevant concentrations over a 7-day period. Hemocyte parameters (cell mortality, enzyme activities and phagocytosis) were monitored using flow cytometry and gene expression was evaluated by real-time PCR (RT-PCR). The expression of 19 genes involved in C gigas hemocyte functions was characterized using RT-PCR. After 7 days of exposure, phagocytosis was significantly reduced and the 19 selected genes were down-regulated in treated animals. As a second step, the experimental contamination method previously developed was used to study interactions between pesticide exposure and bacterial challenge by intramuscular injection of two Vibrio splendidus-related pathogenic strains. Oyster mortality and expression of 10 of the 19 selected genes were followed 4 and 24 h post-injection. Oyster mortality was higher in pesticide-treated oysters compared to untreated oysters after the bacterial challenge. Gene expression was up-regulated in pesticide-treated oysters compared to untreated oysters after the bacterial challenge. We hypothesize that gene over-expression due to an interaction between pesticides and bacteria could lead to an injury of host tissues, resulting in higher mortality rates. In conclusion, this study is the first to show effects of pesticides at environmentally relevant concentrations on C. gigas hemocytes and to hypothesize that pesticides modulate the immune response to a bacterial challenge in oysters. The pathogenicity of two V. penaeicida strains, AM101 and KH-1, with different geographic origin, and V. nigripulchritudo strain AM102, were investigated in juvenile blue shrimp species Litopenaeus stylirostris. Alive bacteria and protein fractions (PFs) obtained from cell-free supernatants (CFS) were used in experimental challenges. Strains AM102, AM101, and KH-1 produced respectively 60, 54 and 12% mortality at 96 h after infection using 104 cfu mL–1 of bacterial suspension. Exocellular toxin-like factors were evidenced in CFS from the New Caledonian strains (AM102 and AM101) but not in the Japanese strain (KH-1). At 48 h post injection of each CFS, mortality rates were respectively 96, 98 and 5% when these strains were cultivated at 20 ºC, whereas only 0, 16 and 5% mortality rates were observed when these strains were cultivated at 30ºC. Clear differences in pathogenicity between both V. penaeicida strains of distinct geographic origin (AM101 and KH-1) were thus emphasized. Protein fractions were obtained from CFS of all the strains tested in this study and cultivated at 20ºC, by ammonium sulfate precipitation. Whatever the used strain, significantly higher mortalities were produced with PFs obtained with 0–40% of ammonium sulfate saturation, in respect to those produced with PFs60 and PFs80. Shrimp injected with PFs40 from strains AM102, AM101 and KH-1, and at a 20-µg org–1 dose produced respectively 100, 90 and 60% mortality 46 h after the challenge. The strain AM101 showed a median lethal dose of approximately 5 µg protein org–1 (1–1.25 µg protein g–1 body weight) 59 h after injection. The PFs40 from New Caledonian strains were found sensitive to heating and proteinase K treatments, reinforcing thus the hypothesis of their proteinaceous nature. Surprisingly, PFs40 from each bacterial strain displayed similar protein bands by SDS-PAGE suggesting that the tested strains share a common exotoxic compound regardless their distinct geographic origin or species.

Experimental infections of Penaeus (Litopenaeus) stylirostris were performed with a Vibrio penaeicida strain (AM101) isolated in New Caledonia from Syndrome 93 diseased shrimp. Cumulative mortalities resulting from intramuscular injection or immersion of shrimp in bacterial suspensions demonstrated high virulence for this bacterial strain and suggested that V. penaeicida could be the etiological agent of Syndrome 93. The median lethal dose (LD50) for AM101 was 1.3 x 10(4) CFU (colony forming units) ml-1 by immersion and less than 5 CFU shrimp-1 by intramuscular challenge, with mortality outbreaks at 48 and 22 h after challenge, respectively. A polymerase chain reaction (PCR) detection assay using a primer set designed from the 16S ribosomal RNA gene of V. penaeicida was developed. It gave an expected amplicon of approximately 310 bp in ethidium bromide-stained agarose gels. The specificity of these primers was assessed with different Vibrio species. Furthermore, DNA extracted by the Chelex method could be used to detect fewer than 20 cultured Vibrio cells in sea-water or shrimp hemolymph by this assay. It appears to be a reliable screening method for detecting V. penaeicida in shrimp and from the aquatic environment.
