Arsenic (As) is a toxic element for plants, animals and humans. The mitigation of As stress has been achieved via selenium (Se) and silica (Si) supplementations. However, the effects of combined application of Se and Si against As stress are not known. The present study was conducted to evaluate the effect of Se (0.5 and 1 mg L−1) and Si (10 and 30 mg L−1) on Oryza sativa L. (rice) subjected to during As (4 mg L−1) stress. The rice plants showed reduction in As accumulation in co-treatment of As + Se, As + Si and As + Se + Si in comparison to As alone treated plants (7 d). The maximum reduction of As accumulation in root and shoot was 57% and 64%, respectively in As + Se + Si treatment. The increase in enzymatic antioxidant system (superoxide dismutase, ascorbate peroxidase, glutathione peroxidase, glutathione reductase and glutathione-s-transferase) along with decrease in oxidative stress markers (lipid peroxidation, dehydroascorbate reductase, ascorbate oxidase and hydrogen peroxide) in As + Se + Si treatment as compared to As treatment signified elevated tolerance of rice plants to As stress. Gene expressions of enzymes involved in antioxidant defence and thiol metabolism were found to be significantly positively correlated with their respective biochemical activities. The present study assessed the utility of Allium cepa based cyto-genotoxicity bioassays in evaluating the arsenic toxicity and remediation potential of Pteris vittata on contaminated soil of Lakhimpur-Kheri district. Untreated and P. vittata treated soil extracts were used for cyto-genotoxicity tests in A. cepa. Results showed that P. vittata extracted high concentration of arsenic, which ranged from 220 to 1420 mgkg−1 in different soils. Cyto-genotoxic assessment of A. cepa showed that extract of P. vittata treated soil had lower cyto-genotoxic effects as compared to untreated soil. A higher mitotic index (10%) while lower mitotic depression (29%), relative abnormality rate (10%), chromosomal aberrations (1%) and micronuclei (2%) were detected in root meristematic cells of A. cepa exposed to remediated soil extract in comparison to untreated soil. The studies provide a simple, rapid and economic cyto-genotoxicity bioassay tool for evaluating toxicity of contaminated soils of contaminated soils as well as revealed the phytoremdiation property of P. vittata against arsenic toxicity. The present study was aimed to analyze the effects of external Zn supply on arsenic (As) toxicity inHydrilla verticillata(L.f.) Royle. The plants were exposed to arsenite (AsIII; 10 μM) with or without 50 and 100 μM Zn. The level of As accumulation (μg g−1dw) after 2 and 4 days was not significantly affected by Zn supply. The plants showed a significant stimulation of the thiol metabolism (nonprotein thiols, cysteine, glutathione-S-transferase activity) upon As(III) exposure in the presence of Zn as compared to As(III) alone treatment. Besides, they did not experience significant toxicity, measured in terms of hydrogen peroxide and malondialdehyde accumulation, which are the indicators of oxidative stress. The minus Zn plants suffered from oxidative stress probably due to insufficient increase in thiols to counteract the stress. Stress amelioration by Zn supply was also evident from antioxidant enzyme activities, which came close to control levels with increasing Zn supply as compared to the increase observed in As(III) alone treatment. Variable Zn supply also modulated the level of photosynthetic pigments and restored them to control levels. Arsenic (As) is a non-essential element whose entry into rice grains is an issue of public concern. The need, therefore, exists to understand the regulatory mechanisms of As accumulation and distribution patterns in plants. This study analyzed the effect of sulfur (S) supply on As accumulation and distribution in rice ( Oryza sativa L. var. IR64) plants. Nine day old seedlings were grown in 0.798 mM S (Normal S), 0.2 mM S (Low S) and 0.003 mM S (Zero S) for 7 days and then subjected to AsIII (20 μM) exposure for 7–15 days. The concentration of As varied significantly in different S treatments with a consistent trend of decrease in As concentration in depleted S supply treatments on both 7 days and 15 days. Arsenic concentration declined from 726 μg g − 1 DW to 537 μg g − 1 DW in roots and from 29 μg g − 1 DW to 8 μg g − 1 DW in shoots at 7 days, and from 1536 μg g − 1 DW to 1062 in roots and from 58 μg g − 1 DW to 16 μg g − 1 DW in shoots at 15 days, when S supply declined from normal to zero. The subcellular distribution of As was also found to vary with alteration in S supply and also differentially in shoot and root. In general, an up-regulation of sulfate transporters of groups 1 and 2 was observed while As transporters (Lsi1 and Lsi2) were down-regulated in response to As exposure to maintain sulfate and to regulate As levels. In spite of S depletion, an increase in cysteine, glutathione (GSH) and phytochelatin (PC) levels was observed upon As exposure. However, some negative impact of S depletion and As could be seen on the growth of plants. The study concluded that even in conditions of decreased S availability, plants continue to rely on thiol metabolism to tackle As levels and its toxicity effectively and altered subcellular distribution of As contributes only partially. enic (As) contamination of the environment has emerged as a concerning issue recently for which phytoremediation has been suggested as a viable solution. Hydrilla verticillata (L.f.) Royle is a widely distributed rapidly growing aquatic weed possessing significant potential to accumulate As and is thus a potential candidate for the purpose of As phytoremediation. In the present study, an investigation of thiol metabolism was conducted in H. verticillata, which revealed differential effects upon exposure to arsenite [As(III)] and arsenate [As(V)]. The accumulation of arsenic was found to be higher upon exposure to As(III) than to As(V). Besides, As(III) was found to induce the activities of enzymes, such as cysteine synthase and γ-glutamylcysteine synthetase and the amounts of cysteine and glutathione (GSH) to higher levels than that observed with As(V). The activity of glutathione-S-transferase was, however, stimulated to a higher level upon exposure to As(V) than As(III). The activity of arsenate reductase was found to increase upon As(V) exposure at all concentrations and durations. In addition, a significant stimulation in the activity of phytochelatin synthase was noticed in vitro with an increase in As/GSH concentration and time of incubation. Arsenic detoxification in H. verticillata thus appeared to involve an induction of thiol synthesis and consumption in a coordinated manner, though differentially upon exposure to As(III) and As(V). The information gained through this study would help in better designing of the pilot experiment at the field level depending on the chemical composition of the contaminated water. The physiological, biochemical, and proteomic changes in germinating rice seedlings were investigated under arsenic stress. A marked decrease in germination percentage, shoot, and root elongation as well as plant biomass was observed with arsenic treatments, as compared to control, whereas accumulation of arsenic and malondialdehyde (MDA) in seedlings were increased significantly with increasing arsenic concentration (both AsIII and AsV). The up-regulation of some antioxidant enzyme activities and the isozymes of superoxide dismutase (SOD, EC 1.15.1.1), ascorbate peroxidase (APX, EC 1.11.1.11), peroxidase (POD, EC 1.11.1.7), and glutathione reductase (GR, 1.6.4.2) substantiated that arsenic accumulation generated oxidative stress, which was more pronounced in As(III) treatment. We also studied the protective effect of reduced glutathione (GSH) and cysteine (Cys) to As(III)/As(V) stressed seedlings. Both GSH and Cys imparted enhanced tolerance to seedlings against arsenic stress. Seedlings growth improved while level of MDA declined significantly when GSH and Cys were supplemented to As(III)/As(V) treatments suggesting GSH and Cys-mediated protection against oxidative stress. The arsenic content was highest in roots of seedlings grown in As(III) in the presence of GSH/Cys. However, in case of As(V) plus GSH or Cys, the arsenic content in seedlings was highest in shoots. Arsenic (As) is a ubiquitous environmental carcinogen that enters the human food chain mainly through rice grains. In the present study, we evaluated the potential of thiourea (TU; non-physiological reactive oxygen species scavenger) in mitigating the negative effects of arsenic (As) stress in indica rice variety IR64, with the overall aim to reduce grain As accumulation. At seedling stage, As + TU treatment induced the formation of more numerous and longer crown roots compared with As alone. The As accumulation in main root, crown root, lower leaf and upper leaf was significantly reduced to 0.1-, 0.14-, 0.16-, 0.14-fold, respectively in As + TU treated seedlings compared with those of As alone. This reduced As accumulation was also coincided with light-dependent suppression in the expression levels of aquaporins and photosynthesis-related genes in As + TU treated roots. In addition, the foliar-supplemented TU under As-stress maintained reducing redox conditions which decreased the rate of As accumulation in flag leaves and, eventually grain As by 0.53-fold compared with those of As treatment. The agronomic feasibility of TU was validated under naturally As contaminated sites of Nadia (West Bengal, India). The tiller numbers and crop productivity (kg seed/ha) of TU-sprayed plants were increased by 1.5- and 1.18-fold, respectively; while, grain As accumulation was reduced by 0.36-fold compared with those of water-sprayed control. Thus, this study established TU application as a sustainable solution for cultivating rice in As-contaminated field conditions. The present study analysed time (0.5 h to 24 h) and tissue [roots, old leaves (OL) and young leaves (YL)] dependent nature of arsenic (As) accumulation and ensuing responses in two contrasting varieties of rice (Oryza sativa L.); Pooja (tolerant) and CO-50 (moderately sensitive). Arsenic accumulation was 5.4-, 4.7- and 7.3-fold higher at 24 h in roots, OL and YL, respectively of var. CO-50 than that in var. Pooja. Arsenic accumulation in YL depicted a delayed accumulation; at 2 h onwards in var. Pooja (0.23 µg g−1 dw) while at 1 h onwards in var. CO50 (0.26 µg g−1 dw). The responses of oxidative stress parameters, antioxidant enzymes, metabolites and ions were also found to be tissue- and time-dependent and depicted differential pattern in the two varieties. Among hormone, salicylic acid and abscisic acid showed variable response in var. Pooja and var. CO-50. Metabolite analysis depicted an involvement of various metabolites in As stress responses of two varieties. In conclusion, an early sensing of the As stress, proper coordination of hormones, biochemical responses, ionic and metabolic profiles allowed var. The present experiment was designed to evaluate physico-chemical characteristics and phyto-genotoxicity of arsenic (As) contaminated soil collected from different sites of Lakhimpur, Uttar Pradesh (UP), India by employing Vicia faba L. The analyses included various biochemical as well as cyto-genotoxicity assays. The results showed that soil pH was slightly acidic to neutral in nature. The bulk density (1.18–1.23 gcm−3), particle density (2.51–2.59 gcm−3) and porosity (44–53%) varied in different places. The level of available nutrients, nitrogen, phosphorus and potassium was found to vary as 124–165 mgkg-1, 173–186 mgkg-1 and 48–98 mgkg-1, respectively. The maximum As levels were found in soil of Fulvareya (27.13 mgkg−1) and Atareya (24.12 mgkg−1), the level of As in water samples of these sites were 0.19 mgl−1and 0.21 mgl−1, respectively. Phytotoxicity of the As present in soils was evident through significant increases in stress metabolites, hydrogen peroxide (H 2 O 2), malondialdehyde (MDA) and carbonyl groups in root and shoot of V. faba. Cyto-genotoxic effects were also seen through reduced mitotic index (MI) and increased mitotic depression (MD), relative abnormality rate (RAR) as well as other chromosomal abnormalities along with micronuclei in root meristematic cells of V. faba. In the present study, arsenic (As) toxicity amelioration potential of a consortium of plant growth promoting rhizobacterium ( Pseudomonas putida ) and alga ( Chlorella vulgaris ) was evaluated during arsenate (AsV) exposure to rice ( Oryza sativa ) plants for 15 d. The consortium mediated amelioration of As toxicity was evident through improved growth of rice plants (root and shoot length and biomass) and reduced oxidative stress [as level of superoxide radicals (O 2 − ), hydrogen peroxide (H 2 O 2 ) and membrane damage]. The positive responses were attributable to a significant decline in As accumulation in root (94 mg kg −1 dw) and shoot (51 mg kg −1 dw) in consortium ( P. putida  +  C. vulgaris ) inoculated seedlings as compared to As alone exposed plants (156 and 98 mg kg −1 dw, respectively). There were also significant changes in the level of various nutrient elements (Mn, Fe, Co, Zn, Mo and Cu), thiols and in the activities of antioxidant and thiol metabolism enzymes in the consortium inoculated seedlings that allowed the plants to tolerate As stress effectively and achieve better growth. Arsenic (As) assessment in agricultural soils and corresponding crops is necessary from the global health safety perspective. To the best of our knowledge, we are reporting for the first time, As flux determining parametric equations for paddy field with seasonal rice cultivation under conventional flooding and dry-wet irrigation approaches. Rigorous field experiments and measuring quantitative parameters, flushed out or percolated into the deeper soil As flux was assessed. A wintery (boro)-monsoonal (aman) study from 2016 to 2019 has been conducted showing the efficiency of dry-wet irrigation on reduction of soil As bioavailability. The reduction in boro was 52.4% in 2016 to 64.8% in 2019 while in aman, it was 61% in 2016 to 74.9% in 2019. Low bioavailability was correlated to plant's internal vascular structure that was found more rigid and firm in dry-wet field grown plants. Observed soil physico-chemical parameters clearly influenced As bioavailability as well as soil microbial community. Assessment of microbial diversity using metagenomics under altered water regime was done by population analysis, relative abundance, species richness, Krona chart comparison.
