The Hexi Corridor is an arid region in northwestern China, where hypoliths are widely distributed, resulting from large amounts of translucent stone pavements. In this region, the water and heat distributions are uneven, with a descent gradient from east to west, which can affect the area's biological composition. The impact of environmental heterogeneity on the distribution of hypolithic microbial communities in this area is poorly understood, and this is an ideal location to investigate the factors that may influence the composition and structure of hypolithic microbial communities. An investigation of different sites with differences in precipitation between east and west revealed that the colonization rate decreased from 91.8% to 17.5% in the hypolithic community. Environmental heterogeneity influenced both the structure and function of the hypolithic community, especially total nitrogen (TN) and soil organic carbon (SOC). However, the effect on taxonomic composition was greater than that on ecological function. The dominant bacterial phyla in all sample sites were Cyanobacteria, Actinobacteria, Proteobacteria, and Deinococcus-Thermus, but the abundances varied significantly between the sampling sites. The eastern site had the highest relative abundance of Proteobacteria (18.43%) and Bacteroidetes (6.32%), while the western site had a higher relative abundance in the phyla Cyanobacteria (62%) and Firmicutes (1.45%); the middle site had a higher relative abundance of Chloroflexi (8.02%) and Gemmatimonadetes (1.87%). The dominant phylum in the fungal community is Ascomycota. Pearson correlation analysis showed that the soil's physicochemical properties were also associated with changes in community diversity at the sample sites. Limited knowledge about the variation patterns of bacterial community composition in the sand and vegetative ecosystems confines our understanding regarding the contribution of the sand dune to desert areas. In this study, 454 pyrosequencing platforms were adopted to determine the community structure of bacteria and diversity of sand dunes in northeastern Qinghai-Tibet Plateau, China: 50 cm deep, rhizosphere, physical crusts, and biological crusts representing sand and vegetative ecosystems, respectively. The findings revealed significant variation in bacterial diversities and the structure of communities in the sand and vegetative ecosystems. The dominant bacterial phyla of sand and vegetative ecosystems were Firmicutes (47%), Actinobacteria (21%), Proteobacteria (16%), and Bacteroidetes (13%), while Lactococcus (50%) was found to be the dominant genus. Furthermore, samples with high alpha-diversity indices (Chao 1 and Shannon) for the vegetative ecosystem have the lowest modularity index and the largest number of biomarkers, with some exceptions. Redundancy analysis exhibited that environmental factors could explain 72% (phyla) and 67% (genera) of the bacterial communities, with EC, TC, and TOC being the major driving factors. This study expands our understanding of bacterial community composition in the desert ecosystem. The findings suggest that variations in the sand and vegetative ecosystems, such as those predicted by environmental factors, may reduce the abundance and diversity of bacteria, a response that likely affects the provision of key ecosystem processes by desert regions. The Qaidam Basin of the Qinghai–Tibet Plateau is a cold, hyper-arid desert that presents extreme challenges to microbial communities. As little is known about variations between surface and subsurface microbial communities, high-throughput DNA sequencing was used in this study to profile bacterial communities of the soil samples collected at different depths in three regions in the Qaidam Basin. The α-diversity indices (Chao, Shannon, and Simpson) indicated that bacterial abundance and diversity were higher in the east and the high-elevation regions compared to the west region. In general, Firmicutes was dominant in the west region, while Proteobacteria and Acidobacteria were dominant in the east and the high-elevation regions. The structure of the bacterial communities differed greatly across regions, being strongly correlated with total organic carbon (TOC) and total nitrogen (TN) content. The differences in bacterial communities between the surface and the subsurface soil samples were smaller than the differences across the regions. Network analyses of environmental factors and bacterial genera indicated significant positive correlations in all regions. Overall, our study provides evidence that TOC and TN are the best predictors of both surface and subsurface bacterial communities across the Qaidam Basin. This study concludes that the bacterial community structure is influenced by both the spatial distance and the local environment, but environmental factors are the primary drivers of bacterial spatial patterns in the Qaidam Basin. Permafrost degradation affects soil properties and vegetation, but little is known about its consequent effects on the soil bacterial community. In this study, we analyzed the bacterial community structure of 12 permafrost-affected soil samples from four principal permafrost types, sub-stable permafrost (SSP), transition permafrost (TP), unstable permafrost (UP) and extremely unstable permafrost (EUP), to investigate the effects of vegetation characteristics and soil properties on bacterial community structure during the process of permafrost degradation. Proteobacteria, Acidobacteria, Actinobacteria and Bacteroidetes were the predominant phyla in all four permafrost soil types. The relative abundance of Proteobacteria decreased in the order SSP > TP> UP > EUP, whereas the abundance of Actinobacteria increased in the order SSP < TP < UP < EUP. Moreover, the Actinobacteria/Proteobacteria ratio increased significantly in the order SSP < TP < UP < EUP along with permafrost degradation, which may be useful as a sign of permafrost degradation. Redundancy analysis (RDA) showed that bacterial communities could be clustered by permafrost types. Analysis of single factors revealed that soil moisture (SM) was the most important factor affecting the bacterial community structure and diversity, followed by soil total nitrogen (STN) and vegetation cover (VC). Partial RDA analysis showed that the soil properties and vegetation characteristics jointly shaped the bacterial community structure. Hence, we can conclude that permafrost degradation, caused by global warming, affects vegetation and soil properties and consequently drives changes in the soil bacterial community structure. The aim of this study was to survey the response of the microbial community to crude oil and the diversity of alkane hydroxylase ( alkB) genes in soil samples from the Qinghai-Tibet Plateau (QTP). The enrichment cultures and clone libraries were used. Finally, 53 isolates and 94 alkB sequences were obtained from 10 pristine soil samples after enrichment at 10 °C with crude oil as sole carbon source. The isolates fell into the phyla Proteobacteria, Actinobacteria, and Bacteroidetes, with the dominance of Pseudomonas and Acinetobacter. The composition of degraders was different from polar habitats where Acinetobacter sp. is not a predominant responder of alkane degradative microbial communities. Phylogenetic analysis showed that the alkB genes from isolates and enrichment communities formed eight clusters and mainly related with alkB genes of Pseudomonas, Rhodococcus, and Acinetobacter. The alkB gene diversity in the QTP was lower than marine environments and polar soil samples. In particular, a total of 10 isolates exhibiting vigorous growth with crude oil could detect no crude oil degradation-related gene sequences, such as alkB, P450, almA, ndoB, and xylE genes. The Shannon-Wiener index of the alkB clone libraries from the QTP ranged from 1.00 to 2.24 which is similar with polar pristine soil samples but lower than that of contaminated soils. These results indicated that the Pseudomonas, Acinetobacter, and Rhodococcus genera are the candidate for in situ bioremediation, and the environment of QTP may be still relatively uncontaminated by crude oil. Variations in the culturable terrestrial bacterial communities and soil biochemical parameters along an altitude gradient (from 1260m to 4111m) upstream of the Shule River, Qinghai-Tibetan Plateau, were investigated. The results showed that the number of cultivable bacteria varied between 0.4×107 and 3.3×107 CFU/g, with an average of 1.6×107 CFU/g. 168 isolates from these soils were clustered into 34 groups by amplified ribosomal DNA restriction analysis (ARDRA). These groups are affiliated to 15 genera that belong to six taxa, α-Proteobacteria, β-Proteobacteria, γ-Proteobacteria, Actinobacteria, Firmicutes and Bacteroides, of which Bacillus and Arthrobacter were the dominant species at the level of the genus. The relative abundance of Arthrobacter increased significantly at high altitude. Correlation analysis showed that the total number of culturable bacteria in the soils decreased first and then increased below 3000m, and these trends significantly positively correlated to the soil organic C, total N and the activities of soil sucrase, and positively correlated to the activities of soil urease and catalase. However, when the altitude exceeded 3000m, cultivable bacterial number dramatically declined, although soil organic carbon, total nitrogen and enzyme activities were relatively high. While the diversity index of bacteria increased along with the increased altitude as a whole and there existed a significantly positive correlation between altitude and bacteria diversity. Together, these results illustrated that culturable bacterial numbers were mainly influenced by soil biochemical properties while bacterial diversity was mainly influenced by altitude in this region. Bacterial communities in cold-desert habitats play an important ecological role. However, the variation in bacterial diversity and community composition of the cold-desert ecosystem in Qinghai–Tibet Plateau remains unknown. To fill this scientific gape, Illumina MiSeq sequencing was performed on 15 soil samples collected from different cold-desert habitats, including human-disturbed, vegetation coverage, desert land, and sand dune. The abundance-based coverage estimator, Shannon, and Chao indices showed that the bacterial diversity and abundance of the cold-desert were high. A significant variation reported in the bacterial diversity and community composition across the study area. Proteobacteria accounted for the largest proportion (12.4–55.7%) of all sequences, followed by Actinobacteria (9.2–39.7%), Bacteroidetes (1.8–21.5%), and Chloroflexi (2.7–12.6%). Furthermore, unclassified genera dominated in human-disturbed habitats. The community profiles of GeErMu, HongLiangHe, and CuoNaHu sites were different and metagenomic biomarkers were higher (22) in CuoNaHu sites. Among the soil physicochemical variables, the total nitrogen and electric conductivity significantly influenced the bacterial community structure. In conclusion, this study provides information regarding variation in diversity and composition of bacterial communities and elucidates the association between bacterial community structures and soil physicochemical variables in cold-desert habitats of Qinghai–Tibet Plateau. Research has shown that forest management can improve the post-drought growth and resilience of Qinghai spruce in the eastern Qilian Mountains, located on the northeastern Tibetan Plateau. However, the impact of such management on the tree-associated phyllosphere microbiome is not yet fully understood. This study provides new evidence of positive forest management effects on the phyllosphere microbiome after extreme drought, from the perspectives of community diversity, structure, network inference, keystone species, and assembly processes. In managed Qinghai spruce forest, the α -diversity of the phyllosphere bacterial communities increased, whereas the β -diversity decreased. In addition, the phyllosphere bacterial community became more stable and resistant, yet less complex, following forest management. Keystone species inferred from a bacterial network also changed under forest management. Furthermore, forest management mediated changes in community assembly processes, intensifying the influence of determinacy, while diminishing that of stochasticity. These findings support the hypothesis that management can re-assemble the phyllosphere bacterial community, enhance community stability, and ultimately improve tree growth. Overall, the study highlights the importance of forest management on the phyllosphere microbiome and furnishes new insights into forest conservation from the perspective of managing microbial processes and effects. Microorganisms can accelerate the deterioration of stone monuments. In this study, we used next-generation sequencing (Illumina MiSeq PE300 Platform) to investigate the microbial biofilm communities that have developed on sandstone in two famous Buddhist cave temples, the Maijishan Grottoes and Tiantishan Grottoes, located on the historical Silk Road in western China. We found high bacterial diversity and relatively low eukaryota diversity based on 16S and 18S ribosomal RNA (rRNA) sequencing analysis. The dominant bacterial groups were Cyanobacteria, Actinobacteria, Proteobacteria, Bacteroidetes, and Chloroflexi. The dominant eukaryota were Ascomycota, Basidiomycota, and Chytridiomycota. The most dominant genes among the bacteria were from Cyanobacteria (unclassified and no rank), Pseudonocardia , Kribbella, and Rubrobacter ; the dominant fungal genes were from unclassified Lecanorales and Capnodiales (unclassified and norank). These microbes form biofilms that will contribute to biodeterioration of the underlying sandstone. Combining with field monitoring and microscopic analysis, we determined that high bioreceptivity of the sandstone and locally available water were two important exogenous and endogenous factors that promoted microbial colonization, proliferation, and subsequent biofilm formation. The aforementioned microbes usually coexisted on the sandstone surfaces by building an inter-connected and dynamic community capable of adapting to and resisting the harsh local environmental conditions to survive and undergo succession. • A comparative analysis of microbial communities of stone biofilms of the two Grottoes. • High bioreceptivity of sandstone and available water are two of the most important factors for biofilm formation. • Strongly positive intra-domain connections contributed to resistance to harsh environment.
