The red mite, Dermanyssus gallinae (Mesostigmata: Dermanyssidae), is one of the most serious pests in chicken farming and causes serious economic losses. Overuse of chemical acaricides has caused pest resistance as well as environmental residual toxicity to chickens and eggs. Therefore, given the warm and humid conditions of chicken farms, alternative application with entomopathogenic Beauveria bassiana sensu lato was investigated to control D. gallinae. Based on the characterization and virulence assays of B. bassiana isolates against red mite, a red mite-pathogenic fungal library was constructed. A total of six highly virulent B. bassiana isolates were selected from the first bioassay. They showed 70–100% mortality in ten days which were higher than cypermethrin, showing 40% mortality. Considering their fungal virulence, conidial production, and thermotolerance, isolate JEF-410 was established to be the best candidate for industrialization. JEF-410 showed a dose-dependent virulence against red mite nymphs. JEF-410 conidia from millet granule-based mass culture were applied to a commercial chicken farm and it showed a significant suppression of population increase under high humidity conditions. In summary, B. bassiana JEF-410 having higher control efficacy than cypermethrin could be effectively used to control D. gallinae in the fungus-friendly humid chicken farms. This work solved challenges of mass production of B. bassiana and management of the red mite in poultry farming under right conditions of RH and temperature.

The melon thrip, Thrips palmi Karny, is a worldwide pest that causes severe damage to a wide range of host plants. Current management mainly relies on sprayable synthetic chemicals that target adult, egg, and larval stages. However, these chemicals have adverse effects on the environment and induce resistance in thrips. An alternative environmentally sound management method with a different mode of action, targeting the soil-dwelling stage of thrips, was considered in this work. Bioassay of seven Beauveria bassiana isolates under laboratory conditions revealed that two isolates, JEF-350 and JEF-341, were highly virulent against melon thrips. Conidial production and thermotolerance of JEF-341 and JEF-350 were compared with those of the commercial isolate B. bassiana ERL836 (Chongchaesak GR). B. bassiana JEF-350 showed similar conidial production to ERL836 but had significantly higher thermotolerance than JEF-341 and ERL836. Dose-dependent virulence of JEF-350 was tested in pot and field conditions, and JEF-350 millet-based fungal granules were applied to the soil surface to target the pupal stage of melon thrips. In the pot and field trials, JEF-350 granules significantly reduced thrip population with greater efficacy than JEF-341 and ERL836 granules. In application, JEF-350 needs to be carefully applied when in combination with chemical fungicides to ensure survival. Our results suggest that B. bassiana JEF-350 could be effectively used to control the pupal stage of melon thrips in soil.

The species of Beauveria bassiana is widely used for the management of agricultural insect pests. In this study, we integrated egfp-double-stranded RNA (dsRNA) to a previously generated egfp-expressing B. bassiana transformant (Bb-egfp#3) using a protoplast integration method. The Bb-egfp#3 protoplast was mixed with the dsRNA under PEG/CaCl2 conditions and liquid-cultured in Sabouraud dextrose broth for 5 days. A control culture followed the same procedure without dsRNA. Bb-egfp#3/egfp-dsRNA cultures showed very low fungal growth (OD630 = 0.2) compared to the control culture, Bb-egfp#3 only (OD630 = 1.1). Screening of possible transformants on Sabouraud dextrose agar revealed a transformant T3, without egfp signal. T3 was confirmed as B. bassiana through sequencing of conserved genes and insect bioassays. Interestingly, the genomic egfp fragment of T3 was disrupted, and the egfp signal was not detected over four subcultures, which was also confirmed by RNA-seq of Bb-egfp#3 and T3. This study provides an interesting observation that protoplast integration with dsRNA could possibly generate significantly reduced gene expression in B. bassiana and it is stable across several generations.

Chemical control is widely used to control the Japanese pine sawyer beetle, Monochamus alternatus, but strong chemical regulations require an environmentally sound management strategy. In this work, we investigated the use of entomopathogenic fungi and their application as a means of practical pest management. Thirty-two diverse species of fungal isolates were assayed against adult pine sawyer beetles using a contact method under laboratory conditions, and four isolates showed over 70% virulence consequently. These isolates, two each of Beauveria bassiana and Metarhizium anisopliae were sprayed on the adult beetles at 1 × 107 conidia/ml in plastic containers, respectively. The M. anisopliae-treated adult beetles showed 67% mortality. M. anisopliae isolates JEF-197 and JEF-279 demonstrated dosage-dependent insecticidal activity. Following the laboratory experiments, semi-field trials were conducted in young pine trees under high (RH 94%) and low (RH 35%) humidity conditions. In the high humidity conditions, most of the adult beetles stayed on the top of the branches. When the two M. anisopliae isolates were sprayed on the beetles, they showed ca. 50–70% insecticidal activity 11 days after application. In contrast, in low humidity conditions, the adult beetles tried to move off the branches and onto the soil. When the beetles reached the JEF-197 and JEF-279-treated soil, we measured >90% insecticidal activity. This work suggests that M. anisopliae was the most virulent entomopathogenic fungus against adult Japanese pine sawyer beetles, and this forest insect could be ecologically controlled by the spray and soil application of the M. anisopliae isolates.

Beauveria bassiana (Balsamo) Vuillemin (Hypocreales: Cordycipitaceae) ERL836 has been commercialized under the name ChongchaeSak to control an agricultural insect pest, the western flower thrips Frankliniella occidentalis Pergande (Thysanoptera: Thripidae), in the Republic of Korea. As soon as it was launched in 2017, it became a popular product and has received a positive response. However, study of the storage stability of the fungus ERL836 has yet to be investigated. To determine the optimum conditions for long-term storage, we assessed conidial viability and insecticidal activity of B. bassiana ERL836 according to storage temperature and culture substrate. Viability of B. bassiana ERL836 conidia from mycotized grains (millet and rice) stored at low (4 °C) and moderate (25 and 30 °C) temperatures was maintained at >85% for 24 and 18 months, respectively, along with insecticidal activity. In contrast, the samples stored at 37 °C showed low germination rate (about 80% germination rate for only 5 months). This result suggests that low and moderate temperatures (4 to 30 °C) conserve B. bassiana ERL836 viability and virulence.

Background Insect-killing fungal species, Beauveria bassiana, is as an environment-friendly pest management tool, and many isolates are on the track of industrialization. However, some of B. bassiana isolates show similar morphology and virulence against insect pests, and so it is hard to differentiate them. Herein we used two patented isolates, ERL836 and JEF-007, and investigated their virulence against western flower thrips, Frankliniella occidentalis, and further analyzed genome structures and transcriptional responses when interacting with cuticles of thrips to see possible differences on the initial step of fungal infection. Results The two isolates showed no significant differences in fungal growth, conidial production, and virulence against thrips, and they were structurally similar in genome. But, in transcription level, ERL836 appeared to infect thrips easily, while JEF-007 appeared to have more difficulty. In the GO analysis of ERL836 DEGs (differentially expressed genes), the number of up-regulated genes was much larger than that of down-regulated genes, when compared to JEF-007 DEGs (more genes down-regulated). Interestingly, in the enrichment analysis using shared DEGs between two infecting isolates, plasma membrane-mediated transporter activity and fatty acid degradation pathway including cytochrome P450 were more active in infecting ERL836. Conclusion The two B. bassiana isolates had similar morphology and virulence as well as genome structure, but in transcription level they differently interacted with the cuticle of western flower thrips. This comparative approach using shared DEG analysis could be easily applied to characterize the difference of the two B. bassiana isolates, JEF-007 and ERL836.

Dermacentor albipictus (Acari: Ixodidae), the winter tick, is a one-host tick that parasitizes large ungulates. They can dramatically affect moose, Alces alces (Artiodactyla: Cervidae), causing significant physiological and metabolic stress and mortality among heavily parasitized individuals. Entomopathogenic fungi in the genera Metarhizium (Hypocreales: Clavicipitaceae) and Beauveria (Hypocreales: Cordycipitaceae) are promising tick biological control agents. We examined the pathogenicity of experimental and commercially formulated isolates of M. anisopliae, M. brunneum and B. bassiana sprayed at concentrations of 106, 107 and 108 conidia/mL against the larval stage of D. albipictus and assessed the efficacy of spraying the commercial product Met52®EC, containing M. brunneum, strain F52, under laboratory conditions. Results showed larval D. albipictus mortality was significantly higher and occurred earlier when treated with M. anisopliae and M. brunneum isolates compared to B. bassiana at 106, 107 and 108 conidia/mL. Mortality was observed as early as 3 days in the M. anisopliae and M. brunneum treatments and after 6 days in the B. bassiana treatments. After 21 days, larval mortality ranged from 74–99% when ticks were treated with M. anisopliae and M. brunneum isolates at 106, 107 and 108 and conidia/mL. In contrast, mortality of ticks treated with B. bassiana ranged from 30 to 64%. When larvae were treated with the commercial product Met52, mortality was ~ 45% after 3 days and ~ 96% after 9 days. These results demonstrate the effectiveness of M. anisopliae and M. brunneum against D. albipictus.

Beauveria bassiana is a species complex whose isolates show considerable natural genetic variability. However, little is known about how this genetic diversity affects the fungus performance. Herein, we characterized the diversity of genes involved in various mechanisms of the infective cycle of 42 isolates that have different growth rates, thermotolerance and virulence. The analysed genes showed general genetic diversity measured as non-synonymous changes (NSC) and copy number variation (CNV), with most of them being subjected to positive episodic diversifying selection. Correlation analyses between NSC or CNV and the isolate virulence, thermotolerance and growth rate revealed that various genes shaped the biological features of the fungus. Lectin-like, mucin signalling, Biotrophy associated and chitinase genes NSCs correlated with the three biological features of B. bassiana. In addition, other genes (i.e. DNA photolyase and cyclophilin B) that had relatively conserved sequences, had variable CNs across the isolates which were correlated with the variability of either virulence or thermotolerance of B. bassiana isolates. The data obtained is important for a better understanding of population structure, ecological and potential impact when isolates are used as mycoinsecticides and can justify industrialization of new isolates.

Background Entomopathogenic Beauveria bassiana has been used as a biocontrol agent for insect pests, but its effect at the molecular level on the hosts has not been studied in detail. Herein, we performed transcriptome analysis of bean bug, Riptortus pedestris (Hemiptera: Alydidae) in response to infection with a highly virulent strain of B. bassiana JEF-007 (Bb JEF-007). Results Based on RNA-seq data from R. pedestris infected with Bb JEF-007 compared with non-infected bean bugs, infection was assumed to strongly activate (i) the energy production pathway by expressing dehydrogenases, (ii) metabolic pathways by expressing secreted proteins, GTPase, MBF2 transcription factor family, pigment-dispersing factor, antioxidants, and cuticle proteins, and (iii) the immune response pathway by expressing serine-threonine kinase in Toll pathway of bean bug. Conclusion We have established the platform for functional studies of the genes required for an immune response against entomopathogenic fungi like B. bassiana in the bean bug, R. pedestris. Moreover, this study also paves the way for genetic modification of B. bassiana to combat with the defense mechanism of R. pedestris. © 2018 Society of Chemical Industry.

Insect-killing fungi have high potential in pest management. A deeper insight into the fungal genes at the whole genome level is necessary to understand the inter-species or intra-species genetic diversity of fungal genes, and to select excellent isolates. In this work, we conducted a whole genome sequencing of Beauveria bassiana (Bb) JEF-007 and characterized pathogenesis-related features and compared with other isolates including Bb ARSEF2860. A large number of Bb JEF-007 genes showed high identity with Bb ARSEF2860, but some genes showed moderate or low identity. The two Bb isolates showed a significant difference in vegetative growth, antibiotic-susceptibility, and virulence against Tenebrio molitor larvae. When highly identical genes between the two Bb isolates were subjected to real-time PCR, their transcription levels were different, particularly in heat shock protein 30 (hsp30) gene which is related to conidial thermotolerance. In several B. bassiana isolates, chitinases and trypsin-like protease genes involved in pathogenesis were highly conserved, but other genes showed noticeable sequence variation within the same species. Given the transcriptional and genetic diversity in B. bassiana, a selection of virulent isolates with industrial advantages is a pre-requisite, and this genetic approach could support the development of excellent biopesticides with intellectual property protection.

The western flower thrip, Frankliniella occidentalis (Pergande) (Thysanoptera: Thripidae), is recognized as a serious pest worldwide, but overuse of chemical pesticides results in environmental pollution and insect resistance. Herein we report the biocontrol of soil-dwelling stages of western flower thrips using entomopathogenic Beauveria bassiana granules. Using a B. bassiana isolate expressing enhanced green fluorescent protein (Bb-egfp), we confirmed that Bb-egfp was highly virulent to the thrips and colonized the soil, which allowed soil-dwelling pupae to come in contact with the colonized fungal mass. In a glasshouse trial, granules of BbERL836 and BbERL1578 isolates were applied to the soil surface of tomatoes growing in pots and female F. occidentalis were released on tomato plants three days after the fungal application. Populations reductions between 75 and 90% were observed in the treatments of BbERL836 and BbERL1578 at 20 and 40 days after application (chemical control: 85% reduction). In the field trial with cucumbers, the two ERL soil treatments showed ca. 90% control efficacy, similar to that of spinetorm, a semi-synthetic insecticide (85% efficacy). This study shows that using entomopathogenic B. bassiana granules to kill soil-dwelling stages of thrips is an effective strategy to manage F. occidentalis populations in tomato and cucumber.

Most entomopathogenic fungal biopesticides have been produced in wettable powder and liquid concentrate formulations. Separation of fungal conidia from granular substrates to manufacture these formulations is costly and labor-intensive, which has hampered growth of the fungal biopesticide industry. Herein to overcome this challenge, we developed a novel screen bag formulation using mycotized granules produced from solid-substrate cultures of Beauveria bassiana JEF-007 and Metarhizium anisopliae JEF-003. The isolates were solid-cultured on Italian millet grains and packed in screen bags (pore size: ~ 15 μm). The bags containing the mycotized granules were immersed in water and agitated to disperse the conidia, after which the number of conidia released into the water was determined. These results were compared to the number of conidia released after vortexing the granules in water as a control. Various surfactants were added to the water to assess their effect on conidial release. Silwet L-77 yielded the highest conidial release. The insecticidal activity against Riptortus pedestris nymphs of suspensions produced from the screen bag formulation and vortexed conidial suspensions was similar, demonstrating that our novel formulation could be a cost-effective replacement for standard WP and ES formulations. The screen bag formulation has several advantages over other standard fungal-based formulations, such as a shorter and lower-cost production process. This formulation technology could also be applied to other conidial-based biopesticides, such as fungal fungicides, herbicides, and fertilizers.

The bean bug, Riptortus pedestris, is a major agricultural pest that reduces crop quality and value. Chemical pesticides have contributed to pest management, but resistance to these chemicals has significantly limited their use. Alternative strategies with different modes of action, such as entomopathogenic fungi, are therefore of great interest. Herein, we explored how entomopathogenic fungi can potentially be used to control the bean bug and focused on identifying virulence-related genes. Beauveria bassiana (JEF isolates) were assayed against bean bugs under laboratory conditions. One isolate, JEF-007, showed >80 % virulence by both spray and contact exposure methods. Agrobacterium tumefaciens-mediated transformation (AtMT) of JEF-007 generated 249 random transformants, two of which (B1-06 and C1-49) showed significantly reduced virulence against Tenebrio molitor and R. pedestris immatures. Both species were used for rapid screening of virulence-reduced mutants. The two transformants had different morphologies, conidial production, and thermotolerance than the wild type. To determine the localization of the randomly inserted T-DNA, thermal asymmetric interlaced (TAIL) PCR was conducted and analysis of the two clones found multiple T-DNA insertions (two in B1-06 and three in C1-49). Genes encoding complex I intermediate-associated protein 30 (CIA30) and the autophagy protein (Atg22) were possibly disrupted by the T-DNA insertion and might be involved in the virulence. This work provides a strong platform for future functional genetic studies of bean bug-pathogenic B. bassiana. The genes putatively involved in fungal virulence should be experimentally validated by knockdown in future studies.

In the present study, an enhanced green fluorescent protein gene (egfp) was expressed in Beauveria bassiana ERL836 based on the Agrobacterium tumefaciens-mediated transformation (AtMT) method. The ERL836 transformants were generated with pCambia-egfp binary vectors. Ten transformants were randomly selected and analyzed for the T-DNA insertion and gene expression. The results revealed that 60% (egfp) of the fungal putative transformants were inserted by the T-DNA fragment. The expressions of egfp in putative transformants were rapidly detected by reverse transcription PCR (RT-PCR) method. Among these transformants, 33.33% (egfp, 2 transformants) expressed the target genes. Two selected egfp transformants showed strong green fluorescence with different expression levels. The results of this study could provide a successful procedure for foreign protein production in B. bassiana by using the AtMT method and accomplished with a rapid selection of fungal transformants using RT-PCR.

Persistence of granular formulations of Beauveria bassiana sensu lato, strains GHA726 and ERL1170, and Metarhizium anisopliae sensu lato, strains ERL824 and ERL701, applied to forest soil was documented. One formulation was prepared by producing fungi on millet and at application adding cooked, dried millet to provide additional nutrients for fungal growth. The other was made by producing fungi on corn starch ‘packing peanuts’, a biodegradable packaging material, adding additional packing peanuts as a nutrient base. The trial was conducted in three sites in a forest predominating in sugar maple (Acer saccharum) in Underhill, VT, USA. In each site, 33 dominant sugar maples were randomly selected. A 1-m2 plot located 1 m from the bole of the tree was established. Soil samples were collected before application, and 1 h, 2, 4, 8, 12 and 16 wk post-application. Beauveria bassiana is an entomopathogenic fungi used in environmentally mindful pest management. Its main active ingredient, conidia, is commercially available as a fungal biopesticide. Many studies of conidia production have focused on how to optimize culture conditions for maximum productivity and stability against unfavorable abiotic factors. However, understanding of how conidiogenesis-related genes provide improved conidial production remains unclear. In this study, we focus on identifying conidiogenesis-related genes in B. bassiana ERL1170 using a random mutagenesis technique. Transformation of ERL1170 using restriction enzyme-mediated integration generated one morphologically different transformant, ERL1170-pABeG #163. The transformant was confirmed to represent B. bassiana, and the binary vector was successfully integrated into the genome of ERL1170. Entomopathogenic fungi have potential for controlling brown planthoppers in paddy. Currently only sprayable formulations can be applied despite the fact that granular formulations may reduce physical labor during application. Herein, granules of a Beauveria bassiana sensu lato (Balsamo) Vuillemin (Hypocreales: Clavicipitaceae) isolate were produced from a millet-based solid culture. The efficacy of the granules to control brown planthoppers, Nilaparvata lugens (Stal) (Hemiptera: Delphacidae), was determined by applying them to water surrounding rice plants in the laboratory and then infesting the plants with brown planthoppers. In a spray-mediated bioassay to select the most virulent of four B. bassiana s.l. isolates, ERL836 was the most virulent and caused minimal damage to rice plants. Dropping the ERL836 granules on water provided similar control to spraying the conidia. In the laboratory ERL836-colonized millet granules were applied to the water and simultaneously infested with brown planthoppers. The granular treatment had 87.7 % control efficacy 15 days post-application and etofenprox 0.5 % granules had 94.6 % efficacy. ERL836 colonized the surface of the water and brown planthoppers underwent mycosis three and 15 days post-application. Mycelial growth on the stems of rice plants was observed using a B. bassiana s.l. mutant that expresses an enhanced green fluorescence protein. Applying millet-based ERL836 granules to water can be an efficient biological strategy to manage brown planthoppers in paddy. This method is economical and requires less work than spraying the fungi directly on rice.

Larvae of mealworms Tenebrio molitor L. (Coleoptera: Tenebrionidae) have been used as animal feed, but fungal pathogens rapidly downsize the populations, resulting in economic losses. In this work, we established an effective management strategy for fungal pathogens. An entomopathogenic fungus, Beauveria bassiana, was isolated from mealworm cadavers. The bioassay of some isolates of this species at >90% relative humidity revealed that the ERL1575 isolate had the highest virulence. At 20–30% RH, ERL1575 conidia when ingested produced 80% mortality but when sprayed topically produced only 95%) for 5 days. BACKGROUND Spraying of entomopathogenic fungi formulated as wettable powders and suspension concentrates based on oils or water has been used for the management of crop pests. However, the use of fungal granules to control paddy field pests has not been fully explored. Herein, several Beauveria bassiana isolates (ERL1170, ERL1578 and ERL836) were produced as granules using a millet-based solid culture. The granules were applied to the rice nursery 3 days before transplanting, and their control efficacy against rice water weevils was determined in paddy fields. RESULTS The solid cultures produced ∼1 × 108 conidia g−1 of millet grains 10 days after the inoculation. The granules were applied to the soil in the rice nursery 3 days before the rice seedlings were transplanted in the paddy fields. Rice in plots with granules of ERL1578 had 17.3% leaf damage (74% control efficacy) 14 days post-application, whereas rice plants in the untreated control had 66.5% damage. Rice plants treated in the nursery with ERL1170 and ERL836 had 52–54% damage. In the rice plots previously treated with ERL1578, the smallest numbers of larvae and adults were observed 38 days post-application. In laboratory conditions, ERL1578-treated larvae turned pink and were covered with mycelial mass. CONCLUSION Applications of millet-based B. bassiana granules on rice nursery soil can be an effective and efficient biological control strategy for the management of rice water weevils. This method can be relatively inexpensive and requires less labor than practices involving the spraying of fungi directly on rice in paddy fields. © 2014 Society of Chemical Industry

Beauveria bassiana (Bb) is used as an environment-friendly biopesticide. However, the molecular mechanisms of Bb-host interactions are not well understood. Herein, RNA isolated from B. bassiana (Bb JEF-007) and Riptortus pedestris (Hemiptera: Alydidae) infected with this strain were firstly subjected to high-throughput next generation sequencing (NGS) to analyze and compare transcriptomes. Due to lack of fungal and host genome information, fungal transcriptome was processed to partially exclude non-infection specific genes and host-flora. Differentially Expressed Gene (DEG) analysis showed that 2381 genes were up-regulated and 2303 genes were down-regulated upon infection. Most DEGs were classified into the categories of single-organism, cellular and metabolism processes by Gene Ontology analysis. Most DEGs were involved in metabolic pathways based on Kyoto Encyclopedia of Genes and Genomes pathway mapping. Carbon metabolism-related enzymes in the glyoxylate cycle were significantly up-regulated, suggesting a possible role for them in Bb growth in the host. Additionally, transcript levels of several fungal genes were dramatically increased after infection, such as cytotoxic lectin-like protein, bacterial-like toxin, proteins related to cell wall formation, hyphal growth, nutrient uptake, and halogenated compound synthesis. This work provides insight into how entomopathogenic B. bassiana grows in agriculturally harmful bean bug at 6 d post infection.

Western flower thrips (WFT), Franklinella occidentalis, is a major pest of ornamentals. Mycotized millet grains with entomopathogenic fungi applied to soil of potted marigold plants were tested to target the soil-dwelling stages of thrips. Two experimental fungal isolates, (Beauveria bassiana [ARS7060] and Metarhizium anisopliae [ERL1171]), were compared with the registered B. bassiana strain GHA [commercialized as BotaniGard®] and untreated controls in greenhouse caged trials. Mycotized millet grains were mixed into the upper surface of the potting soil in pots of flowering ‘Hero Yellow’ marigolds (4 g/pot). One week after application five mated WFT females were released onto each plant (four plants per cage). At 8 weeks post-infestation, the mean total number of thrips per plant was 81% and 90% less in the ERL1171 and ARS 7060 treatments, respectively, than in the control. The mean numbers of thrips per plant for the control and GHA treatments were not significantly different. Plant damage was 60% less on plants treated with the experimental fungi than the control and GHA treatments. At 10 weeks post-application, 75–90% of WFT collected from the treatments of the experimental fungi were infected with the fungal species applied. These results demonstrate that soil applications of entomopathogenic fungi can reduce WFT populations significantly and prevent major damage.

Low thermotolerance in entomopathogenic fungi is the main impediment to their industrialization. This research, for the first time, describes the generation of a thermotolerant colony by pairing and subculturing (cycling) two Beauveria bassiana isolates without sexual reproduction. A mixture of B. bassiana ERL1578 and ERL1576 was inoculated on quarter-strength Sabouraud dextrose agar with yeast extract (¼SDAY). The paired culture (ERL1578 + 1576) was cycled three times to increase the frequency of possible hyphal fusion at the first cycle (c. 5/5 × 10(5) conidia), followed by a heat treatment as a selection pressure. Two non-paired isolates served as controls. Two morphologically different colonies (BbHet1 and BbHet2) were isolated from the pairing. BbHet1 colony had the highest conidial yield. BbHet2 had the most rapid mycelial growth and produced sponge-like mycelial masses (the others were flat), and its conidia were darker than the non-paired colonies under a microscope (400×). BbHet2 conidia had 60.7% germination after exposure to 45 °C for 60 min (the others had 15%) without significant loss of virulence against Western flower thrips, Frankliniella occidentalis; however, there was a slight decrease in conidial yield. The new phenotypes formed suggested that a genetic variation happened as a result of heterokaryosis and/or recombination, more than environmental adaptation, when mixing different conidia. This methodology seems to be very useful for enhancing thermotolerance in fungi.

Enzymes from Hypocrealean entomopathogenic fungi often encounter unfavorable abiotic and biotic factors during pathogenesis. The present work describes the roles of adjuvants, such as corn oil and polyoxyethylene-(3)-isotridecyl ether (TDE-3), in promoting aphicidal activity of enzyme precipitate from Beauveria bassiana SFB-205 supernatant. Supernatant enzymes including chitinase and proteases were lyophilized by attagel-mediated protein precipitation to produce attagel-mediated enzyme powder (AMEP). Corn oil-based AMEP + TDE-3 suspension showed 96.3% control efficacy against cotton aphids, Aphis gossypii Glover (Hemiptera: Aphididae), in glasshouse conditions at 2 days post-application, whereas water-based AMEP + TDE-3 suspension or TDE-3 alone showed d -acetylglucosaminide) under a drying condition compared to water-based AMEP + TDE-3 suspension. TDE-3 made supernatant, a source material of AMEP, degraded more cotton aphid proteins than supernatant alone in SDS-polyacrylamide gel electrophoresis; supernatant was used to clearly show the degradation without interfering with other proteins such as AMEP. These results suggest that 1) corn oil can slow down the evaporation of the diluted suspension drop and provide more time for enzymes from AMEP to degrade more cuticles at the time of application and 2) TDE-3 can disrupt chitin–protein matrixes within procuticle and facilitate enzymes from AMEP in degrading proteins which increases the exposure of chitin fibers to the attack of chitinases. This approach can provide another strategy in developing biopesticides using entomopathogenic fungi.

Hypocrealean entomopathogenic fungal conidia are made up of multi-aged groups given their chronological conidiogenesis. Most thermotolerance assays have been conducted using mixed-age conidia. The present work exploited a polysiloxane polyether copolymer (siloxane) (Silwet L-77®) mediated conidial collection method, validated by a hydrophobicity assay. This was done to divide mixed-age conidia into two groups based on hydrophobicity and test their thermotolerance, relying on the relationship of conidial age with hydrophobicity. Beauveria bassiana GHA and ERL1170 and Metarhizium anisopliae ERL1171 and ERL1540 conidia, produced on millet agar, whey permeate agar, and ¼SDAY were subjected to hydrophobicity assays that included data on yield of conidia/unit of surface area. Conidia were also collected using 0.01% siloxane, and those remaining with 0.08% siloxane. Hydrophobicity was correlated with percent conidia collected in the two siloxane solutions and yield, suggesting a relationship between percent conidia collected and conidial age (maturation). The conidial suspensions were exposed to 45°C for 45min, and conidial germination was examined. Overall, conidia which were collected in 0.08% siloxane had lower germination after heat exposure than those collected in the 0.01% solution. Conidia of both fungi produced by incubation on millet or whey permeate for 14d were more hydrophobic and exhibited greater thermotolerance than those produced on ¼SDAY. These results suggest that conidia can be divided into two groups with different thermotolerance by using a siloxane-mediated conidial collection method based on hydrophobicity. This depends on the types of substrates used that could influence conidial maturation.

Thermotolerance of entomopathogenic (insect-killing) fungi should be seriously considered before industrialization. This work describes the feasibility of millet grain as a substrate for production of thermotolerant Beauveria bassiana (Bb) GHA and ERL1170 and Metarhizium anisopliae (Ma) ERL1171 and ERL1540 conidia. First, conidial suspensions of the Bb isolates, produced on millet grain in polyethylene bags, were exposed to five temperatures (43-47 °C) at 15-min intervals for up to 120 min (experiment I). Agar-based quarter-strength (¼) Sabouraud dextrose agar supplemented with yeast extract (SDAY) and whey permeate media served as controls. Millet-grain-based culture was superior in producing the most thermotolerant Bb conidia, followed by whey permeate agar and ¼SDAY-based cultures. Secondly, to compare the thermotolerance of conidia produced at the same conditions, the Bb isolates were then produced on agar-based millet powder medium, with ¼SDAY and whey permeate agar media as controls, and the two Ma isolates were added (experiment II). They were then exposed to the same temperatures as above. More thermotolerant Bb and Ma conidia were produced on millet powder agar than on whey permeate agar and ¼SDAY overall. These results suggest that millet grain can be used as a substrate to produce thermotolerant conidia in a mass production system.

The downward movement of Beauveria bassiana (Balsamo) Vuillemin GHA, in potting medium with different levels of moisture and top-watering was compared. The population of B. bassiana was greatest in the upper layer of the potting medium without significant movement 18 d post-application, finding no B. bassiana in untreated pots.

Conidia from Beauveria bassiana and Metarhizium anisopliae, produced in quarter-strength Sabouraud dextrose agar amended with yeast extract, whey permeate, and millet agar, were exposed to 45°C for 90 min prior to and at 30 d post-storage at 25°C. B. bassiana produced in whey permeate or millet had better thermotolerance than the other treatments after the storage.

The two FPLC fractions from Beauveria bassiana SFB-205 supernatant, displaying chitinase or Pr1/Pr2 protease activity were bioassayed against Aphis gossypii in different ratios. The decrease of the aphid population was more significantly influenced by the chitinase fraction in a dosage-dependent manner.

The supernatant of Beauveria bassiana SFB-205 reduced the population of cotton aphid, Aphis gossypii Glover, with a dosage-dependant manner, which allowed a quality control (QC) factor to be determined for the evaluation of the supernatant as the first step of a development. Enzymes were assumed as possible QC factors based on 1) the comparable aphicidal activity of the supernatant protein pellet to the raw supernatant, 2) the supernatant-induced degradation of the insect cuticles, observed by transmission electron microscopy, and 3) the confirmation of enzymes related to the fungal penetration – chitinase, and the Pr1- and Pr2 proteases – in the supernatant. Finally, from the bioassay with the enzyme-inhibited supernatants processed by substrate inhibition one by one, decreased aphicidal activities were observed for all three enzyme-inhibited treatments. This phenomenon, furthermore, was more remarkable in the chitinase-inhibited supernatant. This finding provides that those enzymes (and most particularly the chitinase) in the supernatant were strongly involved in the aphicidal activity. Consequently, the amount of the chitinase may be used as one of the QC factors to determine the insecticidal activity of the supernatant of B. bassiana SFB-205 in the optimization of mass production.

The brown marmorated stink bug. Halyomorpha halys, (BMSB) is an exotic pest causing damage to fruits and vegetables. They also transmit diseases. Integrated pest management strategies are lacking for this invasive. For the first time, information is presented on the virulence of several entomopathogenic fungi against BMSB. Three Beauveria bassiana and two Metarhizium anispoliae isolates were bioassayed against adult BMSB. One B. bassiana isolate was the active ingredient in BotaniGard® which produced 85 and 100% mortality in 9 and 12 days post treatment, respectively. Experimental isolates ERL 1170 and ERL 1540 were also efficacious. Metarhizium anispoliae isolates produced lower mortalities than the B. bassiana isolates. Efforts are underway to produce antimicrobial peptides in yellow mealworms (Tenebrio molitor), which can be developed as more effective and safer animal feed additives. In this work, we expressed Bombyx mori (Bm) cecropin-A in mealworms by the infection of transformed entomopathogenic Beauveria bassiana ERL1170. The active domain of Bm cecropin A gene was tagged with a signal sequence of B. bassiana for extracellular secretion, and the fragment was inserted into ERL1170 by the restriction enzyme-mediated integration method. Transformant D-6 showed antibacterial activity against Bacillus subtilis and Listeria monocytogenes. Against T. molitor larvae, D-6 had similar mortality to wild-type, and D6-infected mealworm suspension showed strong antibacterial activity against the two bacteria, but not in the wild-type-infected mealworms, thereby increasing the value of mealworms as animal feed additives.

Conidia of Beauveria bassiana ARSEF-7060 produced in millet amended with plant oils such as sunflower, corn, or cotton seed oil, were exposed to 45 degrees C of wet heat for 90 min. Conidia from millet+corn oil medium had the highest thermotolerance (LT50 (median survival time): 45.7 min). The mycotized millet grains were coated with each of the same plant oils as a granular formulation and subjected to 50 degrees C of dry heat for 8 h. Corn oil coating (LT50: 8.68 h) was superior to sunflower- and cotton seed oil coatings, suggesting the feasibility of using corn oil to increase conidial thermotolerance.

Efforts are underway to develop more effective and safer animal feed additives. Entomopathogenic fungi can be considered practical expression platforms of functional genes within insects which have been used as animal feed additives. In this work, as a model, the enhanced green fluorescent protein (egfp) gene was expressed in yellow mealworms, Tenebrio molitor by highly infective Beauveria bassiana ERL1170. Among seven test isolates, ERL1170 treatment showed 57.1% and 98.3% mortality of mealworms 2 and 5 days after infection, respectively. The fungal transformation vector, pABeG containing the egfp gene, was inserted into the genomic DNA of ERL1170 using the restriction enzyme-mediated integration method. This resulted in the generation of the transformant, Bb-egfp#3, which showed the highest level of fluorescence. Bb-egfp#3-treated mealworms gradually turned dark brown, and in 7-days mealworm sections showed a strong fluorescence. This did not occur in the wild-type strain. This work suggests that further valuable proteins can be efficiently produced in this mealworm-based fungal expression platform, thereby increasing the value of mealworms in the animal feed additive industry.

Insect-killing (entomopathogenic) fungi have high potential for controlling agriculturally harmful pests. However, their pathogenicity is slow, and this is one reason for their poor acceptance as a fungal insecticide. The expression of bumblebee, Bombus ignitus, venom serine protease (VSP) by Beauveria bassiana (ERL1170) induced melanization of yellow spotted longicorn beetles (Psacothea hilaris) as an over-reactive immune response, and caused substantially earlier mortality in beet armyworm (Spodopetra exigua) larvae when compared to the wild type. No fungal outgrowth or sporulation was observed on the melanized insects, thus suggesting a self-restriction of the dispersal of the genetically modified fungus in the environment. The research is the first use of a multi-functional bumblebee VSP to significantly increase the speed of fungal pathogenicity, while minimizing the dispersal of the fungal transformant in the environment.

Beauveria bassiana SFB-205 supernatant can effectively control cotton aphid populations, which is closely associated with its chitinase activity. The present work extends to optimizing a culture medium to produce more efficacious supernatant in flask conditions, followed by scale-up in 7 L, 300 L and 1.2 KL fermentors with the parameter of chitinase. In flask conditions, a combination of soluble starch and yeast extract produced the greatest amount of chitinase (5.1 units/ml) and its supernatant had the highest aphicidal activity. An optimal quantitative combination of the two substrates, estimated by a response surface method, enabled the supernatant to have 15.7 units/ml of chitinase activity and 3.7 ml/l of median lethal concentration (LC50) of toxicity against cotton aphid adults in laboratory conditions. In the scale-up conditions, overall supernatant had 25-28 units/ml of chitinase activity. Decrease in pH and limitation of dissolved oxygen (DO) during cultures were significantly related to the yield of chitinase. These results suggest that the substrate-dependent chitinase production can be background information for optimizing a culture medium, and pH and DO are critical factors in maximizing the production in scale-up conditions.

Insect killing fungi have high potential in controlling agriculturally harmful pest, but their slow progress and high variation in killing insect are major impediments to successful industrialization. The present work describes the use of supernatant from the liquid culture of Beauveria bassiana SFB-205 to surmount this problem, particularly efficient production of thermotolerant chitinase, which is one of the major pathogenesis-related enzymes in the supernatant. The chitinase was precipitated using varying mineral precipitants and followed by lyophilization, which was compared with a salting out method using ammonium sulfate in effectiveness. Incorporating of the supernatant fraction of the Beauveria preparation with attagel at 0.5% (w/v) as a precipitant enabled this treatment to show the greatest chitinase-precipitation efficiency (93.4%), followed up with excellent insecticidal activity against cotton aphids when it was mixed with 0.01% (v/v) polyoxyethylene-(3)-isotridecyl ether (TDE-3) as a spreading agent in laboratory conditions. Consequently, lyophilized attagel-mediated precipitation pellet was superior to lyophilized salting out pellet in maintaining chitinase activity against a thermal stress at 50 degrees C. This finding provides that the attagel-mediated precipitation can be exploited to improve the thermotolerance of B. bassiana SFB-205 chitinase as a novel strategy for biopesticide production.
