Biological membranes are renowned for their intricate complexity, with the formation of membrane domains being pivotal to the successful execution of numerous cellular processes. However, due to their nanoscale characteristics, these domains are often understudied, as the experimental techniques required for quantitative investigation present significant challenges. In this study we employ spot-variation z-scan fluorescence correlation spectroscopy (svzFCS) tailored for artificial lipid vesicles of varying composition and combine this approach with high-resolution imaging. This method has been harnessed to examine the lipid-segregation behavior of distinct types of ceramide-1-phosphate (C1P), a crucial class of signaling molecules, within these membranes. Moreover, we provide a quantitative portrayal of the lipid membranes studied and the domains induced by C1P at both nano and microscales. Given the lack of definitive conclusions from the experimental data obtained, it was supplemented with comprehensive in silico studies—including the analysis of diffusion coefficient via molecular dynamics and domain populations via Monte Carlo simulations. This approach enhanced our insight into the dynamic behavior of these molecules within model lipid membranes, confirming that nano- and microdomains can co-exist in lipid vesicles. Experiments show that elastic constants of lipid bilayers vary greatly during the liquid-to-gel phase transition. This fact forms the cornerstone of the Heimburg–Jackson model of soliton propagation along membranes of axons, in which the action potential is accompanied by a traveling phase transition. However, the dispersion term, which is crucial for the existence of solitons, is added to the Heimburg–Jackson model ad hoc and set to fit experimental observations. In the present paper, we aim to consolidate this view with continuous membrane mechanics. Using literature data, we show that the compression modulus of a DPPC membrane is smaller by approximately an order of magnitude during phase transition. With a series expansion of the compression modulus, we write the action of a membrane and solve the corresponding wave equation analytically using an Exp-function method. We confirm that membrane solitons with speeds around 200 m/s are possible with amplitudes inversely proportional to their speed. We conclude that dispersion necessary for existence of solitons is directly related to a membrane's bending properties, offering a possible explanation for h. In this work, a theoretical model describing the interaction between a positively or negatively charged nanoparticle and neutral zwitterionic lipid bilayers is presented. It is shown that in the close vicinity of the positively charged nanoparticle, the zwitterionic lipid headgroups are less extended in the direction perpendicular to the membrane surface, while in the vicinity of the negatively charged nanoparticle, the headgroups are more extended. This result coincides with the calculated increase in the osmotic pressure between the zwitterionic lipid surface and positively charged nanoparticle and the decrease of osmotic pressure between the zwitterionic lipid surface and the negatively charged nanoparticle. Our theoretical predictions agree well with the experimentally determined fluidity of a lipid bilayer membrane in contact with positively or negatively charged nanoparticles. The prospective significance of the present work is mainly to contribute to better understanding of the interactions of charged nanoparticles with a zwitterionic lipid bilayer, which may be important in the efficient design of the lipid/nanoparticle nanostructures (like liposomes with encapsulated nanoparticles), which have diverse biomedical applications, including targeted therapy (drug delivery) and imaging of cancer cells. The lipid bilayer is a basic building block of biological membranes and can be pictured as a barrier separating two compartments filled with electrolyte solution. Artificial planar lipid bilayers are therefore commonly used as model systems to study the physical and electrical properties of the cell membranes in contact with electrolyte solution. Among them the glycerol-based polar phospholipids which have dipolar, but electrically neutral head groups, are most frequently used in formation of artificial lipid bilayers. In this work the electrical properties of the lipid layer composed of zwitterionic lipids with non-zero dipole moments are studied theoretically. In the model, the zwitterionic lipid bilayer is assumed to be in contact with aqueous solution of monovalent salt ions. The orientational ordering of water, resulting in spatial variation of permittivity, is explicitly taken into account. It is shown that due to saturation effect in orientational ordering of water dipoles the relative permittivity in the zwitterionic headgroup region is decreased, while the corresponding electric potential becomes strongly negative. Shapes of phospholipid vesicles that involve narrow neck(s) were studied theoretically. It is taken into account that phospholipid molecules are intrinsically anisotropic with respect to the membrane normal and that they exhibit quadrupolar orientational ordering according to the difference between the local principal membrane curvatures. Direct interactions between oriented molecules were considered within a linear approximation of the energy coupling with the deviatoric field. The equilibrium shapes of axisymmetric closed vesicles were studied by minimization of the free energy of the phospholipid bilayer membrane under relevant geometrical constraints. The variational problem was stated by a system of Euler-Lagrange differential equations that revealed a singularity in the derivative of the meridian curvature at points where the effect of the orientational ordering exactly counterbalances the effect of the isotropic bending. The system of Euler-Lagrange differential equations was solved numerically to yield consistently related equilibrium orientational distribution of the phospholipid molecules and vesicle shape. According to our estimation of the model constants the formation of the neck is promoted if direct interactions between the oriented molecules are taken into account. Abstract: Coupling between the area density of anisotropic membrane inclusions and local membrane curvature is considered theoretically for a simple case of nearly flat bilayer membrane with thin tubular membrane protrusions. Lateral phase separation, i.e. accumulation of membrane inclusions in tubular membrane protrusions was obtained for strongly anisotropic inclusions if the radius of tubular protrusions is small enough. In accordance with these theoretical predictions we observed persistence of long tubular membrane protrusions devoid of internal rod-like microtubular structure in cells. We suggest that the stability of the tubular membrane protrusions without the inner supporting rod-like cytoskeleton is a consequence of the accumulation of anisotropic membrane components in the bilayer membrane of these protrusions. Based on the presented theoretical and experimental results it is suggested that previously reported concentration of prominin rafts in thin tubular membrane protrusions may be caused by a curvature-induced accumulation of small prominin–lipid complexes (inclusions) in protrusions and their coalescence into larger rafts. Amphiphile-induced tubular budding of the erythrocyte membrane was studied using transmission electron microscopy. No chiral patterns of the intramembraneous particles were found, either on the cylindrical buds, or on the tubular nanoexovesicles. In agreement with these observations, the tubular budding may be explained by in-plane ordering of anisotropic membrane inclusions in the buds where the difference between the principal membrane curvatures is very large. The molecular details of the passive water flux across the hydrophobic membrane interior are still a matter of debate. One of the postulated mechanisms is the spontaneous, water-filled pore opening, which facilitates the hydrophilic connection between aqueous phases separated by the membrane. In the paper, we provide experimental evidence showing that the spontaneous lipid pore formation correlates with the membrane mechanics; hence, it depends on the composition of the lipid bilayer and the concentration of the osmotically active compound. Using liposomes as an experimental membrane model, osmotically induced water efflux was measured with the stopped-flow technique. Shapes of kinetic curves obtained at low osmotic pressure differences are interpreted in terms of two events: the lipid pore opening and water flow across the aqueous channel. The biological significance of the dependence of the lipid pore formation on the concentration difference of an osmotically active compound was illustrated by the demonstration that osmotically driven water flow can be accompanied by the dissipation of the pH gradient. The application of the Helfrich model to describe the probability of lipid pore opening was validated by demonstrating that the probability of pore opening correlates with the membrane bending rigidity. In this review paper, we theoretically explain the origin of electrostatic interactions between lipid bilayers and charged solid surfaces using a statistical mechanics approach, where the orientational degree of freedom of lipid head groups and the orientational ordering of the water dipoles are considered. Within the modified Langevin Poisson–Boltzmann model of an electric double layer, we derived an analytical expression for the osmotic pressure between the planar zwitterionic lipid bilayer and charged solid planar surface. We also show that the electrostatic interaction between the zwitterionic lipid head groups of the proximal leaflet and the negatively charged solid surface is accompanied with a more perpendicular average orientation of the lipid head-groups. We further highlight the important role of the surfaces' nanostructured topography in their interactions with biological material. As an example of nanostructured surfaces, we describe the synthesis of TiO2 nanotubular and octahedral surfaces by using the electrochemical anodization method and hydrothermal method, respectively. The physical and chemical properties of these nanostructured surfaces are described in order to elucidate the influence of the surface topography and other physical properties on the behavior of human cells adhered to TiO2 nanostructured surfaces. In the last part of the paper, we theoretically explain the interplay of elastic and adhesive contributions to the adsorption of lipid vesicles on the solid surfaces. We show the numerically predicted shapes of adhered lipid vesicles corresponding to the minimum of the membrane free energy to describe the influence of the vesicle size, bending modulus, and adhesion strength on the adhesion of lipid vesicles on solid charged surfaces. The magneto-mechanical actuation (MMA) of magnetic nanoparticles with a low-frequency alternating magnetic field (AMF) can be used to destroy cancer cells. So far, MMA was tested on different cells using different nanoparticles and different field characteristics, which makes comparisons and any generalizations about the results of MMA difficult. In this paper we propose the use of giant unilamellar vesicles (GUVs) as a simple model system to study the effect of MMA on a closed lipid bilayer membrane, i.e., a basic building block of any cell. The GUVs were exposed to barium-hexaferrite nanoplatelets (NPLs, ~50 nm wide and 3 nm thick) with unique magnetic properties dominated by a permanent magnetic moment that is perpendicular to the platelet, at different concentrations (1–50 µg/mL) and pH values (4.2–7.4) of the aqueous suspension. The GUVs were observed with an optical microscope while being exposed to a uniaxial AMF (3–100 Hz, 2.2–10.6 mT). When the NPLs were electrostatically attached to the GUV membranes, the MMA induced cyclic fluctuations of the GUVs' shape corresponding to the AMF frequency at the low NPL concentration (1 µm/mL), whereas the GUVs were bursting at the higher concentration (10 µg/mL). Theoretical considerations suggested that the bursting of the GUVs is a consequence of the local action of an assembly of several NPLs, rather than a collective effect of all the absorbed NPLs. The lipid bilayer composed of negatively charged lipid 1-palmitoyl-3-oleoyl- sn -glycero-3-phosphatidylserine (POPS) in contact with an aqueous solution of monovalent salt ions was studied theoretically by using the mean-field modified Langevin–Poisson–Boltzmann (MLPB) model. The MLPB results were tested by using molecular dynamic (MD) simulations. In the MLPB model the charge distribution of POPS head groups is theoretically described by the negatively charged surface which accounts for negatively charged phosphate groups, while the positively charged amino groups and negatively charged carboxylate groups are assumed to be fixed on the rod-like structures with rotational degree of freedom. The spatial variation of relative permittivity, which is not considered in the well-known Gouy–Chapman (GC) model or in MD simulations, is thoroughly derived within a strict statistical mechanical approach. Therefore, the spatial dependence and magnitude of electric potential within the lipid head group region and its close vicinity are considerably different in the MLPB model from the GC model. The influence of the bulk salt concentration and temperature on the number density profiles of counter-ions and co-ions in the lipid head group region and aqueous solution along with the probability density function for the lipid head group orientation angle was compared and found to be in qualitative agreement in the MLPB and MD models. Lipid bilayer is the main constitutive element of biological membrane, which confines intracellular space. The mechanical properties of biological membranes may be characterized by various parameters including membrane stiffness or membrane bending rigidity, which can be measured using flicker noise spectroscopy. The flicker noise spectroscopy exploits the spontaneous thermal undulations of the membrane. The method is based on the quantitative analysis of a series of microscopic images captured during thermal membrane fluctuations. Thus, measured bending rigidity coefficient depends on the image quality as well as the selection of computational tools for image processing and mathematical model used. In this work scanning and spinning disc confocal microscopies were used to visualize fluctuating membranes of giant unilamellar vesicles. The bending rigidity coefficient was calculated for different acquisition modes, using different fluorescent probes and different image processing methods. It was shown that both imaging approaches gave similar bending coefficient values regardless of acquisition time. Using the developed methodology the effect of fluorescent probe type and aqueous phase composition on the value of the membrane bending rigidity coefficient was measured. Specifically it was found that the bending rigidity coefficient of DOPC bilayer in water is smaller than that determined for POPC membrane. It has been found that the POPC and DOPC bending rigidities coefficient in sucrose solution was lower than that in water. The interaction of the divalent calcium ions with the zwitterionic lipid membranes was studied by measuring the lipid order parameter which is inversely proportional to the membrane fluidity. Small unilamellar lipid vesicles were prepared from 1-palmitoyl-2-oleoyl-sn-glycero-3-phosphocholine and then treated with different concentrations of divalent calcium ions. An increase in the order parameter and decrease in the fluidity of the liposomal membranes were observed after treatment with the calcium ions. The presence of positively charged iron oxide nanoparticles in the suspension of liposomes negligibly changed the results. The results of experiments were discussed theoretically within modified Langevin- Poisson-Boltzmann (MLPB) model leading to the conclusion that the membrane fluidity and ordering of the membrane lipids are primarily altered by the accumulation of calcium ions in the region of negatively charged phosphate groups within the head groups of the membrane lipids. Mechanical properties of a lipid bilayer are parameters determined mainly for giant unilamellar vesicles (GUVs). It is not clear if values obtained on the GUV model can be directly translated to submicron large unilamellar vesicles (LUVs). This ambiguity is a major obstacle in exploring the effect of lipid bilayer mechanics on membrane associated processes and effectiveness of liposome-based targeted drug delivery systems. In presented work extrusion, which is a common method to prepare LUVs, was used to study liposomes preparation and stability upon exposure to mechanical stress. The effect of parameters of the extrusion process (temperature, membrane pore size, extrusion force and volumetric flux) on the properties of liposome suspension (average liposome size, polydispersity index and lipid recovery ratio) was determined for model liposomes composed of DPPC lipid. The state of the DPPC lipid bilayer depends on temperature, therefore, the effect of lipid bilayer mechanics on the extrusion process can be quantitated without altering membrane composition. The extrusion process was carried out with the automated extruder delivering quantitative data on the extrusion force and volumetric flux. Obtained results have been interpreted in terms of mechanical properties of the lipid bilayer. Determined mechanical properties of the lipid bilayer and its dependence on temperature are in good agreement with the literature results determined for GUVs. This shows that mechanical properties of the lipid bilayer does not depend on the liposome size in the range from 100 nm to hundreds of microns.
