After vitrectomy, the ideal vitreous substitute should be implanted to maintain the normal function of the eye. However, the existing materials (such as silicone oil, air, perfluorocarbons, etc.) still have some shortcomings and cannot fully meet the clinical needs. In this study, thiolated hyaluronic acid (SH-HA) was prepared based on hyaluronic acid. The SH-HA hydrogel was formed by a simple transformation of the sulfhydryl group to the disulfide bond, which had high transparency, controllable swelling property, suitable mechanical strength, excellent biocompatibility and similar physical and chemical properties to natural vitreous. SH-HA hydrogel was filled into the eyes of experimental rabbits to replace their own vitreous after vitrectomy. During the 90 days follow-up period, SH-HA hydrogel showed excellent intraocular compatibility, maintained normal intraocular pressure (IOP), and no cataract, endophthalmitis, retinal detachment and other complications were observed. In general, SH-HA hydrogel has great potential as a vitreous substitute.

Herein, the thermosensitive hydroxypropyl chitin (HPCT) hydrogel was prepared and the chemical structures, microstructures, rheological properties and degradation in vitro were investigated. The HPCT hydrogel possessed satisfactory biocompatibility in mouse fibroblast cells and Sprague Dawley rats. On the other hand, N-acetylglucosamine (NAG) and carboxymethyl chitosan (CMCS) provided favorable capacity for promoting cell proliferation, delaying cell apoptosis, and facilitating the insulin secretion of rat pancreatic beta cells (RIN-m5F) in three-dimensional culture. Most importantly, the effects of HPCT/NAG and HPCT/CMCS thermosensitive hydrogels as RIN-m5F cells carriers were evaluated via injection into different areas of diabetic rats. Our results demonstrated that HPCT/NAG and HPCT/CMCS hydrogels loaded RIN-m5F cells could keep cells survival, maintain insulin secretion and reduce blood glucose for one week. Overall, the functional thermosensitive hydrogels based on HPCT were effective cell carriers for RIN-m5F cells and might provide novel strategy for the treatment of diabetes via cell engineering.

Novel polymer-drug conjugates (CNC) were prepared from carboxymethyl chitosan (CMCS) and norcantharidin (NCTD) via amidation reaction and characterized by FTIR and 1H NMR spectroscopy. The aim of this study was to elucidate the antitumor efficacy of CNC on gastric cancer and the possible underlying mechanisms. The CNC conjugates possessed significant inhibitory effects on the proliferation of SGC-7901 cells and suppressed the migration as well as tube formation of HUVECs. Besides, Hoechst 33258 staining and Annexin V-FITC/PI detection suggested that the conjugates were more effective in triggering apoptosis of SGC-7901 cells compared with free NCTD. Moreover, CNC remarkably reduced systemic toxicity and enhanced the antitumor efficacy in vivo with a tumor suppression rate of 59.57% against SGC-7901 gastric tumor in BALB/c nude mice. Further investigation about the underlying mechanisms indicated that CNC could upregulate expressions of TNF-α and Bax, and downregulate expressions of VEGF, Bcl-2, MMP-2 and MMP-9, thereby inhibiting tumor metastasis and inducing apoptosis in vivo. Overall, our results demonstrated that CNC might be a promising and feasible polymer therapeutics for gastrointestinal tumor therapy.

Chitosan as a natural bioactive biopolymer has been commonly employed in guidance conduit for repairing peripheral nerve injury, due to its excellent properties of low toxicity, antibacterial properties, high biocompatibility and biodegradability. In this study, chitin and CM-chitosan were prepared from pharmaceutical grade chitosan. Moreover, a novel composite chitosan-based nerve graft comprising microporous chitin-based conduit and internal CM-chitosan fiber was constructed and applied to bridge sciatic nerve across a 10-mm defect in SD rats. The chitin/CM-chitosan artificial nerve graft could promote the proliferation of rat Schwann cells (RSC96) with good cell biocompatibility. After implantation, the artificial nerve graft showed slow degradation. No apparent toxicity was observed, and tissue inflammation was very slight after implantation, indicating favorable bio-safety of the nerve graft. Furthermore, the chitin/CM-chitosan artificial nerve graft could effectively promote restoration of damaged neurons with similar effect compared to the autograft. In conclusion, the composite biodegradable chitin/CM-chitosan nerve grafts possessed favorable biocompatibility and good potential in repairing peripheral nervous injury.

An extracellular chondroitinase ABC (ChSase ABC) produced by Sphingomonas paucimobilis was purified to homogeneity through ammonium sulfate precipitation, DEAE-Sepharose Fast Flow and Sephadex G-100 chromatography. The molecular weight was 82.3 kDa. It showed specific lyase activity toward chondroitin sulfate A (CS-A), CS-B, CS-C and hyaluronan (HA). Using CS-A as substrate, the specific activity was 98.04 U/mg, the maximal reaction rate (Vmax) and Michaelis-Menten constant (Km) were 0.49 μmol/min/ml and 0.79 mg/ml, respectively. Highest activity was obtained at pH 6.5 and 40 °C, and Hg2+ could strongly inhibit the enzyme activity. Mass spectrometry analysis indicated CS-A was degraded to unsaturated disaccharides by ChSase ABC. In vitro cytotoxic tests showed that CS-A oligosaccharide at the concentration of 50 and 100 μg/ml could promote the proliferation of normal H9c2 myocardial cells, decrease the damage induced by isoproterenol (ISO) and accelerate the recovery of cells injured by ISO. These findings suggested that ChSase ABC from Sphingomonas paucimobilis could be a promising tool for the structural analysis and bioactive oligosaccharide preparation of glucosaminoglycans.

Analgesis and wound healing effect of chitosan and carboxymethyl chitosan on scalded rats were investigated. A II degree scald model was established in rats, which was subsequently treated with chitosan and carboxymethyl chitosan solution, respectively. The concentration of bradykinin and 5-hydroxytryptophan was detected by assaying enzyme-linked immunosorbent. Healing condition was observed and pathological sections were made to determine the healing effect of chitosan and carboxymethyl chitosan. Results showed that the concentration of bradykinin and 5-hydroxytryptophan peaked at the third hour post-wound in all groups, while the concentration of hydroxyproline peaked at the seventh day post-wound in both chitosan and carboxymethyl chitosan group. The concentration of bradykinin and 5-hydroxytryptophan of carboxymethyl chitosan group was significantly lower than that of control (P 0.05). These findings indicated that carboxymethyl chitosan reduced the concentration of algogenic substances, resulting in analgesia. During the whole recovery process, the hydroxyproline concentration in chitosan and carboxymethyl chitosan group on day 3 and 7 was significantly higher than that of control (P < 0.01); however the significance of such a highness decreased on day 14 (P < 0.05). These findings indicated that chitosan and carboxymethyl chitosan accelerated tissue repair. Meanwhile, chitosan performed better in healing than carboxymethyl chitosan in both decrustation and healing time. In conclusion, carboxymethyl chitosan showed significant analgesis and wound-healing promotion effect, but chitosan only showed wound-healing promotion effect.

Chitosanase is able to specifically cleave β-1,4-glycosidic bond linkages in chitosan to produce a chito-oligomer product, which has found a variety of applications in many areas, including functional food and cancer therapy. Although several structures for chitosanase have been determined, the substrate-binding mechanism for this enzyme has not been fully elucidated because of the lack of a high-resolution structure of the chitosanase–substrate complex. In the present study we show the crystal structure of a novel chitosanase OU01 from Microbacterium sp. in complex with its substrate hexa-glucosamine (GlcN)6, which belongs to the GH46 (glycoside hydrolyase 46) family in the Carbohydrate Active Enzymes database (http://www.cazy.org/). This structure allows precise determination of the substrate-binding mechanism for the first time. The chitosanase–(GlcN)6 complex structure demonstrates that, from the −2 to +1 position of the (GlcN)6 substrate, the pyranose rings form extensive interactions with the chitosanase-binding cleft. Several residues (Ser27, Tyr37, Arg45, Thr58, Asp60, His203 and Asp235) in the binding cleft are found to form important interactions required to bind the substrate. Site-directed mutagenesis of these residues showed that mutations of Y37F and H203A abolish catalytic activity. In contrast, the mutations T58A and D235A only lead to a moderate loss of catalytic activity, whereas the S27A mutation retains ~80% of the enzymatic activity. In combination with previous mutagenesis studies, these results suggest that the −2, −1 and +1 subsites play a dominant role in substrate binding and catalysis. DSF (differential scanning fluorimetry) assays confirmed that these mutations had no significant effect on protein stability. Taken together, we present the first mechanistic interpretation for the substrate (GlcN)6 binding to chitosanase, which is critical for the design of novel chitosanase used for biomass conversion.

Introduction The prevalence of thromboembolic disease, one of the top 3 leading causes of mortality worldwide, is being reported continually. More effective and safer antithrombotic drugs may overcome the underlying problems in antithrombotic therapy. In the present work, antithrombotic effects of UFEIII, a newly purified fibrinolytic protease from Urechis unicinctus were evaluated. Materials and methods UFEIII was purified from the marine invertebrate, Urechis unicinctus , using anion exchange and gel filtration chromatography. Molecular weight, fibrinolytic activity and fibrinogenolysis pattern of UFEIII were determined. Furthermore, antithrombotic effects of UFEIII in vivo were investigated through electrical induced carotid arterial thrombosis in rats, FeCl 3 induced carotid arterial thrombus model in rabbits and stasis induced vena caval thrombus model in rats. Results SDS-PAGE of the purified enzyme showed a single polypeptide chain with molecular weight of 20.8kDa. In fibrin plate assays, UFEIII could not only directly degrade fibrin but also activate plasminogen. The fibrinogenolysis pattern of UFEIII was Aα-chains>Bβ-chains>γ-chain. Moreover, UFEIII could effectively prolong the time to occlusion in electrical induced carotid arterial thrombosis. Besides, both in rabbits and rats, the administration of UFEIII not only prolonged the activated partial thromboplastin time (APTT) and thrombin time (TT) also decreased the fibrinogen (FIB) content. Further, the thrombus lysis was observed after administration of UFEIII both in rabbits and rats. Conclusion UFEIII can possibly be a new potential source of fibrinolytic agent.

A non-hemorrhagic, chymotrypsin-like serine protease, UFEII, was purified from the marine echiuroid worm, Urechis unicinctus, after a combination of chromatography steps. UFEII was monomeric, with an apparent molecular weight of 26.7 kDa via SDS-PAGE. The isoelectric point of UFEII was 4.03, and the maximum activity of the enzyme was observed at 50 °C and pH 8.0. According to fibrin plate assays, UFEII could not only directly degrade fibrin and fibrinogen but also activate plasminogen. Further, UFEII preferentially hydrolyzed the fibrinogen γ-chain, followed by the Bβ-chains and Aα-chains. Moreover, ufeII, full length of the gene encoding UFEII, was obtained by RT-PCR, degenerated PCR, and nested PCR. The ufeII was determined to be a 906-bp cDNA containing an open reading frame of 795 bp encoding a putative protein of 264 amino acids with a predicted molecular weight of 27.03 kDa. Besides, UFEII exhibited no hemorrhagic effect. Overall, U. unicinctus may represent a potential source of new therapeutic agents in thrombolytic therapy.

A new serine protease with fibrinolytic activity from a marine invertebrate, Urechis unicinctus, was purified to electrophoretic homogeneity using column chromatography. SDS-PAGE of the purified enzyme showed a single polypeptide chain with MW ~20.8 kDa. Its N-terminal sequence was IIGGSQAAITSY. The purified enzyme, UFEIII, was stable at pH 6-10 below 60 °C with an optimum pH of 8.5 at approx. 55 °C. The enzyme activity was significantly inhibited by PMSF and SBTI suggesting that it was a serine protease. In fibrin plate assays, UFEIII was contained 1.46 × 10(3 )U (urokinase units) mg(-1) total fibrinolytic activity, which consisted of 692 U mg(-1) direct fibrinolytic activity and 769 U mg(-1) plasminogen-activator activity. Km and Vmax values for azocasein were 1 mg ml(-1) and 43 μg min(-1) ml(-1), respectively.

Carboxymethyl chitosan (CM-chitosan) is one of water-soluble derivatives of chitosan. Numerous studies have been focused on its applications as pharmaceutical excipient, bioactive reagent and nontoxic drug carrier. Like other polysaccharides, CM-chitosan is inhomogenous in molecular weight. Originations and preparation procedures considerably influence its molecular weight and molecular weight distributions. Understanding the molecular weight related biological behaviour of this inhomogenous glycopolymer in vivo were crucial for the quality control and clinical applications of chitosan and chitosan based medical devices. In this study, we investigated the effects of molecular weights on the absorption, distribution, degradation and urinary excretion of the fluorescein isothiocyanate-labeled CM-chitosan in rats. The results indicated that molecular weight significantly influenced the uptake of CM-chitosan from the lumen of abdomen and blood vessels to peripheral tissues, the distribution of this chemical and urinary excretion after intraperitoneal administration. These findings provided an important reference for the clinical applications of this versatile derivative of chitosan as postsurgical and other biomedical materials and important clues for the exploitation of CM-chitosan based drug targeting and delivery systems.

The aim of the present study was to evaluate the anticancer effects of chitooligosaccharides (COS) and N-acetyl-D-glucosamine (NAG), as well as to investigate the possible mechanisms involved. MTT assay and flow cytometry were used to evaluate the effect of various concentrations of COS and NAG on the proliferation and differentiation of peripheral blood mononuclear cells (PBMCs). In addition, sarcoma 180 cells were transplanted into mice to establish a tumor model. COS and NAG were administered by gavage of various doses. The tumor inhibition rate, thymus and spleen indexes, natural killer (NK) cell activity, and interleukin-2 (IL-2) and interferon-γ (IFN-γ) serum levels were detected. Vascular endothelial growth factor (VEGF) expression levels, an important marker of angiogenesis, were also detected. As shown by immunohistochemistry, VEGF mRNA expression was decreased following treatment with COS and NAG, indicating that COS and NAG have an inhibitory effect on the expression of VEGF. The results from this study indicate that COS administered at a dose of 100 mg/kg and NAG at a dose of 300 mg/kg or 500 mg/kg can not only promote the differentiation of PBMCs and the secretion of IL-2 and IFN-γ, but can also inhibit the expression of VEGF mRNA in sarcoma 180 tumors. Our results show that the antitumor and immunoregulatory effects of COS and NAG are dose-dependent. Furthermore, the antitumor effect is achieved by the improvement of immunoregulation indirectly.

The properties of modulating liver l - γ -glutamyl- l -cysteinylglycine (GSH) homeostasis by thiazolidine derivative N-acetyl-glucosamine-thiazolidine-4(R)-carboxylic acid (GlcNAcCys) and the underlying mechanisms were investigated in l -buthionine-[S,R]-sulfoximine (BSO)-induced mice liver GSH depletion model. The data show that BSO (6 mmol/kg body weight; intraperitoneally) significantly decreased liver total sulfhydryl and GSH concentrations when compared with control. When mice were treated with different doses of GlcNAcCys (200, 400, 900 mg/kg body weight; intraperitoneally, respectively), total sulfhydryl and GSH concentrations were significantly increased when measured 6 hours after treatment. The activities of GSH-associated enzymes were also measured. Liver glutathione S-transferase (GST) activities were significantly decreased by BSO compared with the control, and GlcNAcCys significantly increased GST activity. Moreover, reverse-transcriptase polymerase chain reaction data indicated that GlcNAcCys could significantly induce glutamylcysteine ligase catalytic subunit c mRNA transcription. The mRNA levels of transcription factors c-jun and c-fos were increased by BSO administration but were decreased back to normal after the administration of GlcNAcCys. In a conclusion, GlcNAcCys can modulate liver GSH homeostasis, which may be related to its ability to induce glutamylcysteine ligase catalytic subunit transcription. GlcNAcCys has potential hepatoprotective properties by increasing GSH content, increasing GST activity.

Four isonitrogenous (gross protein content 30%) and isoenergetic (gross energy content 15 kJ/g) diets were formulated to investigate the effects of inclusion of plant proteins on the hybrid tilapia (Oreochromis niloticus×Oreochromis aureus). The plant proteins tested were soybean meal (SBM), rapeseed meal (RSM), cottonseed meal (CSM), and peanut meal (PNM). In each diet, 30% of the protein was supplied by fish meal, and the rest of protein was supplied by the plant protein tested. Each diet was randomly fed to three groups of hybrid tilapia for 8 wk in a recirculating freshwater system. The results showed that the best performance in terms of final weight, specific growth rate, feed efficiency ratio, and protein efficiency ratio were shown in diets SBM and CSM. Four parameters were significantly affected by dietary RSM (P 0.05). The highest activities of glutamate-pyruvate transaminase, glutamate-oxaloacetate transaminase, and superoxide dismutase in the liver were observed in group SBM and the poorest in group RSM. The results show that 39% CSM may be included in the practical diets without inhibiting fish performance. Although the inclusion of 50% RSM resulted in significantly lower growth performance, it produced histopathological damage in the liver.

Carboxymethyl chitosan (CM-chitosan), which is a water-soluble derivative of chitosan, has attracted much attention as a new biomedical material. The safety study of this material was persuasive for its potential application. The present study was conducted to assess the tissue distribution, pharmacokinetics, biodegradation mechanism, and excretion of CM-chitosan in rats. After the rats were intraperitoneally injected at the dose of 50 mg/kg, the fluorescein isothiocyanate (FITC)-labeled CM-chitosan was absorbed rapidly and distributed to different organs, including liver, spleen, and kidney. The highest level of CM-chitosan was found in liver. It was at the level of 1.6 +/- 0.6 mg/liver and made up approximately 10-22% of the total injected FTC-CM-chitosan. Urinary excretion was the predominant way of excretion of FITC-labeled CM-chitosan, and 85% of the dose was excreted in urine over the period of 11 days. The molecular weights of body distributed FTC-CM-chitosan and urinary excreted FTC-CM-chitosan were analyzed by gel chromatography. The results indicated that the FTC-CM-chitosan was degraded in abdominal dropsy. The absorbed CM-chitosan forms were found with a relatively high molecular weight (approximately 300 kDa), whereas the molecular weight of the urinary excreted FTC-CM-chitosan was less than 45 kDa. In vitro research revealed that the CM-Chi was also degradable in plasma and homogenate of liver. The CM-chitosan with a molecular weight of approximately 800k was thoroughly degraded to a small molecule after it was incubated in homogenate of liver at 37 degrees C for 24 h. The results suggested that the liver plays a central role in biodegradation of CM-chitosan. The excellent biodegradability of CM-chitosan could potentially contribute to the clinical applications. The results also provide important clues for further modification of CM-chitosan as the postsurgical and other biomedical materials.

Carboxymethyl-chitosan and carboxymethyl-chitin were prepared with the methods developed in our laboratory. The DS (degree of substitution) and DD (degree of deacetylation) of the carboxymethyl-chitosan were 103.14% and 97.18% respectively, while the DS of the carboxymethyl-chitin was 96.37%. Their effects on human fibroblasts, intradermal irritation test, in vitro and vivo degradability, and biocompatibility were evaluated. The results indicate that the polysaccharides at low concentrations can facilitate the growth of human fibroblasts and the carboxymethyl-chitosan at 100 μg mL−1 is the most effective. The polysaccharides at higher concentrations, however, inhibit the growth of fibroblasts. The PII (Primary Irritation Index) values of CM-chitosan and CM-chitin are both 0.0, which shows that they have no irritation reaction. Both of the polysaccharides show good degradability and biocompatibility. Carboxymethyl-chitin degrades faster in vitro than carboxymethyl-chitosan. The latter, however, has no inflammatory reaction after being implanted in vivo for 7 d and shows better biocompatibility. This study may provide a scientific basis for the use of carboxymethyl-chitosan and carboxymethyl-chitin as biomaterials.

Aqueous purified extract (PE) was isolated from the crude extract (CE) of the marine mollusk, Ruditapes philippinarum. Two water-soluble polysaccharides, PEF1 and PEF2, were purified from PE. The chemical structures were determined using FTIR, GC, HPLC and 13C NMR. The results indicated that PEF1 and PEF2 were homoglucan–protein complexes with a protein content of 26.0 and 8.2%, and their average molecular weights were about 2.0 × 106 and 5.0 × 103, respectively. The glucan moiety of PEF1 was mainly a (1 → 6)-branched (1 → 4)-α-d-glucan, while that of PEF2 contained (1 → 4)-α-d-glucan and (1 → 6)-β-d-glucan. PE fractions contained large quantities of glutamic acid and aspartic acid, but no cysteine. The antitumor activities were tested both in vitro and in vivo. PE showed significantly the highest tumoricidal activity against human hepatoma SMMC-7721 cells, and PEF1 had better cytotoxicity than PEF2. There was no observed cytotoxicity in human hepatocyte HL-7702 cells within the experimental concentration range for all PE fractions. PE showed antitumor activity against solid tumor Sarcoma 180 in a dose-dependent manner. It also demonstrated stimulating effect on murine lymphocyte proliferation induced by concanavalin A. The results demonstrated that the presence of bound protein, compositional glucan and moderate molecular mass would be helpful to the enhancement of antitumor activities. This study suggested that the polysaccharides isolated from the marine mollusk had a potential application as natural antitumor and immunomodulator agents.

Using d-glucosamine hydrochloride (GlcNH2·HCl) as starting material, a new amino acid sugar conjugate, arginine–glucosamine (Arg–GlcNH2), was synthesized and characterized by infrared spectroscopy, 13C NMR and element analysis. Its cytotoxicity in vitro was evaluated by MTT assay. The inhibition ratio against human hepatoma cell SMMC-7721 was higher than that of GlcNH2·HCl. This effect was accompanied by a marked increase in the proportion of S cells as analyzed by flow cytometry. In addition, SMMC-7721 cells treated with Arg–GlcNH2 resulted in the induction of apoptosis as assayed qualitatively by agarose gel electrophoresis. The manner of Arg–GlcNH2 cytocidal activity was concluded to be apoptosis.

A novel potent protease, Urechis unicinctus fibrinolytic enzyme (UFE), was first discovered by our laboratory. In this study, we further investigated the enzymatic properties and dynamic parameters of UFE. As a low molecular weight protein, UFE appeared to be very stable to heat and pH. When the temperature was50 degrees C, the remnant enzyme activity remained almost unchanged, but when the temperature was raised to 60 degrees C the remnant enzyme activity began to decrease rapidly. UFE was quite stable in a pH range of 3.0-12.0, especially at slightly alkaline pH values. Mn(2+), Cu(2+), and Fe(2+) ions were activators of UFE, whereas Fe(3+) and Ag(+) ions were inhibitors. Fe(2+) ion along with Fe(3+) ion might regulate UFE activity in vivo. The optimum pH and temperature of UFE were about 8.0 and 50 degrees C, respectively. When using casein as substrate and a substrate concentration0.1% casein (w/v), the reaction velocity was increased with substrate concentration. Also when using casein as substrate, the determined K(m) and V(max) of UFE were 0.5298 mg/mL and 3.0845 mol of L-tyrosine equivalent, respectively. Our systematic research results are significant when UFE is applied for medical and industrial purposes.

Background A detailed knowledge about the degradation mechanism of chitosanase hydrolysis is critical for the design of novel enzymes to produce well-defined chito-oligosaccharide products. Methods Through the combination of structural and biochemical analysis, we present new findings that provide novel insights into the degradation mechanism of chitosanase OU01. Results We have determined the crystal structure of Asp 43 /Ala mutant of OU01, and have trapped the hydrolyzed product of the reaction. This structure reveals the role of the general acid (Glu 25 ) in catalysis. Two structural features about the mechanisms of the non-processive chitosanases are described for the first time. 1). Structural comparison reveals that the enzyme goes through an open–closed–open conformational transition upon substrate binding and product release; 2). polar residues constitute the substrate binding cleft. Additional site important for polymeric substrate recognition is identified and a three-step polymeric substrate recognition mechanism is proposed. Conclusions Detailed substrate recognition mechanism is described for non-processive chitosanase for the first time. General significance These findings provide new structural insights into the understanding of overall hydrolysis mechanism for non-processive chitosanase, and also will facilitate the design of new enzymes used for industrial purpose.

Carboxymethyl chitosan (CM-chitosan), a derivative of chitosan, was extensively studied in the biomedical materials field for its beneficial biological properties of hemostasis and stimulation of healing. However, studies examining the safety of CM-chitosan in the blood system are lacking. In this study CM-chitosan was implanted into the abdominal cavity of rats to determine blood indexes at different times and to evaluate the effects of CM-chitosan on the blood system of rats. Coagulation function was reflected by thrombin time (TT), prothrombin time (PT), activated partial thromboplatin time (APTT), fibrinogen (FIB) and platelet factor 4 (PF4) indexes; anti-coagulation performance was assessed by the index of antithrombinIII (ATIII); fibrinolytic function was reflected by plasminogen (PLG) and fibrin degradation product (FDP) indexes; and blood viscosity (BV) and plasma viscosity (PV) indexes reflected hemorheology. Results showed that CM-chitosan has no significant effects on the blood system of rats, and provides experimental basis for CM-chitosan to be applied in the field of biomedical materials.

A novel chitosan-based membrane that made of hydroxyethyl chitosan, gelatin and chondroitin sulfate was used as a carrier of corneal endothelial cells. The characteristics of the blend membrane including transparency, equilibrium water content, ion and glucose permeability were determined. The results showed that the optical transparency of the membrane was as good as the natural human cornea. The water content of this scaffold was 81.32% which was remarkably close to the native cornea. The membrane had a good ion permeability and its glucose permeability was even higher than natural human cornea. The cultured rabbit corneal endothelial cells formed a monolayer on the membrane. The results demonstrated that the membrane was suitable for corneal endothelial cells to attach and grow on it. In addition, the membranes in vivo could be degraded steadily with less inflammation and showed a good histocompatibility. These results demonstrated that the hydroxyethyl chitosan-chondroitin sulfate-gelatin blend membrane can potentially be used as a carrier for corneal endothelial cell transplantation.

The growth inhibitory effects of D-glucosamine hydrochloride (GlcNH(2).HCl), D-glucosamine (GlcNH(2)) and N-acetyl glucosamine (NAG) on human hepatoma SMMC-7721 cells in vitro were investigated. The results showed that GlcNH(2).HCl and GlcNH(2) resulted in a concentration-dependent reduction in hepatoma cell growth as measured by MTT (3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide) assay. This effect was accompanied by a marked increase in the proportion of S cells as analyzed by flow cytometry. In addition, human hepatoma SMMC-7721 cells treated with GlcNH(2).HCl resulted in the induction of apoptosis as assayed qualitatively by agarose gel electrophoresis. NAG could not inhibit the proliferation of SMMC-7721 cells. GlcNH(2).HCl exhibited antitumor activity against Sarcoma 180 in Kunming mice at dosage of 125-500 mg/kg, dose of 250 mg/kg being the best. GlcNH(2).HCl at dose of 250 mg/kg could enhance significantly the thymus index, and spleen index and could promote T lymphocyte proliferation induced by ConA. The antitumor effect of GlcNH(2).HCl is probably host-mediated and cytocidal.

Water-soluble hydroxypropyl chitosan (HPCS) derivatives with different degrees of substitution (DS) and weight-average molecular weight (Mw) were synthesized from chitosan and propylene epoxide under basic conditions. Their structure was characterized by IR spectroscopy, NMR spectroscopy, and elemental analysis, which showed that both the OH groups at C-6 and C-3 and the NH2 group of chitosan were alkylated. The DS value of HPCS ranged from 1.5 to 3.1 and the Mw was between 2.1x10(4) and 9.2x10(4). In vitro antimicrobial activities of the HPCS derivatives were evaluated by the Kirby-Bauer disc diffusion method and the macrotube dilution broth method. The HPCS derivatives exhibited no inhibitory effect on two bacterial strains (Escherichia coli and Staphylococcus aureus); however, some inhibitory effect was found against four of the six pathogenic fruit fungi investigated. Some derivatives (HPCS1, HPCS2, HPCS3, HPCS3-1, and HPCS4) were effective against C. diplodiella and F. oxysporum. HPCS3-1 is the most effective one with MIC values of 5.0, 0.31, 0.31, and 0.16mg/mL against A. mali, C. diplodiella, F. oxysporum, and P. piricola, respectively. Antifungal effects were also observed for HPCS2 and HPCS3-1 against A. mali, as well as HPCS3 and HPCS3-1 against P. piricola. The results suggest that relatively lower DS and higher Mw value enhances the antifungal activity of HPCS derivatives.
