Dental caries is a widespread bacterial infectious disease that imposes a significant public health burden globally. The primary culprits in caries development are cariogenic bacteria, notably Streptococcus mutans ( S. mutans ), due to their robust biofilm-forming capabilities. To address this issue, a series of cationic pyridinium-substituted photosensitizers with aggregation-induced emission have been designed. All of these aggregation-induced emission luminogens (AIEgens) exhibit outstanding microbial visualization and photodynamic killing of S. mutans , thanks to their luminous fluorescence and efficient singlet oxygen generation ability. Notably, one of the membrane-anchored AIEgens (TDTPY) can inactivate planktic S. mutans and its biofilm without causing significant cytotoxicity. Importantly, application of TDTPY-mediated photodynamic treatment on in vivo rodent models has yielded commendable imaging results and effectively slowed down caries progression with assured biosafety. Unlike traditional single-mode anticaries materials, AIEgens integrate the dual functions of detecting and removing S. mutans and are expected to build a new caries management diagnosis and treatment platform. To the best of our knowledge, this is also the first report on the use of AIEgens for anticaries studies both in vitro and in vivo.

Evidence suggests that short amyloid-forming peptides derived from bacterial proteomes have functional roles; however, the reported activities are diverse and the underlying mechanisms remain unclear. In this study, we simulated short amyloid-forming peptides from the amyloid-forming truncated protein C123 of Streptococcus mutans (S. mutans), studied their biological functions in microbial proliferation and biofilm formation, and further investigated the underlying mechanism. Fourteen hexapeptides were simulated, 13 of which were successfully synthesized. We found that the amyloid-forming hexapeptides (AFhPs) displayed efficient broad-spectrum antibiofilm activity against the Gram-positive bacteria S. mutans, Streptococcus sanguis and Staphylococcus aureus, Gram-negative bacteria Escherichia coli and fungus Candida albicans, by aggregating into rigid amyloid fibres agglutinating microbes, whereas the non-amyloid-forming hexapeptides (non-AFhPs) did not. The AFhPs did not kill microbes and showed little or no cytotoxicity. Furthermore, a set of AFhPs displayed broad-spectrum antibiofilm activity, regardless of its source. The microbial cell wall carbohydrates, peptidoglycan (PGN), lipoteichoic acid (LTA), glucan and zymosan A, mediated AFhP binding and triggered significant AFhP fibrillation. Although amyloid fibres agglutinated lipid membrane model - large unilamellar vesicles (LUVs) - and LUVs facilitated AFhP fibrillation, the roles of lipid membranes in AFhP antibiofilm activities remain to be elucidated. Background: Streptococcus mutans (S. mutans) is one of the main cariogenic bacteria for caries. It was found that the clinical strains of S. mutans isolated from caries active population have stronger cariogenic ability than the isolates from caries-free (CF) people. Previous studies have found that curcumin can inhibit biofilm formation of S. mutans UA159. The objective of this study is to explore the antibiofilm effect of curcumin on the clinical isolates of S. mutans from severe early childhood caries(SECC)., Results: The isolates from SECC group had more biomass than CF group (t = 4.296, P < 0.001). The acidogenicity and aciduricity of the strains from two groups showed no significant difference. After treatment with curcumin, the viability of biofilm was reduced to 61.865% ± 7.108% in SECC and to 84.059% ± 10.227% in CF group at 24 h (P < 0.05). The net reduction of live bacteria and total bacteria in the SECC group was significantly higher than that of the CF group (live bacteria t = 3.305, P = 0.016; total bacteria t = 2.378, P = 0.045) at 5 min. For 24 h, the net reduction of live bacteria and total bacteria in the SECC group was significantly higher than that of the CF group (live bacteria t = 3.305, P = 0.016; total bacteria t = 2.378, P = 0.045). The reduction of biofilm thickness reduced significantly in 5 min (t = 4.110, P = 0.015) and in 24 h (t = 3.453, P = 0.014). Long-term (24 h) curcumin treatment inhibited the amount of EPS in SECC group from (25.980 ± 1.156) μm 3 /μm 2 to (20.136 ± 1.042) μm 3 /μm 2 , the difference was statistically significant (t = 7.510, P < 0.001). The gene of gtfC, gtfD, ftf, gbpB, fruA and srtA in the CF group and the gtfB, gtfC, gtfD, ftf, gbpB, srtA in SECC group were respectively reduced after 5 min curcumin treatment. After 24 h treatment, the gtfB, gtfC, gtfD, ftf, gbpB, fruA and srtA in both two groups were downregulation, all the differences were statistically significant., Conclusions: Curcumin has antibiofilm activity on clinical strains of S. mutans, especially for those isolated from SECC.

Biofilm formation under cariogenic conditions contributes to dental caries development, in which Streptococcus mutans (S. mutans) is regarded as the major cariogenic bacteria. Here, we synthesized a series of imidazolium salts. Their properties of antimicrobial and anti-biofilm were investigated. The microdilution method crystal violet staining, and cell counting Kit-8 assay were used to screen imidazolium salts. Then, the bacterial composition in multi-species biofilm composed of S. mutans, Actinomyces naeslundii, and Streptococcus gordonii was quantified by quantitative PCR. The exopolysaccharide and morphology of the structure of multi-species biofilm were further observed by confocal laser scanning microscopy and scanning electron microscope, respectively. Imidazolium salts exhibited highly antimicrobial activity against oral pathogens, especially for S. mutans. Compounds with ortho-diisopropyl and para-methoxyl on N-moieties as well as bearing ancenaphthyl skeleton (C5) showed the lowest cytotoxicity and most efficient anti-biofilm activity. C5 inhibited approximately 50% of multi-species biofilm at 0.98 μg/mL. Notably, C5 resulted in 98.97% live S. mutans and 77.65% A. naeslundii decreased. Furthermore, the exopolysaccharide was reduced by 88%, along with a sparse and scattered microstructure. The imidazolium salts present low cytotoxicity and remarkable antimicrobial activity against S. mutans in multi-species biofilm, suggesting that they may have a great potential in anti-biofilm clinical applications. Bacterial biofilms have evoked worldwide attention owing to their serious threats to public health, but how to effectively eliminate bacterial biofilms still remains great challenges. Here, we rationally designed a novel and vigorous chitosan grafted Fe-doped-carbon dots (CS@Fe/CDs) as an efficient artificial nanozyme to combat rigid bacterial biofilms through the selective activation of Fenton-like reaction-triggered peroxidase-like catalytic activity and the synergistic antibacterial activity of CS. On the one hand, the peroxidase-like catalytic activity made CS@Fe/CDs catalyze H 2 O 2 for producing hydroxyl radicals (•OH), resulting in efficient cleavage of extracellular DNA (eDNA). On the other hand, CS was capable of binding with the negatively charged cell membrane through electrostatic interaction, changing the cell membrane permeability and causing cell death within bacterial biofilms. Based on their synergistic effects, the fragments of bacterial biofilm and exposed bacteria were persistently eradicated. Remarkably, CS@Fe/CDs-based nanozyme not only enabled the effective destroying of gram-positive Staphylococcus aureus (S. aureus) biofilms, but also completely eliminated gram-negative Pseudomonas aeruginosa (P. aeruginosa) biofilms, showing great potential as a promising anti-biofilm agent against bacteria biofilms. This proposed synergistic strategy for bacterial biofilm eradication might offer a powerful modality to manage of bacterial biofilm fouling in food safety and environmental protection. A series of well-defined copper-NHCs have been synthesized and characterized. Relationship between the structure of the copper-NHCs and the inhibition effects was investigated, and Copper-NHCs of Cu1 was demonstrated as a broad-spectrum inhibitor to oral bacteria. With the goal of developing high-efficiency oral antimicrobial agents to prevent S. mutans biofilm formation, a collection of copper(I)- N -heterocyclic carbenes (NHCs) complexes with different substitutions was synthesized and characterized. Their inhibitory activities of Cu1–Cu7 were screened. It was found that Cu1 provided significant excellent performance, which suggested that the less lipophilic and less sterically hindered copper complexes would be effective towards the bacteria. This study demonstrates the great potential of copper(I)-containing small molecule complexes as broad-spectrum inhibitors to treat oral bacteria, especially for diminishing S. mutans biofilm formation, which opens promising perspectives for the development of new metal-NHCs for controlling the biofilm formation associated with pathogens. The effect of acetate (HAc) and propionate (HPr) on denitrifying phosphorus removal (DPR) was evaluated in a novel two-sludge A2/O - MBBR (anaerobic/anoxic/oxic - moving bed biofilm reactor) system. Results showed that it was the carbon source transformation and utilization especially the composition of poly-β-hydroxyalkanoates (PHA) (mainly poly-β-hydroxybutyrate (PHB) and poly-bhydroxyvalerate (PHV)) decided DPR performance, where the co-exist of HAc and HPr promoted the optimal nitrogen (85.77%) and phosphorus (91.37%) removals. It facilitated the balance of PHB and PHV and removing 1 mg NO 3 − (PO 4 3−) consumed 3.04–4.25 (6.84–9.82) mgPHA, where approximately 40–45% carbon source was saved. Mass balance revealed the main metabolic pathways of carbon (M An,C (consumed amount in anaerobic stage) and M A-O,C (consumed amount in anoxic and oxic stages): 66.38–76.19%), nitrogen (M DPR,N (consumed amount in DPR): 57.01–65.75%), and phosphorus (M WS,P (discharged amount in waste sludge): 81.05–85.82%). Furthermore, the relative abundance and microbial distribution were assessed to elucidate DPR mechanism (e.g. Accumulibacter , Acinetobacter , Dechloromonas , Competibacter , and Defluviicoccus) in the A2/O reactor and nitrification performance (e.g. Nitrosomonas , Nitrosomonadaceae and Nitrospira) in the MBBR. Carbon source was demonstrated as the key point to stimulate the biodiversity and bioactivity related to DPR potential, and the operational strategy of carbon source addition was proposed based on the utilizing rules of HAc and HPr.
