Computational toxicology is widely applied to screen tens and thousands of potential environmental endocrine disruptors (EDCs) for its great efficiency and rapid evaluation in recent years. Polychlorinated biphenyls (PCBs) with 209 congeners have not been comprehensively tested for their ability to interact with the thyroid receptor (TR), which is one of the most extensively studied targets related to the effects of EDCs. In this study, we aimed to determine the thyroid-disrupting mechanisms of 209 PCBs through the combination of a novel computational ternary classification model and luciferase reporter gene assay. The reporter gene assay was performed to examine the hormone activities of 22 PCBs via TR to classify their profiles as agonistic, antagonistic or inactive. Thus, six agonists, eleven antagonists and seven inactive chemicals against TR were identified in in vitro assays. Then, six relevant variables, including molecular structural descriptors and molecular docking scores, were selected for describing compounds. Machine learning methods (i.e., linear discriminant analysis (LDA) and support vector machines (SVM)) were used to build classification models. The optimal model was obtained by using SVM, with an accuracy of 88.24% in the training set, 80.0% in the test set and 75.0% in 10-fold cross-validation. The remaining 187 PCB congeners' hormone activities were predicted using the obtained models. Out of these PCBs, the SVM model predicted 38 agonists and 81 antagonists. The findings revealed that higher chlorinated PCBs tended to have TR-antagonistic activities, whereas lower chlorinated PCBs were agonists. Pollution of agricultural soils by pesticides, such as organochlorine pesticides (OCPs), can be a significant issue since high detection rates of these compounds were reported in our previous studies. However, more uncertain kinds, quantities and density of pollutants remained in soil samples were unidentified. In this study, the total hormonal activities of complex mixtures of both known and unknown contaminants in agricultural soils in mainland China were measured by applying highly sensitive reporter gene assays for detecting agonists/antagonists for estrogen receptor (ER), androgen receptor (AR), progesterone receptor (PR), glucocorticoid receptor (GR) and mineralocorticoid receptor (MR). High detection rates of estrogenic activities and anti-progestogenic activities were observed among the 123 soil samples, reaching 79% and 73%, respectively. More than half of the soil samples showed obvious antagonistic effects against AR and GR. Approximately a third of tested samples exhibited androgenic, progestogenic and glucocorticoidic effects. A total of 72% and 78% soil extracts had mineralocorticoid-like and anti-mineralocorticoid activities, respectively. Significant positive correlations were observed between estrogenic activity and the concentrations of Σdichlorodiphenyltrichloroethanes (DDTs), Σendosulfans, Σchlordanes, heptachlor and Σdrins, respectively, but not other receptors. As a rapid and convenient pre-caution method, determination of endocrine-disrupting potencies of contaminated soils via bioassay could help to identify and define sites that required further attention for ecological risk assessments., (Copyright © 2017 Elsevier Ltd. Polychlorinated biphenyls (PCBs) are thought to cause adverse health effects, particularly endocrine disruption. However, results on the estrogenic activities of a large set of PCB congeners through hormone receptors have not been fully studied. In this study, we evaluated the anti/estrogenic effects of 20 PCBs using an in vitro dual-luciferase reporter gene assay. PCB 18, 28, 49, 52, 99, 101, 103, 110, and 128 exhibited estrogenic effects, whereas PCB 118, 138, 163, 170, 180, 187, 194, 199 and 203 behaved as anti-estrogens. In particular, PCB 30 and 44 exhibited both agonistic and antagonistic activities in the dual-luciferase reporter gene assay. The results obtained from the dual-luciferase reporter gene assay, yeast two-hybrid assay and E-SCREEN were compared, suggesting that the dual-luciferase reporter gene assay is a useful approach for high-throughput screening. Background: The incorporation of human APOBEC3G (hA3G) into HIV is required for exerting its antiviral activity, therefore the mechanism underlying hA3G virion encapsidation has been investigated extensively. hA3G was shown to form low-molecular-mass (LMM) and high-molecular-mass (HMM) complexes. The function of different forms of hA3G in its viral incorporation remains unclear. Methodology/Principal Findings: In this study, we investigated the subcellular distribution and lipid raft association of hA3G using subcellular fractionation, membrane floatation assay and pulse-chase radiolabeling experiments respectively, and studied the correlation between the ability of hA3G to form the different complex and its viral incorporation. Our work herein provides evidence that the majority of newly-synthesized hA3G interacts with membrane lipid raft domains to form Lipid raft-associated hA3G (RA hA3G), which serve as the precursor of mature HMM hA3G complex, while a minority of newly-synthesized hA3G remains in the cytoplasm as a soluble LMM form. The distribution of hA3G among the soluble LMM form, the RA LMM form and the mature forms of HMM is regulated by a mechanism involving the N-terminal part of the linker region and the C-terminus of hA3G. Mutagenesis studies reveal a direct correlation between the ability of hA3G to form the RA LMM complex and its viral incorporation. Conclusions/Significance: Together these data suggest that the Lipid raft-associated LMM A3G complex functions as the cellular source of viral hA3G. [ABSTRACT FROM AUTHOR]

Abstract: Bone marrow stromal cell antigen 2 (BST-2, also known as Tetherin) inhibits HIV-1 release and thereby severely impairs viral replication. HIV-1 accessory protein Vpu induces the down-regulation of cell surface BST-2, and counteracts the antiviral function of BST-2. Blocking Vpu-mediated down-regulation of cell surface BST-2 is viewed as a new opportunity for developing anti-HIV drugs. In this study, we have developed a high-throughput cell-based ELISA to identify small molecules that antagonize HIV-1 Vpu function and consequently inhibit HIV-1 replication through rescuing the antiviral activity of host BST-2. This cell-ELISA shows an excellent correlation with results obtained by flow cytometry (FACS). Under optimal conditions, a Z′ factor of 0.605 was achieved in a 96-well format. Together, these results demonstrate that this assay can be used to quantify the cell surface level of BST-2 and be adapted to a high-throughput screening for novel anti-HIV compounds.
