(-)-[alpha]-bisabolol (BISA) is a sesquiterpene alcohol, which has several recognized biological activities, including Anti-inflammatory, anti-irritant, and antibacterial properties. In the present study, we investigated the influence of BISA (5, 25, and 250 [micro]mol/L) on rotenone (500 [micro]mol/L)-induced toxicity in Drosophila melanogaster for 7 days. BISA supplementation significantly decreased rotenone-induced mortality and locomotor deficits. The loss of motor function induced by rotenone correlated with a significant change in stress response factors; it decreased thiol levels, inhibited mitochondria complex I, and increased the mRNA expression of antioxidant marker proteins such as superoxide dismutase (SOD), catalase (CAT), and the keap1 gene product. Taken together, our findings indicate that the toxicity of rotenone is likely due to the direct inhibition of complex I activity, resulting in a high level of oxidative stress. Dietary supplementation with BISA affected the expression of SOD mRNA only at a concentration of 250 [micro]mol/L, and did not affect any other parameter measured. Our results showed a protective effect of BISA on rotenone-induced mortality and locomotor deficits in Drosophila; this effect did not correlate with mitochondrial complex I activity, but may be related to the antioxidant protection afforded by eliminating superoxide generated as a result of rotenone-induced mitochondrial dysfunction. L'(-)-[alpha]-bisabolol (BISA) est un alcool sesquiterpenique, dote de plusieurs activites biologiques reconnues, y compris des proprietes anti-inflammatoires, anti-irritantes et antibacteriennes. Dans les presents travaux, nous avons etudie l'influence du BISA (5, 25 et 250 [micro]mol/L) sur la toxicite provoquee par la rotenone (500 [micro]mol/L) sur 7 jours chez Drosophila melanogaster. La supplementation en BISA a entraine une diminution notable de la mortalite et des deficits moteurs provoques par la rotenone. La perte de fonction motrice provoquee par la rotenone etait proportionnelle a une variation marquee des facteurs de reaction au stress; elle a entraine une diminution des taux de thiols, une inhibition du complexe mitochondrial I et une augmentation de l'expression de l'ARNm de marqueurs proteiques anti-oxydants comme la superoxyde dismutase (SOD), la catalase (CAT) et le produit du gene keapl. Dans l'ensemble, nos resultats montrent que la toxicite de la rotenone est probablement le fait d'une inhibition directe de l'activite du complexe I, ce qui se traduit par un degre eleve de stress oxydatif. La supplementation alimentaire en BISA modifiait l'expression de l'ARNm de la SOD uniquement a une concentration de 250 [micro]mol/L, et elle ne modifiait aucun des autres parametres mesures. Nos resultats ont montre un effet protecteur du BISA contre la mortalite et les deficits moteurs provoques par la rotenone chez la drosophile; cet effet n'etait pas proportionnel a l'activite du complexe mitochondrial I, mais il pourrait etre lie a la protection anti-oxydante obtenue en reduisant a neant la generation d'ions superoxyde issue du dysfonctionnement mitochondrial provoque par la rotenone.
