Due to the emergence and development of antibiotic resistance in the treatment of bacterial infections, efforts to discover new antimicrobial agents have increased. One of these antimicrobial agents is a compound produced by a large number of bacteria called bacteriocin. Bacteriocins are small ribosomal polypeptides that can exert their antibacterial effects against bacteria close to their producer strain or even non-closely-relatedstrains. Adequate knowledge of the structure and functional mechanisms of bacteriocins and their spectrum of activity, as well as knowledge of the mechanisms of possible resistance to these compounds, will lead to further development of their use as an alternative to antibiotics. Furthermore, most bacteria that live in the gastrointestinal tract (GIT) have the ability to produce bacteriocins, which spread throughout the GIT. Despite antimicrobial studies in vitro, our knowledge of bacteriocins in the GIT and the migration of these bacteriocins from the epithelial barrier is low. Hence, in this study, we reviewed general information about bacteriocins, such as classification, mechanism of action and resistance, emphasizing their presence, stability, and spectrum of activity in the GIT.

The current study was completed to analyze the effect of natural components on various improvement affecting traits of Kajli sheep in Pakistan. For this reason, we apply two huge contemporary datasets in sheep to explore factors that influence the traits. Therefore, the generation information record of 13715 Kajli sheep lambing accumulated from 1994 to 2010 at Livestock Experimental Stations Khushab and Khizarabad, Punjab. AIM We investigated differential DNA methylation in gingival tissues in periodontal health, gingivitis, and periodontitis, and its association with differential mRNA expression. MATERIALS AND METHODS Gingival tissues were harvested from individuals and sites with clinically healthy and intact periodontium, gingivitis, and periodontitis. Samples were processed for differential DNA methylation and mRNA expression using the IlluminaEPIC (850 K) and the IlluminaHiSeq2000 platforms, respectively. Across the three phenotypes, we identified differentially methylated CpG sites and regions, differentially expressed genes (DEGs), and genes with concomitant differential methylation at their promoters and expression were identified. Myocardial tuberculosis is an exceptionally rare form of extra-pulmonary TB. Few cases were reported world-wide. Here a young snake charmer who had skin tuberculosis 5 yrs back admitted into National institute of diseases of Chest and hospital (NIDCH), Dhaka with the complaints of cough, palpitation and breathlessness for 2 months. He had right axillary firm matted lymphadenopathy, left sided large pleural effusion, left ventricular and septal hypertrophy with band and mass inside the ventricle (evident on CT scan of heart and echocardiography). His ESR was 95 mm in1st hr, Mantaux test was 15mm, Pleural fluid was exudative lymphocyte predominant with adenosin deaminase (ADA) 68.6 U/L. Fine needle aspirates from right axillary LNs showed Mycobacterium tuberculosis on GeneXpert for MTB/RIF testing and caseous granuloma on cytopathological study. Whole Body F18 FDG PET-CT revealed numerous low FDG avid size significant lymph nodes in right side of neck, mediastinum and right axilla with cardiomegaly with focal FDG avid within the left ventricular cavity likely to be prominent papillary muscle. MRI of heart or Myocardial biopsy for histology was not done due to their cost and invasiveness and also for that there was sufficient evidence of having tuberculosis in lymph node, pleura nas myocardium. This patient was treated with anti tubercular medications (3HRZE2S/5HRE) with prednisolone for six months. After treatment, myocardial lesions, pleural effusion and lymphadenopathy were found resolved. Thus a case of fatal and serious tuberculosis was explored and managed successfully.

This study was carried out to evaluate the growth response of guppy Poecilia reticulata to different dietary protein levels. A total of 72 fry of guppy fish of an initial body weight 0.365± 0.01g were distributed randomly into 60 L aquaria at the density rate of 0.05 g/L. Fish were fed to satiation twice daily for eight weeks with diets containing different dietary protein levels (20%, 30%, or 40%.). The survival rate of guppy was unaffected by increasing dietary protein levels. At the end of the experiment, weight gains (g) ranging from 1.93 to 3.55 were obtained corresponding to 20% and 40% crude protein respectively. The growth performance and feeding efficiencies were significantly improved (P < 0.05) by increasing dietary protein levels. However, no significant difference (P > 0.05) was observed in the specific growth rate of fish that were fed 20% and 30% dietary protein levels. The value of the condition factor was significantly high in fish fed at 40% dietary protein. It can be concluded that a 40% dietary protein level is recommended for guppy fish.

The ability of various Streptomyces isolates obtained from soil to induce systemic resistance in tomato (Solanum lycopersicum cv. Supra) plant against Tomato mosaic virus (ToMV) was characterized in current study. Importantly, of nine Streptomyces isolates tested herein, the culture filtrate (CF) of one isolate, designated as Streptomyces ovatisporus LC597360, was the most effective. It exhibited 93.9% biocontrol efficacy and induced a significant decrease (17.6 ± 0.8%) of symptoms severity compared with infected control plants. These finding were confirmed using I-ELISA showing that ToMV concentration was significantly reduced in plants treated with S. ovatisporus LC597360 CF as compared with plants inoculated with ToMV. Moreover, treatment with CF of S. ovatisporus LC597360 not only increased activity of defense-related enzymes such as ascorbate oxidase, catalase, peroxidase, and polyphenol oxidase, but also induced plant growth promotion. The present study is the first one that demonstrates the potential of S. ovatisporus LC597360 in biocontrol of ToMV and investigated its antiviral mechanisms.

The natural product celastrol has also been widely documented as a potent anti-inflammatory, anti-angiogenic, and anti-oxidant.Purpose: To elucidate the antitumor effect of celastrol on pancreatic cancer cells and its modulatory role on whole genome expression.Methods: The antitumor activity of celastrol on a panel of pancreatic cancer cells has been evaluated by Sulforhodamine B assay. Caspase 3/7 and histone-associated DNA fragments assays were done for apoptosis measurement. Additionally, prostaglandin (PGE2) inhibition was evaluated. Moreover, a microarray gene expression profiling was carried out to detect possible key players that modulate the antitumor effects of celastrol on cells of pancreatic cancer.Results: Our findings indicated that celastrol suppresses the cellular growth of pancreatic cancer cells, induces apoptosis, and inhibits PGE2 production. Celastrol modulated many signaling genes and its cytotoxic effect was mainly mediated via over-expression of ATF3 and DDIT3, and down-expression of RRM2 and MCM4.Conclusion: The current study aims to be a starting point to generate a hypothesis on the most significant regulatory genes and for a full dissection of the celastrol possible effects on each single gene to prevent the pancreatic cancer growth.Keywords: celastrol, natural products, pancreatic cancer, RRM2, DDIT3, MCM4

BACKGROUND: The diagnosis of osteomyelitis is a key step of diabetic foot management. Procalcitonin (PCT) is a novel infection marker. This study aimed to investigate the diagnostic value of procalcitonin and other conventional infection markers and clinical findings in diagnosis of osteomyelitis in diabetic foot patients. METHODS AND MATERIALS: This diagnostic value study was carried out on ninety patients with diabetic infected foot ulcers admitted in Kashan Beheshti Hospital, 2016. After obtaining consent, 10 cc blood sample was taken for measuring serum PCT, CBC, ESR, CRP and FBS. Clinical characteristics of the wounds were noted. Magnetic resonance imaging of the foot was performed in all patients to diagnose osteomyelitis. All statistical analyses were done with the use of SPSS-16. RESULTS: PCT levels were 0.13 ± 0.02 ng/mili patients with osteomyelitis (n= 45) and 0.04 ± 0.02 ng/ml in patients without osteomyelitis (n= 45). PCT, Erythrocyte sedimentation rate and C-reactive protein was found significantly higher in patients with osteomyelitis (p< 0.001). The ROC curve was calculated for PCT. The area under the ROC curve for infection identification was 1 (p< 0.001). The best cut-off value for PCT was 0.085 ng/ml. Sensitivity, specificity, and positive and negative predictive values were 100%, 97.8%,97.8% and 100%, respectively. CONCLUSION: In this group of patients, PCT was useful to discriminate patients with bone infection. Also, Erythrocyte sedimentation rate and C-reactive protein can be used as a marker of osteomyelitis in diabetic patients.

Mutational signatures are imprints of pathophysiological processes arising through tumorigenesis. We generated isogenic CRISPR-Cas9 knockouts (Δ) of 43 genes in human induced pluripotent stem cells, cultured them in the absence of added DNA damage, and performed whole-genome sequencing of 173 subclones. ΔOGG1, ΔUNG, ΔEXO1, ΔRNF168, ΔMLH1, ΔMSH2, ΔMSH6, ΔPMS1, and ΔPMS2 produced marked mutational signatures indicative of being critical mitigators of endogenous DNA modifications. Detailed analyses revealed mutational mechanistic insights, including how 8-oxo-dG elimination is sequence-context-specific while uracil clearance is sequence-context-independent. Mismatch repair (MMR) deficiency signatures are engendered by oxidative damage (C>A transversions), differential misincorporation by replicative polymerases (T>C and C>T transitions), and we propose a ‘reverse template slippage’ model for T>A transversions. ΔMLH1, ΔMSH6, and ΔMSH2 signatures were similar to each other but distinct from ΔPMS2. Finally, we developed a classifier, MMRDetect, where application to 7,695 WGS cancers showed enhanced detection of MMR-deficient tumors, with implications for responsiveness to immunotherapies.

Objectives: Demodicosis is a vital skin problem in dogs. The present study has determined the prevalence and associated risk factors of demodicosis in dogs and the response to treatment. Materials and Methods: A total of 100 skin scrapings were collected from dogs having dermatolog¬ical lesions brought to the Teaching and Training Pet Hospital and Research Center of Chattogram Veterinary and Animal Sciences University for treatment purpose. The collected scrapings were dissolved in 10% potassium hydroxide to detect mites through microscopic examination. Various risk factors like breed, age, sex, hair type, health status, and management system (indoor and outdoor) were analyzed using the logistic regression model. Positive cases were treated with oral ivermectin (Scabo®; at 0.6 mg/kg/day) along with amitraz 12.5% (Ridd®) diluted to 0.05% for rub¬bing on the body after bathing with chlorhexidine shampoo (PetHex®). Clavulanate amoxicillin (Moxaclav®) and omega-3 fatty acids (OMG-3®) were also suggested to prevent secondary bacte¬rial infection and to maintain skin and hair coat integrity. The recovery rate was observed every 24 weeks of treatment upto 1113 weeks. Results: The overall prevalence of demodicosis was recorded as 27%. Hair type, health status, and management system were significantly (p < 0.05) associated with the occurrence of demodicosis in dogs. Following treatment, the first negative skin scraping for mite was noticed at 810 weeks of treatment, and in all cases, clinical signs completely disappeared at 8090 days of treatment. Conclusion: A good percentage of the dogs having dermatological lesions was determined as demodicosis. Long-term oral ivermectin and topical amitraz, an oral antibiotic, and nutritional therapy are effective against canine demodicosis. Keeping long-haired dogs with good body con¬dition and indoor management is highly suggestive of preventing and controlling the disease. [J Adv Vet Anim Res 2021; 8(1.000): 116-122]

Coronavirus disease 2019 (COVID-19) is a contagious disease which was first identified in Wuhan, China in December 2019. Severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) is responsible for this ongoing pandemic worldwide. COVID-19 itself has a high mortality rate but in addition with bacterial co-infection, the risk of death amplifies much higher. The diagnosis of COVID-19 along with other respiratory co-infections can be a difficult task due to the similarities in their presentations. Here, we present a case of 60-years-old gentleman who was COVID-19 positive, co-infected with three types of bacterial pathogens, which were Mycobacterium tuberculosis, Enterobacter spp. and Pseudomonas. The patient was isolated and treated according to the pathogens’ culture sensitivity reports and was discharged when his condition improved and advised for routine follow-up. The source of the co-infection could not be identified and may have been hospital acquired. Therefore, every hospital should give utmost priority to infection prevention and control (IPC) strategies.

Klebsiella pneumoniae causes a variety of human infections including pneumonia. Herein, 3 lytic bacteriophages specific for K. pneumoniae designated ΦKpnM-vB1, ΦKpnP-vB2 and ΦKpnM-vB3 were isolated and characterized. Transmission electron microscopy (TEM) analysis revealed that both ΦKpnM-vB1 and ΦKpnM-vB3 belong to family Myoviridae, while ΦKpnP-vB2 is a member of family Podoviridae. The one-step growth curve showed that ΦKpnM-vB1, ΦKpnP-vB2 and ΦKpnM-vB3 exhibited latent period of 10 min. The average burst sizes were 100, 150 and 120 PFU/cell, respectively. Isolated phages showed high thermal and pH stability. The genomic analysis indicated that ΦKpnM-vB1, ΦKpnP-vB2 and ΦKpnM-vB3 contain dsDNA genome with estimated sizes of 55, 40 and 50 Kbp, respectively. Isolated phages had lytic activity on K. pneumoniae and Escherichia coli strains. Isolated phages were highly efficient in reduction of Klebsiella biofilm suggesting their use to control biofilm formation caused by this pathogen. Isolated ΦKpnM-vB1, ΦKpnP-vB2 and ΦKpnM-vB3 are proposed to be suitable candidates for phage therapy applications. These phages offer an effective solution for treatment of infections caused by these drug-resistant bacteria.

LC-HRMS-based metabolomics approach was applied to the river Nile-derived fungus Aspergillus awamori after its fermentation on four different media and using four epigenetic modifiers as elicitors. Thereafter, a comprehensive multivariate statistical analysis such as PCA, PLS-DA and OPLS-DA were employed to explain the generated metabolomic data (1587 features). PCA showed that the fungus displayed a unique chemical profile in each medium or elicitor. Additionally, PLS-DA results revealed the upregulated metabolites under each of these conditions. Results indicated that both rice and malt dextrose agar were recognized as the best media in terms of secondary metabolites diversity and showed better profiles than the four applied epigenetic modifiers, of which nicotinamide was the best secondary metabolite elicitor. Testing the antibacterial and cytotoxic effects of all A. awamori-derived extracts revealed that using epigenetic modifiers can induce antimicrobial metabolites against S. aureus and E. coli, whereas using rice, malt dextrose or nicotinamide can induce groups of cytotoxic metabolites. OPLS-DA results assisted in the putative identification of the induced metabolites that could be responsible for these observed inhibitory activities. This study highlighted how powerful the OSMAC approach in maximizing of the chemical diversity of a single organism. Furthermore, it revealed the power of metabolomics in tracing, profiling and categorizing such chemical diversity and even targeting the possible bioactive candidates which require further scaling up studies in the future.

Momordica charantia (Family: Cucurbitaceae), commonly referred to as Bitter Gourd, Karela, and balsam pear, is a very familiar vegetable of Bangladesh and has long been used in traditional medicine to cure numerous illnesses. Three different extracts of a selected hybrid variety of M. charantia fruit from Bangladesh were prepared and screened to assess cytotoxic activity using the brine shrimp lethality test (BST). Gallic acid was used as a positive control. The LC50 value of standard gallic acid, ethanol, ethyl acetate, and petroleum ether extract from the fruits of Momordica charantia L. found 4.40 µg/ml, 125.89 µg/ml, 146.49 µg/ml, and 194.2 µg/ml, respectively. The ethanol extract demonstrated a prominent cytotoxic activity in comparison with the moderate cytotoxic activity of ethyl acetate and petroleum ether extract. The study results support the predominant presence of cytotoxic bioactive compounds in the more polar solvent of the fruit extract which can be beneficial for our health because of the numerous pharmacological activities of these compounds.

Paclitaxel (PTX) is widely used to treat breast and ovarian cancers, but innate and acquired resistance often compromises its applications. The objective of this study was to screen new-generation taxanes for their efficiency against both PTX-sensitive and PTX-resistant breast cancer cells. From twelve compounds, difluorovinyl-ortataxel (DFV-OTX) displayed potent cytotoxic activities against both PTX-sensitive and PTX-resistant breast cancer cells. Moreover, DFV-OTX effectively induced tubulin/microtubule polymerization and G2/M phase arrest, leading to apoptosis in both PTX-sensitive and PTX-resistant cancer cells. Molecular docking analysis showed that DFV-OTX possesses unique hydrogen-bonding and van der Waals interactions with β-tubulin. LC-MS/MS analysis also demonstrated that the intracellular drug amount of DFV-OTX was lower than that of PTX, which would be critical to overcome PTX-resistance. Furthermore, DFV-OTX exhibited clear efficacy in the MCF-7R and MDA-MB-231R tumor xenografts in mouse models. Taken together, our results demonstrate that the novel taxane, DFV-OTX, can effectively overcome PTX-resistance in MDA-MB-231R cells, wherein the drug resistance was attributed to ABCB1/ABCG2 upregulation and a distinct mode of action in MCF-7R cells. Our results strongly indicate that DFV-OTX is a promising chemotherapeutic agent for the treatment of PTX-resistant cancers.

Methods Ten peri-implant tissue biopsies of patients diagnosed clinically and radiographically with peri-implantitis were obtained from the archives of Oral Pathology Centre, University of Otago. The inclusion criteria involves: bleeding on probing, ≥6 mm probing depth and ≥3 mm radiographic bone loss around the dental implant. Peri-implant tissue samples were evaluated using scanning electron microscopy-energy dispersive x-ray spectroscopy (SEM-EDS) to identify of sites with/without titanium particles. Antibodies against human transforming growth factor beta 1 (TGF-β1), receptor activator of nuclear factor kappa-B ligand (RANKL), interleukin 33 (IL-33) and cluster of differentiation 68 (CD68) were used to stain the specimens. ImageJ software was used to standardise the sampling area, compare and characterise the inflammatory infiltrate in tissues with/without titanium particles. Inflammatory cytokines positivity was assessed using the immunoreactive scores (IRSs). Results Light microscopy and SEM-EDS analysis identified titanium wear particles in 90% of the tissue samples, associated with a mixed chronic inflammatory infiltrate. Introduction.Enterococcus faecalisis a facultative, anaerobic, opportunistic pathogen associated with medical and dental diseases. Bacterial phenotypic traits and pathogenesis are often influenced by lysogeny.Aim.The aim of this study was to characterize both the morphology and complete genome sequences of induced prophages purified fromE. faecalisclinical isolates.Methodology.E. faecalisisolates were recovered from the roots of teeth of patients attending an endodontic clinic. The morphological features of isolated phage were characterized using transmission electron microscopy (TEM). DNA sequencing was performed using the Illumina MiSeq platform.Results.TEM indicated that the isolated φEf-vB1 prophage belongs to the familySiphoviridae. The φEf-vB1 prophage was stable over a wide range of temperatures and pH. Sequencing of φEf-vB1 DNA revealed that the phage genome is 37 561 bp in length with a G+C content of 37.6mol% and contained 53 ORFs. Comparison with previously predicted prophage genomes usingblastrevealed that φEf-vB1 has a high sequence similarity to previously characterized phage genomes. The lysogenicE. faecalisstrain exhibited a higher biofilm formation capacity relative to the non-lysogenic strain.Conclusion.The current findings highlight the role of lysogeny in modification ofE. faecalisproperties and reveal the potential importance of prophages inE. faecalisbiology and pathogenesis.

Escherichia coli is a pathogen commonly encountered in clinical laboratories, and is capable of causing a variety of diseases, both within the intestinal tract (intestinal pathogenic strains) and outside (extraintestinal pathogenic E. coli, or ExPEC). It is associated with urinary tract infections (UTIs), one of the most common infectious diseases in the world. This report represents the first comparative analysis of the draft genome sequences of 11 uropathogenic E. coli (UPEC) strains isolated from two tertiary hospitals located in Dhaka and Sylhet, Bangladesh, and is focused on comparing their genomic characteristics to each other and to other available UPEC strains. Multilocus sequence typing (MLST) confirmed the strains belong to ST59, ST131, ST219, ST361, ST410, ST448 and ST4204, with one of the isolates classified as a previously undocumented ST. De novo identification of the antibiotic resistance genes blaNDM-5, blaNDM-7, blaCTX-M-15 and blaOXA-1 was determined, and phenotypic-genotypic analysis of virulence revealed significant heterogeneity within UPEC phylogroups.

Objectives: The present study estimated the seroprevalence of avian reovirus (ARV) infections in backyard chickens of the Mymensingh district in Bangladesh. Materials and Methods: Considering several risk factors, a total of 460 serum samples were col¬lected from backyard chickens from eight Upazilas of the Mymensingh district in Bangladesh. Blood samples were taken from the wing vein using 3-ml sterile syringes and kept at room tem¬perature for clotting in a slanting position and then transported to the laboratory maintaining the cool chain. Subsequently, the prepared sera were harvested and stored at −20°C until used. Finally, an indirect enzyme-linked immunosorbent assay (ELISA) was performed to detect ARV-specific antibodies using a commercial ARV antibody detection ELISA test kit. Results: The results revealed high prevalence rates of ARV antibodies, with a total seroprevalence of 69.78% (321/460). Area-wise, 74.55% (82/110) seroprevalence was recorded as the highest in Mymensingh Sadar, whereas 64% (32/50) was the lowest in Gauripur Upazila. With regard to sex, female chickens showed a significantly higher (p < 0.05) seroprevalence as 90.33% (271/300) compared to male chickens 31.25% (50/160). With regard to age groups, the seroprevalence of ARV infection was 59.33% (89/150) within 28 weeks, 82% (205/250) within 916 weeks, and 45% (27/60) within 1720 weeks, respectively. Based on hygienic conditions, the highest sero¬prevalence of ARV was noted in backyard chickens housed in poor conditions 80% (120/150) than good conditions 50% (40/80). Backyard chickens reared in free-ranging conditions exhibited a significantly higher seroprevalence 73.33% (220/300) of ARV antibodies compared to rearing in separate houses 63.12% (101/160). The seroprevalence of ARV was higher in crossbreeds 71.67% (43/60), brought from market 76% (38/50), and unhealthy 78.57% (55/70) backyard chickens than non-descriptive indigenous 69.5% (278/400), home-reared 69.02% (283/410), and healthy chick¬ens 68.21% (266/390). Conclusion: The high prevalence of ARV antibodies revealed in the current study indicates an extensive exposure of ARV to backyard chickens in Bangladesh that may be transmitted naturally to other chickens, ultimately leading to ominous economic effects on the poultry sector. The heterogeneity of climate and different agro-ecological conditions in Iran have resulted in development of 27 indigenous sheep breeds. Wild Asiatic mouflon (Ovis orientalis) is believed to be the ancestor of Iranian sheep. Evaluation of genetic diversity and population structure within and among domestic breeds has important implications for animal breeding programs and genetic resources management. Based on 50K SNP genotype data, we studied the genetic diversity of five indigenous Iranian sheep breeds: Afshari (n = 37), Moghani (n = 34), Qezel (n = 35), Zel (n = 46) and Lori-Bakhtiari (n = 46), and Asiatic mouflon (n = 8) sampled from Iran. Furthermore, genetic diversity and the breed admixture of Iranian sheep were assessed on a larger geographic scale using a reference panel comprising: three indigenous Afghan breeds - Arabi (n = 15), Balouchi (n = 15) and Gadik (n = 15); three indigenous breeds from Turkey and Cyprus - Cyprus Fat Tail (n = 30), Karakas (n = 18) and Norduz (n = 20); and three commercial European breeds - Suffolk (n = 19), Comisana (n = 24) and Engadine Red Sheep (n = 24). The results revealed that the investigated breeds are divided into five genetically distinct clusters according to their geographic origin. Afshari was closest to the local mouflon population and showed signs of mouflon admixture. Qezel was identified as a hybrid sheep breed. Much evidence supported the Afghan breeds being identical. Inbreeding values, which were estimated based on ROHs, were highest for Suffolk (FROH = 0.0544) and lowest for Balouchi (FROH = 0.0078). In conclusion, analysis of selected breeds from neighboring countries along with Asiatic mouflon gave a deeper insight into the evolutionary history and origin of Iranian sheep with important implications for future breed management.

The mechanisms that underpin how insertions or deletions (indels) become fixed in DNA have primarily been ascribed to replication-related and/or double-strand break (DSB)-related processes. Here, we introduce a method to evaluate indels, orientating them relative to gene transcription. In so doing, we reveal a number of surprising findings: First, there is a transcriptional strand asymmetry in the distribution of mononucleotide repeat tracts in the reference human genome. Second, there is a strong transcriptional strand asymmetry of indels across 2,575 whole genome sequenced human cancers. We suggest that this is due to the activity of transcription-coupled nucleotide excision repair (TC-NER). Furthermore, TC-NER interacts with mismatch repair (MMR) under physiological conditions to produce strand bias. Finally, we show how insertions and deletions differ in their dependencies on these repair pathways. Serratia marcescens is an opportunistic pathogen that causes diverse nosocomial infections. S. marcescens has developed considerable resistance to different antibiotics and is equipped with an armory of virulence factors. These virulence factors are regulated in S. marcescens by an intercellular communication system termed quorum sensing (QS). Targeting bacterial virulence and QS is an interesting approach to mitigating bacterial pathogenesis and overcoming the development of resistance to antimicrobials. In this study, we aimed to evaluate the anti-virulence activities of secnidazole on a clinical isolate of S. marcescens. The effects of secnidazole at sub-inhibitory concentrations (sub-MICs) on virulence factors, swarming motility, biofilm formation, proteases, hemolysin activity, and prodigiosin production were evaluated in vitro. Secnidazole’s protective activity against S. marcescens pathogenesis was assessed in vivo in mice. Furthermore, a molecular docking study was conducted to evaluate the binding ability of secnidazole to the S. marcescens SmaR QS receptor. Our findings showed that secnidazole at sub-MICs significantly reduced S. marcescens virulence factor production in vitro and diminished its pathogenesis in mice. The insilico docking study revealed a great ability of secnidazole to competitively hinder the binding of the autoinducer to the SmaR QS receptor. In conclusion, secnidazole is a promising anti-virulence agent that may be used to control infections caused by S. marcescens.

Background Serine/threonine kinase 3 (AKT3) is a protein-coding gene that is associated with several cattle immune diseases including different tumors and cancers. The objective of this study was to investigate the differences in structures and functions of AKT3 of cow and buffalo cattle. Methods The sequence differences of gene-coding sequence (CDS) and core promoter region of AKT3 in cow and buffalo were analyzed by using bioinformatics tools and PCR sequencing. Also, the functional analysis of promoter regulating gene expression by RT-PCR was performed using 500 Holstein cows and buffalos. And, evaluation of AKT3 inflammatory response to the lipopolysaccharide (LPS)-induced mastitis was performed between both species. Results The results revealed the variation in 6 exons out of 13 exons of the two species of CDS. Also, 4 different regions in 3-kb promoters of the AKT3 gene were significantly different between cow and buffalo species, in which cow’s AKT3 promoter sequence region was started from − 371 to − 1247, while in buffalo, the sequence was started from − 371 to − 969 of the promoter crucial region. Thus, the promoter was overexpressed in cows compared to buffaloes. As a result, significant differences (P < 0.05) between the two species in the AKT3 gene expression level related to the LPS stimulation in their mammary epithelial cell line. Conclusions This study emphasized the great importance of the structural differences of AKT3 between the animal species on their different responses against immune diseases like mastitis.

BackgroundMastitis is one of the major diseases causing economic loss to the dairy industry by reducing the quantity and quality of milk. Thus, the objective of this scientific study was to find new biomarkers based on genes for the early prediction before its severity.MethodsIn the present study, advanced bioinformatics including hierarchical clustering, enrichment analysis, active site prediction, epigenetic analysis, functional domain identification, and protein docking were used to analyze the important genes that could be utilized as biomarkers and therapeutic targets for mastitis.ResultsFour differentially expressed genes (DEGs) were identified in different regions of the mammary gland (teat cistern, gland cistern, lobuloalveolar, and Furstenberg’s rosette) that resulted in 453, 597, 577, and 636 DEG, respectively. Also, 101 overlapped genes were found by comparing 27 different expressed genes. These genes were associated with eight immune response pathways including NOD-like receptor signaling pathway (IL8, IL18, IL1B, PYDC1) and chemokine signaling pathway (PTK2, IL8, NCF1, CCR1, HCK). Meanwhile, 241 protein-protein interaction networks were developed among overlapped genes. Fifty-seven regulatory events were found between miRNAs, expressed genes, and the transcription factors (TFs) through micro-RNA and transcription factors (miRNA-DEG-TF) regulatory network. The 3D structure docking model of the expressed genes proteins identified their active sites and the binding ligands that could help in choosing the appropriate feed or treatment for affected animals.ConclusionsThe novelty of the distinguished DEG and their pathways in this study is that they can precisely improve the detection biomarkers and treatments techniques of cows’Escherichia colimastitis disease due to their high affinity with the target site of the mammary gland before appearing the symptoms.Graphical abstract

Background Osteosarcoma (OSA) is a devastating disease that is common in the Irish Wolfhound breed. The aim of this study was to use a pedigree-based approach to determine the heritability of OSA in the Irish Wolfhound using data from a large publically available database. Results The pedigree used for this study included 5110 pure-bred Irish Wolfhounds, including 332 dogs diagnosed with OSA and 360 control dogs; dogs were considered controls if they lived over 10 years of age and were not reported to have developed OSA. The estimated heritability of OSA in the Irish Wolfhound was 0.65. Conclusion The results of this study indicate that OSA in the Irish Wolfhound is highly heritable, and support the need for future research investigating associated genetic mutations., Plain English summary Osteosarcoma is a devastating condition that is prevalent in the Irish Wolfhound breed. A series of 3-benzylideneindolin-2-one compounds was designed and synthesized based on combretastatin A-4 and compound IC261, a dual casein kinase (CK1)/tubulin polymerization inhibitor, taking into consideration the pharmacophore required for EGFR-tyrosine kinase inhibition. The new molecular entities provoked significant growth inhibition against PC-3, MCF-7 and COLO-205 at a 10 μM dose. Compounds 6-chloro-3-(2,4,6-trimethoxybenzylidene) indolin-2-one, 4b, and 5-methoxy-3-(2,4,6-trimethoxybenzylidene)indolin-2-one, 4e, showed potent activity against the colon cancer COLO-205 cell line with an IC50 value of 0.2 and 0.3 μM. A mechanistic study demonstrated 4b’s efficacy in inhibiting microtubule assembly (IC50 = 1.66 ± 0.08 μM) with potential binding to the colchicine binding site (docking study). With an IC50 of 1.92 ± 0.09 μg/mL, 4b inhibited CK1 almost as well as IC261. Additionally, 4b and 4e were effective inhibitors of EGFR-TK with IC50s of 0.19 μg/mL and 0.40 μg/mL compared to Gifitinib (IC50 = 0.05 μg/mL). Apoptosis was induced in COLO-205 cells treated with 4b, with apoptotic markers dysregulated. Caspase 3 levels were elevated to more than three-fold, while Cytochrome C levels were doubled. The cell cycle was arrested in the pre-G1 phase with extensive cellular accumulation in the pre-G1 phase, confirming apoptosis induction. Levels of cell cycle regulating proteins BAX and Bcl-2 were also defective. The binding interaction patterns of these compounds at the colchicine binding site of tubulin and the Gifitinib binding site of EGFR were verified by molecular docking, which adequately matched the reported experimental result. Hence, 4b and 4e are considered promising potent multitarget agents against colon cancer that require optimization.

Three approaches were used and compared in terms of prediction accuracy: (1) whole-genome prediction (WGP) using SNP marker information only, (2) whole-methylome prediction (WMP) using methylation profiles only, and (3) whole-genome/methylome prediction (WGMP) with combining both omics layers. The number of SNP and of methylation sites varied in each scenario, with either 1, 10, or 50% of these preselected based on four approaches: randomly, evenly spaced, lowest p value (genome-wide association or epigenome-wide association study), and estimated effect size using a Bayesian ridge regression (BRR) model. To remove effects of high levels of pairwise linkage disequilibrium (LD), SNPs were also preselected with an LD-pruning method. Five Bayesian regression models were studied for classification, including BRR, Bayes-A, Bayes-B, Bayes-C, and the Bayesian LASSO. Adjusting methylation profiles for cellular heterogeneity within whole blood samples had a detrimental effect on the classification ability of the models. Overall, WGMP using Bayes-B model has the best performance. In particular, selecting SNPs based on LD-pruning with 1% of the methylation sites selected based on BRR included in the model, and fitting the most significant SNP as a fixed effect was the best method for predicting disease risk with a classification accuracy of 0.975. Our results showed that multiomics data can be used to effectively predict the risk of RA and identify cases in early stages to prevent or alter disease progression via appropriate interventions. The F2 and F3 populations resulting from Giza171 × Sids12 hybrid were used from 2015/2016 to 2018/2019 seasons at Sakha Agricultural Research Station to study the inheritance of some agronomic characters and resistance to rusts diseases and select new bread wheat families with high yield potential and good grain quality. Giza171 was preferable for yield and its components, while Sids12 was desirable for earliness characters. The ranges of the F2 and F3 populations went out the means of its parents for the studied characters. All characters showed moderate to high values of broad sense heritability in F2 and F3 generations. The best responsive characters to selection in F2 were No. spikes plant-1 and grain yield plant-1, while days to maturity was the least responsive character. F3 families hadhighly significant variations for all studied characters. Correlation coefficients referred that grain filling rate, followed by No. spikes m-2, then kernels weight spike-1, later the No. kernels spike-1 had the most impact on grain yield in F3 families. The resistance of yellow and leaf rusts was controlled by two dominant genes and stem rust was controlled by two complementary dominant genes. Thirteen promising families were high yielding, resistant to the three rusts and have appropriate height were selected to promote to the next advanced of segregating generations and five of them were preferable for grain quality of wheat.

Pseudomonas aeruginosa is a Gram-negative opportunistic pathogen that causes serious infections in humans, notably cystic fibrosis. P. aeruginosa faces various stresses such as oxidative stress either in the environment or within the host during infection. In the present study, the influence of oxidative stress on both Pseudomonas antibiotic susceptibility and host pathogenesis was characterized. Prior exposure to H2O2 significantly altered P. aeruginosa susceptibility to tested antibiotics; colistin, ciprofloxacin, tobramycin, and ceftazidime. The minimum inhibitory concentrations (MICs) of tested antibiotics either increased or decreased following H2O2 exposure. Importantly, RT-qPCR revealed that expression of quorum sensing genes, that regulate virulence factors production in P. aeruginosa, was significantly higher in unstressed relative to H2O2-stressed cells. The impact of P. aeruginosa exposure to oxidative stress by H2O2 on bacterial pathogenesis was investigated using in vivo mice infection model. Interestingly, exposure to oxidative stress markedly reduces P. aeruginosa pathogenesis in mice. Unstressed P. aeruginosa was able to kill more mice as compared to H2O2-stressed bacteria. In addition, body weight of mice infected with unstressed P. aeruginosa was lower than that of mice inoculated with stressed bacteria. Isolated organs (spleen, liver, and kidney) from mice infected with unstressed bacteria exhibited increased weight as well as bacterial load in comparison with mice infected with stressed bacteria. In summary, current data highlight the impact of oxidative stress on P. aeruginosa antibiotic susceptibility as well as host pathogenesis. These findings could be helpful in treatment of infections caused by this important pathogen.

Background and Aim: Houseflies (Musca domestica) are synanthropic insects which serve as biological or mechanical vectors for spreading multidrug-resistant bacteria responsible for many infectious diseases. This study aimed to detect antibiotic-resistant bacteria from houseflies, and to examine their resistance genes. Materials and Methods: A total of 140 houseflies were captured using sterile nylon net from seven places of Mymensingh city, Bangladesh. Immediately after collection, flies were transferred to a sterile zipper bag and brought to microbiology laboratory within 1 h. Three bacterial species were isolated from houseflies, based on cultural and molecular tests. After that, the isolates were subjected to antimicrobial susceptibility testing against commonly used antibiotics, by the disk diffusion method. Finally, the detection of antibiotic resistance genes tetA, tetB, mcr-3, mecA, and mecC was performed by a polymerase chain reaction. Results: The most common isolates were Staphylococcus aureus (78.6%), Salmonella spp., (66.4%), and Escherichia coli (51.4%). These species of bacteria were recovered from 78.3% of isolates from the Mymensingh Medical College Hospital areas. Most of the isolates of the three bacterial species were resistant to erythromycin, tetracycline, penicillin and amoxicillin and were sensitive to ciprofloxacin, ceftriaxone, chloramphenicol, gentamicin, and azithromycin. Five antibiotic resistance genes of three bacteria were detected: tetA, tetB, mcr-3, and mecA were found in 37%, 20%, 20%, and 14% isolates, respectively, and no isolates were positive for mecC gene. Conclusion: S. aureus, Salmonella spp., and E. coli with genetically-mediated multiple antibiotic resistance are carried in houseflies in the Mymensingh region. Flies may, therefore, represent an important means of transmission of these antibiotic-resistant bacteria, with consequent risks to human and animal health.

Background and Aim: Highly pathogenic avian influenza (HPAI) is a deadly virus of zoonotic potential. The study mainly aims to determine the risk pathways (RPs) for the probable incursion of HPAI virus (HPAIV) in backyard poultry in Bangladesh. Materials and Methods: The study involves expert elicitation technique. The concept map determines the possible RPs. The map consists of 16 concepts, each with nodes from which probabilities of an event originates. These probabilities are described by qualitative descriptors ranging from negligible to high. Risk assessment has been performed using the subjective risk assessment tool. Results: The tool demonstrates positive correlation among groups of experts in the level of agreement by scoring RP; however, the level of agreement varies from 71% to 93% among group of experts. The median risk score of viral incursion through the "Exposure of backyard poultry with farm poultry in the trading market" was 11 and ranked as top, followed by "Contaminated live bird market environment" and "Sharing common scavenging space with migratory birds" (median risk score, 10.5; rank, 2), and "Scavenging of infected slaughtered poultry remnants by backyard poultry" (median risk score, 5.3; rank, 3) when no control options were applied along with the RPs. After applying or considering control option along with contaminated live bird market environment, the median risk score was reduced to 5.0. Applying a specific control option along with each RP reduced estimated median risk scores for HPAIV incursions. Conclusion: This study provides an insight into the incursion risks of HPAIV through various RPs in backyard poultry in Bangladesh.

The predatory mites Neoseiulus barkeri, Amblyseius swirskii and Cydnoseius negevi are native phytoseiid mites in Egypt. The biology of these predators was studied using Achroia grisella eggs as food source. Tested phytoseiids were succeeded to develop to adulthood and sustain oviposition on A. grisella eggs. For Cydnoseius negevi, the development was slower and fecundity was lower than for Neoseiulus barkeri and Amblyseius swirskii. The predation rate during immature stages of Cydnoseius negevi was higher than those of Neoseiulus barkeri and Amblyseius swirskii. During oviposition period, N. barkeri consumed more eggs than other phytoseiids did. The higher mean fecundity was recorded for Neoseiulus barkeri and Amblyseius swirskii as opposed to the lower mean for Cydnoseius negevi. When Amblyseius swirskii fed on Achroia grisella eggs, the oviposition period, female longevity and mean generation time were longer than for Cydnoseius negevi and Neoseiulus barkeri. Feeding on eggs of Achroia grisella resulted in the lowest mean fecundity, net reproductive rate, intrinsic rate of natural increase, finite rate of increase and gross reproductive rate for Cydnoseius negevi. The sex ratio of all predators' progeny was in female-biased. Consequently, the moth egg was alternative food for Neoseiulus barkeri and Amblyseius swirskii, while it considers as survival prey for Cydnoseius negevi.

Summary: Heart failure (HF) and subarachnoid hemorrhage (SAH) chronically reduce cerebral perfusion, which negatively affects clinical outcome. This work demonstrates a strong relationship between cerebral artery cystic fibrosis transmembrane conductance regulator (CFTR) expression and altered cerebrovascular reactivity in HF and SAH. In HF and SAH, CFTR corrector compounds (C18 or lumacaftor) normalize pathological alterations in cerebral artery CFTR expression, vascular reactivity, and cerebral perfusion, without affecting systemic hemodynamic parameters. This normalization correlates with reduced neuronal injury. Therefore, CFTR therapeutics have emerged as valuable clinical tools to manage cerebrovascular dysfunction, impaired cerebral perfusion, and neuronal injury. Alzheimer’s disease (AD) is a multifactorial and fatal neurodegenerative disorder. Memory loss and cognitive decline occur due to death of brain cells. Various important hallmarks of AD have reported like deposition of β-amyloid fibril, β-amyloid oligomers formation, hyperactive phosphorylated tau protein, oxidative stress in cell, low levels of acetylcholine, etc. Treatment of AD based on cholinesterase inhibitors is only symptomatic, its efficacy is limited. A multi-targeted ligand may enable therapeutic efficacy, because of being multifactorial nature of AD. Hence, this research has been focused on developing novel components that preferentially block cholinesterase and simultaneously bind with other targets like β-secretase, Monoamine oxidases, Glycogen synthase kinase, etc., which are directly or indirectly associated with AD to offer more efficient treatment than earlier. To select novel targets and therapeutic ligands, computational approaches have proved to be robust and reliable tools. To expose intermolecular binding mode of the compounds, molecular docking studies and molecular dynamics simulation studies performed and the results indicate their substantial interactions with the active sites of AChE and BCHE and other responsible targets. In silico ADME/T, study performed to estimate several pharmacokinetic parameters and toxicity profile of the selected compounds. Amongst the series, compounds 2-(2,2-dimethylchromen-6-yl)-5,7- dihydroxychromen-4-one (PCID5315395) and 7-(1,3-benzodioxol-5-yl)-5-hydroxy-2,2-dimethylpyrano [3,2-g] chromen-6-one (PCID5983661) are the most encouraging multi-targeted candidates which have the ability to increase memory, acetylcholine as well as other neurotransmitter levels and give the protection of the neurons against the cognitive deficit. In this study, we are proposing two new compounds from PubChem database as AChE as well as BCHE, MAO-A, MAO-B, Beta-secretase, GSK-3 and N-Methyl-D-aspartate (NMDA) inhibitors for further investigation and experimental validation.

The present study is aimed at investigating clinical poultry diseases in an Upazila Government Veterinary Hospital of Bangladesh through passive surveillance and to know the frequency distribution of antimicrobial drugs prescription pattern. The study was conducted in Ramu Upazila Government Veterinary Hospital under Cox’s Bazar district of Bangladesh from February to March, 2016. A total of 180 cases including 73 broiler, 84 layer, 18 duck and 5 pigeon were included in this study. Diseases were diagnosed based on clinical history, clinical signs, post mortem lesions and rapid kit test. The most prevalent diseases were Newcastle disease (ND) 17.8% (95% CI 12-24%), colibacillosis 11.5% (95% CI 6-17%), infectious bursal disease (IBD) 10.8% (95% CI 6-16%) and aspergillosis 9.6% (95% CI 5-14%) in broiler and layer chickens; duck plague 69.6% (95% CI 49-90%) in duck and pigeon pox 13% (95% CI 0.2-24%) in pigeon. The major study of this work was to evaluate the chemical composition and the inhibitory components of Parmotrema perlatum against the clinically isolated specimens of Staphylococcus, Streptococcus, Escherichia, Pseudomonas and Candida.. The GC-FID-MS by using a Trace GC Ultra apparatus MS DSQ II detectors and FID-MS splitter under the operating conditions of apolar capillary column was employed to determine the chemical composition of Parmotrema perlatum essential oil. Amino acid (38.1%) and cymene (29.1%) were the most adequate constituents in this oil. The antibacterial activity of Parmotrema perlatum oil was tested against standard strains of bacteria and clinical strains isolated from patients with infections of the oral cavity, abdominal cavity, respiratory, genitourinary tracts, skin, and from the hospital environment. Kirby-Bauer disc diffusion technique was employed to determine the susceptibility of the organism towards the Parmotrema perlatum essential oil exhibited very strong antimicrobial activity to inhibit the growth of the clinical specimens. Application of natural antimicrobial based essential oil from Parmotrema perlatum can be potential source in the treatment & prevention of different types of infections as an alternative to the chemical - based antibiotics based on the investigation.

Pseudomonas aeruginosa is an opportunistic pathogen exhibiting higher resistance to commonly used antibiotics. Microbial resistance to antibiotics is a major problem that hinders attempts to control microbial infections. Quorum sensing inhibitors could help us solve such problem by repressing quorum sensing that controls the production of virulence factors in many pathogens including P. aeruginosa. In this study, the influence of tenoxicam, a non-steroidal anti-inflammatory drug, on quorum sensing in P. aeruginosa was characterized. Treatment of P. aeruginosa with tenoxicam decreased production of many virulence factors such as pyoverdin, rhamnolipids, pyocyanin, elastase, proteases, and hemolysins. Moreover, qRT-PCR revealed a significant reduction in expression of quorum sensing genes in tenoxicam-treated P. aeruginosa in comparison with untreated bacteria. Tenoxicam markedly reduced the capacity of P. aeruginosa to kill mice infection model. Mice injected with tenoxicam-treated P. aeruginosa exhibited higher survival rate as compared with those inoculated with untreated bacteria. Current data clearly demonstrate that tenoxicam has quorum sensing inhibitory effect on P. aeruginosa. Tenoxicam could play a role in reduction of Pseudomonas quorum sensing-dependant virulence factors production, and therefore affect its pathogenesis in the host. In summary, the current study suggests that tenoxicam could be used as adjuvant to antibiotics in the management of diseases caused by P. aeruginosa.

Background A 32-base pair deletion (∆32) in the open reading frame (ORF) of C-C motif chemokine receptor 5 (CCR5) seems to be a protective variant against immune system diseases, especially human immunodeficiency virus type 1 (HIV-1). We aimed to assess the frequency of CCR5∆32 in the healthy Iranian population. Methods In this study, 400 normal samples from Khorasan, northeastern Iran, were randomly selected. The frequency of CCR5∆32 carriers was investigated using PCR analysis. Allele prevalence and the fit to the Hardy-Weinberg equilibrium were analyzed. Results The prevalence of CCR5∆32 in the northeastern population of Iran was 0.016. Four hundred samples were studied, among which one with CCR5∆32/∆32 and 11 with CCR5Wild/∆32 genotype were detected. Conclusion This study was the first investigation for an assessment of the prevalence of CCR5∆32 in northeastern Iran. The low prevalence of CCR5∆32 allele in the Iranian population may result in the increased susceptibility to HIV-1. In addition, this prevalence is the same as that of reported in East Asia, while is lower than that in the Europeans.
