Current literature related to the impact of probiotics on the incidence of gastrointestinal tract infections (GITIs) has shown mixed results and no systematic review available with pooled analysis exists. Thus, the aim of this systematic review was to provide contemporary evidence regarding the overall and strain-specific influence of probiotics in preventing GITIs among infants and children attending childcare centres. The review shortlisted 18 RCTs after screening through the initial search results of 779 articles. However, only 15 trials were deemed eligible, addressing at least one outcome in the pooled analysis. It is concluded that the supplementation of probiotics (overall effect) may reduce the risk of GITI episode by 26%, with Lacticaseibacillus paracasei, Limosilactobacillus reuteri and Lacticaseibacillus rhamnosus GG being specifically potent probiotic strains in reducing GITI episode, duration of infection and absence from childcare respectively. There is insufficient evidence to determine the effect of Bifidobacterium animalis subsp. lactis BB-12 based on the findings of the trials included in this review.

This study was performed to determine the effects of bacteriocin-producing and non-bacteriocin-producing Pediococcus acidilactici strains on the immune system and intestinal flora of normal mice. Two P. acidilactici strains with antibacterial activity (P. acidilactici CCFM28 and CCFM18) were obtained based on the inhibition-zone assay. The produced components were identified as bacteriocins through protease treatment, pH adjustment and hydrogen peroxide treatment. Bacteriocin-producing and non-bacteriocin-producing P. acidilactici strains (P. acidilactici CCFM28, CCFM18 and NT17-3) caused significant changes in serum immune factors and intestinal flora of normal mice. After 14 days of intervention, the relative abundance of Firmicutes was significantly decreased, but that of Proteobacteria was significantly increased at the phylum level. At the genus level, the administration of three P. acidilactici strains resulted in the downregulation of Blautia and the upregulation of Ruminococcus and Lactobacillus. Furthermore, there were also different regulations on some probiotic strains, such as Bifidobacterium, Coprococcus and Akkermansia, which were closely related to the antibacterial ability of the bacteriocin and the type of strain. The results indicated that the intervention of different P. acidilactici strains could differently change the structure of intestinal flora in normal mice, which provided theoretical guidance for the selective use of bacteriocin-producing strains for health regulation in the future.

Long-term sleep deprivation (SD) is a bad lifestyle habit, especially among specific occupational practitioners, characterized by circadian rhythm misalignment and abnormal sleep/wake cycles. SD is closely associated with an increased risk of metabolic disturbance, particularly obesity and insulin resistance. The incretin hormone, glucagon-like peptide-1 (GLP-1), is a critical insulin release determinant secreted by the intestinal L-cell upon food intake. Besides, the gut microbiota participates in metabolic homeostasis and regulates GLP-1 release in a circadian rhythm manner. As a commonly recognized intestinal probiotic, Bifidobacterium has various clinical indications regarding its curative effect. However, few studies have investigated the effect of Bifidobacterium supplementation on sleep disorders. In the present study, we explored the impact of long-term SD on the endocrine metabolism of rhesus monkeys and determined the effect of Bifidobacterium supplementation on the SD-induced metabolic status. Lipids concentrations, body weight, fast blood glucose, and insulin levels increased after SD. Furthermore, after two months of long-term SD, the intravenous glucose tolerance test (iVGTT) showed that the glucose metabolism was impaired and the insulin sensitivity decreased. Moreover, one month of Bifidobacterium oral administration significantly reduced blood glucose and attenuated insulin resistance in rhesus macaques. Overall, our results suggested that Bifidobacterium might be used to alleviate SD-induced aberrant glucose metabolism and improve insulin resistance. Also, it might help in better understanding the mechanisms governing the beneficial effects of Bifidobacterium.

Background::Cancer is a serious problem throughout the world. The pathophysiology of cancer is multifactorial and is also related to gut microbiota. Intestinal microbes are the useful resident of the healthy human. They play various aspects of human health including nutritional biotransformation, flushing of the pathogens, toxin neutralization, immune response, and onco-suppression. Disruption in the interactions among the gut microbiota, intestinal epithelium, and the host immune system are associated with gastrointestinal disorders, neurodegenerative diseases, metabolic syndrome, and cancer. Probiotic bacteria (Lactobacillus spp., Bifidobacterium spp.) have been regarded as beneficial to health and shown to play a significant role in immunomodulation and displayed preventive role against obesity, diabetes, liver disease, inflammatory bowel disease, tumor progression, and cancer.Objective::The involvement of gut microorganisms in cancer development and prevention has been recognized as a balancing factor. The events of dysbiosis emphasize metabolic disorder and carcinogenesis. The gut flora potentiates immunomodulation and minimizes the limitations of usual chemotherapy. The significant role of prebiotics and probiotics on the improvement of immunomodulation and antitumor properties has been considered.Methods::I had reviewed the literature on the multidimensional activities of prebiotics and probiotics from the NCBI website database PubMed, Springer Nature, Science Direct (Elsevier), Google Scholar database to search relevant articles. Specifically, I had focused on the role of prebiotics and probiotics in immunomodulation and cancer prevention.Results::Prebiotics are the nondigestible fermentable sugars that selectively influence the growth of probiotic organisms that exert immunomodulation over the cancerous growth. The oncostatic properties of bacteria are mediated through the recruitment of cytotoxic T cells, natural killer cells, and oxidative stress-induced apoptosis in the tumor microenvironment. Moreover, approaches have also been taken to use probiotics as an adjuvant in cancer therapy.Conclusion::The present review has indicated that dysbiosis is the crucial factor in many pathological situations including cancer. Applications of prebiotics and probiotics exhibit the immune-surveillance as oncostatic effects. These events increase the possibilities of new therapeutic strategies for cancer prevention.

Background & Aims The tumor microbiome of patients with pancreas ductal adenocarcinoma (PDAC) includes bacteria normally present in the upper gastrointestinal tract. If the predominant source of intratumoral bacteria in patients with PDAC is retrograde migration from the duodenum, duodenal fluid could be a representative biospecimen for determining microbiome profiles of patients with PDAC or at risk of developing PDAC. Methods We performed a case-control study comparing bacterial and fungal (16S and 18S rRNA) profiles of secretin-stimulated duodenal fluid collections from 308 patients undergoing duodenal endoscopy including 134 normal pancreas control subjects, 98 patients with pancreatic cyst(s) and 74 patients with PDAC. Results Alterations in duodenal fluid microbiomes with diminished alpha diversity were significantly associated with age >70 and proton pump inhibitor use. Patients with PDAC had significantly decreased duodenal microbial alpha diversity compared with age-matched control subjects with normal pancreata and those with pancreatic cyst(s). There was evidence of enrichment of Bifidobacterium genera in the duodenal fluid of patients with PDAC compared with control subjects and those with pancreatic cyst(s). There were also enrichment of duodenal fluid Fusobacteria and Rothia bacteria among patients with PDAC with short-term survival. Duodenal fluid microbiome profiles were not significantly different between control subjects and patients with pancreatic cyst(s). Conclusion Patients with PDAC have alterations in their duodenal fluid microbiome profiles compared with patients with pancreatic cysts and those with normal pancreata. Bifidobacteria are widely used as a probiotic for their health-promoting effects. To promote their growth, bifidogenic prebiotics, including human milk oligosaccharides (HMOs), have been added to supplements and infant formula. However, the efficacy of both probiotic and prebiotic interventions is often debated, as clinical responses vary significantly by case. Here, we review clinical studies that aimed to proliferate human-residential Bifidobacterium (HRB) strains in the gut, and we highlight the difference between responders and non-responders to such interventions through an ecological, niche-based perspective and an examination of the prevalence of genes responsible for prebiotic assimilation in HRB genomes. We discuss the criteria necessary to better evaluate the efficacy of probiotic and prebiotic interventions and the recent therapeutic potential shown by synbiotics.

Introduction The roles of Bacteroides species in alleviating inflammation and intestinal injury has been widely demonstrated, but few studies have focused on the roles of Bacteroides vulgatus. Objectives In this study, four B. vulgatus strains were selected, based on their genomic characteristics, to assess their ability to alleviate lipopolysaccharide (LPS)-induced acute intestinal injury in C57BL/6J mice. Methods Alterations in the intestinal microbiota, intestinal epithelial permeability, cytokine level, short-chain fatty acid (SCFA) concentration, and immune responses were investigated following LPS-induced acute intestinal injury in C57BL/6J mice. Results Severe histological damage and a significant change in cytokine expression was observed in the mouse colon tissues 24 h after LPS administration. Oral administration of different B. vulgatus strains showed different effects on the assessed parameters of the mice; particularly, only the administration of B. vulgatus FTJS7K1 was able to protect the architectural integrity of the intestinal epithelium. B. vulgatus FTJS7K1 also negated the LPS-induced changes in cytokine mRNA expression in the colon tissues, and in the proportion of regulatory T cells in the mesenteric lymph node. Compared with the LPS group, the B. vulgatus FTJS7K1 group showed significantly increased abundance of Lactobacillus, Akkermansia, and Bifidobacterium, and decreased abundance of Faecalibaculum. The B. vulgatus FTJS7K1 group also showed significantly increased concentration of SCFAs in fecal samples. The results of genomic analysis showed that these protective roles of B. vulgatus FTJS7K1 may be mediated through specific genes associated with defense mechanisms and metabolism (e.g., the secretion of SCFAs). Conclusions Our findings suggest that the protective role of B. vulgatus FTJS7K1 appear to be via modulation of cytokine production in the colon tissue and regulation of the structure of the gut microbiota. These results provide support for the screening of the Bacteroides genus for next-generation probiotics.

BACKGROUND: Type 2 diabetes as the most prevalent metabolic disorder, is one of the major causes of morbidity and mortality worldwide. Recent studies suggest that body microbiota may play a role in developing metabolic disorders including type 2 diabetes. The objective of the present study was to investigate the blood microbiota composition in Iranian pre-diabetic and type 2 diabetic patients compared to healthy individuals. METHODS: Blood samples were taken after 12-h fasting from 90 participants, 30 healthy individuals, 30 type 2 diabetes patients and 30 pre-diabetic participants. The buffy coat layer separated by centrifugation at 800 and DNA was extracted using a column-based method. Composition and load of blood microbiota was evaluated by real-time PCR method using genus specific 16S rRNA primers. RESULTS: The load of Akkermansia, and Faecalibacterium was higher in normal volunteers compared to pre-diabetic and type 2 diabetes group (p< 0.05). The load of Bifidobacterium was higher in normal volunteers compared to type 2 diabetes patients (p= 0.02). In contrast, the load of Lactobacillus and Escherichia coli was higher in pre-diabetics and type 2 diabetes patients compared to normal volunteers (p< 0.05). The load of Bacteroides fragilis was not statistically different between studied groups but it was higher in males compared to female group (p= 0.04). the load of other bacteria was not significantly different between male and female participants. CONCLUSION: There is difference between microbiota composition in white blood cells of pre-diabetic and type 2 diabetes patients compared to healthy people. Determination of blood microbiota pattern may have a role in diagnosis and preventive of type 2 diabetes in a certain population. For more clarification about correlation between blood microbiota and type 2 diabetes, larger studies with more participants in different ethnical populations is suggested.

This study investigated the effects of mung bean protein (MPI) and a MPI-polyphenol complex on oxidative stress levels and intestinal microflora in a D-galactose-induced aging mouse model. MPI and MPI-polyphenol complex intervention significantly increased activity of superoxide dismutase (SOD) and catalase and other antioxidant enzymes, improved the abundance and diversity of intestinal flora, and decreased the Firmicutes to Bacteroidetes ratio. The addition of MPI and MPI-polyphenol complex can help the proliferation of Bacteroidetes, Bifidobacterium and Roseburia in the intestinal tract of aging mice, and inhibit the growth of Firmicutes and Ruminococcus. MPI significantly upregulated five pathways related to lipid and energy metabolism. Roseburia and Muribaculaceae were negatively correlated with malondialdehyde levels and positively correlated with SOD and other antioxidant enzyme indices. Our findings showed that MPI and MPI-polyphenol complexes can delay aging in mice by reducing oxidative damage and regulating intestinal flora. We also found a strong relationship between the abundance of intestinal flora and the levels of oxidative stress-related enzymes. This study provides theoretical support for the health and anti-aging benefits of mung bean food products.

Torreya grandis cv. Merrillii (Taxace, Torreya) is mainly distributed across the hilly areas of subtropical China and is well known for its nutritional value. In this study, the ameliorative effects of T. grandis seed oil on lipid metabolism were investigated, and the underlying mechanism was explored from the perspective of gut microbiota. Mice experiments showed that the rate of body mass gain in the group where the mice were fed a high-fat diet (HFD) and supplemented with 550 mg/(kg·day) T. grandis seed oil (HFD + TO550 group) was 42.27%, while it was 62.25% in the HFD group. Compared with the HFD group, the liver and fat indices, total cholesterol, triglycerides, and low-density lipoprotein cholesterol were reduced in the oil-supplement groups. Moreover, the oil supplement significantly changed the fatty acid composition and alleviated pathological damage to the liver caused by the high-fat diet. Additionally, the distinct clustering of bacteria in the composition of gut microbiota was observed in the oil treatment group compared with that in the HFD group. T. grandis seed oil significantly increased the abundance of the beneficial bacteria and short-chain fatty acid producers, including Lactobacillus, Bifidobacterium, Faecalibaculum and Allobaculum. Our results suggest that the supplements of T. grandis seed oil could alleviate hyperlipidemia caused by HFD. These positive effects are considered to be related with sciadonic acid (SCA) and are partially mediated by alterations in gut microbiota composition and functionality.

Type 2 diabetes mellitus (T2DM) is known as a complex genetic disease characterized by genetic and environmental factors. The imbalanced intestinal flora and intestinal mucosal barrier are considered to be related to T2DM. Curcumin has been proved to affect the progression of T2DM. T2DM animal was established by low-dose streptozotocin intraperitoneal injection combined with high-fat diet (HFD) feeding. Hematoxylin and eosin (HE) staining and transfer electron microscopy (TEM) were used to observe morphological changes of intestinal tissues of T2DM rats. Insulin and glucose tolerance tests were performed to investigate the influence of curcumin on blood glucose. Curcumin significantly improved the intestinal integrity, hyperglycemia and insulin resistance in diabetic rats. The metabolic endotoxemia induced by HFD in diabetic rats was inhibited remarkably. Curcumin reversed gut microbiota dysbiosis in diabetic rats caused by HFD. We demonstrated that curcumin could protect intestinal mucosal barrier, improve insulin resistance and reduce blood glucose in diabetic rats. This study might provide experimental evidence for the prevention and treatment in T2DM.

Much evidence suggests a role for human milk oligosaccharides (HMOs) in establishing the infant microbiota in the large intestine, but the response of particular bacteria to individual HMOs is not well known. Here twelve bacterial strains belonging to the genera Bifidobacterium, Enterococcus, Limosilactobacillus, Lactobacillus, Lacticaseibacillus, Staphylococcus and Streptococcus were isolated from infant faeces and their growth was analyzed in the presence of the major HMOs, 2'-fucosyllactose (2'FL), 3-fucosyllactose (3FL), 2',3-difucosyllactose (DFL), lacto-N-tetraose (LNT) and lacto-N-neo-tetraose (LNnT), present in human milk. Only the isolated Bifidobacterium strains demonstrated the capability to utilize these HMOs as carbon sources. Bifidobacterium infantis Y538 efficiently consumed all tested HMOs. Contrarily, Bifidobacterium dentium strains Y510 and Y521 just metabolized LNT and LNnT. Both tetra-saccharides are hydrolyzed into galactose and lacto-N-triose (LNTII) by B. dentium. Interestingly, this species consumed only the galactose moiety during growth on LNT or LNnT, and excreted the LNTII moiety. Two β-galactosidases were characterized from B. dentium Y510, Bdg42A showed the highest activity towards LNT, hydrolyzing it into galactose and LNTII, and Bdg2A towards lactose, degrading efficiently also 6'-galactopyranosyl-N-acetylglucosamine, N-acetyl-lactosamine and LNnT. Background Gut microbiota dysbiosis and sarcopenia commonly occur in the elderly. Although the concept of the gut–muscle axis has been raised, the casual relationship is still unclear. This systematic review analyses the current evidence of gut microbiota effects on muscle/sarcopenia. Methods A systematic review was performed in PubMed, Embase, Web of Science, and The Cochrane Library databases using the keywords (microbiota* OR microbiome*) AND (sarcopen* OR muscle). Studies reporting the alterations of gut microbiota and muscle/physical performance were analysed. Results A total of 26 pre‐clinical and 10 clinical studies were included. For animal studies, three revealed age‐related changes and relationships between gut microbiota and muscle. Three studies focused on muscle characteristics of germ‐free mice. Seventy‐five per cent of eight faecal microbiota transplantation studies showed that the recipient mice successfully replicated the muscle phenotype of donors. There were positive effects on muscle from seven probiotics, two prebiotics, and short‐chain fatty acids (SCFAs). Ten studies investigated on other dietary supplements, antibiotics, exercise, and food withdrawal that affected both muscle and gut microbiota. Twelve studies explored the potential mechanisms of the gut–muscle axis. For clinical studies, 6 studies recruited 676 elderly people (72.8 ± 5.6 years, 57.8% female), while 4 studies focused on 244 young adults (29.7 ± 7.8 years, 55.4% female). The associations of gut microbiota and muscle had been shown in four observational studies. Probiotics, prebiotics, synbiotics, fermented milk, caloric restriction, and exercise in six studies displayed inconsistent effects on muscle mass, function, and gut microbiota. Conclusions Altering the gut microbiota through bacteria depletion, faecal transplantation, and various supplements was shown to directly affect muscle phenotypes. Probiotics, prebiotics, SCFAs, and bacterial products are potential novel therapies to enhance muscle mass and physical performance. Lactobacillus and Bifidobacterium strains restored age‐related muscle loss. Potential mechanisms of microbiome modulating muscle mainly include protein, energy, lipid, and glucose metabolism, inflammation level, neuromuscular junction, and mitochondrial function. The role of the gut microbiota in the development of muscle loss during aging is a crucial area that requires further studies for translation to patients.

The fungus Sporidiobolus pararoseus not only produces carotenoids, but also produces bioactive extracellular polysaccharides (SPP). However, the relationship between SPP and the metabolism of gut microbiome is unclear. The aim of this study was to investigate the mechanism of SPP regulating intestinal health in vivo and in vitro. Results showed that SPP are nondigestible polysaccharides after the digestion with simulated stomach and small intestinal juice in vitro. After SPP was cultured in an in vitro intestinal simulation system for seven days, the concentration of short-chain fatty acids (SCFAs) increased; the microbial diversity changed; the relative abundance of Bifidobacterium and Streptococcus increased; and that of Escherichia Shigella and Lachnospiraceae NK4A136 decreased. In addition, metabolism of SPP by the mice colonic microbiome showed SPP decreased the relative abundance of Firmicutes and Bacteroidota, while the relative abundance of Verrucomicrobiota, Desulfobacterota, and Actinobacteriota increased. Finally, predicted Kyoto Encyclopedia of Genes and Genomes (KEGG) metabolism results also showed that SPP can enhance the metabolism of cofactors, vitamins, amino acids, starch, and sucrose. In conclusion, SPP can multiply the intestinal beneficial bacteria of humans and mice, promote the production of SCFAs and metabolism of amino acids, and promote intestinal health.

Purpose The development of probiotics has seen tremendous growth over the years, with health benefits ranging from gut health to respiratory. We thus aimed to investigate the effects of probiotic Bifidobacterium lactis Probio-M8 (2 × 1010 log CFU/day) against acute respiratory tract infections (RTI), use of antibiotics, hospitalization period and elucidate the possible mechanisms of action in hospitalized young children. Method A prospective, randomized, double-blind and placebo-controlled study was performed in RTI-hospitalized children. Patients were randomized to either the probiotic (n = 60, mean age 13.81 ± 0.90 months) or placebo (n = 60, mean age 12.11 ± 0.73 months) which were administered upon admission, continued during hospitalization and 4-week post-discharged. RTI and gut health parameters were assessed at these time points using validated questionnaires while concentrations of inflammatory cytokines were assessed via oral swabs. To evaluate the administration of Bifidobacterium animalis subsp. lactis, BB-12® (BB-12) on infant colic in breastfed infants, a double-blind, placebo-controlled randomised study was conducted in Chengdu, China from April 2016 to October 2017 with 192 full-term infants less than 3 months of age and meeting the ROME III criteria for infant colic. After a 1-week run-in the infants were randomly assigned to receive daily BB-12 (1×109 cfu/day) or placebo for 3 weeks. Crying/fussing time were recorded using a 24 h structured diary. The primary endpoint was the proportion of infants achieving a reduction in crying and fussing time of ≥50% from baseline. Parent’s/caregiver’s health related quality of life was measured using a modified PedsQL™ 2.0 Family Impact Module and immunological biomarkers were evaluated from faecal samples at baseline and after the 21-day intervention. Objectives Evidence for the contribution of the brain-gut-microbiota axis to the depression pathophysiology is increasing nowadays. Disturbed gut microbiota equilibrium along with bad dietary habits both lead to kynurenine pathway abnormalities contributing to the depression pathophysiology. In this respect, many studies are found but the interventional clinical trials are limited. The present interventional study aims to evaluate the impact of Bifidobacterium spp. supplementation together with improving dietary intake on depressive mood and well-being and their correlation with kynurenine blood level in adult Egyptian healthy volunteers. Methods A number of 98 healthy female volunteers with a mean age of 46.96 ± 1.82 years were selected and enrolled in this study. They were given yogurt enriched with Bifidobacterium spp. daily for eight weeks. Clinical examination as well as questionnaires for the evaluation of psychological well-being and depression were done at base line and after eight weeks of intervention. Fasting blood samples and stool samples were collected from all subjects at baseline and eight weeks after the intervention for the investigation of serum kynurenine concentration, blood hemoglobin, serum transaminases (ALT & AST) serum urea and creatinine as well as fecal Bifidobacterium count. Results Data revealed that both depression and well-being showed highly significant improvement combined with significant drop in kynurenine blood level after intervention. Also, a significant rise in fecal Bifidobacterium count and a significant improvement in hemoglobin level and activity of liver enzymes were recorded. After intervention, a significant negative correlation was recorded between depression and fecal Bifidobacterium count as well as between serum kynurenine level, and well-being. Conclusion Bifidobacterium spp. supplementation combined with improvement in dietary intake resulted in improvement of depressive mood and well-being and reduced kynurenine blood level.

Oral microbiota can alter cancer susceptibility and progression by modulating metabolism and inflammation. We assessed the association between the oral microbiome and lymph node (LN) metastasis in oral squamous cell carcinoma (OSCC). We collected a total of 54 saliva samples from patients with OSCC before surgery. LN metastasis was assessed based on postoperative pathological examination. We used QIIME2, linear discriminant analysis effect size (LEfSe), and PICRUSt2 methods to analyze microbial dysbiosis. A random forest classifier was used to assess whether the oral microbiome could predict LN metastasis. Among the 54 OSCC samples, 20 had LN metastasis, and 34 had no evidence of metastasis. There was a significant difference in β-diversity between the metastasis and no metastasis groups. Through LEfSe analysis, the metastasis group was enriched in the genera Prevotella, Stomatobaculum, Bifidobacterium, Peptostreptococcaceae, Shuttleworthia and Finegoldia. Pathways related to signal peptidase II were predominant in the no metastasis group. The RF model showed a modestly high accuracy for predicting metastasis. Differences in microbial community composition and functions were observed in the oral microbiome of patients with OSCC with and without LN metastasis. However, the finding that specific taxa may be associated with LN metastasis should be verified in a further prospective study.

Background Low dietary fiber intake has been shown to disturb the gut microbiome community, damage the mucus barrier, and promote pathogen susceptibility. However, little is known about the temporal response of the gut microbiome to dietary fiber deprivation and the recovery induced by dietary fiber inclusion in pigs. Objective In the present study, temporal responses of ileal and fecal microbiota to dietary fiber deprivation were profiled using an ileum cannulated growing pig model. In addition, the potential of dietary-resistant starch, β-glucan, and xylan to alleviate gut dysbiosis throughout the gastrointestinal tract, as well as its possible mechanisms were investigated. Methods Six cannulated growing pigs were fed a fiber deprivation diet for 35 days. Ileal digesta and feces were collected at days 0, 7, 21, and 35 for 16S rRNA sequencing and short-chain fatty acid (SCFA) determination. Another twenty-four healthy growing pigs were assigned to one of four dietary treatments including (1) fiber-free diet, (2) resistant starch diet, (3) β-glucan diet, and (4) xylan diet. These twenty-four pigs were fed a corresponding diet for 35 days and slaughtered. Gut microbiome and SCFA concentration were profiled along the gastrointestinal tract. Results Dietary fiber deprivation-induced consistent microbiota extinction, mainly Bifidobacterium and Lactobacillus, and decreased SCFA concentrations in both ileum and feces. The community structure partially recovered at day 35 compared with baseline while SCFA concentrations remained low. Xylan supplementation alleviated gut dysbiosis by selectively promoting Bifidobacterium pseudocatenulatum within the large intestine. SCFA concentration increased significantly after xylan supplementation and exhibited a positive association with B. pseudocatenulatum abundance. An elevated abundance of xylan degradation-related enzyme genes was also observed in the gut microbiome after xylan supplementation. In vitro growth assay further verified the xylan utilization capacity of B. pseudocatenulatum. Conclusions Dietary fiber deprivation could induce probiotic extinction and loss of the SCFA production while potential pathogen was promoted. Xylan intervention could partially restore dietary fiber deprivation-induced gut dysbiosis through selectively promoting B. pseudocatenulatum and therefore normalizing the gut environment. These findings collectively provide evidence that dietary fiber-driven microbiota metabolism bridges the interplay between microbiome and gut health.

Background Fructo-oligosaccharides (FOS), inulin, and galacto-oligosaccharides (GOS) are widely recognized prebiotics that profoundly affect the intestinal microbiota, including stimulation of bifidobacteria and lactobacilli, and are reported to elicit several health benefits. The combination of dietary FOS and inulin with calcium phosphate was reported to stimulate commensal Lactobacillus populations and protect the host against pathogenic Enterobacteriaceae, but little is known about the effects of GOS in diets with a different level of calcium phosphate. Methods We investigated the microbiome changes elicited by dietary supplementation with GOS or inulin using diets with high (100 mmol/kg) and low (30 mmol/kg) calcium phosphate levels in adult Wistar rats. Rats were acclimatized to the respective experimental diets for 14 days, after which fecal material was collected, DNA was extracted from fecal material, and the V3‑V4 region of the bacterial 16S rRNA gene was amplified with PCR, followed by microbial composition analysis. In tandem, the organic acid profiles of the fecal material were analyzed. Results Feeding rats non-supplemented (no prebiotic-added) diets revealed that diets rich in calcium phosphate favored members of the Firmicutes and increased fecal lactic, succinic, acetic, propionic, and butyric acid levels. In contrast, relatively low dietary calcium phosphate levels promoted the abundance of mucin degrading genera like Akkermansia and Bacteroides, and resulted in increased fecal propionic acid levels and modest increases in lactic and butyric acid levels. Irrespective of the calcium phosphate levels, supplementation with GOS or inulin strongly stimulated Bifidobacterium, while only high calcium phosphate diets increased the endogenous Faecalibaculum populations. Conclusions Despite the prebiotic’s substantial difference in chemical structure, sugar composition, oligomer size, and the microbial degradation pathway involved in their utilization, inulin and GOS modulated the gut microbiota very similarly, in a manner that strongly depended on the dietary calcium phosphate level. Therefore, our study implies that the collection of detailed diet information including micronutrient balance is necessary to correctly assess diet-driven microbiota analysis.

Our objective was to evaluate the effect of colostrum feeding times on genome-wide gene expression of neonatal calves. In total, twenty-seven calves were assigned to three colostrum feeding treatments: within 45 min (TRT0h, n = 9), 6 h (TRT6h, n = 9) and 12 h (TRT12h, n = 9). Ileum tissues were collected at 51 h and transcriptomic analysis was conducted. Uniquely expressed genes were identified in TRT0h group with enriched "Antigen Presentation" function. Meanwhile, the weighted gene co-expression network analysis (WGCNA) identified four significant gene modules (|correlation| > 0.50 and P ≤ 0.05). In particular, Turquoise gene module with the enriched "Cadherin binding involved in cell-cell adhesion" and "Cell-cell adherences junction" GO terms were significantly correlated with Faecalibacterium prausnitzii (R = -0.70, P < 0.01) and Bifidobacterium (R = -0.55, P < 0.01). Our findings suggest feeding colostrum without delay could stimulate the expression of genes involved in immune function development related to host response and microbial colonization in neonatal claves.

Background Parkinson’s disease (PD) is one of the most common neurodegenerative diseases. There is accumulating evidence that link gut microbiota to symptomatology and pathophysiology of PD. The aim of this study was to describe the pattern of gut microbiota and its association with PD and identify the effect of environmental factors on gut microbiota. This case–control study included 46 patients diagnosed as Parkinson’s disease (PD) and 31 healthy volunteers age and sex matched. Detailed history including age of onset, duration of disease, environmental risk factors, diet data, treatment, Unified Parkinson’s Disease Rating Scale (UPDRS), and gastrointestinal tract (GIT) domain of Non‐Motor Symptoms Scale (NMSS) were assessed. After extraction of bacterial DNA from the fecal samples, bacterial abundance was quantified by qPCR using 16S rRNA group-specific primers. Results Significant high abundance of Clostridium cluster IV, Akkermansia, Bifidobacterium, and lactic acid bacteria were found in the PD group compared with the control group (P P P = 0.002), while no significant correlations between different gut microbiota and demographic, environment data, different rating scores or dominant type of PD. There was a significant negative correlation between the Bifidobacterium with the duration of illness (P = 0.012). Conclusion The present study highlighted a significant connection between PD and levels of certain types of gut microbiota, in support of a possible link between gut microbiota and a neurodegenerative cascade of PD.

BackgroundThe composition of the infant gastrointestinal (GI) microbiome has been linked to adverse long-term health outcomes and neonatal sepsis. Several factors are known to impact the composition of the microbiome, including mode of delivery, gestational age, feeding method and exposure to antibiotics. The impact of intrapartum antibiotics (IPAs) on the infant microbiome requires further research.ObjectiveWe aimed to evaluate the impact of IPAs on the infant GI microbiome.MethodsWe searched Ovid MEDLINE and Embase Classic+Embase for articles in English reporting on the microbiome of infants exposed to IPAs from the date of inception to 3 January 2021. Primary outcomes included abundance and colonisation ofBifidobacteriumandLactobacillus, as well as alpha and beta diversity.Results30 papers were included in this review. In the first year of life, following exposure to IPAs, 30% (6/20) of infant cohorts displayed significantly reducedBifidobacterium, 89% (17/19) did not display any significant differences inLactobacilluscolonisation, 21% (7/34) displayed significantly reduced alpha diversity and 35% (12/34) displayed alterations in beta diversity. Results were further stratified by delivery, gestational age (preterm or full term) and feeding method.ConclusionsIPAs impact the composition of the infant GI microbiome, resulting in possible reductionsBifidobacteriumand alpha diversity, and possible alterations in beta diversity. Our findings may have implications for maternal and neonatal health, including interventions to prevent reductions in health-promoting bacteria (eg, probiotics) and IPA class selection.

The exopolysaccharide (EPS) from the mycelial fermentation of a medicinal fungus Cordyceps sinensis Cs-HK1 had shown significant anti-inflammatory activity previously, and EPS-LM was a highly active fraction with a relatively low molecular weight (MW) isolated from the Cs-HK1 EPS. This study was to assess the effects of Bifidobacterial fermentation in anaerobic conditions on the molecular properties and anti-inflammatory activity of EPS-LM. In both Bifidobacterial cultures (B. breve and B. longum), EPS-LM was fractionally consumed as a carbon source, increasing the bacterial growth and acetic acid production. Analytical results from the fermentation digesta (supernatant) suggested that EPS-LM was partially degraded to lower molecular weight (MW) products with modified structures during the Bifidobacterial fermentation. More interestingly, the higher MW digesta fraction containing the partially degraded EPS-LM showed even stronger inhibiting activity than the original EPS-LM on the LPS-induced pro-inflammatory responses in THP-1 cell culture, including NF-κB activation, release of NO, TNF-α and IL-8. The study has shown that the fermentation by selected Bifidobacterial strains is effective to modify natural polysaccharides with enhanced bioactivities.

It has been shown that whole grains and dietary fiber are important for their fermentation characteristics in the large intestine, drawing more and more attention to quinoa and quinoa polysaccharides. In this study, we evaluated the prebiotic effect of quinoa seeds and quinoa polysaccharides after human simulated digestion. The modulatory effect of the quinoa and quinoa polysaccharides (QPs) on the gut microbiota was evaluated by the in vitro fermentation using human fecal microbiota. The yield of polysaccharides extraction was 15.45%. The digestibility of the cooked and uncooked quinoa after simulation of human digestion was 69.04% and 64.09%, respectively. The effect on the microbiota composition and their metabolic products was determined by the assessment of pH, short‐chain fatty acids (SCFAs), and changes in the bacterial population. After 24 hr anaerobic incubation, the total SCFAs of cooked, uncooked quinoa, and quinoa polysaccharides were 82.99, 77.11, and 82.73 mM, respectively with a pH decrease. At the phylum, genus, and class level, it has been found that the quinoa substrates enhance the growth of certain beneficial bacteria such as Prevotella and Bacteroides. Quinoa polysaccharides can be considered prebiotic due to their ability to increase Bifidobacterium and Collinsella. Principal component analysis (PCA) showed that there was a distinct modulating effect on the fecal microbiota which represents different distribution. Our research suggests that quinoa and quinoa polysaccharides have a prebiotic potential due to their association with the positive shifts in microbiota composition and short‐chain fatty acids production, which highlights the importance of further studies around this topic., Quinoa and quinoa polysaccharides can modulate the gut microbiota. The steadily increasing prevalence of Alzheimer's disease (AD) worldwide and the lack of effective therapeutic agent attract novel therapeutic approach in recent years. In view of the close relationships between gut microbiota and AD, probiotics have been suggested as potential therapeutic options for AD in recent years. The present review discussed the research progresses concerning the effects of probiotics administration to combat AD. A total of 35 studies, including 26 animal model studies and 9 human studies, were included herein. Among the 26 animal model studies, 24 used mice model, and 2 used Caenorhabditis elegans and Drosophila melanogaster AD models, respectively. As for probiotics, a total of 13 studies employed single-strain probiotic, and the rest studies used multi-strain probiotics (ranged from 2 to 9 probiotic strains), 4 used probiotic-fermented milk or probiotic-fermented soybean, 2 studies used engineered probiotic strain, and 4 studies focused on the combined effect of probiotics with AD drug memantine, selenium, or exercise. Bifidobacterium and Lactobacillus species were the most frequently used probiotics in the included studies. Overall, currently available studies showed that probiotic administration conferred neuroprotective benefits and could attenuate cognitive deficits and modulate gut microbiota dysbiosis, which may be related to oxidative and inflammatory pathways. Several perspectives on future studies on this topic are proposed. Thus, probiotics seem to be an attractive approach to combat AD, which deserves to be further studied by well-designed large-scale clinical studies. To investigate the correlation between intestinal flora and serum inflammatory factors IL-1, IL-2, IL-6 and hs-CRP in post-stroke depression (PSD) in ischemic stroke patients.Observational study. Study Place and Duration of Study: Jiulongpo District Hospital of Traditional Chinese Medicine, Chongqing City, China, from October 2018 to May 2020.One hundred and sixty-three patients with ischemic stroke were divided into Group A (PSD) and Group B (no PSD), according to whether they had PSD. Intestinal flora indexes (Enterococcus faecalis, Escherichia coli and Bifidobacterium) and serum IL-1, IL-2, IL-6 and hs-CRP were detected.Among 163 patients with ischemic stroke, 67 (41.10%) had PSD (Group A) and 96 (58.90%) had no PSD (Group B). Contents of Enterococcus faecalis and Escherichia coli in Group A were higher than those in Group B (both p0.001), and content of Bifidobacterium in Group A was lower than that in Group B (p0.001). Serum IL-1, IL-2, IL-6 and hs-CRP levels in Group A were higher than those in Group B (all p0.001). Pearson correlation test showed that contents of Enterococcus faecalis and Escherichia coli in Group A were positively correlated with IL-1, IL-2, IL-6 and hs-CRP, and content of Bifidobacterium was negatively correlated with IL-1, IL-2, IL-6 and hs-CRP.There are intestinal flora imbalance and Bifidobacterium undergrowth in patients with PSD, which can lead to overexpression of serum inflammatory factors. In this study, we have unveiled the novel potential of xylanase production by Bacillus velezensis, previously known only for its PGPR traits. According to our investigations, Bacillus velezensis AG20 produced extracellular xylanase with a fold purification of 5.3 and a yield of 21 %. The purified xylanase (45 kDa) aptly cleaved linear β-(1→4)-xylan with a maximum velocity of 21.0 ± 3.0 U/mL, a turnover of 1.75/s, and a catalytic cycle of 0.571 at optimum pH 7. The temperature of 50 °C was found optimum for enzyme activity. Purified endoxylanase was thermostable and retained its significant residual activities at 50 °C and 60 °C. Metal cations such as Ca2+ and Mg2+ enhanced the substrate catalysis up to 1.5 fold. The purified endoxylanase also demonstrated multi-substrate hydrolyzing properties. However, the enzymatic hydrolysis of sugarcane bagasse produced xylooligosaccharides (XOS), including xylobiose, xylotriose, and xylotetrose. The mixture of released XOS has profound stability in gastric juice, intestinal fluid, and α-amylase and facilitates probiotic bacteria Bifidobacterium. The mixed XOS (300 μg/mL) significantly inhibited the HT-29 and Caco-2 cell proliferation by 90 % and 75 %, respectively, at 48 h. The possible mechanism of XOS uptake through the BIAXP receptor of Bifidobacterium has been contemplated and established using the caver analysis, Molecular Dynamics (MD) simulation, and Density Functional Theory (DFT) studies. Thus, here we reveal the utility of this novel bacterial strain in industries as high-temperature catalysts.

Gut microbial fermentation of soluble dietary fibers promotes general and substrate-specific health benefits. In this study, the fermentation characteristics of two soluble branched-dietary fibers, namely, agavin (a type of agave fructans) and digestion-resistant maltodextrin (RD) were investigated against cellulose, using a simulated colonic fermenter apparatus employing a mixed culture of swine fecal bacteria. After 48 h of complete fermentation period, the microbial composition was different among all groups, where Bifidobacterium spp. and Lactobacillus spp. dominated the agavin treatment, while the members of the families Lachnospiraceae and Prevotellaceae dominated the RD treatment. Agavin treatment exhibited a clearly segregated two-phased prolonged fermentation trend compared to RD treatment as manifested by the fermentation rates. Further, the highest short-chain fatty acids production even at the end of the fermentation cycle, acidic pH, and the negligible concentration of ammonia accumulation demonstrated favorable fermentation attributes of agavin compared to RD. Therefore, agavin might be an effective and desirable substrate for the colonic microbiota than RD with reference to the expressed microbial taxa and fermentation attributes. This study revealed a notable significance of the structural differences of fermentable fibers on the subsequent fermentation characteristics.

Background Intermittent fasting (IF) and aerobic training have demonstrated beneficial effects on intestinal microbiota composition, but little is known about benefits to the brain through the gut-brain axis. The present study aimed to evaluate gut-brain axis parameters in Wistar rats submitted to IF associated or not with aerobic training. Methods Male rats were evaluated for training performance and then randomized into 4 groups of ten: sedentary control (SC), trained control (TC), sedentary intermittent fasting (SIF), and trained intermittent fasting (TIF), and evaluated during four weeks. Results The adiposity index was similar among the TC (2.15±0.43%), SIF (1.98±0.69%) and TIF (1.86±0.51%) groups, and differed from SC (2.98±0.80%). TIF had lower counts of lactic acid bacteria, while SIF had higher counts of Bifidobacterium and Enterococcus. TIF had the highest amount of formic acid in faeces (44.44±2.40 μmol/g) and lowest amount of succinic acid in the gut (0.38±0.00 μmol/g), while SIF had the highest propionic acid amount in the faeces (802.80±00.33 μmol/g) and the lowest amount of lactic acid in the gut (0.85±0.00 μmol/g). TIF demonstrated a tendency towards an anxiolytic effect and SIF showed potential antidepressant effect. IF caused different brain and intestinal injuries. TIF rats presented a diffuse and intense marking of IL-1β in the hippocampus. Conclusion IF and aerobic exercise, associated or not, can modulate parameters related to the gut-brain axis of Wistar rats, and some benefits may be related to the amounts of organic acids.

Accumulating evidence has pointed out that the gut-brain axis plays important roles in the etiology of autism spectrum disorder (ASD). Gut dysbiosis was reported in both ASD human patients and animal models. Dip2a was identified as a human ASD candidate gene. Deletion of Dip2a led to dendritic spine dysfunction and autistic-like behaviors in mice. To further investigate if Dip2a deletion leads to gut dysbiosis, we used 16S rDNA sequencing to study the gut microbiota in Dip2a KO mice. In both co-housed and separated breeding conditions, deletion of Dip2a could affect the gut microbiome composition. The probiotic bacteria, Lactobacillus and Bifidobacterium, became less abundant, while some potentially harmful bacteria, Alistipes, Lachnospiraceae_NK4A136_group, Clostridium, Desulfovibrio, and Enterorhabdus, became more abundant. We further found that probiotic treatment could help to reconstitute the gut microbiome composition in Dip2a KO mice. Altogether, these data showed DIP2A is required for the proper composition of gut microbiota, and the probiotics have potential roles in rectifying the gut microbiota in Dip2a KO mice.

This study elucidated the regulatory roles of milk glycoprotein core fucosylation on the gut microbiota of breast-fed infants. In order to investigate the effects of core fucosylation of maternal milk N-glycan on the intestinal functions in newborn mice, Fut8+/+ → Fut8+/+ and Fut8± → Fut8+/+ models were established. The results indicated that the gut microbiota diversity was altered and production of Bifidobacterium and Lactobacillus was reduced in the intestine of Fut8± → Fut8+/+mice. Furthermore, the levels of SCFAs were decreased in the gut of Fut8± → Fut8+/+infant. To further investigate the effects of core fucosylation of breast milk N-glycan on the intestinal inflammation in newborns, NEC models were established, and Fut8+/+ off-springs with experimentally induced NEC were fed with the breast milk of Fut8+/+ and Fut8± maternal mice. Our data revealed that reduced core fucosylation of breast milk N-glycan could be responsible for the intestinal inflammation in NEC offspring, as the production of pro-inflammatory cytokines including IL-1b, IL-6, IL-17A and TNF-a was significantly upregulated in Fut8± → Fut8+/+NEC group, while the levels of anti-inflammatory cytokine IL-10 were remarkably reduced. Furthermore, Treg/Th17 balance in the intestine was disturbed in Fut8± → Fut8+/+NEC newborn, suggesting the impaired immune development caused by reduced core fucosylation of breast milk. These findings indicated that core fucosylation of maternal milk N-glycan could assist the colonization of gut Lactobacillus and Bifidobacterium and regulate the Treg/Th17 balance, subsequently preventing intestinal inflammation in breast-fed newborns.

The human microbiome influences human health in both negative and positive ways. Studies on the transportomes of these organisms yield information that may be utilized for various purposes, including the identification of novel drug targets and the manufacture of improved probiotic strains. Moreover, these genomic analyses help to improve our understanding of the physiology and metabolic capabilities of these organisms. The present study is a continuation of our studies on the transport proteins of the major gut microbes. Bifidobacterium species are essential members of the human gut microbiome, and they initiate colonization of the gut at birth, providing health benefits that last a lifetime. In this study we analyze the transportomes of nine bifidobacterial species: B. adolescentis, B. animalis, B. bifidum, B. breve, B. catenulatum, B. dentium, B. longum subsp. infantis, B. longum subsp. longum, and B. pseudocatenulatum. All of these species have proven probiotic characteristics and exert beneficial effects on human health. Surprisingly, we found that all nine of these species have similar pore-forming toxins and drug exporters that may play roles in pathogenesis. These species have transporters for amino acids, carbohydrates, and proteins, essential for their organismal lifestyles and adaption to their respective ecological niches. The strictly probiotic species, B. bifidum, however, contains fewer such transporters, thus indicative of limited interactions with host cells and other gut microbial counterparts. The results of this study were compared with those of our previous studies on the transportomes of multiple species of Bacteroides, Escherichia coli/Salmonella, and Lactobacillus. Overall, bifidobacteria have larger transportomes (based on percentages of total proteins) than the previously examined groups of bacterial species, with a preference for primary active transport systems over secondary carriers. Taken together, these results provide useful information about the physiologies and pathogenic potentials of these probiotic organisms as reflected by their transportomes.

Bifidobacterium longum NCIM 5672 is a probiotic strain isolated from the Indian infant feces. The probiotic efficacy of Bifidobacteria is majorly affected by its acid tolerance. This study determined the probiotic properties and acid-tolerance mechanism of B. longum NCIM 5672 using whole-genome sequencing. The genome annotation is carried out using the RAST web server and NCBI PGAAP. The draft genome sequence of this strain, assembled in 63 contigs, consists of 22,46,978 base pairs, 1900 coding sequences and a GC content of 59.6%. The genome annotation revealed that seven candidate genes might be involved in regulating the acid tolerance of B. longum NCIM 5672. Furthermore, the presence of genes associated with immunomodulation and cell adhesion support the probiotic background of the strain. The analysis of candidate acid- tolerance-associated genes revealed three genes, argC, argH, and dapA, may play an essential role in high acid tolerance in B. longum NCIM 5672. The results of RT-qPCR supported this conclusion. Altogether, the results presented here supply an effective way to select acid-resistant strains for the food industry and provide new strategies to enhance this species' industrial applications and health-promoting properties.

Background. The pathogenesis of chronic inflammatory bowel disease (IBD) is still not fully clarified. It is known that disorders of the intestinal microbiota lead to an increased intestinal permeability, activation of mucous and adaptive immunity, impaired production and intestinal absorption of short-chain fatty acids (SCFA). The ratio of acetic, propionic, butyric acids is an important indicator of the integrity of the intestinal microbial community. Thus, the study of the composition of the intestinal microbiota and the concentrations of fecal SCFA is a very promi­sing approach to broadening the understanding of IBD pathoge­nesis. The purpose of our study was to determine the features of the production of fecal SCFA and the composition of colon microbiota in patients with IBD. Materials and methods. The study, which was carried out at the Department of Intestinal Diseases of the Institute of Gastroenterology of the National Academy of Medical Sciences of Ukraine, involved 74 patients with IBD with an average age of (43.2 ± 1.8) years, who according to nosology were divided into 2 groups: group I — those with ulcerative colitis (UC) (n = 66), group II — individuals with Crohn’s disease (CD) (n = 8). The diagnoses of CD and UC were established accor­ding to generally accepted diagnostic standards in gastroenterology. Determination of fecal SCFA was carried out by chromatographic method with the use of hardware-software complex for medical researches on the basis of the gas chromatographer Chromateс Crystal 5000. The intestinal microflora was evaluated using a microbiological study of the colon content. Results. Патогенез хронічних запальних захворювань кишечника (ХЗЗК) досі залишається до кінця не вивченим. Відомо, що порушення мікробіоти кишечника призводять до підвищеної проникності кишечника, порушення продукції та кишкового всмоктування коротколанцюгових жирних кислот (КЖК), співвідношення яких є важливим показником цілісності мікробної спільноти кишечника. Таким чином, вивчення складу мікробіоти кишечника і концентрацій фекальних КЖК є дуже перспективним підходом до розширення розуміння патогенезу ХЗЗК. Метою дослідження було визначити особливості продукції фекальних КЖК та складу мікробіоти товстої кишки у хворих на ХЗЗК. Матеріали та методи. У дослідження, що проводилось у відділенні захворювань кишечника ДУ «Інститут гастроентерології НАМН України», були залучені 74 пацієнти з ХЗЗК, які були розподілені на 2 групи: І група — хворі на неспецифічний виразковий коліт (НВК) (n = 66), II — хворі на хворобу Крона (ХК) (n = 8). Діагнози ХК та НВК були встановлені згідно із загальноприйнятими стандартами діагностики в гастроентерології. В статье рассматриваются вопросы, связанные с кишечной микрофлорой и короткоцепочечными жирными кислотами у больных с синдромом раздраженного кишечника. Установлено, что концентрация уксусной, пропионовой, масляной кислот в кале выше у больных с синдромом раздраженного кишечника с диареей по сравнению с больными без диарейного синдрома. У 83,3–88,9 % больных с различными формами синдрома раздраженного кишечника наблюдаются дисбиотические изменения; у пациентов с диарейной формой чаще отмечается пониженное содержание бифидобактерий и, особенно, лактобактерий. Отмечается определенная взаимозависимость между концентрацией короткоцепочечных жирных кислот и содержанием бифидобактерий, лактобактерий, кандидозной и условно-патогенной флоры.

The dietary supplementation of prebiotics is considered a promising strategy for the modulation of gut microbiota. Due to the wide variety of animal models and tremendous inter-individual variability from human investigations, the prebiotic effect of fibers is often difficult to compare between studies. Here, the effects of 11 dietary fibers on human fecal microbiota were studied using an in vitro human fecal fermentation model under well-controlled conditions. All fibers showed positive regulatory effects on short chain fatty acids (SCFAs) and several beneficial bacteria, including Parabacteroides distasonis and Bifidobacterium spp. Cultures supplemented with xylo-oligosaccharide and konjac flour showed the highest SCFAs. According to regulatory effects, fibers were divided into three groups, with 13 indicator OTUs (operational taxonomic units) identified. Fecal microbiota regulated by isomalto-oligosaccharide and chitosan-oligosaccharide were similar to fructo-oligosaccharide and inulin outputs. As a supplement to in vivo studies, our results comprehensively summarized the similarities and distinctiveness of fibers in regulating fecal microbiota structures. Розвиток та перебіг пептичної виразки шлунка та дванадцятипалої кишки супроводжуються наявністю штамів H. pylori (СagA+ VacA+, СagA+ vacА–, CagA– VacA+, CagA– VacA–), що виявлено у хворих за наявності супутньої патології та коли її немає. У групі хворих на пептичну виразку шлунка та дванадцятипалої кишки в поєднані з артеріальною гіпертензією і цукровим діабетом 2-го типу виявлено штами СagA+ VacA+ у 14 осіб (45,16 %), СagA+ VacA– — у 4 (12,9 %), CagA– VacA+ — у 13 (41,43 %) та в групі хворих на пептичну виразку шлунка та дванадцятипалої кишки — відповідно в 4 (13,79 %), у 8 (27,59 %), у 16 (55,17 %) осіб. Вплив штамів H. pylori супроводжується змінами протеолітичної та фібринолітичної активності. З урахуванням супутньої патології та ролі штамів при розвитку основної патології виявлено вірогідне зниження рівня лізису альбуміну, лізису азоказеїну та підвищення лізису азоколу порівняно зі зниженням сумарної фібринолітичної активності, неферментативної фібринолітичної активності та підвищенням ферментативної фібринолітичної активності. Використання антихелікобактерної базисної терапії та пробіотиків (Lactobacterium, Bifidobacterium) дає позитивний результат.

Background. Type 1 diabetes mellitus (DM) results in the development and unfavorable clinical course of chronic inflammatory diseases, including those of ENT organs. Aim of the study — to explore the species composition and population level of the large intestine microflora in patients with type 1 DM depending on the severity of its clinical course. Materials and methods. By means of bacteriological and mycological methods, we have examined the content of the large intestine cavity in 50 patients aged 15 to 60 years with moderate and severe DM type 1, as well as in 30 apparently healthy individuals of the same age, who considered themselves to be healthy and were not affected by any diseases during the previous six months. The severe form of DM was diagnosed in 39 of 50 patients, and the clinical course of moderate severity — in 11 patients. Results. In moderate and severe course of type 1 DM, the deficiency of autochtonous bacteria of the genus Bifidobacterium, Lactobacillus, Peptostreptococcus, Enterococcus is formed in the content of the large intestine cavity, they are the most important by their representation in the large intestine microbiocenosis. Against this background, there is a contamination of the large intestine cavity with pathogenic (E.coli Hly+, EPEC) and opportunistic (Enterobacter, Citrobacter, Serratia, Proteus) enterobacteria, as well as opportunistic bacteria of the genus Bacteroides, Clostridium, P.niger, Staphylococcus, and yeast-like fungi of the Candida genus reaching a high population level. Conclusions. In the microbiocenosis of the large intestine cavity of patients with type 1 DM, there is a significant increase in the level of enterotoxigenic and enterotoxic (61.38 %) and opportunistic bacteria (71.24 %). Background. Production of numerous biologically active compounds and their metabolites by intestinal microflora, interaction with the immune and other systems is of great importance while studying its changes in various diseases, one of which is thyrotoxicosis. So, the purpose of this study was to determine the severity of intestine microbioma disorder in white rats with experimental thyrotoxicosis (ET). Materials and methods. Studies were carried out on 25 mature male white rats (15 — control group, 10 — research group). ET was simulated by intragastric administration of L-thyroxine for 14 days. Under sterile conditions a laparotomy was performed, a section (2–3 cm) of the large intestine with its contents was taken. Sterile 0.9% NaCl solution was added to the content. Series of ten-fold dilutions with a concentration of the initial mixture of 10–2 to 10–11 was prepared. From each test tube 0.01 ml was seeded on solid nutrient media with subsequent isolation and identification of microbes according to morphological, tinctorial, cultural and biochemical properties. Results. The results of the study demonstrated that in ET animals the main microbioma is represented by bacteria Bifidobacterium, Lactobacillus, Bacteroides, and also opportunistic enterobacteria (Escherichia, Proteus, Klebsiella), peptococcus, staphylococci and clostridia. This is accompanied by the elimination of Peptostreptococcus, Enterococcus from bacterial biotope and the contamination of K. oxytoca and staphylococci. There was a pronounced deficit of bifidobacteria by 42.81 %, lactobacillus by 22.57 %, normal intestinal bacillus by 16.48 %. By the population level, the coefficient of quantitative dominance and the significance factor, the leading place is occupied by bacteroids, role of which is increased by 21.72 %, and lactobacillus role decreases by 39.31 %, bifidobacteria decreases by 51.48 % and E. coli decreases by 57.49 %. In this case, the role of peptococcus 3.37-fold increases, clostridia by 4.53, and by 73.93 % by the number of proteus. Conclusions. Under conditions of ET, there is an elimination of Bifidobacterium, Lactobacillus, Peptostreptococcus, Enterococcus and contamination of the biotope with conditionally pathogenic enterobacteria (Proteus, Klebsiella) and staphylococci. Deficiency of bifidobacteria and lactobacilli leads to changes in taxonomic structure and formation of dysbiosis of II and III stage.

We investigated the ability of lotus seed resistant starch (LRS) to affect the conversion of sodium taurocholate (STCA) by regulating the intestinal flora, using glucose (GLU) and high amylose corn starch (HAMS) as controls. The dominant microbiota in LRS group were mainly Lactobacillus and Escherichia_Shigella, with a small proportion of Bifidobacterium. Meanwhile, Lactobacillus, Bifidobacterium and Enterococcus were dominant microbiota in the HAMS group. Lactobacillus, Burkholderia-Caballeronia-Paraburkholderia and Sphingomonas were found in the GLU group. Furthermore, Bifidobacterium, Enterococcus and Escherichia_Shigella were negatively correlated with STCA and sodium taurodeoxycholate (STDCA), while these bacteria were positively correlated with bile salt hydrolase (BSH) and hydroxysteroid dehydrogenase (HSDH) content. Meanwhile Burkholderia-Caballeronia-Paraburkholderia and Sphingomonas were positively correlated with STCA and STDCA, while these bacteria were negatively correlated with BSH and HSDH content. LRS promoted the proliferation of Bifidobacterium and Escherichia_Shigella to secret more BSH and HSDH, accelerating the hydrolysis of STCA and reducing the conversion of STDCA.

Background. The microecosystem of human organism closely cooperates with immune and endocrine systems, and in particular the microbiome of the pre­epithelial biofilm of the large intestine is one of the most sensitive biotopes that is able to change in different diseases. The purpose was to establish the microbiological structure (species composition and population level of microflora) of the pre­epithelial biofilm of the large intestine in albino rats with experimental hypothyroidism. Materials and methods. Experiments were carried out on 25 mature male albino rats weighing 220–240 g, of which 15 animals were included to the control group (intact animals), and 10 rats — to the main group. The experimental thyrotoxicosis was simulated by intragastric administration of mercazolilum for 14 days. Under sterile conditions, laparotomy was performed; a sample (up to 3 cm) of the large intestine with its contents was taken. The washed portion of the intestine was homogenized with a sterile 0.9% NaCl solution. A series of ten­fold dilutions with 10 –2 to 10 –7 concentrations of the initial mixture were prepared. From each tube, 0.01 ml were seeded on solid optimal nutrient media with subsequent isolation and identification of microbes according to morphological, tinctorial, cultural and biochemical properties. Results . Colonization resistance of the pre­epithelial biofilm of the large intestine changed in experimental animals when modeling hypothyroi­dism: the population level of lactobacteria reduced by 51.58 %, bifidobacteria — by 37.35 %, bacteroides — by 22.82 %, peptostreptococci — by 6.5 %. Pre­epithelial biofilm was contaminated by collibacillus and other enterobacteria (Proteus, Klebsiella), P.niger , staphylococci. The increase was detected in the population level of collibacillus by 10.31 % and clostridia by 8.2 %. Dysbiotic violations of I, II and III degrees were determined that requires a correction by means of probiotics. Conclusions. In the experimental research of mercazolilum­induced hypothyroidism on albino rats, the elimination of Bifidobacterium , Lactobacillus was established; contamination and colonization of biotope by opportunistic pathogenic microorganisms of Proteus , Klebsiella , Staphylococcus , Peptococcus were detected on the background of increased population level of collibacillus. There are violations of microbiocenosis in the form of dysbacteriosis degree I–III.

Mucormycosis is an acute fungal infection with 90% of cases in the form of rhino-orbito-cerebellar. It is an aggressive and life-threatening fungal infection causing 50% mortality in people with coronavirus disease 2019 (COVID-19). In COVID-infected patients due to, diabetic ketoacidosis, epithelial damage, ciliary dysfunction, dysfunctional phagocytic mechanism, and immunosuppression, there is impaired chemotaxis and defective intracellular killing leads to fungal spores to invade, germinate and penetrate in surrounding tissues. The use of broad-spectrum antibiotics disrupts the normal microbiomes and increases the probability of growth of Rhizopus spp. Commercially available probiotics such as Lactobacillus, Bifidobacterium, Enterococcus, Streptococcus, and Saccharomyces when administered in adequate quantities form siderophores which induces iron stress in fungus and inhibits spore germination.

INTRODUCTION : Inbalance of microbiota in gastrointestinal tract have the risk of getting gastrointestinal infections, one of them is diarrhea. AIM : The aim of this study was to determine bifidobacterium and eschericia coli microbiota and compare the microbiota obtained between exclusive breast fed and formula fed Indonesian infants village andalas. METHODS : This study was an observational study with cross sectional comparative design. Samples faeses of infant was taken by Multistage Simple Random Sampling with total sample of 28 infants in range 0-6 months. This study was conducted in Pauh community health center areas, Village Andalas . The infant’s feces was taken then send to Microbiology Laboratorium of Dr. M. Djamil Hospital Padang city, Indonesia. The instruments used were a questionnaire and a colony counter. Data were processed and analyzed by using independent t test and Mann Whitney test. RESULTS : The fecal microbiota of the 0-6 months Indonesian infants village andalas two species including bifidobacterium and eschericia coli were common commensal intestinal microbiota in all infants. The predominant intestinal microbiota in the breast fed infants bifidobacterium average 3,59 x 109 CFU/g (p < 0,05). the mean eschericia coli microbiota from the formula fed infants 66,8 x 109 CFU/g (p < 0,05) CONCLUSION : Exclusive breastfeeding infant’s faeses contain more bifidobacteria and formula breastfed infant’s faeces contain more escherichia coli bacteria. These findings advance our understanding of the gut microbiota in healthy infants. They also provide new evidence infant diet as determinants of this essential microbial community in early life

Antibiotic-induced dysbiosis of the microbial community has been associated with several gastrointestinal symptoms. The impact of repeated administration of Lacticaseibacillus rhamnosus GG (CNCM-I-4798) (formerly known as Lactobacillus rhamnosus GG), Saccharomyces cerevisiae boulardii (CNCM-I-1079) and their combination (associated in Smebiocta/Smectaflora Protect®) in supporting recovery of gut microbiota functionality and composition during and following amoxicillin:clavulanic acid administration was evaluated in vitro. Antibiotic dosage negatively affected SCFA production, coinciding with detrimental effects on Bacteroidetes, Firmicutes and Bifidobacterium spp. in the simulated proximal colon, while Akkermansia muciniphila was significantly reduced in the distal colon. L. rhamnosus GG and S. boulardii were able to thrive in both colon regions upon dosing, with S. boulardii even showing protective effects on the survival of L. rhamnosus GG during antibiotic administration. The impact of the probiotic strains on microbiome recovery revealed that supplementation with L. rhamnosus GG and/or S. boulardii resulted in a stimulating effect on the most abundant bacterial groups within the bacterial community of each donor. For one of the donors tested, co-dosing of L. rhamnosus GG and S. boulardii resulted in superior short-chain fatty acid recovery accompanied by a stronger increase in abundance of Bifidobacteriaceae. Overall, the current study provides first evidence that combined supplementation of L. rhamnosus GG and S. boulardii might be an interesting candidate in limiting detrimental effects of amoxicillin:clavulanic acid on the human gut microbiome, though further studies are warranted to confirm these findings.

Context Obesity-associated chronic metabolic disease is a leading contributor to mortality globally. Plants belonging to the genera Acacia are routinely used for the treatment of diverse metabolic diseases under different ethnomedicinal practices around the globe. Objective The current review centres around the pharmacological evidence of intestinal-level mechanisms for metabolic health benefits by Acacia spp. Results Acacia spp. increase the proportions of gut commensals (Bifidobacterium and Lactobacillus) and reduces the population of opportunistic pathobionts (Escherichia coli and Clostridium). Acacia gum that is rich in fibre, can also be a source of prebiotics to improve gut health. The intestinal-level anti-inflammatory activities of Acacia are likely to contribute to improvements in gut barrier function that would prevent gut-to-systemic endotoxin translocation and limit "low-grade" inflammation associated with metabolic diseases. Conclusion This comprehensive review for the first time has emphasised the intestinal-level benefits of Acacia spp. which could be instrumental in limiting the burden of metabolic disease.
