Biomedical science is rapidly developing in terms of more transparency, openness and reproducibility of scientific publications. This is even more important for all studies that are based on results from basic semen examination. Recently two concordant documents have been published: the 6th edition of the WHO Laboratory Manual for the Examination and Processing of Human Semen, and the International Standard ISO 23162:2021. With these tools, we propose that authors should be instructed to follow these laboratory methods in order to publish studies in peer-reviewed journals, preferable by using a checklist as suggested in an Appendix to this article.

Cover crop nitrogen (N) cycling has an important role in agricultural production and contributes to peach [Prunus persica (L.) Batsch] N nutrition. This study evaluated black oat (Avena strigosa Schreb) and ryegrass (Lolium multiflorum L.) residue decomposition dynamics, N recovery from cover crop residues, and N compartmentalization in peach tree organs. A 2-year field trial was developed with labeled (3.6?4.0 atom% 15N excess) cover crop shoot biomass application in a 5-year-old peach orchard. The region?s climate is warm temperate (Cfb), and the soil is classified as a Typic Hapludalf. Litter bags with unlabeled shoot residues were also deposited in the orchard to assess biomass, carbon (C), N, lignin, cellulose, and non-structural biomass decomposition dynamics. After 13 months, the leaves, trunk, and roots showed the greatest proportion of N derived from residues (Ndfr) (35.4, 25.1, and 22.4%, respectively) while the greatest concentrations of 15N and Ndfr occurred in roots

BACKGROUND Dementia is a chronic progressive neurological disease affecting millions of people worldwide, and represents a relevant economic burden for healthcare systems. Although its pathogenesis is still unknown, recent findings have reported that a dysregulated gut-brain axis communication, a fundamental relationship mediated by several host and microbial molecules, is associated with cognitive disorders. In addition, gut microbiota manipulation reduces neuroinflammation, improving cognitive function by restoring the functional gut-brain axis. AIM To better define the effects of probiotics, prebiotics, synbiotics, and fecal microbiota transplant (FMT) on cognitive function. METHODS We performed a literature search of human randomized clinical trials to examine the effects of the administration of probiotics, prebiotics, synbiotics, or FMT on cognition outcomes in healthy or sick people of every age, sex, and nationality. We systematically searched Embase, Medline/PubMed, Cochrane Library, central and clinicaltrials.gov databases with a combination of comprehensive terms related to cognition and gut microbiota manipulation. Then we carefully reviewed and synthesized the data by type of study design and setting, characteristics of the studied population, kind of intervention (strain type or mixture type, dosage, and frequency of administration), control treatment, inclusion and exclusion criteria, follow-up duration, and cognitive or memory outcomes. RESULTS After examining the titles and abstracts, the initial literature screening identified 995 articles, but we added 23 papers in our systematic review. The analyses of these selected studies highlighted that both probiotic supplementation and FMT improved cognitive function regardless of the type and posology of administration and the adopted cognitive tests and questionnaires. We found that most of the studies conducted in healthy people showed a significant positive effect of the intervention on at least one of the performed cognitive tests. Regarding unhealthy subjects, while FMT and especially probiotic administration had multiple beneficial effects on different cognitive functions, supplementation with prebiotics did not provide any cognitive improvement. CONCLUSION Probiotic supplementation and FMT may represent a promising strategy to restore gut eubiosis and enhance the cognitive functions of healthy people and patients with neurological disorders.

BAP1-inactivated melanocytic tumor (BIMT) is a group of melanocytic neoplasms with epithelioid cell morphology molecularly characterized by the loss of function of BAP1, a tumor suppressor gene located on chromosome 3p21, and a mutually exclusive mitogenic driver mutation, more commonly BRAF. BIMTs can occur as a sporadic lesion or, less commonly, in the setting of an autosomal dominant cancer susceptibility syndrome caused by a BAP1 germline inactivating mutation. Owing to the frequent identification of remnants of a conventional nevus, BIMTs are currently classified within the group of combined melanocytic nevi. "Pure" lesions can also be observed. We studied 50 BIMTs from 36 patients. Most lesions were composed of epithelioid melanocytes of varying size and shapes, resulting extreme cytomorphological heterogeneity. Several distinctive morphological variants of multinucleated/giant cells were identified. Some hitherto underrecognized microscopic features, especially regarding nuclear characteristics included nuclear blebbing, nuclear budding, micronuclei, shadow nuclei, peculiar cytoplasmic projections (ant-bear cells) often containing micronuclei and cell-in-cell structures (entosis). In addition, there were mixed nests of conventional and BAP1-inactivated melanocytes and squeezed remnants of the original nevus. Of the 26 lesions studied, 24 yielded a BRAF mutation, while in the remaining two cases there was a RAF1 fusion. BAP1 biallelic and singe allele mutations were found in 4/22 and 16/24 neoplasms, respectively. In five patients, there was a BAP1 germline mutation. Six novel, previously unreported BAP1 mutations have been identified. BAP1 heterozygous loss was detected in 11/22 lesions. Fluorescence in situ hybridization for copy number changes revealed a related amplification of both RREB1 and MYC genes in one tumor, whereas the remaining 20 lesions studied were negative; no TERT-p mutation was found in 14 studied neoplasms. Tetraploidy was identified in 5/21 BIMTs. Of the 21 patients with available follow-up, only one child had a locoregional lymph node metastasis. Our results support a progression of BIMTs from a conventional BRAF mutated in which the original nevus is gradually replaced by epithelioid BAP1-inactivated melanocytes. Some features suggest more complex underlying pathophysiological events that need to be elucidated.

AIMS: Controversial data have been published regarding the prognostic role of cardiac troponins in patients who need hospitalization because of coronavirus disease 2019 (COVID-19). The aim of the study was to assess the role of high-sensitivity troponin plasma levels and of respiratory function at admission on all-cause deaths in unselected patients hospitalized because of COVID-19. METHODS: We pooled individual patient data from observational studies that assessed all-cause mortality of unselected patients hospitalized for COVID-19. The individual data of 722 patients were included. The ratio of partial pressure arterial oxygen to fraction of inspired oxygen (PaO2/FiO2) and high-sensitivity troponins was reported at admission in all patients. This meta-analysis was registered on PROSPERO (CRD42020213209). RESULTS: After a median follow-up of 14 days, 180 deaths were observed. Cilia are dynamic subcellular systems, with core structural and functional components operating in a highly coordinated manner. Since many environmental stimuli sensed by cilia are circadian in nature, it is reasonable to speculate that genes encoding cilia structural and functional components follow rhythmic circadian patterns of expression. Using computational methods and the largest spatiotemporal gene expression atlas of primates, we identified and analyzed the circadian rhythmic expression of cilia genes across 22 primate brain areas. We found that around 73% of cilia transcripts exhibited circadian rhythmicity across at least one of 22 brain regions. In 12 brain regions, cilia transcriptomes were significantly enriched with circadian oscillating transcripts, as compared to the rest of the transcriptome. The phase of the cilia circadian transcripts deviated from the phase of the majority of the background circadian transcripts, and transcripts coding for cilia basal body components accounted for the majority of cilia circadian transcripts. In addition, adjacent or functionally connected brain nuclei had large overlapping complements of circadian cilia genes. Most remarkably, cilia circadian transcripts shared across the basal ganglia nuclei and the prefrontal cortex peaked in these structures in sequential fashion that is similar to the sequential order of activation of the basal ganglia-cortical circuitry in connection with movement coordination, albeit on completely different timescales. These findings support a role for the circadian spatiotemporal orchestration of cilia gene expression in the normal physiology of the basal ganglia-cortical circuit and motor control. Studying orchestrated cilia rhythmicity in the basal ganglia-cortical circuits and other brain circuits may help develop better functional models, and shed light on the causal effects cilia functions have on these circuits and on the regulation of movement and other behaviors.

Systematic pre-participation screening of subjects practicing sports activity has the potential to identify athletes at risk of sudden cardiac death. However, limited evidence are present concerning the yield of echocardiography as a second-line exam in athletes with abnormal pre-participation screening.Consecutive athletes were screened (2011-2017) in a community-based sports medicine center in Tuscany, with familial history, physical examination and ECG. Patients with abnormal/1 borderline ECG findings, symptoms/signs of cardiovascular diseases, cardiovascular risk factors or family history of juvenile/genetic cardiac disease underwent echocardiography.A total of 30109 athletes (age 21 [15;31]) were evaluated. Of these, 6234 (21%) were aged 8-11 years, 18309 (61%) 12-18 years, 4442 (15%) 19-35 years, 1124 (4%)35 years. A total of 2569 (9%) athletes were addressed to echocardiography. Referral rates increased significantly with age (5% in preadolescents to 38% in master athletes, P 0.01). Subclinical heart diseases were found in 290/30109 (0.8%) and were common35 years (135/1124, 11%), but rare at 19-35 years (91/4442, 2%), very rare18 years (64/24 543, 0.2%; P 0.01). Seventy-four (0.3%) athletes were disqualified because of the structural alterations identified, 29 (0.1%) with cardiac structural diseases at risk for sudden death.Italian community-based pre-participation screening showed an age-dependent yield, with a three-fold increase in referral in athletes 35 years. Subclinical structural abnormalities potentially predisposing to sudden death were rare (0.01%), mostly in post-pubertal and senior athletes. Age-specific pre-participation screening protocols may help optimize resources and improve specificity.

However, the actual nutritional contribution of different N supply modes applied to peach crops, or the contribution of residual N in the subsequent cycle, is yet to be fully known. The current study aims to assess the annual and residual urea N contribution to the nutrition of peach trees grown under subtropical climate. Forty kilograms of N per hectare supplied as enriched urea (3.0 at. % N-15), were applied to peach in full production at a single rate (100 % at budding) or split in two rates (50 % at budding and 50 % at flowering). Total N-15 and N concentrations in the application year and in the year after treatments were assessed in peach leaves throughout the cycle, in fruits (pulp and stone) at harvest and in stratified soil samples. Total N-15, N concentrations and total dry mass were measured in annual and perennial tree organs in the year after treatment application. Peach trees evidenced higher N deriving from fertilizer (NDFF) in leaves and fruits (pulp and stone) in the year N was applied to the soil, as well as in the following year when N application was split into two rates. The highest NDFF amounts recorded in the year following N application were observed in leaves and fruits (annual organs) and thick roots (perennial organ), mainly when N was splitted. However, mainly in the year following its application, due to the small residual N, the N found in trees derived from sources other than N fertilizer; this justifies annual N applications, whenever necessary. The cultivation of cover crops and the preservation of organic matter could help N peach nutrition that seems to take advantage more of residual N in soil than on fertilizers. Sustainable plant production with the integration of eco-friendly agricultural practices, low chemical inputs, minimal deleterious effects on human health and low cost methods, is the need of the moment. In this direction, scientific/industrial community, continuously exploring novel and reliable methods. Plant growth promoting microbes are proving more promising to achieve eco-friendly and sustainable agricultural outcomes. In the present study, individuals of Foeniculum vulgare, a valuable aromatic plant species with potential medicinal value, were inoculated with different plant growth promoting microorganisms: (1) Arbuscular mycorrhizal like fungi (Sebacina vermifera) (2) Phosphate solubilizing bacteria (Pseudomonas fluorescens) (3) Azotobacter (Azotobacter chroococcum). Response of individual microorganism species was evaluated with reference to the emergence, plant growth and yield of essential oil along with qualitative effects on essential oil. Comparatively, significant response of Sebacina vermifera, in the stimulation of emergence of seeds, growth of plant and yield enhancement of essential oil was observed. An enhanced synthesis of anethole (major chemical constituent) was also recorded. Moreover, the enhancement in growth of plants was dependent on the extent of colonization percentage. A periodic study of growth parameters indicates plant’s health and vitality influenced by Sebacina vermifera. The enhanced essential oil of seeds along with enhanced synthesis of anethole was in agreement with the assumption that Sebacina vermifera trigger defensive responses and hence improve phytochemical production. A mechanistic insight is also illustrated. In conclusion Sebacina vermifera possesses immense potentials in the pursuit of agro-ecological attributes of medicinal plant cultivation and crop production. It exerts excellent growth effects and enhances phytochemical production in medicinal plants.

During the initial stages of drug use, cocaine-induced neuroadaptations within the ventral tegmental area (VTA) are critical for drug-associated cue learning and drug reinforcement processes. These neuroadaptations occur, in part, from alterations to the transcriptome. Although cocaine-induced transcriptional mechanisms within the VTA have been examined, various regimens and paradigms have been employed to examine candidate target genes. In order to identify key genes and biological processes regulating cocaine-induced processes, we employed genome-wide RNA-sequencing to analyze transcriptional profiles within the VTA from male mice that underwent one of four commonly used paradigms: acute home cage injections of cocaine, chronic home cage injections of cocaine, cocaine-conditioning, or intravenous-self administration of cocaine. We found that cocaine alters distinct sets of VTA genes within each exposure paradigm. Using behavioral measures from cocaine self-administering mice, we also found several genes whose expression patterns corelate with cocaine intake. In addition to overall gene expression levels, we identified several predicted upstream regulators of cocaine-induced transcription shared across all paradigms. Although distinct gene sets were altered across cocaine exposure paradigms, we found, from Gene Ontology (GO) term analysis, that biological processes important for energy regulation and synaptic plasticity were affected across all cocaine paradigms. Coexpression analysis also identified gene networks that are altered by cocaine. These data indicate that cocaine alters networks enriched with glial cell markers of the VTA that are involved in gene regulation and synaptic processes. Our analyses demonstrate that transcriptional changes within the VTA depend on the route, dose and context of cocaine exposure, and highlight several biological processes affected by cocaine. Overall, these findings provide a unique resource of gene expression data for future studies examining novel cocaine gene targets that regulate drug-associated behaviors.

Indicine breeds are the main source of beef products in tropical and subtropical regions. However, genetic improvement for carcass- and meat-quality traits in zebu cattle have been limited and genomics studies concerning structural variations that influence these traits are essential. Aim. The aim of this study was to perform a genome-wide association study between copy number variation regions (CNVRs) and carcass- and meat quality-traits in Nellore cattle. Methods. In total, 3794 animals, males and females included, were genotyped using a 777 962 single-nucleotide polymorphism platform of BovineHD BeadChip (777k; Illumina Inc.). Of these, 1751 Nellore bulls were slaughtered at 24 months of age for further carcass beef analysis. The following traits were studied: beef tenderness, marbling, rib-eye area, backfat thickness and meat colour (lightness, redness and yellowness). The CNV detection was conducted through PennCNV software. The association analyses were performed using CNVRuler software. Key results. Several identified genomic regions were linked to quantitative trait loci associated with fat deposition (FABP7) and lipid metabolism (PPARA; PLA2 family; BCHE), extracellular matrix (INS; COL10A1), contraction (SLC34A3; TRDN) and muscle development (CAPZP). The gene-enrichment analyses highlighted biological mechanisms directly related to the metabolism and synthesis of lipids and fatty acids. Conclusions. The large number of potential candidate genes identified within the CNVRs, as well as the functions and pathways identified, should help better elucidate the genetic mechanisms involved in the expression of beef and carcass traits in Nellore cattle. Several CNVRs harboured genes that might have a functional impact to improve the beef and carcass traits. Implications. The results obtained contribute to upgrade the sensorial and organoleptic attributes of Nellore cattle and make feasible the genetic improvement of carcass- and meat-quality traits. Phosphorus (P) availability in subtropical soils does not often meet the nutritional demand of native tree species such as the ipê-roxo tree (Handroanthus heptaphyllus), therefore, it is necessary to supply P at planting. However, the impact of P on root system growth remains unknown. The aim of the current study was to investigate the effect of P application on root morphology of H. heptaphyllus plants over a 36-month period in a subtropical climate region. During the experiment, the plants subjected to fertilization with 40 kg P ha−1 were compared to untreated control. Plant roots were scanned through minirhizotron system 18 and 36 months after transplant (MAT), and generated images were used to determine total root length, mean root diameter and total root volume. Plant height and leaf P concentrations were also evaluated. Phosphorus application enhanced root and whole plant growth with a more evident effect at 36 MAT, when soil P availability decreased. The results give important information on the cultivation of H. heptaphyllus plants in soils presenting low P availability.

Toxoplasmosis is a zoonotic food-borne disease caused by Toxoplasma gondii, a land-derived protozoan parasite that infects a broad range of terrestrial and aquatic hosts. T. gondii may reach coastal waters via contaminated freshwater runoff and its oocysts may enter into the marine food web. Marine invertebrates as mussels being filter feeders are exposed and may concentrate T. gondii oocysts representing a potential source of infection for animals and humans. The present works investigated the prevalence, parasite burden and genotypes of T. gondii in the Mediterranean mussels (Mytilus galloprovincialis) from southern Italy. We sampled a total of 382 individual Mediterranean mussels from May to August 2018 from seven production sites in the Gulf of Naples (Campania region). An additional sample including 27 farmed Mediterranean mussels was obtained in February 2018 from a mollusk depuration plant in Corigliano Calabro (Calabria region). T. gondii DNA was detected in 43 out of 409 (10.5%) Mediterranean mussels from seven out of eight sampling sites. The number of T. gondii copies/g in the digestive gland ranged from 0.14 to 1.18. Fragment analysis of Short Tandem Repeats (STRs) at 5 microsatellite loci was performed from 10 T. gondii PCR positive samples revealing the presence of five distinct genotypes including one corresponding to type I and four atypical genotypes. These findings suggest potential implications of epidemiological importance for human and animal health because both type I and atypical genotypes could be highly pathogenic.

Background : Cutaneous tumors are rarely described in avian and are frequently of viral origin. Solid tumors of vascular origin are seldom reported and usually result in difficult management by surgery alone. We describe the outcome of a subcutaneous low-grade epithelioid hemangioendothelioma (EHE) treated with the combination of surgery and electrochemotherapy. Case Description : A 10-year-old male budgerigar parakeet ( Melopsittacus undulatus ) was referred for evaluation of a 2-month non-healing exophytic mass on the left wing. The bird was bright, alert, and responsive, with a 2 × 1 cm proliferative lesion on the wing. Signs of discomfort were elicited by the clinical manipulation of the wing; no other abnormalities were detected during physical evaluation. Following hematological and imaging analysis, the parakeet was anesthetized and the mass was surgically removed. The histopathology report came back with a diagnosis of incompletely excised subcutaneous low-grade EHE. A surgical revision was not feasible due to the anatomical location and tumor extension. Adjuvant electrochemotherapy was chosen to increase the chance of tumor control. Two sessions of electrochemotherapy were performed with a 2-week interval between treatments using intralesional bleomycin followed by trains of permeabilizing electric pulses. Side effects were not observed and the parakeet was disease-free for 12 months when he died of acute renal failure. Conclusion : In lieu of the incomplete surgical excision of the tumor, electrochemotherapy resulted in good local control and cosmetic appearance and should be added to the standard oncological therapies for avian. Brown spot (Stemphylium vesicarium) is a fungal disease widespread in European pear industry. The high number of fungicide applications required to control the disease can promote strains with fungicide resistance. The aim of the present study was to assess the effectiveness of soil- applied water solution of calcium chloride in combination with rootstock to control the incidence of brown spot in Abbé Fétel pear. Soil salinity, stem water and osmotic potential, along with fruit fraction of calcium and defence-mechanism related-enzymes such as peroxidase, polyphenol oxidase, phenyl-alanine ammonia-lyase, and β-1,3-glucanase were investigate in tree grafted on Fox 11 (Pyrus communis L.) and Sydo®, a quince (Cydonia oblonga) rootstock. Both grafting combinations were fertigated or not with calcium chloride. Sydo® showed a positive effect in reducing brown spot severity on Abbé Fétel. Compared to Fox 11, fruits from trees grafted on Sydo® showed higher calcium concentrations, fraction of calcium pectate, fruit firmness, polyphenol oxidase, phenyl- alanine ammonia-lyase and β-1,3-glucanase levels. Fruit calcium concentration was positively correlated with disease fruit tolerance. However, soil calcium applications were ineffective in promoting fruit calcium partitioning. Calcium chloride applications decreased leaf osmotic and stem water potential in trees grafted onto Sydo®, but this wasn’t found in those grafted on Fox 11. In conclusion, Sydo® rootstock promoted Ca acquisition, osmotic adjustment at leaf and fruit level, fruit synthesis of defence-related enzymes that all together reduced brown spot severity in Abbé Fétel pear.

Purpose AL2106 is a new medical device based on a mixture of chondroitin sulphate in a xyloglucan and glycerol solution made to maximize its bioadhesive capability to the esophageal mucosa. The aim of the present study was twofold to evaluate the AL2106 protective effect on the esophageal mucosa when exposed to an acidic solution mimicking gastric reflux and to assess the resilience of this effect to saline washing. Materials and Methods A porcine ex vivo model was used and the effects of the new medical device were compared to a sodium alginate suspension (SAS) already present on the market which was assumed as reference. Mucosal damage was induced in 19 porcine esophagi by perfusion with an acidic solution added with pepsin, and Evans blue dye (EBD) tissue uptake was used as an indicator of mucosal permeability. The EBD penetration, expressed as EBD µg/g of dry tissue, was assessed in specimens of untreated damaged mucosa and in specimens treated with AL2106 or SAS. The same evaluation was carried out after washing with normal saline. Rapid antimicrobial susceptibility testing (AST) is an integral tool to mitigate the unnecessary use of powerful and broad-spectrum antibiotics that leads to the proliferation of multi-drug-resistant bacteria. Using a sensor platform composed of surface-enhanced Raman scattering (SERS) sensors with control of nanogap chemistry and machine learning algorithms for analysis of complex spectral data, bacteria metabolic profiles post antibiotic exposure are correlated with susceptibility. Deep neural network models are able to discriminate the responses of Escherichia coli and Pseudomonas aeruginosa to antibiotics from untreated cells in SERS data in 10 min after antibiotic exposure with greater than 99% accuracy. Deep learning analysis is also able to differentiate responses from untreated cells with antibiotic dosages up to 10-fold lower than the minimum inhibitory concentration observed in conventional growth assays. In addition, analysis of SERS data using a generative model, a variational autoencoder, identifies spectral features in the P. aeruginosa lysate data associated with antibiotic efficacy. From this insight, a combinatorial dataset of metabolites is selected to extend the latent space of the variational autoencoder. This culture-free dataset dramatically improves classification accuracy to select effective antibiotic treatment in 30 min. Unsupervised Bayesian Gaussian mixture analysis achieves 99.3% accuracy in discriminating between susceptible versus resistant to antibiotic cultures in SERS using the extended latent space. Discriminative and generative models rapidly provide high classification accuracy with small sets of labeled data, which enormously reduces the amount of time needed to validate phenotypic AST with conventional growth assays. Thus, this work outlines a promising approach toward practical rapid AST.

Wild boars represent a potential dissemination source of important pathogens to public and animal health, since they can host several pathogens. This study aimed to evaluate the health profile of wild boars hunted for population control in the state of Santa Catarina (SC), south of Brazil. For this study, tissue and blood samples were collected from 61 wild boars hunted from October 2017 to November 2018. All 61 serum samples were screened for antibodies by different assays, presenting the following seroprevalence: 52.4% for porcine circovirus type 2 (PCV2); 21.3% for Leptospira spp.; 19.7% for Mycoplasma hyopneumoniae; 13.1% for hepatitis E virus (HEV); and 9.8% for influenza A virus (IAV). No seroreaction was observed for Brucella spp. and classical swine fever virus (CSFV). At necropsy, the main lesions observed were evidence of pulmonary metastrongylid parasites, haemorrhagic lymph nodes and white-spotted liver. Histopathological analyses mainly showed interstitial pneumonia, bronchopneumonia and bronchi-associated lymphoid tissue (BALT) hyperplasia. There was a significant association between metastrongylid parasitism and BALT hyperplasia in lungs. Pathological findings of wild boars indicate that the assessed individuals were not exposed to notifiable diseases, such as CSFV, African swine fever virus and foot-and-mouth disease, which cause economic damage by non-tariff trade barriers. However, the detection of antibodies against PCV2 and M. hyopneumoniae, pathogens that impact commercial production, indicates the circulation of these agents, with frequency variation in wild populations. This shows the potential risk of pathogen transmission among domestic swine populations and free-living ones. The detection of antibodies against zoonotic agents such as HEV, IAV and Leptospira spp., other than characterizing the pathogens' circulation in these populations, suggests a potential risk to public health and pig farming, depending on the interactions established with the environment, humans and domestic swine.

Several protein-coding genes were identified within the genomic regions that explain more than 0.5% of the additive genetic variance for these traits. Pine tree invasions threaten many natural ecosystems of the Southern Hemisphere, modifying their structure and functioning through shifts in fire regimes, water balance, and biodiversity. The magnitude of such impacts depends on how much of the landscape has been invaded, thus a better understanding of the dispersal ability of pines and predictions of their future invasions are needed. Here we depict the spatio-temporal patterns of Pinus elliottii and Pinus taeda invading a new environment away from planted plots (i.e., invasion front), and discuss the underlying mechanisms that lead to a very concerning, yet poorly documented, pine invasion in central Argentina. Combining high-resolution imagery, allometric field data, and dendrochronology, we reconstructed the pine invasion into mountain grasslands from its onset in 1990. We found that even though the maximum density of invading pines (80 trees ha−1) was very low compared to adjacent plantation (1000 trees ha−1), density decreases exponentially with distance from the plantation edge. Remarkably, invading pines were found throughout the sampling plots showing high dispersal capacity, with no differences in age with increasing distance. The observed low density and spatially widespread exotic pine establishment, create a stealth type of invasion that is difficult to perceive in its early stages and challenging to manage once large areas are compromised. As invasion continues, long-distance dispersal will possibly become a major agent of landscape transformation and may lead to large pine-dominated neo-ecosystems, such as the savanna-like formation described here that replaced native grasslands in only three decades.

Rapid analyses methods for the assessment of soil microbiota are lacking. In a commercial farm tomato plants were subjected to different fertilization strategies: 1. mineral Control (C); 2. Organic amendment (O); 3. Organic amendment + Micosat F © biofertilizer (OM). A first rapid method (Litterbag-NIRS) concerned hay litterbags coupled with a smart SCiOTM device. A second method (Foliar-NIRS) used the same device on the leaves. The plants showed positive responses to the amendment and biofertilization in the yield: C 60.5.1 t ha-1vs. 70.8 in O (+17%) and 74.2 in OM (+23% from C and + 5% (P 0.08) from O). The use of Litterbag-NIRS fingerprinting, completed with litterbags phenotyping and elaborated with a multivariate support vector machine classifier provided a similar knowledge to that obtained from microbial and chemical analyses of the soil. The reason for this response is that the analyses were embedded in the Litterbag-NIRS at medium-high precision. A polydromic function was hypothesized in order to disentangle the activities of different soil microbial populations from each other. The organic amendment delayed the functionality of the rapid r-strategist microbial populations, but at the same time activated slow k-strategists to intake the walls of the hay inside the litterbags. In this sense, the Litterbag-NIRS test can provide an effective “swamp” of the microbial fertility of the soil. Briefly, the Litterbag-NIRS coupled with Foliar-NIRS accounted for 95% of the average yield results, and both are therefore recommended for a rational assessment of microbial soil fertility.

Objective: The present study was designed to investigate phytopromotional effects of Sebacina vermifera on economically and medicinally important aromatic plant - Coriandrum sativum (coriander). Methods: Phytopromotional effects of Sebacina vermifera were evaluated on coriander, under greenhouse and field conditions. The evaluations were carried out with reference to emergence, growth promotion and quantitative as well as the qualitative composition of essential oil. Beside this the overall effects were comparatively assessed with the effects of (a) Phosphate solubilizing bacteria (Pseudomonas fluorescens) (b) Nitrogen-fixing bacteria (Azotobacter chroococcum) on coriander using same parameters. Results: Mycorrhizal fungus (Sebacina vermifera) was observed with the most significant effect in all aspects viz. emergence, growth promotion and quantitative as well as the qualitative composition of essential oil. Conclusion: Based upon the observations, Sebacina vermifera is highly recommended as a potential biological agent that could be applied for phytopromotional effects and economic cultivation of aromatic plants.

This review addresses the potential role of antioxidants and methyl-group sources in optimising the metabolic health of dairy ruminants. The productivity of high-yielding dairy cows has increased over the past 40 years and the milk yield has doubled. Such increases in milk production have been observed not only in dairy cows but also to some extent in other dairy ruminants such as ewes, goats and buffaloes (Bubalus bubalis). As a consequence, in all specialized dairy ruminants it is essential to optimize the macro and micro-nutrient supply, especially during the most critical period in the animals' production cycle i.e. from parturition until the peak of lactation. In this critical phase, an array of factors can enhance the balance between the intake and demand for nutrients, although the availability and supply of the selected micronutrients is also important. The supplementation of dietary antioxidants or boosting the endogenous methyl group status, via vitamin E, selenium and choline are proposed as possible strategies in maintaining stable metabolic health and optimising milk production.

Understanding how the genome is structurally organized as chromatin is essential for understanding its function. Here, we review recent developments that allowed the readdressing of old questions regarding the primary level of chromatin structure, the arrangement of nucleosomes along the DNA and the folding of the nucleosome fiber in nuclear space. In contrast to earlier views of nucleosome arrays as uniformly regular and folded, recent findings reveal heterogeneous array organization and diverse modes of folding. Local structure variations reflect a continuum of functional states characterized by differences in post-translational histone modifications, associated chromatin-interacting proteins and nucleosome-remodeling enzymes. Technological advances have led to new insights into genome-wide arrangements of nucleosomes along the DNA and the folding of the chromosome fiber in nuclear space, revealing unexpected diversity.

Nutrient management is vital for food, feed, fiber, and fuel production. However, excessive application and loss (volatilization, leaching, run-off, etc.) of inorganic and organic sources of nutrients have detrimental environmental impacts, while increasing prices for petroleum-based and mined fertilizers further limit opportunities for their utilization in developing nations. This study evaluated a novel, alternative type of nutrient source through pretreatment processes of torrefaction and pyrolysis by converting high-biomass feedstocks into renewable soil amendments. Napiergrass (Pennisetum purpureum Schumach., [cv. Merkeron]) and pearl millet—napiergrass (Pennisetum glaucum [L.] R. Br. × P. purpureum Schumach.) (PMN) were converted under atmospheric pressure with minimal oxygen at 250°C and 400°C, ground to 1 millimeter (mm) and 2 mm particle sizes, and compared to urea in a full-season field trial and short-season nursery trial growing maize (Zea mays L.) and PMN for fertility response. When compared to urea in the field trial, the torrefied biomass amendment (TBA) and biochar had similar responses despite lower nitrogen (N) application rates. The nursery trial also produced equivalent responses from urea and TBA regardless of lower N application with the exception being phosphorus (P). Finally, maize and PMN had higher P uptake with the TBA in both trials.

The State of Espirito Santo (Brazil) has 9.4 % of its area cultivated in full sun, monocrop coffee. Coffee has a great importance for the State and a dependenceon just one product can make farmers vulnerable to price fluctuations. Integrating coffee (Coffea canephora Pierre ex Froehner) intercropped withAmbarella (Spondias dulcis Parkinson) offers environmental benefits and an extra financial combination through the commercialization of Ambarellafruits. The goal of this study was to verify the effects of the distance and orientation (East-West versus North-South) between Ambarella on coffee cropyield. We assessed two different coffee crops spaced (3 x 1.5) m. Both were intercropped with Ambarella spaced (15 x 7.5) m. The average coffee productivityvaried from (2,177 to 7,346) kg plant-1. The lowest productivity was measured in the North-South orientation, during 2016, and the highest in theEast-West orientation during 2017. Our findings also indicate that regardless of planting distance, Ambarella did not significantly affected coffee productivitydemonstrating that it can be a promising fruit tree to intercrop with Conilon coffee.

Ця стаття узагальнює аргументи та контраргументи в межах наукової дискусії з питання менеджменту інновацій в публічному управлінні. Вказано, що специфіка впровадження інновацій в публічному управління має свої характерні особливості, які полягають у впровадженні системи організаційного контролю. Здійснено аналіз елементів системи організаційного контролю: система управління якістю, адміністративний контроль, внутрішній аудит, які впливають на процеси прийняття рішень та результативності діяльності. Окреслюються специфічні функціональні можливості досліджуваних інструментів: внутрішній аудит застосовується для перевірки відповідності процедур; адміністративний контроль застосовується в області управління бюджетуванням; система якісного менеджменту застосовується для регламентації процедур. Зазначено, що попри значні потенційні можливості вищезазначених інструментів, недостатня ефективність та фрагментарність їх використовуються не приносять значного синергетичного ефекту. Основною метою проведеного дослідження є виявлення факторів, що дозволяють сприятливо впливати на ефективність впровадження системи організаційного контролю в громадських організаціях. Об’єктом дослідження є три громадські організації в Марокко. В статті представлено результати емпіричного аналізу, який засвідчив необхідність зміни системи менеджменту досліджуваних організацій на основі врахування дії двох факторів: одночасності (тобто одночасному впровадженні досліджуваних інструментів) та оригінальності асигнувань. За результатами дослідження запропоновано низку доповнень до моделі асигнувань Де Вауяні, а саме уточнення факторів прогнозування, які впливають на кожен етап процесу асигнування досліджуваних інструментів контролю (внутрішній аудит, адміністративний контроль та система якісного менеджменту). Результати проведеного дослідження можуть бути корисними для громадських організацій при проведенні оцінки ефективності запровадження інструментів організаційного контролю та вибору напрямів стимулювання їх використання. The race for performance has led some public service organizations to put in place some management innovations such as the implementation of organizational control. Organizational control is a system, a set of mechanisms and tools such as Quality management system, Management controlling, Internal auditing etc that affects all levels of the organization (strategic and operational levels), to master the decision-making processes, actions and results. Those organizational control tools don’t bring all expected effects because they are under-used. Internal auditing is used mainly for compliance procedures, management controlling for budgeting and Quality Management, a system for the prescription of procedures which is only a small percentage of the potential functionalities of these tools used punctually or inducing inappropriate behaviours. Those behaviours deprive these organizations of a significant source of synergies and performance. Those observations led to the questions of change management, learning organization, etc that can be included in a general question of appropriation of controlling tools considering Simultaneity Dimension (Simultaneous deployment of those tools), an original dimension little studied in research on appropriation and which corresponds more to the reality of those companies. Appropriation refers to two conceptions. Appropriation as a process inducing the acquisition of knowledge or behaviours concerning an object. Appropriation as a state which implies the assessment of the gap between how an object is integrated by users and its conceptual definition. Appropriation implies consistency between an object and its use. This research is a multiple case study comparing 3 companies, which allowed the enrichment of the appropriation model proposed by De Vaujany (2006) by specifying the predictive factors affecting each of the phases of this process of appropriation of control tools (Internal Auditing, Management controlling and Quality Management System) including the impact of simultaneity in the context of public service organizations in Morocco. For managerial use, those models constitute a frame or a diagnosis methodology to assess the appropriation of organizational control tools for the stimulation of their usage and therefore improve the multidimensional performance of the public service and citizen’s satisfaction.

Long noncoding RNAs (lncRNAs) have been identified in all eukaryotes and are most abundant in the human genome. However, the functional importance and mechanisms of action for human lncRNAs are largely unknown. Using comparative sequence, structural, and functional analyses, we characterize the evolution and molecular function of human lncRNA JPX. We find that human JPX and its mouse homolog, lncRNA Jpx, have deep divergence in their nucleotide sequences and RNA secondary structures. Despite such differences, both lncRNAs demonstrate robust binding to CTCF, a protein that is central to Jpx's role in X chromosome inactivation. In addition, our functional rescue experiment using Jpx-deletion mutant cells shows that human JPX can functionally complement the loss of Jpx in mouse embryonic stem cells. Our findings support a model for functional conservation of lncRNAs independent from sequence and structural divergence. This study provides mechanistic insight into the evolution of lncRNA function.

Thus, this trait is generally not included in animal breeding programs. The use of molecular techniques to elucidate intramuscular fat deposition may help improve this trait. In this respect, transcriptome studies and differential gene expression analysis by RNA-Seq can contribute to advances in this area. The objective of this study was to use RNA-Seq to identify differentially expressed genes (DEGs) in muscle tissue (longissimus thoracis) of Nellore cattle divergently ranked on marbling, in order to increase our understanding of genes involved in the expression of this trait. The results revealed 49 DEGs and three hub genes (CISH, UFM1, TSHZ1), all of them involved in insulin and diabetes mellitus metabolism. These results indicating key genes and pathways, which may help to develop strategies designed to select animals with greater marbling. Listeria monocytogenes is a bacterium that infect humans and animals and causes a zoonotic disease characterized by encephalitis, septicemia or abortion. In addition, listeriosis leads to significant economic losses due to animal death and sacrifice. This research compared the technique of immunofluorescence (IF) and immunohistochemistry (IHC) for the diagnosis of L. monocytogenes in formalin-fixed and paraffin-embedded (FFPE) tissues. A total of 30 tissue blocks from 15 animals with history and/or lesions compatible with listeriosis were selected. For both IHC and IF, the same diluted (1:200) polyclonal primary antibody was used against L. monocytogenes serotypes 1 and 4. For IHC, a polymer secondary antibody conjugated to peroxidase (HRP) was used. For IF, samples were incubated with a fluorescein-labeled anti-rabbit IgG secondary antibody. Each sample was classified according to the presence and percentage of immunolabeling area. From 30 samples, 10 were positive at least for one technique, whereas eight samples were positive for both IHC and IF with similar score. There was strong immunolabeling in tissue samples from bovines experimentally infected with L. monocytogenes ATCC 7644, as well as in nervous tissues from naturally infected ruminants. Additionally, IF did not show any difference in sensitivity when compared to IHC. Using processed biological materials for IF, instead of fresh tissues, is a quite unique technique, since there are few protocols described. Therefore, this study demonstrated that both techniques are efficient to detect L. monocytogenes in FFPE tissues. RESUMO: Listeria monocytogenes é uma bactéria que infecta humanos e animais, podendo causar uma doença zoonótica caracterizada por encefalite, septicemia e abortos. Além disso, a listeriose resulta em perdas econômicas significativas pela morte e sacrifício de animais. O objetivo deste trabalho foi comparar a técnica de imunofluorescência (IF) e imuno-histoquímica (IHQ) para detecção de L. monocytogenes em tecidos fixados e parafinizados. Foram selecionados 30 blocos de tecidos de 15 animais com histórico e/ou lesões compatíveis com listeriose. Para ambas as técnicas, foi utilizado o mesmo anticorpo primário policlonal diluído (1:200) para detecção de L. monocytogenes sorotipos 1 e 4. Para a IHQ foi utilizado anticorpo secundário em polímero acoplado a peroxidase (HRP). Para a IF, as amostras foram incubadas com anticorpo secundário anti-IgG de coelho marcado com fluoresceína. Cada amostra foi classificada quanto à presença de imunomarcação e porcentagem de área positiva. Das 30 amostras, 10 foram positivas em pelo menos uma das técnicas, sendo oito amostras positivas em ambas IHQ e IF com o mesmo escore. Houve forte imunomarcação tanto em amostras de bovino experimentalmente infectado com L. monocytogenes ATCC 7644, como em amostras de tecido nervoso de ruminantes naturalmente infectados. Além disso, a IF não apresentou diferença na sensibilidade quando comparada com a IHQ. O uso de materiais biológicos processados para IF, ao invés de tecidos frescos, é algo inovador, uma vez que existem poucos protocolos descritos. Portanto, este estudo demonstrou que as duas técnicas foram eficientes para detectar L. monocytogenes em tecidos fixados em formol e parafinizados.

Spaghetti meat (SM) is a recent muscular abnormality that affects the Pectoralis major muscle of fast-growing broilers. As the appellative suggests, this condition phenotypically manifests as a loss of integrity of the breast muscle, which appears soft, mushy, and sparsely tight, resembling spaghetti pasta. The incidence of SM can reach up to 20% and its occurrence exerts detrimental effects on meat composition, nutritional value, and technological properties, accounting for an overall decreased meat value and important economic losses related to the necessity to downgrade affected meats. However, due to its recentness, the causative mechanisms are still partially unknown and less investigated compared to other muscular abnormalities (i.e., White Striping and Wooden Breast), for which cellular stress and hypoxia caused by muscle hypertrophy are believed to be the main triggering factors. Within this scenario, the present review aims at providing a clear and concise summary of the available knowledge concerning SM abnormality and concurrently presenting the existing research gaps, as well as the potential future developments in the field.

Background: In patients with Multiple Sclerosis (pwMS) disease-modifying therapies (DMTs) affects immune response to antigens. Therefore, post-vaccination serological assessments are needed to evaluate the effect of the vaccine on SARS-CoV-2 antibody response. Methods: We designed a prospective multicenter cohort study enrolling pwMS who were scheduled for SARS-Cov-2 vaccination with mRNA vaccines (BNT162b2, Pfizer/BioNTech,Inc or mRNA-1273, Moderna Tx,Inc). A blood collection before the first vaccine dose and 4 weeks after the second dose was planned, with a centralized serological assessment (electrochemiluminescence immunoassay, ECLIA, Roche-Diagnostics). The log-transform of the antibody levels was analyzed by multivariable linear regression. Findings: 780 pwMS (76% BNT162b2 and 24% mRNA-1273) had pre- and 4-week post-vaccination blood assessments. 87 (11·2%) were untreated, 154 (19·7%) on ocrelizumab, 25 (3·2%) on rituximab, 85 (10·9%) on fingolimod, 25 (3·2%) on cladribine and 404 (51·7%) on other DMTs. 677 patients (86·8%) had detectable post-vaccination SARS-CoV-2 antibodies. At multivariable analysis, the antibody levels of patients on ocrelizumab (201-fold decrease (95%CI=128–317), p < 0·001), fingolimod (26-fold decrease (95%CI=16–42), p < 0·001) and rituximab (20-fold decrease (95%CI=10–43), p < 0·001) were significantly reduced as compared to untreated patients. Vaccination with mRNA-1273 resulted in a systematically 3·25-fold higher antibody level (95%CI=2·46–4·27) than with the BNT162b2 vaccine (p < 0·001). The antibody levels on anti-CD20 therapies correlated to the time since last infusion, and rituximab had longer intervals (mean=386 days) than ocrelizumab patients (mean=129 days). Interpretation: In pwMS, anti-CD20 treatment and fingolimod led to a reduced humoral response to mRNA-based SARS-CoV-2 vaccines. As mRNA-1273 elicits 3·25-higher antibody levels than BNT162b2, this vaccine may be preferentially considered for patients under anti-CD20 treatment or fingolimod. Combining our data with those on the cellular immune response to vaccines, and including clinical follow-up, will contribute to better define the most appropriate SARS-CoV-2 vaccine strategies in the context of DMTs and MS. The aim of the present experiment was to determine if the supply of biofertilizers could differently stimulate the native microbiota, thus determining different patterns of organic material decomposition processes. The microbial composition of soil and litter was investigated by next generation sequencing using a metabarcoding approach. The chemical structure of the decomposing litterbags was investigated through the TG-DTA analysis and NIR spectroscopy. The study was conducted in an apricot orchard in Italy, and two different type of biofertilizers (AMF and Trichoderma spp.) were compared to unfertilized control over one year. Bacteria and fungi in soil, 162 days from litter deposition, evidenced differentiated clusters for control and both biofertilizers, on the other hand, only fungal composition of litterbags was modified as a consequence of Trichoderma spp. supply, no effect was observed in the bacterial community of litterbags. NIR and TG-DTA analysis evidenced a significant change over time of the chemical composition of litterbags with a faster degradation as a consequence of Trichoderma spp. supply testified by a higher degradation coefficient (1.9) than control (1.6) and AMF (1.7). The supply of biofertilizers partially modified the bacteria community of soil, while Trichoderma spp. Influenced the fungal community of the litter. Moreover, Trichoderma spp. Evidenced a faster and higher degradation of litter than AMF-biofertilizers, laying the foundation for an efficient use in orchard.

The present study aimed at assessing the impact of lysine restriction performed during different feeding phases on growth performances, meat quality traits and technological properties as well as on the incidence and severity of breast muscle abnormalities. For this purpose, a total of 945 one-day-old Ross 308 male chicks was randomly divided into three experimental groups: CONT, fed a four feeding phases commercial diet, GRW I, and GRW I + II fed CONT diet with the depletion of synthetic lysine during grower I and grower I and II feeding phases, respectively. Productive performances were recorded throughout the whole rearing cycle and the incidence of breast muscle growth-related abnormalities assessed at slaughter (49 d) on 280 breasts/group. Quality traits and technological properties of breast meat were measured on a total of 54 Pectoralis major muscles. Lysine restriction only marginally affected the productive performances and the quality parameters of breast meat. The increased (p &lt, 0.05) solubility of the protein fraction along with the remarkably higher (p &lt, 0.05) anserine content found in GRW I + II suggests an increased energy requirement in the pectoral muscles belonging to lysine-restricted birds and supports the hypothesis of a reduced protein synthesis taking place within these muscles.

Immune checkpoint blockade (ICB) therapies have significantly prolonged patient survival across multiple tumor types, particularly in melanoma. Interestingly, sex-specific differences in response to ICB have been observed, with males receiving a greater benefit from ICB than females, although the mechanism or mechanisms underlying this difference are unknown. Mining published transcriptomic data sets, we determined that the response to ICBs is influenced by the functionality of intratumoral macrophages. This puts into context our observation that estrogens (E2) working through the estrogen receptor α (ERα) stimulated melanoma growth in murine models by skewing macrophage polarization toward an immune-suppressive state that promoted CD8+ T cell dysfunction and exhaustion and ICB resistance. This activity was not evident in mice harboring macrophage-specific depletion of ERα, confirming a direct role for estrogen signaling within myeloid cells in establishing an immunosuppressed state. Inhibition of ERα using fulvestrant, a selective estrogen receptor downregulator (SERD), decreased tumor growth, stimulated adaptive immunity, and increased the antitumor efficacy of ICBs. Further, a gene signature that determines ER activity in macrophages predicted survival in patients with melanoma treated with ICB. These results highlight the importance of E2/ER signaling as a regulator of intratumoral macrophage polarization, an activity that can be therapeutically targeted to reverse immune suppression and increase ICB efficacy.

Lead (Pb) contamination risks to crops grown in urban and peri-urban soils is a great concern that should be better evaluated to define the Pb maximum levels in soils for safe cultivation and to identify suitable strategies to remediate Pb polluted urban soils. The objective of this work was to evaluate the potential risk for human health from the ingestion of the edible portions of barley, castor bean, common bean, Indian mustard, sorghum, spinach, and tomato grown in an unpolluted soil (initial Pb content 32.6 mg kg−1) spiked with 0, 300, 650, 1000 mg Pb kg−1, respectively. The potential possibility of using these plants to phyto-remediate the soil of Pb was also assessed. Pot trials were conducted for two years (2008 and 2009). Results highlighted that all the investigated species were able to attain growth to maturity in high Pb spiked soil, although Pb influenced dry matter accumulation. Even in soils with low Pb concentrations, Pb accumulated the edible parts. Noteworthy, even in untreated control soils, all tested species revealed a Pb concentration in the edible parts that was higher than the safe limit set by FAO/WHO. None of the investigated species were considered Pb hyperaccumulators, but all were shown to be potentially suitable for phyto-stabilization.

No. of bitstreams: 0 Previous issue date: 2021-11-01 This study aimed to identify structural variations in the form of runs of homozygosity (ROH) and copy number variants (CNVs) in the genome of the Brazilian Senepol cattle and to better explore the potential biological functions of the genes located within such regions. A total of 140 animals were genotyped with the GeneSeek® Genomic Profiler™ Bovine 50 K. Autosomal ROH and CNVs were detected after appropriated quality control. A total of 5,531 ROH were identified, with an average number of 40.37 per animal and an average length of 5.09 Mb. BTA1 had the greatest number of ROH per chromosome (n = 458), while BTA20 displayed the greatest fraction of the chromosome covered with ROH (22.06%). A total of 181 CNVs were identified, with an average length size of 184.67 kb, with values ranging from 26.61 to 334.48 kb. A total of 16 CNV regions (CNVRs) were seen distributed among eleven autosomes, and the largest number of CNVRs (n = 4) was described on BTA12. Autozygosity islands were identified using an outlier approach, and none of them overlapped with CNVRs. Our study revealed that the limited genetic basis together with the narrow number of imported animals to disseminate the breed might have strongly contributed to the low effective population size and the high genomic autozygosity proportion described in this population. International audience; The molecular complexity of human breast cancer (BC) renders the clinical management of the disease challenging. Long non-coding RNAs (lncRNAs) are promising biomarkers for BC patient stratification, early detection, and disease monitoring. Here, we identified the involvement of the long intergenic non-coding RNA 01087 (LINC01087) in breast oncogenesis. LINC01087 appeared significantly downregulated in triple-negative BCs (TNBCs) and upregulated in the luminal BC subtypes in comparison to mammary samples from cancer-free women and matched normal cancer pairs. Interestingly, deregulation of LINC01087 allowed to accurately distinguish between luminal and TNBC specimens, independently of the clinicopathological parameters, and of the histological and TP53 or BRCA1/2 mutational status. Moreover, increased expression of LINC01087 predicted a better prognosis in luminal BCs, while TNBC tumors that harbored lower levels of LINC01087 were associated with reduced relapse-free survival. Furthermore, bioinformatics analyses were performed on TNBC and luminal BC samples and suggested that the putative tumor suppressor activity of LINC01087 may rely on interferences with pathways involved in cell survival, proliferation, adhesion, invasion, inflammation and drug sensitivity. Altogether, these data suggest that the assessment of LINC01087 deregulation could represent a novel, specific and promising biomarker not only for the diagnosis and prognosis of luminal BC subtypes and TNBCs, but also as a predictive biomarker of pharmacological interventions.

We describe an original electroporation protocol for in vivo plasmid DNA transfection. The right hind limbs of C57 mice are exposed to a specifically designed train of permeabilizing electric pulses by transcutaneous application of tailored needle electrodes, immediately after the injection of pEGFP-C1 plasmid encoding GFP (Green Fluorescente Protein). The electroporated rodents show a greater GFP expression than the controls at three different time points (4, 10, and 15 days). The electroporated muscles display only mild interstitial myositis, with a significant increase in inflammatory cell infiltrates. Finally, mild gait abnormalities are registered in electroporated mice only in the first 48 h after the treatment. This protocol has proven to be highly efficient in terms of expression levels of the construct, is easy to apply since it does not require surgical exposure of the muscle and is well tolerated by the animals because it does not cause evident morphological and functional damage to the electroporated muscle.

Cystic echinococcosis (CE) is a severe zoonosis, caused by the larval stage of the tapeworm Echinococcus granulosus. This helminth infection is of increasing public health and socio-economic concern due to the considerable morbidity rates that cause economic losses both in the public health sector and in the livestock industry. Control programmes against E. granulosus are considered long-term actions which require an integrated approach and high expenditure of time and financial resources. Since 2010, an integrated approach to control CE has been implemented in a highly endemic area of continental southern Italy (Campania region). Innovative procedures and tools have been developed and exploited during the control programme based on the following strategies: i) active and passive surveillance in livestock (using geospatial tools for georeferencing), ii) diagnosis in dogs (using the FLOTAC techniques and molecular analysis), iii) targeted treatment of farm dogs (using purpose-built confinement cages), iv) early diagnosis in livestock (by ultrasonography), v) surveillance in humans (through hospital discharge records analysis), vi) monitoring the food chain (analysing raw vegetables), vii) outreach activities to the general public (through dissemination material, e.g. brochures, gadgets, videos, virtual reality). Over eight years, the integrated approach and the new strategies developed have resulted in a noteworthy reduction of the parasite infection rates in livestock (e.g. up to 30 % in sheep). The results obtained so far highlight that using a one health multidisciplinary and multi-institution effort is of pivotal importance in preparing CE control programmes at regional level and could be extended to other endemic Mediterranean areas.

Over the past decade, the poultry industry has faced an increasing occurrence of growth-related muscular abnormalities that mainly affect fast-growing genotypes selected for their production performances (high growth rate and breast yield). These abnormalities, termed white striping (WS), wooden breast (WB) and spaghetti meat (SM), primarily affect the superficial portion of pectoralis major muscles. Despite their distinctive phenotypes, WS, WB, and SM conditions entail common histological features, i.e., they might share common causative mechanisms underpinning their occurrence. Meat affected by growth-related abnormalities is harmless for human nutrition since no specific biological or chemical hazards have been found to be related to its consumption. However, WS, WB, and SM abnormalities negatively affect both quality traits and technological properties of raw and processed meat, causing relevant economic damages in the poultry industry. This paper aims to provide an update about the current status of poultry meat abnormalities, giving useful insights about their impact on meat quality, the possible causative mechanisms, methods for mitigation, and future perspectives.

Turkey meat is the second most consumed poultry meat worldwide and represents an economic source of high-quality protein for human consumption. To fulfill the increasing demand for turkey meat, breeding companies have been selecting genetic lines with increased growth potential and breast muscle proportion. Moreover, the progressive shift toward further processed products has emphasized the need for higher standards in poultry meat to improve its technological characteristics and functional properties (i.e., water-holding capacity). However, as observed for broiler chickens, a growing body of scientific evidence suggests that the intense selection for the aforementioned traits could be associated with a greater occurrence of growth-related myopathies and abnormalities and, consequently, to increased downgrading rates and overall reduction of meat quality characteristics. In the past, muscle abnormalities such as deep pectoral myopathy, pale-soft-and-exudative-like meat, and focal myopathy have been reported in turkey lines selected for increased growth rate. In addition, the presence of white striations in the superficial layer of pectoralis major muscle, as well as the tendency of muscle fiber bundles to separate resulting in an altered breast muscle structure, has been detected in commercial turkey abattoirs. Furthermore, past investigations revealed the presence of another quality issue depicted by an overall toughening of the breast muscle. These meat abnormalities seem to macroscopically overlap the white striping, spaghetti meat, and wooden breast conditions observed in pectoral muscle of fast-growing, high-breast-yield chicken hybrids, respectively. Considering the high economic impact of these growth-related abnormalities in broilers, there is an increasing interest of the turkey industry in estimating the occurrence and the impact of these meat quality issues also in the modern turkey lines. Studies have been recently conducted to assess the effect of the genotype on the occurrence of these emerging growth-related defects and to evaluate how meat quality properties are affected by white-striping condition in turkeys, respectively. Therefore, this review aims to provide a critical overview of the current understanding regarding the growth-related abnormalities and their impact on meat quality in modern turkey hybrids with the hope that this information may improve the knowledge concerning their overall effect on poultry meat.

In this study, whole-genome re-sequencing data from indicine and Brazilian locally adapted taurine cattle breeds were used to detect genomic regions under selective pressure. Within-population and cross-population statistics were combined separately in a single score using the de-correlated composite of multiple signals (DCMS) method. Putative sweep regions were revealed by assessing the top 1% of the empirical distribution generated by the DCMS statistics. RESULTS: A total of 33,328,447 biallelic SNPs with an average read depth of 12.4X passed the hard filtering process and were used to access putative sweep regions. Admixture has occurred in some locally adapted taurine populations due to the introgression of exotic breeds. The genomic inbreeding coefficient based on runs of homozygosity (ROH) concurred with the populations' historical background. Signatures of selection retrieved from the DCMS statistics provided a comprehensive set of putative candidate genes and revealed QTLs disclosing cattle production traits and adaptation to the challenging environments. Additionally, several candidate regions overlapped with previous regions under selection described in the literature for other cattle breeds. CONCLUSION: The current study reported putative sweep regions that can provide important insights to better understand the selective forces shaping the genome of the indicine and Brazilian locally adapted taurine cattle breeds. Such regions likely harbor traces of natural selection pressures by which these populations have been exposed and may elucidate footprints for adaptation to the challenging climatic conditions. The elevated ultimate pH (pH(u)) found in wooden breast (WB) meat suggests an altered muscular energetic status in WB but also could be related to a prematurely terminated post-mortem pH decline. The aims of this study were to explore the factors contributing to the elevated pH(u) and establish whether the occurrence of WB defect alters muscle post-mortem carbohydrate metabolism and determine if the contractile apparatus reflects such changes. A total of 24 carcasses from Ross 308 male chickens were obtained from a commercial producer and harvested using commercial processing procedures. Carcasses were categorized into unaffected (NORM) and WB groups (n = 12 each), and samples were collected from cranial bone-in pectoralis major (PM) muscles at 15 min and 24 h post-mortem for the determination of pH, glycolytic metabolites, adenonucleotides, buffering capacity, phosphofructokinase (PFK) activity, and in vitro pH decline. Twenty-four additional deboned PM samples (12 NORM and 12 WB) were collected from the same processing plant to assess muscle histology and sarcomere length at four different locations throughout the PM muscle. Data show that the reduced glycolytic potential of WB muscles only partially explains the higher (P < 0.001) pH(u) of WB meat, as residual glycogen along with unaltered PFK activity suggests that neither glycogen nor a deficiency of PFK is responsible for arresting glycolysis prematurely. The dramatic reduction in ATP concentrations in the early post-mortem period suggests a defective ATP-generating pathway that might be responsible for the reduced pH decline in WB samples. Further, the addition of excess of ATPase extended post-mortem glycolysis of WB meat in an in vitro glycolytic system. WB-affected samples have longer (P < 0.001) sarcomeres compared to NORM, indicating the existence of compromised energy-generating pathways in myopathic muscles that may have had consequences on the muscle contraction and tension development, as in vivo, also during the post-mortem period. Considering the overall reduced glycolytic potential and the myodegenerative processes associated with WB condition, we speculate that the higher pH(u) of WB meat might be the outcome of a drastically impaired energy-generating pathway combined with a deficiency and/or a dysfunction of muscle ATPases, having consequences also on muscle fiber contraction degree.

Multiple myeloma (MM) is a hematologic malignancy produced by a clonal expansion of plasma cells and characterized by abnormal production and secretion of monoclonal antibodies. This pathology exhibits an enormous heterogeneity resulting not only from genetic alterations but also from several epigenetic dysregulations. Here we provide evidence that Che-1/AATF (Che-1), an interactor of RNA polymerase II, promotes MM proliferation by affecting chromatin structure and sustaining global gene expression. We found that Che-1 depletion leads to a reduction of “active chromatin” by inducing a global decrease of histone acetylation. In this context, Che-1 directly interacts with histones and displaces histone deacetylase class I members from them. Strikingly, transgenic mice expressing human Che-1 in plasma cells develop MM with clinical features resembling those observed in the human disease. Finally, Che-1 downregulation decreases BRD4 chromatin accumulation to further sensitize MM cells to bromodomain and external domain inhibitors. These findings identify Che-1 as a promising target for MM therapy, alone or in combination with bromodomain and external domain inhibitors.
