Digital, Palm Oil Mill Effluent (POME) requires treatment before disposal due to its high organic matter content. In this study, the electrical performance and wastewater treatment efficiency were evaluated for Microbial Fuel Cells (MFC) treating unsterile POME with chemical oxygen demand (COD) from 200 to 10 000 mg/L. Since the inoculum type is a key factor in MFC performance, three types of sludge (methanogenic sludge (MS), facultative sludge (FS), and dry sludge (DS), obtained from the current POME treatment ponds were evaluated as inoculum. Dry sludge (DS) developed a maximum power output of 3.30 W/m3 by oxidizing 71% out of the COD provided by POME (1000 mg/L). Also, raw POME microbiota contributed to an enrichment of the community in DS inoculum along with the operation, in which Geobacter was the predominant genus reaching a current generation of 247 mA/m2 and a power density of 2.36 W/m3. Conversely, pure electrogenic (Shewanella sp.) inoculation led to a diversification process, resulting in a lower current generation of 52 mA/m2 and a power density of 0.10 W/m3. Consequently, microbial community dynamics revealed that MFC inoculation tends to a microbial equilibrium wherein generation of high current density was achieved by gradual microbial enrichment rather than external electrogenic invasion., Ciencias Exacta y Naturales

Yellow passion fruit (Passiflora edulis f. flavicarpa) is rich in nutrients. In March 2018, anthracnose symptoms were observed in a storeroom in Baoting, Hainan province, China. The pathogen namely Colletotrichum brevisporum was isolated and identified from symptomatic fruits based on the morphological and multi-gene phylogenetic analyses derived from an internal transcribed spacer (ITS), beta-tubulin (β-tub), fragments of actin (ACT), and glyceraldehyde-3-phosphate dehydrogenase (GAPDH). A pathogenicity test and revalidation of Koch’s postulates indicated that the fungus was cause of disease on yellow passion fruit. And the cause of rot on yellow passion fruit is C. brevisporum in China.

Hydrothermal pretreatment (HTP) of sewage sludge (SS) has been shown to improve the subsequent biogas production by anaerobic digestion (AD), but the effect of catalysts on HTP performance was less explored. This study intended to investigate the SS pretreatment by wet air oxidation (WAO) with the addition of K2CO3 as a catalyst on the performance of methane production by AD. WAO was found to improve the solubilization of SS, the soluble chemical oxygen demand, dissolved organic carbon, and total dissolved nitrogen. The methane yield from WAO increased from 202 mL/gVSin with no catalyst added to 277 mL/gVSin with 10 wt% of K2CO3 added at 180 °C with 30 min of residence time. Under this pretreatment condition, the highest methane production rate could achieve 15.8 mL/gVSin day, and the percentage of methane reached 73%. The structure of the microbial community involved in the AD was affected by the residence time, working gas, and catalyst of the HTP process. The results showed that Bacteroidetes, Bacteroidia, and SC103 were the dominant phylum, class, and genus of bacteria, respectively, of almost all of the samples. In addition, the most abundant archaeal order was Methanosarcinales, while Methanosaeta was the dominant archaeal genus of most of the samples. However, Methanosarcina largely increased the relative abundance, corresponding to the amount of K2CO3 catalyst used. The findings in this study demonstrated the potential use of K2CO3 during WAO of SS and implied the link between shift of methanogen community and the enhanced methane yield in AD.

Two Gram-stain-negative, aerobic, motile and rod-shaped bacteria, one designated as strain AXBT, capable of degrading estrogens, and another, YL23T, capable of degrading estrogen and bisphenol A, were isolated from activated sludge in Xiamen City, PR China. The optimum temperature and pH of both strains were 25–35 °C and pH 7.0–8.0. While strain AXBT could tolerate 3 % (w/v) NaCl, YL23T could only grow between 0–1 % (w/v) NaCl. They contained ubiquinone-10 as the major quinone, spermidine as the major polyamine, summed feature 8 (comprising C18:1ω6c and/or C18:1ω7c) as the major fatty acids and diphosphatidylglycerol, phosphatidylcholine, phosphatidyldimethylethanolamine, phosphatidylethanolamine, phosphatidylglycerol and sphingoglycolipid as the major polar lipids. The DNA G+C contents of strains AXBT and YL23T were 63.6 and 63.7 mol%, respectively. Based on the results of 16S rRNA gene sequence analysis, strains AXBT and YL23T belonged to the genus Sphingobium . Strain AXBT was most closely related to Sphingobium chlorophenolicum NBRC 16172T (97.5 %) and Sphingobium chungbukense DJ77T (97.2 %), and strain YL23T was most closely related to S. chlorophenolicum NBRC 16172T (97.4 %) and S. quisquiliarum P25T (97.1 %). Average nucleotide identity values between these two strains and S. chlorophenolicum NBRC 16172T, S. chungbukense DJ77T, Sphingobium chinhatense IP26T, Sphingobium quisquiliarum P25T and Sphingobium japonicum UT26ST were from 80.7 to 85.8 %. In conclusion, strains AXBT and YL23T represent novel species of the genus Sphingobium , for which the names Sphingobium estronivorans sp. nov. and Sphingobium bisphenolivorans sp. nov. are proposed, respectively. The type strains of S. estronivorans and S. bisphenolivorans are AXBT (=MCCC 1K01232T=DSM 102173T) and YL23T (=MCCC 1K02300T=DSM 102172T), respectively.

This study investigated the torrefaction of de-oiled Jatropha seed residue after a two-stage sequential process consisting of mechanical screw pressing and solvent extraction using n-hexane (denoted as JMS). The optimal torrefaction temperature (Tr) and torrefaction time (tr) were determined in the ranges of 260–300 °C and 10–60 min, respectively, so to achieve a better heating value and satisfactory energy densification (ED) with acceptable mass loss. Thermogravimetric analysis was employed to elucidate the thermal decomposition behaviors of JMS. By comparison with the torrefaction of Jatropha seed residue after mechanical oil extraction by screw pressing only (namely, JMET), the results indicated that the ED of the torrefaction of JMS yielding the torrefied product JMST (two-stage product) was higher than that of the torrefaction of JME giving the torrefied product JMET (single-stage product). Further, it was found that JMET contained some tar, which was attributed to a thermal reaction in the residual oil in JME during torrefaction. The tar/oil content of JMET was about 1.0–1.8 wt.% in the determined optimal conditions. Thus, the enhanced recovery of the residual oil is advantageous not only because it allows obtaining more oil from Jatropha seed residue with a positive net energy gain but also because it prevents the formation of tar in torrefied biomass products.

Summary Steroid hormones modulate development, reproduction and communication in eukaryotes. The widespread occurrence and persistence of steroid hormones have attracted public attention due to their endocrine‐disrupting effects on both wildlife and human beings. Bacteria are responsible for mineralizing steroids from the biosphere. Aerobic degradation of steroid hormones relies on O2 as a co‐substrate of oxygenases to activate and to cleave the recalcitrant steroidal core ring. To date, two oxygen‐dependent degradation pathways – the 9,10‐seco pathway for androgens and the 4,5‐seco pathways for oestrogens – have been characterized. Under anaerobic conditions, denitrifying bacteria adopt the 2,3‐seco pathway to degrade different steroid structures. Recent meta‐omics revealed that microorganisms able to degrade steroids are highly diverse and ubiquitous in different ecosystems. The aim of the present study was to investigate the role of microRNA-21 (miR-21) in regulating the classical WNT/β-catenin signaling pathway by targeting low-density lipoprotein-related receptor 6 (LRP6) in non-alcoholic fatty liver disease (NAFLD). For this purpose, we established a NAFLD model by feeding C57BL/6J mice a methionine-choline-deficient diet. Antagomir-21 was then injected via the tail vein, and the expression levels of WNT/β-catenin signaling pathway-related proteins, such as LRP6, glycogen synthase kinase-3β (GSK3β), p-β-catenin, β-catenin and the downstream protein, peroxisome proliferator-activated receptor γ (PPAR-γ), and lipid metabolism-related genes, including sterol regulatory element-binding transcription factor 1c (SREBP1c), fatty acid synthase (FAS), carnitine palmitoyl transferase 1α (CPT1α) and adenosine 5-monophosphate (AMP)-activated protein kinase α (AMPKα), were detected. The results revealed that in the NAFLD model, LRP6 expression was negatively associated with miR-21 expression. After antago-nizing the expression of miR-21, the protein level of LRP6 was increased. In addition, the WNT/β-catenin signaling pathway was activated, and lipid accumulation and inflammation were alleviated in the liver. However, the expression of PPAR-γ was not inhibited following the upregulation of the WNT signaling pathway. Taken together, the results of this study demonstrate that the inhibition of miR-21 expression can alleviate NAFLD by targeting LRP6 to activate the WNT/β-catenin signaling pathway.

Natural and synthetic estrogens have been widely detected in wastewater treatment plant (WWTP) influent and effluent as well as in the corresponding receiving aqueous environment and other ecosystems. Microalgae can be used to remove nitrogen and phosphorus in wastewater, but the species-dependent removal of estrogens needs further investigation. In this study we investigated estrone, 17β-estradiol and 17α-ethynylestradiol removals and transformation products by four common microalgae Haematococcus pluvialis, Selenastrum capricornutum, Scenedesmus quadricauda, and Chlorella vulgaris. It was found that H. pluvialis, S. capricornutum and S. quadricauda could more effectively remove all three estrogens in synthetic wastewater effluent. The estrogenic activities i.e. 17β-estradiol equivalency determined by yeast estrogenic screening assay showed substantial estrogenic activity reductions after biotransformation by H. pluvialis, S. capricornutum, and S. quadricauda. Quadrupole Time-of-flight Mass Spectrometry results identified several possible ring-cleavage metabolites as well as their metabolic pathways, which had not been reported yet, confirming the estrogen degradation rather than mere absorption or uptake by microalgae. The findings demonstrate that not only can some specific bacteria degrade estrogens, but also the widely living microalgae are able to degrade these emerging pollutants, suggesting that microalgae could be an advanced treatment of WWTPs to remove nutrients and estrogens.

Objective: Comorbidity of diabetes and depression is a critical problem. Decreased glial-derived neurotrophic factor (GDNF) has been demonstrated in depression, but no evidence of a relationship between GDNF and diabetes has been shown. The present studies were designed to investigate the relationship between GDNF and metabolism. Methods: In Study 1, we performed a case-control study in which subjects with type 2 diabetes mellitus (T2DM), prediabetes (p-DM), and normal glucose tolerance (NGT) were included. In Study 2, we performed a cross-sectional study in 296 patients having pre-existing diabetes in whom the levels of serum GDNF, blood glucose, blood lipids, blood pressure, body mass index, scores from the Patient Health Questionnaire (PHQ-9), the EuroQol-5 scale, and the diabetes distress scale were measured, as well as single-nucleotide polymorphisms of GDNF including rs884344, rs3812047, and rs2075680. Results: In Study 1, serum GDNF concentration was significantly lower in the T2DM group than in the NGT group (NGT: 11.706 ± 3.918 pg/mL; p-DM: 10.736 ± 3.722 pg/mL; type 2 diabetes mellitus [T2DM group]: 9.884 ± 2.804 pg/mL, P = .008). In Study 2, significantly decreased serum GDNF levels were observed in subjects with poor glycemic control or depression (glycated hemoglobin [HbA1c]

The tomato red spider mite, Tetranychus evansi, is an important invasive pest of solanaceous plants worldwide. China is a potentially suitable region for T. evansi but there was no official record of its distribution. In this study, we reported five new records of T. evansi from eggplant, tomato and cucumber in southern and southwest China. The invasion of T. evansi constitutes a new threat to solanaceous crops in mainland China.

Food waste (FW) contributes to a great proportion in the municipal solid waste and is generated during disposal from different life cycles of food processing and consumption. FW treatment is a big challenge and there is an urgent need to develop a suitable treatment technology. The microbial fuel cell (MFC) is a promising bioelectrochemical technology using bacteria as the catalyst, which has been developed to effectively generate bioelectricity from diverse organic wastes. The present study investigated the treatment of food waste collected from a Chinese canteen using MFCs under different conditions. It was observed that the highest closed circuit voltage and maximum power density obtained were 775 ± 21 mV and 422 mW m−2, respectively, when using food waste with nutrient medium as the anolyte and permanganate as the catholyte. Under this condition, biodegradation processes in the MFCs could achieve 69 ± 18% COD, 88 ± 5% carbohydrate, 76 ± 9% protein, 65 ± 8% TOC and 71 ± 8% total nitrogen removal. Microbial community analysis using 16S rRNA gene high-throughput sequencing showed Firmicutes, Bacteroidetes and Proteobacteria were the dominant phyla and Geobacter was the most abundant genus. Therefore, our microbial community results suggested that the mixture of exoelectrogenic and fermentative bacteria have a pronounced effect on MFCs system treating FW while affecting on organic degradation and energy production. A power management system was used to demonstrate that electricity from FW-fed MFCs can be successfully harvested to provide intermittent electricity to loads. Overall, this study demonstrated the potential of using MFCs for food waste treatment to achieve electricity production and waste reduction.

In the present study the synthesis of an inexpensive solar light responsive Co2+ and Fe3+ co-doped TiO2 (Co Fe TiO2) for efficient photocatalytic degradation of ofloxacin (OFL) in addition to its excellent activity as peroxidase mimics has been reported. The as-synthesized material was fully investigated morphologically and structurally by X-ray diffraction (XRD) analysis, field emission-scanning electron microscopy (FE-SEM), Fourier transform infrared (FTIR) and UV diffuse reflectance spectroscopy (UV-DRS). The morphology of the as-synthesized Co Fe TiO2 appeared to be more dispersed, porous and uniform. The UV-DRS analysis showed that the band gap of the as-synthesized TiO2, Fe TiO2, Co TiO2, Co Fe TiO2 was measured as 3.31, 3.22, 3.15 and 3.04 eV, respectively. The pilot scale experiments were conducted in a simple non-concentrating solar photoreactor by using the as-synthesized Co Fe TiO2 having total volume of 5 L. To synergize the photocatalytic process the photocatalytic reactor was added with peroxymonosulfate (HSO5−). The effect of various process parameters like [HSO5−]0, flow rate and [Co Fe TiO2]0 were optimized. The assessment of degradation products (DPs) of OFL and then their toxicity analysis suggested the as-synthesized Co Fe TiO2 provides a safe route for the solar light induced photocatalytic degradation of OFL. The as-synthesized Co-Fe TiO2 was also applied as novel peroxidase mimetic that can catalyze 3,3′,5,5′-tetramethylbenzidine (TMB) when H2O2 serves as oxidant. The results showed that the color response and the peroxidase like response of Co Fe TiO2 highly rely on the concentration of H2O2 and thus the as-synthesized material can successfully be applied for the calorimetric sensing of H2O2.

Understanding the underlying mechanism that drives the microbial community mediated by substrates is crucial to enhance the biostimulation in trichloroethene (TCE)-contaminated sites. Here, we investigated the performance of stable TCE-dechlorinating consortia by monitoring the variations in TCE-related metabolites and explored their underlying assembly mechanisms using 16S rDNA amplicon sequencing and bioinformatics analyses. The monitoring results indicated that three stable TCE-dechlorinating consortia were successfully enriched by lactate-containing anaerobic media. The statistical analysis results demonstrated that the microbial communities of the enrichment cultures changed along with time and were distinguished by their sample sources. The deterministic and stochastic processes were simultaneously responsible for shaping the TCE-dechlorinating community assembly. The indicator patterns shifted with the exhaustion of the carbon source and the pollutants, and the tceA-carrying Dehalococcoides, as an indicator for the final stage samples, responded positively to TCE removal during the incubation period. Pseudomonas, Desulforhabdus, Desulfovibrio and Methanofollis were identified as keystone populations in the TCE-dechlorinating process by co-occurrence network analysis. The results of this study indicate that lactate can be an effective substrate for stimulated bioremediation of TCE-contaminated sites, and the reduction of the stochastic forces or enhancement of the deterministic interventions may promote more effective biostimulation.

A bisphenol A-degrading bacterium, designated as strain H4T, was isolated from surface seawater, which was sampled from the Jiulong River estuary in southeast PR China. Strain H4T is Gram-stain-negative, aerobic, short rod-shaped, lacking bacteriochlorophyll a, motile with multifibrillar stalklike fascicle structures and capable of degrading bisphenol A. Growth of strain H4T was observed at 24–45 °C (optimum, 32 °C), at pH 5.5–9 (optimum, pH 7.0) and in 0–7 % NaCl (optimum, 2 %; w/v) . The 16S rRNA gene sequence of strain H4T showed highest similarity to Croceicoccus pelagius Ery9T (98.7 %), Croceicoccus sediminis (98.3 %), Croceicoccus naphthovorans PQ-2T (98.1 %) and Croceicoccus ponticola GM-16T (97.6 %), followed by Croceicoccus marinus E4A9T (96.7 %) and Croceicoccus mobilis Ery22T (96.0 %). Phylogenetic analysis revealed that strain H4T fell within a clade comprising the type strains of Croceicoccus species and formed a phyletic line with them that was distinct from other members of the family Erythrobacteraceae . The sole respiratory quinone was quinone 10 (Q-10). The predominant fatty acids (>5 % of the total fatty acids) of strain H4T were summed feature 8 (C18 : 1 ω6c and/or C18 : 1 ω7c), summed feature 3 (C16 : 1 ω6c and/or C16 : 1 ω7c), C17 : 1 ω6c and C14 : 02-OH. The genomic DNA G+C content was 62.8 mol%. In the polar lipid profile, phosphatidylcholine, phosphatidylethanolamine, phosphatidylglycerol, two unidentified phospholipids, two sphingoglycolipids and three unknown lipids were the major compounds. Based on the genotypic and phenotypic data, strain H4T represents a novel species of the genus Croceicoccus , for which the name Croceicoccus bisphenolivorans sp. nov. is proposed. The type strain is H4T (=DSM 102182T=MCCC1 K02301T).

Lung cancer is the leading cause of cancer-related death worldwide. Glypican-5 (GPC5) is a member of heparan sulfate proteoglycans, and its biological importance in initiation and progression of lung cancer remains controversial. In the present study, we revealed that GPC5 transcriptionally enhanced the expression of CTDSP1 (miR-26b host gene) via AhR-ARNT pathway, and such up-regulation of CTDSP1 intracellularly contributed to the inhibited proliferation of lung cancer cells. Moreover, exosomes derived from GPC5-overexpressing human lung cancer cells (GPC5-OE-derived exosomes) had an extracellular repressive effect on human lymphatic endothelial cells (hLECs), leading to decreased tube formation and migration. Comparison between GPC5-WT- and GPC5-OE-derived exosomes showed that miR-26b (embedded within introns of CTDSP1 gene) was significantly up-regulated in GPC5-OE-derived exosomes and critical to the influence on hLECs. On the mechanism, we demonstrated that miR-26b transferred into hLECs directly targeted to PTK2 3'-UTR and led to PTK2 down-regulation, resulting in defects in tube formation and migration of hLECs. By uncovering the regulation network among GPC5, miR-26b, miR-26b host gene (CTDSP1), and target gene (PTK2), our findings demonstrated that GPC5 functioned as a tumor suppressor in human lung cancer.

Trauma or neurodegenerative diseases trigger the retrograde death of retinal ganglion cells (RGCs), causing an irreversible functional loss. AT-rich interaction domain 1A (ARID1A), a subunit of the SWItch/Sucrose Non-Fermentable (SWI/SNF) chromatin remodeling complex, has been shown to play crucial roles in cell homeostasis and tissue regeneration. However, its function in adult RGC regeneration remains elusive. Here, we show that optic nerve injury induces dynamic changes of Arid1a expression. Importantly, deleting Arid1a in mice dramatically promotes RGC survival, but insignificantly impacts axon regeneration after optic nerve injury. Next, joint profiling of transcripts and accessible chromatin in mature RGCs reveals that Arid1a regulates several genes involved in apoptosis and JAK/STAT signaling pathway. Thus, our findings suggest modulation of Arid1a as a potential therapeutic strategy to promote RGC neuroprotection after damage.

Mammalian retinal ganglion cells (RGCs) in the central nervous system (CNS) often die after optic nerve injury and surviving RGCs fail to regenerate their axons, eventually resulting in irreversible vision loss. Manipulation of a diverse group of genes can significantly boost optic nerve regeneration of mature RGCs by reactivating developmental-like growth programs or suppressing growth inhibitory pathways. By injury of the vision pathway near their brain targets, a few studies have shown that regenerated RGC axons could form functional synapses with targeted neurons but exhibited poor neural conduction or partial functional recovery. Therefore, the functional restoration of eye-to-brain pathways remains a greatly challenging issue. Here, we review recent advances in long-distance optic nerve regeneration and the subsequent reconnecting to central targets. By summarizing our current strategies for promoting functional recovery, we hope to provide potential insights into future exploration in vision reformation after neural injuries.

The prevalence of non-alcoholic fatty liver disease (NAFLD) has been increasing in recent years. Previous studies have suggested that micro (mi)RNAs may be involved in the pathogenesis of NAFLD. To investigate the role of miRNAs in rat NAFLD, a total of 16 male Sprague Dawley rats were randomly divided into a control group and a model group. Rats in the control group were fed a normal diet for 12 weeks, whereas the rats in the model group were fed a high‑fat and high‑sugar diet for 12 weeks. Following this, the animals were sacrificed and liver tissues were rapidly removed to investigate the severity of NAFLD. Blood samples were collected to investigate liver function, in addition to total cholesterol, total triglyceride and fasting plasma glucose levels. Total RNA from three fresh liver samples per experimental group was extracted for subsequent miRNA gene chip analysis using GeneChip miRNA 4.0 to investigate differentially expressed miRNAs, and miRNA expression was further verified via reverse transcription‑quantitative polymerase chain reaction (RT‑qPCR). Compared with the control group, the results revealed that there were 10 differentially expressed miRNAs in the model group, five of which were overexpressed and five of which were underexpressed compared with the control group. The results of the RT‑qPCR analysis revealed that miR‑182, miR‑29b‑3p and miR‑741‑3p were significantly overexpressed in the model group compared with the control group, which was largely consistent with the results of the microarray analysis. The results suggested that the differentially expressed microRNAs demonstrated in the present study may be involved in the pathogenesis of NAFLD; however, the mechanism underlying the differential expression of miRNAs in NAFLD requires further investigation.

Sulfamethoxazole (SMX) is a common medicine prescribed to treat infections. Due to vast use, SMX has been detected in different parts of the world. Hence, it has become a high risk because of its long term persistence with high biological activity in the ecosystem. Therefore, it is necessary to understand the mechanism of SMX degradation in different genus of bacteria, which is presently unclear. In the present study, degradation of 5 mg L −1 SMX was studied in three isolated pure bacterial cultures, Ochrobactrum sp. SMX-PM1-SA1, Labrys sp. SMX-W1-SC11 and Gordonia sp. SMX-W2-SCD14 and results showed up to 45.2%, 62.2% and 51.4% degradation, respectively within 288 h. Additionally, strain SA1 and strain SCD14 showed up to 66.2% and 69.2% of 4-aminophenol degradation at an initial concentration of 5 mg L −1 within 216 h whereas Labrys sp. SMX-W1-SC11 completely degraded 4-aminophenol at the same concentration within 120 h. Moreover, all three pure bacteria also completely degraded 3-amino-5-methylisoxazole at initial concentration of 4 mg L −1 within 120 h. Furthermore, gas chromatography-mass spectrometry and quadrupole time-of-flight mass spectrometry analysis results revealed that 3-amino-5-methylisoxazole, 4-aminophenol and hydroquinone were the three main by-products of SMX catabolism. In addition, cell free extracts of both Labrys sp. SMX-W1-SC11 and Gordonia sp. SMX-W2-SCD14 showed hydroquinone dioxygenase activity. Besides, all three bacterial strains showed resistance to different heavy metals. Moreover, all three pure bacterial cultures also showed positive chemotactic response toward 3-amino-5-methylisoxazole and hydroquinone based on the drop plate assay. The results of this study recommend these microorganisms for bioremediation of SMX contaminated sites.

Plant microbial fuel cell (PMFC) is an emerging biotechnological device that integrates plants and microorganisms to convert chemical energy into electrical energy. In this study, we focused on the performance of PMFCs to remediate Cd(II)-contaminated soil. The wetland plants, cattail (Typha orientalis), and wild rice (Oryza rufipogon) were utilized in PMFCs to compare the effects of different plants under closed and open-circuit during 150 days. The cattail PMFCs and wild rice PMFCs under 20 mg/kg Cd(II) achieved the maximum voltage at 137.12 mV ± 13.08 and 350.50 ± 74.89 mV, respectively. For the Cd remediation, the cattail PMFCs and wild rice PMFCs could achieve 30.2% and 22.8% of Cd removal in the soil, respectively, and wild rice PMFCs with 5% chestnut biochar showed the highest Cd removal in the soil (31.7 %). The application of PMFCs for Cd(II) remediation could lead to the removal of metals in contaminated soil via bioelectrochemical processes and plant uptake. The Cd uptake by both cattail and wild rice was mainly accumulated in roots, reaching concentrations of 67.29 mg/kg and 16.49 mg/kg, and bioconcentration factors of 5.02 and 1.17, respectively. Furthermore, we observed that PMFCs enhanced immobilization of Cd contamination and reduced bioavailability in soil. Additionally, the microbial community analysis showed Proteobacteria including genera Anaeromyxobacter, Geobacter, Phenylobacterium, and Azospirillum were dominant at the anode region. Overall, this study has demonstrated that PMFC systems can be a promising technology for the treatment of soil contamination with Cd(II).

Predicted decrease in water availability for crop production and uncertainty in climatic conditions require devising the irrigation strategies to increase water use efficiency (WUE) for sustainable crop production. The development of crop cultivars with higher WUE is a pre-requisite for such strategies, particularly in developing countries, including Pakistan, who face stern food security challenges. A two-year field study was conducted following a split-plot randomized complete block design to understand the effects of wheat cultivars (hybrid cultivars, 18A-1 and 18A-2, and local cultivar Ghaneemat IBGE-2016), sowing dates (15th November, 30th November, and 15th December), and irrigation regimes [I (103 mm), II (175 mm), III (254 mm), and IV (330 mm)] at four different growth stages of tillering, booting, anthesis and grain filling on wheat productivity, biomass production and grain yield, and crop-water relations. Early sown hybrid cultivars 18A-1 and 18A-2 showed significantly higher biological and grain yields compared to the local cultivar (59% and 69% higher than the local cultivar). Trends in biomass production and grain yield were also similar at later sowing dates of 30th November and 15th December. However, biological and grain yields decreased with delay in sowing for each cultivar. The data also revealed that hybrid cultivars were better suited to deficient irrigation and generally produced significantly higher biological and grain yields under each moisture regime. Cultivars, sowing dates, and irrigation regime differed significantly for their effects on the Soil Plant Analysis Development (SPAD) values, chlorophyll a and b contents but not for carotenoids. Sowing dates and irrigation regimes had significant effects on relative water content (RWC), water saturation deficit (WSD), water uptake capacity (WUC), and water retention capacity (WRC), however, only WUC varied significantly between the cultivars. The phenological data show that hybrid cultivars took more days to maturity and grain filling than the local cultivar, and days decreased with delayed sowing. The biological and grain yields show significant positive correlations with SPAD values (p &lt, 0.001) and days to maturity (p &lt, 0.001). Our study shows that hybrid wheat cultivars can be opted for higher biomass production and grain yields under deficit irrigation scenarios of semi-arid climatic conditions in Pakistan. Moreover, the hybrid wheat cultivars can perform better than the indigenous cultivar even for delayed sowing dates of 30th November and 15th December.

Summary Although the health of rivers is threatened by multiple anthropogenic stressors with increasing frequency, it remains an open question how riverine microbial communities respond to emerging micropollutants. Here, by using 16S rDNA amplicon sequencing of 60 water samples collected during different hydrological seasons, we investigated the spatio-temporal variation and the co-occurrence patterns of microbial communities in the anthropogenically impacted Jiulong River in China. The results indicated that the riverine microbial co-occurrence network had a non-random, modular structure, which was mainly shaped by the taxonomic relatedness of co-occurring species. Fecal indicator bacteria may survive for prolonged periods of time in river water, but they formed an independent module which had fewer interactions with typical freshwater bacteria. Multivariate analysis demonstrated that nutrients and micropollutants (i.e. pharmaceuticals and personal care products, PPCPs) exerted combined effects in shaping α- and β-diversity of riverine microbial communities. Remarkably, we showed that a hitherto unrecognized disruptive effect of PPCPs on the abundance variations of central species and module communities was stronger than the influence of physico-chemical factors, suggesting the key role played by micropollutants for the microbial co-occurrence relationships in lotic ecosystems. The dissolved CO 2 that causes ocean acidification has great potential for bioenergy production. In this study, we demonstrate that activated methanogens in waste sewage sludge (WSS) are useful for converting bicarbonate in seawater into methane. These activated methanogens were adapted in different seawater sources for methane production through repeated batch experiments that resulted in an increase of 300–400 fold in the methane yield. During these repeated batch experiments, the microbial communities in WSS adapted to the high salinity of seawater to generate more methane. Microbial community analysis showed the dominance of Achromobacter xylosoxidans, Serrati sp. and methanogens including Methanobacterium sp., Methanosarcina sp., and Methanosaeta concillii . Using a 13 C-labeled isotope, we demonstrate that 81% of the methane is derived from microbial conversion of NaH 13 CO 2 in artificial seawater. Therefore, this study shows that oceans, with the largest surface area on Earth, have a potential as a substrate for methane energy production via an acclimated consortium approach.

Background Pulmonary hypertension (PH) is a proliferative disorder associated with enhanced proliferation and suppressed apoptosis of pulmonary artery smooth muscle cells (PASMCs). Our lately study demonstrated that let-7g inhibited hypoxia-induced proliferation of PASMCs via repressing c-myc-Bmi-1-p16 signaling pathway. However, the upstream of let-7g has not yet been fully defined. Previous studies have shown that LOX-1, a target of let-7g, could also regulate the expression of let-7g in human aortic endothelial cells. In this present study, we aimed to investigate whether there is a negative feedback regulation between microRNA let-7g and LOX-1 in hypoxia-induced proliferation of PASMCs. Methods SD Rats were exposed to hypoxia (10% O2, 3 weeks) to induce PH. HE staining was used to evaluate pulmonary artery remodeling. in situ hybridization and immunohistochemistry were performed to assess the expression and distribution of let-7g and LOX-1, respectively. MTS, EDU and flow cytometry were performed to evaluate PASMCs proliferation. Quantitative real-time polymerase chain reaction (qRT-PCR) and Western blotting were conducted to assess the expression of let-7g, LOX-1, calpain-1,-2,-4, and OCT-1. Results The expression of let-7g was significantly down-regulated in pulmonary arteries of hypoxia-induced PH rats accompanied by pulmonary vascular remodeling, whereas let-7g mimic inhibited hypoxia-induced proliferation of PASMCs and up-regulation of LOX-1 expression. LOX-1 blocking reversed hypoxia-induced down-regulation of let-7g expression. Calpains, protein kinase C and OCT-1 were involved in negative feedback regulation between let-7g and LOX-1. Conclusion Negative feedback regulation between let-7g and LOX-1 mediated hypoxia-induced proliferation of in PASMCs.

Mediator complex subunit 19 (Med19), a RNA polymerase II‐embedded coactivator, is reported to be involved in bladder cancer (BCa) progression, but its functional contribution to this process is poorly understood. Here, we investigate the effects of Med19 on malignant behaviours of BCa, as well as to elucidate the possible mechanisms. Med19 expression in 15 BCa tissues was significantly higher than adjacent paired normal tissues using real‐time PCR and Western blot analysis. Immunohistochemical staining of 167 paraffin‐embedded BCa tissues was performed, and the results showed that high Med19 protein level was positively correlated with clinical stages and histopathological grade. Med19 was knocked down in BCa cells using short‐hairpin RNA. Functional assays showed that knocking‐down of Med19 can suppress cell proliferation and migration in T24, UM‐UC3 cells and 5637 in vitro, and inhibited BCa tumour growth in vivo. TOP/FOPflash reporter assay revealed that Med19 knockdown decreased the activity of Wnt/β‐catenin pathway, and the target genes of Wnt/β‐catenin pathway were down‐regulated, including Wnt2, β‐catenin, Cyclin‐D1 and MMP‐9. However, protein levels of Gsk3β and E‐cadherin were elevated. Our data suggest that Med19 expression correlates with aggressive characteristics of BCa and Med19 knockdown suppresses the proliferation and migration of BCa cells through down‐regulating the Wnt/β‐catenin pathway, thereby highlighting Med19 as a potential therapeutic target for BCa treatment.

Background and objective Diabetic pulmonary fibrosis is a severe disease that increases mortality risk of diabetes. However, the molecular mechanisms leading to pulmonary fibrosis in diabetes are poorly understood. This study investigated the roles of epithelial-mesenchymal transition (EMT) and the associated molecular mechanisms in streptozotocin (STZ)-induced rat pulmonary fibrosis. Methods The rat model of diabetic pulmonary fibrosis was established by intraperitoneal injection of a single dose of STZ (35 mg/kg). Typical lesions of diabetic pulmonary fibrosis were observed 8 weeks after STZ injection by hematoxylin-eosin (HE) and Masson staining. Human bronchial epithelial cells (HBECs) and A549 cells were treated by high glucose. Gene or protein expression was measured by real-time PCR, Western blot, immunohistochemistry or immunofluorescence. The knockdown of lectin-like oxidized low density lipoprotein receptor-1 (LOX-1) or transforming growth factor-β1 (TGF-β1) was conducted by siRNA. Results Activation of EMT was observed in lung tissues of STZ-induced diabetic rats, exhibiting a loss in the epithelial cell marker E-cadherin and an increase in the mesenchymal marker Vimentin. The protein and mRNA levels of LOX-1, TGF-β1 and kruppel-like factor 6 (KLF6) in the lung tissues were increased. Incubation of HBECs and A549 cells with high glucose activated EMT and induced an increase in LOX-1, TGF-β1 and KLF-6 expression. LOX-1 siRNA inhibited high glucose-induced EMT in HBECs and A549 cells, which correlated with the reduction of TGF-β1. TGF-β1 siRNA decreased the expression of LOX-1 and KLF6. Conclusions EMT was involved in the pathological process of diabetic pulmonary fibrosis, which was activated by LOX-1/TGF-β1/KLF6 signaling pathway.

Aim Chronic hypoxia leads to right ventricular hypertrophy (RVH). RVH is believed to result from hypoxia-induced pulmonary hypertension. However, if hypoxia impacts RVH directly awaits clarification. Hypoxia triggers oxidative stress, and lectin-like oxidized low-density lipoprotein receptor-1 (LOX-1) mediates reactive oxygen species (ROS) generation in different cells. Therefore, this study aims to explore whether LOX-1-mediated oxidative stress accounts for hypoxia-induced RVH. Main methods Rats developed RVH after 3 weeks of hypoxia (10% O2). Immunofluorescence staining was performed to evaluate H9C2 cell hypertrophy induced by hypoxia (3% O2). Real-time PCR and Western-blot were performed to assess LOX-1, NADPH oxidases (NOX), collagen I/III, atrial natriuretic peptide (ANP) and B-type natriuretic peptide (BNP) expression. DCFH-DA staining was performed to measure ROS generation. Key findings Hypoxia induced RVH and cardiac fibrosis in rats, as indicated by enlarged cardiomyocytes and deposition of extracellular matrix. Interestingly, hypoxia treatment directly induced H9C2 cardiomyocyte hypertrophy, implying direct effects of hypoxia on cell hypertrophy. Rat and H9C2 hypertrophy model revealed that cell hypertrophy was accompanied by marked increase in LOX-1 expression. Knockdown of LOX-1 significantly ameliorated H9C2 cell hypertrophy. Mechanistically, hypoxia induced prominent oxidative stress in rat right ventricles and H9C2 cells, most likely as a result from increased expression of NOX2/4, contributing to RVH. Knockdown of LOX-1 significantly attenuated H9C2 cell oxidative stress, with a concomitant decrease in NOX2/4 expression. Significance LOX-1/NOX/ROS pathway could represent a novel mechanism underlying hypoxia-induced RVH. Therapeutic targeting of LOX-1 would be exploited to treat RVH owing to chronic hypoxia exposure.

Recent studies revealed that sirtuin 1 (SIRT1) is involved in the regulation of energy metabolism and its agonist resveratrol showed anti-obesity effect. This study aims to determine whether BTM-0512, a novel derivative of resveratrol, acts as an antagonist of obesity and to explore its possible mechanisms. High-fat diet (HFD)-induced obese mice were intragastrically administered with BTM-0512 (5, 10, and 20 mg/kg/day) or resveratrol (10 mg/kg/day). It was found that the body weight, Lee's index, ratio of visceral adipose tissue (VAT) to body weight, and blood glucose were significantly reduced in BTM-0512-treated mice when compared with those in mice treated with resveratrol. BTM-0512 up-regulated the expressions of SIRT1, full length PRDM16 (fPRDM16), total PRDM16 (tPRDM16, including fPPRDM16 and other PRDM16 isoforms), and uncoupling protein 1 (UCP1) in both brown and subcutaneous adipose tissues. Although BTM-0512 and resveratrol also up-regulated SIRT1 and tPRDM16 levels in VAT of HFD-induced obese mice, the expressions of fPRDM16, UCP1, and TMEM26 were down-regulated. In mouse primary subcutaneous preadipocytes cultured with or without adipogenic medium, BTM-0512 up-regulated fPRDM16, tPRDM16, and UCP1 expressions, which was reversed by SIRT1 antagonists. But in cultured brown and visceral adipocytes, the UCP1 protein level showed no significant change after treatment with 1 μM of BTM-0512. Moreover, transfection with human SIRT1 plasmid reduced lipid deposit, as well as the mRNA levels of fPRDM16, UCP1, and TMEM26, in cultured human visceral adipose-derived stem cells. In conclusion, BTM-0512 has stronger anti-obesity effect than resveratrol, which might be associated with activation of beige remodeling in subcutaneous adipose tissue.

In our study we examined the role of microRNA-294 (miR-294) in bladder cancer and related mechanisms. Real-time polymerase chain reaction (RT-PCR) was performed to determine the expression level of miR-294. Western blot was used to determine the expression of NRAS, mainly factors in the PI3K/AKT and JAK/STAT pathways. Cell counting kit-8 assay, clonogenic assay, wound-healing assay, transwell and flow cytometry were used to explore, respectively, cell proliferation, survival, migration, invasion, and apoptosis of bladder cancer cell line T24. The expressions of miR-294 in bladder cancer cells including J82, HT1376, T24, and SW780 were significantly increased compared to those in human bladder epithelium cells (both HCV29 and SV-HUC-1). The proliferation rate, surviving fraction, migration, and invasion of T24 cells in miR-294 mimetic transfected group were significantly increased, while they were significantly decreased by miR-294 inhibitor transfection. Moreover, miR-294 suppression could increase the apoptotic rate of T24 cells. In addition, drug resistance of T24 cells to cisplatin was increased in miR-294 mimetic-treated group, while it was decreased by miR-294 inhibitor compared to empty control. Overexpression of miR-294 could upregulate NRAS expression in T24 cells and activate PI3K/AKT and JAK/STAT pathways. We found that miR-294 expression was positively related with proliferation and motility of T24 cells. Moreover, miR-294 suppression could promote the sensitivity of T24 cells to cisplatin. We also found miR-294 could upregulate NRAS and activate the PI3K/AKT and JAK/STAT pathways in T24 cells.

Public concern regarding silver nanoparticles (AgNPs) in the environment has been increasing since they can cause adverse effects in some aquatic species. However, few data are actually available on the effects of AgNPs on the germ cells. In the present study, we used the zebrafish ovarian follicle as a model to assess the potentially adverse effects of AgNPs on oocyte maturation (germinal vesicle breakdown, GVBD) in vitro. Similar to the maturation inducing hormone (17α, 20β-dihydroxy-4-pregnen-3-one), AgNPs induced GVBD, and reduced the total cyclic adenosine monophosphate (cAMP) concentration in zebrafish ovarian follicles. The results from transmission electron microscope observation and Hoechst 33342 staining clearly indicated that AgNPs induced apoptosis in ovarian follicle cells surrounding the oocyte. Similar to AgNPs, AgNO3 also induced GVBD, decreased cAMP concentration and induced apoptosis of ovarian follicle cells. However, the results from gene expression analysis showed that transcript levels of oxidative stress related genes were more sensitive to AgNPs than AgNO3. Further more, H2O2 has an ability to induce zebrafish oocytes maturation by induction of apoptosis in ovarian follicle cells. Taken together, the results from our study indicated that oxidative stress appeared to be one of important mechanisms in AgNP induced apoptosis in ovarian follicle cells, which further triggered the GVBD.

Microbial treatment for vanadium contamination of soils is a favorable and environment-friendly method. However, information of the resistant mechanism of the strains in soils to vanadium, especially to tetravalent vanadium [vanadium(IV)], is still limited. Herein, potential of the vanadium(IV) biosorption and biotransformation of the strains (4K1, 4K2, 4K3 and 4K4) which were capable of tolerating vanadium(IV) was determined. For biosorption, the bioadsorption and the bioabsorption of vanadium(IV) occur on the bacterial cell wall and within the cell, respectively, were taken into consideration. Comparison of the vanadium(IV) adsorbed on the bacterial cell walls and remained in the cells after sorption indicated the major bacterial vanadium(IV) sorption role of the bioadsorption which was at least one order of magnitude higher than the bioabsorption amount. Isotherm study using various isotherm models revealed a monolayer and a multilayer vanadium(IV) biosorption by 4K2 and the others (4K1, 4K3 and 4K4), respectively. Higher biosorption was observed in acidic conditions than in alkaline conditions, and the maximum biosorption was 2.41, 9.35, 7.76 and 8.44 mg g−1 observed at pH 6 for 4K1, at pH 3 for 4K2, and at pH 4 for 4K3 and 4K4, respectively. At the present experimental range of the initial vanadium(IV) concentration, optimal biosorption capacity of the bacteria was observed at the vanadium(IV) level of 100–250 mg L−1. Different biotransformation level of vanadium(IV) in soils by the stains was observed during a 28-d pot incubation of the soils mixed with the strains, which can be attributed to the discrepancy of both soil properties and bacterial species. Present study can help to fill up the gaps of the insufficient knowledge of the vanadium(IV) resistant mechanism of the strains in soils.

The diversity of pathogens associated with acute respiratory infection (ARI) makes diagnosis challenging. Traditional pathogen screening tests have a limited detection range and provide little additional information. We used total RNA sequencing (“meta-transcriptomics”) to reveal the full spectrum of microbes associated with paediatric ARI. Throat swabs were collected from 48 paediatric ARI patients and 7 healthy controls. Samples were subjected to meta-transcriptomics to determine the presence and abundance of viral, bacterial, and eukaryotic pathogens, and to reveal mixed infections, pathogen genotypes/subtypes, evolutionary origins, epidemiological history, and antimicrobial resistance. We identified 11 RNA viruses, 4 DNA viruses, 4 species of bacteria, and 1 fungus. While most are known to cause ARIs, others, such as echovirus 6, are rarely associated with respiratory disease. Co-infection of viruses and bacteria and of multiple viruses were commonplace (9/48), with one patient harboring 5 different pathogens, and genome sequence data revealed large intra-species diversity. Expressed resistance against eight classes of antibiotic was detected, with those for MLS, Bla, Tet, Phe at relatively high abundance. In summary, we used a simple total RNA sequencing approach to reveal the complex polymicrobial infectome in ARI. This provided comprehensive and clinically informative information relevant to understanding respiratory disease.

Rivers are a significant reservoir of antibiotic resistance genes (ARGs), yet the biogeographic pattern of riverine ARGs and its underlying driving forces remain poorly understood. Here, we used metagenomic approach to investigate the spatio-temporal variation of ARGs in two adjacent sub-watersheds viz. North River (NR) and West River (WR), China. The results demonstrated that Bacitracin (22.8 % of the total ARGs), multidrug (20.7 %), sulfonamide (15.2 %) and tetracycline (10.9 %) were the dominant ARG types. SourceTracker analysis indicated that sewage treatment plants as the main source of ARGs, while animal feces mainly contributed in spreading the ARGs in the upstream of NR. Random forest and network analyses confirmed that NR was under the influence of fecal pollution. PCoA analysis demonstrated that the composition of ARGs changed along with the anthropogenic gradients, while the Raup-Crick null model showed that homogenizing selection mediated by class 1 integron intI1 resulted in stable ARG communities at whole watershed scale. Structural equation models revealed that microbial community, grassland and several non-antibiotic micropollutants may also play certain roles in influencing the distribution of ARGs. Overall, the observed deterministic formation of ARGs in riverine systems calls effective management strategies to mitigate the risks of antibiotic resistance on public health.

Dissipation potential of four algae viz. Haematococcus pluvialis, Selenastrum capricornutum, Scenedesmus quadricauda and Chlorella vulgaris was investigated against ten antibiotics (sulfamerazine, sulfamethoxazole, sulfamonomethoxine, trimethoprim, clarithromycin, azithromycin, roxithromycin, lomefloxacin, levofloxacin and flumequine) in a series of synthetic wastewater batch culture experiments, maintained at 20, 50 and 100 μg L−1 initial concentration levels and incubated over a period of 40 days. Generally, the antibiotic removal was achieved with overall dissipation percentage (%) varying among the algal species and different antibiotics. Biodegradation was the major antibiotic removal mechanism from the dissolved fraction, with minor contributions of bioadsorption, bioaccumulation, and abiotic factors. The antibiotics dissipation followed the pseudo-first-order-kinetics with the fastest antibiotic degradation rate achieved by H. pluvialis. The Monod kinetics was successfully applied to explain the relationship between the algal growth and the removal of antibiotics and nutrients in the batch cultures. S. capricornutum and C. vulgaris showed more affinity for the macrolides and fluoroquinolones than sulfonamides, while, H. pluvialis and S. quadiricauda showed relatively higher preference for sulfonamides than the other antibiotic groups. A total of 10 transformation products were identified and the transformation pathway was proposed, accordingly. Most of the transformation products had lower toxicity compared with their parent antibiotics.

Wastewater treatment plants (WWTPs) rely mainly on the microbial assemblages to contribute significantly for the removal of organic pollutants and nutrients. However, limited information is available on the ecological driving forces underlying the turnover of prokaryotic communities across wastewater treatment processes (i.e., from influents (IFs) and effluents (EFs)) within WWTPs. Here, we used a combination of the 16S rRNA gene amplicon sequencing and a quantitative ecological null model analysis to explore the ecological processes governing the turnover of the prokaryotic communities and the dominant taxonomic taxa across wastewater treatment processes of five full-scale WWTPs in China. Our results indicated that a significant variation in the composition of prokaryotic communities and the dominant taxa between IFs and EFs. The analysis of the environmental sources of indicator OTUs showed that a relatively lower abundance of the sludge/sewage and human guts associated OTUs in EFs than in IFs. Ecological null models revealed that among the ecological processes, deterministic processes were dominant in controlling the turnover of the overall communities from IFs to EFs, whereas the relative importance of deterministic processes varied among the dominant taxa (i.e., Bacteroidetes > Proteobacteria > Gammaproteobacteria > Firmicutes > Betaproteobacteria). However, the assembly of IF and EF communities was influenced mainly by the deterministic and stochastic processes, respectively. In addition, our results indicated that EF communities have a higher phylogenetic diversity than those of the IF communities, but the abundance of prokaryotic 16S rRNA genes was lower in EFs than in IFs. Overall, our study provides a novel insight of the assembly mechanisms underlying the turnover of prokaryotic communities during wastewater treatment processes.

Limited information is currently available on the assembly processes (deterministic vs. stochastic) shaping the compositions of key microbial communities in activated sludge (AS). The relative importance of deterministic and stochastic processes for key bacterial and archaeal assemblages (i.e., core-satellite and habitat generalist-specialist) in AS from 13 wastewater treatment plants in China was investigated using 16S rDNA amplicon sequencing. The results obtained indicated 1, 388 and 369 core operational taxonomic units (OTUs), 1, 038 and 1, 683 satellite OTUs, 255 and 48 habitat generalist OTUs, and 192 and 111 habitat specialist OTUs for Bacteria and Archaea, respectively. The proportions of shared OTUs between core and habitat specialist communities were similar to or higher than those between core and habitat generalist communities, suggesting a stronger inter-linkage between the former two groups. Deterministic processes, indicated by abundance-based β-null models, were responsible for shaping core communities, in which NH4-N, OrgC/OrgN, Cr, and Ni were the main controlling factors. In contrast, satellite communities were predominantly influenced by stochastic processes. Moreover, we found that deterministic and stochastic processes were mainly responsible for shaping the assembly of habitat specialists and generalists, respectively. However, the influence of deterministic factors on habitat specialists remains unclear. The present study provides novel insights into the assembly mechanisms of AS microbial communities.

It has well been demonstrated that E3 ubiquitin ligase cullin7 plays important roles in cancer cell growth control via down-regulating p53 expression. The noncanonical function or the pathogenic role of p53 has more recently been implicated in pulmonary vascular remodeling. Therefore, whether cullin7 participates in hypoxia-induced pulmonary vascular remodeling deserves to be elucidated. The present study found that hypoxia up-regulated the expression of cullin7 mRNA and protein in pulmonary arteries and pulmonary artery smooth muscle cells, and knockdown of cullin7 inhibited hypoxia-induced proliferation and migration of pulmonary artery smooth muscle cells and reversed hypoxia-induced inhibition of p53 expression. Notably, administration of proteasome inhibitor MG132 significantly inhibited the expression of cullin7 and up-regulated the expression of p53 in pulmonary arteries concomitantly with improvement of hypoxia-induced pulmonary vascular remodeling. Our study demonstrated that hypoxia induced up-regulation of cullin7 expression resulting to the proliferation and migration of pulmonary artery smooth muscle cells via down-regulating p53 expression, which contributed to pulmonary vascular remodeling.

In the present study, we found that our isolate Shewanella decolorationis NTOU1 is able to degrade acetate under anaerobic condition with concomitant implementation of extracellular electron transfer (EET). With +0.63 V (vs. SHE) poised on the anode, in a 72-h experiment digesting acetate, only 2 mM acetate was consumed, which provides 6% of the electron equivalents derived from the initial substrate mass to support biomass (5%) and current generation (1%). To clarify the effects on EET of the addition of electron-shuttles, riboflavin, anthraquinone-2,6-disulfonate (AQDS), hexaammineruthenium, and hexacyanoferrate were selected to be spiked into the electrochemical cell in four individual experiments. It was found that the mediators with proton-associated characteristics (i.e., riboflavin and AQDS) would not enhance current generation, but the metal-complex mediators (i.e., hexaammineruthenium, and hexacyanoferrate) significantly enhanced current generation as the concentration increased. According to the results of electrochemical analyses, the i-V graphs represent that the catalytic current induced by the primitive electron shuttles started at the onset potential of -0.27 V and continued increasing until +0.73 V. In the riboflavin-addition experiment, the catalytic current initiated at the same potential but rapid saturated beyond -0.07 V; this indicated that the addition of riboflavin affects mediator secretion by S. decolorationis NTOU1. It was also found that the current was eliminated after adding 48 mM N-acetyl-L-methionine (i.e., the cytochrome inhibitor) when using acetate as a substrate, indicating the importance of outer-membrane cytochrome.

In this study, we have immobilized bacterial cells with mediators as bioaugmentation to increase the numbers of bacterial cells in anodic biofilm for better microbial fuel cell (MFC) performance. Four individual bacterial strains (strain E1, strain SCS5, strain B2 and Shewanella putrefaciens) and their mixed culture were first evaluated for electricity production in MFCs. The mixed culture MFC produced higher power density (3.021 Wm−3) than pure strains. The pre-colonized (PC) biofilms together with mediator immobilized mixed culture (MIMC) beads as bioanodes further enhanced the performance of MFCs. Integrated PC/MIMC MFCs containing anthraquinine-2,6-disulfonate (AQDS) and Fe3O4 immobilized mixed culture beads produced 1.50 and 1.47 times power density with respect to mixed culture biofilm, respectively. AQDS immobilization showed better performance compared to Fe3O4 in energy output. The integrated use of PC biofilms and MIMC exhibited lower charge transfer resistance than corresponding individual anode biofilms in MFCs according to electrochemical impedance spectroscopy analysis. The MFC having PC/MIMC as anode showed the ability to store electrons generated by bacteria, but the biocapacitance of anode was altered after integrating MIMC bead particles. Thus, the collective use of PC biofilm and MIMC beads will be one of the potential choices for improving MFC performance.

Plant microbial fuel cells (PMFCs) is a sustainable technology that can convert sunlight to electricity through the integration of plants, microorganism and electrode systems. Urban greening, such as green roofs, is considered as one of the measures to resolve the urban heat island effect caused by the increasing urbanization. In this study, PMFCs were installed as green roofs in a subtropical metropolis. During the operation, the biomass of Chinese pennisetum, Dwarf rotala, and Narrowleaf cattail increased from spring to summer. Furthermore, the maximum daily average output voltage of Chinese pennisetum and Narrowleaf cattail PMFCs was 667.94 ± 128.65 mV in March and 451.12 ± 94.37 mV in June, respectively. For no plant conditions, the maximum daily average output voltage of soil MFCs was 243.70 ± 128.93 mV in March and 100.16 ± 23.43 mV in June. However, little output voltage of Dwarf rotala PMFCs indicated different plant species in PMFC systems would result in varied efficiencies of electricity generation. The trends of electricity generation in Chinese pennisetum and Narrowleaf cattail PMFCs were influenced by net solar radiation and air temperature, respectively according to the results of correlation analysis. The PMFCs based green roofs could lower the temperature of underneath floor slabs as many as 24.81 °C and 29.37 °C compared with bare slabs at noon in March and June. Vegetation of the PMFCs could relieve soil heat flux, and simulated results showed Chinese pennisetum PMFCs with higher vegetation had lower U-value for energy savings of air conditioning. Microbial community analysis showed Geobacter was among the dominant genera and had higher relative abundance in anode soils than cathode soils in Chinese pennisetum and Narrowleaf cattail PMFCs, which generated higher output voltage. Our roof-top research demonstrated that using PMFCs based green roofs for urban greening is promising and warrants the potential for future application.

MicroRNAs (miRNAs), a large family of small and highly conserved non-coding RNAs, regulate gene expression through translational repression or mRNA degradation. Aberrant expression of miRNAs underlies a spectrum of diseases including organ fibrosis. Recent evidence suggests that miRNAs contribute to organ fibrosis through mediating epithelial-mesenchymal transition (EMT). Alleviation of EMT has been proposed as a promising strategy against fibrotic diseases given the key role of EMT in fibrosis. miRNAs impact the expression of specific ligands, receptors, and signaling pathways, thus modulating EMT and consequently influencing fibrosis. This review summarizes the current knowledge concerning how miRNAs regulate EMT and highlights the specific roles that miRNAs-regulated EMT plays in fibrotic diseases as diverse as pulmonary fibrosis, hepatic fibrosis, renal fibrosis and cardiac fibrosis. It is desirable that a more comprehensive understanding of the functions of miRNAs-regulated EMT will facilitate the development of novel diagnostic and therapeutic strategies for various debilitating organ fibrosis.

In this study, the decolorization and degradation of methyl red (MR) by suspended and immobilized cells of Aeromonas jandaei strain SCS5 under anaerobic and aerobic conditions have been investigated. The complete decolorization of MR at a concentration of 100 mg L−1 by A. jandaei strain SCS5 was obtained within 6 h for both anaerobic and aerobic suspended cultures, where the decolorization rate was faster in acidic conditions than basic conditions. The decolorization efficiency under 6 h increased with increasing cell mass of inoculation and decreased with increasing initial dye concentrations. Immobilized cells of A. jandaei strain SCS5 could decolorize MR, and the decolorization rate was significantly enhanced by cells immobilized with mediators such as anthraquinone-2,6-disulphonate and magnetic Fe3O4 nanoparticles compared to immobilized cells only. Moreover, the immobilized bacterial beads with mediators retained high decolorization activity up to more than 10 repeating cycles. UV–visible spectra (200–800 nm) and gas chromatography-mass spectrometry analysis demonstrated that MR was degraded by A. jandaei strain SCS5 through reductive cleavage of azo bond. MR degradation products showed less phytotoxicity against Triticum aestivum and Phaseolus mungo compared to untreated MR. This study has demonstrated that A. jandaei strain SCS5 could be a promising microbiological agent for the removal of azo dyes from the environment.

Archaea play an important role in global carbon and nitrogen cycles. Archaeal lipids, such as isoprenoid glycerol diakyl glycerol tetraethers (iGDGTs), are important biomarkers tracing change in archaeal community structure and biogeochemical processes in the natural environments. In this research, the spatial distributions of archaeal lipids in the surface sediments of the Jiulong River (JR) and the Jiulong River estuary (JRE) were examined. GDGT-0 (containing zero cyclopentyl ring) and crenarchaeol were the most abundant iGDGTs in the JR and JRE. From the rivers to the estuary, the total iGDGTs, GDGT-0, crenarchaeol and archaeol concentrations showed significant spatial variation; in particular, GDGT-0 and archaeol in the river may be predominantly derived in situ from methanogens, whereas crenarchaeol in the estuary mainly derived in situ from Thaumarchaeota . We inferred that archaeal community was dominated by methanogens in the Jiulong River and by Thaumarchaeota in the Jiulong River estuary, which are consistent with change in archaeal community structure observed in other estuarine environments.

In this paper,using water extraction acid sinking method to extract the active components of scutellaria baicalensis.Taking a certain amount of scutellaria baicalensis powder, adding water to boil, adding hydrochloric acid to the filtrate,through a series of steps,the effective components of scutellaria baicalensis could be obtained. Through infrared spectrum and mass spectrum analysis means,it was known that the effective components was baicalin.By using the grinding method, the chitosan and baicalin were mixed to prepare the mixture.Through the cutting of mice tail hemostasia test , the performance of the product was studied.The results showed that the performance of the mixture of chitosan and baicalin was better than that of chitosan,while chitosan and baicalin optimum ratio was 1: 0.8.

Restoration of blood supply to ischemic myocardium causes cardiomyocyte damage, a process known as ischemia-reperfusion injury. Excess reactive oxygen species and intracellular calcium contribute to cell damage but the involvement of sirt1, a versatile protein deacetylase in reperfusion-induced cell damage remains unknown. Here, we found that hypoxia-reoxygenation, an in vitro model of ischemia-reperfusion injury, induced H9c2 cardiomyocyte apoptosis as revealed by caspase-3 assay, Hoechst 33258 staining, flow cytometric analysis and JC-1 staining. Molecular docking analysis showed that, pterostilbene, a natural dimethyl ether derivative of resveratrol, binds to the enzymatic active pocket of sirt1. Importantly, application of pterostilbene at low concentrations of 0.1-3.0 μM rescued H9c2 cells from apoptosis, an effect comparable with resveratrol at 20 μM. Mechanistically, pterostilbene exerted its cardioprotective effects via 1) stimulation of sirt1 activity, since pretreatment of H9c2 cells with splitomicin, an antagonist of sirt1, removed the effects of pterostilbene, and 2) enhancement of sirt1 expression. Therefore, the present study demonstrates that activation of sitr1 during ischemia-reperfusion is cardioprotective and that the natural compound-pterostilbene-could be used therapeutically to alleviate ischemia-reperfusion injury.

In the present study, wastewater and sludge samples of two major hospitals of Lahore, Pakistan were analyzed by developing an HPLC-UV method for the possible occurrence of five frequently used fluoroquinolone antibiotics i.e. ofloxacin, ciprofloxacin, sparfloxacin, moxifloxacin and gemifloxacin. The highest detected concentration was for moxifloxacin in both wastewater (224 μg/L) and sludge samples (219 μg/kg. The highest concentration of ofloxacin, ciprofloxacin, sparfloxacin and gemifloxacin were found to be 66, 18, 58 and 0.2 μg/L respectively. Risk quotient (RQ) was also calculated based on maximum measured concentrations and the RQ values were very high particularly for ofloxacin and ciprofloxacin. The maximum RQ values for ofloxacin against Vibrio fisheri, Pseudomonas putida, fish, Daphnia, Green algae and Pseudokirchneriella subcapitata were 3300, 66,000, 124, 46, 3300 and 6000, respectively. In case of ciprofloxacin, RQ values were found to be 1750 and 3500 against green algae and Microcystis aeruginosa, respectively.

The occurrence, dissemination and assembly processes of antibiotic resistance genes (ARGs) in urban water ecosystems are far from being understood. Here, we examined the diversity and abundance of ARGs in urban water ecosystems including landscape ponds, drinking water reservoirs, influents (IFs) and effluents (EFs) of wastewater treatment plants of a coastal city, China through high-throughput quantitative PCR. A total of 237 ARGs were identified, where multidrug, aminoglycoside and beta-lactamase resistance genes were the most abundant. Urban ponds had a comparatively high diversity and large numbers of shared ARGs with IFs and EFs. The average absolute abundance of ARGs (1.38 × 107 copies/mL) and mobile genetic elements (MGEs) (4.19 × 106 copies/mL) in ponds were only one order of magnitude lower than those of IFs, but higher than those of EFs and reservoirs. Stochastic processes dominated the ARG community assembly in IFs and ponds due to the random horizontal gene transfer caused by MGEs. These results imply that urban ponds are hotspots of ARGs. We further identified 25, 3, and 11 indicator ARGs for tracing the ARG contamination from IFs, EFs and ponds, respectively. Our study represents the first to highlight the role of urban ponds in the dissemination of ARGs.

Estrogens are among the most concerned emerging contaminants in the wastewater treatment effluent due to their sexual disruption in aquatic wildlife. The use of microalgae for secondary wastewater effluent polishing is a promising approach due to the economic benefit and value-added products. In this study, three microalgae species, including Selenastrum capricornutum, Scenedesmus quadricauda and Chlorella vulgaris were selected to conduct batch experiments to examine important mechanisms, especially the role of algal extracellular organic matter (AEOM) on two selected estrogens (17β-estradiol, E2 and 17α-ethynylestradiol, EE2) removal. Results showed that estrogens could not be significantly degraded under visible light irradiation and adsorption of estrogens by microalgae was negligible. All three living microalgae cultures have ability to remove E2 and EE2, and Selenastrum capricornutum showed the highest E2 and EE2 removal efficiency of 91% and 83%, respectively, corresponding to the reduction of predicted estrogenic activity of 86%. AEOM from three microalgae cultures could induce photodegradation of estrogens, and AEOM from Selenastrum capricornutum and Chlorella vulgaris achieved 100% of E2 and EE2 removal under visible light irradiation. Fluorescence excitation–emission matrix spectroscopy identified humic/fulvic-like substances in AEOM from three microalgae cultures, which might be responsible for inducing the indirect photolysis of E2 and EE2. Therefore, in the living microalgae cultures, the major estrogens removal mechanisms should include biotransformation as well as AEOM meditated photocatalytic degradation. Since removal rates through photodegradation could be faster than biotransformation, the AEOM mediated photocatalytic degradation can play a potential role to remove emerging contaminants when using microalgae technology for wastewater effluent treatment.

Nanomaterials based colorimetric detection is an area of vital importance in the field of sensing applications. The nanoparticles are the main component of colorimetric sensor in replacing the natural enzyme based sensor. In this context, zero valent nanoparticles have revolutionized the field of optical sensing especially due to easily shift of electron, facile and low cost of preparation, and ease of surface modification. In this work, zero valent manganese nanoparticles (ZV-Mn NPs) are prepared through a simple and very quick method and modulated with new type of ionic liquid (IL). As-prepared materials were characterized through FE-SEM, HR-TEM, BET, FTIR, and XRD. Subsequently, the peroxidase like catalytic activity of pure and modified ZV-Mn NPs to catalyze oxidation of N,N′,N,N′-tetramethylbenzidine (TMB) in the presence of hydrogen peroxide (H2O2) investigated. Moreover, the absorbance peak is observed at wavelength 652 nm. The enhanced catalytic activity of ZV-Mn NPs was attributed to the fast transfer of electron mechanism in between substrate and H2O2. The coating of IL on ZV-Mn NPs permitted a low limit of detection 0.2 μM with a linear range of 10–280 μM. This work can find wide spread interest in the colorimetric sensing applications. In order to verify the successful demonstration of H2O2 sensor, we have applied it in the dairy milk products with satisfactory results.
