Crotalaria lanceolata E. Mey. and Crotalaria pallida Aiton. are leguminous plants of family Fabaceae found in most of the Brazilian territory. They were initially used as green manure and due their easy spread they are currently considered weeds in crops. Soybean and corn contamination can occur through the mechanical harvesting of these grains along with seeds of the Crotalaria species, which end up in the formulation of feed for production animals. Crotalaria spp. genus has toxic pyrrolizidine alkaloids (PA). Most plant species belonging to this genus can cause acute or chronic liver injury. In a first stage, one-day old broilers were divided into three groups: Group A (C. pallida seeds), Group B (C. lanceolata seeds), and Group C (Control). Groups A and B were divided into five subgroups, each with eight broilers, which received the following doses of the respective seeds in feed as of the 7th day of age: daily doses of 0.4%, 0.8% and 2.5%, and single doses of 15% and 25%. Four broilers in each study group were euthanized at 28 days of age - completing 21 days of seed consumption, and the four remaining broilers were euthanized at 42 days of age - completing 35 days of seed consumption. In a second stage, experiments were conducted using seeds of both the aforementioned plants with 28-day old broilers. These were divided into three groups of four animals each: Group D (C. pallida seeds) and Group E (C. lanceolata seeds), which received the respective seeds at daily doses of 1% and 2% in feed for 20 days, and Group F (Control). These broilers were euthanized when they were 80 days old. C. lanceolata seeds showed higher toxicity to broilers than C. pallida seeds, both supplied as of the 7th day of life. Clinical signs included inappetence, ruffled feathers, and brown diarrhea. The following gross lesions were observed: subcutaneous edema, ascites, hydropericardium, yellowish liver with hypertrophy or atrophy and enhanced lobular pattern, and distended gallbladder. Histologic lesions present in all birds in varying degrees were characterized by tumefaction and vacuolar degeneration of hepatocytes. The following clinical conditions and gross lesions were observed in the broilers: hepatocyte megalocytosis and karyomegaly, slight biliary epithelial hyperplasia, eosinophilic spheroids, and nuclear invagination with loss of hepatocyte cord architecture. RESUMO: Crotalaria lanceolata E. Mey. e Crotalaria pallida Aiton. são leguminosas da família Fabaceae presentes na maioria do território brasileiro. Inicialmente foram utilizadas como adubação verde e devido sua fácil disseminação são consideradas invasoras de culturas. Através da colheita mecanizada da soja e milho pode ocorrer a contaminação destes grãos com sementes dessas espécies e entrar na formulação de ração para animais de produção. O gênero Crotalaria spp. possui alcaloides pirrolizidínicos de ação tóxica. A maioria das espécies desse gênero causam lesões hepáticas, com evolução aguda, ou, crônica. Frangos de corte de um dia de vida foram divididos em três grupos: Grupo A (sementes de C. pallida - doses diárias de 0,4%, 0,8%, 2,5% e doses únicas de 15% e 25%), Grupo B (sementes de C. lanceolata - doses diárias de 0,4%, 0,8%, 2,5% e doses únicas de 15% e 25%) e Grupo C (Controle). Os Grupos A e B foram divididos em cinco subgrupos, com oito frangos cada, que a partir do sétimo dia de vida, receberam as doses estabelecidas. Quatro frangos de cada grupo foram sacrificados aos 28 dias, e os quatro restantes aos 42 dias de vida. Também foram conduzidos experimentos com as sementes dessas duas plantas com aves de 28 dias, as quais foram divididas em três grupos: Grupo D (C. pallida - doses de 1% e 2% diariamente, durante vinte dias), Grupo E (C. lanceolata - doses de 1% e 2% diariamente, durante vinte dias) e Grupo F (Controle). Cada grupo composto por quatro aves. Estas aves foram sacrificadas ao completarem 80 dias de vida. Sementes de C. lanceolata demonstraram maior toxicidade para frangos de corte do que sementes de C. pallida. Os sinais clínicos foram inapetência, penas arrepiadas e diarreia acastanhada. Na macroscopia observou-se edema subcutâneo, ascite, hidropericárdio, fígado de coloração amarelada com hipertrofia e, ou, atrofia, evidenciação do padrão lobular e vesícula biliar distendida. As lesões histológicas presentes em todas as aves, em diferentes graus caracterizaram-se por tumefação e degeneração vacuolar de hepatócitos. Nas aves que manifestaram alterações clínicas e lesões macroscópicas, havia megalocitose, cariomegalia, hiperplasia do epitélio biliar, leve, esferoides eosinofílicos e invaginação nuclear com perda da arquitetura dos cordões de hepatócitos.

This study described the epidemiological and clinical-pathological aspects of 25 outbreaks of neurological diseases in cattle caused by plants and mycotoxins in Santa Catarina state. Six of them were due to Sida carpinifolia poisoning, five to Solanum fastigiatum, five to Phalaris angusta, three to Claviceps paspali, three to Claviceps purpurea, and three outbreaks were of unknown etiology. The clinical signs observed in the affected cattle were mild to severe and characterized by generalized muscle tremors, incoordination, hypermetria, wide-based stance, intentional head tremors, dull staring eyes, and frequent ear twitching, with convulsions in some cases. At necropsy, lesions were observed only for P. angusta poisoning, characterized by gray-greenish discoloration in thalamus and midbrain. Microscopically, rarefaction and/or disappearance of Purkinje neurons with substitution by Bergmann cells were observed for S. carpinifolia and S. fastigiatum poisoning. For P. angusta poisoning, thin granular brown-yellowish pigment was observed in the cytoplasm of some neurons. Gross and microscopic findings were not observed in three outbreaks of tremorgenic disease of unknown etiology. Experiments conducted with leaves, flowers and seeds of Ipomoea indivisa and Ipomoea triloba, as well as with maize and soybean residues contaminated with Ipomoea spp. did not reproduced clinical signs.

This research reports the use of different diagnostic tests in cattle, naturally infected by Rabies lyssavirus (RABV), and correlates the positivity of the tests with the clinical moment of euthanasia, the intensity of the inflammatory lesion and viral load. It also highlights the possibility of euthanasia in early stages of the disease as a way to improve animal welfare. For that, samples of 34 bovine brains were collected for analysis, preserved in 10% buffered formaline and refrigerated with subsequent freezing. The samples were subjected to direct immunofluorescence antibody technique (DFAT) tests, viral isolation in cell culture (VICC), histopathology with hematoxylin and eosin staining (HE), immunohistochemistry (IHC), Shorr stainied neural tissue smears (DSS), Reverse transcription polymerase chain reaction (RT-PCR) and polymerase chain reaction by quantitative reverse transcriptase (qRT-PCR). The areas used for analysis were the cerebellum, parietal telencephalon and thalamus. Samples with Negri bodies (NBs) or immunostaining in at least one of the analyzed areas were considered positive. For the study of the intensity of histological lesions, the lesions were classified into grades 0, 1, 2 and 3 and the positivity of the test in the presence or absence of NBs in one of the three areas analyzed. To verify the influence of the disease clinical evolution, 4-four groups of analysis were created according to the animal’s clinical status at moment of the euthanasia, being: M1 = animal euthanized while standing, M2 = euthanized when in sternal recumbence, M3 = euthanized when in lateral recumbence, M4 = animal with natural death. Of the 34 brains evaluated, IHC was positive in 100% of cases, DFAT was positive in 97.05% of them, and in this negative sample the presence of RABV was confirmed by VICC. NBs ere seen in 88.23% of the cases, and the DSS test was positive in 82.35% of them. All diagnostic techniques showed positive cases in all groups analyzed. Each case was positive in at least two diagnostic methods. All cases that contained NBs were positive for rabies in the other tests. In this study, it was observed that the variables analyzed (intensity of injury and clinical evolution at the moment of euthanasia) had an influence only on HE and DSS techniques, which are based on NB research to form the diagnosis, but did not interfere with the effectiveness of the diagnosis performed by detecting the viral antigen performed by DFAT and IHC. All isolated RABV samples included in the present study have a genetic lineage characteristic of hematophagous bats Desmodus rotundus. The evaluation of qRT-PCR showed that the amount of virus did not interfere in the positivity of the tests. This work shows that IHC and DFAT are safe diagnostic techniques. They are capable of detecting RABV even in euthanized animals in the early stages of clinical evolution with mild intensities of histological lesions. RESUMO: Esta pesquisa relata a utilização de diferentes testes de diagnóstico em bovinos, naturalmente infectados pelo Rabies lyssavirus (RABV), e correlaciona a positividade dos testes com o momento clínico da eutanásia, a intensidade da lesão inflamatória, e a carga viral. Salienta também a possibilidade da eutanásia em estágios precoces da doença como forma de melhorar o bem-estar animal. Para isso amostras de 34 encéfalos bovinos foram coletados para análise, conservadas em formol tamponado 10% e sob refrigeração com posterior congelamento. As amostras foram submetidas aos testes de imunofluorescência direta (IFD), isolamento viral em cultivo de células (IVCC), histopatologia com coloração de hematoxilina e eosina (HE), imuno-histoquímica (IHQ), esfregaço direto com coloração de Shorr (EDS), reação da polimerase em cadeia por transcriptase reversa (RT-PCR) e reação da polimerase em cadeia por transcriptase reversa quantitativo (qRT-PCR). As áreas utilizadas para análise foram o cerebelo, telencéfalo parietal e tálamo. Foram consideradas positivas as amostras que apresentaram Corpúsculo de Negri (CNs) ou imuno-marcação em ao menos uma das áreas analisadas. Para o estudo da intensidade das lesões histológicas, as lesões foram classificadas em graus 0, 1, 2 e 3 e a positividade do teste na presença ou ausência de CN em uma das três áreas analisadas. Para verificar a influência da evolução clínica da doença foram criados 4 grupos de análise conforme o estado clínico do animal no momento da eutanásia, sendo: M1 = animal eutanasiado em estação, M2 = eutanasiado em decúbito esternal, M3 = eutanasiado em decúbito lateral, M4 = animal com morte natural. Dos 34 encéfalos avaliados a IHQ foi positiva em 100% dos casos, a IFD foi positiva em 97,05%, sendo que na amostra negativa a presença de RABV foi confirmada por IVCC. A histologia com HE, através da visualização das CNs, foi positiva em 88,23 % dos casos, e o teste de EDS, foi positivo em 82,35%. Todas as técnicas de diagnóstico apresentaram casos positivos em todos os grupos analisados. Cada caso foi positivo em, pelo menos, dois métodos de diagnóstico. Todos os casos que continham CN foram positivos para raiva nos demais testes. Nesse estudo observou-se que as variáveis analisadas intensidade de lesão e evolução clínica no momento da eutanásia tiveram influência somente nas técnicas de HE e EDS, que se baseiam na pesquisa do CN para formação do diagnóstico, mas não interferiram na eficácia do diagnóstico realizado através da detecção do antígeno viral realizado por IFD e IHQ. Todas as amostras RABV isoladas incluídas no presente estudo apresentam linhagem genética característica de morcegos hematófagos Desmodus rotundus. A avaliação de qRT-PCR demostrou que a quantidade de vírus não interferiu na positividade dos testes. Esse trabalho mostra que a IHQ e a IFD são técnicas seguras de diagnóstico e que mesmo em animais eutanasiados em estágios iniciais de evolução clínica com intensidades leve de lesões histológicas, são capazes de detectar o RABV.

This study identified the different forms of ingestion of Senecio brasiliensis in cattle, diagnosed by the Animal Pathology Laboratory at CAV-UDESC, in the state of Santa Catarina, Brazil. A retrospective evaluation from 1987 to 2016 showed that ingestion has occurred voluntarily due to the presence of the adult plant in native field and/or involuntary, due to the presence of the plant in hay and silage, provided in the trough together with other pastures or by contamination in grain residues. These different forms of ingestion demonstrate the importance of epidemiologic investigation in the diagnosis of seneciosis.

We experimentally reproduced star grass (Cynodon nlemfuensis Vanderyst var. nlemfuensis cv. “Florico”) poisoning in cattle. Star grass was planted in four shaded (1, 2, 3, and 4) and three sunny paddocks (5, 6, and 7) and were fertilized with the equivalent of 0, 750, 200, or 100 kg/ha urea. Fresh plant leaves were collected from each paddock and fed to 8 calves aged 5 months to 1 year at 10g/kg live weight. Clinical examinations were performed pre- and post-feeding. Fresh leaves were also subjected to sodium picrate paper and diphenylamine tests to evaluate hydrocyanic acid and nitrate concentration, respectively. Clinical signs of poisoning by the star grass cv. “Florico” in cattle were observed 5-15 minutes after ingestion; these included wobbling, muscle tremors, frequent and/or dripping urination, tachycardia, ruminal atony, depression, red ocular mucous membranes, and bright red venous blood. Three animals became severely ill, 4 manifested mild to moderate clinical signs, and 1 did not become ill. The 3 severely ill bovines received a thiosulfate and sodium nitrite solution at a dose of 40ml/100 kg live weight, intravenously (IV). Florico star grass poisoned bovines and the reaction to the sodium picrate paper and diphenylamine tests was more significant for samples cultivated in the shade and with high nitrogen fertilization. Florico grass utilization should be avoided in a silvopastoral system. RESUMO: Descreve-se a reprodução experimental da intoxicação cianogênica por grama estrela (Cynodon nlemfuensis vanderyst var. nlemfuensis cv. ‘Florico’) em bovinos. A grama estrela foi plantada em quatro piquetes em área sombreada (1, 2, 3 e 4) e em três piquetes em área ensolarada (5, 6 e 7) e foram adubados com o equivalente a 0, 750, 200 ou 100kg/ha de ureia. Folhas da planta foram coletadas de cada piquete e imediatamente fornecidas na dose de 10g/Kg de peso vivo para 8 bezerros com idades entre 5 meses a 1 ano. Exames clínicos foram realizados antes e durante fornecimento da planta. Folhas verdes de estrela africana foram submetidas ao teste do papel picro-sódico para verificar a presença de ácido cianídrico e ao teste da difenilamina para avaliar a concentração de nitrato. Sinais clínicos de intoxicação pela grama estrela cv. “Florico” nos bovinos foram observados entre 5 e 15 minutos após o término da ingestão, estes incluíram andar cambaleante, tremores musculares, micção frequente e/ou em gotejamento, taquicardia, atonia ruminal, depressão, mucosas oculares e sangue venoso de coloração vermelho-vivo. Três animais adoeceram gravemente, quatro manifestaram sinais clínicos leves a moderados e um não adoeceu. Os 3 bovinos que adoeceram gravemente receberam solução de tiossulfato e nitrito de sódio na dose de 40ml/100kg de peso vivo, por via endovenosa (IV). Grama estrela florico produziu intoxicação em bovinos e a reação ao teste do papel picro-sódico e difenilamina foi mais significativa para as amostras cultivadas à sombra e com alta adubação nitrogenada. Utilização de grama florico deve ser evitada em sistema silvipastoril.

This study reports the epidemiological data and the clinical-pathological condition of five outbreaks of cyanogenic poisoning in cattle spontaneously ingesting star grass (Cynodon nlemfuensis Vanderyst var. nlemfuensis cv. ‘Florico’). In all outbreaks, the areas where the plant was previously fertilized with high concentrations of nitrogen and the properties adopted the silvipastoral system. The first clinical signs appeared between 10 and 15 minutes after the first introduction of cattle and were characterized by muscular tremors, dyspnea, moderate tympanism, staggering gait, forced breathing with open mouth, sternal recumbency followed by death after 15 to 30 minutes and/or recovery in a few hours after the signs started. In total, 43 cows have become ill and 18 died. Two necropsies were performed and no significant changes were found except for the presence of the plant near the esophageal sphincter region. No histological lesions were seen through microscopy. Green leaves of the star grass were collected from all properties where the outbreaks occurred and the test of the picro-sodium paper was performed, revealing red-brick coloration in 20 minutes after maceration of the leaves. RESUMO: Descrevem-se os dados epidemiológicos e quadro clínico-patológico de cinco surtos de intoxicação cianogênica em bovinos que ingeriram espontaneamente grama estrela (Cynodon nlemfuensis Vanderyst var. nlemfuensis cv. ‘Florico’). Em todos os surtos, as áreas onde a planta se encontrava haviam sido previamente adubadas com altas concentrações de nitrogênio e as propriedades adotavam o sistema silvipastoril com Eucaliptus sp. Os primeiros sinais clínicos surgiram entre 10 e 15 minutos após a primeira introdução dos bovinos e caracterizou-se por tremores musculares, dispneia, timpanismo moderado, andar cambaleante, respiração forçada com a boca aberta, decúbito esternal seguido de morte após 15 a 30 minutos e/ou, recuperação em poucas horas após início dos sinais. No total, adoeceram 43 vacas e destas 18 morreram. Duas necropsias foram realizadas e não foram encontradas alterações significativas, exceto a presença da planta próxima a região do esfíncter esofágico. Através da microscopia não foram visualizadas lesões histológicas. Folhas verdes da grama estrela foram coletadas de todas as propriedades onde os surtos ocorreram e realizadas o teste do papel picro-sódico, o qual revelou coloração vermelho-tijolo em 20 minutos após maceração das folhas.

Hovenia dulcis Thunberg (uva-Japão) é uma árvore caducifolia nativa da China e de alguns lugares do Japão. Nos últimos anos essa planta foi utilizada como forma de sombreamento para aviários no Oeste e Meio-Oeste de Santa Catarina e passou a disseminar-se por toda a região. Os bovinos comem avidamente seus frutos maduros quando caem ao chão. Suspeitas de intoxicação ocorreram no outono e início de inverno e coincidiam com a maturação dos frutos. A doença foi reproduzida em 2004 por outros autores através da administração dos frutos a bovinos, em dose única de 24,5g/kg. Nos anos subsequentes não ocorreram reclamações sobre a intoxicação pelos frutos dessa planta, embora, muitos criadores afirmavam que os bovinos, na temporada de maturação, continuavam a ingerir os frutos. Experimentalmente, nove bovinos que receberam frutos da planta em doses únicas entre 30 e 50g/kg, apenas dois bovinos adoeceram gravemente e um morreu. O quadro clínico e as lesões foram semelhantes à intoxicação reproduzida em 2004; porém, a dose necessária para reproduzirmos a doença foi 100% superior à dose tóxica preconizada como letal em 2004. Hovenia dulcis Thunberg (Japan grape) is a deciduous tree native of China and some parts of Japan. Over the last years, the tree has been used for shading in poultry farms of western and middle western Santa Catarina, spreading in the region. Cattle ingest the ripe fruits of H. dulcis fallen to the ground. The fruits were suspected to cause poisoning during fall and early winter, period of their maturation. The disease has been reproduced in 2004 by other authors with a single dose of 24.5g/kg. In the subsequent years no complaints regarding the toxicity of H. dulcis fruits have been reported, although many breeders observed that cattle continued to consume them during the ripening season. We administered the fruits at single doses of 30-50g/kg to nine calves; only two of them became seriously ill and one died. The clinical picture and lesions were similar to the poisoning reproduced in 2004; however, the dose required to reproduce the poisoning was 100% higher than the lethal dose found in 2004.

RESUMO: Tifton 68 (Cynodon nlemfuensis Vanderyst) é uma gramínea cultivada na região sul do Brasil, responsável por manifestações clínicas superagudas de dispneia, dificuldade de deglutição, tremores musculares, timpanismo e decúbito em bovinos. A morte ocorre rapidamente após o início dos primeiros sinais e não são encontradas alterações macro e microscópicas significativas. O presente estudo descreve os aspectos epidemiológicos, clínicos e lesionais da intoxicação espontânea por tifton 68 que ocorreu nos municípios de Rio do Sul, Pouso Redondo, Taió e Rio do Campo, estado de Santa Catarina, Brasil, nos anos de 1996, 1997, 1998 e 2010, respectivamente. Experimentalmente foram avaliadas a presença de ácido cianídrico nas folhas verdes e secas desta planta, através do teste do papel picrossódico e a reversão da intoxicação pela utilização de uma solução antídoto específica de tiossulfato de sódio e nitrito de sódio. A reprodução experimental consistiu na administração de folhas verdes de tifton 68 para dois bovinos com doses a partir de 10,3g/kg. O quadro de intoxicação cianogênica foi confirmado pela imediata resposta ao tratamento intravenoso com a solução antídoto. As amostras da planta verde forneceram resposta positiva ao teste do papel picrossódico. O feno de tifton 68 não demonstrou qualquer toxicidade, mesmo em altas doses (18 e 27g/kg), sendo seguro para a alimentação de bovinos. ABSTRACT: Tifton 68 (Cynodon nlemfuensis Vanderyst) is a grass cultivated in southern Brazil which causes peracute clinical manifestations including dyspnea, swallowing difficulties, muscular tremors, tympanism and recumbency in cattle. Animal’s death occurs rapidly after the onset of clinical signs and neither macroscopic nor microscopic lesions have been found. This paper describes the epidemiology, clinical findings and lesions of spontaneous poisoning by tifton 68 in cattle in the municipalities of Rio do Sul, Pouso Redondo, Taió and Rio do Campo in the state of Santa Catarina, Brazil, in 1996, 1997, 1998 and 2010 respectively. The presence of hydrocyanic acid was experimentally estimated in green and dried leaves through the picric acid paper test and poisoning treatment using a specific antidote solution of sodium thiosulfate and sodium nitrite. The poisoning was experimentally reproduced by the administration of tifton 68 green leaves to two cattle at a starting dose of 10.3 g/kg. The cyanide poisoning was confirmed by the immediate response to treatment with intravenous antidote. The samples of tifton 68 green leaves gave positive response in the picric acid paper test. The hay of tifton 68 showed no toxicity even at high doses (18 and 27 g/kg) and it is safe for cattle consumption.

Plants of the genus Senecio sp. are known to produce hepatic lesions in different animal species, including man. To evaluate the toxicity of three species of Senecio found in regions where cattle are bred in the state of Santa Catarina, experiments were conducted on broiler chickens (Gallus domesticus). Green leaves of S. conyzaefolius, S. vernonioides and S. paulensis were collected and dried in the shade. After drying, the leaves were ground, mixed into the feed and fed to the chickens divided into 3 groups. Group 1 and Group 2 received single doses of 5g/kg and 20g/kg, respectively. Group 3 was given daily doses of 1g/kg for 20 days (values corresponding to green plant) and Group 4 (control) received free feed of the plant. For each group, five chickens were submitted to necropsy at 30 days and five to 60 days after planting, and five chickens that consumed S. vernonioides and S. conyzaefolius at the dose of 1g/kg for 20 days were necropsied at 90 days after the beginning of the experiment. Macroscopic lesions were observed in chickens that consumed S. vernonioides at a dose of 1 g/kg for 20 days in birds necropsied at 30, 60 and 90 days and were characterized mainly by liver of firm consistency, diminished in size with yellowish coloration and evident lobular pattern, ascites and hydropericardium. In microscopy the main lesions observed were megalocytosis, hepatocyte swelling, fibrosis and biliary hyperplasia and were more intense at the dosage of 1 g/kg for 20 days for S. vernonioides and less severe in the chickens that ingested S. conyzaefolius. The chickens that received S. paulensis showed no macroscopic and microscopic lesions.

This study aimed to describe and discuss the results of an experiment carried out in two stages with pregnant cows fed 25kg/apple pomace/day. The first stage involved 16 pregnant Holstein Friesian cows divided into four groups: Group 0 - Control (5 cows); Group I - 1 month-gestation (4 cows); Group II - 3 month-gestation (4 cows); Group III - 6 month-gestation (3 cows) and was performed from September to December 2015. The second stage comprised 12 pregnant Holstein Friesian cows divided into three groups: Group 0 - Control (6 cows), Group I - 1 month-gestation (3 cows), and Group II - 3 month-gestation (3 cows) and was conducted from April 2016 to February 2017. All study animals received apple pomace at a dose of 25kg/day. As for the first experiment stage, a cow in Group III bred a calf with complete absence of the coccygeal vertebrae and tail, slight bending of the hind limbs, scoliosis in the thoracic spine, and limited mobility. At 30 days, it presented with diarrhea and underdevelopment, and was euthanized for necropsy. At gross examination, malformations were observed in the thoracic spine, coxofemural joint, and genitourinary tract. Regarding the second experiment stage, a cow in Group I gave birth to a calf with curved pelvic and thoracic limbs with thick joints and flattening hooves. Microscopic examination of the femur showed disorganized, irregular hypertrophic zone and scarce growth zone, in addition to primary spongy zone with short, slightly mineralized trabeculae. Samples of the apple pomace used in this study were frozen and sent for laboratory evaluation of pesticide residues, which showed a positive result for the fungicide carbendazim.
