Fungi are rich in complexes of cryptic species that need a combination of different approaches to be delimited, including genomic information. Beauveria (Cordycipitaceae, Hypocreales) is a well-known genus of entomopathogenic fungi, used as a biocontrol agent. In this study we present a polyphasic taxonomy regarding two widely distributed complexes of Beauveria: B. asiatica and B. bassiana s.lat. Some of the genetic groups as previously detected within both taxa were either confirmed or fused using population genomics. High levels of divergence were found between two clades in B. asiatica and among three clades in B. bassiana, supporting their subdivision as distinct species. Morphological examination focusing on the width and the length of phialides and conidia showed no difference among the clades within B. bassiana while conidial length was significantly different among clades within B. asiatica. The secondary metabolite profiles obtained by liquid chromatography-mass spectrometry (LC-MS) allowed a distinction between B. asiatica and B. bassiana, but not between the clades therein. Based on these genomic, morphological, chemical data, we proposed a clade of B. asiatica as a new species, named B. thailandica, and two clades of B. bassiana to respectively represent B. namnaoensis and B. neobassiana spp. nov. Such closely related but divergent species with different host ranges have potential to elucidate the evolution of host specificity, with potential biocontrol application.

Nine species in Cordyceps sensu lato producing orange to red stromata were discovered during diversity surveys of arthropod-pathogenic fungi in Thailand. The phylogenetic analyses of combined ITS, LSU, RPB1, RPB2 and TEF1 sequence data indicated four novel species belonging to Blackwellomyces and five novel species of Cordyceps. All Blackwellomyces species produce filiform ascospores with septations. Blackwellomyces aurantiacus and Blackwellomyces roseostromatus occur on lepidopteran larvae, and both species produce an Evlachovaea-like conidial arrangement of the anamorph and produce a red pigment that diffuses across agar medium. Blackwellomyces calendulinus and Blackwellomyces minutus are found on coleopteran larvae producing Acremonium-like and Mariannaea-like conidial arrangements of their anamorphs and do not produce any pigment. Novel members of Cordyceps include the species Cordyceps brevistroma, Cordyceps inthanonensis, Cordyceps neopruinosa, and Cordyceps parvistroma that are pathogenic on lepidopteran larvae and cocoons, whereas C. araneae is pathogenic on spiders. All Cordyceps species produce bola-shaped, whole ascospores, except for C. inthanonensis that produces ascospores dissociating into part-spores. A common feature of the novel Cordyceps species presented herein is the production of Evlachovaea-like or Mariannaea-like conidial arrangement of the anamorphs.

Two new fungal genera and six species occurring on insects in the orders Orthoptera and Phasmatodea (superorder Orthopterida) were discovered that are distributed across three families in the Hypocreales. Sixty-seven sequences generated in this study were used in a multi-locus phylogenetic study comprising SSU, LSU, TEF, RPB1 and RPB2 together with the nuclear intergenic region (IGR). These new taxa are introduced as Metarhizium gryllidicola, M. phasmatodeae, Neotorrubiella chinghridicola, Ophiocordyceps kobayasii, O. krachonicola and Petchia siamensis. Petchia siamensis shows resemblance to Cordyceps mantidicola by infecting egg cases (ootheca) of praying mantis (Mantidae) and having obovoid perithecial heads but differs in the size of its perithecia and ascospore shape. Two new species in the Metarhizium cluster belonging to the M. anisopliae complex are described that differ from known species with respect to phialide size, conidia and host. Neotorrubiella chinghridicola resembles Torrubiella in the absence of a stipe and can be distinguished by the production of whole ascospores, which are not commonly found in Torrubiella (except in Torrubiella hemipterigena, which produces multiseptate, whole ascospores). Ophiocordyceps krachonicola is pathogenic to mole crickets and shows resemblance to O. nigrella, O. ravenelii and O. barnesii in having darkly pigmented stromata. Ophiocordyceps kobayasii occurs on small crickets, and is the phylogenetic sister species of taxa in the 'sphecocephala' clade.

Thailand has rich natural resources and diverse ecosystems. From our surveys of entomopathogenic fungi in Thailand over the last decade, 66 Beauveria strains in BIOTEC Culture Collection (BCC) were studied to account for the diversity of this genus in Thailand. Examination of morphological characteristics from various media and phylogenetic analyses based on partial sequence of ITS, TEF, RBP1, RPB2, and the nuclear intergenic region Bloc, were realized. Among the strains from Thailand, we described here a new species, namely B. mimosiformis sp. nov., isolated from Coleoptera larva in the North of Thailand. This new species has a unique characteristic of phialides and conidia forming an ensemble similar to plant leaves of the genus Mimosa; it has pale yellow to light yellow stromata, cylindrical with rounded apices or oblong conidia, cylindrical part-ascospores. We also found a teleomorph linked to B. asiatica and documented new hosts (Lepidoptera larva and adult beetle) of B. gryllotalpidicola, as well as reported B. malawiensis for the first time in Thailand. Beside those species, B. bassiana and B. loeiensis were also identified, adding up to six species from Thailand. Through molecular phylogenetic analyses, we revealed (1) diverging lineages within B. asiatica and B. bassiana which, despite lack of support to be erected as any higher taxonomical unit, demonstrated complex intra-specific molecular diversity undergoing differentiation and (2) ambiguous relations between certain known species, particularly for B. majiangensis that we synonymized as B. asiatica.

Over the last two decades the molecular phylogeny and classification of Metarhizium has been widely studied. Despite these efforts to understand this enigmatic genus, the basal lineages in Metarhizium are still poorly resolved. In this study, a phylogenetic framework is reconstructed for the Clavicipitaceae focusing on Metarhizium through increased taxon-sampling using five genomic loci (SSU, LSU, tef, rpb1, rpb2) and the barcode marker ITS rDNA. Multi-gene phylogenetic analyses and morphological characterisation of green-spored entomopathogenic Metarhizium isolates from Thailand and soil isolates of M. carneum and M. marquandii reveal their ecological, genetic and species diversity. Nineteen new species are recognised in the Metarhizium clade with narrow host ranges: two new species are found in the M. anisopliae complex – M. clavatum on Coleoptera larvae and M. sulphureum on Lepidoptera larvae; four new species are found in the M. flavoviride complex – M. biotecense and M. fusoideum on brown plant hoppers (Hemiptera), M. culicidarum on mosquitoes, M. nornnoi on Lepidoptera larvae; three new species M. megapomponiae, M. cicadae, M. niveum occur on cicadas; five new species M. candelabrum, M. cercopidarum, M. ellipsoideum, M. huainamdangense M. ovoidosporum occur on planthoppers, leafhoppers and froghoppers (Hemiptera); one new species M. eburneum on Lepidoptera pupae; and four new species M. phuwiangense, M. purpureum, M. purpureonigrum, M. flavum on Coleoptera. Of these 19 new species, seven produce a sexual morph (M. clavatum, M. eburneum, M. flavum, M. phuwiangense, M. purpureonigrum, M. purpureum, and M. sulphureum) and asexual morphs are found in the remaining new species and also in M. sulphureum, M. purpureonigrum and M. purpureum. Metarhizium blattodeae, M. koreanum and M. viridulum are new records for Thailand. An alternative neotype for Metarhizium anisopliae is proposed based on multi-gene and 5′tef analyses showing that CBS 130.71 from Ukraine is more suitable, being from a much closer geographical location to Metchnikoff’s Metarhizium anisopliae. This isolate is distinct from the neotype of Metarhizium anisopliae var. anisopliae proposed by M. Tulloch from Ethiopia (ARSEF 7487). Six new genera are established for monophyletic clades subtending the core Metarhizium clade, including Keithomyces, Marquandomyces, Papiliomyces, Purpureomyces, Sungia, and Yosiokobayasia. Metarhizium carneum, M. aciculare, and M. neogunnii are combined in Keithomyces and one new combination for M. marquandii in Marquandomyces is proposed. Purpureomyces is introduced for species producing purple stromata including a new combination for M. khaoyaiense and two new species P. maesotensis and P. pyriformis. Papiliomyces contains two new combinations for M. liangshanense and Metacordyceps shibinensis. The genus Sungia is proposed for the Korean species M. yongmunense on Lepidoptera pupa and Yosiokobayasia for the Japanese species M. kusanagiense also on Lepidoptera pupa. A synoptic and dichotomous key to the accepted taxa is provided together with tables listing distinguishing morphological characters between species, host preferences, and geography.

The genus Moelleriella mostly occur on scale insects and whiteflies. It is characterized by producing brightly colored stromata, obpyriform to subglobose perithecia, cylindrical asci, disarticulating ascospores inside the ascus, and fusiform conidia, predominantly found in tropical and occasionally subtropical regions. Scale insects and whiteflies’ pathogens were found during our surveys and collections of entomopathogenic fungi. Investigations of morphological characters and multi-locus phylogenetic analyses based on partial sequences of LSU, TEF, and RPB1 were made. Five new species of Moelleriella are described here, including M. chiangmaiensis, M. flava, M. kanchanaburiensis, M. nanensis, and M. nivea. They were found on scale insects, mostly forming flat to thin, umbonate, whitish stromata. Their anamorphic and teleomorphic states were mostly often on the same stroma, possessing obpyriform perithecia, cylindrical asci with disarticulating ascospores. Their conidiomata are widely open with several locules per stroma, containing cylindrical phialides and fusiform conidia. However, M. flava has yellow stromata with one conidioma per stroma, and M. kanchanaburiensis was found only in its anamorphic state. Furthermore, the perithecia of M. nanensis were completely embedded in stromata as opposed to other species with semi-immersed perithecia. We also found a teleomorph link to M. sinensis, a species described based on anamorphic stage previously reported from southeastern China.

We also found a teleomorph link to M. sinensis, an anamorphic species reported from southeastern China.

Three new fungal species in the Clavicipitaceae (Hypocreales, Ascomycota) associated with plants were collected in Thailand. Morphological characterisation and phylogenetic analyses based on multi-locus sequences of LSU, RPB1 and TEF1 showed that two species belong to Aciculosporium and Shimizuomyces. Morakotia occupies a unique clade and is proposed as a novel genus in Clavicipitaceae. Shimizuomyces cinereus and Morakotia fusca share the morphological characteristic of having cylindrical to clavate stromata arising from seeds. Aciculosporium siamense produces perithecial plates and occurs on a leaf sheath of an unknown panicoid grass.

Thailand is known to be a part of what is called the Indo-Burma biodiversity hotspot, hosting a vast array of organisms across its diverse ecosystems. This is reflected by the increasing number of new species described over time, especially fungi. However, a very few fungal species from the specialized spider-parasitic genusGibellulahave ever been reported from this region. A survey of invertebrate-pathogenic fungi in Thailand over several decades has led to the discovery of a number of fungal specimens with affinities to this genus. Integration of morphological traits into multi-locus phylogenetic analysis uncovered four new species:G. cebrennini,G. fusiformispora,G. pigmentosinum, andG. scorpioides. All these appear to be exclusively linked with torrubiella-like sexual morphs with the presence of granulomanus-like asexual morph inG. pigmentosinumandG. cebrennini. A remarkably high host specificity of these new species towards their spider hosts was revealed, and for the first time, evidence is presented for manipulation of host behavior inG. scorpioides.

The Ophiocordyceps myrmecophila complex is composed of pathogens specific to ants, found on the leaf litter or buried in soil and produce Hymenostilbe asexual morph. Species in this complex are morphologically highly similar and can hardly be distinguished macroscopically. To date, it has only been observed on formicine ants of the genera Polyrhachis and Camponotus. In this study, observations were conducted in three sites at Khao Yai National Park of Thailand where three new species are proposed. Molecular phylogenies based on the large subunit of the ribosomal DNA (LSU), partial sequences of translation elongation factor 1-α (TEF), and the largest and second largest subunits of the RNA polymerase Π (PRB1 and RPB2) revealed distinct clades separating these new species, namely Ophiocordyceps megacuculla, Ophiocordyceps khaoyaiensis, and Ophiocordyceps granospora. The morphological features of these three new species are clearly different from Ophiocordyceps thanathonensis, a recently described species of the complex, but mostly overlap between these three. However, they are proposed as distinct species based on molecular phylogenetic relationships, genetic distance, and minor morphological characters related to sexual structures.

Ophiocordyceps is a heterogeneous, species-rich genus in the order Hypocreales (Sordariomycetes, Ascomycota) that includes invertebrate-pathogenic taxa. In this study, seven new species in Ophiocordyceps producing superficial perithecia infecting various insect hosts (Lepidoptera, Hemiptera) are described from Thailand – Ophiocordyceps brunneinigra, O. brunneiperitheciata, O. geometridicola, O. multiperitheciata, O. pauciovoperitheciata, O. pseudoacicularis and O. spataforae. Phylogenetic analyses based on multigene loci comprising the large subunit of the ribosomal DNA (LSU), partial sequences of elongation factor 1-alpha (TEF) and the largest and second largest subunit of the RNA polymerase (RPB1, PRB2) strongly support these new species of Ophiocordyceps in the Ophiocordycipitaceae. They differ from species previously described species Ophiocordyceps acicularis, O. atewensis, O. cochlidiicola, and O. crinalis, in the shape and sizes of distinguishing characters such as perithecia, ascospores and conidia. We also report a new record of O. macroacicularis in Thailand. A new genus pathogenic to scale insects (Coccidae, Hemiptera), Helicocollum, is described from Thailand. It includes the three species, H. chanthaburiensis, H. krabiensis and H. surathaniensis. Phylogenetic inferences on the basis of partial sequences of the large subunit of the nuclear ribosomal DNA, translation elongation factor 1-α and RNA polymerase II subunit 1 place the new genus in the Clavicipitaceae, although morphological features of the phialides resemble Hirsutella in the Ophiocordycipitaceae. Within the Clavicipitaceae, Helicocollum is closely related to Aschersonia, Moelleriella, Regiocrella, and Samuelsia, all known as pathogens of scale insects. Helicocollum differs from other scale insect pathogens in having phialides with helical necks and synnematous or sporodochial conidiomata, but pycnidia. Morphological characteristics are evaluated and compared with other asexual morphs producing twisted, undulated phialidic necks or occurring on scale insects.

Seven new species occurring on termites are added to Ophiocordyceps – O.asiatica, O.brunneirubra, O.khokpasiensis, O.mosingtoensis, O.pseudocommunis, O.pseudorhizoidea and O.termiticola, based on morphological and molecular phylogenetic evidence. O.brunneirubra possesses orange to reddish-brown immersed perithecia on cylindrical to clavate stromata. O.khokpasiensis, O.mosingtoensis and O.termiticola have pseudo-immersed perithecia while O.asiatica, O.pseudocommunis and O.pseudorhizoidea all possess superficial perithecia, reminiscent of O.communis and O.rhizoidea. Phylogenetic analyses based on a combined dataset comprising the internal transcribed spacer regions (ITS) and the largest subunit (LSU) of the ribosomal DNA, partial regions of the elongation factor 1-α (TEF) and the largest and second largest subunits for the RNA polymerase genes (RPB1, RPB2) strongly support the placement of these seven new species in Ophiocordyceps.

In several surveys in the tropical forests in Thailand, specimens that looked morphologically similar to Metarhizium martiale and Cordyceps variegata, as well as other Metarhizium species were collected and cultured in vitro. A combined phylogeny of several genes including the small (18S) and large (28S) subunits of the ribosomal DNA, elongation factor 1-α (TEF), RNA polymerase II subunits 1 and 2 (RPB1, RPB2) genes has shown these to be new taxa in the Clavicipitaceae. Nigelia is described as a new genus closely related to Metarhizium, to the scale insect pathogens Aschersonia (Hypocrella), Samuelsia and Moelleriella, and to plant pathogens in Claviceps and Balansia, and other relatives. Nigelia comprises M. martiale and a new species Nigelia aurantiaca, which has been found infecting lepidopteran larvae and which produces pseudoimmersed, obliquely arranged, obpyriform perithecia with curved or bent ostioles and with whole (non-separating) cylindric ascospores. Metarhizium chaiyaphumense, M. kalasinense, M. prachinense, M. samlanense, and M. takense are described as new species of Metarhizium. Metarhizium martiale is transferred to Nigelia, and Paecilomyces reniformis is transferred to Metarhizium.

Conoideocrella krungchingensis , a fungal pathogen infecting armored scale insects (Diaspididae) on a dicotyledonous plant, is described and illustrated as a new species from Thailand. The new taxon morphologically resembled C. luteorostrata but could be distinguished from the latter using the sequences of the partial nuclear large subunit rRNA (LSU) and the elongation factor (EF-1α) genes. Apart from it, asci and ascospores of C. krungchingensis were found notably shorter than those of C. luteorostrata and C. tenuis . All three species of Conoideocrella have been reported from Thailand, including C. krungchingensis .

Two new species of Moelleriella were discovered during diversity surveys of entomopathogenic fungi in the south of Thailand and are described as M. alba and M. chumphonensis based on morphological characters and molecular study. They have effuse white to orange stromata. Their teleomorphs are unique in producing a single perithecium inside a tubercle resembling Moelleriella raciborskii, a common species in Thailand. A combined matrix of nuclear large subunit rRNA gene (LSU) and RNA polymerase II subunit one (RPB1) gene shows that both species cluster in a distinct lineage as sister to M. raciborskii/“Aschersonia” placenta.

Two new species found in northern Thailand, Cordyceps chiangdaoensis and Cordyceps morakotii, pathogenic to Coleoptera larvae and Odontomachus ant pupae, respectively, are described using morphological and molecular phylogenetic data. Both species produce narrowly ovoid superficial perithecia at the end of a cylindrical stroma, bola ascospores, Evlachovaea-like conidial morph, and cylindrical conidia with rounded ends. These two species differ from other bola ascospore-producing species in the genus Cordyceps in the sizes of the ascospores and perithecia, as well as the host. Phylogenetic analyses based on internal transcribed spacer (ITS) regions of the rDNA and partial sequences of translation elongation factor 1-alpha (TEF1) data strongly support these two fungi as two distinct new species in the Cordycipitaceae.

New phenolic mono- and digalactopyranosides (1 and 2), their aglycone KS-501a (3), and a new phenolic 4-O-methylglucopyranoside (4) were isolated from the filamentous fungus Acremonium sp. BCC 14080. Structures of these compounds were elucidated by extensive MS and NMR spectroscopic analyses. Compound 1 displayed anti-HSV-1 activity with an IC(50) value of 7.2 microM. Compound 3 exhibited activity against Plasmodium falciparum K1 with an IC(50) value of 9.9 microM.

Ophiocordyceps unilateralis sensu lato infects ants, modifies their behavior, and is found in many countries around the world. One unifying concept of all such parasitic associations is that both the parasite and the host adapt to maximize their fitness and reproductive output. However, little is known about the reproductive life span of this pathogen or about its alternation between asexual and sexual states, even though these two states affect host population dynamics differently. To address these issues, a permanent plot in a tropical rainforest was surveyed over the course of two years to examine the development of O. unilateralis s.l. and the incidence of infection of Polyrhachis and Camponotus ants, which were found to be specifically attacked by this fungus in Thailand. We document here for the first time the life cycle of this pathogen over the long term, which provides fundamental knowledge for the understanding of this fascinating host–parasite interaction.
