The Levantine Middle Paleolithic period displays significant archaeological variability across a series of cave and open-air sites encompassing ca. 200,000 years. Faunal remains are an important source of knowledge regarding hunting and mobility patterns but have mostly been studied in the deep stratigraphic sequences of the Levantine caves. This research addresses questions of hunting, carcass transport, butchery patterns and use of space as they occurred at the Middle Paleolithic open-air site of Nesher Ramla in central Israel. The site is an eight-meter-thick sequence within a karst sinkhole, dating to MIS6/5. We conducted the first detailed taphonomic and zooarchaeological study at the site, focusing on a sample from Unit III, which is a thin layer with dense lithics and faunal remains, combustion features, manuports, and ochre. Our results reveal an anthropogenic accumulation that is dominated by aurochs, equid, and tortoise remains. The large ungulates’ (aurochs) skeletal-element representation is biased in favor of meat- and fat-rich body parts, coupled with abundant evidence of dismemberment, filleting and marrow extraction. These parts were imported to the sinkhole for processing and consumption. Tortoises are abundant. The lithic assemblages exhibit high frequency of retouched tools and low typological diversity and are characterized by high visibility of personal toolkit components. Coupled with use-wear analysis that suggests low spectrum of activities and massive occurrence of hammerstones, anvils and manuports, these characteristics indicate that the Unit III occupation represents an intensive camp centering on aurochs processing and butchering.

This paper focuses on new findings from Middle Paleolithic Geula Cave, Israel, located in the northern part of Mt. Carmel. The cave, consists of several small chambers that are remnants of a larger cave system, initially excavated between 1958 and 1964. The occupation at the cave was ascribed to Middle Paleolithic modern humans. In 2016, a salvage excavation was conducted in areas of the cave that were not previously explored. Analyses of the new excavation revealed a consistency in lithic technology throughout the new excavation areas, with an emphasis on Levallois production using mainly bidirectional and centripetal core preparation modes. The faunal study identified intensive hyena and porcupine activities in small chambers, probably at the back part of the cave, while fractured bones in association with flint artifacts were found at what appears as the living area in the cave. Luminescence ages indicated that Geula Cave was occupied as early as 175 ka, but that the major occupation was from 120 to 100 ka. The major occupation at Geula chronologically overlaps with the Skhul and Qafzeh Caves thus reinforcing the notion that Homo sapiens dominated the southern Levant during early MIS 5. This study demonstrates the importance of reinvestigating and reevaluating past excavated prehistoric sites and their contents to enhance our understanding of the regional, cultural and biological history.

In cnidarians, long-term ecological success relies on sexual reproduction. The sea anemone Nematostella vectensis, which has emerged as an important model organism for developmental studies, can be induced for spawning by temperature elevation and light exposure. To uncover molecular mechanisms and pathways underlying spawning, we characterized the transcriptome of Nematostella females before and during spawning induction. We identified an array of processes involving numerous receptors, circadian clock components, cytoskeleton, and extracellular transcripts that are upregulated upon spawning induction. Concurrently, processes related to the cell cycle, fatty acid metabolism, and other housekeeping functions are downregulated. Real-time qPCR revealed that light exposure has a minor effect on expression levels of most examined transcripts, implying that temperature change is a stronger inducer for spawning in Nematostella. Our findings reveal the mechanisms that may enable the mesenteries to serve as a gonad-like tissue for the developing oocytes and expand our understanding of sexual reproduction in cnidarians.Summary statementAnalysis of transcriptional changes during spawning induction in Nematostella vectensis, revealed upregulation of processes related to signal perception and cytoskeleton rearrangement and downregulation of fatty acid metabolism and housekeeping processes.

We report an observation at Jhalana Leopard Reserve (JLR), Jaipur, India. On 16 March 2019, we saw a female walking up the mountain while calling her two, 4-month-old, male and female cubs. This allowed several safari jeeps to park in the shade of nearby Acacia trees. Two Striped Hyenas (Hyaena hyaena) approached and sniffed at the base of one of the trees near the parked jeeps. Upon seeing the Hyaenas, the female ran down the mountain, passed between the jeeps, and climbed into the tree. That is when we noticed the body of the female cub at a height of approximately 4.5 m. The female licked the body of the cub for several minutes and then picked it up in her mouth, climbed down from the tree, passed again between the jeeps, and walked up the mountain towards a dense stand of Thor (Euphorbia caducifolia), a thorny cactus. Our observations display how a mother leopard that has lost a cub refuses to abandon the carcass, cached it in a tree, and when discovered by scavengers removed it to a thicket of cacti. We consider the behavior towards her dead cub, and subsequent caching when discovered by other animals, to be the first evidence for thanatological expression in leopards.

When the reproductive interests of males and females conflict, males can evolve traits that are harmful to females, and females can coevolve traits to resist this harm. In the fruit fly, Drosophila melanogaster, there is genetic variation in female resistance traits, which can affect the pre- and post-mating success of males that try to mate with them. However, it is not clear to what extent the expression of these phenotypes can be modified by environmental factors such as sociosexual experience. Here, we tested how the genetic background of a female and her previous mating experience interact to affect the mating success of focal males. In the experience phase, we placed females from 28 distinct genetic backgrounds individually either with a single male (low conflict) or with three males (high conflict) for 48 hr. In the subsequent test phase, we measured the mating and post-mating fertilization success of focal males paired individually with each female. We found that focal males paired with females from the high-conflict treatment were less successful at mating, took longer to mate when they were successful, and had a lower proportion of paternity share. Furthermore, we identified significant female genetic variation associated with male mating success. These results indicate that female experience, along with intrinsic genetic factors, can independently influence different fitness components of her subsequent mates and has implications for our understanding of plastic female mating strategies and the evolution of sexually antagonistic traits in males and females.

The ecology and predator–prey dynamics of large felids in the tropics have largely been studied in natural systems where wild ungulates constitute the majority of the prey base. However, in tropical countries where communities are primarily agrarian, the high density of domestic animals in human-dominated landscapes can be a potential prey source for large carnivores. We demonstrate almost complete dependence of the Vulnerable leopard Panthera pardus fusca in the Jhalana Reserve Forest in Jaipur, north-west India on domestic animals as prey. We analysed 132 leopard scats collected during the dry season of November 2017–April 2018. Domestic animals comprised the majority of the leopards' prey (89.5% frequency of occurrence): dogs Canis lupus familiaris (44%), cats Felis catus (13%), goats Capra aegagrus hircus (16%) and cattle Bos taurus (15%). Wild species, which occurred in the leopards' diet at a relatively low frequency, were rodents, the hare Lepus nigricollis, small Indian civet Viverricula indica, rhesus macaque Macaca mulatta, northern plains grey langur Semnopithecus entellus and mongoose Herpestes edwardsii. Diet is also a function of availability of potential prey, but no data are available on the density of the leopard's wild prey species in Jhalana Reserve Forest. Nevertheless, our results suggest that abundance of domestic prey around Jhalana Reserve Forest sustains the c. 25 known leopards. We conclude that these leopards, by preying on feral dogs in an urban environment, could be considered as suppliers of a service to the human population amongst whom they thrive, although this potentially exposes the leopards to the canine distemper virus.

While the biogenesis of microRNAs (miRNAs) in both animals and plants depends on the RNase III Dicer, its partner proteins are considered distinct for each kingdom. Nevertheless, recent discovery of homologs of Hyponastic Leaves1 (HYL1), a “plant-specific” Dicer partner, in the metazoan phylum Cnidaria, challenges the view that miRNAs evolved convergently in animals and plants. Here we show that the HYL1 homolog Hyl1-like a (Hyl1La) is crucial for development and miRNA biogenesis in the cnidarian model Nematostella vectensis. Inhibition of Hyl1La by morpholinos resulted in metamorphosis arrest in Nematostella embryos and a significant reduction in levels of most miRNAs. Further, meta-analysis of morphants of miRNA biogenesis components, like Dicer1, shows clustering of their miRNA profiles with Hyl1La morphants. Strikingly, immunoprecipitation of Hyl1La followed by quantitative PCR revealed that in contrast to the plant HYL1, Hyl1La interacts only with precursor miRNAs and not with primary miRNAs. This was complemented by an in vitro binding assay of Hyl1La to synthetic precursor miRNA. Altogether, these results suggest that the last common ancestor of animals and plants carried a HYL1 homolog that took essential part in miRNA biogenesis and indicate early emergence of the miRNA system before plants and animals separated.Impact statementPresence of a functional homolog of HYL1 in Nematostella vectensis, a basal animal model, indicates divergent evolution of miRNA biogenesis pathway in plants and animals from an ancestral miRNA system

Mutations in more than 150 genes are responsible for inherited hearing loss, with thousands of different, severe causal alleles that vary among populations. The Israeli Jewish population includes communities of diverse geographic origins, revealing a wide range of deafness-associated variants and enabling clinical characterization of the associated phenotypes. Our goal was to identify the genetic causes of inherited hearing loss in this population, and to determine relationships among genotype, phenotype, and ethnicity. Genomic DNA samples from informative relatives of 88 multiplex families, all of self-identified Jewish ancestry, with either non-syndromic or syndromic hearing loss, were sequenced for known and candidate deafness genes using the HEar-Seq gene panel. The genetic causes of hearing loss were identified for 60% of the families. One gene was encountered for the first time in human hearing loss: ATOH1 (Atonal), a basic helix-loop-helix transcription factor responsible for autosomal dominant progressive hearing loss in a five-generation family. Our results demonstrate that genomic sequencing with a gene panel dedicated to hearing loss is effective for genetic diagnoses in a diverse population. Comprehensive sequencing enables well-informed genetic counseling and clinical management by medical geneticists, otolaryngologists, audiologists, and speech therapists and can be integrated into newborn screening for deafness.

We demonstrate a methodology for replicating and projecting the path of COVID-19 using a simple epidemiology model. We fit the model to daily data on the number of infected cases in China, Italy, the United States, and Brazil. These four countries can be viewed as representing different stages, from later to earlier, of a COVID-19 epidemic cycle. We solve for a model-implied effective reproduction number R t each day so that the model closely replicates the daily number of currently infected cases in each country. For out-of-sample projections, we fit a behavioral function to the in-sample data that allows for the endogenous response of R t to movements in the lagged number of infected cases. We show that declines in measures of population mobility tend to precede declines in the model-implied reproduction numbers for each country. This pattern suggests that mandatory and voluntary stay-at-home behavior and social distancing during the early stages of the epidemic worked to reduce the effective reproduction number and mitigate the spread of COVID-19.

Neonatal autoimmune subepidermal blistering disease is rare. Mucosal involvement is more common in neonatal linear immunoglobulin A (IgA) bullous dermatosis. We describe a neonate with subepidermal cutaneous blistering disease with severe laryngeal and esophageal involvement leading to acute respiratory distress. Histopathology demonstrated a subepidermal blister with neutrophils and eosinophils at the dermal base. Collagen IV was detected at the dermal floor, and direct immunofluorescence showed linear IgG, IgA, and C3 deposits at the basement membrane zone. The patient demonstrated markedly increased serum levels of anti-BP180 NC16A and anti-BP230 IgG antibodies (Abs) but failed to show anti-LAD-1 IgA Abs. His healthy mother showed serum anti-LAD-1 IgA Abs but did not show anti-BP180 and anti-BP230 Abs. The neonate responded promptly to systemic corticosteroid therapy. A review of the literature detected 11 cases of neonatal subepidermal blistering disease with linear IgA deposits. Nine of these cases demonstrated coexisting linear IgG deposits, often with C3. Respiratory compromise was present in most of the cases. Neutrophils and eosinophils were commonly present in the inflammatory cell infiltrates. Besides our case, 2 cases of neonatal IgG/IgA subepidermal blistering disease with esophageal involvement were previously described. IgA Abs were present in the sera of both cases. Anti-LAD-1 IgA Abs were detected in the mother's serum of our case alone, but IgA Abs do not cross the placenta. Our case was consistent with neonatal IgG/IgA pemphigoid. Neonatal IgG/IgA subepidermal blistering disease may be associated with severe laryngeal and esophageal involvement leading to respiratory compromise. Expedited diagnosis and prompt treatment are warranted.

Squamate (lizard and snake) remains are abundant in the terminal Pleistocene Natufian archaeological sites of the Levant, raising the question of whether they constitute part of the broad-spectrum diet characteristic of this period. However, the role of squamates in Natufian diets remains unclear, as they are taphonomically under-studied. We conducted a series of experiments and actualistic observations that tested the impact of pre- and post-depositional processes on squamate vertebrae. We emphasized the multiple destruction processes that leave overlapping or altered marks on the bones, such as digestion marks that were modified by trampling. The resulting bone modification typology provides a tool for studying archaeological squamate remains. The experimental data were compared to the archaeological bone samples of the Natufian sequence of el-Wad Terrace (Mount Carmel, Israel, 15,000–12,000 cal BP). The Natufian squamate samples deviate from all actualistic ones in their lesser evidence of digestion and much greater indications for trampling, erosion and breakage. The taphonomic study, coupled with intra-site analysis, has unraveled the complex depositional history of el-Wad Terrace, enabling us to differentiate between cultural and non-cultural contexts and to identify possible human consumption of the European glass lizard and the large whip snake in the Natufian.

Forced copulation is an extreme form of sexual aggression that can affect the evolution of sex-specific anatomy, morphology, and behavior. To characterize mechanistic and evolutionary aspects of forced copulation, we artificially selected male fruit flies based on their ability to succeed in the naturally prevalent behavior of forced matings with newly eclosed (teneral) females. The low and high forced copulation lineages showed rapid divergence, with the high lineages ultimately showing twice the rates of forced copulation as the low lineages. While males from the high lineages spent more time aggressively pursuing and mounting teneral females, their behavior toward non-teneral and heterospecific females was similar to that of males from the low lineages. Males from the low and high lineages also showed similar levels of male-male aggression. This suggests little or no genetic correlations between sexual aggression and non-aggressive pursuit of females, and between male aggression toward females and males. Surprisingly however, males from the high lineages had twice as high mating success than males from the low lineages when allowed to compete for consensual mating with mature females. In further experiments, we found no evidence for trade-offs associated with high forced mating rates: males from the high lineages did not have lower longevity than males from the low lineages when housed with females, and four generations of relaxed selection did not lead to convergence in forced mating rates. Our data indicate complex interactions among forced copulation success and consensual mating behavior, which we hope to clarify in future genomic work.

Sexual conflict is common in nature and sometimes results in sexual aggression. An extreme case is forced copulation, where one individual forcibly mates with another individual who resists the mating. To understand what makes some males sexually aggressive, we established an experimental system that allowed us to quantify the characteristics that contribute to males' forced copulation success. In fruit flies (Drosophila melanogaster), sexually mature females can choose to accept or reject courting males; however, males can forcibly copulate with newly eclosed, sexually immature, teneral females. We tested males from 59 genotypes and found significant genetic variation in forced copulation success, with a broad-sense heritability of 0.16. We then chose three genotypes with the lowest and three genotypes with the highest forced copulation success rates and compared the behaviour of males from these two groups. Males from genotypes with high forced copulation success were more persistent in their pursuit of teneral females and mounted them more frequently than did males from the low-success genotypes. Males of the two categories, however, were similar in their attractiveness to both teneral and sexually mature females. Our results suggest that males vary in their pursuit strategies. Some males respond to female rejection signals by giving up and searching for receptive females, while other males persist in pursuit and coercion in spite of female objection. Our work highlights the practicality of using forced copulation in fruit flies as a model for further research on the mechanisms affecting variation in sexual coercion and forced copulation success and their evolutionary consequences.

The broad impairments in cognitive and neurologic functioning found in Autism Spectrum Disorder (ASD) patients are thought to originate during early prenatal developmental stages. Indeed, postmortem and imaging studies in ASD patients detected white-matter abnormalities, as well as prefrontal and temporal cortex deficits, evident from early childhood. Here, we used Maternal Immune Activation (MIA), a mouse model for ASD, in which the offsprings exhibit Autistic-like behaviors as well as cortical abnormalities. However, the dynamics that influence the number and the identity of newly born cortical neurons following maternal inflammation remains unknown. Our study shows early changes in the duration of the S-phase of PAX6+ progenitors, leading to an increased proportion of neurogenic divisions and a reciprocal decrease in the proliferative divisions. In two different time points of maternal inflammation, MIA resulted in an overproduction of CTIP2+ cortical neurons, which remained overrepresented at the end of gestation and in postnatal mice. Interestingly, MIA-resistant IL6-KO mice did not exhibit these changes. Lastly, we propose that elevated levels of the transcription factor PAX6 following MIA supports the overproduction of CTIP2+ neurons. Taken together, our data reveals a possible link between maternal immune activation and the excess of cortical neurons found in the cortex of ASD patients.

To understand the prevalence and conditional use of aggression among animals, one has to know its costs and benefits. The obvious cost of aggression in animals that possess teeth, claws or other specialized weaponry is injury. Many species, however, do not have such body parts and thus cannot readily injure others. The cost of aggression in these animals is not well studied. We tested whether aggression has a fitness cost in fruit flies, which can serve as a model species for animals without weapons that engage in aggression. In three experiments employing distinct protocols, we allowed focal flies to fight for control of an attractive food patch over 4 days and then compared their survivorship to that of flies not engaged in conflict. In all three experiments, fly survivorship was lower in the aggression than no‐aggression treatments. Microscopic examination revealed no differences in wing damage between flies of the aggression and no‐aggression treatments. The two most likely, non‐mutually exclusive explanations for lower survivorship post‐fighting are physiological changes due to stress, and metabolic alterations associated with a life‐history strategy optimized for high‐conflict settings.

International audience; Pectobacterium brasiliense (Pbr) infects a wide range of crops worldwide, causing potato blackleg and soft rot and vegetable soft rots. This study aimed to characterize the genetic diversity and virulence variability among 68 Pbr strains isolated from either symptomless potato progeny tubers, diseased potato plants, ware potatoes wash water, or vegetables grown in Israel, as well as strains isolated from symptomless seed tubers grown in Europe, or diseased potato plants grown in France. The collection was typed using PCR and TaqMan real-time PCR analyses, dnaX sequence analysis, pulsed-field gel electrophoresis (PFGE), and pectolytic activity. dnaX phylogeny grouped almost all strains in a common genetic clade related to Pbr, which was distinct from the other Pectobacterium species. PFGE analysis identified two main clusters, including one major group of 47 strains with 95%-100% similarity. Maceration assays on two potato cultivars showed significant differences between strains but with no correlations with the source of the strains nor the status of the host (with/without symptoms). Molecular (dnaX sequences and PFGE profiles) and phenotypic analyses (tuber maceration tests) showed that the tested Pbr strains are not a homogeneous group. Analysis of the tested Pbr strains isolated from potato and vegetables grown in fields with a history of potato cultivation suggests that seed tubers imported from Europe may be the main source for Pbr in Israel. To the best of our knowledge, this is the first study that describes biodiversity and population structure of P. brasiliense isolated from potato and vegetables under hot climate conditions

Staphylococcus aureus is a major bacterial cause of clinical infections and foodborne illnesses.Through the synthesis of a group of Staphylococcal enterotoxins (SEs), gastroenteritis occurs and the SEs function as superantigens to massively activate T cells. The ability to rapidly detect and quantify SEs is imperative in order to learn the causes of staphylococcal outbreaks and to stop similar outbreaks in the future. Also, the ability to discern active toxin is essential for development of food treatment and processing methods. Here, we discuss the various methodologies for detection and analysis of SEs.

Sonic booms or disruptive explosions cause differing response in wildlife. In May 2021, when missiles were fired at urban areas in Israel, we observed the responses of breeding common swifts (Apus apus). In both nests, the initial boom resulted in a startled awakening of the swifts. In one case, a parent engaged in displacement behaviour of feeding the young while in the second case a parent tried to clamber up the walls. Our data support earlier studies that although auditory booms are disruptive, they do not result in nest abandonment or nest failure.

Isoprenoids are the largest group of natural products, found in all living organisms and play an essential role in numerous cellular processes. These compounds are synthesized by prenyltransferases, catalyzing the condensation reaction between an allylic diphosphate primer and a variable number of isopentenyl diphosphate (C5) units. This superfamily of enzymes can be subdivided into trans- or cis-prenyltransferases according to the stereoisomerism of the product. The cis branch can be further classified according to product length. While the active site volume was suggested to determine the final length in enzymes synthesizing short- and medium-chain products (up to C60), long-chain enzymes (up to C120) and rubber synthases (>C10,000) fail to conform to this paradigm. Here, to resolve the structural basis for long-chain isoprenoid synthesis, we focused on the human cis-prenyltransferase complex (hcis-PT). This enzyme, peripheral to the endoplasmic reticulum membrane, produces the precursor for dolichol phosphate, a membrane residing glycosyl carrier. In line with its crucial role in the cellular protein glycosylation machinery, disease-causing mutations in hcis-PT were shown to result in a wide spectrum of clinical phenotypes. The crystallographic structures of hcis-PT in four different substrate/product-bound conformations revealed an outlet enabling product elongation into the bulk solvent. Moreover, hydrogen-deuterium exchange mass spectrometry analysis in solution showed that the hydrophobic active site core is flanked by dynamic regions consistent with separate inlet and outlet orifices. Finally, using a fluorescent substrate analog and a fluorescently-labeled lipid nanodiscs, we show that product elongation and membrane association are closely correlated. Together, our results support directional product synthesis in long-chain enzymes and rubber synthases, with a distinct substrate inlet and product outlet, allowing direct membrane insertion of the elongating isoprenoid during catalysis. This mechanism uncouples active site volume from product length and circumvents the need to expulse hydrophobic product into a polar environment prior to membrane insertion.

The aim of this study was to analyze the biological role of different transforming growth factor-β (TGFβ) receptor splice variants in ovarian carcinoma (OC). Specific receptor variant knockouts (KO) were prepared using the CRISPR/Cas9 genome editing system in two OC cell lines, TβRI variant 1 (TβRIv1) KO in ES-2 cells and TβRII variant 1 (TβRIIv1) KO in OVCAR-8 cells. Control and KO cells were compared by proteomic analysis, functional tests, analysis of epithelial–mesenchymal transition (EMT) drivers, and Western blot of signaling proteins. Proteomic analysis revealed significant changes in protein pathways in the KO cells. TβRIv1 KO resulted in a significant reduction in both cellular motility and invasion, while TβRIIv1 KO significantly reduced cellular motility and increased Reactive Oxygen Species (ROS) production. Both receptor variant KOs reduced MET protein levels. Of the EMT drivers, a significant decrease in TWIST protein expression, and increase in SNAIL protein and MALAT1 mRNA levels were observed in the TβRIIv1 KO compared to control. A significant decrease in JNK1 and JNK2 activation was found in the TβRIv1 KO compared to control cells. These findings provide new insight regarding the biological role of the TGFβ receptor variants in the biology and potentially the progression of OC.

There has been extensive research on the ecology and evolution of social life in animals that live in groups. Less attention, however, has been devoted to apparently solitary species, even though recent research indicates that they also possess complex social behaviors. To address this knowledge gap, we artificially selected on sociability, defined as the tendency to engage in nonaggressive activities with others, in fruit flies. Our goal was to quantify the factors that determine the level of sociability and the traits correlated with this feature. After 25 generations of selection, the high-sociability lineages showed sociability scores about 50% higher than did the low-sociability lineages. Experiments using the evolved lineages indicated that there were no differences in mating success between flies from the low and high lineages. Both males and females from the low lineages, however, were more aggressive than males and females from the high lineages. Finally, the evolved lineages maintained their sociability scores after 10 generations of relaxed selection, suggesting no costs to maintaining low and high sociability, at least under our settings. Sociability is a complex trait, which we currently assess through genomic work on the evolved lineages.

Changes in ecosystems resulting from anthropause caused by Covid-19 relate to both abiotic and biotic factors which have both a positive or negative effect on wildlife. The lockdown was manifested by reduced air and water pollution, lower mortality of animals on the roads, an increase in animals' body condition and reproduction success. On the other hand, the closures lead to an increase in the populations of invasive species or poaching. We studied the behavioural reaction of natural, desert-dwelling Nubian ibex (Capra nubiana) on the appearance of a new element in the environment - the facial-masks. We hypothesized that the mask would trigger a response expressed through differences in the vigilance towards a potentially new threat. We applied the flight initiation distance (FID) technique to check the reaction at the approach of a human with a facial-mask and without it. The average FID was 8.8 m and was longer when the observer was wearing a mask (10.7 m) as compared to trials without the mask (6.9 m). Our study indicates that wildlife, even if habituated to human silhouette at a distance, appear to notice unusual accessories when up-close and respond by increased vigilance and what may affect their overall fitness.

Behavioral handedness is known to enhance an individual’s handling capabilities. However, the ecological advantages in brachyuran crustaceans remain unclear, despite the Ocypode species having been studied extensively. Thus, in this study, we analyzed the laterality of the endemic Red Sea ghost crab on one beach in Eilat, Israel. We successfully documented the laterality of the large cheliped in 125 crabs, in 60 (48.0%), the right cheliped was larger, and in 64 (51.2%), the left. We also observed temporal segregation between the right- and left-clawed crabs. The right-handed crabs start activity just after sunrise, while left-handed crabs appear ca. 40 min after it. Similarly, temporal segregations were also observed in the evening. The right-clawed crab activity peaked ca. 20 min before sunset, while the left-clawed crabs were active uniformly. Additionally, burrow entrances corresponded to the larger cheliped of the resident individual and is probably a self-defense-related behavior. We conclude that cheliped laterality in O. saratan populations should be considered as a bimodal trait, where left- and right-handedness is not under natural selection pressure.

Recent studies on the oral, anaerobic, gram-negative bacterium Fusobacterium nucleatum revealed its presence and involvement in colorectal, esophageal and breast cancer. We previously demonstrated that F. nucleatum binds and activates the human inhibitory receptors TIGIT and CEACAM1 leading to inhibition of T and NK cell anti-tumor immunity. CEACAM1 was found to be bound and activated by the fusobacterial trimeric autotransporter adhesin CbpF. Here we report the generation of a recombinant E. coli expressing full-length CbpF that efficiently binds and activates CEACAM1.

Kaposi’s sarcoma-associated herpesvirus (KSHV), also familiar as human herpesvirus 8 (HHV-8), is one of the well-known human cancer-causing viruses. KSHV was originally discovered by its association with Kaposi’s sarcoma (KS), a common AIDS-related neoplasia. Additionally, KSHV is associated with two B-lymphocyte disorders; primary effusion lymphoma (PEL) and Multicentric Castlemans Disease (MCD). DNA methylation is an epigenetic modification that is essential for a properly functioning human genome through its roles in chromatin structure maintenance, chromosome stability and transcription regulation. Genomic studies show that expressed promoters tend to be un-methylated whereas methylated promoters tend to be inactive. We have previously revealed the global methylation footprint in PEL cells and found that many cellular gene promoters become differentially methylated and hence differentially expressed in KSHV chronically infected PEL cell lines. Here we present the cellular CpG DNA methylation footprint in KS, the most common malignancy associated with KSHV. We performed MethylationEPIC BeadChip to compare the global methylation status in normal skin compared to KS biopsies, and revealed dramatic global methylation alterations occurring in KS. Many of these changes were attributed to hyper-methylation of promoters and enhancers that regulate genes associated with abnormal skin morphology, a well-known hallmark of KS development. We observed six-fold increase in hypo-methylated CpGs between early stage of KS (plaque) and the more progressed stage (nodule). These observations suggest that hyper-methylation takes place early in KS while hypo-methylation is a later process that is more significant in nodule. Our findings add another layer to the understanding of the relationship between epigenetic changes caused by KSHV infection and tumorigenesis.
