Lagerstroemia ovalifolia Teijsm. & Binn. (LO) (crape myrtle) has reportedly been used as traditional herbal medicine (THM) in Java, Indonesia. Our previous study revealed that the LO leaf extract (LOLE) exerted anti-inflammatory effects on lipopolysaccharide (LPS)-stimulated RAW264.7 macrophages. Based on this finding, the current study aimed to evaluate the protective effects of LOLE in a mouse model of LPS-induced acute lung injury (ALI). The results showed that treatment with LPS enhanced the inflammatory cell influx into the lungs and increased the number of macrophages and the secretion of the inflammatory cytokines in the bronchoalveolar lavage fluid (BALF) of mice. However, these effects were notably abrogated with LOLE pretreatment. Furthermore, the increase of inducible nitric oxide synthase (iNOS), cyclooxygenase-2 (COX-2) and monocyte chemoattractant protein-1 (MCP-1) expression in the lung tissues of mice with ALI was also reversed by LOLE. In addition, LOLE significantly suppressed the LPS-induced activation of the MAPK/NF-κB signaling pathway and led to heme oxygenase-1 (HO-1) induction in the lungs. Additionally, in vitro experiments showed that LOLE enhanced the expression of HO-1 in RAW264.7 macrophages. The aforementioned findings collectively indicate that LOLE exerts an ameliorative effect on inflammatory response in the airway of ALI mice.

Bischofia javanica (Blume) has been traditionally used to treat inflammatory diseases such as tonsillitis and ulcers throughout Asia, including China, Indonesia, and the Philippines: however, the molecular mechanisms by which B. javanica exerts its antioxidant and anti-inflammatory properties remain largely unknown. In this study, we analyzed the antioxidant and anti-inflammatory mechanisms of methanol extracts of B. javanica leaves (MBJ) in vitro and in vivo. MBJ decreased nitric oxide (NO) production and the expression of pro-inflammatory cytokines, including interleukin (IL)-1β, IL-6, and tumor necrosis factor-α, in lipopolysaccharide (LPS)-treated RAW 264.7 cells. The observed suppression of inflammatory responses by MBJ was correlated with an inhibition of the nuclear factor-κB (NF-κB) and the mitogen-activated protein kinase (MAPK) pathways. Additionally, MBJ induced nuclear translocation of the nuclear factor erythroid 2-related factor 2 (Nrf2), a transcription factor that upregulates the expression of anti-inflammatory and antioxidant genes. Furthermore, MBJ exhibited antioxidant and anti-inflammatory effects in an acute hepatitis mouse model. In conclusion, our results confirm the medicinal properties of B. javanica, and therefore MBJ could be applied to improve inflammatory and redox imbalances in different types of pathologies.

Hyperuricemia is a disease that is characterized by high uric acid levels, in which the number of victim increase year by year in the worldwide. Flavonoid is an active compound with inhibitory activity towards Xanthine Oxidase Enzyme which is a compound that plays a role in the formation of uric acid in the body.Sansevieria trifasciatais an ornamental plant which is also useful as a source of antibacterial and antioxidant agent. Studies of S.trifasciataas Xanthine Oxidase Enzyme inhibitor have not been reported. This research isolate flavonoid compounds using open column chromatography from crude extract of S.trifasciataleaves that extracted by sonication method. There are six eluent used to isolate flavonoid which are methanol : ethyl acetate, chloroform : ethyl acetate, chloroform : ethyl acetate : methanol. Wilstater test is used to select the fraction that rich of flavonoid. The best result from isolation step that contains flavonoid is assessed the inhibitory activity of xanthine oxidase. It is analyzed qualitative using Liquid Chromatography Mass Spectrometry (LCMS). The inhibition percentage showed that fraction 3 was potential to inhibit XO by 85.48 %. LC-MS chromatogram can show that crude extract and positive fraction of isolation were containing falvonoid.

Hiperuresemia prevalence tends to increase in society. A combined extract of Uncaria gambir (gambir) and Caesalinia sappan (secang), had been proven to reduce blood uric acid level in vivo. This study aimed to evaluate the subchronic toxicity of this combination in male and female Sprague Dawley rat strain. Animals were randomly grouped into four groups, namely, DOSE-1 (75 mg/kg bw), DOSE-2 (300 mg/kg bw), DOSE-3 (1200 mg/kg bw) and control group gavaged with carrier. The tested sample was given for 7 weeks orally. The result of blood biochemical parameters were not different significantly compared to control (p> 0.05), as well as the results of hematology analysis. However, the NEUT level of female of DOSIS-2 showed lower and significantly different compared to control (p 0,05). Furthermore, this dose did not influence feed intake and body weight of animals in each sex. From this study, it could be concluded that the combination administrated at the dose of 75 mg/kg bw for 7 consecutive weeks did not affect blood biochemistry and hematology and also organ histopathology of kidney, liver, heart, small intestine, and stomach.

A methanol soluble fraction extracted from Kayu Manik leaves (Trema orientalis) from Seluma, Bengkulu, exhibited an antiradical activity 69.73% (scavenging activity of the stable 1,1-diphenyl-2-picryl hydrazyl (DPPH) free radical) that was almost similar to that of 1 mM ascorbic acid. On the other hand, the iron chelating activity was 40.74%. We believe that it would be useful to take the results as an alternative for processing industries and can be observed as a good source of new agent for iron chelator.

Objective: Hyperuricemia (high uric acid levels) prevalence increased year by year. This study was aimed to elaborate the in vitro xanthine oxidase (XO) inhibitory activity and in vivo lowering hyperuricemic effect of Uncaria gambir (Hunter) Roxb) (gambir), Caesalpinia sappan L. (secang) and the combined extract of secang and gambir (formulae extract [FE]). Methods: Gambir and secang extracts were prepared by maceration with ethanol and FE was the proportioned combination of these two extracts. XO inhibitory activity was determined by measuring the formation of uric acid in the xanthine/XO system in vitro using allopurinol as a positive control at 100 ug/mL. Antioxidant activity was by 1,1-diphenyl-2-picrylhydrazyl radical reducing methods. The in vivo experiments were conducted in the oxonate-induced hyperuricemia rat model, in which FE was gavaged p.o. at the arrange dose of 75, 150, and 300 mg/kg bw for 2 weeks. Polyphenol content was measured using Folin–Ciocalteu reagent spectrophotometrically. Results: The XO inhibitory activity of FE was 80% of allopurinol, while secang and gambir were 98% and 50%, respectively. The strength was appropriate to the total polyphenol content, in which it decreased in the order of secang (99%) > FE (86%) > gambir (46%). Furthermore, FE at all tested doses was able to decrease uric acid levels. FE also demonstrated antioxidant activity with a value of 74% relative to Vitamin C at 4 ug/mL. Conclusion: These studies could be concluded that FE exhibited the ability to decrease uric acid level so that it was potential to be developed further as a uric acid-lowering agent.
