Marine invasive species and their bioactive metabolites have become critical ecological issues in the Mediterranean Sea. In particular, the highly invasive green algae Caulerpa taxifolia and Caulerpa cylindracea are known to contain the bioactive sesquiterpene caulerpenyne (1) and the bisindolic alkaloid caulerpin (2), potentially acting as chemical stressors for native species. The recent spread of a variety of C. taxifolia, Caulerpa taxifolia var. distichophylla, also raises urgent questions about its chemical composition. Indeed, the only chemical data available for this alga are limited to the seasonal variations of caulerpenyne (1) in samples collected in the Eastern Mediterranean. In this study, we confirmed the presence of 1 also in C. taxifolia var. distichophylla collected along the Sicilian coast, while 2 was not detected in the alga. However, caulerpin(2) was found both in a Mediterranean specimen of C. taxifolia, and at a much higher level in the congeneric C. cylindracea. This suggests that C. taxifolia var. distichophylla differs from C. taxifolia in its secondary metabolism, potentially exerting dissimilar chemically mediated impacts on native biota. Further chemical investigations on the terpenoidic content of C. taxifolia var. distichophylla led to isolate and identify squalene 2,3 oxide (3), phytol (4), and plastoquinone (5), along with the two unreported sesquiterpene lactones 6 and 7. Finally, chemoecological assays clarified that caulerpenyne (1) at its natural concentration in C. taxifolia var. distichophylla elicits avoidance responses in native shrimp, although sensitization was a prerequisite to significantly induce food rejection.

The ethno-pharmaceutical use of the edible fixed oil produced from lentisk (Pistacia lentiscus) berries covers a long tradition in several Mediterranean regions. Many of the health-promoting properties of lentisk berry oil (LBO) have been associated with the content of polar (poly)phenolic compounds. However, the polar fraction (methanol 80%, v/v) of LBO (LBO-pf) remains poorly and inadequately characterized. We assessed the phytochemical composition (fatty acids, phytosterols and polyphenols) of cold-pressed LBO produced in Cilento (Campania region, Italy) over four years of production (2015-2018). Main phenolic compounds present in LBO-pf were identified and semi-quantified combining ultra-high performance liquid chromatography-electrospray ionization-tandem mass spectrometry (UHPLC-ESI-MS/MS) and HPLC with diode array detection. Phenolic compounds, also responsible for oil stability and antioxidant properties, are relatively abundant in LBO, compared to other edible oils. LBO-pf induced clear dose-dependent effects on the growth of HT-29 cell line derived from human colorectal adenocarcinoma, as evidenced by the cell cycle arrest. Our data support the health-promoting properties of cold-pressed LBO, which is obtained with good yield from spontaneous plants growing in semiarid regions.

An unprecedented phosphatidylmonogalactosyldiacylglycerol pool (PGDG, 1) rich in polyunsaturated fatty acids was isolated from the marine diatoms Thalassiosira weissflogii. Here we report for the first time the NMR characterization of this rare lipid from marine organisms along with a synthetic strategy for the preparation of a PGDG analog (2). PGDG 1 exhibited immunostimulatory activity in human dendritic cells (DCs) and the synthetic PGDG 2 was prepared to explore its mechanism of action. A Toll-like receptor-4 (TLR-4) agonistic activity was evidenced in human and murine DCs underlying the antigen-specific T-cell activation of this class of molecules.

We have exploited the properties of filamentous bacteriophage fd to deliver immunologically active lipids together with antigenic peptides. Filamentous bacteriophages resemble for size, capability to be permeable to blood vessels and high density antigen expression, a nature-made nanoparticle. In addition, their major coat protein pVIII, which is arranged to form a tubular shield surrounding the phage genome, has a high content of hydrophobic residues promoting lipid association. We conjugated bacteriophages to alpha-GalactosylCeramide (alpha-GalCer), a lipid antigen stimulating invariant Natural Killer T (iNKT) cells and capable of inducing their anti-tumoral activities. We found that bacteriophage fd/alpha-GalCer conjugates could repeatedly stimulate iNKT cells in vitro and in vivo, without inducing iNKT anergy. Moreover, co-delivery of alpha-GalactosylCeramide and a MHC class I restricted tumor associated antigenic determinant to APCs via bacteriophages strongly boosted adaptive CD8+ T cell response and efficiently delayed tumor progression. Co-delivery of a tumor antigen and iNKT-stimulatory lipid on the surface of filamentous bacteriophages is a novel approach to potentiate adaptive anti-cancer immune responses, overcoming the current limitations in the use of free alpha-GalactosylCeramide and may represent an attractive alternative to existing delivery methods, opening the path to a potential translational usage of this safe inexpensive and versatile tool.

Prostaglandins (PGs) are hormone-like mediators in many physiological and pathological processes that are present in all vertebrates, in some terrestrial and aquatic invertebrates, and have also been identified in some macroalgae. They have recently been reported also in marine microalgae but their role as chemical mediators is largely unknown. Here we studied the expression pattern of the PG biosynthetic pathway during different growth phases of the centric diatom Thalassiosira rotula and assessed the release of PGs in the surrounding environment for the first time. We show that enzymes responsible for PGs formation such as cyclooxygenase, prostaglandin E synthase 2-like and prostaglandin F synthase are mainly expressed at the end of the exponential phase and that PGs are released especially during the stationary and senescent phases, suggesting a possible signaling function for these compounds. Phylogenetic analysis of the limiting enzyme, COX, indicate the presence in diatoms of more than one enzyme related to the oxidative metabolism of fatty acids belonging to the peroxidase-cyclooxygenase superfamily. These findings suggest a more complex evolution and diversity of metabolic pathways leading to the synthesis of lipid mediators in diatoms.

Two new polypropionates, named exiguapyrone (2) and exiguaone (3) along with the known haminol-1 (4) and -2 (5), have been isolated from the lipidic extract of the Mediterranean cephalaspidean mollusc Haminoea exigua. The regular propionate skeletons, structurally related to other polypropionates from the congener Haminoea fusari, have been elucidated by means of NMR techniques as natural (2 and 3) and alpha-/gamma-pyrone methyl derivatives (2a and 2b). This is a further report showing the co-occurrence of alkyl-pyridines and polypropionates in Haminoea molluscs strengthening the role of polypropionates as chemical markers among cephalaspideans. Wheat beer is a traditional light-colored top-fermenting beer brewed with at least 50% malted (e.g., German Weissbier) or unmalted (e.g., Belgian Witbier) wheat (Triticum aestivum) as an adjunct to barley (Hordeum vulgare) malt. For the first time, we explored the proteome of three Weissbier samples, using both 2D electrophoresis (2DE)-based and 2DE-free strategies. Overall, 58 different gene products arising from barley, wheat, and yeast (Saccharomyces spp.) were identified in the protein fraction of a representative Weissbier sample analyzed in detail. Analogous to all-barley-malt beers (BMB), barley and wheat Z-type serpins and nonspecific lipid transfer proteins dominated the proteome of Weissbier. Several alpha-amylase/trypsin inhibitors also survived the harsh brewing conditions. During brewing, hundreds of peptides are released into beer. By liquid chromatography-electrospray tandem mass spectrometry (LC-ESI MS/MS) analysis, we characterized 167 peptides belonging to 44 proteins, including gliadins, hordeins, and high- and low-molecular-weight glutenin subunits. Because of the interference from the overabundant yeast-derived peptides, we identified only a limited number of epitopes potentially triggering celiac disease. However, Weissbier samples contained 374, 372, and 382 ppm gliadin-equivalent peptides, as determined with the competitive G12 ELISA, which is roughly 10-fold higher than a lager BMB (41 ppm), thereby confirming that Weissbier is unsuited for celiacs. Western blot analysis demonstrated that Weissbier also contained large-sized prolamins immunoresponsive to antigliadin IgA antibodies from the pooled sera of celiac patients (n = 4).

Chemical communication in bacteria relies on the production, release, and detection of small molecules called autoinducers (AIs) that regulate gene expression in a cell-density-dependent manner (quorum sensing, QS). QS not only serve as intraspecies but also in interspecies communication. Furthermore, the clinical significance of interkingdom crosstalk between bacteria and human host is becoming more and more evident. Typically, signal molecules are produced at a very low concentration and their detection requires highly sensitive techniques. Biosensors have allowed fast screening of microorganisms for AI production; however, for their specific identification, the gold choice are mass spectrometry-based methodologies, coupled with either gas or liquid chromatography. Sample extraction, cleanup, and preconcentration greatly improve autoinducer identification from complex matrices allowing reliable quantification. The present chapter provides an overview on the most recent and advanced analytical approaches to identify and quantify these bacterial chemical messages with a special focus on N-acyl-L-homoserine lactones (AHLs/Acyl-HSLs).

Accurate characterization of biomass constituents is a crucial aspect of research in the biotechnological application of natural products. Here we report an efficient, fast and reproducible method for the identification and quantitation of fatty acids and complex lipids (triacylglycerols, glycolipids, phospholipids) in microalgae under investigation for the development of functional health products (probiotics, food ingredients, drugs, etc.) or third generation biofuels. The procedure consists of extraction of the biological matrix by modified Folch method and direct analysis of the resulting material by proton nuclear magnetic resonance (H-1 NMR). The protocol uses a reference electronic signal as external standard (ERETIC method) and allows assessment of total lipid content, saturation degree and class distribution in both high throughput screening of algal collection and metabolic analysis during genetic or culturing studies. As proof of concept, the methodology was applied to the analysis of three microalgal species (Thalassiosira weissflogii, Cyclotella cryptica and Nannochloropsis salina) which drastically differ for the qualitative and quantitative composition of their fatty acid-based lipids.

Olfaction is considered a distance sense; hence, aquatic olfaction is thought to be mediated only by molecules dissolved in water. Here, we challenge this view by showing that shrimp and fish can recognize the presence of hydrophobic olfactory cues by a “tactile” form of chemoreception. We found that odiferous furanosesquiter- penes protect both the Mediterranean octocoral Maasella edwardsi and its specialist predator, the nudibranch gastropod Tritonia striata, from potential predators. Food treated with the terpenes elicited avoidance responses in the cooccurring shrimp Palaemon elegans. Rejection was also induced in the shrimp by the memory recall of postingestive aversive effects (vomiting), evoked by repeatedly touching the food with chemosensory mouthparts. Consistent with their emetic properties once ingested, the compounds were highly toxic to brine shrimp. Further experiments on the zebrafish showed that this vertebrate aquatic model also avoids food treated with one of the terpenes, after having experienced gastrointestinal malaise. The fish refused the food after repeatedly touching it with their mouths. The compounds studied thus act simultaneously as (i) toxins, (ii) avoidance-learning inducers, and (iii) aposematic odorant cues. Although they produce a characteristic smell when exposed to air, the compounds are detected by direct contact with the emitter in aquatic environments and are perceived at high doses that are not compatible with their transport in water. The mouthparts of both the shrimp and the fish have thus been shown to act as “aquatic noses,” supporting a substantial revision of the current definition of the chemical senses based upon spatial criteria.

Diatoms are among the most successful primary producers in ocean and freshwater environments. Deriving from a secondary endosymbiotic event, diatoms have a mixed genome containing bacterial, animal and plant genes encoding for metabolic pathways that may account for their evolutionary success. Studying the transcriptomes of two strains of the diatom Skeletonema marinoi, we report, for the first time in microalgae, an active animal-like prostaglandin pathway that is differentially expressed in the two strains. Prostaglandins are hormone-like mediators in many physiological and pathological processes in mammals, playing a pivotal role in inflammatory responses. They are also present in macroalgae and invertebrates, where they act as defense and communication mediators. The occurrence of animal-like prostaglandins in unicellular photosynthetic eukaryotes opens up new intriguing perspectives on the evolution and role of these molecules in the marine environment as possible mediators in cell-to-cell signaling, eventually influencing population dynamics in the plankton.The ISME Journal advance online publication, 28 March 2017; doi:10.1038/ismej.2017.27.

Bathydoris hodgsoni and Doris kerguelenensis are two of the largest Antarctic nudibranchs. They are both common circumpolar species with broad bathymetric distributions, although B. hodgsoni is restricted to deep waters in the Antarctic high latitude. Egg masses and juveniles of these species were collected over multiple years (1998-2012) in the eastern Weddell Sea and the South Shetland Islands, and here new data are provided about egg mass characteristics and ontogeny using histological techniques. The egg mass of B. hodgsoni has a maximum length of 12.4 cm with one or two egg capsules with a mean diameter of 4.9 cm. The capsules either contained non-developing eggs or ready-to-hatch juveniles up to 2.9 cm long. The egg mass of D. kerguelenensis is a semicircular ribbon-like structure including 1,500-2,400 oval capsules (similar to 1.7 x 1.2 mm) containing various stages of development up to ready-to-hatch juveniles 2.5 mm in length. Based on their morphology and development in egg masses maintained in the laboratory, the embryonic period for B. hodgsoni is estimated to be up to 10 years, and for D. kerguelenensis 13 months. Thus, B. hodgsoni has the largest egg capsules and probably the largest hatchlings of any mollusc. Chemical analyses of D. kerguelenensis egg masses showed no trace of terpenoid acylglycerols, although these compounds were present in field-collected juveniles and adults. None of four sponges that likely serve as food for D. kerguelenensis had the glycerides, or their precursors, found in the nudibranch.

Recent studies have shown that the Mediterranean white sea bream Diplodus sargus includes the invasive green alga Caulerpa cylindracea in its diet, with consequent metabolic and enzymatic alterations. As a result of this novel alimentary habit, the bioactive algal red pigment caulerpin has been detected in its tissues. However, this may not be an isolated case: other fish species have also been reported to feed on C. cylindracea, although the possible accumulation of caulerpin in their tissues has not yet been investigated. In this report, we analysed stomach con- tents and caulerpin levels in the native sparid species Spondyliosoma cantharus, Sarpa salpa, and Diplodus vulgaris, and in the scarid Sparisoma cretense, along with the Lessepsian siganid Siganus luridus. C. cylindracea was found in the stomachs of all but one fish species, the exception being S. cretense, in which prey items could not be determined due to the high degree of diges- tion. Chemical analysis of fish tissues revealed that only S. cantharus and S. salpa accumulated caulerpin, while no traces of the compound were detected in the other species. Despite intense research efforts on natural products obtained from C. cylindracea, a complete picture of the impacts caused by fish including this alga in their diet has not been elucidated. The identification of caulerpin in other Mediterranean native fish suggests a need for further research in order to assess the possible transfer of such molecules to humans through seafood consumption.

Green microalgae contain many active pigments such as carotenoids having antioxidant and protective activity on human cells. Here we investigate the biological activity of an ethanol/water extract of the marine green microalga Tetraselmis suecica containing high levels of carotenoids such as the xanthophylls lutein, violaxanthin, neoxanthin, antheraxanthin and loroxanthin esters. This extract has a strong antioxidant and repairing activity in the human lung cancer cell line (A549) as shown by the increased expression of dehydrocholesterol reductase-24 (DHCR24) and prostaglandin reductase 1 (PTGR1) genes and proteins. The extract also reduces prostaglandin E2 (PGE2) levels in cells damaged by H2O2 and has tissue repairing effects on reconstructed human epidermal tissue cells (EpiDermTM) indicating a potential cosmeceutical activity of this microalgal species.

The chemical study of the Brazilian brittle star Ophionereis reticulata led to the isolation of three chamigrene sesquiterpenes, including the partially characterized isoobtusadiene (1), its unreported acetyl derivative (2), and the known (+)-elatol (3). The complete elucidation of the structures 1 and 2 was accomplished by 1D and 2D NMR spectroscopy. The first assignment of the absolute configuration of the isoobtusadiene skeleton is suggested as 6S,9R,10S on the basis of the NMR analysis of the Mosher's ester derivatives of 1 and the ECD study of the acetyl derivative 2. Chamigrenes are typical constituents of Laurencia red algae. O. reticulata is a predator with a preference for algae. Thus, the origin of these metabolites can be likely ascribed to diet.

Benthic cnidarians are colonial marine animals that host a rich population of associated and symbiotic microorganisms. In a recent paper we described for the first time the isolation of amphidinolide P (1) from the Brazilian octocoral Stragulum bicolor. Amphidinolides and similar compounds had been previously reported only from dinoflagellates of the genus Amphidinium; thus the presence of 1 in the invertebrate opens intriguing questions on the role and occurrence of these molecules in marine ecosystems. Here we report the identification of four further amphidinolides from the same soft coral, including the known amphidinolide T1 (2) and the new analogues here named amphidinolides C4 (3), B8 (4), and B9 (5). The chemical structures have been elucidated mainly by extensive study of spectroscopic data. Cytotoxic activities of 3 and 4 were evaluated against the colon adenocarcinoma cell line HCT-116.

The loss of the shell in nudibranch gastropods has been related to the acquisition of chemical defensive strategies during evolution, such as the use of natural products to deter predation. In the present study, we investigated the origin, location, and putative role of granuloside (1), a homosesterterpene lactone, recently isolated from the Antarctic nudibranch Charcotia granulosa Vayssiere, 1906. Several adults, egg masses, and its bryozoan prey, Beania erecta Waters, 1904, were chemically analyzed by chromatographic and spectroscopic techniques. Light- and transmission electron microscopy of the mantle revealed complex glandular structures, which might be associated with the storage of defensive compounds in analogy to mantle dermal formations described in other nudibranchs. Although preliminary in situ repellence bioassays with live specimens of the nudibranch showed avoidance against the Antarctic generalist sea star predator Odontaster validus, the specific role of the terpene granuloside requires further investigation. The egg masses do not present granuloside, and the glandular structures are absent in the trochophore larvae. Our results suggest that C. granulosa synthesizes granuloside de novo in early stages of its ontogeny, instead of obtaining it from the prey. Considering the wide geographic area inhabited by this slug, this may be advantageous, because natural products produced by the slug will not be affected by fluctuant food availability. Overall, the Antarctic sea slug C. granulosa seems to possess defensive strategies that are similar, in terms of production and storage, to nudibranchs from other regions of the world. This species is one of the few cladobranchs investigated so far that present de novo biosynthesis of a defensive compound.

Opisthobranchs belong to a subclass of highly evolved and specialised marine gastropods that rely on the use of secondary metabolites for their survival. Here we report the full elucidation of the biosynthesis of aromatic metabolites, lignarenones, in one of these gastropods, the cephalaspidean Scaphander lignarius. Feeding experiments with 2H- and 13C-labelled precursors revealed a mixed acetate/propionate polyketide pathway primed by benzoic acid. Phenylalanine ammonia lyase (PAL), unprecedented in animals, is central to the synthesis of this aromatic precursor by oxidative deamination of L-phenylalanine to cinnamic acid. Lignarenones are synthesised in the cytoplasm of specialised eukaryotic cells named Blochmann's glands, which are distributed in biosynthetic tissue localised in the vulnerable mantle of the mollusc. This result supports the hypothesis that this lineage of gastropods has acquired the genetic information to produce the chemical substances that they use for their survival.

Polypropionates that incorporate pyrones are a family of polyketides featuring the chemistry of a few marine molluscs capable of phototrophic CO2 fixation as a result of storing viable symbiotic chloroplasts in their bodies. The role and origin of these molecules is poorly investigated, although the unusual biological ACHTUNGTRENUNGactivities and chemistry of these natural products have recently received renewed interest. Here, we report the results of in vivo studies on production of g-pyrone-containing polypropionates in the Mediterranean mollusc Elysia viridis. Biosynthesis of the metabolites in the sacoglossan is shown to proceed through condensation of eight intact C3 units by polyketide synthase assembly. LC–MS and NMR spectroscopic studies demonstrate that the process involves a pyrone tetraene (10) as key intermediate, whereas the levels of the final polypropionates (6, 7 and 9) are related to each other and show a significant dependence upon light conditions.

Egg production rates and/or hatching success in the copepods Acartia clausi, Calanus helgolandicus and Temora longicornis were negatively affected by a late spring (May-June 2003) phytoplankton bloom in the North Adriatic Sea, dominated mainly by the large diatom Cerataulina pelagica. Highest total concentrations of 3.3- 104 cells-ml(-1) were located in the vicinity of the Po River, which also corresponded to the area where the highest numbers of phaeophorbides were measured (0.779, 0.528 and 0.419 mu g.l(-1), respectively, compared to an average of the remaining stations of 0.183 +/- 0.049 SD), suggesting some grazing on the bloom. Phytoplankton biomass in terms of carbon was dominated by diatoms, representing on average 42% of total phytoplankton carbon and more than 80% at several stations. Cerataulina pelagica, Cyclotella spp., Chaetoceros spp. and small unidentified centric diatoms dominated the diatom community numerically but C. pelagica was by far the dominant diatom in terms of carbon due to its large cell size. This species represented more than 60% of the diatom biomass at nine of the 14 stations sampled, and was absent only at one station, which was the most offshore station sampled during the cruise. Although polyunsaturated aldehydes (PUAs) were not detected, other oxylipins which are hydroxy and keto derivatives of eicosapentaenoic and docosahexaenoic acids that affect copepod reproduction were found in these samples. Hence, we can attribute the negative impact of diatoms not only to PUAs, as previously believed, but also to these compounds. This is the first direct evidence of the presence of oxylipins other than PUAs in marine blooms dominated by diatoms.

The biosynthesis of lignarenones 1 and 2, the major polyketides of the Mediterranean mollusc Scaphander lignarius is described. The process is primed by benzoic acid and requires acetate and propionate as extender units. The labeling pattern suggests PKS-like synthesis of an unusual E,Z,E-triene chain and origin of the benzoate unit from phenylalanine. 13C-13C NMR COSY has been used to establish the labeling positions due to incorporation of 13C2-acetate.

Quorum sensing (QS) mechanisms regulate a variety of vital functions such as motility, growth inhibition, biofilm development, virulence expression and plasmid conjugation. Many of these activities are essential for the well-being of organisms, in particular in extreme conditions such as hypersaline environments. Halophiles are able to synthesize products that are stable, and to exploit their activities under such “extreme” conditions, namely saline environments such as marine habitat, salt lakes, brines and saline soils. The versatility of these products leads researchers to address their interest in biotechnological applications. In this study, the QS activity of Halomonas smyrnensis AAD6—a moderately halophilic and exopolysaccharide-producing bacterium—was investigated. Dichloromethane extracts from stationary phase cultures of Halomonas smyrnensis AAD6 were partially purified, and C16-AHL was identified by LC-MS analysis. The synthesis of autoinducers was assayed and their production was detected starting from 48 h of growth. The growth-phase dependent production of exopolysaccharide (EPS) and signal molecules suggested that these activities could be linked.

Phenolic profiling of ten plant samples of Mentha rotundifolia (L.) Huds. collected from different bioclimatic areas of Tunisia, was for the first time carried out by using a fast ultra-high-performance liquid chromatography (UHPLC)-high resolution tandem mass spectrometry (HRMS/MS) method on a Q Exactive platform equipped with an electrospray ionization (ESI) source. An intraspecific, interpopulation variability was evidenced and a total of 17 polyphenolic metabolites were identified and quantified by using the UHPLC-HRESIMS/MS method, here validated for specificity, linearity, limit of detection, limit of quantitation, accuracy and precision. The quantitative method resulted sensitive at the nM level and reliable for rapid polyphenol quantification in vegetal matrices. The metabolomic study allowed us to identify a new compound, named salvianolic acid W, which was isolated and characterized mainly by NMR and MS analysis. A statistical correlation of the phenolic composition with antioxidant and anti-acetylcholinesterase activities was provided.

Secondary metabolites of the Mediterranean mollusc Scaphander lignarius from different collection sites have been investigated, proving the constant presence of a number of minor metabolites correlated to the already known lignarenones. Complete characterization of the new metabolites has been supported by enantioselective synthesis. The data are consistent with the origin of this unique class of ω-phenyloctanoids from a common polyketide pathway.

Dinoflagellates are an important source of unique bioactive secondary metabolites. Symbiotic species, commonly named zooxanthellae, transfer most of their photosynthetically fixed carbon to their host. The mutualistic relationship provides the organic metabolites used for energy production but there are very few reports of the role of the dinoflagellates in the production of secondary metabolites in the symbiotic association. Corals and other related cnidarians are the most well-known animals containing symbiotic dinoflagellates. In the present paper we describe the isolation of amphidinolide P (1) from the octocoral Stragulum bicolor and its prey, the nudibranch Marionia limceana, collected off the coasts of Fortaleza (CearA, Brazil). The coral extracts also contained 3-O-methyl derivative (2) of amphidinolide P, together with minor compounds still under investigation. Amphidinolides have been so far reported only in laboratory cultures of Amphidinium sp., thus compounds 1 and 2 represents the first identification of these polyketides in invertebrates. The finding proves the possibility to isolate amphidinolides from a natural symbiosis, enabling further biological and biotechnological studies. (C) 2015 Sociedade Brasileira de Earmacognosia. A new homosesterterpene with a unique linear skeleton, named granuloside (1), has been fully characterized from the Antarctic nudibranch Charcotia granulosa Vayssiere, 1906 (Mollusca: Gastropoda). The planar structure of 1 was determined by extensive spectroscopic techniques on the methyl derivatives (la and 1b), and the R absolute configuration at C-4 is suggested by comparison of experimental and calculated ECD spectra of 1b. Granuloside (1) is the first linear homosesterterpene skeleton ever reported and, despite the low molecular complexity, its chemical structure poses many questions about its biogenesis and origin in the nudibranch.

The green alga Caulerpa racemosa is a non-native, invasive species in the Mediterranean, and an important stressor for several native organisms. The algal capacity to produce secondary metabolites has been suggested to modulate success of the C. racemosa invasion, although many of potentially involved biological pathways still remain unexplored. In this respect, the aim of the present study was to investigate some molecular and cellular effects in the white seabream Diplodus sargus , an ecologically key species, which included the alien C. racemosa in its diet. Organisms were sampled in 2 seasonal periods from 3 locations of Southern Italy, each characterized by different levels of algal abundance. The level of caulerpin, one of the main secondary algal metabolites, in fish tissues has been used as an indicator of the trophic exposure to the seaweed and related with molecular and cellular responses. Chemical analyses indicated that fish from invaded sites can accumulate caulerpin, with liver concentrations ranging from a few up to hundreds of μg/g. Biomarkers analyses revealed only limited alterations of the main antioxidant defences, such as glutathione reductase and levels of glutathione; on the other hand, increased enzymatic activities of cytochrome P450, glutathione S-transferases and acyl CoA oxidase, as well as enhanced gene transcription for peroxisome proliferator-activated receptor alpha, cytochrome P4501A and vitellogenin 1 were observed in fish more exposed to C. racemosa as indicated by liver concentrations of caulerpin higher than 50 μg/g. Despite a direct molecular relationship with this algal metabolite could not be established, our results suggest that a C. racemosa enriched diet can modulate biotransformation and fatty acids metabolism of D. sargus. Assessing whether similar effects represent short- or long-term effects will be of crucial importance to understand consequences on the general health status and reproductive performance of exposed key fish species in the Mediterranean region.

Diatom oxylipins have been observed to deleteriously impact copepod reproductive success. However, field studies have revealed very variable and case-dependent results. Therefore, the plasticity of diatom oxylipin metabolism was studied among four clones of the marine diatom Skeletonema marinoi Sarno et Zingone. Diatom oxylipin metabolism was studied by two lipoxygenase (LOX) activity assays carried out at different pH values and by oxylipin quantification. The four clones showed no major metabolic differences in terms of protein content or growth rate. However, two of the clones produced significantly higher levels of oxylipins than the other two. LOX activity measurements also indicated clonal variability in fatty acid oxidative metabolism. The presence of clone-specific differences in oxylipin metabolism may play a role in shaping diatom population dynamics by conferring selective advantages to certain clones.

Oxylipins are oxygenated derivatives of polyunsaturated fatty acids (PUFAs) that act as chemical mediators in many ecological and physiological processes in marine and freshwater diatoms. The occurrence and distribution of these molecules are relatively widespread within the lineage with considerable species-specific differences due to the variability of both the fatty acids recognized as substrates and the enzymatic transformations. The present review provides a general introduction to recent studies on diatom oxylipins and describes an analytical method for the detection and assessment of these elusive molecules in laboratory and field samples. This methodology is based on selective enrichment of the oxylipin fraction by solvent extraction, followed by parallel acquisition of full-scan UV and tandem mass spectra on reverse phase liquid chromatography (LC) peaks. The analytical procedure enables identification of potential genetic differences, enzymatic regulation, and ecophysiological conditions that result in different oxylipin signatures, thus providing an effective tool for probing the functional relevance of this class of lipids in plankton communities. Examples of oxylipin measurements in field samples are also provided as a demonstration of the analytical potential of the methodology.

Two new polyketides of the amphidinol family, amphidinol 18 (wAM18, 1) and its corresponding 7-sulfate derivative (AM19, 2), have been isolated from the MeOH extract of the dinoflagellate Amphidinium carterae. Structure elucidation of the two polyoxygenated molecules has been accomplished by extensive use of spectroscopic and spectrometric techniques. AM18 exhibited antifungal activity against Candida albicans at 9 mu g/mL.

Oxylipins are a large and diverse family of fatty acid derivatives exhibiting different levels of oxidation of the carbon chain. They are involved in many biological functions in mammals, plants and diatoms. In this last group of organisms, they are suggested to play a role in the reproductive failure of copepod predators, showing clear pro-apoptotic effects on newborn nauplii. In this work, these compounds were tested for the ability to induce mitotic arrest in sea urchin embryos. We show for the first time that oxylipins have an increased efficacy in their corresponding methylated form. Natural oxylipins were also used as an inspiration for the rational design and synthesis of stable chemical analogs with apoptotic activity against tumor cell lines. This approach led to the synthesis of the linear C15-ketol (22) that was shown to induce apoptosis in human leukemia U-937 cells. These results are proof of the concept of the use of eco-physiological considerations as a platform to guide the search for novel drug candidates.

From the Antarctic sponge Lyssodendoryx flabellata, a new polycyclic compound, which we named flabellone, related to the oestrogenic hormone oestrone, has been elucidated by spectrometric and spectroscopic means. Along with flabellone, a glycosphingolipid (GSL) mixture featuring an unusual alpha-fucofuranosyl-3-beta-glucopyranoside unit structurally identical to the previously reported terpioside has been isolated and identified. Chemical studies in the octocoral Renilla octodentata from San Jorge Gulf (Patagonia, Argentina) have been carried out for the first time. Two conjugated polyunsaturated fatty acids, 1 and 2, named renillenoic acids (RAs), were isolated and characterized from the sea pen. These compounds, analogs of eicosapentaenoic acid (EPA) and arachidonic acid (AA), accounted for a large fraction (20%) of the organic extract of the octocoral. Renillenoic acids were uniform distributed in rachis and peduncle and showed comparable volumetric concentrations, 1.61 +/- 0.51 mg/cm(3) and 2.00 +/- 1.01 mg/cm(3) respectively (Paired Student's t-test, two tailed, p>0.05). The green alga Caulerpa racemosa var. cylindracea has invaded Mediterranean seabed including marine reserves, modifying the structure of habitats and altering the distributional patterns of associated organisms. However, the understanding of how such invasion can potentially affect functional properties of Mediterranean subtidal systems is yet to be determined. In this study, we show that C. racemosa changes foraging habit of the native white seabream, Diplodus sargus. In invaded areas, we found a high frequency of occurrence of C. racemosa in the stomach contents of this omnivorous fish (72.7 and 85.7%), while the alga was not detected in fish from a control area. We also found a significant accumulation of caulerpin, one of the main secondary metabolites of C. racemosa, in fish tissues. The level of caulerpin in fish tissues was used here as an indicator of the trophic exposure to the invasive pest and related with observed cellular and physiological alterations. Such effects included activation of some enzymatic pathways (catalase, glutathione peroxidases, glutathione S-transferases, total glutathione and the total oxyradical scavenging capacity, 7-ethoxy resorufin O-deethylase), the inhibition of others (acetylcholinesterase and acylCoA oxidase), an increase of hepatosomatic index and decrease of gonadosomatic index. The observed alterations might lead to a detrimental health status and altered behaviours, potentially preventing the reproductive success of fish populations. Results of this study revealed that the entering of alien species in subtidal systems can alter trophic webs and can represent an important, indirect mechanism which might contribute to influence fluctuations of fish stocks and, also, the effectiveness of protection regimes.

Structural similarity or even the identity of polyketide compounds does not necessarily imply unique biosynthesis. Feeding experiments with a (13)C labeled precursor establish that the C(3) units in 7-methyl-cyercene-1 (1) are derived from intact propionate in the marine mollusc Ercolania funerea. The same compound in the terrestrial fungus Leptosphaeria maculans/Phoma lingam is synthesized by an acetate/SAM pathway thus proving for the first time metabolic convergence of polyketide biosynthesis in eukaryotes. Traditional (1)H-(13)C NMR correlation spectroscopy has been successfully applied to estimate (13)C incorporation in biosynthetic experiments.

One of the most promising alternative technologies to antifouling (AF) biocides based on toxic heavy metals lies in the development of natural eco-friendly biocides. The present study evaluates the AF potential of structurally different compounds containing a 3-alkylpyridine moiety. The products, namely poly 3-alkylpyridinium salts, saraine, and haminols, were either extracted or derived from natural sources (the sponges Haliclona sp. and Reniera sarai and the mollusc Haminoea fusari), or obtained by chemical synthesis. All the molecules tested showed generally good anti-settlement activity against larvae of the barnacle Amphibalanus (=Balanus) amphitrite (EC50 values between 0.19 and 3.61 μg ml−1) and low toxicity (LC50 values ranging from 2.04 to over 100 μg ml−1) with non-target organisms. For the first time, the AF potential of a synthetic monomeric 3-alkylpyridine was demonstrated, suggesting that chemical synthesis is as a realistic way to produce large amounts of these compounds for future research and development of environmentally-friendly AF biocides.

The polar extract of the Vanuatu sponge Reniera n. sp., which showed immunomodulating activity in preliminary tests, was found to contain a cyclic tripeptide, which we named renieramide (1). This metabolite is identical to a synthetic derivative mentioned in a patent concerning the preparation of cyclic peptides of the OF4949 family of anticancer agents. We describe here the first isolation of this metabolite from natural sources and its complete characterization by spectroscopic and chemical approaches. Renieramide (1) possesses a 17-membered cyclic side-chain-linked biphenyl ether skeleton, typical of the class that includes the natural products OF4949 I-IV, K13, and eurypamides. A tridimensional model of 1, obtained by NMR restrained molecular mechanics and dynamics, is also presented.

Structural diversity of polypropionates arises from the amazing combination of acetate and propionate units. Feeding experiments with single and doubly 13 C -labelled precursors prove that the Mediterranean slug Placida dendritica utilizes intact C 3 -units for the biosynthesis of placidenes (e.g., 1 – 5 ), prototypes of a family of pyrone-containing polypropionates largely represented in fungi and marine invertebrates. These results show that fungi and molluscs have elaborated two distinct polyketide pathways for the synthesis of similar or even identical compounds.

Summary • In recent years, oxylipins (lipoxygenase-derived oxygenated fatty acid products) have been reported in several bloom-forming marine diatoms. Despite increasing attention on the ecophysiological role of these molecules in marine environments, their biosynthesis is largely unknown in these microalgae. • Biochemical methods, including tandem mass spectrometry, nuclear magnetic resonance and radioactive probes were used to identify structures, enzymatic activities and growth-dependent modulation of oxylipin biosynthesis in the pennate diatom Pseudo-nitzschia delicatissima. • Three major compounds, 15S-hydroxy-(5Z,8Z,11Z,13E,17Z)-eicosapentaenoic acid (15S-HEPE), 15-oxo-5Z,9E,11E,13E-pentadecatetraenoic acid and 13,14-threo-13R-hydroxy-14S,15S-trans-epoxyeicosa-5Z,8Z,11Z,17Z-tetraenoic acid (13,14-HEpETE), were produced by three putative biochemical pathways triggered by eicosapentaenoic acid-dependent 15S lipoxygenase. Oxylipin production increases along the growth curve, with remarkable changes that precede the demise of the culture. At least one of the compounds, namely 15-oxoacid, is formed only in the stationary phase immediately before the collapse of the culture. • Synthesis and regulation of phyco-oxylipins seem to correspond to a signaling mechanism that governs adaptation of diatoms along the growth curve until bloom termination. Factors triggering the process are unknown but synthesis of 15-oxoacid, constrained within a time-window of a few days just before the collapse of the culture, implies the involvement of a physiological control not directly dependent on distress or death of diatom cells.

This paper reports the study of terpene biosynthesis in the marine nudibranch Doriopsilla pelseneeri. In ViVo feeding experiments with 13C-glucose proved the de noVo origin of the terpene metabolites via a mevalonate pathway. Characterization of the stereochemical relationship of the new acetyl pelseneeriols (7 and 8) suggests a two-step mechanism for the sesquiterpene cyclization, leading to a slight refinement of terpene tailoring in this family of marine invertebrates.

Some marine diatoms negatively affect the reproduction of dominant zooplankton grazers such as copepods, thus compromising the transfer of energy through the marine food chains. In this paper, the metabolic mechanism that leads to diatom-induced toxicity is investigated in three bloom-forming microalgae. We show that copepod dysfunctions can be induced by highly reactive oxygen species (hROS) and a blended mixture of diatom products, including fatty acid hydroperoxides (FAHs); these compounds display teratogenic and proapoptotic properties. The process is triggered by the early onset of lipoxygenase activities that elicit the synthesis of species-specific products, the basic structures of which were established (1-20); these compounds boost oxidative stress by massive lipid peroxidation. Our study might explain past laboratory and field results showing how diatoms damage zooplankton grazers even in the absence of polyunsaturated aldehydes, a class of molecules that has been formerly implicated in mediating the toxic activity of diatoms on copepods.

A novel regular polypropionate skeleton has been characterized in two new pyrones, named fusaripyrone A (1) and fusaripyrone B (2), from the lipid extracts of the mollusc Haminoea fusari. Here we report elucidation of the polyketide structures of 1 and 2, isolated as either natural products or isomeric pairs of alpha-/gamma-pyrone methyl derivatives.

Enzymatic preparations and specialized analytical tools have shown that chloroplast-derived glycolipids are the main substrates for the biosynthetic pathway that produces antiproliferative polyunsaturated aldehydes in broken cells of the marine diatom Thalassiosira rotula. This process, which is associated with the formation of free fatty acids and lyso compounds from polar lipids but not triglycerides, is largely dependent on glycolipid hydrolytic activity, rather than phospholipase A(2) as previously suggested. Preliminary characterization of lipolytic enzymes has revealed protein bands of 40-45 kDa. Under native conditions these proteins seem to be associated with soluble aggregates that have an apparent molecular weight of approximately 200 kDa. The biochemical process, which is similar to that described in the algal-bloom forming diatom Skeletonema costatum, suggests a mechanism based on decompartmentalization and mixing of preexisting enzymes and substrates.

Intermediates of the aldehyde biosynthesis in Thalassiosira rotula are investigated. Use of labeled precursors and cell preparations proves production of 2 E ,4 Z -octadienal ( 1a ) from 6 Z ,9 Z ,12 Z -hexadecatrienoic acid (C16:3 ω − 4) through the lipoxygenase-dependent intermediate (9 S )-9-hydroperoxyhexadeca-6,10,12-trienoic acid. On the contrary, synthesis of 2 E ,4 Z ,7 Z -decatrienal ( 2a ) involves mainly EPA (C20:5 ω − 3) by a 11 R -lipoxygenase, as suggested by identification of chiral 11 R -HEPE (12% e.e.) in the diatom extracts. Consistently with the necessity to have a rapid transport and metabolization of the intermediate hydroperoxides, we show that lipoxygenase and lyase activities are both found in the same subcellular fraction of the microalga.

A new unusual phytotoxic bis-amino acid N-glucoside, named ascaulitoxin, was isolated from the culture filtrate of Ascochyta caulina, the causal agent of leaf and stem necrosis of Chenopodium album, a promising mycoherbicide for the biological control of this common noxious weed. Ascaulitoxin, characterized by extensive use of NMR techniques and chemical methods as N 2 - (2,4,7-triamino-5-hydroxy)-octanedioyl-β- d -glucopyranoside , showed phytotoxic activity against host and non-host plants.

Diatoms are unicellular plants broadly present in freshwater and marine ecosystems, where they play a primary role in sustaining the marine food chain. In the last 10 years, there has been accumulating evidence that diatoms may have deleterious effects on the hatching success of zooplankton crustaceans such as copepods, thus affecting dynamics of planktonic populations and limiting secondary production. At the molecular level, failure to hatch is ascribed to the presence of a family of inhibitory oxylipins, which we propose to collectively name polyunsaturated short-chain aldehydes (abbreviated here as PUSCAs). Here we describe the origin of PUSCAs produced by the marine diatom Skeletonema costatum via a lipoxygenase-mediated pathways involving non-esterified polyunsaturated fatty acids (PUFA). Experiments with complex lipids proved the pivotal role of chloroplast-derived glycolipids, especially monogalactosyldiacylglycerol (MGDG), in providing hexadecatrienoic acid (C16:3 omega-4), hexadecatetraenoic acid (C16:4 omega-1) and eicosapentaenoic acid (C20:5 omega-3) to the downstream process leading to 2E,4Z-octadienal (C8:2 omega-4), 2E,4Z,7-octatrienal (C8:3 omega-1) and 2E,4Z-heptadienal (C7:2 omega-3), respectively. Under physiological conditions, the hydrolytic process is associated to galactolipid hydrolyzing enzyme capable of removing fatty acids from both sn positions of glycerol.

The paper reports the results of biosynthetic experiments with the marine nudibranch Doriopsilla areolata. Biogensis of the enantiomeric sesquiterpenes 1 and 5 was investigated by feeding of [1-13C]-glucose, [1,2-13C2]-glucose and [1,2-13C2]-acetate. Evidence is presented that supports de novo origin of both compounds via an single metabolic pathway based on mevalonic acid.

The biogenesis of verrucosins, diterpenoid glycerides of the marine mollusc Doris verrucosa, has been investigated by feeding experiments with labelled precursors. Incorporation of radioactive D-[U-14C]-glucose into the diterpenes proved the de novo origin of verrucosins in the mollusc. Four new alpha-pyrone-containing propionates (5-8) and an unprecedented hydroperoxide 9 have been isolated from the mantle extract of Placida dendritica , a Mediterranean sacoglossan that lives upon the green alga Bryopsis plumosa . The new metabolites co-occur with the related compounds 1-4, which have been described in previous studies of the mollusc. The presence of 9 opens intriguing perspectives on the ecological role of placidenes. This paper reports the isolation and structural elucidation of the new compounds 5-9
