The global production of cereal straw as an agricultural by-product presents a significant source of biomass, which could be used as feedstock for the production of second generation biofuels by fermentation. The production of sugars for fermentation is an important measure of straw quality and in its suitability for biofuel production. In this paper, we present a characterization of straw digestibility from a wide range of cereal. Our main objective is to evaluate the variability of fermentable sugars released from different species including wheat (Triticum durum L., Triticum aestivum L.), barley (Hordeum vulgare L.) and triticale (X Triticosecale Wittmack). To this end, we adapted a saccharification method (IAS Method) capable of detecting significant differences of released sugars between cultivars and species, while using separately another method that would serve as a control and with which we could contrast our results (CNAP method). ANOVA analyses revealed that barley has a higher saccharification potential than wheat and triticale and shows more variation between genotypes. Thus, populations derived from crosses among them such as Steptoe × Morex and OWB Dominant × OWB Recessive hold potential for the identification of genetic basis for saccharification-related traits. The correlation of glucose released between the two methods was moderate (R2 = 0.57). An evaluation of the inter- and intra- specific correlation between a number of chemical and agronomical parameters and saccharification suggests that the cell wall thickness and lignin content in straw could be used in breeding programs for the improvement of the saccharification potential. Finally, the lack of correlation between grain yield and saccharification suggests that it would be possible to make a selection of genotypes for dual purpose, low recalcitrance and grain yield. Background: Miscanthus sinensis is a high yielding perennial grass species with great potential as a bioenergy feedstock. One of the challenges that currently impedes commercial cellulosic biofuel production is the technical difficulty to efficiently convert lignocellulosic biomass into biofuel. The development of feedstocks with better biomass quality will improve conversion efficiency and the sustainability of the value-chain. Progress in the genetic improvement of biomass quality may be substantially expedited by the development of genetic markers associated to quality traits, which can be used in a marker-assisted selection program. Results: To this end, a mapping population was developed by crossing two parents of contrasting cell wall composition. The performance of 182 F1 offspring individuals along with the parents was evaluated in a field trial with a randomized block design with three replicates. Plants were phenotyped for cell wall composition and conversion efficiency characters in the second and third growth season after establishment. A new SNP-based genetic map for M. sinensis was built using a genotyping-by-sequencing (GBS) approach, which resulted in 464 short-sequence uniparental markers that formed 16 linkage groups in the male map and 17 linkage groups in the female map. A total of 86 QTLs for a variety of biomass quality characteristics were identified, 20 of which were detected in both growth seasons. Twenty QTLs were directly associated to different conversion efficiency characters. Marker sequences were aligned to the sorghum reference genome to facilitate cross-species comparisons. Analyses revealed that for some traits previously identified QTLs in sorghum occurred in homologous regions on the same chromosome. Conclusion: In this work we report for the first time the genetic mapping of cell wall composition and bioconversion traits in the bioenergy crop miscanthus. These results are a first step towards the development of marker-assisted selection programs in miscanthus to improve biomass quality and facilitate its use as feedstock for biofuel production. The aim of producing sustainable liquid biofuels and chemicals from lignocellulosic biomass remains high on the sustainability agenda, but is challenged by the costs of producing fermentable sugars from these materials. Sugars from plant biomass can be fermented to alcohols or even alkanes, creating a liquid fuel in which carbon released on combustion is balanced by its photosynthetic capture. Large amounts of sugar are present in the woody, nonfood parts of crops and could be used for fuel production without compromising global food security. However, the sugar in woody biomass is locked up in the complex and recalcitrant lignocellulosic plant cell wall, making it difficult and expensive to extract. In this paper,wereview what is known about the major polymeric components of woody plant biomass, with an emphasis on the molecular interactions that contribute to its recalcitrance to enzymatic digestion. In addition, we review the extensive research that has been carried out in order to understand and reduce lignocellulose recalcitrance and enable more cost-effective production of fuel from woody plant biomass. Background: Miscanthus is a major bioenergy crop in Europe and a potential feedstock for second generation biofuels. The most efficient and realistic method to produce fermentable sugars from lignocellulosic biomass is by enzymatic hydrolysis, assisted by thermo-chemical pretreatment. Recently, microwave technology has drawn growing attention, because of its unique effects and performance on biomass. Result: In this work, microwave energy was applied to facilitate NaOH and H2SO4 pretreatment for Miscanthus under different temperatures (130-200 °C) for 20 min. The yields of reducing sugars from Miscanthus during the pretreatment process increased up to 180 °C and then declined with increasing temperature. Out results here showed a remarkable sugar yield from available carbohydrate (73 %) at the temperature of 180 °C by using 0.2 M H2SO4. In comparison with conventional heating pretreatment studied at same temperature with same biomass material, the reducing sugar release in this study was 17 times higher within half the time. It was highlighted that the major sugar component could be tuned by changing pretreatment temperature or pretreatment media. Optimally, the glucose and xylose yield from available carbohydrate are 47 and 22 % by using 0.2 M H2SO4 and NaOH respectively when temperature was 180 °C. The digestibility of pretreated Miscanthus was 10 times higher than that of untreated biomass. 68-86 % of the lignin content was removed from biomass by 0.2 M NaOH. Simultaneous saccharification fermentation (SSF) results showed an ethanol production of 143-152 mg/g biomass by using H2SO4/NaOH microwave assisted pretreatment, which is 7 times higher than that of untreated Miscanthus. Biomass morphology was studied by SEM, showing temperature has a strong influence on lignin removal process, as different lignin deposits were observed. At the temperature of 180 °C, NaOH pretreated biomass presented highly exposed fibres, which is a very important biomass characteristic for improved enzymatic hydrolysis. Conclusion: Compared to conventional pretreatment, microwave assisted pretreatment is more energy efficient and faster, due to its unique heating mechanism leading to direct interaction between the polar part of biomass and electromagnetic field. Background The search for promising and renewable sources of carbohydrates for the production of biofuels and other biorenewables has been stimulated by an increase in global energy demand in the face of growing concern over greenhouse gas emissions and fuel security. In particular, interest has focused on non-food lignocellulosic biomass as a potential source of abundant and sustainable feedstock for biorefineries. Here we investigate the potential of three Brazilian grasses (Panicum maximum, Pennisetum purpureum and Brachiaria brizantha), as well as bark residues from the harvesting of two commercial Eucalyptus clones (E. grandis and E. grandis x urophylla) for biofuel production, and compare these to sugarcane bagasse. The effects of hot water, acid, alkaline and sulfite pretreatments (at increasing temperatures) on the chemical composition, morphology and saccharification yields of these different biomass types were evaluated. Results The average yield (per hectare), availability and general composition of all five biomasses were compared. Compositional analyses indicate a high level of hemicellulose and lignin removal in all grass varieties (including sugarcane bagasse) after acid and alkaline pretreatment with increasing temperatures, whilst the biomasses pretreated with hot water or sulfite showed little variation from the control. For all biomasses, higher cellulose enrichment resulted from treatment with sodium hydroxide at 130°C. At 180°C, a decrease in cellulose content was observed, which is associated with high amorphous cellulose removal and 5- hydroxymethyl-furaldehyde production. Morphological analysis showed the effects of different pretreatments on the biomass surface, revealing a high production of microfibrillated cellulose or cellulose nanocrystals on grass surfaces, after treatment with 1% sodium hydroxide at 130°C for 30 minutes. This may explain the higher hydrolysis yields resulting from these pretreatments, since these cellulosic nanoparticles can be easily accessed and cleaved by cellulases. Conclusion Our results show the potential of three Brazilian grasses with high productivity yields as valuable sources of carbohydrates for ethanol production and other biomaterials. Sodium hydroxide at 130°C was found to be the most effective pretreatment for enhanced saccharification yields. It was also efficient in the production of microfibrillated cellulose on grass surfaces, thereby revealing their potential as a source of natural fillers used for bionanocomposites production. Background: In recent years, the growing demand for biofuels has encouraged the search for different sources of underutilized lignocellulosic feedstocks that are available in sufficient abundance to be used for sustainable biofuel production. Much attention has been focused on biomass fromgrass. However, large amounts of timber residues such as eucalyptus bark are available and represent a potential source for conversion to bioethanol. In the present paper, we investigate the effects of a delignification process with increasing sodium hydroxide concentrations, preceded or not by diluted acid, on the bark of two eucalyptus clones: Eucalyptus grandis (EG) and the hybrid, E. grandis x urophylla (HGU). The enzymatic digestibility and total cellulose conversion were measured, along with the effect on the composition of the solid and the liquor fractions. Barks were also assessed using Fourier-transform infrared spectroscopy (FTIR), solid-state nuclear magnetic resonance (NMR), X-Ray diffraction, and scanning electron microscopy (SEM). Results: Compositional analysis revealed an increase in the cellulose content, reaching around 81% and 76% of glucose for HGU and EG, respectively, using a two-step treatment with HCl 1%, followed by 4% NaOH. Lignin removal was 84% (HGU) and 79% (EG), while the hemicellulose removal was 95% and 97% for HGU and EG, respectively. However, when we applied a one-step treatment, with 4% NaOH, higher hydrolysis efficiencies were found after 48 h for both clones, reaching almost 100% for HGU and 80% for EG, in spite of the lower lignin and hemicellulose removal. Total cellulose conversion increased from 5% and 7% to around 65% for HGU and 59% for EG. NMR and FTIR provided important insight into the lignin and hemicellulose removal and SEM studies shed light on the cell-wall unstructuring after pretreatment and lignin migration and precipitation on the fibers surface, which explain the different hydrolysis rates found for the clones. Conclusion: Our results show that the single step alkaline pretreatment improves the enzymatic digestibility of Eucalyptus bark. Furthermore, the chemical and physical methods combined in this study provide a better comprehension of the pretreatment effects on cell-wall and the factors that influence enzymatic digestibility of this forest residue. The digestion of lignocellulose is attracting attention both in terms of basic research into its metabolism by microorganisms and animals, and also as a means of converting plant biomass into biofuels. Limnoriid wood borers are unusual because, unlike other wood-feeding animals, they do not rely on symbiotic microbes to help digest lignocellulose. The absence of microbes in the digestive tract suggests that limnoriid wood borers produce all the enzymes necessary for lignocellulose digestion themselves. In this study we report that analysis of ESTs from the digestive system of Limnoria quadripunctata reveals a transcriptome dominated by glycosyl hydrolase genes. Indeed, >20% of all EST5 represent genes encoding putative cellulases. including glycosyl hydrolase family 7 (GH7) cellobiohydrolases. These have not previously been reported in animal genomes. but are key digestive enzymes produced by wood-degrading fungi and symbiotic protists in termite guts. We propose that limnoriid GH7 genes are important for the efficient digestion of lignocellulose in the absence of gut microbes. Hemocyanin transcripts were highly abundant in the hepatopancreas transcriptome. Based on recent studies indicating that these proteins may function as phenoloxidases in isopods. we discuss a possible role for hemocyanins in lignin decomposition. [ABSTRACT FROM AUTHOR]

Background: Cell wall resistance represents the main barrier for the production of second generation biofuels. The deconstruction of lignocellulose can provide sugars for the production of fuels or other industrial products through fermentation. Understanding the biochemical basis of the recalcitrance of cell walls to digestion will allow development of more effective and cost efficient ways to produce sugars from biomass. One approach is to identify plant genes that play a role in biomass recalcitrance, using association genetics. Such an approach requires a robust and reliable high throughput (HT) assay for biomass digestibility, which can be used to screen the large numbers of samples involved in such studies. Results: We developed a HT saccharification assay based on a robotic platform that can carry out in a 96-well plate format the enzymatic digestion and quantification of the released sugars. The handling of the biomass powder for weighing and formatting into 96 wells is performed by a robotic station, where the plant material is ground, delivered to the desired well in the plates and weighed with a precision of 0.1 mg. Once the plates are loaded, an automated liquid handling platform delivers an optional mild pretreatment (< 100°C) followed by enzymatic hydrolysis of the biomass. Aliquots from the hydrolysis are then analyzed for the release of reducing sugar equivalents. The same platform can be used for the comparative evaluation of different enzymes and enzyme cocktails. The sensitivity and reliability of the platform was evaluated by measuring the saccharification of stems from lignin modified tobacco plants, and the results of automated and manual analyses compared. Conclusions: The automated assay systems are sensitive, robust and reliable. The system can reliably detect differences in the saccharification of plant tissues, and is able to process large number of samples with a minimum amount of human intervention. The automated system uncovered significant increases in the digestibility of certain lignin modified lines in a manner compatible with known effects of lignin modification on cell wall properties. We conclude that this automated assay platform is of sufficient sensitivity and reliability to undertake the screening of the large populations of plants necessary for mutant identification and genetic association studies. Sugarcane bagasse represents one of the best potential feedstocks for the production of second generation bioethanol. The most efficient method to produce fermentable sugars is by enzymatic hydrolysis, assisted by thermochemical pretreatments. Previous research was focused on conventional heating pretreatment and the pretreated biomass residue characteristics. In this work, microwave energy is applied to facilitate sodium hydroxide (NaOH) and sulphuric acid (H 2 SO 4 ) pretreatments on sugarcane bagasse and the efficiency of sugar production was evaluated on the soluble sugars released during pretreatment. The results show that microwave assisted pretreatment was more efficient than conventional heating pretreatment and it gave rise to 4 times higher reducing sugar release by using 5.7 times less pretreatment time. It is highlighted that enrichment of xylose and glucose can be tuned by changing pretreatment media (NaOH/H 2 SO 4 ) and holding time. SEM study shows significant delignification effect of NaOH pretreatment, suggesting a possible improved enzymatic hydrolysis process. However, severe acid conditions should be avoided (long holding time or high acid concentration) under microwave heating conditions. It led to biomass carbonization, reducing sugar production and forming ‘humins’. Overall, in comparison with conventional pretreatment, microwave assisted pretreatment removed significant amount of hemicellulose and lignin and led to high amount of sugar production during pretreatment process, suggesting microwave heating pretreatment is an effective and efficient pretreatment method. Abstract: The potential of biofuels contributing to the UK emission reduction targets in the formulated UK Low Carbon Transition Plan (LCTP) and the UK’s obligation in the wider EU emissions reduction targets are assessed using four scenarios. The scenarios were evaluated using hybrid lifecycle assessment developed in a multi-regional input–output (MRIO) framework. In the hybrid MRIO LCA framework, technology-specific processes in the biofuels and fossil fuels LCA systems are integrated into a generalised 2-region (UK and Rest of the World) environmental-economic input–output framework in order to account for economy-wide indirect GHG emissions in the biofuels and fossil fuels LCA systems in addition to other indirect impacts such as indirect land use change. The lifecycle greenhouse gas emissions of biodiesel (soybean, palm, rape, waste cooking oil) and bio-ethanol (sugarcane, sugarbeet, corn) were assessed and compared to fossil fuel (diesel and petrol) baseline. From one of the scenarios, biodiesel production from waste cooking oil and bioethanol from sugarbeet offer the biggest potential for emissions savings relative to fossil fuel equivalent and offering a maximum emission savings of 4.1% observed with a biofuel market share of 10% reached in 2020. It was also established that under current biofuel feedstock mix, to achieve the 6% emissions saving primarily from biofuels as proposed in the LCTP, 23.8% of the transport fuels market would be required to be held by biofuels by 2020.
