Ectopic mineralization refers to the deposition of mineralized complexes in the extracellular matrix of soft tissues. Calcific aortic valve disease, vascular calcification, gallstones, kidney stones, and abnormal mineralization in arthritis are common examples of ectopic mineralization. They are debilitating diseases and exhibit excess mortality, disability, and morbidity, which impose on patients with limited social or financial resources. Recent recognition that inflammation plays an important role in ectopic mineralization has attracted the attention of scientists from different research fields. In the present review, we summarize the origin of inflammation in ectopic mineralization and different channels whereby inflammation drives the initiation and progression of ectopic mineralization. The current knowledge of inflammatory milieu in pathological mineralization is reviewed, including how immune cells, pro-inflammatory mediators, and osteogenic signaling pathways induce the osteogenic transition of connective tissue cells, providing nucleating sites and assembly of aberrant minerals. Advances in the understanding of the underlying mechanisms involved in inflammatory-mediated ectopic mineralization enable novel strategies to be developed that may lead to the resolution of these enervating conditions. Matrix vesicles (MVs) are extracellular membrane-bound vesicles of about ~ 50–200 nm in diameter that play a role in the bio-mineralization process of hard tissue formation. The present review is based on the empirical phenomenon of primary mineralization process via matrix vesicle-mediated mechanism with special reference to crystal ghosts as well as the mechanism on the organic–inorganic relationship between matrix vesicles and crystal ghosts, and the transformation that these structures undergo during bio-mineralization. Lower Cambrian assemblages of phosphatic and chitinous tubular fossils of probable cnidarian affinities were recovered from the Yanjiahe, Shuijingtuo and Shipai formations in the Three Gorges area, South China. Terreneuvian Cambrorhytium cf. C . major co-occur with an unnamed taxon in the Yanjiahe Formation. They can be compared with specimens from the Ediacaran Miaohe Fauna in the Three Gorges area. The most abundant and diversified cnidarian assemblages, including Cambrorhytium gracilis n. sp., Cambrorhytium cf. major , Cambrorhytium minor , Sphenothallus songlinensis , Sphenothallus taijiangensis ?, Sphenothallus kozaki , ? Sphenothallus sp., ? Byronia sp., and ? Mackenzia sp. are reported from the overlying, Cambrian Stage 3–4, Shuijingtuo and Shipai formations. These new fossils show previously unknown evidence for cnidarian affinities, such as an attachment disk, clonal clusters and probable budding structures. These diversified assemblages of probable cnidarian fossils further fill major gaps in the Cambrian record of stem metazoans and document associated early evolutionary patterns. In addition, our discovery suggests that along with sponges, these tubular fossils represent another important component of benthic ecosystems that colonized often dysoxic, deep-settings during the Cambrian explosion. The observed increases of average body-size and biomineralization among these probable cnidarians are tentatively linked to oxygenation of deep oceanic settings and appearance of predatory pressure during the Ediacaran–Cambrian transition, respectively. Purpose/Aim: The most common kidney stone composed of calcium oxalate forms on interstitial calcium phosphate mineral known as a Randall’s plaque (RP). Due to limited information about events leading to the initial deposition of nanometer size interstitial calcium phosphate pre-clusters, there continues to be a debate on the initial site of calcium phosphate deposition and factors leading to stone formation. Materials and Methods: High-resolution X-ray micro-computed tomography (CT), and light and electron microscopy techniques were used to characterize human renal pyramids and five representative kidney stones with identifiable stems. Mineral densities of mineralized aggregates within these specimens were correlated with micro- and ultra-structures as seen using light and electron microscopy techniques. Results: The earliest detectable biominerals in the human renal papilla were proximal intratubular plate-like calcium phosphate deposits. Unoccluded tubules in stems connected to calcium phosphate stones were observed by electron microscope and X-ray micro-CT. These tubules were similar in diameter (30-100 μm) and shape to those observed in the distal regions of the renal papilla. Conclusions: Observations were patterned through a novel and unified theory of stepwise-architecture guided biomineralization (a combination of smaller structures leading to a larger but similar structural framework). A plausible stepwise progression in renal biomineralization is proposed; proximal intratubular calcium phosphate deposits can lead to interstitial yet calcium phosphate rich RP and mature into a stem on which a calcium oxalate stone grows within the collecting system of a kidney. Bone and dentin development requires temporal and spatial deposition of calcium phosphate mineral. A host of proteins works in concert to contribute to this tightly regulated process while malfunction in this scheme often leads to pathological defects. We have reported earlier that DMP1 stimulation of preosteoblasts leads to calcium release from internal Ca2+ stores and this store depletion is sensed by the ER Ca2+ sensor STIM1 (stromal interaction molecule 1). In this study, we first assessed the temporal and spatial localization of STIM1 protein during the development of bone and dentin by immunohistochemical methods. We further analyzed the function of STIM1 by establishing a stable MC3T3-E1 cell-line by overexpressing STIM1 (MC3T3-E1/STIM1 OE). Under mineralizing conditions, STIM1 overexpressing cells showed increased calcium deposits with higher expression of key osteogenic markers, such as Runx2 and type I collagen, BMP4 when compared with the control cells. Our results demonstrate that during mineralized matrix formation STIM1, the key ER sensor protein, can promote cellular differentiation in the presence of extracellular calcium. Aims The aim of this work is to analyse the effect of pH, fungal identity and P chemical nature on microbial development and phosphatase release, discussing solubilization and mineralization processes in P cycling. Methods and Results P solubilizing fungi ( Talaromyces flavus, T. helicus L, T. helicus N, T. diversus and Penicillium purpurogenum) were grown under three pH conditions (6, 6·5 and 8·5) and with different inorganic (calcium, iron, aluminium and rock) and organic (lecithin and phytate) P sources. P solubilization, mineralization, growth and phosphatase production were recorded. Acid and neutral environments maximized fungal development and P recycling. P chemical nature changed the phosphatases release pattern depending on the fungal identity. Acid phosphatase activity was higher than alkaline phosphatases, regardless of pH or sample times. Alkaline phosphatases were affected by a combination of those factors. Conclusions P chemical nature and pH modify fungal growth, P mineralization and solubilization processes. The underlying fungal identity-dependent metabolism governs the capacity and efficiency of P solubilization and mineralization. P solubilization and mineralization processes are interrelated and simultaneously present in soil fungi. Significance and Impact of the study This study constitutes a reference work to improve the selection of fungal bioinoculants in different environmental conditions, highlighting their role in P cycling. Biomineralization is a complicated process which involves multiple gene products inPinctada fucata. Here,Pinctada fucatamantle gene 2 (pfmg2) was characterized inP. fucata. The putative PFMG2 protein has a Calponin Homology (CH) domain and an EF hand that can bind calcium. PFMG2 shares homology with transgelin in other species. Multi-tissue reverse transcriptase polymerase chain reaction (RT-PCR) indicates thatpfmg2mRNA is expressed ubiquitously, but its expression is high in the mantle tissue ofP. fucata. Furthermore,in situhybridization shows thatpfmg2localized at the edge of the mantle. Our results suggest that PFMG2 plays an important role in the shell mineralization process in pearl oyster. Negatively charged macromolecules play a starring role in Ca 2 + saturation at interfaces: they act as organized points for biomineral growth in nature. However, the role positively charged interfaces play in biomineralization remains unknown. Therefore, we decided to use a cationic Langmuir-Blodgett (LB) film composed of dioctadecyldimethylammonium bromide (DOMA) and type-I collagen in the presence of CaCl 2 solutions to guide CaCO 3 growth on titanium surfaces. Ca 2 + was able to interact with DOMA monolayers, leading to impressive cation excess at the film surface, as evidenced by the increased positive zeta potential of the LB films. Additionally, molecular dynamics simulations showed that Ca 2 + co-ions were able to accumulate on the positively charged LB interface, especially because configurations like R-N(+)-counterion-Ca 2 + existed therein. Counterion-mediated Ca 2 + accumulation on the surface was mainly driven by Cl − ions, which were much closer to Ca 2 + ions than Br − ions. Ca 2 + excess on the surface was sufficient to grow continuous and particulate CaCO 3 films on titanium surfaces. In addition, collagen presence induced changes in CaCO 3 film morphology and organization. Finally, we were able to confirm that positively charged organic interfaces can act as templates for biomineral growth because counterion-mediated Ca 2 + accumulation on these surfaces can drive subsequent crystal growth. The synthesis of crystals through biomineralization is a process of protection and support preserved in animals, protists, moneras, plants and fungi. The genome of every species has evolved to preserve and/or modify the formation of one or another type of crystal, which may be of the organic or inorganic type. The most common inorganic crystals identified in organisms include calcium carbonate (CaCO 3 ), calcium phosphate (CaP), calcium oxalate (CaOx), magnetite or greigite, and sulfides of cadmium (CdS), mercury (HgS) and lead (PbS). Organic crystals are of the protein or ice type. The formation of both types of crystals requires biomolecules such as proteins. This paper reviews the proteins involved in the synthesis of different crystals in distinct biological systems, in order to understand how each organism has adapted its genome to preserve essential mechanisms such as biomineralization, which has enabled them to survive in a changing environment for millions of years. Hydroxyapatite (HAp) is an excellent biomaterial for bone repair and regeneration. The biological functions of HAp particles, such as biomineralization, cell adhesion, and cell proliferation, can be enhanced when their size is reduced to the nanoscale. In this work, HAp nanoparticles were synthesized by the hydrothermal technique with addition of cetyltrimethylammonium bromide (CTAB). These particles were also characterized, and their size controlled by modifying the CTAB concentration and hydrothermal duration. The results show that most HAp nanoparticles were rod-like in shape, exhibiting the most uniform and smallest size (mean diameter and length of 39 nm and 125 nm, respectively) at optimal conditions of 0.64 g CTAB and hydrothermal duration of 12 h. Moreover, good biomineralization capability of the HAp nanorods was confirmed through in␣vitro tests in simulated body fluid. A bone-like mineral layer of synthesized HAp nanorods formed rapidly after 7 days. This study shows that highly bioactive HAp nanorods can be easily prepared by the hydrothermal method, being a potential nanomaterial for bone regeneration. [ABSTRACT FROM AUTHOR]

Moonmilk is a karstic speleothem mainly composed of fine calcium carbonate crystals (CaCO3) with different textures ranging from pasty to hard, in which the contribution of biotic rock-building processes is presumed to involve indigenous microorganisms. The realmicrobial input in the genesis of moonmilk is difficult to assess leading to controversial hypotheses explaining the origins and the mechanisms (biotic vs. abiotic) involved. In this work, we undertook a comprehensive approach in order to assess the potential role of filamentous bacteria, particularly a collection of moonmilk-originating Streptomyces, in the genesis of this speleothem. Scanning electron microscopy (SEM) confirmed that indigenous filamentous bacteria could indeed participate in moonmilk development by serving as nucleation sites for CaCO3 deposition. The metabolic activities involved in CaCO3 transformation were furthermore assessed in vitro among the collection of moonmilk Streptomyces, which revealed that peptides/amino acids ammonification, and to a lesser extend ureolysis, could be privileged metabolic pathways participating in carbonate precipitation by increasing the pH of the bacterial environment. Additionally, in silico search for the genes involved in biomineralization processes including ureolysis, dissimilatory nitrate reduction to ammonia, active calcium ion transport, and reversible hydration of CO2 allowed to identify genetic predispositions for carbonate precipitation in Streptomyces. Finally, their biomineralization abilities were confirmed by environmental SEM, which allowed to visualize the formation of abundant mineral deposits under laboratory conditions. Overall, our study provides novel evidences that filamentous Actinobacteria could be key protagonists in the genesis of moonmilk through a wide spectrum of biomineralization processes. In recent years, scientists have developed various biomaterials to remineralize human teeth to treat dentine hypersensitivity. Poly(amido amine) (PAMAM) dendrimers have become a research focus in this field. It has been demonstrated that PAMAM is able to create precipitates both on the surface of and within the dentinal tubules, however, there is little information about its effect on reducing dentine permeability in vitro. This study aimed to evaluate the in vitro effectiveness and stability of the fourth generation amine-terminated PAMAM on dentinal tubule occlusion, especially on dentine permeability. Sodium fluoride (NaF), which has been widely used as a desensitizing agent, is regarded as positive control. Demineralized sensitive dentine samples were coated with PAMAM or sodium fluoride solutions and soaked in artificial saliva (AS) at 37 °C for different periods. Four weeks later, samples in each group were then equally split into two subgroups for testing using a brushing challenge and an acid challenge. Dentine permeability of each specimen was measured before and after each challenge using a fluid filtration system. Dentine morphology and surface deposits were characterized by scanning electron microscope (SEM) and analyzed with Image-Pro Plus software. Data were evaluated through multifactorial ANOVA with repeated measures and pair-wise comparisons at a level of 5%. The results showed that PAMAM and NaF significantly reduced dentine permeability to 25.1% and 20.7%. Both of them created precipitates on dentine surfaces after AS immersion for 28 days. PAMAM-induced biomineralization not only on dentine surfaces, but also deeper in dentinal tubules, significantly reduced dentine permeability. Moreover, PAMAM-induced biomineralization elicited excellent stable occlusion effects after acid challenge. X-ray structures of homopolymeric L-ferritin obtained by freezing protein crystals at increasing exposure times to a ferrous solution showed the progressive formation of a triiron cluster on the inner cage surface of each subunit. After 60 min exposure, a fully assembled (μ3-oxo)Tris[(μ2-peroxo)(μ2-glutamato-κO:κO')](glutamato- κO)(diaquo)triiron(III) anionic cluster appears in human L-ferritin. Glu60, Glu61, and Glu64 provide the anchoring of the cluster to the protein cage. Glu57 shuttles incoming iron ions toward the cluster.We observed a similar metallocluster in horse spleen L-ferritin, indicating that it represents a common feature of mammalian L-ferritins. The structures suggest a mechanism for iron mineral formation at the protein interface. The functional significance of the observed patch of carboxylate side chains and resulting metallocluster for biomineralization emerges from the lower iron oxidation rate measured in the E60AE61AE64A variant of human L-ferritin, leading to the proposal that the observed metallocluster corresponds to the suggested, but yet unobserved, nucleation site of L-ferritin. Study Design: A study examining the clinical protocol of scoliosis wound irrigation, demonstrating povidone-iodine's (PVI) effect on human osteoblast cells. Primary and immortal cell line osteoblasts were treated with 0.35% PVI for 3 minutes, and analyzed for proliferation rate, oxidative capacity, and mineralization.Objective: To model spinal wound irrigation with dilute PVI in vitro, in order to investigate the effect of PVI on osteoblast proliferation, metabolism, and bone mineralization.Summary Of Background Data: Previously PVI irrigation has been proposed as a safe and effective practice to avoid bacterial growth after spinal surgery. However, recent evidence in multiple cell types suggests that PVI has a deleterious effect on cellular viability and cellular function.Methods: Primary and immortal human osteoblast cells were exposed to either phosphate buffered saline control or with 0.35% PVI for 3 minutes. Cellular proliferation was measured over the duration of 7 days by MTS assay. Oxygen consumption rate, extracellular acidification rate, and proton production rate were analyzed using a Seahorse XF24 Bioanalyzer. Protein expression of the electron transport chain subunits CII-SDHB, CIII-UQRCR2, and CV-ATP5A was measured via Western blotting. Mineralized bone nodules were stained with alizarin red.Results: Expressed as a percentage of normal osteoblast proliferation, osteoblasts exposed to 0.35% PVI exhibited a significant 24% decrease in proliferation after 24 hours. This was a sustained response, resulting in a 72% decline in cellular proliferation at 1 week. There was a significant reduction in oxygen consumption rate, extracellular acidification rate, and proton production rate (P < 0.05), in osteoblasts that had been exposed to 0.35% PVI for 3 minutes, coupled with a marked reduction in the protein expression of CII-SDHB. Osteoblasts exposed to 0.35% PVI exhibited reduced bone nodule mineralization compared to control phosphate buffered saline exposed osteoblasts (P < 0.01).Conclusion: PVI has a rapid and detrimental effect on human osteoblast cellular proliferation, metabolic function, and bone nodule mineralization.Level Of Evidence: NA. The emergence of metal-organic frameworks (MOFs) as potential candidates to supplant existing adsorbent types in real-world applications has led to an explosive growth in the number of compounds available to researchers, as well as in the diversity of the metal salts and organic linkers from which they are derived. In this context, the use of carbonate-based precursors as metal sources is of interest due to their abundance in mineral deposits and their reaction chemistry with acids, resulting in just water and carbon dioxide as side products. Here, we have explored the use of calcium carbonate as a metal source and demonstrate its versatility as a precursor to several known frameworks, as well as a new flexible compound based on the 2,5-dihydroxybenzoquinone (H2dhbq) linker, Ca(dhbq)(H2O)2. Furthermore, inspired by the ubiquity and unique structures of biomineralized forms of calcium carbonate, we also present examples of the preparation of superstructures of Ca-based MOFs via the coordination replication technique. In all, the results confirm the suitability of carbonate-based metal sources for the preparation of MOFs and further expand upon the growing scope of coordination replication as a convenient strategy for the preparation of structuralized materials. The structural organization of the palliostracum--the dominant part of the shell which is formed by the mantle cells--of Glycymeris glycymeris (Linné 1758) is comprised of five hierarchical levels with pronounced structural commonalities and deviations from other crossed-lamellar shells. The hierarchical level known as second order lamellae, present within other crossedlamellar shells, is absent highlighting a short-coming of the currently used nomenclature. On the mesoscale, secondary microtubules penetrate the palliostracum and serve as crack arrestors. Moreover, the growth lamellae follow bent trajectories possibly impacting crack propagation, crack deflection, and energy dissipation mechanisms whilst circumventing delamination. Finally, at least two structural elements are related to external circatidal and circaanular stimuli. This emphasizes that endogeneous rhythms may contribute and (co-)control the self-organization of a complex mineralized tissue and that it is insufficient to rely fully on a reductionistic approach when studying biomineralization. We present in this paper accurate and reliable Raman and IR spectral identification of mineral constituents in nine samples of renal calculi (kidney stones) removed from patients suffering from nephrolithiasis. The identified mineral components include Calcium Oxalate Monohydrate (COM, whewellite), Calcium Oxalate Dihydrate (COD, weddellite), Magnesium Ammonium Phosphate Hexahydrate (MAPH, struvite), Calcium Hydrogen Phosphate Dihydrate (CHPD, brushite), Pentacalcium Hydroxy Triphosphate (PCHT, hydroxyapatite) and Uric Acid (UA). The identification is based on a satisfactory assignment of all the observed IR and Raman bands (3500–400 cm − 1 ) to chemical functional groups of mineral components in the samples, aided by spectral analysis of pure materials of COM, MAPH, CHPD and UA. It is found that the eight samples are composed of COM as the common component, the other mineral species as common components are: MAPH in five samples, PCHT in three samples, COD in three samples, UA in three samples and CHPD in two samples. One sample is wholly composed of UA as a single component; this inference is supported by the good agreement between ab initio density functional theoretical spectra and experimental spectral measurements of both sample and pure material. A combined application of Raman and IR techniques has shown that, where the IR is ambiguous, the Raman analysis can differentiate COD from COM and PCHT from MAPH. [ABSTRACT FROM AUTHOR]

Microbially induced calcium carbonate precipitation (MICP) is emerging as a sustainable technology for improved construction materials. In this technique, calcium carbonate is deposited in the pores of substrates such as sand and concrete. As the technology moves from laboratory to field new metrics and field measurement techniques are required to ensure that the results achieved at the laboratory are replicated. In the scaled up technique it is not possible to collect samples from deep inside the substrate for direct measurement. This paper explores some indirect indicators of MICP that can be easily monitored in large scale applications. Bacterial fluid has been passed through sand columns of varying grain sizes. The rate of flow is monitored for ten days. The effluent has been tested for pH and electrical conductivity to evaluate their potential for ensuring MICP. The residual urea and calcium in the effluent has been monitored to have a quantitative estimate of MICP. At the end of the tests the quantity of carbonate deposited in the sand columns is measured. A correlation between the residual reactants and the carbonate deposition is developed. An approximate expression for different grain sizes has been developed. This study demonstrates the utility of indirect measurement techniques as flow rate, pH, conductivity, urea and calcium consumption as potential indicators of efficient MICP in sand media. Sea urchin embryos sequester calcium from the sea water. This calcium is deposited in a concentrated form in granule bearing vesicles both in the epithelium and in mesenchymal cells. Here we use in vivo calcein labeling and confocal Raman spectroscopy, as well as cryo-FIB-SEM 3D structural reconstructions, to investigate the processes occurring in the internal cavity of the embryo, the blastocoel. We demonstrate that calcein stained granules are also present in the filopodial network within the blastocoel. Simultaneous fluorescence imaging and Raman spectroscopy show that these granules do contain a calcium mineral. By tracking the movements of these granules, we show that the granules in the epithelium and primary mesenchymal cells barely move, but those in the filopodial network move long distances. We could however not detect any unidirectional movement of the filopodial granules. We also show the presence of mineral containing multivesicular vesicles that also move in the filopodial network. We conclude that the filopodial network is an integral part of the mineral transport process, and possibly also for sequestering calcium and other ions. Although much of the sequestered calcium is deposited in the mineralized skeleton, a significant amount is used for other purposes, and this may be temporarily stored in these membrane-delineated intracellular deposits. Sharks, rays and other elasmobranch fishes are characterized by a skeletal type that is unique among living vertebrates, comprised predominantly of an unmineralized cartilage, covered by a thin outer layer of sub-millimeter, mineralized tiles called tesserae. The mineralized portion of the skeleton appears to grow only by apposition, adding material at the edges of each tessera; maintenance of non-mineralized joints between tesserae is therefore vital, with precise control of mineral deposition and inhibition at the many thousands of growth fronts in the skeleton. Yet, we have only scattered evidence as to how the elasmobranchs mineralize and grow their skeletons. In this review, we take an “environment to skeleton” approach, drawing together research from a vast range of perspectives to track calcium and phosphate from the typical elasmobranch habitats into and through the body, to their deposition at tesseral growth fronts. In the process, we discuss the available evidence for skeletal resorption capability, mineral homeostasis hormones, and nucleation inhibition mechanisms. We also outline relevant theories in crystal nucleation and typical errors in measurements of serum calcium and phosphate in the study of vertebrate biology. We assemble research that suggests consensus in some concepts in elasmobranch skeletal development, but also highlight the very large gaps in our knowledge, particularly in regards to endocrine functional networks and biomineralization mechanisms. In this way, we lay out frameworks for future directions in the study of elasmobranch skeletal biology with stronger and more comparative links to research in other disciplines and into other taxa. Osteocare, a herbal formulation, has been found to be very effective in bone mineralization and support of the microstructure of bone tissue. The water-soluble components of Osteocare (WSCO) induced osteogenic activity in human osteoblast-like SaOS-2 cells. The addition of WSCO (100 μg/ml) to SaOS-2 cells was effective in increasing the cell proliferation by 41.49% and DNA content by 1.9-fold. WSCO increased matrix mineralization in SaOS-2 cells by increased alkaline phosphatase levels and calcium-rich deposits as observed by Alizarin red staining. WSCO markedly increased mRNA expression for osteopontin (OPN), osteocalcin (OCN), type I collagen (Col I) in SaOS-2 cells, and it down-regulated IL-6 mRNA levels in SaOS-2 cells. The present study showed that WSCO plays an important role in osteoblastic bone formation through enhanced activities of ALP, Col I, bone matrix proteins such as OPN and OCN, down-regulation of cytokines like IL-6, as well as promoting mineralization in SaOS-2 cells. Abstract: This work aimed to better understand the ozonation process of a typical antibiotic pharmaceutical, trimethoprim in aqueous solution. The parent compound was almost completely degraded with ozone dose up to 3.5 mg/L with no mineralization. Twenty one degradation products were identified using an electrospray quadrupole time-of-flight mass spectrometer. Several ozonation pathways were proposed including hydroxylation, demethylation, carbonylation, deamination and methylene group cleavage. Two species of luminescent bacteria Photobacterium phosphoreum and Vibrio qinghaiensis were selected to assess the toxicity of ozonation products. For P. phosphoreum, higher level of toxicity was observed compared to the parent compound, but a negligible toxicity change was observed for V. qinghaiensis, indicating different modes of action for the same water sample. This was further confirmed by quantitative structure–active relationship analysis. This work proves the dominant role of ozone rather than hydroxyl radicals in the reaction and the potential risk after ozonation. [Copyright &y& Elsevier]

Abstract: Ectopic mineralization of soft tissues is known to be a typical response to systemic imbalance of various metabolic factors as well as tissue injury, leading to severe clinical consequences. In this study, coxsackievirus B3 (CVB3) infection in mice resulted in significant tissue injury, especially in the heart and pancreas. Inflammatory damage and apoptotic cell death were observed in CVB3-infected heart and pancreas tissues. Along with tissue damage, substantial ectopic calcification was detected in CVB3-infected heart, pancreas, and lung tissues, as determined by von Kossa staining and calcium content quantification. In addition, CVB3 infection induced upregulation of osteogenic signals, including six genes (BMP2, SPARC, Runx2, osteopontin, collagen type I, and osterix) in the heart, three genes (SPARC, osteopontin, and collagen type I) in the pancreas, and two genes (BMP2 and alkaline phosphatase) in the lung, as determined by quantitative real-time PCR analysis. Intriguingly, we showed that α-lipoic acid diminished CVB3-mediated inflammatory and apoptotic tissue damage, subsequently ameliorating ectopic calcification via the suppression of osteogenic signals. Collectively, our data provide evidence that ectopic calcification induced by CVB3 infection is implicated in the induction of osteogenic propensity, and α-lipoic acid may be a potential therapeutic agent to ameliorate pathologic calcification. Application of a sequential anaerobic and aerobic process in a constructed wetland can enhance the treatment performance of textile wastewater. In this study, two laboratory-scale recirculated up-flow constructed wetland (UFCW) reactors planted with Phragmites australis were constructed to investigate the treatment performance between aerated and non-aerated reactors for treating 50mg/L Reactive Black 5 (RB5)-containing wastewater. Results showed that the non-aerated reactor demonstrated an almost entire anaerobic environment yielded a better RB5 removal efficiency (89%) compared to the aerated reactor (81%). However, the aerated reactor performed higher removal on chemical oxygen demand (COD) and aromatic amines than the non-aerated reactor. Proper design ratio of anaerobic/aerobic region in UFCW is a key to accomplish not only color reduction but also COD and aromatic amines in textile wastewater treatment. [ABSTRACT FROM AUTHOR]

Background: Calabash chalk, a popularly consumed geophagic material in Nigeria has been reported to contain lead, arsenic, alpha lindane, endrin, and endosulfan 11 among other pollutants. Aim: The continuous exposure of young children to this chalk necessitated this study on the bone morphometry and mineralization in young Wistar rats. Materials and Methods: Fourteen young (weanling) Wistar rats of both sexes weighing 54-72 g were assigned into two groups of seven animals each. Group I served as control, while group II was the test group (TG). 40 mg/ml of C. chalk was administered as suspension to the test animals in group II. Animals in the control group were orally treated with 1ml of distilled water . Administration of the C. chalk in the animals lasted for 28 days, and the animals were sacrificed on day 29, using chloroform anaesthesia. The femur bones were dissected out, cleaned of flesh and sun-dried. The lengths and weights of the femur bones were measured using graphite furnace atomic mass spectrophotometer . Results: Results showed 1.6% decrease in body weight change in the TG, insignificant decreases in the weights and lengths of both the right and left femur bones, and significant decreased (P < 0.0126) organ-somatic index, and femur bones concentrations (mg/l) of zinc, phosphate, carbonate, calcium, sodium, and potassium (P < 0.05). Conclusion: In conclusion, this study showed that C. chalk may alter growth rate, and cause de-mineralization in the femur bone, hence, it may be detrimental to bone growth. [ABSTRACT FROM AUTHOR]

Calcanei of mature mule deer have the largest mineral content (percent ash) difference between their dorsal 'compression' and plantar 'tension' cortices of any bone that has been studied. The opposing trabecular tracts, which are contiguous with the cortices, might also show important mineral content differences and microscopic mineralization heterogeneity (reflecting increased hemi-osteonal renewal) that optimize mechanical behaviors in tension vs. compression. Support for these hypotheses could reveal a largely unrecognized capacity for phenotypic plasticity - the adaptability of trabecular bone material as a means for differentially enhancing mechanical properties for local strain environments produced by habitual bending. Fifteen skeletally mature and 15 immature deer calcanei were cut transversely into two segments (40% and 50% shaft length), and cores were removed to determine mineral (ash) content from 'tension' and 'compression' trabecular tracts and their adjacent cortices. Seven bones/group were analyzed for differences between tracts in: first, microscopic trabecular bone packets and mineralization heterogeneity (backscattered electron imaging, BSE); and second, trabecular architecture (micro-computed tomography). Among the eight architectural characteristics evaluated [including bone volume fraction (BVF) and structural model index (SMI)]: first, only the 'tension' tract of immature bones showed significantly greater BVF and more negative SMI (i.e. increased honeycomb morphology) than the 'compression' tract of immature bones; and second, the 'compression' tracts of both groups showed significantly greater structural order/alignment than the corresponding 'tension' tracts. Although mineralization heterogeneity differed between the tracts in only the immature group, in both groups the mineral content derived from BSE images was significantly greater ( P < 0.01), and bulk mineral (ash) content tended to be greater in the 'compression' tracts (immature 3.6%, P = 0.03; mature 3.1%, P = 0.09). These differences are much less than the approximately 8% greater mineral content of their 'compression' cortices ( P < 0.001). Published data, suggesting that these small mineralization differences are not mechanically important in the context of conventional tests, support the probability that architectural modifications primarily adapt the tracts for local demands. However, greater hemi-osteonal packets in the tension trabecular tract of only the mature bones ( P = 0.006) might have an important role, and possible synergism with mineralization and/or microarchitecture, in differential toughening at the trabeculum level for tension vs. compression strains. Calcium phosphates have important role in biological and pathological mineralization. While only one of calcium phosphates, carbonate apatite, represents the main mineral component of teeth and bones, octacalcium phosphate, calcium hydrogenphosphate dihydrate and beta-tricalcium phosphate occur in pathological deposits. From the standpoint of chemists, processes of biological and pathological mineralization could be considered as deposition of inorganic phase within organic matrix, i.e. formation of inorganic-organic composites. Although this approach is very simplified at first glance, it allows clarification of important issues related to biomineralization (e.g. what is the role of individual components of organic matrix in the emerging solid tissue), and design and preparation of new materials for hard tissue regeneration (e.g. process of transformation after implantation). The importance of investigation about calcium phosphates will be presented through the overview of basic physico-chemical reactions related to the formation and transformation of biologically relevant calcium phosphates and their interaction with various organic additives in the laboratory. The Transforming growth factor b (Tgf-β) pathway, by signaling via the activation of Smad transcription factors, induces the expression of many diverse downstream target genes thereby regulating a vast array of cellular events essential for proper development and homeostasis. In order for a specific cell type to properly interpret the Tgf-b signal and elicit a specific cellular response, cell-specific transcriptional co-factors often cooperate with the Smads to activate a discrete set of genes in the appropriate temporal and spatial manner. Here, via a conditional knockout approach, we show that mice mutant for Forkhead Box O transcription factor FoxO1 exhibit an enamel hypomaturation defect which phenocopies that of the Smad3 mutant mice. Furthermore, we determined that both the FoxO1 and Smad3 mutant teeth exhibit changes in the expression of similar cohort of genes encoding enamel matrix proteins required for proper enamel development. These data raise the possibility that FoxO1 and Smad3 act in concert to regulate a common repertoire of genes necessary for complete enamel maturation. This study is the first to define an essential role for the FoxO family of transcription factors in tooth development and provides a new molecular entry point which will allow researchers to delineate novel genetic pathways regulating the process of biomineralization which may also have significance for studies of human tooth diseases such as amelogenesis imperfecta. Corallium species have ecological, cultural and commercial importance and thus require tools to assist with their identification for both management and trade. The organic matrix (OM) of the skeletons of four Corallium species ( C. rubrum, C. konojoi, C. secundum and C. elatius) was examined to provide insight into the biomineralization process and to develop a new tool of identification. The pattern of OM and the set of soluble organic matrix proteins (SOM) in the skeletons were examined by gel electrophoresis. Staining of cross-sections of skeletons showed a common cyclic, concentric pattern of OM during growth. Differences in molecular weight and isoelectric point were observed for proteins in the SOM in different Corallium species but not among different populations of Corallium rubrum. Immunolabeling with antibodies against the SOM of C. rubrum showed labeling of the OM of the three other Corallium species suggesting the presence of common epitopes. [ABSTRACT FROM AUTHOR]

Contents of mineral substance, silica, and a range of bioelements and toxic elements (Mg, Na, K, Ca, Ba, Zn, Cr, P Al, Cd, Mn Cu, Ni, Pb, Sr, Fe) in 38 livers of donors from the Upper Silesia Coal Basin (southern Poland) are presented. Elements were determined by inductively coupled plasma-atomic emission spectrometry (ICP-AES) with the exception of silica that was estimated colorimetrically. Concentrations, concentration variability, and correlations between selected liver components determined for the total population are related to donor age, gender, and lesion occurrence. Correlations between particular elements were found using correlation coefficient values and the Fisher transformation. Mineral substance in the livers lies in the range 0.40-5.03 wt%. With increasing donor age, mineral-matter content decreases to a minimum for the 40-60 years of age range. Microbioelement contents show a similar tendency, while microbioelements and toxic elements reach maximum contents in donors aged 60-80 years. All elements show content decreases in livers from the oldest group (>80 years). Silica contents increase with age. Variability of element contents is lowest in the older subpopulations. Livers with lesions show lower element contents and variability. The results are compared to literature data for regions of Poland assumed to be of low pollution and to data from comparable regions in Japan and Hungary. Elemental sulfur (S) is deposited each summer onto surface ice at Borup Fiord pass on Ellesmere Island, Canada, when high concentrations of aqueous HS are discharged from a supraglacial spring system. 16S rRNA gene clone libraries generated from sulfur deposits were dominated by β-Proteobacteria, particularly Ralstonia sp. Sulfur-cycling micro-organisms such as Thiomicrospira sp., and ε -Proteobacteria such as Sulfuricurvales and Sulfurovumales spp. were also abundant. Concurrent cultivation experiments isolated psychrophilic, sulfide-oxidizing consortia, which produce S in opposing gradients of NaS and oxygen. 16S rRNA gene analyses of sulfur precipitated in gradient tubes show stable sulfur-biomineralizing consortia dominated by Marinobacter sp. in association with Shewanella, Loktanella, Rubrobacter, Flavobacterium, and Sphingomonas spp. Organisms closely related to cultivars appear in environmental 16S rRNA clone libraries; none currently known to oxidize sulfide. Once consortia were simplified to Marinobacter and Flavobacteria spp. through dilution-to-extinction and agar removal, sulfur biomineralization continued. Shewanella, Loktanella, Sphingomonas, and Devosia spp. were also isolated on heterotrophic media, but none produced S alone when reintroduced to NaS gradient tubes. Tubes inoculated with a Marinobacter and Shewanella spp. co-culture did show sulfur biomineralization, suggesting that Marinobacter may be the key sulfide oxidizer in laboratory experiments. Light, florescence and scanning electron microscopy of mineral aggregates produced in Marinobacter experiments revealed abundant cells, with filaments and sheaths variably mineralized with extracellular submicron sulfur grains; similar biomineralization was not observed in abiotic controls. Detailed characterization of mineral products associated with low temperature microbial sulfur-cycling may provide biosignatures relevant to future exploration of Europa and Mars. Bone is a hierarchically structured composite consisting of a protein phase (type I collagen) and a mineral phase (carbonated apatite). The objective of this study was to investigate the hierarchical structure of mineral in mature bone. A method to completely deproteinize bone without altering the original structure is developed, and the completion is confirmed by protein analysis techniques. Stereoscopy and field emission electron microscopy are used to examine the structural features from submillimeter- to micrometer- to nanometer-length scales of bovine femur cancellous bone. Stereoscopic images of fully deproteinized and demineralized bovine femur cancellous bone samples show that fine trabecular architecture is unaltered and the microstructural features are preserved, indicating the structural integrity of mineral and protein constituents. SEM revealed that bone minerals are fused together and form a sheet-like structure in a coherent manner with collagen fibrils. Well-organized pore systems are observed at varying hierarchical levels. Mineral sheets are peeled off and folded after compressive deformation, implying strong connection between individual crystallites. Results were compared with commercially available heat-deproteinized bone (Bio-Oss(®)), and evidence showed consistency in bone mineral structure. The formation of defined shapes by cells is one of the challenging questions in biology. Due to the anisotropy of cell walls and of certain biominerals, the LC-PolScope represents a promising tool for tracking dynamic structural changes in vivo non-invasively and, to some extent, quantitatively. A complex three-dimensional biogenic system, the in vitro precipitation of calcium oxalate induced by cellulose stalks produced by Dictyostelium discoideum, was analyzed. Although the retardance values and orientation of the crystals with respect to the stalk were quickly and easily detected, this study raised a number of issues that were addressed in this work. The effect of the refractive index of the embedding medium was examined by taking advantage of the homogeneous size and shape distribution of kiwifruit raphides, a biologically controlled calcium oxalate biomineral and of cotton ( Gossypium) seed fibers. The retardance remained consistent when embedding these samples in media with increasing refractive indices from 1.33 to 1.42 or 1.47 for sucrose or glycerol gradients, respectively. The general applicability of LC-PolScope image processing for biominerals and cell wall formation during development in vivo was demonstrated in a particular group of green algae, the Desmidiaceae. Various organization levels of the cell wall were identified, thus confirming earlier findings based on electron microscopy and immunostaining investigations. It can be concluded that LC-PolScope microscopy is an attractive tool for studying dynamic ordering of biomolecules, such as plant cell walls, when additional parameters regarding the structure, composition, and refractive indices of the specimen are available. Abstract: Four non-magnetic mutants of Magnetospirillum gryphiswaldense strain MSR-1 were isolated by transposon mutagenesis and found to contain interruption of either the mamO or mamE gene within the mamAB operon. Studies indicated that mamO and mamE genes are essential for magnetosome crystal biomineralization in MSR-1. Biomineralization is the process by which hydroxyapatite is deposited in the extracellular matrix. Physiological mineralization occurs in hard tissues, whereas pathological calcification occurs in soft tissues. The first step of mineralization is the formation of hydroxyapatite crystals within matrix vesicles that bud from the surface membrane of hypertrophic chondrocytes, osteoblasts, and odontoblasts. This is followed by propagation of hydroxyapatite into the extracellular matrix and its deposition between collagen fibrils. Extracellular inorganic pyrophosphate, provided by NPP1 and ANKH, inhibits hydroxyapatite formation. Tissue- nonspecific alkaline phosphatase (TNAP) hydrolyzes pyrophosphate and provides inorganic phosphate to promote mineralization. Inorganic pyrophosphate, pyridoxal phosphate, and phosphoethanolamine are thought to be the physiologic substrates of TNAP. These accumulate in the event of TNAP deficiency, e.g., in cases of hypophosphatasia. The gene encoding TNAP is mapped to chromosome 1, consists of 12 exons, and possesses regulatory motifs in the 5′-untranslated region. Inhibition of TNAP enzymatic activity suppresses TNAP mRNA expression and mineralization in vitro. Hypophosphatasia is an inherited systemic bone disease characterized by hypomineralization of hard tissues. The phenotype of hypophosphatasia is varied. To date, more than 200 mutations in the TNAP gene have been reported. Knockout mice mimic the phenotypes of severe hypophosphatasia. Among the mutations in the TNAP gene, c.1559delT is frequent in the Japanese population. This frameshift mutation results in the expression of an abnormally long protein that is degraded in cells. DNA-based prenatal diagnosis using chorionic villus sampling has been developed, but requires thorough genetic counseling. Although hypophosphatasia is untreatable at present, the recent success of enzyme replacement therapy offers promise. The problems presented by impaired mineralization in age-related chronic diseases, such as pathologic calcification and decreasing physiological mineralization are growing in importance. Strategies for preventing pathologic calcification using TNAP and NPP1 are in development. The aim of this study was to prepare bone like mineral (BLM) layers rapidly on the exterior surfaces of chitosan (CS) microparticles (MPs). The CS MPs were fabricated using a scale-up double emulsification method. The CS MPs were in the spherical shape and the size of 30–60 μm. The MPs were then placed in 5× concentrated simulated body fluid (5 × SBF) and allowed to undergo biomineralization to form a BLM layers on the surface of CS MPs at 37°C over a 24 h period. The BML layers on the exterior surface of CS MPs were characterized using wide angle X-ray diffraction (XRD), Fourier transform infrared microscopy (FTIR), and scanning electron microscopy (SEM). Insulin like growth factor-1 (IGF-1) was dissolved at a concentration of 1 μg/ml in 5 × SBF to incorporate into the BLM layer. The CS MPs (100 mg) were incubated in a sample of 4 ml of 5 × SBF containing IGF-1 at a concentration of 1 μg/ml for 24 h. The IGF-1 release from BML layers on CS MPs were studied by placing MPs in 4 ml of phosphate buffered saline (PBS) and incubating MPs at 37°C for 30 days. Samples (100 μl) were taken over the course of the 30 days and analyzed using Enzyme-linked Immunosorbent assay (ELISA). The release IGF-1 from BML layers was in a burst manner followed by a sustained release during the 30-day period. This study suggests that the CS MPs have the potential to be used to help deliver therapeutic drugs to localized areas and hence increase and accelerate bone growth. [ABSTRACT FROM AUTHOR]

The aim of this study was to evaluate dogs with urolithiasis in which mineral composition of calculi was quantitatively analyzed. Quantitative mineral composition was performed in 156 canine uroliths. Simple uroliths represented 79.5% (n=124) of the cases, 18% were compound (n=28) and only 2.5% (n=4) of the calculi were mixed. Struvite was the most frequent mineral type of simple uroliths (47.6%; n=59) as well as in all mixed (100%; n=4) and in the core and stone uroliths (32.1% and 75%, respectively). Calcium oxalate was the second more frequent mineral composition of simple uroliths (37.9%; n=47). Unlike simple uroliths, recommendation for the treatment of compound uroliths is more complex, and diet protocols for calculi dissolution may be different when the calculus is composed by different minerals that are possible to be dissolved (e.g. urate and stuvite). Besides, dissolution may not be feasible if it occurs in presence of insoluble material involving urolith or if it represents more than 20% of the layer. Twenty two compound uroliths (78.7%) presented an external layer that was not possible to be dissolved (calcium oxalate or calcium phosphate); two calculi (7.1%) had superficial layers dissolvable (struvite or urate), but inner layers were not soluble, which allowed only partial dissolution of urolith. Knowledge of all urolith layers mineral composition is essential for the understanding of calculus formation and for the adequate treatment indication as well as for the procedures to prevent recurrence. Background: The shell of the pearl-producing bivalve Pinctada margaritifera is composed of an organic cell-free matrix that plays a key role in the dynamic process of biologically-controlled biomineralization. In order to increase genomic resources and identify shell matrix proteins implicated in biomineralization in P. margaritifera, highthroughput Expressed Sequence Tag (EST) pyrosequencing was undertaken on the calcifying mantle, combined with a proteomic analysis of the shell. Results: We report the functional analysis of 276 738 sequences, leading to the constitution of an unprecedented catalog of 82 P. margaritifera biomineralization-related mantle protein sequences. Components of the current "chitin-silk fibroin gel-acidic macromolecule" model of biomineralization processes were found, in particular a homolog of a biomineralization protein (Pif-177) recently discovered in P. fucata. Among these sequences, we could show the localization of two other biomineralization protein transcripts, pmarg-aspein and pmarg-pearlin, in two distinct areas of the outer mantle epithelium, suggesting their implication in calcite and aragonite formation. Finally, by combining the EST approach with a proteomic mass spectrometry analysis of proteins isolated from the P. margaritifera shell organic matrix, we demonstrated the presence of 30 sequences containing almost all of the shell proteins that have been previously described from shell matrix protein analyses of the Pinctada genus. The integration of these two methods allowed the global composition of biomineralizing tissue and calcified structures to be examined in tandem for the first time. Conclusions: This EST study made on the calcifying tissue of P. margaritifera is the first description of pyrosequencing on a pearl-producing bivalve species. Our results provide direct evidence that our EST data set covers most of the diversity of the matrix protein of P. margaritifera shell, but also that the mantle transcripts encode proteins present in P. margaritifera shell, hence demonstrating their implication in shell formation. Combining transcriptomic and proteomic approaches is therefore a powerful way to identify proteins involved in biomineralization. Data generated in this study supply the most comprehensive list of biomineralization-related sequences presently available among protostomian species, and represent a major breakthrough in the field of molluskan biomineralization. The skeleton, teeth, and otoconia are normally the only mineralized tissues or organs in the human body. We describe physiological biomineralization in collagenous matrices as well as a more derived noncollagenous matrix. The origin of the collagenous matrices used in mineralized skeletal tissues can be traced to a soft tissue in early Metazoa. In early vertebrates, a genetic system coding for ancient soft collagenous tissue was-co-opted for biomineralization using redundant-genes resulting from whole genome duplication. However, genes more specific to mineralized tissues arose subsequent to the genome duplication by genomically local tandem duplication. These new genes are the basis for a novel genetic system for various mineralized tissues in skeleton and teeth. In addition, any tissue can be abnormally mineralized, and many pathologies of mineralization in humans are known. [ABSTRACT FROM AUTHOR]

Iron oxidation at neutral pH by the phototrophic anaerobic iron-oxidizing bacterium Rhodobacter sp. strain SW2 leads to the formation of iron-rich minerals. These minerals consist mainly of nano-goethite (α-FeOOH), which precipitates exclusively outside cells, mostly on polymer fibers emerging from the cells. Scanning transmission X-ray microscopy analyses performed at the C K-edge suggest that these fibers are composed of a mixture of lipids and polysaccharides or of lipopolysaccharides. The iron and the organic carbon contents of these fibers are linearly correlated at the 25-nm scale, which in addition to their texture suggests that these fibers act as a template for mineral precipitation, followed by limited crystal growth. Moreover, we evidence a gradient of the iron oxidation state along the mineralized fibers at the submicrometer scale. Fe minerals on these fibers contain a higher proportion of Fe(III) at cell contact, and the proportion of Fe(II) increases at a distance from the cells. All together, these results demonstrate the primordial role of organic polymers in iron biomineralization and provide first evidence for the existence of a redox gradient around these nonencrusting, Fe-oxidizing bacteria. The inhibition of mineralization of urinary stone forming minerals by medicinal plants i.e. Achyranthes aspera Linn, Passiflora leschenaultii DC, Solena amplexicaulis (Lam.) Gandhi, Scoparia dulcis Linn and Aerva lanata (Linn.) been investigated. The inhibition efficiency was studied. The entire tube wall of Dodecaceria caulleryi and D. coralii is composed of thin calcareous lamellae with a spherulitic prismatic ultrastructure. In Diplochaetetes mexicanus the tube ultrastructure is similar, but contains also some lamellae with a homogeneous structure. The biomineralization system of cirratulids has not evolved since the appearance of calcareous tubes in the Oligocene. Abstract: Antler is a unique mineralized tissue, with extraordinary toughness as well as an ability to annually regenerate itself in its entirety. The high fracture resistance enables it to fulfill its biological function as a weapon and defensive guard during combats between deer stags in the rutting season. However, very little is quantitatively understood about the structural origin of the antler''s high toughness. We used a unique combination of time-resolved synchrotron small angle X-ray diffraction together with tensile testing of antler cortical tissue under physiologically wet conditions. We measured the deformation at the nanoscale from changes in the meridional diffraction pattern during macroscopic stretch-to-failure tests. Our results show that on average fibrils are strained only half as much as the whole tissue and the fibril strain increases linearly with tissue strain, both during elastic and inelastic deformation. Most remarkably, following macroscopic yielding we observe a straining of some fibrils equal to the macroscopic tissue strain while others are hardly stretched at all, indicating an inhomogeneous fibrillar strain pattern at the nanoscale. This behavior is unlike what occurs in plexiform bovine bone and may explain the extreme toughness of antler compared to normal bone. [Copyright &y& Elsevier]

Pseudoxanthoma elasticum (PXE) is an autosomal recessive multisystem disorder characterized by ectopic connective tissue mineralization, with clinical manifestations primarily in the skin, eyes, and cardiovascular system. There is considerable, both intra- and interfamilial, variability in the spectrum of phenotypic presentation. Previous studies have suggested that mineral content of the diet may modify the severity of the clinical phenotype in PXE. In this study, we utilized a targeted mutant mouse (Abcc6−/−) as a model system for PXE. We examined the effects of changes in dietary phosphate and magnesium on the mineralization process using calcification of the connective tissue capsule surrounding the vibrissae as an early phenotypic biomarker. Mice placed on custom-designed diets either high or low in phosphate did not show changes in mineralization, which was similar to that noted in Abcc6−/− mice on control diet. However, mice placed on diet enriched in magnesium (fivefold) showed no evidence of connective tissue mineralization in this mouse model of PXE. The inhibitory capacity of magnesium was confirmed in a cell-based mineralization assay system in vitro. During the mineralization process of enamel, gene expression controls the activities of ameloblasts, the secretion and assembly of an extracellular protein matrix, affecting the final structure and functions. In this study, the enamel in the maxillary and mandibular incisors of wild-type and transgenic (col1-caPPR) mice, in which a constitutively active PTH/PTHrP receptor (PPR) was targeted to osteoblastic cells, was observed by scanning electron microscopy (SEM), Fourier transform infrared microscopy (FTIRM), and nanoindentation. The SEM studies showed that several different patterns of aberrations in crystal arrangement, disturbed prism organization without decussation, as well as abnormal enamel distribution were encountered in transgenic enamel. FTIRM analysis revealed poorer crystallinity/maturity after mutation. Nanoindentation measurement disclosed that transgenic enamel had 24.6% lower hardness and 12.3% lower elastic modulus. We attributed the inferior properties to the loosely packing crystals and abnormal prism organization. Furthermore, the col1-caPPR mouse model was substantiated to be useful to study how genes modulate the biomineralization process. [ABSTRACT FROM AUTHOR]
