Background: MiRNA primarily acts to repress gene expression at the post-transcriptional level through imperfect complementarity of its 5′ region to the “seed site” in the 3′ untranslated region of target mRNAs, with its “3′–supplementary site” and “center site” also playing important roles under certain circumstances. The aim of this study was to test if artificial miRNA mimics (miR-Mimics) that are designed to target the “centered sites” without “seed sites” complementarity are able to repress gene expression as natural miRNAs. Methods: We designed miR-Mimics carrying centered-site matches (CS–miR-Mimics) or seed-site matches (SS–miR-Mimics) and siRNA to two antiapoptotic genes BCL2 and AKT1. We tested the gene targeting of these constructs using real-time RT-PCR and Western blot to quantify mRNA and protein levels of BCL2 and AKT1, respectively, luciferase reporter gene assay to investigate the interaction between miR-Mimics and their target sites, and cell survival assay to study the functional outcomes of the miR-Mimics. Results: We found that CS-miR-Mimic, SS-miR-Mimic and siRNA, all down regulated the mRNA and protein levels of their cognate target BCL2 or AKT1 in a concentration-dependent manner. Luciferase reporter gene assay further confirmed the functional interactions of CS–miR-Mimic, SS-miR-Mimic and siRNA with their target sites. We then observed that the miR-Mimics and siRNAs were all able to induce cell death, as indicated by the reduced survival rate of cells. Conclusions: We have provided evidence for the feasibility of CS–miR-Mimics for post-transcriptional repression of genes, which can be designed to have reduced numbers of seed type off-target sites compared to the number of target sites from an average endogenous seed–site miRNA. Two new sesquiterpene pyridine alkaloids hypoglaunines E (1) and F (2) and a new triterpenoid saponin hypoglaside A (3), together with a known diterpenoid glucoside 11-O-β-d-glucopyranosyl neotriptophenolide (4) and two known triterpenoids 23-nor-oxopristimerol (5) and 2,3-dihydroxy-6-oxo-D:A-froedo-24 nor-1,3,5(10),7-oleanatetraen-29-oic acid (6), have been isolated from the root barks of Tripterygium hypoglaucum. Their structures were elucidated by NMR spectroscopy and MS analysis. Compounds 1–3 were screened for their cytotoxic activities against five cancer cell lines, but all of them were inactive. Afforestation activities in the Kubuqi Desert, Inner Mongolia, China, have substantially increased tree and shrub coverage in this region. In this study, the response of the surface temperature to afforestation is simulated with the Weather Research and Forecasting model. The surface temperature changes are decomposed into contributions from the intrinsic surface biophysical effect and atmospheric feedback, using the theory of intrinsic biophysical mechanism. The effect of afforestation on the surface temperature is 1.34 K, −0.48 K, 2.09 K and 0.22 K for the summer daytime, the summer nighttime, the winter daytime and the winter nighttime, respectively, for the grid cells that have experienced conversion from bare soil to shrub. The corresponding domain mean values are 0.15 K, −0.2 K, 0.67 K, and 0.06 K. The seasonal variation of surface temperature change is mainly caused by changes in roughness and Bowen ratio. In the daytime, the surface temperature changes are dominated by the biophysical effect, with albedo change being the main biophysical factor. In the nighttime, the biophysical effect (mainly associated with roughness change) and the atmospheric feedback (mainly associated with change in the background air temperature) contribute similar amounts to the surface temperature changes. Abstract: Helicobacter pylori is a bacterium that causes chronic active gastritis and peptic ulcers. Drugs targeting H. pylori phosphopantetheine adenylyltransferase (HpPPAT), which is involved in CoA biosynthesis, may be useful. Herein, we report the expression in Escherichia coli and purification of recombinant HpPPAT and describe a crystal structure for an HpPPAT/CoA complex. As is the case for E. coli PPAT (EcPPAT), HpPPAT is hexameric in solution and as a crystal. Each protomer has a well-packed dinucleotide-binding fold in which CoA binds. Structural characterisation demonstrated that CoA derived from the E. coli expression system bound tightly to HpPPAT, presumably to initiate feedback inhibition. However, the interactions between the active-site residues of HpPPAT and CoA are not identical to those of other PPATs. Finally, CoA binding affects HpPPAT thermal denaturation. Abstract: Four new neolignans, wilfordiols A–D (1–4), together with five known compounds (5–9), were isolated from an aqueous extract of the dried leaves of Tripterygium wilfordii. Their structures were determined by spectroscopic methods, including 1D and 2D NMR, HRESIMS, and CD experiments.
