Design dark fraction reflects the unlit part of a microalgal culture system, as for example a hydraulic loop used for temperature or pH regulation, or a circulating pump for mixing purposes. This study investigates the impact of design dark fraction on photosynthetic biomass productivity of the eukaryotic microalgae Chlorella vulgaris. The effect of the volume of the dark fraction and the residence time spent in this dark fraction was investigated with two different nitrogen sources (N-NH 4 + , N-NO 3 - ). Results showed a decrease of biomass productivity when the volume of the dark fraction and the dark residence time increased. Up to 47% loss of biomass productivity could be reached for a design dark fraction [Formula: see text] = 30% of the total culture system volume. This loss was explained as a result of metabolic reactions related to an increase of respiration activity or a decrease of photosynthetic activity in the cells.

Industrial development of microalgae biomass valorization relies on process optimization and controlled scale-up. Both need robust modeling: (i) for biomass production and (ii) for integrated processes in the downstream processing (DSP). Cell disruption and primary fractionation are key steps in DSP. In this study, a kinetic model, including microalgal cell size distribution, was developed for Chlorella sorokiniana disruption in continuous bead milling. Glass beads of 0.4 mm size at impeller tip velocity of 14 m.s -1 were used as optimal conditions for efficient cell disruption. These conditions allowed faster disruption of big cells than small ones. A modified expression of the Stress Number, including cell size effect, was then proposed and validated. Separation of starch, proteins and chlorophyll by mild centrifugation was studied as function of the disruption parameters. Low energy consumption conditions led to extreme comminution. Modeling was done to simulate whole-year running of solar rectangular photobioreactors (PBRs). Introducing the concept of ideal reactor, the maximal biomass productivity that could be achieved on Earth on nitrate as N-source was calculated. Two additional factors were also analyzed with respect to dynamic calculations over the whole year: the effect of PBR location and the effects of given operating conditions on the resulting decrease in productivity compared with the ideal one. Simulations were conducted for the cyanobacterium Arthospira platensis, giving an ideal productivity (upper limit) in the range 55-60 tX ha(-1) year(-1) for a sun tracking system (and around 35-40 tX ha(-1) year(-1) for a fixed horizontal PBR). For an implantation in France (Nantes, west coast), the modification in irradiation conditions resulted in a decrease in biomass productivity of 40%. Various parameters were investigated, with special emphasis on the influence of the incident angle of solar illumination on resulting productivities, affecting both light capture and light transfer inside the bulk culture. The aim of this study was to establish and validate a model for the photosynthetic growth of Chlamydomonas reinhardtii in photobioreactors (PBRs). The proposed model is based on an energetic analysis of the excitation energy transfer in the photosynthesis apparatus (the Z-scheme for photosynthesis). This approach has already been validated in cyanobacteria (Arthorspira platensis) and is extended here to predict the volumetric biomass productivity for the microalga C. reinhardtii in autotrophic conditions, taking into consideration the two metabolic processes taking place in this eukaryotic microorganism, namely photosynthesis and respiration. The kinetic growth model obtained was then coupled to a radiative transfer model (the two-flux model) to determine the local kinetics, and thereby the volumetric biomass productivity, in a torus PBR. The model was found to predict PBR performances accurately for a broad set of operating conditions, including both light-limited and kinetic growth regimes, with a variance of less than 10% between experimental results and simulations., (Copyright © 2012 American Institute of Chemical Engineers (AIChE).)

We describe a methodology to investigate the potential of given microalgae species for biodiesel production by characterizing their productivity in terms of both biomass and lipids. A multi-step approach was used: determination of biological needs for macronutrients (nitrate, phosphate and sulphate), determination of maximum biomass productivity (the "light-limited" regime), scaling-up of biomass production in photobioreactors, including a theoretical framework to predict corresponding productivities, and investigation of how nitrate starvation protocol affects cell biochemical composition and triggers triacylglycerol (TAG) accumulation. The methodology was applied to two freshwater strains, Chlorella vulgaris and Neochloris oleoabundans, and one seawater diatom strain, Cylindrotheca closterium. The highest total lipid content was achieved with N. oleoabundans (25-37% of DW), while the highest TAG content was found in C. vulgaris (11-14% of DW). The validity of a simple, reliable, and useful recently published formula enabling to calculate the maximum volumetric biomass productivities in photobioreactors (PBRs) was investigated through the cultivation of the microalga Chlamydomonas reinhardtii. Experimental maximum kinetic performances accurately obtained in two different, artificially lightened torus-plane and cylindrical reactors having the same specific illuminated area confirmed the availability, power, and robustness of such formula. The predictive kinetic parameters previously proposed and validated with cyanobacteria were then proved general and robust in case of eukaryotic microalgae, as postulated in the founding article. In this case, an additional criterion requiring rigorous control of the working illuminated fraction gamma = 1 +/- (15%) inside the reactor is demonstrated. For this, the usefulness and reliability of a generalized two-flux model accurately describing the radiation field inside turbid culture media of C. reinhardtii were also established in this article. These important results contribute to identify the main engineering factors governing light-limited PBRs functioning and then to clarify some misinterpretations widely reported in the literature. Together with the referenced previous work, this article gives a framework toward optimal conception of PBRs on a strong physical basis.

The fresh water microalga Neochloris oleoabundans was investigated for its ability to accumulate lipids and especially triacylglycerols (TAG). A systematic study was conducted, from the determination of the growth medium to its characterization in an airlift photobioreactor. Without nutrient limitation, a maximal biomass areal productivity of 16.5 g m(-2) day(-1) was found. Effects of nitrogen starvation to induce lipids accumulation was next investigated. Due to initial N. oleoabundans total lipids high content (23% of dry weight), highest productivity was obtained without mineral limitation with a maximal total lipids productivity of 3.8 g m(-2) day(-1). Regarding TAG, an almost similar productivity was found whatever the protocol was: continuous production without mineral limitation (0.5 g m(-2) day(-1)) or batch production with either sudden or progressive nitrogen deprivation (0.7 g m(-2) day(-1)). The decrease in growth rate reduces the benefit of the important lipids and TAG accumulation as obtained in nitrogen starvation (37% and 18% of dry weight, respectively).

This study deals with the scale of a new photobioreactor for continuous microalgal production in hatcheries. The combination of the state-of-art with the constraints inherent to hatcheries has turned the design into a closed, artificially illuminated and external-loop airlift configuration based on a succession of elementary modules, each one being composed of two transparent vertical interconnected columns. The liquid circulation is ensured pneumatically (air injections) with respect to a swirling motion (tangential inlets). A single module of the whole photobioreactor was built-up to scale its geometry (diameter and length) and to optimize its design (air sparger, tangential inlets). The volumetric productivities were predicted by modeling radiative transfer and growth of Isochrysis affinis galbana (clone Tahiti). The hydrodynamics of the liquid phase was modeled in terms of global flow behavior (circulation and mixing times, Péclet number) and of swirling motion decay along the column (Particle Image Velocimetry). The aeration performances were determined by overall volumetric mass transfer measurements. Continuous cultures of Isochrysis affinis galbana (clone Tahiti) were run in two geometrical configurations, generating either an axial or a swirling flow. Lastly, the definitive options of design are presented as well as a 120-L prototype, currently implemented in a French mollusk hatchery and commercialized.

The light attenuation in a photobioreactor is determined using a fully predictive model. The optical properties were first calculated, using a data bank of the literature, from only the knowledge of pigments content, shape, and size distributions of cultivated cells which are a function of the physiology of the current species. The radiative properties of the biological turbid medium were then deduced using the exact Lorenz-Mie theory. This method is experimentally validated using a large-size integrating sphere photometer. The radiative properties are then used in a rectangular, one-dimensional two-flux model to predict radiant light attenuation in a photobioreactor, considering a quasi-collimated field of irradiance. Combination of this radiative model with the predictive determination of optical properties is finally validated by in situ measurement of attenuation profiles in a torus photobioreactor cultivating the microalgae Chlamydomonas reinhardtii, after a complete and proper characterization of the incident light flux provided by the experimental set-up., (Copyright 2005 Wiley Periodicals, Inc)

Light is the main limiting factor in photoautotrophic-intensive production of microorganisms, and improvement of its use is an important concern for photobioreactor design and operation. Swirling flows, which are known to improve mass and photon transfers, were applied to annular light chambers of a photobioreactor and studied by simulation and microalgal culture. Two hydrodynamic conditions were compared: axial flow generating poor radial mixing, and tangential flow generating three-dimensional swirling motion. Batch and continuous cultures of the Rhodophyte Porphyridium cruentum were performed in a 100-L, 1.5-m(2), fully controlled photobioreactor with eight light chambers. The inlet design of these chambers was modified to create the hydrodynamic conditions for comparison. Various intensities of swirling motion were used, characterized by the velocity factor (VF), defined as the ratio between annular chamber flow and inlet aperture sections. Experiments were performed within the range of photon flux densities (PFD) optimizing the yield of light energy transformation into living substance for the species and the temperature used. Culture kinetics with swirling flows generated by apertures of VF = 2, 4, and 9 were compared with pseudoaxial VF = 2 chosen as reference. Batch cultures with VF = 4 swirling flow showed no significant difference, whereas continuous cultures proved more discriminating. Although no significant difference was obtained for VF = 2, a 7% increase of steady-state productivity and a 26% decrease in time required to reach this steady state were obtained with VF = 4 swirling flow. This beneficial effect of swirling flow could have accounted for increased mixing. Conversely, VF = 9 swirling flow resulted in a 9% decrease of steady-state productivity and a 9% increase in the time required to reach this steady state, a negative effect that could have accounted for increased shear stress. CO(2) bioconversion yield at steady state showed a 34% increase for VF = 4. These results suggest that swirling motion makes microalgal cultures more efficient, provided that the resulting adverse effects remain acceptable. Experimental investigation was completed by a theoretical approach in which simulation of continuous cultures of P. cruentum was based on the hydrodynamic conditions achieved in the photobioreactor. Although the results obtained with pseudoaxial flow were correctly predicted, simulations with swirling flow showed a marked enhancement of productivity not observed experimentally. The influence of side effects induced by increased mixing (particularly hydrodynamic shear stress) was considered with respect to modeling assumptions. Comparison of experimental results with theoretical simulation provided a better understanding of the mixing effect, a key factor in improving the efficiency of such bioprocesses., (Copyright 2004 Wiley Periodicals, Inc. A deterministic Markov process was developed to estimate the efficiency of a fully controlled photobioreactor by calculating the growth of the Rhodophyte Porphyridium cruentum. It was assumed that microalgal growth, in strictly controlled and non-nutrient-limited conditions, is a function of the amount of light energy received. The light sources delivered a continuous 206 microE m(-2) s(-1) average photon flux density to cultures reaching concentrations of 3 g l(-1) in batch and 0.7 g l(-1) in chemostat. The concentration time-courses calculated compared satisfactorily with measured results for both batch and continuous cultures. The quality of simulation in these two cases validated the hypotheses made, especially for the linearity of light bioconversion, and allowed the model to be used for further exploration of the photobioreactor-operating domain. Distribution of the specific growth rate as a function of time and radial position was compared for the two simulated cases. The use of an initial batch of several days prior to the dilution of a continuous culture proved theoretically beneficial for overall production. The influence of the dilution rate and of light-path length on surface productivity and concentration at steady state were demonstrated.
