Cryptochromes are ubiquitously expressed in various animal tissues including the retina. Some cryptochromes are involved in regulating circadian activity. Cryptochrome proteins have also been suggested to mediate the primary mechanism in light-dependent magnetic compass orientation in birds. Cryptochrome 1b (Cry1b) exhibits a unique carboxy terminus exclusively found in birds so far, which might be indicative for a specialised function. Cryptochrome 1a (Cry1a) is so far the only cryptochrome protein that has been localised to specific cell types within the retina of migratory birds. Here we show that Cry1b, an alternative splice variant of Cry1a, is also expressed in the retina of migratory birds, but it is primarily located in other cell types than Cry1a. This could suggest different functions for the two splice products. Using diagnostic bird-specific antibodies (that allow for a precise discrimination between both proteins), we show that Cry1b protein is found in the retinae of migratory European robins (Erithacus rubecula), migratory Northern Wheatears (Oenanthe oenanthe) and pigeons (Columba livia). In all three species, retinal Cry1b is localised in cell types which have been discussed as potentially well suited locations for magnetoreception: Cry1b is observed in the cytosol of ganglion cells, displaced ganglion cells, and in photoreceptor inner segments. The cytosolic rather than nucleic location of Cry1b in the retina reported here speaks against a circadian clock regulatory function of Cry1b and it allows for the possible involvement of Cry1b in a radical-pair-based magnetoreception mechanism.

The most studied comparison of aging and maximum lifespan potential (MLSP) among endotherms involves the 7-fold longevity difference between rats (MLSP 5y) and pigeons (MLSP 35y). A widely accepted theory explaining MLSP differences between species is the oxidative stress theory, which purports that reactive oxygen species (ROS) produced during mitochondrial respiration damage bio-molecules and eventually lead to the breakdown of regulatory systems and consequent death. Previous rat-pigeon studies compared only aspects of the oxidative stress theory and most concluded that the lower mitochondrial superoxide production of pigeons compared to rats was responsible for their much greater longevity. This conclusion is based mainly on data from one tissue (the heart) using one mitochondrial substrate (succinate). Studies on heart mitochondria using pyruvate as a mitochondrial substrate gave contradictory results. We believe the conclusion that birds produce less mitochondrial superoxide than mammals is unwarranted. We have revisited the rat-pigeon comparison in the most comprehensive manner to date. We have measured superoxide production (by heart, skeletal muscle and liver mitochondria), five different antioxidants in plasma, three tissues and mitochondria, membrane fatty acid composition (in seven tissues and three mitochondria), and biomarkers of oxidative damage. The only substantial and consistent difference that we have observed between rats and pigeons is their membrane fatty acid composition, with rats having membranes that are more susceptible to damage. This suggests that, although there was no difference in superoxide production, there is likely a much greater production of lipid-based ROS in the rat. We conclude that the differences in superoxide production reported previously were due to the arbitrary selection of heart muscle to source mitochondria and the provision of succinate. Had mitochondria been harvested from other tissues or other relevant mitochondrial metabolic substrates been used, then very different conclusions regarding differences in oxidative stress would have been reached.

Oxidative stress, an imbalance between endogenous levels of oxygen radicals and antioxidative defense, increases with aging. However, it is not clear which of these two factors is the more critical. To clarify the production of oxygen radicals increases with age, we examined oxygen radical-dependent chemiluminescent signals in ex vivo brain slices using a novel photonic imaging method. The chemiluminescent intensity was significantly decreased by the membrane permeable superoxide dismutase (SOD)/catalase mimic, but not by Cu,Zn-SOD. Inhibitors for complex I, III, and IV of the mitochondrial electron transport chain transiently enhanced the chemiluminescent signal. The superoxide-dependent chemiluminescent intensity in senescence accelerated mouse (SAM) brain tissues increases with age. Moreover, the slope of the age-dependent increase was steeper in SAMP10, a strain characterized by a short lifespan and atrophy in the frontal cerebral cortex, than the senescence-resistant strain SAMR1, which has a longer lifespan. An increase in chemiluminescence with age was also observed in C57/BL6 mice, Wistar rats, and pigeons, although levels of chemiluminescence were lower in the pigeons than murines. The rate of age-related increases of superoxide-dependent chemiluminescence was inversely related to the maximum lifespan of the animals. The activity of superoxide dismutase was unchanged during the aging process in the brain. This suggested that superoxide production itself may increase with age. We speculated that reactive oxygen may be a signal to determine the aging process.

Androgens and estrogens are crucial for the differentiation and function of the vocal control system of songbirds. A major source of estrogens in songbirds is the cerebral aromatization of circulating testosterone by aromatase (ARO). In the vocal control system, songbirds have a unique estrogen receptor (ER)-containing area, the nucleus hyperstriatalis ventrale pars caudale (HVC) of the caudal neostriatum. Work in the zebra finch has demonstrated ARO expression adjacent to but not in the HVC. Compared with other songbirds, such as the canary, the HVC of adult zebra finches contains only few ERs. To determine whether the disjunctive distribution of ERs and ARO in the forebrain is a songbird-specific feature, the authors investigated ARO and ER mRNA expression in songbirds (canary, house sparrow, and zebra finch) and in nonsongbirds (budgerigar, ring dove, swift, grey partridge, and barn owl) of five avian orders. In addition, the coexpression of androgen receptor (AR) and ARO mRNAs was studied. Preoptic hypothalamic areas showed similar expression of ARO in all species. In the caudal neostriatum, ARO, AR, and ER transcripts were found only in songbirds. ARO and ER mRNA expression in the caudal forebrain was spatially separated, i.e., the HVC contained ER mRNA but very little or no ARO mRNA, and the caudomedial neostriatum contained high levels of ARO mRNA but few if any ERs. ARO and AR mRNAs, however, were coexpressed in the caudomedial neostriatum. The coexpression of ARO mRNA with AR mRNA but not with ER mRNA was found in further brain areas, such as the nucleus posterior lateralis hypothalami. The area-specific coexpression of AR, ER, and ARO suggests various possibilities for the steroid-dependent regulation of ARO and for the role of ARO in controlling AR- and ER-dependent mechanisms.

2,3,7,8-Tetrachlorodibenzo-p-dioxin (TCDD) markedly induced cytochrome P450 (CYP)-dependent arachidonic acid metabolism in liver microsomes from hatchlings of four avian species belonging to four different orders: chick, pigeon, cormorant, and great blue heron, increasing formation of arachidonic acid epoxides (EETs), monohydroxyeicosatetraenoic acids (HETEs), omega-1, and omega-2 OH arachidonic acid products by fivefold or more. Microsomes from TCDD-induced hatchling chicks had the highest activity and the least restricted EET regioselectivity. omega-OH arachidonic acid, the principal constitutive metabolite in chick and pigeon liver microsomes and a major product for cormorant and great blue heron was not induced by TCDD. Constitutive EET formation in avian liver microsomes was very low except in cormorant microsomes where 8,9-EET was generated almost exclusively. Western blots of liver microsomes using polyclonal antisera to chick embryo-derived CYP1A4 and 1A5 recognized two TCDD-induced bands in each of the species. The chick bands had the same molecular weights as CYP1A4 and 1A5 (55 and 55.5 kDa, respectively) but those of the other species differed. Immunopurified antiserum monospecific for CYP1A5 recognized a band in microsomes from all of the avian species, and monospecific antiserum for CYP1A4 recognized a band in microsomes from chick, pigeon, and great blue heron. AntiCYP1A4 and 1A5 IgG immunoinhibited TCDD-induced mixed function oxidase activity completely in chick and chick embryo microsomes and only partially in the other avian microsomes. The results demonstrate that (1) TCDD causes much greater induction of CYP-dependent arachidonic acid metabolism, and of arachidonic acid epoxygenation in particular, in avian than in mammalian species; (2) TCDD induces two CYP1A-related enzymes in birds as in mammals; (3) CYP1A enzymes in the birds other than chicks are not identical to CYP1A4 and 1A5 but share some enzymatic and immunochemical characteristics with them; (4) constitutive omega-OH arachidonic acid in all of the avian species and 8,9-EET in cormorant are formed by CYP enzymes unrelated to CYP1A; and (5) two distinct characteristics of avian CYP1A enzymes are the acquisition by avian CYP1A4-related P450 of unique epitope(s) and by CYP1A5-related P450 of unusual catalytic effectiveness for arachidonic acid epoxygenation.

The hepatic microsomal ethoxyresorufin O-deethylase (EROD)-inducing potency in ovo of 2,3,7,8-tetrachlorodibenzo-p-dioxin (TCDD) was determined in the domestic chicken (Gallus gallus), domestic pigeon (Columba livia), great blue heron, and double-crested cormorant. Dose-response curves were produced by injecting various doses of [3H]TCDD into the air sac of developing eggs during the latter third part of incubation. Hepatic EROD activities were measured in day-old hatchlings. Liver, yolk, and whole blood were analyzed for [3H]TCDD; no distributional differences among species were found. The ED50 for EROD induction was between one and two orders of magnitude lower in the chick (0.1 microgram/kg egg) than in the heron and cormorant (3-10 micrograms/kg egg). Consistent with this, the apparent affinity of TCDD for the hepatic cytosolic Ah receptor was about 15 times higher in the domestic chick (Kd = 0.75-1.6) than in the other avian species (pigeon, Kd = 11-14; heron, Kd = 10-20; cormorant, Kd = 12-16). Receptor binding affinities in the pigeon, heron, and cormorant were of the same order of magnitude as that reported for human placenta (D.K. Manchester, S.K. Gordon, C.L. Golas, E.A. Roberts, and A.B. Okey, 1987, Cancer Res. 47, 4861-4868). Subcutaneous edema was observed in TCDD-treated hatchlings of the chick, heron, and cormorant, but not of the pigeon, within the dose range examined. The laboratory dose-response relationships demonstrated that the heron and cormorant hatchlings that were exposed to TCDD and related chemicals in the Strait of Georgia (J.T. Sanderson, R.J. Norstrom, J.E. Elliott, L.E. Hart, K.M. Cheng, and G.D. Bellward (1994b) J. Toxicol. Environ. Health 41, 245-263; and J.T. Sanderson, J.E. Elliott, R.J. Norstrom, P.E. Whitehead, L.E. Hart, K.M. Cheng, and G.D. Bellward (1994a) J. Toxicol. Environ. Health 41, 435-450) had hepatic EROD activities at the lower end of the linear part of their respective dose-response curves. A further increase in levels of TCDD and related compounds in the environment would lead to a large increase in EROD activity and further increases in TCDD-induced toxicities, such as body weight loss and subcutaneous edema.

We compared the circadian oscillation of melatonin release from cultured pineal cells in Japanese quail, pigeon and house sparrow, to determine whether the pineal gland of these species retains the circadian oscillator function in vitro. After dissociated pineal cells have been cultured for 4 days under 12 h: 12 h light-dark (LD) cycle, they were perfused at a flow rate of 0.25 ml/h for 6-7 days under LD or constant darkness (DD). Melatonin release increased during the dark period and low during the light period in all pineal cell cultures. Under DD conditions, the circadian rhythm of melatonin release persisted for up to 3-4 cycles in pigeon and house sparrow pineal cells, but the amplitude of the rhythm decreased gradually. However, in Japanese quail pineal cell culture the circadian oscillation of melatonin release was weak or abolished under DD conditions. These results strengthen the argument that the avian pineal gland's role in circadian organization differs between species. The direct demonstration of species-specific differences in the mechanism of the circadian oscillation of melatonin release from pineal cells should provide a useful model for the analysis of the pineal's circadian system at the cellular level.

The melatonin-binding sites in membrane preparations of the bursa of Fabricius of birds were studied using [125I]iodomelatonin as a radioligand. The binding sites were stable, saturable, reversible and of high affinity. Scatchard analysis of specific binding revealed equilibrium binding constants (Kd) of (means +/- S.E.M.) 43.1 +/- 5.9, 73.3 +/- 7.6 and 35.3 +/- 4.8 pmol/l respectively at the mid-point of the light period (mid-light) in chickens, pigeons and quail, with a total number of binding sites (Bmax) of 1.23 +/- 0.15, 1.33 +/- 0.18 and 0.94 +/- 0.07 fmol/mg protein. The diurnal variation in [125I]iodomelatonin binding showed that the Bmax was 45, 115 and 70% higher at mid-light than at mid-dark in the bursae of chickens, pigeons and quail respectively. The Kd value determined from kinetic analysis was 49.0 +/- 6.4 pmol/l at mid-light in the chicken bursa. The [125I]iodomelatonin-binding sites of chicken bursal membranes had the following order of pharmacological affinities: 2-iodomelatonin > melatonin > 6-chloromelatonin > 6-hydroxymelatonin > N-acetyl-serotonin > 5-hydroxytryptamine, tryptamine, 5-methoxytryptophol, 1-acetylindole-3-carboxaldehyde, 3-acetylindole, L-tryptophan, 5-hydroxyindole-3-acetic acid, 5-hydroxytryptophan suggesting that the [125I]iodomelatonin-binding sites were highly specific for melatonin. The subcellular distribution of binding sites in the chicken bursa was in the following descending order: nuclear > mitochondrial > microsomal > cytosolic fraction. There was an age-related decrease in [125I]iodomelatonin-binding in chicken bursal membranes, with higher densities in the neonate.(ABSTRACT TRUNCATED AT 250 WORDS)

Alanine:glyoxylate aminotransferase has been reported to be present as the apo enzyme in the peroxisomes and as the holo enzyme in the mitochondria in chick (white leghorn) embryonic liver. However, surprisingly, birds were found to be classified into two groups on the basis of intraperoxisomal forms of liver alanine:glyoxylate aminotransferase. In the peroxisomes, the enzyme was present as the holo form in group 1 (pigeon, sparrow, Java sparrow, Australian budgerigar, canary, goose, and duck), and as the apo form in group 2 (white leghorn, bantam, pheasant, and Japanese mannikin). In the mitochondria, the enzyme was present as the holo form in both groups. The peroxisomal holo enzyme was purified from pigeon liver, and the peroxisomal apo enzyme from chicken (white leghorn) liver. The pigeon holo enzyme was composed of two identical subunits with a molecular weight of about 45,000, whereas the chicken apo enzyme was a single peptide with the same molecular weight as the subunit of the pigeon enzyme. The peroxisomal holo enzyme of pigeon liver was not immunologically cross-reactive with the peroxisomal apo enzyme of chicken liver, the mitochondrial holo enzymes from pigeon and chicken liver, and mammalian alanine:glyoxylate aminotransferases 1 and 2. The mitochondrial holo enzymes from both pigeon and chicken liver had molecular weights of about 200,000 with four identical subunits and were cross-reactive with mammalian alanine:glyoxylate aminotransferase 2 but not with mammalian alanine:glyoxylate aminotransferase 1.

A survey of rhodanese activity (thiosulphate: cyanide sulphur transferase) in the tissues of the domestic fowl revealed that the highest activity occurred in the kidney, approximately twice that in the liver, 316 and 141 mumol SCN formed/min g protein, respectively. 2. In sparrows, pigeons and ducks, liver and kidney activities tended to be similar and increased with body size. 3. Relationships were found between enzyme activity and body weight to a fractional power, indicating relatively higher activity in smaller birds.

The work was devoted to a histochemical studying of the covering lamina and other epithelial secretions of the avian muscular stomach. It has been shown that in its histochemical properties the secreted mucin is similar to mucous secretions of the enterodermal lining of the mammalian stomach and contains neutral polysaccharides, sulfo- and slilosaccharides. The protein component is represented by aminoacid residues of lysin, cystine, cystein. Formation of the mechanically firm structure--lamina is based on the substances of carbohydrate--protein nature secreted by the epithelial lining of the stomach.

Studies were conducted to establish the differences in yolk cholesterol concentrations in eggs from various avian species. Cholesterol was determined by a modification of the colorimetric procedure of Pearson et al. (1953). Species listed in increasing concentrations of cholesterol per gram of yolk, were guinea fowl, chicken, pheasant, quail, turkey, duck, goose, and dove with an overall range of 12.77 to 21.99 mg of cholesterol per gram of yolk. Significant differences in cholesterol concentrations also were found between domestic and wild genetic groups for turkeys and ducks. In a second study, eggs from 7 inbred lines of chickens showed significant differences in yolk cholesterol. The same general result was found in an analysis of eggs from 17 commercial test strains of chickens at two different ages. Yolk cholesterol tended to decrease as age of hen increased. These results confirm the possibility of genetically selecting for decreased yolk cholesterol if economic or other conditions warrant. The increased efforts required to decrease yolk cholesterol by this approach would undoubtedly be expensive.

Acetylcholinesterase (AChE) activity was studied in the glycogen bodies of the spinal cords of 2 birds namely Streptopelia senegalensis and Passer domesticus. A possible functional significance of AChE in the light of relative enzymatic localization especially in Hoffmann-Kolliker nuclei (motor cell groups), substantia gelatinosa and other regions of gray matter of 2 avian glycogen bodies has been discussed.

Tissue and plasma concentrations and the biological half-life of tylosin in avian species of a variety of body sizes and metabolic rates were studied. The species chosen were eastern bobwhite quail (Colinus virginianus virginianus), pigeons (Columba livia), greater sandhill cranes (Grus canadensis tabida), and emus (Dromaius novaehollandiae). In the 1st phase of this study, tylosin was administered IM to quail, pigeons, and emus at a dosage rate of 25 mg/kg of body weight and to cranes at a dosage rate of 15 mg/kg. The average peak plasma concentrations of tylosin in quail, pigeons, cranes, and emus were 4.31, 5.63, 3.62, and 3.26 microgram/ml, respectively. These peak concentrations occurred at 0.5 to 1.5 hours after administration. The biological half-life of tylosin averaged 1.2 hours in quail, pigeons, and cranes, and was 4.7 hours in emus. In the 2nd phase of this study, tylosin concentrations in the tissues of quail, pigeons, and cranes were markedly higher than were plasma concentrations at corresponding sampling times. Six hours after antibiotic administration, tissue concentrations of tylosin in all species remained within the minimum inhibitory concentration for most pathogenic organisms. Dosage regimens of 25 mg of tylosin/kg 4 times daily for quail and pigeons, 15 mg/kg 3 times daily for cranes, and 25 mg/kg 3 times daily for emus would be needed to establish and maintain therapeutic tissue concentrations.

Residues of BHC and DDT were estimated by gas-liquid chromatographic analysis of the internal body organs, depot fat, and blood plasma of a few species of Indian wild birds, captured in and around the urban area of Lucknow. Total BHC and gamma-BHC (lindane) levels were high in breast muscle, liver, heart, and lung tissues of pigeon, crow, and vulture, compared with the respective tissues of chicken, cattle egret, and kite. More lindane and total BHC was found in tissues of vulture compared with other species. The major part of BHC isomers in the brain of birds examined was accounted for by alpha-BHC. Total BHC detected in depot fat of crows was 29.7 ppm; lesser amounts were found in vulture, kite, and cattle egret, respectively. Total DDT levels were comparable in the blood plasma of chicken, pigeon, crow, and cattle egret, although residues generally showed the following order in the tissues examined: chicken less than pigeon less than cattle egret less than crow less than kite less than vulture. High levels of DDT were detected in depot fat of crow, kite, and vulture (50.8, 67.0 and 95.3 ppm, respectively). Avian species thus reflect biological magnification of BHC and DDT residues, presumably due to their food habits.

Plasma concentrations and the biological half-lives of cephalothin and cephalexin in avian species of a variety of body sizes and metabolic rates were studied. The species chosen were eastern bobwhite quail (Colinus v virginianus), pigeons (Columba livia), hybrid rosybill ducks (Netta sp), greater sandhill cranes (Grus canadensis tabida), and emus (Dromiceius novaehollandiae). In the 1st phase of the study, cephalothin sodium was given IM in a dose of 100 mg/kg of body weight. Plasma concentrations reached peak (av 18 micrograms/ml) at 0.5 hour and were measurable 2.5 to 5.5 hours after drug administration. The biological half-life of cephalothin was 16 to 54 minutes; the half-life varied directly with increased species body weight, with the exception of the ducks studied. In the 2nd phase, cephalexin monohydrate was given orally in doses of 25, 35, and 50 mg/kg of body weight. Plasma concentrations reached peak (av 20 micrograms/ml) at 0.5 to 1 hour and were measurable 2.5 to 5.5 hours after drug administration. The biological half-life of cephalexin was 36 to 126 minutes. In the 3rd phase, differences in plasma concentrations and the half-lives of cephalexin between fed quail and fasted quail were insignificant. Dosage regimens for cephalothin of 100 mg/kg 4 times a day and for cephalexin of 35 to 50 mg/kg 4 times a day would be expected to establish and maintain therapeutic plasma concentrations in large birds (pigeons, cranes, and emus). These same doses, administered every 2 to 3 hours, would be expected to establish and maintain therapeutic plasma concentrations in smaller birds (quail, ducks).

1. Benzo[a]pyrene hydroxylase, aminopyrine demethylase and glutathione S-transferase activities in hepatic and extrahepatic tissues of five wild birds, rat and mouse were compared. 2. Hepatic benzo[a]pyrene hydroxylase of wild pigeon was at least three times higher than that of rat or mouse. Hepatic aminopyrine demethylase and GSH S-transferase activities of wild birds were lower than those of rodents. 3. Renal GSH S-transferase of wild birds was 2-3 times more than in rat and mouse. 4. Avian benzo[a]pyrene hydroxylase activity of hepatic and renal tissue was linear up to 3.0 mg enzyme protein with pH optima of 7.4 and 7.2, at 42 degrees C. Apparent Km values were 11.76 and 3.33 microM respectively. 5. Hepatic enzyme activity was induced four-fold by 3-methylcholanthrene and about two-fold by phenobarbitone administration.

In the retina of vertebrates, carbonic anhydrase (CA) is present in the cells of the pigmented epithelial layer and in Muller cells. Its activity has been demonstrated to increase during development, corresponding to the maturity of the tissue. We have studied CA activity, either during embryonic development or after hatching, in the retina of 3 species of birds, having precocial or atricial broods. The research has been carried out, by histochemical and biochemical methods, in pigeon as an example of atricial brood, and in fowl and quail, that represent examples of precocial broods. Our results confirmed that CA activity may be taken as a marker of retina maturity, or, more precisely, of Muller cells maturity, for any species of vertebrates, depending solely on maturity of the offspring at birth time.

No data are available on the protamines of birds, with the exception of galline. We have characterized the protamines from four species of birds belonging to four different orders. All of them have very similar properties. They have been purified by carboxymethylcellulose chromatography and analyzed with respect to amino acid composition and electrophoretic behaviour. They are very arginine-rich proteins (63.4-67.3%) but do not contain lysine. Serine (12.0-18.2%), tyrosine (5.8-9.0%) and glycine (4.5-7.1%), along with arginine, make up the bulk of the amino acid residues in these molecules. The electrophoretic mobility of bird protamines in acetic acid-urea-polyacrylamide gels is intermediate between that of somatic histones and salmine. The molecular size, estimated from amino acid analysis and electrophoretic migration, is 65 +/- 5 amino acid residues.

Previous studies of cecal sugar and amino acid transport in the domestic chicken led to a widely held generalization that the avian cecum is unimportant as a site of nutrient transport. In fact, we found that the uptake capacity of the cecum for hexose sugars and amino acids is substantial in some species of birds. Cecal transport of glucose was measurable in all five study species (Canada goose, sage grouse, domestic chicken, red-necked phalarope, and rock dove), approached or exceeded intestinal levels in the grouse and phalarope, and accounted for between 0.1% (rock dove) and 49% (sage grouse) of the whole gut's integrated uptake capacity. Proline uptake averaged higher in the proximal portion of the cecum than in any region of the small intestine for all species but the goose. The ceca contributed between 2% (rock dove) and 25% (sage grouse) of the gut's integrated uptake capacity for proline. Similar ranges were found for fructose, lysine, leucine, and aspartate. Future studies should be undertaken to search for phylogenetic and ecological correlates of the interspecific variation in cecal transport and to determine how nutrient transport integrates with other functions of the avian cecum.
