Background and Purpose: The histamine H4 receptor is the most recently identified of the G protein-coupled histamine receptor family and binds several neuroactive drugs, including amitriptyline and clozapine. So far, H4 receptors have been found only on haematopoietic cells, highlighting its importance in inflammatory conditions. Here we investigated the possibility that H4 receptors may be expressed in both the human and mouse CNS., Methods: Immunological and pharmacological studies were performed using a novel anti-H4 receptor antibody in both human and mouse brains, and electrophysiological techniques in the mouse brain respectively. Pharmacological tools, selective for the H4 receptor and patch clamp electrophysiology, were utilized to confirm functional properties of the H4 receptor in layer IV of the mouse somatosensory cortex., Results: Histamine H4 receptors were prominently expressed in distinct deep laminae, particularly layer VI, in the human cortex, and mouse thalamus, hippocampal CA4 stratum lucidum and layer IV of the cerebral cortex. In layer IV of the mouse somatosensory cortex, the H4 receptor agonist 4-methyl histamine (20 micromol x L(-1)) directly hyperpolarized neurons, an effect that was blocked by the selective H4 receptor antagonist JNJ 10191584, and promoted outwardly rectifying currents in these cells. Monosynaptic thalamocortical CNQX-sensitive excitatory postsynaptic potentials were not altered by 4-methyl histamine (20 micromol x L(-1)) suggesting that H4 receptors did not act as hetero-receptors on thalamocortical glutamatergic terminals., Conclusions and Implications: This is the first demonstration that histamine H4 receptors are functionally expressed on neurons, which has major implications for the therapeutic potential of these receptors in neurology and psychiatry.

D2 dopamine-like receptors have been purified from five bovine brain regions (caudate nucleus, putamen, olfactory tubercle, frontal cortex, cerebellum) and the anterior and neurointermediate lobes of the pituitary gland using a combined ligand-affinity and lectin-affinity chromatography procedure. In all the brain regions except cerebellum and in the neurointermediate lobe of the pituitary gland the purified species appeared as a M(r) 95,000 doublet on SDS-PAGE. In the anterior lobe of the pituitary an additional M(r) 142,000-145,000 species was seen. The M(r) 95,000 species had a low affinity for the lectin wheat germ agglutinin (WGA) whereas the M(r) 142,000-145,000 species had a higher affinity for WGA and additionally showed some affinity for concanavalin A. It is concluded that both the M(r) 95,000 and 142,000-145,000 species are D2 dopamine-like receptors and that the differences between the species are mainly at the oligosaccharide level. Some evidence was also obtained for heterogeneity at the protein level which may correspond to the D2(short) and D2(long) isoforms of these receptors.

The rat NMDAR1 (N-methyl-D-aspartate receptor) was expressed transiently in human embryonic kidney cells. Transfected cell homogenates showed saturable [3H]MK-801 binding activity that was best fit by a single high-affinity site with a KD of 9 nM and a Bmax of 113 fmol of binding sites/mg of protein. Antibodies raised against the peptide sequence NMDAR1 (929-938) coupled to keyhole limpet haemocyanin specifically recognised a single band with M(r) 117,000 in immunoblots from adult rat brain. In the transfected cells, the antibody recognised two bands: one with M(r) 117,000, which was coincident with that from brain membranes, and one with M(r) 97,000, which was identified as nonglycosylated NMDAR1 subunit. These results identify the NMDAR1 of rat brain and further show that the homooligomer binds MK-801, albeit at low efficiency.

The effects of the thiol group reagent 5,5'-dithiobis-(2-nitrobenzoic acid) (DTNB) on D2 dopamine receptors have been examined in three brain regions (caudate nucleus, putamen and olfactory tubercle), and in the anterior and neurointermediate lobes of the pituitary gland. Whereas the receptors in brain were insensitive to DTNB, a dose-dependent inhibition of [3H]spiperone binding to D2 receptors in both lobes of the pituitary gland was observed. The effects in the pituitary could be reversed by subsequent treatment with dithiothreitol and could be prevented by prior occupancy of the receptor binding site with a ligand. The effect of DTNB was to decrease the number of ligand-binding sites without altering the affinity of those remaining. The results show that modification of a thiol group of the D2 dopamine receptor in the two lobes of the pituitary gland tested here significantly affects ligand binding. There is therefore a difference between the D2 dopamine receptor populations in brain and pituitary in their sensitivity to modification by DTNB, and this may imply the existence of different receptor isoforms in the two issues.

Summary: Cortical spreading depression (CSD) is a transient suppression of neuronal activity, resulting from a temporary disruption of local brain ionic homoeostasis by a propagating wave of neuronal hyperexcitability over the cerebral cortex. This activity has been shown to induce mechanisms which precondition the rodent brain structures affected by the CSD (protects them from the damaging effects of a stroke). A number of groups are using this strategy to identify the underlying protective mechanisms which will ultimately lead to the identification of new targets and strategies for neuroprotection in many acute and chronic neuropathologies. Mechanisms thus far identified include activation of adenosine A1 receptors which then activates the ATP-sensitive K+ (KATP) channels, and modulation of neuronal nicotinic receptors.
