Illusory figures demonstrate the visual system's ability to integrate disparate parts into coherent wholes. We probed this object integration process by either presenting an integrated diamond shape or a comparable ungrouped configuration that did not render a complete object. Two tasks were used that either required localization of a target dot (relative to the presented configuration) or discrimination of the dot's luminance. The results showed that only when the configuration was task relevant (in the localization task), performance benefited from the presentation of an integrated object. Concurrent functional magnetic resonance imaging was performed and analyzed using dynamic causal modeling to investigate the (causal) relationship between regions that are associated with illusory figure completion. We found object-specific feedback connections between the lateral occipital cortex (LOC) and early visual cortex (V1/V2). These modulatory connections persisted across task demands and hemispheres. Our results thus provide direct evidence that interactions between mid-level and early visual processing regions engage in illusory figure perception. These data suggest that LOC first integrates inputs from multiple neurons in lower-level cortices, generating a global shape representation while more fine-graded object details are then determined via feedback to early visual areas, independently of the current task demands., (© 2021 The Authors. Hemispatial neglect, after unilateral lesions to parietal brain areas, is characterized by an inability to respond to unexpected stimuli in contralesional space. As the visual field's horizontal meridian is most severely affected, the brain networks controlling visuospatial processes might be tuned explicitly to this axis. We investigated such a potential directional tuning in the dorsal and ventral frontoparietal attention networks, with a particular focus on attentional reorientation. We used an orientation-discrimination task where a spatial precue indicated the target position with 80% validity. Healthy participants (n = 29) performed this task in two runs and were required to (re-)orient attention either only along the horizontal or the vertical meridian, while fMRI and behavioral measures were recorded. By using a general linear model for behavioral and fMRI data, dynamic causal modeling for effective connectivity, and other predictive approaches, we found strong statistical evidence for a reorientation effect for horizontal and vertical runs. However, neither neural nor behavioral measures differed between vertical and horizontal reorienting. Moreover, models from one run successfully predicted the cueing condition in the respective other run. Our results suggest that activations in the dorsal and ventral attention networks represent higher-order cognitive processes related to spatial attentional (re-)orientating that are independent of directional tuning and that unilateral attention deficits after brain damage are based on disrupted interactions between higher-level attention networks and sensory areas., (© 2020 The Authors. The processing of congruent stimuli, such as an object or action in its typical location, is usually associated with reduced neural activity, probably due to facilitated recognition. However, in some situations, congruency increases neural activity-for example, when objects next to observed actions are likely versus unlikely to be involved in forthcoming action steps. Here, we investigated using fMRI whether the processing of contextual cues during action perception is driven by their (in)congruency and, thus, informative value to make sense of an observed scene. Specifically, we tested whether both highly congruent contextual objects (COs), which strongly indicate a future action step, and highly incongruent COs, which require updating predictions about possible forthcoming action steps, provide more anticipatory information about the action course than moderately congruent COs. In line with our hypothesis that especially the inferior frontal gyrus (IFG) subserves the integration of the additional information into the predictive model of the action, we found highly congruent and incongruent COs to increase bilateral activity in action observation nodes, that is, the IFG, the occipitotemporal cortex, and the intraparietal sulcus. Intriguingly, BA 47 was significantly stronger engaged for incongruent COs reflecting the updating of prediction in response to conflicting information. Our findings imply that the IFG reflects the informative impact of COs on observed actions by using contextual information to supply and update the currently operating predictive model. In the case of an incongruent CO, this model has to be reconsidered and extended toward a new overarching action goal.

Action recognition involves not only the readout of body movements and involved objects but also the integration of contextual information, e.g. the environment in which an action takes place. Notably, inferring superordinate goals and generating predictions about forthcoming action steps should benefit from screening the actor's immediate environment, in particular objects located in the actor's peripersonal space and thus potentially used in following action steps. Critically, if such contextual objects (COs) afford actions that are semantically related to the observed action, they may trigger or facilitate the inference of goals and the prediction of following actions. This fMRI study investigated the neural mechanisms underlying the integration of COs in semantic and spatial relation to observed actions. Specifically, we tested the hypothesis that the inferior frontal gyrus (IFG) subserves this integration. Participants observed action videos in which COs and observed actions had common overarching goals or not (goal affinity) and varied in their location relative to the actor. High goal affinity increased bilateral activity in action observation network nodes, i.e. the occipitotemporal cortex and the intraparietal sulcus, but also in the precuneus and middle frontal gyri. This finding suggests that the semantic relation between COs and actions is considered during action observation and triggers (rather than facilitates) processes beyond those usually involved in action observation. Moreover, COs with high goal affinity located close to the actor's dominant hand additionally engaged bilateral IFG, corroborating the view that IFG is critically involved in the integration of action steps under a common overarching goal., (Copyright © 2017 Elsevier Inc. Older individuals typically display stronger regional brain activity than younger subjects during motor performance. However, knowledge regarding age-related changes of motor network interactions between brain regions remains scarce. We here investigated the impact of ageing on the interaction of cortical areas during movement selection and initiation using dynamic causal modelling (DCM). We found that age-related psychomotor slowing was accompanied by increases in both regional activity and effective connectivity, especially for 'core' motor coupling targeting primary motor cortex (M1). Interestingly, younger participants within the older group showed strongest connectivity targeting M1, which steadily decreased with advancing age. Conversely, prefrontal influences on the motor system increased with advancing age, and were inversely correlated with reduced parietal influences and core motor coupling. Interestingly, higher net coupling within the prefrontal-premotor-M1 axis predicted faster psychomotor speed in ageing. Hence, as opposed to a uniform age-related decline, our findings are compatible with the idea of different age-related compensatory mechanisms, with an important role of the prefrontal cortex compensating for reduced coupling within the core motor network.

Background and Objective: Apraxia is a deficit of motor cognition leading to difficulties in actual tool use, imitation of gestures, and pantomiming object use. To date, little data exist regarding the recovery from apraxic deficits after stroke, and no statistical lesion mapping study investigated the neural correlate of recovery from apraxia. Accordingly, we here examined recovery from apraxic deficits, differential associations of apraxia task (imitation vs. pantomime) and effector (bucco-facial vs. limb apraxia) with recovery, and the underlying neural correlates., Methods: We assessed apraxia in 39 patients with left hemisphere (LH) stroke both at admission and approximately 11 days later. Furthermore, we collected clinical imaging data to identify brain regions associated with recovery from apraxic deficits using voxel-based lesion-symptom mapping (VLSM)., Results: Between the two assessments, a significant recovery from apraxic deficits was observed with a tendency of enhanced recovery of limb compared to bucco-facial apraxia. VLSM analyses revealed that within the lesion pattern initially associated with apraxia, lesions of the left insula were associated with remission of apraxic deficits, whereas lesions to the (inferior) parietal lobe (IPL; supramarginal and angular gyrus) and the superior longitudinal fasciculus (SLF) were associated with persistent apraxic deficits., Conclusions: Data suggest that lesions affecting the core regions (and white matter) of the fronto-parietal praxis network cause more persistent apraxic deficits than lesions affecting other regions (here: the left insula) that also contribute to motor cognition and apraxic deficits.

Humans flexibly attend to features or locations and these processes are influenced by the probability of sensory events. We combined computational modeling of response times with fMRI to compare the functional correlates of (re-)orienting, and the modulation by probabilistic inference in spatial and feature-based attention systems. Twenty-four volunteers performed two task versions with spatial or color cues. Percentage of cue validity changed unpredictably. A hierarchical Bayesian model was used to derive trial-wise estimates of probability-dependent attention, entering the fMRI analysis as parametric regressors. Attentional orienting activated a dorsal frontoparietal network in both tasks, without significant parametric modulation. Spatially invalid trials activated a bilateral frontoparietal network and the precuneus, while invalid feature trials activated the left intraparietal sulcus (IPS). Probability-dependent attention modulated activity in the precuneus, left posterior IPS, middle occipital gyrus, and right temporoparietal junction for spatial attention, and in the left anterior IPS for feature-based and spatial attention. These findings provide novel insights into the generality and specificity of the functional basis of attentional control. They suggest that probabilistic inference can distinctively affect each attentional subsystem, but that there is an overlap in the left IPS, which responds to both spatial and feature-based expectancy violations., (Copyright © 2016 Elsevier Inc. Joint attention, the shared attentional focus of at least two people on a third significant object, is one of the earliest steps in social development and an essential aspect of reciprocal interaction. However, the neural basis of joint attention (JA) in the course of development is completely unknown. The present study made use of an interactive eye-tracking paradigm in order to examine the developmental trajectories of JA and the influence of a familiar interaction partner during the social encounter. Our results show that across children and adolescents JA elicits a similar network of "social brain" areas as well as attention and motor control associated areas as in adults. While other-initiated JA particularly recruited visual, attention and social processing areas, self-initiated JA specifically activated areas related to social cognition, decision-making, emotions and motivational/reward processes highlighting the rewarding character of self-initiated JA. Activation was further enhanced during self-initiated JA with a familiar interaction partner. With respect to developmental effects, activation of the precuneus declined from childhood to adolescence and additionally shifted from a general involvement in JA towards a more specific involvement for self-initiated JA. Similarly, the temporoparietal junction (TPJ) was broadly involved in JA in children and more specialized for self-initiated JA in adolescents. Modelling psychophysical data using the Theory of Visual Attention (TVA) allows for a quantification of attentional sub-processes, such as the resolution of competition amongst multiple stimuli by top-down control signals for target selection (TVA-parameter α). This fMRI study investigated the neural correlates of α by comparing activity differences and changes of effective connectivity between conditions where a target was accompanied by a distractor or by a second target. Twenty-five participants performed a partial report task inside the MRI scanner. The left angular gyrus (ANG), medial frontal, and posterior cingulate cortex showed higher activity when a target was accompanied by a distractor as opposed to a second target. The reverse contrast yielded activation of a bilateral fronto-parietal network, the anterior insula, anterior cingulate cortex, and left inferior occipital gyrus. A psychophysiological interaction analysis revealed that the connectivity between left ANG and the left and right supramarginal gyrus (SMG), left anterior insula, and right putamen was enhanced in the target-distractor condition in participants with worse attentional top-down control. Dynamic causal modelling suggested that the connection from left ANG to right SMG during distractor presence was modulated by α. The human brain readily perceives fluent movement from static input. Using functional magnetic resonance imaging, we investigated brain mechanisms that mediate fluent apparent biological motion (ABM) perception from sequences of body postures. We presented body and nonbody stimuli varying in objective sequence duration and fluency of apparent movement. Three body postures were ordered to produce a fluent (ABC) or a nonfluent (ACB) apparent movement. This enabled us to identify brain areas involved in the perceptual reconstruction of body movement from identical lower-level static input. Participants judged the duration of a rectangle containing body/nonbody sequences, as an implicit measure of movement fluency. For body stimuli, fluent apparent motion sequences produced subjectively longer durations than nonfluent sequences of the same objective duration. This difference was reduced for nonbody stimuli. This body-specific bias in duration perception was associated with increased blood oxygen level-dependent responses in the primary (M1) and supplementary motor areas. Moreover, fluent ABM was associated with increased functional connectivity between M1/SMA and right fusiform body area. We show that perceptual reconstruction of fluent movement from static body postures does not merely enlist areas traditionally associated with visual body processing, but involves cooperative recruitment of motor areas, consistent with a "motor way of seeing"., (© The Author 2015. Conventional mass-univariate analyses have been previously used to test for group differences in neural signals. However, machine learning algorithms represent a multivariate decoding approach that may help to identify neuroimaging patterns associated with functional impairment in "individual" patients. We investigated whether fMRI allows classification of individual motor impairment after stroke using support vector machines (SVMs). Forty acute stroke patients and 20 control subjects underwent resting-state fMRI. Half of the patients showed significant impairment in hand motor function. Resting-state connectivity was computed by means of whole-brain correlations of seed time-courses in ipsilesional primary motor cortex (M1). Lesion location was identified using diffusion-weighted images. These features were used for linear SVM classification of unseen patients with respect to motor impairment. SVM results were compared with conventional mass-univariate analyses. Resting-state connectivity classified patients with hand motor deficits compared with controls and nonimpaired patients with 82.6-87.6% accuracy. Classification was driven by reduced interhemispheric M1 connectivity and enhanced connectivity between ipsilesional M1 and premotor areas. In contrast, lesion location provided only 50% sensitivity to classify impaired patients. Hence, resting-state fMRI reflects behavioral deficits more accurately than structural MRI. The mechanisms driving cortical plasticity in response to brain stimulation are still incompletely understood. We here explored whether neural activity and connectivity in the motor system relate to the magnitude of cortical plasticity induced by repetitive transcranial magnetic stimulation (rTMS). Twelve right-handed volunteers underwent functional magnetic resonance imaging during rest and while performing a simple hand motor task. Resting-state functional connectivity, task-induced activation, and task-related effective connectivity were assessed for a network of key motor areas. We then investigated the effects of intermittent theta-burst stimulation (iTBS) on motor-evoked potentials (MEP) for up to 25 min after stimulation over left primary motor cortex (M1) or parieto-occipital vertex (for control). ITBS-induced increases in MEP amplitudes correlated negatively with movement-related fMRI activity in left M1. Control iTBS had no effect on M1 excitability. Subjects with better response to M1-iTBS featured stronger preinterventional effective connectivity between left premotor areas and left M1. In contrast, resting-state connectivity did not predict iTBS aftereffects. Plasticity-related changes in M1 following brain stimulation seem to depend not only on local factors but also on interconnected brain regions. Motor skills are mediated by a dynamic and finely regulated interplay of the primary motor cortex (M1) with various cortical and subcortical regions engaged in movement preparation and execution. To date, data elucidating the dynamics in the motor network that enable movements at different levels of behavioral performance remain scarce. We here used functional magnetic resonance imaging (fMRI) and dynamic causal modeling (DCM) to investigate effective connectivity of key motor areas at different movement frequencies performed by right-handed subjects (n=36) with the left or right hand. The network of interest consisted of motor regions in both hemispheres including M1, supplementary motor area (SMA), ventral premotor cortex (PMv), motor putamen, and motor cerebellum. The connectivity analysis showed that performing hand movements at higher frequencies was associated with a linear increase in neural coupling strength from premotor areas (SMA, PMv) contralateral to the moving hand and ipsilateral cerebellum towards contralateral, active M1. In addition, we found hemispheric differences in the amount by which the coupling of premotor areas and M1 was modulated, depending on which hand was moved. Other connections were not modulated by changes in motor performance. We used fMRI to investigate both common and differential neural mechanisms underlying two distinct types of switching requirements, namely switching between stimulus categorizations (color vs. form) and switching between response modalities (hand vs. foot responses). Both types of switching induced similar behavioral shift costs. However, at the neural level, switching between stimulus categorizations led to left-hemispheric activations including the inferior frontal gyrus as well as the intraparietal sulcus extending to the superior parietal gyrus and the supramarginal gyrus. In contrast, switching between response modalities was associated mainly with left-hemispheric activation of the intraparietal sulcus and the supramarginal gyrus. A conjunction analysis indicated common activation of the left intraparietal sulcus and the supramarginal gyrus for both types of switching. Together, these results qualify previous claims about a general role of the left prefrontal cortex in task control by suggesting that the left inferior frontal gyrus is specifically involved in switching between stimulus categorizations, whereas parietal cortex is more generally implicated in the selection of action rules., (Copyright © 2012 Wiley Periodicals, Inc.)

Individuals with autism spectrum disorder (ASD) often fail to attach context to their memories and are specifically impaired in processing social aspects of contextual information. The aim of the present study was to investigate the modulatory influence of social vs. non-social context on neural mechanisms during encoding in ASD. Using event-related fMRI, 13 boys with ASD and 13 typically developing boys comparable for age and IQ were investigated during encoding of neutral objects presented either with a social (faces) or a non-social (houses) context. A memory paradigm was then applied to identify brain activation patterns associated with encoding of subsequently recollected versus non-recollected objects. On the behavioural level, no significant between-group differences emerged. In particular, no differential effects of context on memory performance were observed. Neurally, however, context-specific group differences were observed in several brain regions. During encoding of subsequently recollected objects presented with a face, ASD subjects (compared to controls) showed reduced neural activation in the bilateral inferior frontal gyrus, bilateral middle frontal gyrus and right inferior parietal lobule. Neural activation in the right inferior frontal gyrus was positively correlated with memory performance in controls, but negatively in ASD individuals. During encoding of subsequently non-recollected objects presented in the non-social context, ASD subjects showed increased activation in the dorsal MPFC. Our findings suggest that in ASD subjects, fronto-parietal brain regions subserving memory formation and the association of contextual information are activated atypically when a social context is presented at encoding. The data add to findings from related research fields indicating that in ASD, socioemotional impairment extends into domains beyond social cognition. Previous brain imaging studies identified the neural networks underlying free choice of self-initiated actions. In contrast, the neural mechanisms underlying free choice of external objects remain to be elucidated. In this event-related functional magnetic resonance imaging study, participants had to choose one out of two different single-colored target squares presented at two of three possible locations. In 50% of the trials the choice was either free or specified, which was indicated by a preceding cue. In order to disentangle processes associated with object choice from those related to motor responses, object-response mapping was orthogonally varied. Processes related to the freedom of choice were isolated by means of an adaptive algorithm: based on the subjects individual choices in the free trials, specified trials were continuously generated in a way that matched the free trials in all aspects but the freedom to choose the object. Comparing free and specified trials revealed enhanced neural activity of a bilateral symmetrical network including the dorsolateral prefrontal, medial frontal, and medial parietal cortex. This network overlaps with that shown previously to be associated with free motor selection. It includes the pre-supplementary motor area (pre-SMA), suggesting that this area is rather associated with a supramodal function of initiating choice than with a specific motor function. Neural activity specifically associated with the freedom to choose an object was found bilaterally in the lateral superior parietal cortex. How the human brain reconstructs the three-dimensional (3D) world from two-dimensional (2D) retinal images has received a great deal of interest as has how we shift attention in 2D space. In contrast, it remains poorly understood how visuospatial attention is shifted in depth. In this fMRI study, by constructing a virtual 3D environment in the MR scanner and by presenting targets either close to or far from the participants in an adapted version of the Posner spatial-cueing paradigm, we investigated the behavioral and neural mechanisms underlying visuospatial orienting/reorienting in depth. At the behavioral level, although covering the same spatial distance, attentional reorienting to objects unexpectedly appearing closer to the observer and in the unattended hemispace was faster than reorienting to unexpected objects farther away. At the neural level, we found that in addition to the classical attentional reorienting system in the right temporoparietal junction, two additional brain networks were differentially involved in aspects of attentional reorienting in depth. First, bilateral premotor cortex reoriented visuospatial attention specifically along the third dimension of visual space (i.e., from close to far or vice versa), compared with attentional reorienting within the same depth plane. Second, a network of areas reminiscent of the human "default-mode network," including posterior cingulate cortex, orbital prefrontal cortex, and left angular gyrus, was involved in the neural interaction between depth and attentional orienting, by boosting attentional reorienting to unexpected objects appearing both closer to the observer and in the unattended hemispace.

Attentional orientation to a spatial cue and reorientation-after invalid cueing-are mediated by two distinct networks in the human brain. A bilateral dorsal frontoparietal network, comprising the intraparietal sulcus (IPS) and the frontal eye fields (FEF), controls the voluntary deployment of attention and may modulate visual cortex in preparation for upcoming stimulation. In contrast, reorienting attention to invalidly cued targets engages a right-lateralized ventral frontoparietal network comprising the temporoparietal junction (TPJ) and ventral frontal cortex. The present fMRI study investigated the functional architecture of these two attentional systems by characterizing effective connectivity during lateralized orienting and reorienting of attention, respectively. Subjects performed a modified version of Posner's location-cueing paradigm. Dynamic causal modeling (DCM) of regional responses in the dorsal and ventral network, identified in a conventional (SPM) whole-brain analysis, was used to compare different functional architectures. Bayesian model selection showed that top-down connections from left and right IPS to left and right visual cortex, respectively, were modulated by the direction of attention. Moreover, model evidence was highest for a model with directed influences from bilateral IPS to FEF, and reciprocal coupling between right and left FEF. Invalid cueing enhanced forward connections from visual areas to right TPJ, and directed influences from right TPJ to right IPS and IFG (inferior frontal gyrus). These findings shed further light on the functional organization of the dorsal and ventral attentional network and support a context-sensitive lateralization in the top-down (backward) mediation of attentional orienting and the bottom-up (forward) effects of invalid cueing.

Studying cognitive processes underlying synesthesia, a condition in which stimulation of one sensory modality automatically leads to abnormal additional sensory perception, allows insights into the neural mechanisms of normal and abnormal cross-modal sensory processing. Consistent with the notion that synesthesia results from hyperconnectivity, functional connectivity analysis (adopting independent component analysis and seed-based correlation analysis) of resting-state functional magnetic resonance imaging data of 12 grapheme-color synesthetes and 12 nonsynesthetic control subjects revealed, in addition to increased intranetwork connectivity, both a global and a specific (medial and lateral visual networks to a right frontoparietal network) increase of intrinsic internetwork connectivity in grapheme-color synesthesia. Moreover, this increased intrinsic network connectivity reflected the strength of synesthetic experiences. These findings constitute the first direct evidence of increased functional network connectivity in synesthesia. In addition to this significant contribution to the understanding of the neural mechanisms of synesthesia, our results have important general implications. In combination with data derived from clinical populations, our data strongly suggest that altered differences in intrinsic network connectivity are directly related to the phenomenology of human experiences.

Functional magnetic resonance imaging (fMRI) and transcranial magnetic stimulation (TMS) are well-established tools for investigating the human motor system in-vivo. We here studied the relationship between movement-related fMRI signal changes in the primary motor cortex (M1) and electrophysiological properties of the hand motor area assessed with neuronavigated TMS in 17 healthy subjects. The voxel showing the highest task-related BOLD response in the left hand motor area during right hand movements was identified for each individual subject. This fMRI peak voxel in M1 served as spatial target for coil positioning during neuronavigated TMS. We performed correlation analyses between TMS parameters, BOLD signal estimates and effective connectivity parameters of M1 assessed with dynamic causal modeling (DCM). The results showed a negative correlation between the movement-related BOLD signal in left M1 and resting as well as active motor threshold (MT) obtained for left M1. The DCM analysis revealed that higher excitability of left M1 was associated with a stronger coupling between left supplementary motor area (SMA) and M1. Furthermore, BOLD activity in left M1 correlated with ipsilateral silent period (ISP), i.e. the stronger the task-related BOLD response in left M1, the higher interhemispheric inhibition effects targeting right M1. DCM analyses revealed a positive correlation between the coupling of left SMA with left M1 and the duration of ISP. The data show that TMS parameters assessed for the hand area of M1 do not only reflect the intrinsic properties at the stimulation site but also interactions with remote areas in the human motor system., (Copyright © 2011 Wiley-Liss, Inc.)

Empirical evidence for an essential role of the hippocampal system in arbitrary visuo-motor mapping suggests that acquisition and retrieval of arbitrary visuo-motor mapping might be impaired in mild cognitive impairment (MCI) and Alzheimer's disease (AD). The present pilot study investigated whether MCI of amnestic type or AD impact upon the capacity to scale grip force in a predictive manner to the mass of an object to be lifted based on learned associations between arbitrary colour cues and mass. Patients with MCI (n=8) and AD (n=8) grasped and lifted two different masses (400g and 600g) in random order using a precision grip between index finger and thumb. In a "no cue" experiment, a non-informative neutral visual stimulus was presented prior to each lift, thereby disallowing any prediction about which of the two masses was going to be lifted in the next trial. In a "cue" experiment an arbitrary colour cue provided advance information about which of the two masses to be lifted. In the "no cue" condition patients scaled their grip force according to the mass of the preceding lift. In the "cue" experiment neither patients with amnestic MCI nor those with AD were able to adjust their grip force based on visuo-motor mappings with arbitrary colour cues. In the past decade several multivariate causality measures based on Granger causality have been suggested to assess directionality of neural signals. To date, however, a detailed evaluation of the reliability of these measures is largely missing. We systematically evaluated the performance of five different causality measures (squared partial directed coherence (sPDC), partial directed coherence (PDC), directed transfer function (DTF), direct directed transfer function (dDTF) and transfer function) depending upon data length, noise level, coupling strength, and model order and performed simulations based on four different neural data recording procedures (magnetoencephalography, electroencephalography, electromyography, intraoperative local field potentials). Moreover, we analyzed the effect of two common numerical methods to determine the significance of the particular causality measure (random permutation and the leave one out method (LOOM)). The simulations showed the sPDC combined with the LOOM to be the most reliable and robust choice for assessing directionality in neural data. While DTF and H by construction were unable to distinguish between direct and indirect connections, the dDTF also failed this test. Finally, we applied the causality measures to a real data set. This showed the usefulness of our simulation results for practical applications in order to draw correct inferences and distinguish between conflicting evidence obtained with different causality measures., (Copyright © 2011 Elsevier B.V. The motor system comprises a network of cortical and subcortical areas interacting via excitatory and inhibitory circuits, thereby governing motor behaviour. The balance within the motor network may be critically disturbed after stroke when the lesion either directly affects any of these areas or damages-related white matter tracts. A growing body of evidence suggests that abnormal interactions among cortical regions remote from the ischaemic lesion might also contribute to the motor impairment after stroke. Here, we review recent studies employing models of functional and effective connectivity on neuroimaging data to investigate how stroke influences the interaction between motor areas and how changes in connectivity relate to impaired motor behaviour and functional recovery. Based on such data, we suggest that pathological intra- and inter-hemispheric interactions among key motor regions constitute an important pathophysiological aspect of motor impairment after subcortical stroke. We also demonstrate that therapeutic interventions, such as repetitive transcranial magnetic stimulation, which aims to interfere with abnormal cortical activity, may correct pathological connectivity not only at the stimulation site but also among distant brain regions. In summary, analyses of connectivity further our understanding of the pathophysiology underlying motor symptoms after stroke, and may thus help to design hypothesis-driven treatment strategies to promote recovery of motor function in patients.

Stroke may trigger a number of cellular and molecular events in perilesional and remote brain regions enabling cortical reorganization and recovery of function. We here investigated the pattern and time course of acute stroke-induced changes in motor system activity during motor recovery using functional magnetic resonance imaging. Hand movement-related neural activity was assessed in 11 acute stroke patients scanned 3 times during the first 2 weeks starting within 72 h after symptom onset. A motor recovery score was computed based on the action research arm test and the maximum grip force. Increases of activity in primary motor cortex, premotor cortex (dorsal and ventral), and supplementary motor area in both hemispheres significantly correlated with behavioral recovery. These longitudinal changes depended upon the degree of initial motor impairment: Patients with mild deficits did not differ from healthy subjects. In contrast, patients with severe deficits were characterized by a global reduction of task-related activity, followed by increases in ipsilesional as well as contralesional motor areas. The finding that the gradually increasing activity in contralesional primary motor and premotor cortex correlated with improved functional recovery in severely affected patients indicates early cortical reorganization supporting motor function of the affected hand.

Besides the fact that RTs in cognitive tasks are affected by the specific demands of a trial, the context in which this trial occurs codetermines the speed of the response. For instance, invalid spatial cues generally prolong RTs to targets in the location-cueing paradigm, whereas the magnitude of these RT costs additionally varies as a function of the preceding trial types so that RTs for invalid trials may be increased when preceded by valid rather than invalid trials. In the present fMRI study, we investigated trial sequence effects in a combined oddball and location-cueing paradigm. In particular, we tested whether RTs and neural activity to infrequent invalid or deviant targets varied as a function of the number of preceding valid standard trials. As expected, RTs in invalid and deviant trials were significantly slower when more valid standard trials had been presented beforehand. This behavioral effect was reflected in the neural activity of the right inferior/middle frontal gyrus where the amplitude of the hemodynamic response in invalid and deviant trials was positively related to the number of preceding valid standard trials. In contrast, decreased activity (i.e., a negative parametric modulation effect) was observed when more valid standard trials were successively presented. Further positive parametric effects for the number of preceding valid standard trials were observed in the left caudate nucleus and lingual gyrus. The data suggest that inferior frontal cortex extracts both event regularities and irregularities in event streams.

Cognitive impairment is a common and largely undiagnosed finding in a significant number of dialysis patients. These alterations may result from concomitant cerebrovascular disease, hemodynamic instability, the uremic milieu, or changes induced by the dialysis process. In order to gain further insight into this, we recruited 12 stable chronic hemodialysis patients (without clinical neurological disease) and an age- and gender-matched cohort of 12 control individuals (without renal or neurological problems) in a prospective, single-center study. In order to disentangle the influence of dialysis itself on memory function, each dialysis patient was tested twice: once immediately before dialysis following a long weekend (t1) and again the day after this dialysis (t2). The control individuals were tested in the same time frame. Neuropsychological testing found that the control individuals performed significantly better in verbal learning, motor speed, task switching, verbal comprehension, word fluency, spatial visualization, spatial perception, and reasoning; all independent of the time point. Functional magnetic resonance imaging of the whole brain in seven hemodialysis patients found significantly more bilateral activation of the hippocampus during the verbal working memory task at t2 relative to t1 compared with their seven matched control counterparts. Thus, our study found differential and task-specific activation of memory-relevant brain areas during a dialysis cycle.

Autobiographical memories enable us to mentally reconstruct and relive past events, which is essential for one's personal identity. Unfortunately, this complex memory system is susceptible to age-related deterioration, possibly changing the way episodic information is being processed in older adults. The aim of this study was to investigate whether age influences the neural activity associated with content (episodic versus semantic) and remoteness (recent versus remote) of memories. Using functional magnetic resonance imaging in healthy older and young adults, we found significant age-dependent differences in the neural networks underlying memory content but not remoteness. Our data suggest an age-associated functional reorganization in the neural networks underlying long-term declarative memory. Relative increase in activity of posterior brain regions could reflect changes in visuospatial processing during episodic memory retrieval in older adults.

Although empathy is rooted early in life, the ability to understand and share the emotions of others continues to develop after childhood. Here, we aimed at exploring developmental changes in the neural mechanisms underlying empathy from childhood to early adulthood. Using functional magnetic resonance imaging, 47 healthy male subjects aged 8-27 years were investigated during an explicit empathy task. Emotional faces were presented and participants were either asked to infer the emotional state from the face (other-task) or to judge their own emotional response to the face (self-task). A perceptual decision on the width of faces was used as a control condition. Age-related activity increases were observed in the fusiform gyrus and inferior frontal gyrus, depending on whether subjects attributed emotions to self or other. During the self-task, activity in the right precuneus and right intraparietal sulcus decreased as a function of age. No age-related differences were observed in behavioral performance measures. Increased activity in the fusiform gyrus and in the frontal component of the human mirror neuron system with increasing age may be explained by greater experience and expertise accumulated during socio-emotional interactions. Greater recruitment of right parietal structures in younger as compared to older subjects might reflect developmental differences in the cognitive strategies to infer one's own emotional response. This study is the first to show developmental changes in the neural mechanisms supporting empathy. Our findings may have important implications for the development of novel therapeutic interventions in clinical conditions characterized by empathy deficits, such as autism spectrum disorder.

A deficit in global motion processing caused by a specific dysfunction of the visual dorsal pathway has been suggested to underlie perceptual abnormalities in subjects with autism spectrum disorders (ASD). However, the neural mechanisms associated with abnormal motion processing in ASD remain poorly understood. We investigated brain responses related to the detection of coherent and random motion in 15 male subjects with ASD and 15 age- and IQ-matched healthy controls (aged 13-19 years) using event-related functional magnetic resonance imaging (fMRI). Behaviorally, no significant group differences were observed between subjects with ASD and controls. Neurally, subjects with ASD showed increased brain activation in the left primary visual cortex across all conditions compared with controls. A significant interaction effect between group and condition was observed in the right superior parietal cortex resulting from increased neural activity in the coherent compared with the random motion conditions only in the control group. In addition, neural activity in area V5 was not differentially modulated by specific motion conditions in subjects with ASD. Functional connectivity analyses revealed positive correlations between the primary visual cortex and area V5 within both hemispheres, but no significant between-group differences in functional connectivity patterns along the dorsal stream. The data suggest that motion processing in ASD results in deviant activations in both the lower and higher processing stages of the dorsal pathway. In the past, causality measures based on Granger causality have been suggested for assessing directionality in neural signals. In frequency domain analyses (power or coherence) of neural data, it is common to preprocess the time series by filtering or decimating. However, in other fields, it has been shown theoretically that filtering in combination with Granger causality may lead to spurious or missed causalities. We investigated whether this result translates to multivariate causality methods derived from Granger causality with (a) a simulation study and (b) an application to magnetoencephalographic data. To this end, we performed extensive simulations of the effect of applying different filtering techniques and evaluated the performance of five different multivariate causality measures in combination with two numerical significance measures (random permutation and leave one out method). The analysis included three of the most widely used filters (high-pass, low-pass, notch filter), four different filter types (Butterworth, Chebyshev I and II, elliptic filter), variation of filter order, decimating and interpolation. The simulation results suggest that preprocessing without a strong prior about the artifact to be removed disturbs the information content and time ordering of the data and leads to spurious and missed causalities. Only if apparent artifacts like a current or movement artifact are present, filtering out the respective disturbance seems advisable. While oversampling poses no problem, decimation by a factor greater than the minimum time shift between the time series may lead to wrong inferences. The field of social neuroscience has made remarkable progress in elucidating the neural mechanisms of social cognition. More recently, the need for new experimental approaches has been highlighted that allow studying social encounters in a truly interactive manner by establishing 'online' reciprocity in social interaction. In this article, we present a newly developed adaptation of a method which uses eyetracking data obtained from participants in real time to control visual stimulation during functional magnetic resonance imaging, thus, providing an innovative tool to generate gaze-contingent stimuli in spite of the constraints of this experimental setting. We review results of two paradigms employing this technique and demonstrate how gaze data can be used to animate a virtual character whose behavior becomes 'responsive' to being looked at allowing the participant to engage in 'online' interaction with this virtual other in real-time. Possible applications of this setup are discussed highlighting the potential of this development as a new 'tool of the trade' in social cognitive and affective neuroscience.

The human amygdala plays a pivotal role in the processing of socially significant information. Anatomical studies show that the human amygdala is not a single homogeneous structure but is composed of segregable subregions. These have recently been functionally delineated by using a combination of functional magnetic resonance imaging (fMRI) and cytoarchitectonically defined probabilistic maps. However, the response characteristics and individual contribution of these subregions to the processing of social-emotional stimuli are little understood. Here, we used this novel technique to segregate intra-amygdalar responses to facial expressions and nonsocial control stimuli. We localized facial expression-evoked signal changes bilaterally in the superficial amygdala, which suggests that this subregion selectively extracts the social value of incoming sensory information.

This study disentangles the prefrontal network underlying executive functions involved in the Wisconsin Card Sorting Test (WCST). During the WCST, subjects have to perform two key processes: first, they have to derive the correct sorting rule for each trial by trial-and-error, and, second, they have to detect when this sorting rule is changed by the investigator. Both cognitive processes constitute key components of the executive system, which is subserved by the prefrontal cortex. For the current fMRI experiment, we developed a non-verbal variant of the WCST. Subjects were instructed either to respond according to a given sorting rule or to detect the correct sorting rule, like in the original version of the WCST. Data were obtained from 14 healthy male volunteers and analysed using SPM and a random effects model. All conditions activated a fronto-parietal network, which was generally more active when subjects had to search for the correct sorting rule than when the rule was announced beforehand. Significant differences between these two conditions were seen in the dorsolateral prefrontal cortex (PFC) and the parietal lobe. In addition, the data provided new evidence for the assumption of differentiated roles of the left and right prefrontal cortex. Voluntary orienting of visual spatial attention has been shown to be associated with activation in a distributed network of frontal and parietal brain areas. Neuropsychological data suggest that at least some of these areas should be sensitive to the direction in which attention is shifted. The aim of this study was to use rapid event-related functional magnetic resonance imaging to investigate whether spatial attention in the auditory modality is subserved by the same or different brain areas as in the visual modality, and whether the auditory and visual attention networks show any degree of hemispheric lateralisation or sensitivity to the direction of attention shifts. The results suggest that auditory and visual spatial attention shifts are controlled by a supramodal network of frontal, parietal and temporal areas. Areas activated included the precuneus and superior parietal cortex, the inferior parietal cortex and temporo-parietal junction, as well as the premotor and supplementary motor areas and dorsolateral prefrontal cortex (DLPFC). In the auditory task, some of these areas, in particular the precuneus as well as the inferior parietal cortex and temporo-parietal junction, showed 'relative' asymmetry, in that they responded more strongly to attention shifts towards the contralateral than the ipsilateral hemispace. Neuropsychological studies suggest that the human brain is endowed with two body representations: the body schema (BS), coding the orientation of one's body parts in space, and the body structural description (BSD), coding the location of body parts relative to a standard body. We used fMRI to disentangle the neural mechanisms underlying these putatively distinct body representations. Participants saw an arm or a pot's handle (stimulus: arm, handle) rotated at different angles (angle: 30-150 degrees). If the stimulus was an arm, subjects were instructed to imagine (1) rotating their own arm until it matched the stimulus orientation (comparing the seen arm to their own) or (2) seeing the stimulus moving toward its appropriate position on a simultaneously presented human body [comparing the arm to the one of a standard body (strategy: motor, visual imagery)]. If the stimulus was a handle, subjects were instructed to imagine (1) placing the handle on its appropriate position on a simultaneously presented pot or (2) seeing it moving toward its pot's position. The analysis of the interaction stimulus x strategy revealed activation of left secondary somatosensory cortex (SII), specifically when comparing the stimulus arm to one's own. The analysis of the parameters describing the linear effect of angle revealed that neural activity of left posterior intraparietal sulcus was modulated by the stimulus's rotation, but only when relating the arm to a standard body. The results associate BS and BSD with differential neural substrates, thereby suggesting that these are independent body representations, and furthermore extend current concepts of SII function.

Within the parietal cortex, the temporo-parietal junction (TPJ) and the intraparietal sulcus (IPS) seem to be involved in both spatial and nonspatial functions: Both areas are activated when misleading information is provided by invalid spatial cues in Posner's location-cueing paradigm, but also when infrequent deviant stimuli are presented within a series of standard events. In the present study, we used functional magnetic resonance imaging to investigate the distinct and shared brain responses to (i) invalidly cued targets requiring attentional reorienting, and (ii) to target stimuli deviating in color and orientation leading to an oddball-like distraction effect. Both unexpected location and feature changes were accompanied by a significant slowing of manual reaction times. Bilateral TPJ and right superior parietal lobe (SPL) activation was observed in response to invalidly as compared to validly cued targets. In contrast, the bilateral inferior occipito-temporal cortex, the left inferior parietal cortex, right frontal areas, and the cerebellum showed stronger activation in response to deviant than to standard targets. Common activations were observed in the right angular gyrus along the IPS and in the right inferior frontal gyrus. We conclude that the superior parietal and temporo-parietal activations observed here as well as previously in location-cueing paradigms do not merely reflect the detection and processing of unexpected stimuli. Furthermore, our data suggest that the right IPS and the inferior frontal gyrus are involved in attentional selection and distractor processing of both spatial and nonspatial features.

In humans, transcranial direct current stimulation (tDCS) can be used to induce, depending on polarity, increases or decreases of cortical excitability by polarization of the underlying brain tissue. Cognitive enhancement as a result of tDCS has been reported. The purpose of this study was to test whether weak tDCS (current density, 57 microA/cm(2)) can be used to modify language processing. Fifteen healthy subjects performed a visual picture naming task before, during and after tDCS applied over the posterior perisylvian region (PPR), i.e. an area which includes Wernicke's area [BA 22]. Four different sessions were carried out: (1) anodal and (2) cathodal stimulation of left PPR and, for control, (3) anodal stimulation of the homologous region of the right hemisphere and (4) sham stimulation. We found that subjects responded significantly faster following anodal tDCS to the left PPR (p<0.01). No decreases in performance were detected. Our finding of a transient improvement in a language task following the application of tDCS together with previous studies which investigated the modulation of picture naming latency by transcranial magnetic stimulation (TMS) and repetitive TMS (rTMS) suggest that tDCS applied to the left PPR (including Wernicke's area [BA 22]) can be used to enhance language processing in healthy subjects. Whether this safe, low cost, and easy to use brain stimulation technique can be used to ameliorate deficits of picture naming in aphasic patients needs further investigations.

Purpose: To investigate the neural correlates associated with recovery from acute spatial neglect resulting from right hemispheric stroke., Methods: Four neglect patients were investigated both behaviourally and by fMRI at an acute (18 +/- 5 days) and at a chronic stage (123 +/- 18 days) post stroke., Results: At the second assessment all patients showed substantial behavioural improvements. These were associated with an increase of neural activity in the right middle frontal gyrus, right inferior parietal cortex, right inferior temporal gyrus/fusiform gyrus, left superior temporal gyrus/angular gyrus and left anterior cingulate gyrus. Decreased neural activity at the second assessment was found in the right parahippocampal gyrus and left fusiform gyrus., Conclusions: The pattern of neural reorganisation comprises areas of a right hemisphere fronto-parietal attentional network and corresponding left hemisphere areas suggesting a compensatory recruitment of analogous contralesional areas. Interestingly, a more complex pattern of neural changes was observed in the fusiform gyri which have previously been implicated in lateralised directed spatial attention. There was an increase in the right hemisphere and a decrease in the left hemisphere. This pattern of recovery is reminiscent of a "push-pull" pattern previously described for the dorsal parietal cortex by Corbetta et al. (2005) in the recovery from spatial neglect.

Research investigating risk perception suggests that not only the quantitative parameters used in technical risk assessment (i.e., frequency and severity of harm) but also 'qualitative' aspects such as the dread a hazard provokes or its controllability influence risk judgments. It remains to be elucidated, however, which neural mechanism underlie risk ratings in healthy subjects. Using fMRI to detect changes in neural activity we compared the neural activations elicited by risk ratings with those elicited by a letter detection task performed on the same stimuli. The latter task served to control for basic stimulus processing, response selection and button-pressing during task performance. Risk ratings differentially activated the medial prefrontal cortex, the inferior frontal gyrus, the cerebellum (P<0.05, FWE corrected, whole brain approach), and in an additional ROI analysis the amygdala (P<0.05, FWE corrected). Of these structures, particularly the amygdala and the prefrontal cortex have been previously associated with decisions about affective interference. Furthermore our data suggest both, similarities and differences between the neural correlates of risk ratings and risk taking as involved, for e.g., in gambling tasks.

Attending to a visual stimulus feature, such as color or motion, enhances the processing of that feature in the visual cortex. Moreover, the processing of the attended object's other, unattended, features is also enhanced. Here, we used functional magnetic resonance imaging to show that attentional modulation in the auditory system may also exhibit such feature- and object-specific effects. Specifically, we found that attending to auditory motion increases activity in nonprimary motion-sensitive areas of the auditory cortical "where" pathway. Moreover, activity in these motion-sensitive areas was also increased when attention was directed to a moving rather than a stationary sound object, even when motion was not the attended feature. An analysis of effective connectivity revealed that the motion-specific attentional modulation was brought about by an increase in connectivity between the primary auditory cortex and nonprimary motion-sensitive areas, which, in turn, may have been mediated by the paracingulate cortex in the frontal lobe. The current results indicate that auditory attention can select both objects and features. The finding of feature-based attentional modulation implies that attending to one feature of a sound object does not necessarily entail an exhaustive processing of the object's unattended features.

Impaired retrieval of conceptual knowledge for actions has been associated with lesions of left premotor, left parietal, and left middle temporal areas [Tranel, D., Kemmerer, D., Adolphs, R., Damasio, H., & Damasio, A. R. Neural correlates of conceptual knowledge for actions. Cognitive Neuropsychology, 409-432, 2003]. Here we aimed at characterizing the differential contribution of these areas to the retrieval of conceptual knowledge about actions. During functional magnetic resonance imaging (fMRI), different categories of pictograms (whole-body actions, manipulable and nonmanipulable objects) were presented to healthy subjects. fMRI data were analyzed using SPM2. A conjunction analysis of the neural activations elicited by all pictograms revealed ( p<.05, corrected) a bilateral inferior occipito-temporal neural network with strong activations in the right and left fusiform gyri. Action pictograms contrasted to object pictograms showed differential activation of area MT+, the inferior and superior parietal cortex, and the premotor cortex bilaterally. An analysis of psychophysiological interactions identified contribution-dependent changes in the neural responses when pictograms triggered the retrieval of conceptual action knowledge: Processing of action pictograms specifically enhanced the neural interaction between the right and left fusiform gyri, the right and left middle temporal cortices (MT+), and the left superior and inferior parietal cortex. These results complement and extend previous neuropsychological and neuroimaging studies by showing that knowledge about action concepts results from an increased coupling between areas concerned with semantic processing (fusiform gyrus), movement perception (MT+), and temporospatial movement control (left parietal cortex).

Reorienting of visuospatial attention can be investigated by comparing reaction times to validly and invalidly cued targets ("validity effect"). The cholinergic agonist nicotine reduces the validity effect and neural activity in the posterior parietal cortex. Behavioral effects of nicotine in nonsmokers are weak and it has been suggested that differences in baseline behavior before nicotine exposure may influence the effect of nicotine. This study investigates whether individual differences in reorienting-related neural activity under placebo may be used to predict individual nicotine effects. Individual nicotine effects are defined as the behavioral effects under nicotine that cannot be predicted by the behavioral data under placebo. Fifteen nonsmoking subjects were given either placebo or nicotine gum (2 mg) prior to performing a cued target detection task inside a magnetic resonance imaging scanner. The results of a partial least square analysis suggest that neural data under placebo can be used to predict individual behavioral effects of nicotine. Neural activity in the left posterior cingulate cortex, the right superior parietal cortex, the right dorsal medial prefrontal cortex, and the left ventral medial prefrontal cortex significantly contributes to that prediction. We conclude that nicotine effects on reorienting attention depend on individual differences in reorienting-related neural activity under placebo and suggest that functional magnetic resonance imaging data can contribute to the prediction of individual drug effects.

The mechanisms underlying interhemispheric integration (IHI) remain poorly understood, particularly for lateralized cognitive processes. To test competing theories of IHI, we constructed and fitted dynamic causal models to functional magnetic resonance data from two visual tasks that operated on identical stimuli but showed opposite hemispheric dominance. Using a systematic Bayesian model selection procedure, we found that, in the ventral visual stream, which was activated by letter judgments, interhemispheric connections mediated asymmetric information transfer from the nonspecialized right to the specialized left hemisphere when the latter did not have direct access to stimulus information. Notably, this form of IHI did not engage all areas activated by the task but was specific for areas in the lingual and fusiform gyri. In the dorsal stream, activated by spatial judgments, it did not matter which hemisphere received the stimulus: interhemispheric coupling increased bidirectionally, reflecting recruitment of the nonspecialized left hemisphere. Again, not all areas activated by the task were involved in this form of IHI; instead, it was restricted to interactions between areas in the superior parietal gyrus. Overall, our results provide direct neurophysiological evidence, in terms of effective connectivity, for the existence of context-dependent mechanisms of IHI that are implemented by specific visual areas during task-driven lateralization.

Unlabelled: The role of the primary motor cortex (M1) in tasks involving action words remains controversial. Therefore, we investigated whether the previously reported involvement of M1 in processing of action words results from the semantic representation of action words per se, or if M1 activation may actually depend on whether or not subjects (explicitly or automatically) adopt a strategy of simulating the movements. Subjects silently read short phrases describing a situation which either involved a motor scene or not (STIMULUS: motor, non-motor phrases) and performed a secondary task: either they were explicitly asked to imagine the situation or they performed letter detection preventing them from using a simulation strategy (TASK: imagery vs. letter detection). In addition, phrases were presented both in 1st and 3rd person singular (, Perspective: 1st vs. 3rd person). This allowed us to investigate the influence of the secondary tasks (letter detection versus explicit motor imagery) on the neural activity in M1 during the processing of motor and non-motor phrases. We found differential left M1 activity in the task by stimulus interaction with enhanced M1 activation for imagery in the presence of motor phrases (vs. non-motor phrases) compared to letter detection of motor vs. non-motor phrases. This M1-activity was not differentially modulated by perspective. Therefore, M1 activation previously found in experiments of silent reading of action words may have resulted from the subjects' strategy to mentally simulate the movements during the processing of action words.

Since the brain may engage different neuronal systems for a single behavioral goal, activation may show intersubject variability expressed in the systematic recruitment of multiple distinct networks. We apply a principal component analysis (PCA) to activation over task conditions and subjects to reveal cortical networks that may underlie this intersubject variability. Normal subjects were presented with novel meaningless objects, which appeared in personally familiar or unfamiliar places. During a subsequent, event-related functional MRI (fMRI) experiment, each subject was presented with learned or novel objects in isolation and performed a learned/novel judgment. Recollection of places was not essential for the task, and may exhibit large intersubject variations. The right posterodorsal posterior cingulate cortex (pPCC) and left retrosplenial cortex, whose involvement in place-recognition has been previously established, were selected as regions of interest for the PCA. Neural responses to objects associated with familiar relative to unfamiliar places in pPCC, but not in the retrosplenial cortex, were negatively correlated with task-related activation (common over all objects) in the right anterolateral prefrontal cortex and the left intraparietal sulcus. The latter areas have been implicated previously in cognitive control and object recognition, respectively. These results suggest right prefrontal control over neural processes both in the left parietal cortex, related to object-recognition (enhancement), and pPCC, related to nonessential recollection of place-memory (suppression), but not in the retrosplenial cortex, related to the sense of familiarity. This analysis revealed an important aspect of functional anatomy that was not detectable using a conventional analysis of average activations.

The left hemispheric dominance for complex motor behavior is undisputed. Clinical observations of complex motor deficits in patients with right hemispheric lesions, however, suggest an additional contribution of the right hemisphere to higher motor control. We assessed, using functional MRI (fMRI), which brain regions are implicated in processing the spatial aspects of complex, object-related actions. Using a blocked, factorial design, 17 healthy volunteers were asked to detect either spatial or sequential errors (factor ERROR) in complex activities of daily living, presented as video sequences with the appropriate object(s) or as pantomimes (factor STIMULUS). Observing complex actions (irrespective of stimulus type) activated a bilateral frontoparietal network. Observing actions with objects (relative to pantomimes) differentially increased neural activity in the fusiform gyrus and inferior occipital cortex bilaterally. Observing pantomimes, i.e., the same actions but without any object, differentially activated right prefrontal cortex, anterior cingulate cortex, the precuneus, and left cerebellum. The left cingulate cortex was differentially activated when subjects assessed the sequencing of actions. By contrast, assessing the spatial configuration of complex actions differentially increased neural activity in right posterior parietal cortex. A significant interaction of ERROR and STIMULUS was revealed for the right inferior parietal cortex only. These findings suggest a specific role of the right hemisphere, especially of right posterior parietal cortex, in processing spatial aspects of complex actions and thus provide a physiological basis for the observed apraxic motor deficits in patients with right hemispheric damage., ((c) 2006 Wiley-Liss, Inc.)

Human self-consciousness relies on the ability to distinguish between oneself and others. We sought to explore the neural correlates involved in self-other representations by investigating two critical processes: perspective taking and agency. Although recent research has shed light on the neural processes underlying these phenomena, little is known about how they overlap or interact at the neural level. In a two-factorial functional magnetic resonance imaging (fMRI) experiment, participants played a ball-tossing game with two virtual characters ("avatars"). During an active/agency (ACT) task, subjects threw a ball to one of the avatars by pressing a button. During a passive/nonagency (PAS) task, they indicated which of the other avatars threw the ball. Both tasks were performed from a first-person perspective (1PP), in which subjects interacted from their own perspective, and a third-person perspective (3PP), in which subjects interacted from the perspective of an avatar with another location in space. fMRI analyses revealed overlapping activity in medial prefrontal regions associated with representations of one's own perspective and actions (1PP and ACT), and overlapping activity in temporal-occipital, premotor, and inferior frontal, as well as posterior parietal regions associated with representation of others' perspectives and actions (3PP and PAS). These findings provide evidence for distinct neural substrates underlying representations of the self and others and provide support for the idea that the medial prefrontal cortex crucially contributes to a neural basis of the self. The lack of a statistically significant interaction suggests that perspective taking and agency represent independent constituents of self-consciousness.

Anatomical studies in the macaque cortex and functional imaging studies in humans have demonstrated the existence of different cortical areas within the intraparietal sulcus (IPS). Such functional segregation, however, does not correlate with presently available architectonic maps of the human brain. This is particularly true for the classical Brodmann map, which is still widely used as an anatomical reference in functional imaging studies. The aim of this cytoarchitectonic mapping study was to use previously defined algorithms to determine whether consistent regions and borders can be found within the cortex of the anterior IPS in a population of 10 post-mortem human brains. Two areas, the human intraparietal area 1 (hIP1) and the human intraparietal area 2 (hIP2), were delineated in serial histological sections of the anterior, lateral bank of the human IPS. The region hIP1 is located posterior and medial to hIP2, and the former is always within the depths of the IPS. The latter, on the other hand, sometimes reaches the free surface of the superior parietal lobule. The delineations were registered to standard reference space, and probabilistic maps were calculated, thereby quantifying the intersubject variability in location and extent of both areas. In the future, they can be a tool for analyzing structure-function relationships and a basis for determining degrees of homology in the IPS among anthropoid primates. Four-dot masking is a new form of visual masking that does not involve local contour interactions or spatial superimposition of the target stimulus and the mask (as, e.g., in pattern or metacontrast masking). Rather, the effective masking mechanism is based on object substitution. Object substitution masking occurs when low-level visual information representations are altered before target identification through iterative interaction with high-level visual processing stages has been completed. Interestingly, object substitution interacts with attention processes: Strong masking effects are observed when attentional orientation toward the target location is delayed. In contrast, no masking occurs when attention can be rapidly shifted to and engaged onto the target location. We investigated the neural basis of object substitution masking by studying the interaction of spatial attention and masking processes using functional magnetic resonance imaging. Behavioral data indicated a two-way interaction between the factors Spatial Attention (valid vs. invalid cueing) and Masking (four-dot vs. pattern masking). As expected, spatial attention improved performance more strongly during object substitution masking. Functional correlates of this interaction were found in the primary visual cortex, higher visual areas, and left intraparietal sulcus. A region-of-interest analysis in these areas revealed that the largest blood oxygenation level-dependent signal changes occurred during effective four-dot masking. In contrast, the weakest signal changes in these areas were observed when target visibility was highest. The data suggest that these areas represent an object substitution network dedicated to the generation and testing of a perceptual hypotheses as described by the object substitution theory of masking of Di-Lollo et al. [Competition for consciousness among visual events: The psychophysics of reentrant visual processes. Journal of Experimental Psychology: General, 129, 481-507, 2000].

To characterize the neural correlates of being personally involved in social interaction as opposed to being a passive observer of social interaction between others we performed an fMRI study in which participants were gazed at by virtual characters (ME) or observed them looking at someone else (OTHER). In dynamic animations virtual characters then showed socially relevant facial expressions as they would appear in greeting and approach situations (SOC) or arbitrary facial movements (ARB). Differential neural activity associated with ME>OTHER was located in anterior medial prefrontal cortex in contrast to the precuneus for OTHER>ME. Perception of socially relevant facial expressions (SOC>ARB) led to differentially increased neural activity in ventral medial prefrontal cortex. Perception of arbitrary facial movements (ARB>SOC) differentially activated the middle temporal gyrus. The results, thus, show that activation of medial prefrontal cortex underlies both the perception of social communication indicated by facial expressions and the feeling of personal involvement indicated by eye gaze. Our data also demonstrate that distinct regions of medial prefrontal cortex contribute differentially to social cognition: whereas the ventral medial prefrontal cortex is recruited during the analysis of social content as accessible in interactionally relevant mimic gestures, differential activation of a more dorsal part of medial prefrontal cortex subserves the detection of self-relevance and may thus establish an intersubjective context in which communicative signals are evaluated.
