Introduction: Replicability has become an increasing focus within the scientific communities with the ongoing "replication crisis." One area that appears to struggle with unreliable results is resting-state functional magnetic resonance imaging (rs-fMRI). Therefore, the current study aimed at improving the knowledge of endogenous factors that contribute to inter-individual variability. Methods: Arterial blood pressure (BP), body mass, hematocrit, and glycated hemoglobin were investigated as potential sources of between-subject variability in rs-fMRI, in healthy individuals. Whether changes in resting-state networks (rs-networks) could be attributed to variability in the blood-oxygen-level-dependent (BOLD)-signal, changes in neuronal activity, or both was of special interest. Within-subject parameters were estimated by utilizing dynamic-causal modeling, as it allows to make inferences on the estimated hemodynamic (BOLD-signal dynamics) and neuronal parameters (effective connectivity) separately. Results: The results of the analyses imply that BP and body mass can cause between-subject and between-group variability in the BOLD-signal and that all the included factors can affect the underlying connectivity. Discussion: Given the results of the current and previous studies, rs-fMRI results appear to be susceptible to a range of factors, which is likely to contribute to the low degree of replicability of these studies. Interestingly, the highest degree of variability seems to appear within the much-studied default mode network and its connections to other networks. Impact statement We believe that thanks to the evidence that we have collected by analyzing the well-controlled data of the Human Connectome Project with dynamic-causal modeling (DCM) and by focusing not only on the effective connectivity, which is the typical way of using DCM, but also by analyzing the underlying hemodynamic parameters, we were able to explore the underlying vascular dependencies in a much broader perspective. Our results challenge the premise for studying changes in the default mode network as a clinical marker of disease, and we add to the growing list of factors that contribute to resting-state network variability.

Using fMRI, Hugdahl et al. (2015) reported the existence of a general-domain cortical network during active task-processing which was non-specific to the cognitive task being processed. They labelled this network the extrinsic mode network (EMN). The EMN would be predicted to be negatively, or anti-correlated with the classic default mode network (DMN), typically observed during periods of rest, such that while the EMN should be down-regulated and the DMN up-regulated in the absence of demands for task-processing, the reverse should occur when demands change from resting to task-processing. This would require alternating periods of task-processing and resting and analyzing data continuously when demands change from active to passive periods and vice versa. We were particularly interested in how the networks interact in the critical transition points between conditions. For this purpose, we used an auditory task with multiple cognitive demands in a standard fMRI block-design. Task-present (ON) blocks were alternated with an equal number of task-absent, or rest (OFF) blocks to capture network dynamics across time and changing environmental demands. To achieve this, we specified the onset of each block, and used a finite-impulse response function (FIR) as basis function for estimation of the fMRI-BOLD response. During active (ON) blocks, the results showed an initial rapid onset of activity in the EMN network, which remained throughout the period, and faded away during the first scan of the OFF-block. During OFF blocks, activity in the DMN network showed an initial time-lag where neither the EMN nor the DMN was active, after which the DMN was up-regulated. Objective: This paper aims to provide a review of studies using neuroimaging to measure functional-structural reorganisation of the neuronal network for auditory perception after unilateral hearing loss., Design: A literature search was performed in PubMed. Search criterions were peer reviewed original research papers in English completed by the 11th of March 2015., Study Sample: Twelve studies were found to use neuroimaging in subjects with unilateral hearing loss. An additional five papers not identified by the literature search were provided by a reviewer. Thus, a total of 17 studies were included in the review., Results: Four different neuroimaging methods were used in these studies: Functional magnetic resonance imaging (fMRI) (n = 11), diffusion tensor imaging (DTI) (n = 4), T1/T2 volumetric images (n = 2), magnetic resonance spectroscopy (MRS) (n = 1). One study utilized two imaging methods (fMRI and T1 volumetric images)., Conclusion: Neuroimaging techniques could provide valuable information regarding the effects of unilateral hearing loss on both auditory and non-auditory performance. fMRI-studies showing a bilateral BOLD-response in patients with unilateral hearing loss have not yet been followed by DTI studies confirming their microstructural correlates. Developmental dyslexia is a common reading disorder that negatively impacts an individual's ability to achieve literacy. Although the brain network involved in reading and its dysfunction in dyslexia has been well studied, it is unknown whether dyslexia is caused by structural abnormalities in the reading network itself or in the lower-level networks that provide input to the reading network. In this study, we acquired structural magnetic resonance imaging scans longitudinally from 27 Norwegian children from before formal literacy training began until after dyslexia was diagnosed. Thus, we were able to determine that the primary neuroanatomical abnormalities that precede dyslexia are not in the reading network itself, but rather in lower-level areas responsible for auditory and visual processing and core executive functions. Abnormalities in the reading network itself were only observed at age 11, after children had learned how to read. The findings suggest that abnormalities in the reading network are the consequence of having different reading experiences, rather than dyslexia per se, whereas the neuroanatomical precursors are predominantly in primary sensory cortices., (© The Author (2014). Valuable stimuli are invariably localized in space. While our knowledge regarding the neural networks supporting value assignment and comparisons is considerable, we lack a basic understanding of how the human brain integrates motivational and spatial information. The amygdala is a key structure for learning and maintaining the value of sensory stimuli and a recent non-human primate study provided initial evidence that it also acts to integrate value with spatial location, a question we address here in a human setting. We measured haemodynamic responses (fMRI) in amygdala while manipulating the value and spatial configuration of stimuli in a simple stimulus-reward task. Subjects responded significantly faster and showed greater amygdala activation when a reward was dependent on a spatial specific response, compared to when a reward required less spatial specificity. Supplemental analysis supported this spatial specificity by demonstrating that the pattern of amygdala activity varied based on whether subjects responded to a motivational target presented in the ipsilateral or contralateral visual space. Verbal communication does not rely only on the simple perception of auditory signals. It is rather a parallel and integrative processing of linguistic and non-linguistic information, involving temporal and frontal areas in particular. This review describes the inherent complexity of auditory speech comprehension from a functional-neuroanatomical perspective. The review is divided into two parts. In the first part, structural and functional asymmetry of language relevant structures will be discus. The second part of the review will discuss recent neuroimaging studies, which coherently demonstrate that speech comprehension processes rely on a hierarchical network involving the temporal, parietal, and frontal lobes. Further, the results support the dual-stream model for speech comprehension, with a dorsal stream for auditory-motor integration, and a ventral stream for extracting meaning but also the processing of sentences and narratives. Specific patterns of functional asymmetry between the left and right hemisphere can also be demonstrated. The review article concludes with a discussion on interactions between the dorsal and ventral streams, particularly the involvement of motor related areas in speech perception processes, and outlines some remaining unresolved issues. Conflict monitoring and motor inhibition are engaged in the performance of complex tasks. The midcingulate cortex (MCC) has been suggested to detect conflicts, whereas the right inferior frontal cortex (IFC) seems to be of relevance for the inhibition process. The current experiment investigates the neural underpinnings of their interplay via a modified flanker paradigm. Conflict was manipulated by the congruency of flanking stimuli relative to a target (congruent vs. incongruent) and motor inhibition by a within-trial response change of the initiated response (keep response vs. stop-change). We used event-related functional magnetic resonance imaging, decomposition with high model order ICA, and single trial analysis to derive a functional parcellation of the whole-brain data. Results demonstrate the segmentation of the MCC into anterior and posterior subregions, and of the IFC into the pars opercularis, pars triangularis, and pars orbitalis. The pars opercularis and pars triangularis of the right IFC constituted the foundation of inhibition-related networks. With high conflict on incongruent trials, activity in the posterior MCC network, as well as in one right IFC network was observed. Stop-change trials modulated both the MCC as well as networks covering extended parts of the IFC. Whereas conflict processing and inhibition most often are studied separately, this study provides a synopsis of functionally coupled brain regions acting in concert to enable an optimal performance in situations involving interference and inhibition., (Copyright © 2012 Wiley Periodicals, Inc.)

Purpose: Children with epilepsy have a significant risk for attention-deficit/hyperactivity disorder (ADHD), which is often accompanied by deficits in working memory performance. However, it is not yet clear whether there are specific differences in the underlying mechanisms of working memory capability between children with epilepsy-related ADHD and those with developmental ADHD. There is evidence that methylphenidate can improve the behavioral difficulties in children with developmental ADHD. Whether this medication has the same effect on ADHD symptoms in patients with epilepsy is not yet well understood. The aim of the present study is, therefore, to evaluate whether boys with epilepsy-related ADHD and developmental ADHD share a common behavioral, pharmacoresponsive, and neurofunctional pathophysiology., Methods: Seventeen boys with diagnosed combined epilepsy/ADHD, 15 boys with developmental ADHD, and 15 healthy controls (aged 8-14 years) performed on working memory tasks (N-back) while brain activation was recorded using functional magnetic resonance imaging. Each patient was tested twice: once after the intake of methylphenidate and once without in a counterbalanced order., Key Findings: On a behavioral level, we show that boys with epilepsy-related ADHD as well as those with developmental ADHD performed similarly poorly on tasks with high cognitive load when compared to healthy controls, and that intake of methylphenidate improved performance almost to normal levels in both ADHD groups. On the functional level, both patient groups showed similar reductions of activation in all relevant parts of the functional network of working memory when compared to controls. Of interest, intake of methylphenidate did not significantly alter this activity pattern., Significance: Our data show strong similarities between epilepsy-related and developmental ADHD on the behavioral, pharmacoresponsive, and neural level, favoring the view that ADHD with and without epilepsy shares a common underlying neurobehavioral pathophysiology., (Wiley Periodicals, Inc. © 2012 International League Against Epilepsy.)

The independent component analysis (ICA) method was applied to fMRI data from two synaesthetes and their matched controls while they performed the coloured-word Stroop task and the single-letter (synaesthetic) Stroop task. ICA identified an 'attention' network, a 'perceptual' network as well as a 'conflict monitoring' network. Increased activity was observed in right V4 during the single-letter Stroop task for synaesthetes only. Face recognition is a primary social skill which depends on a distributed neural network. A pronounced face recognition deficit in the absence of any lesion is seen in congenital prosopagnosia. This study investigating 24 congenital prosopagnosic subjects and 25 control subjects aims at elucidating its neural basis with fMRI and voxel-based morphometry. We found a comprehensive behavioral pattern, an impairment in visual recognition for faces and buildings that spared long-term memory for faces with negative valence. Anatomical analysis revealed diminished gray matter density in the bilateral lingual gyrus, the right middle temporal gyrus, and the dorsolateral prefrontal cortex. In most of these areas, gray matter density correlated with memory success. Decreased functional activation was found in the left fusiform gyrus, a crucial area for face processing, and in the dorsolateral prefrontal cortex, whereas activation of the medial prefrontal cortex was enhanced. Hence, our data lend strength to the hypothesis that congenital prosopagnosia is explained by network dysfunction and suggest that anatomic curtailing of visual processing in the lingual gyrus plays a substantial role. The dysfunctional circuitry further encompasses the fusiform gyrus and the dorsolateral prefrontal cortex, which may contribute to their difficulties in long-term memory for complex visual information. Despite their deficits in face identity recognition, processing of emotion related information is preserved and possibly mediated by the medial prefrontal cortex. Congenital prosopagnosia may, therefore, be a blueprint of differential curtailing in networks of visual cognition.

A common assumption is that phonetic sounds initiate unique processing in the superior temporal gyri and sulci (STG/STS). The anatomical areas subserving these processes are also implicated in the processing of non-phonetic stimuli such as music instrument sounds. The differential processing of phonetic and non-phonetic sounds was investigated in this study by applying a "sound-morphing" paradigm, where the presence of phonetic features were parametrically varied, creating a step-wise transition from a non-phonetic sound into a phonetic sound. The stimuli were presented in an event-related fMRI design. The fMRI-BOLD data were analysed using parametric contrasts. The results showed a higher sensitivity for sounds containing phonetic features compared to non-phonetic sounds in the middle part of STG, and in the anterior part of the planum temporale (PT) bilaterally. Although the same areas were involved in the processing of non-phonetic sounds, a difference in activation was evident in the STG, with an increase in activation related to increment of phonetic features in the sounds. In the competition for limited processing resources, top-down attention and cognitive control processes are needed to separate relevant from irrelevant sensory information and to interact with the environment in a meaningful way. The demands for the recruitment of top-down control processes depend on the relative salience of the competing stimuli. In the present event-related functional magnetic resonance imaging (fMRI) study we investigated the dynamics of neuronal networks during varying degrees of top-down control demands. We tested 20 participants with a dichotic auditory discrimination task in which the relative perceptual salience of two simultaneously presented syllables was parametrically varied by manipulating the inter-aural intensity differences (IIDs) and instructing the subjects to selectively attend to either the louder or weaker of the two stimuli. A significant interaction of IID manipulation and attentional instruction was detected bilaterally in the inferior parietal lobe and pre-supplementary motor area, and in the precentral gyrus, anterior cingulate cortex, and inferior frontal gyrus of the right hemisphere. The post hoc analysis of the interaction pattern allowed for an assignment of these regions to either of two sets of regions which can be interpreted to constitute two different brain networks: a fronto-parietal attention control network, involved in the integration of saliency-based and instruction-based processing preferences, and a medial-lateral frontal cognitive control network, involved in the processing of the conflicts arising in the attempt to follow the attentional instruction in face of the varying inter-aural stimulus salience., (Copyright 2010 Elsevier Ltd. Glutamate is critically involved in the regulation of cognitive functions in humans. There is, however, sparse evidence regarding how blocking glutamate action at the receptor site during a cognitive task affects brain activation. In the current study, the effects of the glutamate antagonist memantine were examined with functional magnetic resonance imaging (fMRI). Thirty-one healthy adults were scanned twice in a counter-balanced design, either in a no-drug session or after administration of memantine for 21 days. The subjects performed a simple auditory perception task with consonant-vowel stimuli. Group-level spatial independent component analysis (ICA) was used to decompose the data and to extract task-related activations. The focus was on four task-related ICA components with frontotemporal localization. The results showed that glutamate-blockage resulted in a significant enhancement in one component, with no significant effect in the other three components. The enhanced effect of memantine was in the middle temporal gyrus, superior frontal gyrus, and middle frontal gyrus. It is suggested that the results reflect effects of glutamatergic processes primarily through non-N-methyl-D-aspartate (NMDA) receptor pathways. Moreover, the results demonstrate that memantine can be used as a probe which allows for studying the effect of excitatory neurotransmission on neuronal activation.

Contrary to the classical view, recent neuroimaging studies claim that phonological processing, as part of auditory speech perception, is subserved by both the left and right temporal lobes and not the left temporal lobe alone. This study seeks to explore whether there are variations in the lateralization of response to verbal and nonverbal sounds by varying spectral complexity of those sounds. White noise was gradually transformed into either speech or music sounds using a "sound morphing" procedure. The stimuli were presented in an event-related design and the evoked brain responses were measured using fMRI. The results demonstrated that the left temporal lobe was predominantly sensitive to gradual manipulation of the speech sounds while the right temporal lobe responded to all sounds and manipulations. This effect was especially pronounced within the middle region of the left superior temporal sulcus (mid-STS). This area could be further subdivided into a more posterior area, which showed a linear response to the manipulation of speech sounds, and an anteriorly adjacent area which showed the strongest interaction between the speech and music sound manipulations. Such a differential and selective response was not seen in other brain areas and not when the sound "morphed" into a music stimulus. This gives further experimental evidence for the assumption of a posterior-anterior processing stream in the left temporal lobe. In addition, the present findings support the notion that the left mid STS area is more sensitive to speech signals compared to the right homologue.

Previous functional activation studies in patients with aphasia have mostly relied on standard group comparisons of aphasic patients with healthy controls, which are biased towards regions showing the most consistent effects and disregard variability within groups. Groups of aphasic patients, however, show considerable variability with respect to lesion localisation and extent. Here, we use a novel method, joint independent component analysis (jICA), which allowed us to investigate abnormal patterns of functional activation with O(15)-PET during lexical decision in aphasic patients after middle cerebral artery stroke and to directly relate them to lesion information from structural MRI. Our results demonstrate that with jICA we could detect a network of compensatory increases in activity within bilateral anterior inferior temporal areas (BA20), which was not revealed by standard group comparisons. In addition, both types of analyses, jICA and group comparison, showed increased activity in the right posterior superior temporal gyrus in aphasic patients. Further, whereas standard analyses revealed no decreases in activation, jICA identified that left perisylvian lesions were associated with decreased activation of left posterior inferior frontal cortex, damaged in most patients, and extralesional remote decreases of activity within polar parts of the inferior temporal gyrus (BA38/20) and the occipital cortex (BA19). Taken together, our results demonstrate that jICA may be superior in revealing complex patterns of functional reorganisation in aphasia.

In working memory (WM), functional imaging studies demonstrate cerebellar involvement indicating a cognitive role of the cerebellum. These cognitive contributions were predominantly interpreted as part of the phonological loop within the Baddeley model of WM. However, those underlying investigations were performed in the context of visual verbal WM which could pose a bias when interpreting the results. The aim of this fMRI study was to address the question of whether the cerebellum supports additional aspects of WM in the context of higher cognitive functions. Furthermore, laterality effects were investigated to further disentangle the cerebellar role in the context of the phonological loop and the visuospatial sketchpad. A direct comparison of verbal and abstract visual WM was performed in 17 young volunteers by applying a 2-back paradigm and extracting the % change in BOLD signal from the fMRI data. To minimize potential verbal strategies, Attneave and Arnoult shapes of non-nameable objects were chosen for the abstract condition. The analyses revealed no significant differences in verbal vs. abstract WM. Moreover, no laterality effects were demonstrated in both verbal and abstract WM. These results provide further evidence of a broader cognitive involvement of the cerebellum in WM that is not only confined to the phonological loop but also supports central executive subfunctions. The fact that no lateralization effects are found might be attributed to the characteristics of the n-back paradigm which emphasizes central executive subfunctions over the subsidiary slave systems.

There is little knowledge regarding the underlying mediating factors for cognitive control at the synaptic level, especially the role played by glutamatergic neurotransmission. In the current study the effect of the NMDA receptor antagonist memantine was examined in a pharmacological fMRI study. Thirty-one healthy adults were scanned twice in a counter-balanced design, either drug naïve or after administration of memantine for 21 days, to build up a steady-state plasma concentration in all participants. The participants performed an auditory attention control task while in the MR scanner. The results showed significant reduction in activation in prefrontal cortex and anterior cingulate cortex after memantine administration compared to the drug naïve session. We suggest that the present results may have implications for the understanding of deficits in cognitive control in psychiatric disorders, like schizophrenia, through altered glutamatergic neurotransmission.

This study disentangles the prefrontal network underlying executive functions involved in the Wisconsin Card Sorting Test (WCST). During the WCST, subjects have to perform two key processes: first, they have to derive the correct sorting rule for each trial by trial-and-error, and, second, they have to detect when this sorting rule is changed by the investigator. Both cognitive processes constitute key components of the executive system, which is subserved by the prefrontal cortex. For the current fMRI experiment, we developed a non-verbal variant of the WCST. Subjects were instructed either to respond according to a given sorting rule or to detect the correct sorting rule, like in the original version of the WCST. Data were obtained from 14 healthy male volunteers and analysed using SPM and a random effects model. All conditions activated a fronto-parietal network, which was generally more active when subjects had to search for the correct sorting rule than when the rule was announced beforehand. Significant differences between these two conditions were seen in the dorsolateral prefrontal cortex (PFC) and the parietal lobe. In addition, the data provided new evidence for the assumption of differentiated roles of the left and right prefrontal cortex. In this fMRI-study, 6-year-old children considered at risk for dyslexia were compared with an age-/gender-matched control group for differences in brain activation when presented with visual stimuli differing in demands for literacy processing. Stimuli were nameable pictures, brand logos familiar to children, and written words - these were either regularly spelled using early-acquired rules ("alphabetic") or more complex ("orthographic"). Brain responses distinguished between the presentation conditions, as a function of group, within many cortical areas. Activation in the alphabetic and orthographic conditions in the left angular gyrus correlated with individual at-risk index scores, and activation in inferior occipito-temporal regions further indicated differential activation for the two groups related to orthographic processing, especially. Since similar patterns are reported in adult dyslexics when processing written words, it appears that sensitivity to the cortical differentiation of reading networks is established prior to formal literacy training.

Recent models of speech processing claim a ventral and dorsal stream for processing language as for the visual domain. Patients with aphasia are demonstrating selective disturbances within parts of these streams. Here, we are presenting an fMRI study, which mainly focuses on the ventral stream, i.e. the phonological, lexical, and semantic processing of auditory speech. We used three linguistic conditions: (1) listening to reversed words, (2) listening to real words and, (3) covert naming after short description. We analysed the temporal dynamics of the BOLD signal as well as the temporal dynamics of lateralisation. In addition, an independent component analysis (ICA) was applied. By combining the results from these methods, we were able to separate the areas for pure auditory-phonetic perception, located bilaterally in the middle and posterior aspects of the superior temporal lobe, from predominantly left-lateralised areas for semantic and lexical processing, located in posterior-inferior as well as anterior regions of the temporal lobe, at the temporal-parietal-junction, and in the inferior frontal gyrus. These two networks differed with respect to their time course and temporal dynamics of lateralisation, and they were separated into two components of the ICA analysis. By being able to separate these two networks, we further conclude that this complementary analysis method might also be relevant for exploring recovery processes in patients with aphasia.

Humans engaged in monotonous tasks are susceptible to occasional errors that may lead to serious consequences, but little is known about brain activity patterns preceding errors. Using functional MRI and applying independent component analysis followed by deconvolution of hemodynamic responses, we studied error preceding brain activity on a trial-by-trial basis. We found a set of brain regions in which the temporal evolution of activation predicted performance errors. These maladaptive brain activity changes started to evolve approximately 30 sec before the error. In particular, a coincident decrease of deactivation in default mode regions of the brain, together with a decline of activation in regions associated with maintaining task effort, raised the probability of future errors. Our findings provide insights into the brain network dynamics preceding human performance errors and suggest that monitoring of the identified precursor states may help in avoiding human errors in critical real-world situations.

Concurrent event-related EEG-fMRI recordings pick up volume-conducted and hemodynamically convoluted signals from latent neural sources that are spatially and temporally mixed across the brain, i.e. the observed data in both modalities represent multiple, simultaneously active, regionally overlapping neuronal mass responses. This mixing process decreases the sensitivity of voxel-by-voxel prediction of hemodynamic activation by the EEG when multiple sources contribute to either the predictor and/or the response variables. In order to address this problem, we used independent component analysis (ICA) to recover maps from the fMRI and timecourses from the EEG, and matched these components across the modalities by correlating their trial-to-trial modulation. The analysis was implemented as a group-level ICA that extracts a single set of components from the data and directly allows for population inferences about consistently expressed function-relevant spatiotemporal responses. We illustrate the utility of this method by extracting a previously undetected but relevant EEG-fMRI component from a concurrent auditory target detection experiment.

The authors used voxel-based morphometry (VBM) to study GM volume differences in the whole brain volume between a group of patients with schizophrenia and a healthy control group. There were 12 patients and 12 control subjects. The subjects were scanned in a 1.5 T MR scanner. The patients had all been evaluated by a senior psychiatrist on the brief psychiatric rating scale (BPRS). The VBM data was correlated with reports of rate and frequency of hallucinations based on their scores on the BPRS hallucination item. There were significant grey matter volume reductions in the schizophrenia patient group in the left superior (transverse) temporal gyrus, the left middle frontal gyrus, and in the right cuneus. Areas of grey matter volume reduction that correlated negatively with hallucinations were found in the left superior (transverse) temporal gyrus, left thalamus, and left and right cerebellum. This article proposes that significant reductions in grey matter volume may be instrumental in generating spontaneous neuronal activity that is associated with speech perception experiences in the absence of an external acoustic stimulus that may cause hallucinations.

To date, the delineation of the human visual "motion area" still relies on functional paradigms originally devised to identify monkey area MT. Using fMRI, we have identified putative human area V5/MT+ in normals by modelling the BOLD responses to alternating radially moving and stationary dot patterns. Functional activations were compared with cytoarchitectonic probability maps of its putative correlate area hOc5, which was calculated based upon data from histological sections of ten human post-mortem brains. Bilateral visual cortex activations were seen in the single subject dynamic versus stationary contrasts and in the group random-effects analysis. Comparison of group data with area hOc5 revealed that 19.0%/39.5% of the right/left functional activation was assigned to the right/left hOc5. Conversely, 83.2%/53.5% of the right/left hOc5 was functionally activated. Comparison of functional probability maps (fPM) with area hOc5 showed that 28.6%/18.1% of the fPM was assigned to hOc5. In turn, 84.9%/41.5% of the area hOc5 was covered by the respective fPM. Thus, random-effects data and fPMs yielded similar results. The present study shows for the first time the correspondence between the functionally defined human V5/MT+ and the post-mortem cytoarchitectonic area hOc5.

Little and controversial evidence is available from neuroimaging studies in progressive nonfluent aphasia (PNA). The goal of this study was to combine information from different imaging modalities in PNA compared with Alzheimer's disease (AD). Chemical shift imaging (CSI), voxel-based morphometry (VBM) and fluorodeoxyglucose positron emission tomography (FDG-PET) were used in 5 PNA, 10 AD patients and 10 normal subjects. Group comparisons revealed left anterior lateral temporal abnormalities (BA20/21) in PNA using CSI, VBM and PET in comparison to normal subjects. AD patients showed more limited hypometabolism within the same area. In addition left lateral parietal (BA40) abnormalities were demonstrated in our PNA as well as our AD group using PET and VBM (AD group only). Combining information from all imaging modalities on a single case basis revealed pathology within the left anterior lateral temporal and lateral parietal lobe both in PNA and AD. PNA and AD patients differed significantly, however, with respect to the frequency of medial temporal lobe and posterior cingulate/precuneus involvement. Although our results might not be generalizable to all subgroups of PNA, we conclude that medial temporal and posterior cingulate/precuneus cortex pathology as assessed by CSI and VBM or PET distinguish PNA from AD, whereas lateral temporal and parietal areas are involved in both conditions.

The present study used functional and structural MRI to investigate differences in neuronal substrates underlying shifts of attention in young and old subjects, studied with dichotic listening. Two different consonant-vowel syllables were presented and the subjects were instructed to attend to and report from either the left or right ear stimulus. Typically, a right-ear advantage is observed when attending to the right-ear stimulus, and a left-ear advantage when attending to the left-ear stimulus. The behavioral results showed that the old group had difficulties with attentional modulation of the right-ear advantage in the attend left condition. This is interpreted as a failure of an important aspect of attentional control; the top-down biasing of attention for selection of task-relevant stimulus. The fMRI results showed that an area in the left middle frontal gyrus was more activated in the young group compared to the old group in the attend left condition. The structural MRI data showed reduced gray matter density of the same area in the old group. Based on these converging findings, we suggest that the left middle frontal gyrus plays an important role in top-down biasing of selecting task-relevant stimuli, and to inhibit processing of task-irrelevant stimuli. To our knowledge, this is one of the first studies addressing the question on how age-related changes in attentional processing is reflected in both functional and structural differences in the brain.

Background: fMRI is becoming a standard tool for the presurgical lateralization and mapping of brain areas involved in language processing. However, its within-subject reproducibility has yet to be fully explored., Objective: To evaluate within-test and test-retest reliability of language fMRI in consecutive patients undergoing evaluation for epilepsy surgery., Methods: Thirty-four unselected patients were investigated once (within-test reliability) and 12 patients twice (test-retest reliability). The imaging series consisted of an alternating 25-second synonym judgment condition with a 25-second letter-matching condition repeated 15 times. Reproducibility of activation maps of the first and second half of session 1 or activation maps of sessions 1 and 2 was evaluated by comparing one global and three regional lateralization indexes (Broca's area, remaining prefrontal cortex, temporoparietal area) and on a voxel-by-voxel basis (intraclass correlation coefficient, percentage overlap, correlation of t-values)., Results: Global and regional language lateralization was achieved with high reliability within and across sessions. Reproducibility was evenly distributed across both hemispheres but not within each hemisphere. Frontal activations were more reliable than temporoparietal ones. Depending on the statistical threshold chosen, the voxel-by-voxel analysis revealed a mean overlap of activations derived from the first and second investigation of up to 48.9%., Conclusion: Language fMRI proved sufficiently reliable for the determination of global and regional lateralization of language representation in individual unselected patients with epilepsy.

Whole-head functional MR images were acquired while 10 subjects were asked to tap with their right index finger in synchrony with a visual stimulus appearing regularly with a frequency of 1.5 Hz, or irregularly with a mean frequency of 1.5 Hz. Performance data show that during regular tapping most taps were close to stimulus onset. However, when the subjects paced their tapping according to the irregular stimuli, most taps appeared about 300 ms after the onset of the pacing stimuli. Comparing the brain activations resulting from regular tapping with those from irregular tapping, we found increased activation in left precuneus only. Comparing irregular versus regular tapping shows increased activity in right cerebellar nuclei and vermis, left ventrolateral thalamus, left sensorimotor cortex, left and right pre-SMA and left SMA proper. These results show that during irregular pacing the motor areas are more strongly activated than during regular pacing. In addition, further neural systems are involved in the motor control during irregular pacing: cerebellar vermis and a cerebello-thalamo-cortical system. The latter is supposedly involved in error correction in the context of visually guided movements.

The relationship between activity within the human auditory cortices and the duration of heard tones was investigated by measuring the hemodynamic response with functional magnetic resonance imaging. We demonstrate that there is no significant influence of stimulus duration as used here on the intensity and spatial extent of the hemodynamic response in the auditory cortices. We found however, that the time course of the hemodynamic response to the repeated stimulus presentation exhibited a characteristic decline after the first stimulus exposure during the activation period. The possible reasons for this time course are currently unknown, however, several factors may be involved, including top-down mechanisms and/or the interplay of tissue perfusion and oxygen consumption., (Copyright 1999 Elsevier Science B.V.)

Abstract: Using functional MRI, we characterized field sign maps of the occipital cortex and created three-dimensional maps of these areas. By averaging the individual maps into group maps, probability maps of functionally defined V1 or V2 were determined and compared to anatomical probability maps of Brodmann areas BA17 and BA18 derived from cytoarchitectonic analysis (Amunts, K., Malikovic, A., Mohlberg, H., Schormann, T., Zilles, K., 2000. Brodmann''s areas 17 and 18 brought into stereotaxic space—where and how variable? NeuroImage 11, 66–84). Comparison of areas BA17/V1 and BA18/V2 revealed good agreement of the anatomical and functional probability maps. Taking into account that our functional stimulation (due to constraints of the visual angle of stimulation achievable in the MR scanner) only identified parts of V1 and V2, for statistical evaluation of the spatial correlation of V1 and BA17, or V2 and BA18, respectively, the a priori measure κ was calculated testing the hypothesis that a region can only be part of functionally defined V1 or V2 if it is also in anatomically defined BA17 or BA18, respectively. κ = 1 means the hypothesis is fully true, κ = 0 means functionally and anatomically defined visual areas are independent. When applying this measure to the probability maps, κ was equal to 0.84 for both V1/BA17 and V2/BA18. The data thus show a good correspondence of functionally and anatomically derived segregations of early visual processing areas and serve as a basis for employing anatomical probability maps of V1 and V2 in group analyses to characterize functional activations of early visual processing areas.
