Nicotine addiction and the occurrence of lymph node spread are two major significant factors associated with esophageal cancer’s poor prognosis; however, nicotine’s role in inducing lymphatic metastasis of esophageal cancer remains unclear. Here we show that OTU domain-containing protein 3 (OTUD3) is downregulated by nicotine and correlates with poor prognosis in heavy-smoking esophageal cancer patients. OTUD3 directly interacts with ZFP36 ring finger protein (ZFP36) and stabilizes it by inhibiting FBXW7-mediated K48-linked polyubiquitination. ZFP36 binds with the VEGF-C 3-‘UTR and recruits the RNA degrading complex to induce its rapid mRNA decay. Downregulation of OTUD3 and ZFP36 is essential for nicotine-induced VEGF-C production and lymphatic metastasis in esophageal cancer. This study establishes that the OTUD3/ZFP36/VEGF-C axis plays a vital role in nicotine addiction-induced lymphatic metastasis, suggesting that OTUD3 may serve as a prognostic marker, and induction of the VEGF-C mRNA decay might be a potential therapeutic strategy against human esophageal cancer., The mechanism of how VEGF-C is regulated to promote lymphatic metastasis of oesophageal cancer is unclear. GALNT2 is a GalNAc transferase that regulates insulin signaling, lipogenesis, and serum lipid fractions. The objective of this study was to investigate the association of GALNT2 rs2144300 and rs4846914 single nucleotide polymorphisms (SNPs) with the risk of polycystic ovary syndrome (PCOS) and related traits. The two SNPs were genotyped in 616 PCOS patients and 482 control subjects. Genetic associations with related traits were also analyzed. The genotype distributions of the two SNPs in PCOS patients were similar to those of normal controls. However, significant differences were noted across the three groups of genotypes with respect to the examined variables. Objective Huang-Lian-Jie-Du-Tang (HLJDT), a traditional Chinese medicine, improves cognitive ability in rat models of Alzheimer’s disease (AD). The objective of this study was to evaluate the protective effects of HLJDT on learning and memory impairment that are caused by Aβ25–35. Methods Rats were randomly assigned to the following groups: control (water), Aβ25–35, donepezil hydrochloride 1.05 mg/kg, HLJDT 6 g/kg, HLJDT 3 g/kg, and HLJDT 1.5 g/kg and the corresponding drugs were administered for 28 days by oral gavage. HLJDT for the prevention of Aβ25–35-induced injury in rats and the underlying mechanisms were assessed. Aβ25–35 and amyloid precursor protein (APP) levels were measured in the hippocampal specimens. Total superoxide dismutase (T-SOD), glutathione (GSH), and malondialdehyde (MDA) levels in the hippocampus were also measured. The ultrastructure of CA1 hippocampal region was observed using electron microscopy. Results HLJDT treatment ameliorated impaired learning and memory significantly, decreased Aβ25–35, and APP levels in the hippocampus, increased T-SOD and GSH activity and decreased the MDA concentration, and alleviated the nuclear and cytoplasmic abnormalities of the hippocampal CA 1 region that were induced by Aβ25–35 injection. Conclusions HLJDT might decrease hippocampal vulnerability to Aβ25–35, suggesting its potential neuroprotective effect in AD.

We report on erbium (Er) related electroluminescence (EL) in the visible and near infrared (NIR) regions from the light-emitting device (LED) based on the Er-doped ZnO (ZnO:Er)/n-Si isotype heterostructure formed by sputtering ZnO:Er film on n-Si/n+-Si epitaxial wafer. Herein, the ZnO:Er film exhibits n-type in electrical conduction. The aforementioned LED is electroluminescent only under sufficiently high forward bias with the negative voltage connecting to n+-Si substrate. Such forward bias enables the electrons from n-Si to enter into the ultra-thin SiOx (x ≤ 2) layer inherently existing between the ZnO:Er film and n-Si via Poole-Frenkel conduction mechanism and, subsequently, to drift into the ZnO:Er film thus becoming hot electrons, which impact-excite the Er3+ ions to emit characteristic visible and NIR light. Furthermore, the Er-related EL from the aforementioned LED can be significantly enhanced through adopting the strategy of co-doping F− ions into the ZnO host, which brings about twofold primary effects. Firstly, due to the atomic size compensation between F− and Er3+ ions, the ZnO crystal grains become larger to accommodate much more optically active Er3+ ions. Secondly, the partial substitution of F− ions for O2- ions around the Er3+ ion reduces the symmetry of pseudo-octahedral crystal field of Er3+ ion, thus increasing the probabilities of intra-4f transitions of Er3+ ions. We believe that this work sheds light on developing efficient silicon-based LEDs using the Er-doped oxide semiconductors.

Context and objective Preeclampsia is a pregnancy-specific disorder associated with pro-inflammatory and pro-thrombotic events. The growth arrest-specific 6 (GAS6) has been implicated in systemic inflammation and coagulation. Common genetic polymorphisms of GAS6 gene have previously been reported. The aim of this study was to investigate the association of GAS6 gene polymorphisms with the risk of preeclampsia in Chinese subjects. Study design The case–control population consists of 551 subjects. The genotyping of the single-nucleotide polymorphisms of GAS6 gene, GAS6 834 +7G/A(rs8191974) and +1332C/T (rs1803628), was carried out on genomic DNA using polymerase chain reaction-restriction fragment length polymorphisms (PCR-RFLP) analysis. Results There were significant differences in the +1332C/T genotype and allele frequencies between the patients with preeclampsia and the controls (P=0.03 and 0.02, respectively). The +1332 TT genotype was found to be protective from the development of preeclampsia (odds ratios 0.271, 95% confidence interval 0.077–0.953; P=0.03). Further analysis showed that the TT genotype of the GAS6 +1332C/T conferred a risk of severe preeclampsia (OR=0.597, 95% confidence interval 0.416–0.855; P=0.01). However, there were no differences in the 834+7G/A genotype and allele frequencies between the patients with preeclampsia and the controls. Conclusion Our data suggest that a TT genotype at +1332C/T polymorphism might be associated with decreased risk for preeclampsia, but the 834+7G/A polymorphism is not associated with the disorder, in the Chinese population.

In recent years, extensive reports have been published concerning the molecular mechanism underlying the occurrence and progression of colorectal cancer. Circular RNAs (circRNAs) have been identified as important modulators in the biological processes of colorectal cancer. Microarray analysis unveiled that differential circ-0004277 expression was identified in tissue samples of colorectal cancer. High circ-0004277 expression was then verified in tissue samples and cell lines of colorectal cancer via qRT-PCR. Kaplan-Meier analysis was used for identifying the association between circ-0004277 expression and the overall survival rate of colorectal cancer patients. A relationship existed between higher circ-0004277 expression and decreased overall survival rate of colorectal cancer patients. From a functional perspective, circ-0004277 knockdown accelerated cell apoptosis and restrained cell proliferation of colorectal cancer. From mechanistic perspective, circ-0004277 upregulated PTMA by sponging miR-512-5p. Rescue assay was used for verifying the roles of the circ-0004277-miR-512-5p-PTMA axis. Both miR-512-5p and PTMA participated in circ-0004277-mediated colorectal cancer cell proliferation based on experiments. In summary, our study showed that circ-0004277 promoted the proliferation of colorectal cancer cells as a miR-512-5p sponge to upregulate the PTMA expression.

Background: Amnestic mild cognitive impairment (aMCI) is a syndrome characterized by significant forgetfulness that does not meet the criteria of dementia. Individuals with aMCI are at a bigger risk of progressing to dementia. Current studies suggest good sleep quality is linked with preserved cognition in the elderly, and sleep complaints are common among the elderly with amnesia. Therefore, improving their sleep may be helpful for maintaining and improving their cognitive capacity. According to the theory of traditional Chinese medicine, Yi-Zhi-An-Shen is an herbal compound which may ameliorate forgetfulness and sleep disorders. As growing evidence indicates that gut microbiome is associated with major mental symptoms, a hypothesis was proposed that Yi-Zhi-An-Shen Granules (YZASG) might work by alternating microbial abundance and diversity. In this study, the investigators intend to assess the efficacy of YZASG on global cognition in the elderly suffering from aMCI, and evaluate its safety as well as its potential mechanisms via sleep quality, fecal microbial 16SrDNA and metagenomics analyses, and serum markers. Methods: This is a randomized, double-blind, placebo-controlled clinical trial. A total of 80 patients (aged 60~85 years) will be recruited and allocated randomly to a treatment group and a placebo-matched group in a 1:1 ratio, then will be administered YZASG or isodose placebo three times a day. The intervention course is 16 weeks, with an 18-month follow-up. The primary outcome is Alzheimer Disease Assessment Scale-Cognitive subscale (ADAS-cog11). Secondary outcome measures are mini-mental state examination (MMSE), Montreal cognitive assessment (MoCA), Pittsburgh Sleep Quality Index (PSQI), serum concentrations of immunological factors and inflammatory cytokines, fecal microbiota. Fecal microbiota will only be collected at the baseline and endpoint of the intervention. Discussion: The results of this trial will be conducive to assessing the safety and effectiveness on cognition of YZASG in intervening aMCI among the elderly and determining if it takes effect via the improvement of sleep quality, regulation of gut microbita, concentration of certain serum markers. Context and objective Preeclampsia is a pregnancy-specific disorder associated with oxidative stress. The glutathione S-transferases (GST) are a group of enzymes that protect cells from oxidative stress. Functional genetic polymorphisms of GST genes (GSTT1, GSTM1) have previously been reported. The aim of this study was to investigate the association of GST gene polymorphisms with the risk of preeclampsia in Chinese subjects. Study design The case-control population consists of 525 subjects. The genotyping of the GSTT1 and GSTM1 polymorphisms was carried out on genomic DNA using the multiplex polymerase chain reaction (PCR). We calculated odds ratios (ORs), adjusted for the confounding variables, to estimate the association between gene polymorphisms and preeclampsia. Results The GSTT1 null genotype was found to be protective from the development of preeclampsia (odds ratios 0.645, 95% confidence interval 0.421–0.989; P = 0.044). Further analysis showed that a combination of deletion genotypes of the GSTT1 and GSTM1 genes conferred an even lower risk of preeclampsia (OR = 0.470, 95%CI = 0.255-0.866; P = 0.015). There is no relationship between the GSTT1 and GSTM1 gene polymorphisms and blood pressure levels in pregnant women with and without preeclampsia. Conclusion Our data suggest that a GSTT1 null polymorphism might be associated with decreased risk for preeclampsia in the Chinese population, and that this risk decreases with the combination of both GSTT1 and GSTM1 null polymorphisms.

A type of metal–insulator–semiconductor (MIS) structured light-emitting device (LED), where a semitransparent gold (Au) film, a polymethyl methacrylate film, and an erbium (Er)-doped ZnO (ZnO:Er) film on a silicon substrate act as the “M”, “I”, and “S” components, respectively, has been prepared. With increasing forward bias with the positive voltage connected to the semitransparent “M” (Au) electrode, such LED first exhibits random lasing (RL) from the ZnO host itself and is then electroluminescent with the characteristic emissions from the Er3+ ions incorporated into the ZnO host. Based on the energy band diagram and the analysis of carrier transportation for the ZnO:Er-based LED applied with different forward bias voltages, the evolution from the RL to the Er-related electroluminescence as mentioned above has been tentatively explained.

We demonstrate the synthesis of Co–Ge three-dimensional nanowire arrays directly anchored on the current collector via the growth of Co nanowire arrays and subsequent RF-deposition of Ge. When used as anode materials for lithium-ion batteries, the Co–Ge nanowire arrays show a reversible capacity of 1535 mAh g−1 after 100 cycles at a current density of 320 mA g−1 and a capacity of 1239 mAh g−1 even at 8000 mA g−1. With respect to sodium-ion batteries, cycling capacity of ∼350mAh g−1 after 100 cycles is obtained. The excellent electrochemical properties may be attributed to the unique three-dimensional array nanostructures that can provide robust mechanical support, fast electron transport, and good strain accommodation.

This case study of an insulin pump is to describe our solution of the following difficulties. Firstly, how to model features to obtain a family of products. Secondly, how to handle complex constraints and synchronization of components when composing features. Thirdly, how to construct the continuous environment for the individual features as well as for the composed system.

We have previously developed silicon-based light-emitting devices (LEDs) with luminescent erbium (Er)-doped TiO2 (TiO2:Er) films [Yang et al., Appl. Phys. Lett. 100, 031103 (2012)]. In an LED therein, the TiO2:Er film is sandwiched between the ITO film and heavily boron-doped p-type silicon (p+-Si). In this work, we have investigated the electroluminescence (EL) from two LEDs with the TiO2:Er films annealed at 650 and 850 °C, respectively. It is revealed that between the TiO2:Er film and p+-Si, there is an intermediate silicon oxide (SiOx, x ≤ 2) layer and its thickness increases from ∼4 to 8 nm with the increase of annealing temperature from 650 to 850 °C. Interestingly, the thickness of the intermediate SiOx layer is found to exhibit a profound impact on the EL from the LED with the TiO2:Er film on p+-Si. The EL from the LED with the 650 °C-annealed TiO2:Er film is activated only under the forward bias with the positive voltage connecting to the p+-Si substrate. Such EL consists of the oxygen-vacancy-related emissions from TiO2 host and the characteristic visible and ∼1540 nm emissions from the Er3+ ions, while the EL from the LED with the 850 °C-annealed TiO2:Er film can only be enabled by the reverse bias with the negative voltage applied on the p+-Si substrate. Such EL features only the visible and ∼1540 nm emissions from the Er3+ ions. We have recently reported erbium (Er)-related visible and near-infrared (∼1540 nm) electroluminescence (EL) from the light-emitting device (LED) with a structure of ITO/TiO2:Er/SiO2/Si in which TiO2:Er refers to the Er-doped TiO2 film [Zhu et al., Appl. Phys. Lett. 107, 131103 (2015)]. In this work, we codope yttrium (Y) into the TiO2:Er film in the aforementioned LED to achieve the significantly enhanced Er-related visible and ∼1540 nm EL. It is suggested that the Y-codoping leads to a less symmetric and distorted crystal field around the Er3+ ions incorporated into the TiO2 host, thus increasing the probabilities of intra-4f transitions of Er3+ ions. Moreover, the Y-codoping facilitates the dispersion of Er3+ ions into the TiO2 host, which is favorable for reducing the concentration quenching effect on the Er-related EL. For the aforementioned two reasons, the EL from the LED based on the TiO2:Er film can be enhanced by means of codoping Y into the TiO2 host.
