Background: In recent years, fatty acid oxidation has been considered as an important energy source for tumorigenesis and development. There are extensive studies on CPT1A, a kind of fatty acid oxidation rate-limiting enzyme. However, prognostic value and regulatory network of another subtype CPT1B in AML remains elusive. This submission aims to clarify the independent prognostic role of the metabolic molecule CPT1B in CN-AML from cilinical data and molecule levels including mRNA, miRNA and lncRNA. Method: First, we analysed the CPT1B expression in AML conhort via online database “GEPIA”. Following the assessments on differential mRNA, miRNA, lncRNA expression analysis on the paired CPT1B high and low expression groups, there were abnormal changes at different molecular levels. Moreover, we determine the preliminary predictions on the regulatory network of CPT1B in AML by constructing miRNA-mRNA and ceRNA networks. The study had yielded a series of molecules that potentially proved the prognostic prediction and metabolic function of CPT1B from the side. Finally, we analysed the CPT1B expression in our own conhort to make a verification. Result: Notably, there was a significantly high expression of CPT1B in AML patients than normal people and was associated with the poor outcome. Molecules from different levels verified our finding and revealed the possible regulatory mechanism of CPT1B in AML. Conclusion: CPT1B is a potential prognostic factor as well as a therapeutic target of AML.

We described the spatial and temporal trends of the annual leukemia incidence, prevalence, mortality, and disability-adjusted life years (DALYs) from 1990 to 2017. Leukemia case numbers and age-standardized rates (ASRs) were extracted from the Global Burden of Disease (GBD) study 2017. The estimated annual percentage change (EAPC) in the ASR was calculated using a generalized linear model with a Gaussian distribution. The risk factors for death and DALYs due to leukemia were estimated within the comparative risk assessment framework of the GBD study. Supercritical water gasification technology can realize efficient conversion of biomass, coal and other organics into hydrogen rich gas. But the efficiency of non-catalytic gasification at relative low temperature is not high. Besides, as for catalytic gasification, catalysis mechanism is complex. Thus how to improve efficiency and master the catalysis mechanism is a challenging issue. In this thesis, supercritical water gasification of depolymerizing slag experiments with the catalysis of different kinds of catalysts are conducted and the catalysis mechanism is analyzed. The results indicate that catalyst mechanism of K2CO3 is that it can promote the swelling and hydrolysis of lignocellulose and increase the amounts of phenolic intermediates. Ru/Al2O3 presents some different catalytic properties. It facilitates hydrogenation reaction of hydrolysis products, ring-opening reaction and the cleavage of carbon-carbon bonds then enhances gasification degree and increases gasification efficiency. Moreover, the binary catalyst displays a good synergic effect and the catalytic activity is higher than that of any single catalyst since these two catalysts promote various gasification stages. The gasification efficiency and hydrogen yield increase 13.22 mmol g−1 and 66.46% respectively with the synergic catalyst of K2CO3 and Ru/Al2O3.

Violates containing polycyclic aromatic hydrocarbons (PAHs) were precipitated in the process of fast pyrolysis and gasification of coal and organic substances. PAHs are one of bottlenecks of entire coal gasification for hydrogen production. In current work, the solubility of PAH oil droplets in supercritical water/hydrogen circumstances were investigated based on molecular dynamics simulation, which was beneficial for understanding the solubility behaviors of PAHs in supercritical water/hydrogen environment. The results showed that heavy PAHs were rather stable in the water phase. Supercritical water along with hydrogen promoted the miscibility of PAHs compared with that of pure supercritical water. Furthermore, high density and high temperature facilitated the rapid solvation of PAHs in supercritical water/hydrogen environment. This paper is expected to provide a theoretical support for the development of complete coal gasification technology for hydrogen production.

Behçet’s disease (BD) is a chronic refractory multisystem autoinflammatory disease, characterized by typical clinical features of non-specific vasculitis, oral and genital ulcers, uveitis, as well as skin lesions. The exact etiopathogenesis of BD remains unknown, existing studies have indicated that genetics and environmental factors contribute to the increased development of BD. Recently, several studies have shown that external environmental factors can affect the process of epigenetic modification, and abnormalities of epigenetic factors have been confirmed to be involved in the occurrence of BD. At the same time, abnormalities of gut microbiota (GM) in the body, have also been confirmed to participate in the pathogenesis of BD by regulating the balance of Th17/Tregs. This article reviews the pathogenesis of BD and summarizes numerous clinical studies, focusing on the mechanism of GM and epigenetic factors impacting on BD, and providing new ideas for further elucidating the pathogenesis of BD.

Gestational diabetes mellitus (GDM), a type of pregnancy-specific glucose intolerance or hyperglycemia, is one of the most common metabolic disorders in pregnant women with 16.9% of the global prevalence of gestational hyperglycemia. Not only are women with GDM likely to develop T2DM, but their children are also at risk for birth complications or metabolic disease in adulthood. Therefore, identifying the potential risk factors for GDM is very important in the prevention and treatment of GDM. Previous studies have shown that genetic predisposition is an essential component in the occurrence of GDM. In this narrative review, we describe the role of polymorphisms in different functional genes associated with increased risk for GDM, and available evidence on genetic factors in the risk of GDM is summarized and discussed.

Background: Acute myeloid leukemia (AML) is a group of heterogeneous hematologic malignancies correlates with poor prognosis. It is important to identify biomarkers for effective treatment of AML. Kinases participate in many regulatory pathways and biological activities in AML. Previous studies demonstrated that MAP4K1, a serine/threonine kinase, was associated with immune regulation and cancer progression. However, its role and mechanism in acute myeloid leukemia (AML) have not been explored. Methods: RNA-seq profiling was performed for Homoharringtonine (HHT)-resistant and Homoharringtonine (HHT)-sensitive cell lines. Bioinformatic tools were used for differential analysis. Cell culture and transfection, Cell proliferation, apoptosis and Cell cycle assay, Quantitative RT-PCR, and Western blotting analysis were used to explore biological phenotypes in vitro. Findings: We found that MAP4K1 was highly expressed in HHT-induced resistant AML cell lines. In addition, overexpression of MAP4K1 in AML cells induced resistance of AML cells against HHT. Not only that, the findings of this study showed that overexpression of MAP4K1 was an independent risk factor that predicts poor prognosis of AML. Further, In vitro studies showed that MAP4K1 modulated cell cycle through MAPK and DNA damage/repair pathways. Therefore, MAP4K1 is a potential target for developing therapies for AML. Interpretation: This study demonstrates that MAP4K1 not only regulates HHT resistance but also independently predicts AML prognosis. In addition, understanding the regulatory mechanism of MAP4K1 reveals novel treatment strategies for resistant and refractory AML. Background Fatty acid oxidation (FAO) provides an important source of energy to promote the growth of leukemia cells. Carnitine palmitoyltransferase 1a(CPT1a), a rate-limiting enzyme of the essential step of FAO, can facilitate cancer metabolic adaptation. Previous reports demonstrated that CPT1a acts as a potential molecular target in solid tumors and hematologic disease. However, no systematic study was conducted to explore the prognostic value of CPT1a expression and possible treatment strategies with CPT1a inhibitor on acute myeloid leukemia (AML). Methods The expression of CPT1a in 325 cytogenetically normal AML (CN-AML) patients was evaluated using RT-PCR. The combination effects of ST1326 and ABT199 were studied in AML cells and primary patients. MTS was used to measure the cell proliferation rate. Annexin V/propidium iodide staining and flow cytometry analysis was used to measure the apoptosis rate. Western blot was used to measure the expression of Mcl-1. RNAseq and GC-TOFMS were used for genomic and metabolic analysis. Results In this study, we found AML patients with high CPT1a expression (n = 245) had a relatively short overall survival (P = 0.01) compared to patients in low expression group (n = 80). In parallel, downregulation of CPT1a inhibits proliferation of AML cells. We also conducted genomic and metabolic interactive analysis in AML patients, and found several essential genes and pathways related to aberrant expression of CPT1a. Moreover, we found downregulation of CPT1a sentitized BCL-2 inhibitor ABT199 and CPT1a-selective inhibitor ST1326 combined with ABT199 had a strong synergistic effect to induce apoptosis in AML cells and primary patient blasts for the first time. The underlying synergistic mechanism might be that ST1326 inhibits pGSK3β and pERK expression, leading to downregulation of Mcl-1. Conclusion Our study indicates that overexpression of CPT1a predicts poor clinical outcome in AML. CPT1a-selective inhibitor ST1326 combined with Bcl-2 inhibitor ABT199 showed strong synergistic inhibitory effects on AML.

Previous studies have identified that Th17/Treg cells were involved in the occurrence and development of Graves’ disease (GD). This study aimed at clarifying the association between GD susceptibility and nine single nucleotide polymorphisms (SNPs) of Th17/Treg cell-related genes, including IL2RA, miR27a, miR182, and FoxO1. A two-stage association study was performed in 650 GD patients and 1300 healthy controls. PCR–RFLP assays, real-time PCR, and ELISA were performed. In the first stage, association analysis has identified that IL2RA/rs3118470 TT genotype (Pc = 0.027, OR = 1.688) and IL2RA/rs2104286 AA genotype (Pc = 0.027, OR = 1.658) has significantly increased frequencies in patients with GD than control subjects. In the second stage, the result of rs2104286 was consistent with the first-stage results (AA genotype: Pc = 0.006, OR = 1.618). The combined data showed that IL2RA/rs2104286 AA genotype had increased frequencies in patients with GD (Pc = 8.772 × 10−6, OR = 1.636). Stratification analysis also revealed that rs2104286 AA genotype was significantly associated with Graves’ ophthalmopathy (GO) susceptibility (Pc = 9.150 × 10−4, OR = 1.851). Functional studies showed that carriers of the rs2104286 AA genotype had lower IL2RA mRNA expression than AG genotype carriers (P = 0.021). Cytokine analyses revealed that the rs2104286 AA genotype individuals had lower IL-10 levels (P = 0.015) and increased IL-17 levels than AG genotype carriers (P = 1.467 × 10−4). In conclusion, our findings suggested that IL2RA/rs2104286 was associated with GD and GO susceptibility in Southwest Chinese Han population, which may be involved in the occurrence of GD and GO by affecting the mRNA expression of IL2RA gene and the cytokine production.

Objective: To develop a risk prediction model to identify the high-risk individuals of large niche formation after cesarean section (CS). Design A retrospective study. Setting Women’s health research in Anhui, China. Population: Women received CS between Jan 2012 to Jun 2017. Methods: Women were arranged to receive uterine scar examination by transvaginal ultrasonography, and those diagnosed with niche were divided into two groups according to whether they suffer from postmenstrual spotting. The cut-off values of depth, RMT (residual myometrium thickness), and depth/AMT (adjacent myometrium thickness) were chosen to define a large niche. Then, all participants were classified into three groups, including a control, a small niche, and a large niche group. The scores of each variable in the prediction model were calculated by dividing the minimum β-coefficient from the multivariate logistic analysis. Main outcome: Primary outcome was a prediction scoring model for large niche formation. Results: In total, 727 women were recruited in this study, and the large niche was defined as more than 0.50 cm in depth, less than 0.21 cm in RMT, more than 0.56 in depth/AMT. The large niche prediction model included eight variables of age at delivery, retroflexed uterine, meconium-stained amniotic fluid, history of CS, B-Lynch suture, operation duration, premature rupture of membranes and cervical dilatation more than 4 cm. The cut-off value of 5 in this score-based model presented sensitivity and specificity as 67.48% and 90.07% respectively. Conclusions: This score-based risk prediction model could present the risk of large niche formation of individuals after CS.

Ovarian function is important for optimizing endocrine treatment in patients with hormone receptor-positive (HR+) early breast cancer (eBC). The aim of the study was to determine whether patients’ pretreatment levels of anti-Mullerian hormone (AMH) were associated with menses status after chemotherapy and to build a predictive nomogram model for amenorrhea in women with HR+ eBC. Between August 2013 and December 2014, 120 premenopausal patients with HR+ eBC were included retrospectively. The associations among age, prechemotherapy levels of AMH, follicle-stimulating hormone (FSH),and estradiol (E2) and the 2-year postchemotherapy menses status were analyzed. We determined the cutoff values of hormone levels by using the biostatistical tool (Cutoff Finder). A novel nomogram was established to predict the 2-year amenorrhea status based on the logistic analysis. Concordance index (C-index) was used to validate the capacity. One hundred nine women (90.8%) experienced amenorrhea after chemotherapy. AMH

Zhundong coal (ZDC) has attracted much attention due to its high alkali metal content which can lead to a series of problems such as furnace slagging and ash fouling. Supercritical water gasification (SCWG) become a better choice for ZDC coal utilization because of its unique chemical and physical properties. The transformation mechanism of alkali metals during SCWG process was different from conventional ways of coal utilization. Systematic research about it could hardly be found. In this study, ZDC was used to explore sodium transformation mechanism and kinetics during supercritical water gasification under typical conditions. We got four kinds of sodium including the water-soluble fraction (L1), the carboxylic matrix-associated fraction (L2), the macromolecular organic group-associated fraction (L3), and the inorganic silicate mineral fraction (L4) through sequential extraction method after SCWG. A reaction pathway of sodium transformation in supercritical water gasification was proposed. A quantitative kinetic model for describing sodium transformation mechanism was developed. Finally, it was found that, L1 played an important role in catalytic process and mineral in coal weaken the catalytic process by combining with L1. L2 and L3 served as the two important intermediate products in the coal gasification, which explained the catalytic mechanism of sodium. L3 showed better reactivity. Sodium finally tended to deposit in the form of NaSiAlO4 (L4) which was stable and environmentally friendly. All of these could provide basis for high-efficiency utilization of ZDC and the design of a reactor.

Supercritical water gasification technology provides a promising way for the efficient and clean utilization of high-sodium Zhundong coal. Pore structure evolution rules of char in the gasification process can provide important information for the heat and mass transfer in char particles and the optimization of kinetic models. In this work, supercritical water gasification experiments of Zhundong coal were conducted and the increase of temperature was found to promote the hydrogen production and gas yield increasing. Residual chars were characterized using techniques of nitrogen adsorption at 77 K, mercury intrusion porosimetry and micro-computed tomography. The results indicated that pores mainly appeared in two regions of 1.7–2 nm and 600–2000 nm. The specific surface area showed an increasing trend in the gasification process with micropores dominating the total surface area. The increase of temperature promoted the pore volume development, and a good linear relationship between pore volume and gas production was found.

Supercritical water gasification (SCWG) is a promising technology for coal utilization and has drawn lots of attention. K2CO3 is an outstanding catalyst for coal gasification and can significantly improve pore structure. The evolution of pore structure in coal particles affects heat and mass transfer efficiency, but little research has investigated the effect of K2CO3 on gasification results from the perspective of pore structure. Hence, this study revealed the variation of pore structure of coal chars with stepped K2CO3 loading during SCWG of coal by conducting experiment in a high-capacity autoclave reactor. Gas products and coal chars were characterized by GC, SEM, and BET to obtain the gasification result, surface morphology, and pore structure characteristics of coal chars. The results show that K2CO3 improved gasification efficiency, gas percentage, and gas yield of H2 and CO2. The time when K2CO3 changes pore structure is during the gasification process rather than before gasification. Macropores are dominant in coal chars. 0 wt%-8 wt% K2CO3 favors the formation of mesopores and macropores, while 10 wt% K2CO3 blocks the pore structure. 0 wt%-4 wt% K2CO3 loading makes a pore structure of the ink-bottle type, and 6 wt%-10 wt% K2CO3 loading makes the narrow-slit pore structure. When K2CO3 increases from 0 wt% to 8 wt%, production rate of C-containing gases has a good linear relationship with specific surface area, and pore volume also has a good linear relationship with carbon gasification efficiency. 10 wt% K2CO3 destroys the linear relationship by the blocking effect.

Supercritical water gasification (SCWG) technology is one of the most promising ways to realize efficient and clean utilization of coal because of the special physical and chemical properties of supercritical water. K2CO3 is the most preferable catalyst to improve gasification efficiency. However, K2CO3 leads to ash agglomeration and massive volatility of alkali metals. The accumulation of agglomerated ash prevents the continuous operation of the system, and the volatility of alkali metals weakens the coupling of high-grade H2/CO2/H2O mixture with downstream process. In this study, K2CO3-catalytic SCWG of coal with NaAlO2 loading was firstly investigated in a laboratory-scale continuous reaction system to check whether NaAlO2 can inhibit ash agglomeration and lower the volatility of alkali metals without decreasing the gasification efficiency. The concentration of coal slurry was 20 wt%-35 wt% and the loading content of NaAlO2 was 0 wt%-7.5 wt% of dry coal. By doing GC, PSD, XRD, SEM and ICP-OES analysis for three-phase products collecting from the experiment, it was found that, when NaAlO2 loading increased from 0 wt% to 7.5 wt%, the carbon gasification efficiency of 20 wt%, 30 wt% and 35 wt% coal slurry improved from 90.7% to 93.9%, from 83.8% to 88.6% and from 72.6% to 80.5%, the percentage of H2 increased and the percentage of CO2, CH4 and CO decreased. The addition of NaAlO2 transformed K and Si of coal into K(Na,K)3Al4Si4O16 instead of K2Si2O5, which contributes to the distribution of ash particles into smaller size. 5 wt% NaAlO2 loading decreased the total volatility of alkali metals, and 7.5 wt% NaAlO2 loading will increase the total volatility of alkali metals.

Supercritical water gasification (SCWG) technology shows huge advantages to achieve efficient and clean utilization of coal. Ash agglomeration caused by K2CO3 addition makes ash discharging process more difficult and inhibits gasification reaction of carbon in agglomerated ash, which prevents the constant operation of the system and decreases gasification efficiency. This study investigated the formation mechanism of ash agglomeration in potassium carbonate-catalyzed SCWG of coal and find a solution to inhibit this problem. Experiments were conducted in an autoclave to figure out the effect of K2CO3 (0 wt%-10 wt%) and Al2O3 (0 wt%-20 wt%) on ash agglomeration level at 750 °C. After every single experiment, solid residue was dried and classified into different sizes (0–100 μm, 100–1000 μm, >1000 μm). The ash agglomeration characteristics was examined by X-ray diffraction (XRD), scanning electron microscopy (SEM) and energy dispersive X-ray (EDS). Results indicated that K2CO3 reacted with clay and quartz in raw coal, then formed K2Si2O5, KAlSiO4, KAlSi2O6 and KAlSi3O8. K2Si2O5 plays a cohesive role between different ash particles. Adding Al2O3 can effectively solve this problem by forming KAlSiO4 instead of K2Si2O5. Besides, Al2O3 enhances carbon gasification efficiency by increasing heating rate of feedstock temperature. Simulation work was also done to investigate the boundary condition for ash agglomeration.

Objective This meta-analysis aimed to determine the associations between the rs3761547, rs3761548, and rs3761549 single-nucleotide polymorphisms (SNPs) of the forkhead box P3 ( FOXP3) gene and susceptibility to Graves’ disease (GD). Methods Case–control studies with information on the associations between the rs3761547, rs3761548, and rs3761549 FOXP3 SNPs and GD published before 01 May 2020 were identified in the PubMed, Embase, Web of Science, and China National Knowledge Infrastructure databases. Data from the studies were analyzed using RevMan version 5.3. Results Seven independent case–control studies including 4051 GD patients and 4569 controls were included in the meta-analysis. The overall pooled analysis indicated that FOXP3/rs3761548 and FOXP3/rs3761549 polymorphisms were significantly associated with GD susceptibility (rs3761548: A vs. C, odds ratio [OR] = 1.32, 95% confidence interval [CI] 1.05–1.67; rs3761549: TT vs. CC, OR = 1.98, 95%CI 1.49–2.65; (TT + TC) vs. CC, OR = 1.44, 95%CI 1.11–1.88). In contrast, the FOXP3/rs3761547 polymorphism was not associated with GD susceptibility. Subgroup analysis according to ethnicity showed that rs3761548 was associated with GD in Asians but not in Caucasians, whereas rs3761549 was associated in both Asians and Caucasians. Conclusion This meta-analysis demonstrated that FOXP3/rs3761548 and FOXP3/rs3761549 SNPs were significantly associated with susceptibility to GD, at least in Asian populations.

Nitrogen is an important element related to coal conversion process due to its considerable impact on the environment. In this study, indole, a nitrogen-containing compound of coal, was utilized to explore nitrogen transformation during supercritical water gasification. Experiments were conducted with K2CO3 as catalyst under temperature conditions from 650 °C to 750 °C, pressure of 25 MPa, and residence time from 1 min to 30 min with feedstock concentration of 2 wt%; products and nitrogen transformation characteristics were then investigated in detail. First, we found that carbon gasification efficiency of 95.89% was obtained under conditions of 750 °C, 25 MPa for 30 min, and that gaseous products mainly contained H2, CO, CO2 and CH4. Nitrogen-containing products were not detected in gaseous products. Second, the main liquid intermediate organic products were phenol, aniline, and other nitrogen-containing compounds, and the liquid inorganic product was ammonia. Then a reaction pathway of indole gasification in supercritical water was proposed. The quantitative kinetic model for describing the gaseous products and nitrogen transformation mechanisms of indole was developed. The activation energy of carbon gasification reaction with K2CO3 as catalyst was 209.1 ± 33.2 kJ/mol.

Char conversion is the rate-determining step for coal gasification in supercritical water, because the main components of char are polycyclic aromatic hydrocarbons (PAHs) with chemical stability. Anthracene was selected as the model PAH compound to explore the gasification mechanism of coal in supercritical water. The experimental parameters investigated with autoclaves were within the following ranges: temperature, 600–750 °C; residence time, 5–30 min; and pressure, 23–25 MPa. The main gas products were H2 and CO2. The liquid intermediates were analyzed by gas chromatography–mass-selective detection (GC-MSD) for qualitative analysis and quantitative analysis, respectively. Benzene and naphthalene were found to be relatively stable compounds in the liquid products. Effective catalyst and appropriate temperature were obtained to enhance the gasification process.

The regulation of magnetic domains and high-frequency property in Fe20Ni80 thin films sputtered on anodic aluminum oxide (AAO) substrates with different apertures and a Si substrate were investigated. The obvious stripe domain structure was observed in FeNi thin film sputtered on AAO (pore in diameter d ~ 20 nm) substrate. The distinct cross-sectional columnar structures prepared on three different substrates and measured by scanning electron microscope were shown, which arose from different growth mechanisms on various kinds of substrates. The structure of AAO substrate could modulate the growth mechanism of thin films and the appearance of stripe domains structure. Objective: To systematically evaluate the efficacy and safety of bortezomib with chemotherapy in the treatment of mantle-cell lymphoma and follicular lymphoma. Methods: Randomized controlled clinical trials are conducted. Odd ratios and 95% confidence intervals were applied to pool the effect size. Results: This study identified 3 eligible studies, including 1179 patients with mantle-cell lymphoma or follicular lymphoma. Bortezomib-based chemotherapy treatment was presented to increase the proportion of complete response (OR=1.56, 95%CI=1.20-2.02) in a fixed-effect model, with no heterogeneity among the three studies (I2=0.0%, P=0.883). Odd ratios of overall response (OR=1.61, 95%CI=0.81-3.20) had no statistically significant difference; however, the heterogeneity was significant among the three studies (I2=64.5%, P=0.060). The risk of any adverse events (grade≥3) and peripheral neuropathy (all grades) were estimated by Odd ratios. There was an increasing risk of any adverse events (grade≥3) in the group with bortezomib (OR=3.15, 95%CI=2.26-4.40), with no heterogeneity (I2=0.0%, P=0.594). No significant difference was founded to estimate the risk of peripheral neuropathy (all grades) (OR=4.14, 95%CI=0.43-40.23), and we examined a considerable heterogeneity (I2=92.2%, P=0.000). Conclusion: The chemotherapy protocol with bortezomib is superior to the protocol without bortezomib according to CR. The regimen with bortezomib increases the incidence of any adverse effects (grade≥3) with no statistical difference of peripheral neuropathy (all grades).

Tellurium film was successfully electrodeposited on to macroporous phenolic foam with the assistance of a conducting polyaniline coating layer. The thermopower of the obtained macroporous Te film was measured. The dense and uniform Te film was composed of columnar crystals that preferably oriented along c-axis direction. The seebeck coefficient of the macroporous Te film showed a peak value of 342 μV/K at 473 K.

A novel conductive process for resistive random access memory cells is investigated based on nanoporous anodized aluminum oxide template. Bipolar resistive switching characteristic is clearly observed in CoFe2O4 thin film. Stable and repeatable resistive switching behavior is acquired at the same time. On the basis of conductive filament model, possible generation mechanisms for the resistive switching behaviors are discussed intensively. Besides, the magnetic properties of samples (before and after the annealing process) are characterized, and the distinct changes of magnetic anisotropy and coercive field are detected.
