Myeloid-derived suppressor cells (MDSCs) play a critical role in tumor immune escape because of its remarkable immunosuppressive effect. However, the mechanism of MDSCs migrated into tumor microenvironment remains unclear. In this study, we demonstrated the recruitment of MDSCs can be promoted by exosomes derived from prostate cancer cells, which could upregulate chemokine (CXC motif) receptor 4 (CXCR4) via the TLR2/NF- κ B signalling pathway. Flow cytometry detected that the percentage of MDSCs in the mice spleen and tumor tissue was significantly increased after injection with exosomes via mouse tail vein. Transwell chemotaxis assay showed the recruitment of MDSCs toward the lower chamber was enhanced after stimulation with exosomes, and the migration ability could be inhibited by AMD3100 (a CXCR4 specific inhibitor) both in vivo and in vitro. Additionally, Western blot and flow cytometry verified a remarkably increase of CXCR4 in MDSCs after incubation with exosomes; meanwhile, the protein level of TLR2 and activation of NF- κ B were also strengthened obviously. Nevertheless, after blocking TLR2 by C29 (a TLR2-specific inhibitor), the expression of p-p65 and CXCR4, which were hypothesized as the downstream target of TLR2, was prominently reduced. In conclusion, prostate cancer-derived exosomes could reinforce CXCR4 expression in MDSCs through the TLR2/NF- κ B signalling pathway, eventually promoting migration of MDSCs into tumor microenvironment in a CXCR4-CXCL12 axis-dependent manner.

Objectives MicroRNA-199b-3p (miR-199b-3p) plays a crucial role in the malignant development of various cancers, but little known in prostate cancer (PCa). The aim of our study was to demonstrate the function of miR-199b-3p in PCa. Methods Quantitative real-time polymerase chain reaction (RT-qPCR) was used to detect miR-199b-3p expression in PCa and benign prostatic hyperplasia (BPH) tissue samples. In addition, we examined the relationship between the poor prognosis in PCa and miR-199b-3p. Western blot was used to analyze the expression of Phospholipase Ce (PLCe). CCK8 and colony-forming assays were applied to detect the proliferation of PCa. EdU assay is used to detect PCa cells uptake of EdU. Luciferase reporter assay was applied to analyze the binding between miR-199b-3p and PLCe. Results It has been shown that miR-199b-3p in PCa was significantly lower than that in benign prostatic hyperplasia and correlated with poor prognosis. Meanwhile, upregulation of miR-199b-3p can prominently inhibit the proliferation of PCa cells, while its down-regulation triggered opposite result. PLCe was identified as the downstream binding target gene and negatively associated with that of miR-199b-3p. Conclusion miR-199b-3p suppresses malignant proliferation by inhibiting PLCe in prostate cancer in vitro and vivo.

Graves’ disease (GD), an organ-specific autoimmune disease, is the most common cause of hyperthyroidism. Tumour necrosis factor-alpha (TNF-α) exhibits immunological and metabolic activities involved in the induction and maintenance of immune responses. We attempted to evaluate the relationship between GD and serum TNF-α and its soluble receptors (sTNFRs), soluble TNF receptor 1 and 2 (sTNF-R1 and sTNF-R2). A total of 72 GD patients and 72 matched healthy individuals were recruited for this study. Serum TNF-α and sTNFRs were measured by sandwich ELISA. In our study, no significant difference was observed in TNF-α, but sTNFRs were found to be significantly elevated in GD patients compared to healthy individuals. Serum sTNFR levels were positively correlated with free triiodothyronine (FT3) and free thyroxine (FT4), and TNF-α was negatively correlated with thyroid-stimulating hormone (TSH) in the GD group. It was also shown that thyrotropin receptor antibody (TRAb) was positively correlated with TNF-α and sTNFRs. Spearman’s correlation analysis showed that only sTNF-R1 was positively correlated with complement C3. Multiple linear regression analysis suggests that serum levels of sTNF-R1 and FT4 may play an important role in the serum level of FT3. According to the median value of FT3 level, GD patients were further divided into a high FT3 group and a low FT3 group. The serum levels of sTNF-R1 in the high FT3 GD group were significantly higher than those in the low FT3 GD group. In conclusion, sTNFRs may play an important role in anti-inflammatory and immune response in GD.

Background Senile osteoporosis (SOP), which is caused by unbalanced bone remodeling, leads to significant economic and societal burdens globally. The combination of Epimedii Folium (EF) and Ligustri Lucidi Fructus (LLF) serves as a commonly-used prescription for SOP in Traditional Chinese Medicine (TCM). This study aimed to evaluate the osteoprotective effects of EF and LLF in combination on SOP rats based on the constructed multilayer perception (MLP)-artificial neural network (ANN) model. Methods 15 month old male Sprague–Dawley rats were administrated with EF, LLF or the combination of EF and LLF (EF&LLF) for 2 months, while 17 month old rats were used as the aging control group. All the rats were anesthetized with 25% ethyl carbamate, then their serum liver and bone tissues were taken. We detected bone mass, bone mineral density (BMD), biomechanics and the microstructure of bone trabecula by micro-CT and H&E staining to evaluate the degree of osteoporosis. Blood lipids and serum alanine aminotransferase (ALT), aspartate aminotransferase (AST) and γ-glutamyl transferase (GGT) and liver pathology were use to assess the side effects of drugs. Levels of alkaline phosphatase (ALP) and Tartrate-resistant acid phosphatase (TRACP) and the ratio of ALP to TRACP both in serum and bone were measured for the evaluation of bone turnover rate. The bone mRNA and protein expression of osteoprotegerin (OPG), nuclear factor-kappa B ligand (RANKL), macrophage colony-stimulating factor (M-CSF), d2 isoform of vacuolar (H+) ATPase (ATP6V0d2), insulin-like growth factor (IGF-1), bone morphogenetic protein-2 (BMP2), M-CSF, Wnt5a, transforming growth factor-β1 (TGF-β1) were detected for evaluating bone metabolism. Results The results showed that EF&LLF improved bone mass and bone quality by preventing bone loss, increasing maximal load as well as protecting the micro-structural retrogressive change of trabecular bone in SOP rats; ameliorated the steatosis in the liver and decreased blood lipids and serum ALT, AST and GGT; enhanced bone remodeling by stimulating the expression of ALP and TRACP. At the molecular levels, EF&LLF stimulated the osteoclastogenesis by upregulating the protein and mRNA expression of OPG, RANKL, M-CSF and ATP6V0d2; meanwhile, EF&LLF stimulated osteoblastogenesis by enhancing the expression of TGF-β1, BMP2, Wnt5a and IGF-1. According to our established MLP model, EF&LLF has a better effect on osteoclastogenesis or steoblastogenesis in SOP rats than EF or LLF. Conclusions These findings demonstrate that the systemic bone protective effects of EF&LLF by promoting bone remodeling in aging rats might be a substitute medicine for the treatment of SOP.

Thermoreflectance imaging technique is a noncontact measurement technology that has attracted much attention in the field of thermal characteristics measurement of semiconductor devices. This technology is often used for steady-state temperature measurement and transient temperature measurement of semiconductor devices, with the advantages of high spatial resolution, high temperature resolution, and high time resolution. However, during the measurement of the device, the thermal expansion of the temperature control table and the sample caused by the temperature rise makes the optical image appear defocused. In order to ensure that the optical image collected by the CCD is not distorted, it is necessary to design an image alignment system for focusing so that the semiconductor device can be measured normally. In this paper, an image alignment system consisting of an imaging module, a high-precision motion control component and an image alignment algorithm is proposed to solve the problem of image defocusing caused by thermal expansion, which can provide references for the optimal design and precise measurement of the thermal reflection imaging system based on CCD.

Castration‑resistant prostate cancer (CRPC) is a major challenge in the treatment of prostate cancer (PCa). Phospholipase Cε (PLCε), an oncogene, has been found to be involved in the carcinogenesis, tumor proliferation and migration of several types of cancer. The effects, however, of PLCε on CRPC remains unclear. In the present study, the expression of PLCε and glioma‑associated homolog (Gli)‑1/Gli‑2 in benign prostatic hyperplasia (BPH), PCa and CRPC tissues and cells was investigated, and the correlations between PLCε and Gli‑1/Gli‑2 in CRPC tissues and cell lines were further explored. In addition, the effect of PLCε on cell proliferation and invasion was assessed in CRPC cell lines, and the sensitivity of EN‑R and 22RV1 cells to enzalutamide following the downregulation of PLCε expression was determined using lentivirus‑mediated shPLCε and/or treatment with specific Gli inhibitor GANT61. It was found that the PLCε expression was excessively upregulated in the majority of CRPC tissues, and PLCε positivity was linked to poor progression‑free survival (PFS) and overall survival (OS) in patients with PCa. Furthermore, PLCε knockdown significantly suppressed CRPC cell proliferation and invasion. Of note, it was found that PLCε knockdown increased the sensitivity of CRPC cells to enzalutamide in vitro by suppressing androgen receptor (AR) activities via the non‑canonical Hedgehog/Gli‑2 and p‑STAT3 signaling pathways. PLCε knockdown was shown to increase the sensitivity of CRPC cell xenografts to enzalutamide in vivo. Finally, the combination of PLCε knockdown with GANT61 significantly sensitized CRPC cells to enzalutamide. Collectively, the results of the present study suggest that PLCε is a potential therapeutic target for CRPC.

Background: The relationship between fasting hyperglycemia (FHG) and new-onset atrial fibrillation (AF) in patients with acute myocardial infarction (AMI) is unclear, and whether their co-occurrence is associated with a worse in-hospital and long-term prognosis than FHG or AF alone is unknown.Objective: To explore the correlation between FHG and new-onset AF in patients with AMI, and their impact on in-hospital and long-term all-cause mortality.Methods: We performed a retrospective cohort study comprising 563 AMI patients. The patients were divided into the FHG group and the NFHG group. The incidence of new-onset AF during hospitalization was compared between the two groups and sub-groups under different Killip grades. Logistic regression was used to assess the association between FHG and new-onset AF. In-hospital mortality and long-term all-cause mortality were compared among patients with FHG, AF, and with both FHG and AF according to 10 years of follow-up information.Results: New-onset AF occurred more frequently in the FHG group than in the NFHG group (21.6 vs. 9.2%, p < 0.001). This trend was observed for Killip grade I (16.6 vs. 6.5%, p = 0.002) and Grade II (17.1 vs. 6.9%, p = 0.005), but not for Killip grade III–IV (40 vs. 33.3%, p = 0.761). Logistic regression showed FHG independently correlated with new-onset AF (OR, 2.56; 95% CI, 1.53–4.30; P < 0.001), and 1 mmol/L increased in fasting glucose was associated with a 5% higher rate of new-onset AF, after adjustment for traditional AF risk factors. AMI patients complicated with both fasting hyperglycemia and AF showed the highest in-hospital mortality and long-term all-cause mortality during an average of 11.2 years of follow-up. Multivariate Cox regression showed FHG combined with AF independently correlated with long-term all-cause mortality after adjustment for other traditional risk factors (OR = 3.13, 95% CI 1.64–5.96, p = 0.001), compared with the group with neither FHG nor new-onset AF.Conclusion: FHG was an independent risk factor for new-onset AF in patients with AMI. AMI patients complicated with both FHG and new-onset AF showed worse in-hospital and long-term all-cause mortality than with FHG or AF alone.

The past decade has witnessed a rapid development of open government data practices and academic research. However, there is no systematic survey of existing research to understand the evolution of open government data. Such research can facilitate knowledge transfer within and across domains, and foster learning for countries in the early stages of open government data development. This study quantitively extracted the evolution trajectory of open government data based on the main path analysis method and then analysed the underlying motivations. The results show that open government data research went through four main phases and that the open government data movement has spread towards developing countries and smart cities. Different challenges and issues faced by the researchers in each phase drove the evolution of open government data research. Finally, we discuss future directions of open government data research based on our findings and recent development. There is a tendency to create sustainable open government data and smartness by employing artificial intelligence and creating data marketplaces. Points for practitioners Open government data efforts have evolved over the years into a global phenomenon. Countries have learned from each other and more and more efforts are focused on innovating with open government data by stimulating co-creation and using other incentives. The way that data are opened should focus on achieving goals like innovation, participation, transparency and accountability. There is a tendency to create sustainable open government data and smartness by employing artificial intelligence and creating data marketplaces.

In sea battlefield search and rescue operation problems, the risk assessment for ships is a vital procedure to get a decision support as an important intelligence to combat search and rescue. In this paper, based on back propagation (BP) neural network, a risk assessment indicator system is proposed first to facilitate the hazard, the vulnerability, and the resilience. Next, by developing two types of network structure, the main neural network for estimating the risk has been modeled. Then, the corresponding learning algorithm is designed. Finally, 69 samples for search and rescue ships are given as the input of the neural network. The performance and regression state of the obtained training results demonstrate that the proposed risk assessment method is valid and effective.

Most prostate cancers (Pcas) develop into castration-resistant prostate cancer (CRPC) after receiving androgen deprivation therapy (ADT). The expression levels of PLCε and wnt3a are increased in Pca and regulate androgen receptor (AR) activity. However, the biological function and mechanisms of PLCε and wnt3a in CRPC remain unknown. In this study, we found that the expression levels of PLCε, wnt3a, and AR were significantly increased in CRPC tissues as well as bicalutamide-resistant-LNCaP and enzalutamide-resistant-LNCaP cells. In addition, PLCε knockdown partly restored the sensitivity of drug-resistant cells to bicalutamide and enzalutamide by inhibiting the activity of the wnt3a/β-catenin/AR signaling axis. Interestingly, the resistance of LNCaP cells docetaxel is related to PLCε but not the wnt3a/β-catenin pathway. We also found that the combination of PLCε knockdown and enzalutamide treatment synergistically suppressed cell proliferation, tumor growth, and bone metastasis using in vitro and in vivo experiments. Our study revealed that PLCε is involved in the progression of drug-resistance in CRPC and could be a new target for the treatment of CRPC.

In this study, an expanded granular sludge blanket (EGSB) reactor was used for the treatment of low-strength domestic sewage and the sludge granulation process was systematically investigated. At an optimized hydraulic retention time (HRT) of 5 h, up-flow velocity (Vup) of 1.9 m/h, and organic loading rate (OLR) of 2.16 kg COD/m3/d, the average COD removal efficiency was 71.5 ± 2.3%. Completely granular sludge can be observed after 107 d of continuous operation. Analysis of the distribution and composition of the extracellular polymeric substances (EPS) indicates that the tightly bound EPS (TB-EPS) content shows an increasing trend, while the loosely bound EPS (LB-EPS) content did not significantly alter after the granular sludge was formed. The three-dimensional excitation-emission matrix technique (3D-EEM) confirms that aromatic protein-like substances are of key importance to sludge granulation. High-throughput sequencing analysis indicates that the metabolism shifted from hydrogenotrophic (Methanobaterium) to aceticlastic methanogens (Methanosaeta) during sludge granulation.

Turnip, one of the oldest groups of cultivated Brassica rapa species, is a traditional crop as well as a form of animal fodder, a vegetable, and a herbal medicine that is widely cultivated in farming and farming-pastoral regions in Tibet. Different regions of the Qinghai–Tibet Plateau (QTP) are home to a rich diversity of turnip owing to their high altitudes and variable climate types. However, information on the morphology and genetic diversity of Tibetan turnip remains limited. Therefore, the genetic diversity of 171 turnip varieties from China and elsewhere (Japan, Korea, and Europe) was analyzed using 58 morphological characteristics and 31 simple sequence repeat (SSR) markers in this study. The varieties showed that the genetic distance ranged from 0.12 to 1.00, and the genetic similarity coefficient ranged between 0.73 and 0.95. Cluster tree showed two distinct clusters. Both morphotype and geography contributed to the group classification. A combination of morphological traits and molecular markers could refine the precision of accurate identification compared to the separate morphological and molecular data analyses. The sampling ratio of 15% to utmost precisely represent the initial population was compared to ratios of 10% and 20%, and the sampling ratio of 15% is recommended for future works when a primary core collection of turnip resources is constructed. These results could furnish a foundation for germplasm conservation and effective turnip breeding in future studies.

Wireless ultraviolet (UV) non-line-of-sight (NLOS) communication network is increasingly being used in three-dimensional (3D) space, such as formation flying and holding, underwater communication networks and ground-to-air communications. In this article, a networking strategy for three-dimensional wireless ultraviolet communication network (UVNNS) is proposed to optimize the coverage, connectivity and the survivability of the UV network. The performance of UVNNS with different communication parameters (apex angle, transmitted power, data rate, error probability and node density), modulation and noise model is simulated and analyzed in the 3D UV network. The simulation results show that the approach of UVNNS can improve the coverage ratio, connectivity and the survivability. Compared with the random deployment approach, the performance metrics of UVNNS are better than that of random deployment approach (RDA) in the same UV network. Therefore, the deployment of the 3D UV network can be accomplished efficiently with a minor impact of obstacles.

Castration-resistant prostate cancer (CRPC) is a progressed stage of prostate cancer, which requires better understanding of the mechanisms and remains an unmet clinical need. As a common oncogene, K-Ras is associated with malignant behavior in different types of tumors but its role in CRPC is unknown. The present study aims to find the mechanism of K-Ras in CRPC and whether it can be used as a crucial molecule for the treatment of CRPC. For this purpose, tissue samples from primary prostate cancer (PPC) and CRPC patients were analyzed by immunohistochemistry and the data showed that K-Ras was elevated in CRPC. More importantly, higher K-Ras expression was related to a shorter recurrence-free survival time in patients with CRPC. In addition, K-Ras promoted the invasion, migration, and drug resistance of CRPC cells by activation of PLCε/PKCε signaling pathway. Meanwhile, the inhibitor of K-RasG12C mutants was able to inhibit malignant behavior of CRPC cells in vitro and in vivo. Inhibitors of K-RasG12C mutants have entered clinical trials. Taken together, the study shows that K-Ras may activate PKCε through PLCε, resulting in the alterations of malignant behavior of CRPC. Inhibitor 9, an inhibitor of the K-RasG12C mutant, has a strong anti-tumor effect in CRPC, which potentially suggests that inhibitors of this nature may serve as a promising treatment for CRPC.

Senile osteoporosis (SOP) is a related disease of systematic degenerative changes in bones during natural aging. Increasing age is an important factor in its pathogenesis. This experiment was to evaluate the comprehensive effect of calcium with vitamin D3 (CaD) on SOP based on multilayer perception (MLP)-artificial neural network (ANN) methods. 15-month-old male Sprague-Dawley rats were administered CaD for 2 months, while 3-, 6-, 9-, 12-, 15- and 17-month-old rats were used as the mature or aging control groups. We detected the bone mass and bone mineral density (BMD), performed biomechanical testing and measured micro-CT properties to evaluate the degree of osteoporosis. Levels of alkaline phosphatase (ALP) and tartrate-resistant acid phosphatase (TRACP), and the ratio of ALP to TRACP both in serum and bone were measured for the evaluation of the bone turnover rate. The bone mRNA and protein expression of ATP6v0d2, IGF-1, BMP2, M-CSF, Wnt5a and TGF-β1 were detected by western blotting (WB), immunofluorescence (IF) and quantitative real time polymerase chain reaction (qRT-PCR) for evaluating bone metabolism in the bone microenvironment. The MLP-ANN model was constructed and used to evaluate the importance of related parameters and the comprehensive action of CaD. Our data showed that bone mass, BMD, maximal load, ultimate displacement, ALP and TRACP in serum and tibia, and the protein and mRNA expressions of ATP6v0d2, IGF-1, BMP2, M-CSF, Wnt5a and TGF-β1 in tibia reached a peak in 6 m rats, and then were gradually decreased with the increase of age to the lowest in 17 m rats. This study demonstrated the degeneration of the bone structure and bone metabolism in SOP rats during the aging process of rats aged 3 to 17 months. CaD could effectively increase bone mass and bone strength, alleviate the degradation of the bone microstructure and rebalance bone remodeling. In addition, the MLP model was a comprehensive method for evaluating the effects of drugs on SOP, which provided a new direction for future drug and nutrition evaluation.

Hand tracking is an important part of Human-Computer Interaction (HCI). Computer vision based hand tracking can free the users and help HCI more natural. Hand area should be cropped from the image before hand tracking. Inspired by the success of deep learning, we use CNN to crop the hand area. We proposed a new method named FACPM to crop hand area with attention mechanism and Feature pyramid networks. We adopt VGG as base network, to improve the speed of the framework, unimportant network is cut off. FPN model is used for different scale features and can perform better on tiny feature. Attention model is used for distributing the weights of different features in FPN. A better cropped image can lead to a more precise 3D hand pose estimation and can be used in hand tracking.

Terrorism is a common threat to mankind, and fighting terrorism is the responsibility of every country. Aiming at suspects prediction of terrorist attacks, five methods based on machine learning are used to sort the recorded data of terrorist attacks quantitatively. Based on Logical Regression, Decision Tree, Gaussian Bayesian Network, AdaBoost and Random Forest fitting model, the terrorist attacks recorded, Global Terrorism Database(GTD), in 2015 and 2016 were used as experimental data. The most likely suspects for each attack and the possibility of these attacks, which may be launched by the five most harmful terrorist organizations, will be sorted out and the ranking of these attacks launched by each terrorist organization in the incident will be shown out. The results show that Decision Tree has the highest accuracy and the precision among the candidates, and Gaussian Bayesian Network is close behind.

Ethnopharmacological relevance Kidney deficiency is the main pathogenesis of osteoporosis based on the theory of “kidney governing bones” in traditional Chinese medicine (TCM). Osteoporosis is a systemic disease; kidney deficiency influences the growth, aging and reproduction of human body, reflecting in endocrine, nerve, immunity, metabolism and other functions. Multi-target drugs composed of natural non-toxic products from kidney-reinforcing herbs, are being investigated for the treatment of osteoporosis. Therefore, it is necessary and imperative to develop an objective and comprehensive method to evaluate and compare the effects of herbs with listed drugs. Aim of the study This study was designed to evaluate and compare the therapeutic effects and the underlying molecular mechanism of the combined extracts of Epimedii Folium and Ligustri Lucidi Fructus (EL) with Raloxifene hydrochloride (RH) in ovariectomy (OVX)-induced postmenopausal osteoporosis (PMOP) rats based on the multi-layer perception (MLP)-artificial neural network (ANN) model. Materials and methods Female SD rats were subjected to either sham surgery (n = 8) or bilateral OVX (n = 48). One week after recovering from surgery, the OVX-induced rats were randomly divided into three groups: OVX model group (n = 32, every 8 rats were killed at the end of the 5th, 9th, 11th or 13th week after OVX), EL group (treated with EL 0.35 g/kg, n = 8), and RH group (treated with RH 6.25 mg/kg, n = 8). The rats in the treatment groups were administrated once a day for 12 weeks, then sacrificed. We observed bone mass and quality, bone remodeling, the function of estrogen and TGF-β1/Smads pathway in all rats. Results Both EL and RH could increase bone mineral density, enhance bone strength, relieve bone micro-structure degeneration, re-balance bone remodeling, regulate estrogen dysfunction, and up-regulate TGF-β1 expression. The evaluation of the MLP-ANN model showed that EL and RH had markedly anti-PMOP effects, and there was no significant difference in the comprehensive evaluation of anti-osteoporosis between the two drugs. However, RH had better effects on bone mass and quality and TGF-β1/Smads pathway than EL; EL had better effects on estrogen function than RH. Conclusion Combined extracts of Epimedii Folium and Ligustri Lucidi Fructus (EL) exhibited bone-protective effects on PMOP. The MLP-ANN method evaluated the efficacy of drugs more comprehensively, which provided a new direction for the evaluation and comparison of drugs.

Background Context To our knowledge, there is no study that has systematically analyzed the relationship between C1 transpedicular screw trajectory and V3 segment of vertebral artery (VA V3 segment). Purpose To study the relationship between C1 transpedicular screw trajectory and VA V3 segment. Study Design A morphologic computed tomography angiography (CTA) analysis of the spatial relationship between C1 transpedicular screw trajectory and VA V3 segment. Methods Measurements were made on a workstation by using CTA data of 62 patients. Firstly, parameters related to the relationship between C1 vertebral artery groove (VAG) and vertebral artery (VA) were measured: (A) the shortest distance between the posterosuperior aspect of C1 posterior arch and VA; (B) distance between the outer aspect of VAG and VA; (C) distance between midpoint of VAG and VA; and (D) distance between the inner aspect of the VAG and VA. Then, the central axis of trajectory perpendicular to the coronal plane (axis P) and the central axis of trajectory with a medial inclination (axis M) were designed for the basis of measurements. Parameters related to the relationship between axis P/M and VA V3 segment were measured respectively: (E, E′), distance between insertion point and anterior aspect of VA along axis P/M; (F, F′), the shortest distance between axis P/M and the outer cortex of C1 transverse foramen; and (G, G′), the narrowest width of C1 internal medullary canal along axis P/M. Result A, B, C, and D were 1.7±1.0 mm, 1.6±0.9 mm, 1.5±0.7 mm, 2.3±1.1 mm, respectively. E, E′ were 5.5±1.7 mm and 4.1±2.3 mm. F, F′ were 1.9±0.7 mm and 2.9±0.7 mm. G, G′ were 3.7±1.4 mm and 4.8±1.2 mm. There was a little interspace between atlas VAG and VA, which was mainly filled with venous plexus. Conclusions There is a close relationship between C1 transpedicular screw trajectory and VA V3 segment. Trajectory with medial inclination technique is suggested especially for female patients.

Ethnopharmacological relevance Fructus Arctii is the dried ripe fruit of Arctium lappa L. (family Asteraceae). It is a well-known Chinese Materia Medica that was included in the Chinese pharmacopoeia because of its traditional therapeutic actions, such as heat removal, detoxification, and elimination of swelling. Since ancient times Fructus Arctii has been used extensively in a number of classical drug formulas to treat type 2 diabetes mellitus. Modern pharmacological studies have shown that certain components of Fructus Arctii have multiple physiological activities on type 2 diabetes and its complications. Aim of the study We have reported the inhibitory effect of total lignans from Fructus Arctii (TLFA) on aldose reductase, the key enzyme in the polyol pathway, which is considered to be closely related to the onset of diabetic retinopathy (DR). The present study aimed to observe the preventive and therapeutic effects of TLFA on DR in Streptozotocin (STZ)-induced DR rats. Materials and methods TLFA was prepared from Fructus Arctii and its content was determined using UV spectrophotometry. The DR model was induced by STZ in Wistar rats. For DR prevention, the animals were gavaged once daily for 9 weeks with TLFA (1.38, 0.69, and 0.35 g/kg/day) as soon as they were confirmed as diabetes models. Pathological changes to retinal tissues and the expression of vascular endothelial growth factor (VEGF) and protein kinase C (PKC) in the retina were detected after TLFA treatment. The effects of TLFA on blood glucose levels and body weight were also observed. For DR treatment, the animals were gavaged once daily for 12 weeks with TLFA (1.38 and 0.69 g/kg/day) at 3 months after they were confirmed as diabetes models. The therapeutic effect was studied using quantitative detection of blood-retina barrier (BRB) breakdown via an Evans Blue leakage assay. Results For DR prevention, after 9 weeks of TLFA administration, histopathological examination of retinal tissue showed that TLFA improved the lesions in the retina. Changes to retinal microstructures such as capillaries, ganglion cells, bipolar cells, and the membrane disk examined by electron microscopy further confirmed that TLFA has a preventive effect on retinopathy. Terminal deoxynucleotidyl Transferase-mediated dUTP nick end labeling (TUNEL) detection showed that TLFA could inhibit retinal cell apoptosis in the diabetic rats, and fasting blood glucose (FBG) levels of rats in the TLFA-treated groups decreased during the experiment. For DR treatment, after 3 months of administration, the amount of dye leakage in the TLFA-administered groups was reduced by more than 50% compared with that in the model group, which indicated that TLFA has a therapeutic effect on middle and late DR. Messenger RNA (mRNA) expression of VEGF and PKCβ2 in the retina detected by real-time fluorescent quantitative reverse transcription-polymerase chain reaction (FQ-RT-PCR) showed that TLFA could inhibit the expression of them, which was consistent with the results of immunohistochemistry (IHC). Conclusion TLFA has a preventive and therapeutic effect on DR. Its mechanism of action on DR is related to inhibiting PKC activation and blocking VEGF elevation.

Meibomian gland dysfunction (MGD) is an epidemic chronic ocular inflammation. However, little is known about its effective treatment. Here, this study identified important MGD-related genes, core regulators, and potential drugs and their targets though integrating a series of bioinformational analyses. First, there were 665 differentially expression genes (DEGs) were identified. Then, 56 coexpression modules were exacted based on the expression of DEGs and their interactors. Moreover, core transcription factors (TF) significantly regulated modules were identified, including RELA, HIF1A, SIRT1, and MYC, which related to variety of eye diseases. Finally, the prediction of potential drugs and the identification of their target were performed. The results showed that artenimol, copper, and glutathione may have the remarkable curative effect or the toxicology to MGD. Moreover, their targets module gene LDHA (lactate dehydrogenase A), ENO1 (enolase 1), ALB (albumin), and PKM (pyruvate kinase M) are play important role in eye diseases. It suggests that these potential drugs may be useful for the treatment of MGD by acting on their targets. It provides valuable references for drug redirection and new drug development for drug developers, and provides individualized treatment strategies for tarsal gland dysfunction.

Objective To investigate how hepatocyte cell adhesion molecule (hepaCAM) regulates cancer energy metabolism through hypoxia-inducible factor (HIF-1α) in renal cell carcinoma (RCC). Materials and Methods The expression of hepaCAM and HIF-1α in RCC tissue samples was examined by immunohistochemistry. Glucose consumption and lactate production assays were used to detect metabolic activity in RCC cell lines. P65 and IκB kinase (IKKβ) mRNA and protein expression were detected using quantitative real-time polymerase chain reaction and western blotting, respectively. Nuclear translocation of P65 was observed by immunofluorescence staining after re-expressing hepaCAM. The luciferase reporter assay was applied to validate the transcriptional activity of HIF-1α. Results HIF-1α expression was elevated and hepaCAM suppressed in RCC compared with adjacent normal tissues. Furthermore, hepaCAM re-expression significantly decreased glycolytic metabolism in RCC cell lines, and reduced HIF-1α, IKKβ, and P65 expression. The expression of HIF-1α, GLUT1, LDHA, and PKM2 were further reduced with combined hepaCAM overexpression and treatment with the NF-κB inhibitor BAY11-7082, compared to hepaCAM overexpression alone. Additionally, hepaCAM decreased the transcriptional activity of HIF-1α and blocked P65 nuclear translocation by the NF-κB pathway. Conclusion Our data suggest that hepaCAM suppresses the Warburg effect via the HIF-1α/NF-κB pathway in RCC, which is a facilitating factor in hepaCAM-reduced tumorigenesis.

Gesture recognition is an important way of human-computer interaction. With time going on, people are no longer satisfied with gesture recognition based on wearable devices, but hope to perform gesture recognition in a more natural way. Computer vision-based gesture recognition can transfer human feelings and instructions to computers conveniently and efficiently, and improve the efficiency of human-computer interaction significantly. The gesture recognition based on computer vision is mainly based on hidden Markov, dynamic time rounding algorithm and neural network algorithm. The process is roughly divided into three steps: image collection, hand segmentation, gesture recognition and classification. This paper reviews the computer vision-based gesture recognition methods in the past 20 years, analyses the research status at home and abroad, summarizes its current development, the advantages and disadvantages of different gesture recognition methods, and looks forward to the development trend of gesture recognition technology in the next stage.

SLE might affect all aspects of life including sexual functioning; previous study found that body image disturbance (BID) was the most powerful predictors of impaired partner relationships and sexual function. The current study investigated the relationship among disease parameters, quality of life, the psychological status, BID, and sexual problems in Chinese patients with SLE. A self-report survey design was administered to 168 SLE patients and 210 healthy individuals. Our results showed that 86 (55.1%) SLE patients reported impaired relationships with a sexual partner or partners, and 100 (64.1%) patients reported impaired sexual function which were significantly higher than the control group (31.6%, 35.7%, rep.). Age, marital status, depression, and BIDQ were the most powerful predictors of impaired partner relationships, while BIDQ3 and education, disease activity, and depression were the most significant causes of impaired sexual function. The study for first time reported Chinese SLE patients had sexual problems and BID was associated with sexual problems. So, early detection and interventions might not only rehabilitate the patients and their loved ones, but also improve overall health outcomes and reduce the direct and indirect costs of their medical care.

Premature ovarian failure (POF) is a heterogeneous disorder characterized by amenorrhea, infertility, lack of estrogen, and elevated gonadotropin levels before the age of 40. Most POF females have fertility problems due to defects of germ cell development or complete ovarian failure. Though hormonal therapy could partially restore ovarian function, effects of these treatments are transient, much less for regeneration and repair of the ovaries. With the rise of regenerative medicine, stem cells transplantation gives new hope for repairing POF related ovaries damage. Studies from animal models indicate that stem cell transplantation can, at least in part, repair ovarian structure, improve ovarian function and fertility in POF, i.e., the bone marrow mesenchymal stem cells (BMSCs) and umbilical cord mesenchymal stem cells (UC-MSCs) can differentiate to ovaries and facilitate fertility recovery. However, the underlying mechanisms remain unclear, which limits their clinical applications. Here, we discuss the recent progress on the clinical applications of stem cells for POF treatment, which would become promising therapies for POF patients.

Ethnopharmacological relevance Aerial parts of Peganum harmala Linn (APP) is used as traditional medical herb in Uighur medicine in China, and it is traditionally used for treatment of cough and asthma.The aim of the present study is to evaluate the antitussive, expectorant and bronchodilating effects of extract and fractions (alkaloids and flavonoids) from APP, and to support its folk use with scientific evidence, and lay a foundation for its further researches. Materials and methods APP was extracted with 50% ethanol by reflux, extracts were concentrated in vacuum to afford total extract of APP (EXT). EXT was separated to provide alkaloid fraction (ALK) and flavonoid fraction (FLA) by macroporous resin. Antitussive evaluations were carried out with cough models in mice and guinea pigs induced by ammonia liquor, capsaicin, and citric acid. Phenol red secretion experiments in mice were performed to evaluate the expectorant activity. Bronchodilating activities were evaluated with a bronchoconstrictive challenge induced by acetylcholine chloride and histamine in guinea pigs. Results In all the three antitussive tests, the EXT and ALK could significantly inhibit the frequency of cough, and prolong the cough latent period in animals. High dose of EXT (1650 mg/kg) and ALK (90 mg/kg) in mice and in guinea pigs created therapeutic activities as good as that of codeine phosphate (30 mg/kg). EXT could significantly increase phenol red secretion in mice for 0.64, 1.08 and 1.29 fold averagely at dosages of 183, 550, and 1650 mg/kg, ALK for 0.63, 0.96, 1.06 fold averagely at dosages of 10, 30, and 90 mg/kg, and ammonium chloride (1500 mg/kg, standard expectorant drug) for 0.97 fold, comparing with control group. Aminophylline could dramatically prolong the preconvulsive time for 162.28% in guinea pigs, EXT for 67.34%, 101.96% and 138.00% at dosages of 183, 550, and 1650 mg/kg, ALK for 55.47%, 97.74% and 126.77% at dosages of 10, 30, and 90 mg/kg, and FLA for 84.69%, 95.94% and 154.52% at dosages of 10, 30, and 90 mg/kg, comparing with pretreatment. Conclusions APP is an effective traditional folk medicine for the treatment of cough with potent antitussive, expectorant and bronchodilating activities. The alkaloid fraction is proved to be the most effective components of APP. The alkaloids from APP may be valuable lead compounds for drug development of respiratory diseases.

A multi-perspective describing method for emotional speech is proposed. Three perspectives - cognitive, psychological, physiological ones and seven phonetic evaluation features are involved, with which it is expected to describe speech emotion more completely and to bridge the gap between emotions and acoustic signals. A series of experiments including cognition tests and corpus annotation are implemented. Although preliminarily, the results still reveal the effectiveness of the method.

This paper examined the UNESCO PBL teacher training's effect through the investigation, and what problem still exist in the training, then we found the training effect is good, and this training really effective. So under the project based learning environment, the teacher training pattern can promote high effective teacher training, this can provide reference for the later teacher training.

A novel design of combination of hybrid system and zoom system in hyperspectral detection imaging is discribed based on the application of harmonic diffractive element (HDE) compared with conventional system. The principles of HDE and infrared zoom hyspectral detection system are presented and the results of a example design are given. It is show that the resolution of this hybrid system is improved and the light power received by Cassegrain system is enlarged. Wavefront aberrations are less than 1/4 wavelength and Modulation Transfer Function in the dual band on each zoom focus location approaches or attains the diffraction limit on 20 cycles/mm.
