Introduction: Artificial intelligence (AI) is one of the most considered topics of the current time. AI has the power to bring revolutionary improvements to the world of technology not only in the field of computer science but also in other fields like medical sciences. Objectives: This paper assumes the adoption of appropriate AI engineering principals in previous studies, and focusses on providing a structured review of the impact of AI on human society and the individual human being as a technology user. Additionally, it opens a window on how the future will look like in terms of AI and personalised medicine. Methods: The paper employed a qualitative research approach and data were collected through a structured literature review. Twenty-three peer reviewed papers were identified and analysed in relation to their relevance to the study. Results: Previous studies show a positive impact on users' behaviour is expected in supporting their healthcare needs especially in decision-making, personalised treatment and future diseases prediction, and that integrating users in studying AI impact is essential to test possible implications of the technology. Conclusion: Results indicate that without a clear understanding of why patients need AI, or how AI can support individuals with their healthcare needs, it is difficult to visualise the kinds of AI applications that have a meaningful and sustainable impact the daily lives of individuals. Therefore, there is an emerging need to understand the impact of AI technology on users' behaviour to maximise the potential benefits of AI technology.

The maintenance of visual acuity during active and passive body motion is ensured by gaze-stabilizing reflexes that aim at minimizing retinal image slip. For the optokinetic reflex (OKR), large-field visual motion of the surround forms the essential stimulus that activates eye movements. Properties of the moving visual world influence cognitive motion perception and the estimation of visual image velocity. Therefore, the performance of brainstem-mediated visuo-motor behaviors might also depend on image scene characteristics. Employing semi-intact preparations of mid-larval stages of Xenopus laevis tadpoles, we studied the influence of contrast polarity, intensity, contour shape and different motion stimulus patterns on the performance of the OKR and multi-unit optic nerve discharge during motion of a large-field visual scene. At high contrast intensities, the OKR amplitude was significantly larger for visual scenes with a positive contrast (bright dots on a dark background) compared with those with a negative contrast. This effect persisted for luminance-matched pairs of stimuli, and was independent of contour shape. The relative biases of OKR performance along with the independence of the responses from contour shape were closely matched by the optic nerve discharge evoked by the same visual stimuli. However, the multi-unit activity of retinal ganglion cells in response to a small single moving vertical edge was strongly influenced by the light intensity in the vertical neighborhood. This suggests that the underlying mechanism of OKR biases related to contrast polarity directly derives from visual motion-processing properties of the retinal circuitry.

Perera et al. report that balance impairment relates to deficits in intermittent and continuous control systems proposed to maintain upright stance. The deficits may be reversible with the experimental therapy, pedunculopontine nucleus deep brain stimulation., Impaired balance is a major contributor to falls and diminished quality of life in Parkinson’s disease, yet the pathophysiology is poorly understood. Here, we assessed if patients with Parkinson’s disease and severe clinical balance impairment have deficits in the intermittent and continuous control systems proposed to maintain upright stance, and furthermore, whether such deficits are potentially reversible, with the experimental therapy of pedunculopontine nucleus deep brain stimulation. Two subject groups were assessed: (i) 13 patients with Parkinson’s disease and severe clinical balance impairment, implanted with pedunculopontine nucleus deep brain stimulators; and (ii) 13 healthy control subjects. Patients were assessed in the OFF medication state and blinded to two conditions; off and on pedunculopontine nucleus stimulation. Postural sway data (deviations in centre of pressure) were collected during quiet stance using posturography. Intermittent control of sway was assessed by calculating the frequency of intermittent switching behaviour (discontinuities), derived using a wavelet-based transformation of the sway time series. Continuous control of sway was assessed with a proportional–integral–derivative (PID) controller model using ballistic reaction time as a measure of feedback delay. Clinical balance impairment was assessed using the ‘pull test’ to rate postural reflexes and by rating attempts to arise from sitting to standing. Patients with Parkinson’s disease demonstrated reduced intermittent switching of postural sway compared with healthy controls. Patients also had abnormal feedback gains in postural sway according to the PID model. Pedunculopontine nucleus stimulation improved intermittent switching of postural sway, feedback gains in the PID model and clinical balance impairment. Clinical balance impairment correlated with intermittent switching of postural sway (rho = − 0.705, P < 0.001) and feedback gains in the PID model (rho = 0.619, P = 0.011). These results suggest that dysfunctional intermittent and continuous control systems may contribute to the pathophysiology of clinical balance impairment in Parkinson’s disease. Clinical balance impairment and their related control system deficits are potentially reversible, as demonstrated by their improvement with pedunculopontine nucleus deep brain stimulation.

Previous evidence indicates that the brain stores memory in two complementary systems, allowing both rapid plasticity and stable representations at different sites. For memory to be established in a long-lasting neocortical store, many learning repetitions are considered necessary after initial encoding into hippocampal circuits. To elucidate the dynamics of hippocampal and neocortical contributions to the early phases of memory formation, we closely followed changes in human functional brain activity while volunteers navigated through two different, initially unknown virtual environments. In one condition, they were able to encode new information continuously about the spatial layout of the maze. In the control condition, no information could be learned because the layout changed constantly. Our results show that the posterior parietal cortex (PPC) encodes memories for spatial locations rapidly, beginning already with the first visit to a location and steadily increasing activity with each additional encounter. Hippocampal activity and connectivity between the PPC and hippocampus, on the other hand, are strongest during initial encoding, and both decline with additional encounters. Importantly, stronger PPC activity related to higher memory-based performance. Compared with the nonlearnable control condition, PPC activity in the learned environment remained elevated after a 24-h interval, indicating a stable change. Our findings reflect the rapid creation of a memory representation in the PPC, which belongs to a recently proposed parietal memory network. The emerging parietal representation is specific for individual episodes of experience, predicts behavior, and remains stable over offline periods, and must therefore hold a mnemonic function.

Understanding the mechanisms of symptoms that are insufficiently explained by organic dysfunction remains challenging. Recently, it has been proposed that such "functional symptoms" are based on erroneous sensory processing in the central nervous system (CNS), with internal expectations dominating sensory inputs. In a pilot study, we used a head motor control set-up to assess the interplay between sensory input and expectation on the example of patients with functional dizziness. Eight patients and 11 age-matched healthy controls performed large active eye-head gaze shifts towards visual targets in the natural situation and with the head moment of inertia 3.3-fold increased. The latter induces head oscillations and the expected sensory outcome of the movement, estimated in the CNS, does not match the actual sensory input. Head oscillations were assessed in patients and in healthy subjects and compared to prior results from patients with organic disease (vestibular loss and cerebellar ataxia). Passive and interactive virtual reality (VR) environments are becoming increasingly popular in the field of behavioral neuroscience. While the technique was originally developed for human observers, corresponding applications have been adopted for the research of visual-driven behavior and neural circuits in animals. RGB color reproduction using red, green and blue primary color pixels is generally calibrated for humans, questioning if the distinct parameters are also readily transferable to other species. In particular, a visual image in the RGB color space has a clearly defined contrast pattern for humans, but this may not necessarily be the case for other mammals or even non-mammalian species, thereby impairing any interpretation of color-related behavioral or neuronal results. Here, we present a simple method to estimate the sensitivity of animals to the three primary colors of digital display devices based on the performance of object motion-driven visuo-motor reflexes and demonstrate differences in the color sensitivity between Xenopus laevis and Ambystoma mexicanum (Axolotl). Human navigation is generally believed to rely on two types of strategy adoption, route-based and map-based strategies. Both types of navigation require making spatial decisions along the traversed way although formal computational and neural links between navigational strategies and mechanisms of value-based decision making have so far been underexplored in humans. Here we employed functional magnetic resonance imaging (fMRI) while subjects located different objects in a virtual environment. We then modelled their paths using reinforcement learning (RL) algorithms, which successfully explained decision behavior and its neural correlates. Our results show that subjects used a mixture of route and map-based navigation and their paths could be well explained by the model-free and model-based RL algorithms. Furthermore, the value signals of model-free choices during route-based navigation modulated the BOLD signals in the ventro-medial prefrontal cortex (vmPFC), whereas the BOLD signals in parahippocampal and hippocampal regions pertained to model-based value signals during map-based navigation. Our findings suggest that the brain might share computational mechanisms and neural substrates for navigation and value-based decisions such that model-free choice guides route-based navigation and model-based choice directs map-based navigation. These findings open new avenues for computational modelling of wayfinding by directing attention to value-based decision, differing from common direction and distances approaches.

To explore mechanisms of object integration, the present experiments examined how completion of illusory contours and surfaces modulates the sensitivity of localizing a target probe. Observers had to judge whether a briefly presented dot probe was located inside or outside the region demarcated by inducer elements that grouped to form variants of an illusory, Kanizsa-type figure. From the resulting psychometric functions, we determined observers' discrimination thresholds as a sensitivity measure. Experiment 1 showed that sensitivity was systematically modulated by the amount of surface and contour completion afforded by a given configuration. Experiments 2 and 3 presented stimulus variants that induced an (occluded) object without clearly defined bounding contours, which gave rise to a relative sensitivity increase for surface variations on their own. Experiments 4 and 5 were performed to rule out that these performance modulations were simply attributable to variable distances between critical local inducers or to costs in processing an interrupted contour. Collectively, the findings provide evidence for a dissociation between surface and contour processing, supporting a model of object integration in which completion is instantiated by feedforward processing that independently renders surface filling-in and contour interpolation and a feedback loop that integrates these outputs into a complete whole. Acute unilateral lesions of vestibular graviceptive pathways from the otolith organs and semicircular canals via vestibular nuclei and the thalamus to the parieto-insular vestibular cortex regularly cause deviations of perceived verticality in the frontal roll plane. These tilts are ipsilateral in peripheral and in ponto-medullary lesions and contralateral in ponto-mesencephalic lesions. Unilateral lesions of the vestibular thalamus or cortex cause smaller tilts of the perceived vertical, which may be either ipsilateral or contralateral. Using a neural network model, we previously explained why unilateral vestibular midbrain lesions rarely manifest with rotational vertigo. We here extend this approach, focussing on the direction-specific deviations of perceived verticality in the roll plane caused by acute unilateral vestibular lesions from the labyrinth to the cortex. Traditionally, the effect of unilateral peripheral lesions on perceived verticality has been attributed to a lesion-based bias of the otolith system. We here suggest, on the basis of a comparison of model simulations with patient data, that perceived visual tilt after peripheral lesions is caused by the effect of a torsional semicircular canal bias on the central gravity estimator. We further argue that the change of gravity coding from a peripheral/brainstem vectorial representation in otolith coordinates to a distributed population coding at thalamic and cortical levels can explain why unilateral thalamic and cortical lesions have a variable effect on perceived verticality. Finally, we propose how the population-coding network for gravity direction might implement the elements required for the well-known perceptual underestimation of the subjective visual vertical in tilted body positions.

The human head and eye rotate in coordination to rapidly project images of stationary targets from the visual space. Once a target is fixated, the head/eye complex maintains stability of the image, even while the head continues to move toward the target. The orientation of the head is constrained by Donders’ law, whereas the final orientation of the eye satisfies Listing’s constraint with respect to the final orientation of the head. The eye is rotated opposite to the movement of the head to compensate and maintain image stability. Using dynamic modeling, this brief investigates the underlying target fixating control mechanisms behind the head and eye movements. Finally, model output trajectories of the head and eye are compared with the corresponding experimentally observed trajectories.

Here we characterize persistent apogeotropic type of central positional nystagmus, and compare it with the apogeotropic nystagmus of benign paroxysmal positional vertigo involving the lateral canal. Nystagmus was recorded in 27 patients with apogeotropic type of central positional nystagmus (22 with unilateral and five with diffuse cerebellar lesions) and 20 patients with apogeotropic nystagmus of benign paroxysmal positional vertigo. They were tested while sitting, while supine with the head straight back, and in the right and left ear-down positions. The intensity of spontaneous nystagmus was similar while sitting and supine in apogeotropic type of central positional nystagmus, but greater when supine in apogeotropic nystagmus of benign paroxysmal positional vertigo. In central positional nystagmus, when due to a focal pathology, the lesions mostly overlapped in the vestibulocerebellum (nodulus, uvula, and tonsil). We suggest a mechanism for apogeotropic type of central positional nystagmus based on the location of lesions and a model that uses the velocity-storage mechanism. During both tilt and translation, the otolith organs can relay the same gravito-inertial acceleration signal. This inherent ambiguity can be resolved by a ` tilt-estimator circuit' in which information from the semicircular canals about head rotation is combined with otolith information about linear acceleration through the velocity-storage mechanism. An example of how this mechanism works in normal subjects is the sustained horizontal nystagmus that is produced when a normal subject is rotated at a constant speed around an axis that is tilted away from the true vertical (off-vertical axis rotation). We propose that when the tilt-estimator circuit malfunctions, for example, with lesions in the vestibulocerebellum, the estimate of the direction of gravity is erroneously biased away from true vertical. If the bias is toward the nose, when the head is turned to the side while supine, there will be sustained, unwanted, horizontal positional nystagmus (apogeotropic type of central positional nystagmus) because of an inappropriate feedback signal indicating that the head is rotating when it is not.

Objective. Many algorithms have been suggested for detection and sorting of spikes in extracellular recording. Nevertheless, it is still challenging to detect spikes in low signal-to-noise ratios (SNR). We propose a spike detection algorithm that is based on the fractal properties of extracellular signals and can detect spikes in low SNR regimes. Semi-intact spikes are low-amplitude spikes whose shapes are almost preserved. The detection of these spikes can significantly enhance the performance of multi-electrode recording systems. Approach. Semi-intact spikes are simulated by adding three noise components to a spike train: thermal noise, inter-spike noise, and spike-level noise. We show that simulated signals have fractal properties which make them proper candidates for fractal analysis. Then we use fractal dimension as the main core of our spike detection algorithm and call it fractal detector. The performance of the fractal detector is compared with three frequently used spike detectors. Main results. We demonstrate that in low SNR, the fractal detector has the best performance and results in the highest detection probability. It is shown that, in contrast to the other three detectors, the performance of the fractal detector is independent of inter-spike noise power and that variations in spike shape do not alter its performance. Finally, we use the fractal detector for spike detection in experimental data and similar to simulations, it is shown that the fractal detector has the best performance in low SNR regimes. Significance. The detection of low-amplitude spikes provides more information about the neural activity in the vicinity of the recording electrodes. Our results suggest using the fractal detector as a reliable and robust method for detecting semi-intact spikes in low SNR extracellular signals.

The automatic tendency to anthropomorphize our interaction partners and make use of experience acquired in earlier interaction scenarios leads to the suggestion that social interaction with humanoid robots is more pleasant and intuitive than that with industrial robots. An objective method applied to evaluate the quality of human–robot interaction is based on the phenomenon of motor interference (MI). It claims that a face-to-face observation of a different (incongruent) movement of another individual leads to a higher variance in one’s own movement trajectory. In social interaction, MI is a consequence of the tendency to imitate the movement of other individuals and goes along with mutual rapport, sense of togetherness, and sympathy. Although MI occurs while observing a human agent, it disappears in case of an industrial robot moving with piecewise constant velocity. Using a robot with human-like appearance, a recent study revealed that its movements led to MI, only if they were based on human prerecording (biological velocity), but not on constant (artificial) velocity profile. However, it remained unclear, which aspects of the human prerecorded movement triggered MI: biological velocity profile or variability in movement trajectory. To investigate this issue, we applied a quasi-biological minimum-jerk velocity profile (excluding variability in the movement trajectory as an influencing factor of MI) to motion of a humanoid robot, which was observed by subjects performing congruent or incongruent arm movements. The increase in variability in subjects’ movements occurred both for the observation of a human agent and for the robot performing incongruent movements, suggesting that an artificial human-like movement velocity profile is sufficient to facilitate the perception of humanoid robots as interaction partners.

Several regions of the brain are involved in smooth-pursuit eye movement (SPEM) control, including the cortical areas MST (medial superior temporal) and FEF (frontal eye field). It has been shown that the eye-movement responses to a brief perturbation of the visual target during ongoing pursuit increases with higher pursuit velocities. To further investigate the underlying neuronal mechanism of this nonlinear dynamic gain control and the contributions of different cortical areas to it, we recorded from MSTd (dorsal division of the MST area) neurons in behaving monkeys ( Macaca mulatta) during step-ramp SPEM (5–20°/s) with and without superimposed target perturbation (one cycle, 5 Hz, ±10°/s). Smooth-pursuit–related MSTd neurons started to increase their activity on average 127 ms after eye-movement onset. Target perturbation consistently led to larger eye-movement responses and decreasing latencies with increasing ramp velocities, as predicted by dynamic gain control. For 36% of the smooth-pursuit–related MSTd neurons the eye-movement perturbation was accompanied by detectable changes in neuronal activity with a latency of 102 ms, with respect to the eye-movement response. The remaining smooth-pursuit–related MSTd neurons (64%) did not reflect the eye-movement perturbation. For the large majority of cases this finding could be predicted by the dynamic properties of the step-ramp responses. Almost all these MSTd neurons had large visual receptive fields responding to motion in preferred directions opposite to the optimal SPEM stimulus. Based on these findings it is unlikely that MSTd plays a major role for dynamic gain control and initiation of the perturbation response. However, neurons in MSTd could still participate in SPEM maintenance. Due to their visual field properties they could also play a role in other functions such as self-motion perception.

Humans can discriminate whether a change in the direction of gravito-inertial force (GIF) is caused by body tilt or by linear translation. This ability, attributed to vestibular sensory fusion, is often examined by asking subjects to adjust an indicator to match their subjective earth-fixed vertical (SV). We used two different modalities, visual and haptic, to examine continuous adjustment during different combinations of roll rotation and linear translation on a hexapod motion device. We conclude that, in conditions of combined translational and rotational motion, the modality of indication plays a major role for the perception of verticality of the indicator.

ZusammenfassungVertikaler Nystagmus ist ein klinisches Zeichen einer zentralen Läsion. Downbeat und Upbeat Nystagmus unterscheiden sich jedoch bezüglich Lokalisation, Ätiologie, Pathophysiologie und Prognose. Diese sowie die aktuellen Therapieansätze sollen im folgenden Übersichtsartikel dargestellt werden.

Dynamic signals from multiple sensory channels must be integrated by the central nervous system to create a unified perception of self-motion and spatial orientation. Using immersive virtual environments, we altered the relative contribution of visual and inertial inputs and evaluated the effects on perceptuomotor outputs. Subjects seated in a tilting chair were exposed to a combined 0.25 Hz sinusoidal roll-tilt (+/-7.5 degrees) about the naso-occipital axis while viewing one of four visual conditions. One visual condition was in darkness, and the other three depicted 2 m of sinusoidal horizontal or vertical linear motion either synchronous or asynchronous with the roll-tilt. Subjects performed a perceptuomotor task of aligning a handheld object to gravitational vertical (GV) with the entire arm being free to move in six degrees of freedom. Subjects were tested with two objects, a joystick and glass of water, in counter-balanced order. Specific visual effects were as follows: (1) the phase leads of object tilt relative to chair/subject roll-tilt were affected by visual condition, (2) horizontal translation of the object was entrained with visual velocity, rather than with visual acceleration or maximum roll-tilt, and (3) when vertical visual motion was viewed during chair/subject roll-tilt, vertical object translation increased. Although the head-fixed scene meant visual vertical cues were always aligned with the subject's median sagittal plane, object tilt showed sensitivity to inertial roll-tilt (Gain0.5) which was not significantly different from the dark condition. Two object effects were found: (1) tilt deviation from GV was greater when wielding a joystick compared to a full glass of water, and (2) the phase of horizontal visual motion relative to subject roll tilt affected the joystick amplitude of horizontal translation but not the glass of water. In conclusion, an attentional shift driven by postural assumptions can account for the two object effects, however, the visual effects suggest that a process for deriving the net gravitoinertial force from visual and inertial cues is involved. Inertial signals dominated the perception of verticality, but visual linear translation affected the spatiotemporal dynamics of the manual motor responses during object wielding.

Nicotine has wellknown, unpleasant side effects, e.g., transient dizziness, nausea, and nicotine-induced nystagmus (NIN). To investigate factors influencing these effects, we addressed three questions: (1) Is the intensity of dizziness, nausea, NIN, and unsteadiness dependent on nicotine dosage? (2) Does the intensity of perceptual, ocular motor, vegetative effects, and postural imbalance correlate? (3) Do visual or vestibular motion stimuli produce and/or aggravate distressing dizziness and nausea? Sixty healthy non-smokers or occasional smokers participated; 40 were tested once before and six times after application of a nicotine nasal spray in doses of 1 mg or 2 mg with or without motion stimulation; 20 received a placebo nasal spray. Plasma nicotine concentrations were significantly related to nicotine dosage. Dizziness, nausea, NIN, and unsteadiness also depended on the nicotine dosage (p < 0.01).Nicotine blood concentration was a better predictor for the temporal dependence of nystagmus than nicotine dosage. Dizziness correlated highly with nausea (R = 0.63, p < 0.001). The degree of nicotine-induced nausea significantly correlated with postural imbalance. The time course of postural sway differed according to nicotine dosage and gender: for women, there was no clear relationship between sway magnitude and nicotine dosage, while men showed increased sway with higher dosage. Motion stimulation increased nicotine-induced dizziness and nausea, but did not significantly influence NIN or postural imbalance. Our data support the view that all measured adverse effects reflect dose-dependent nicotine-induced vestibular dysfunction. Additional motion stimulation aggravates dizziness and nausea, i.e., nicotine increases sensitivity to motion sickness.

Information about the future trajectory of a visual target is contained not only in the history of target motion but also in static visual cues, e.g., the street provides information about the car's future trajectory. For most natural moving targets, this information imposes strong constraints on the relation between velocity and acceleration which can be exploited by predictive smooth pursuit mechanisms. We questioned how cue-induced predictive changes in pursuit direction depend on target speed and how cue- and target-induced pursuit interact. Subjects pursued a target entering a +/-90 degrees curve and moving on either a homogeneous background or on a low contrast static band indicating the future trajectory. The cue induced a predictive change of pursuit direction, which occurred before curve onset of the target. The predictive velocity component orthogonal to the initial pursuit direction started later and became faster with increasing target velocity. The predictive eye acceleration increased quadratically with target velocity and was independent of the initial target direction. After curve onset, cue- and target-induced pursuit velocity components were not linearly superimposed. The quadratic increase of eye acceleration with target velocity is consistent with the natural velocity scaling implied by the two-thirds power law, which is a characteristic of biological controlled movements. Comparison with linear pursuit models reveals that the ratio between eye acceleration and actual or expected retinal slip cannot be considered a constant gain factor. To obey a natural velocity scaling, this acceleration gain must linearly increase with target or pursuit velocity. We suggest that gain control mechanisms, which affect target-induced changes of pursuit velocity, act similarly on predictive changes of pursuit induced by static visual cues.

Vestibular input is required for accurate locomotion in the dark, yet blind subjects’ vestibular function is unexplored. Such investigation may also identify visually dependent aspects of vestibular function. We assessed vestibular function perceptually in six congenitally blind (and 12 sighted) subjects. Cupula deflection by a transient angular, horizontal acceleration generates a related vestibular nerve signal that declines exponentially with time constant ≈4–7 s, which is prolonged to 15 s in the evoked vestibular-ocular reflex by the brain stem “velocity storage.” We measured perceptual velocity storage in blind subjects following velocity steps (overall perceptual vestibular time constant, experiment 1) and found it to be significantly shorter (5.34 s; range: 2.39–8.58 s) than in control, sighted subjects (15.8 s; P < 0.001). Vestibular navigation was assessed by subjects steering a motorized Bárány-chair in response to imposed angular displacements in a path-reversal task, “go-back-to-start” (GBS: experiment 2); and a path-completion task, “complete-the-circle” (CTC: experiment 3). GBS performances (comparing response vs. stimulus displacement regression slopes and r2) were equal between groups ( P > 0.05), but the blind showed worse CTC performance ( P < 0.05). Two blind individuals showed ultrashort perceptual time constants, high lifetime physical activity scores and superior CTC performances; we speculate that these factors may be inter-related. In summary, the vestibular velocity storage as measured perceptually is visually dependent. Early blindness does not affect path reversal performance but is associated with worse path completion, a task requiring an absolute spatial strategy. Although congenitally blind subjects are overall less able to utilize spatial mechanisms during vestibular navigation, prior extensive physical spatial activity may enhance vestibular navigation.

Throughout the last decade, a large number of electronic driver-assistance systems have been developed in order to improve vehicles’ comfort and safety. Up to now, they more or less all rely on odometric data. However in the future, a new form of information is likely to be used: visual sensors, e.g. cameras, can provide additional knowledge on traffic and the vicinity. This paper presents an inertially stabilized camera platform for high precision tracking of points of interest, that has been developed within the project FORBIAS (Research cooperation for biomimetic assistance systems). The inertial stabilization is based on a specially designed sensor system that uses MEMS components in connection with a biologically inspired control algorithm for adaptive enhancement and online calibration of the gaze stabilization system.

Downbeat nystagmus (DBN) is caused by an impairment of Purkinje cells in the flocculus. The decreased cerebellar inhibitory input affects otolith pathways. Since ocular and cervical vestibular evoked myogenic potentials (o-/cVEMP) test the otoliths, the VEMP were measured in DBN patients and in controls. Sixteen patients with DBN, 14 cerebellar oculomotor disorder patients without DBN (COMD), and 16 healthy controls were examined with o-/cVEMP. Computational modeling was used to predict VEMP differences between groups. DBN patients had significantly higher oVEMP peak-to-peak (PP) amplitudes than COMD patients without DBN and controls. Cervical VEMP did not differ. The computational model of DBN predicted a twofold oVEMP increase for DBN patients. These findings suggest an enhancement of the utriculo-ocular response. The unchanged cVEMP indicate no effect on the otolith-cervical reflex in DBN. Computational modeling suggests that the utriculo-ocular enhancement is caused by an impaired vertical neural integrator resulting in the increased influence of utricular signals. This also explains the gravitational dependence of DBN.

Downbeat nystagmus (DN), a fixation nystagmus with the fast phases directed downward, is usually caused by cerebellar lesions, but the precise etiology is not known. A disorder of the smooth-pursuit system or of central vestibular pathways has been proposed. However, both hypotheses fail to explain why DN is usually accompanied by gaze-holding nystagmus, which implies a leaky neural velocity-to-position integrator. Because three-dimensional (3-D) analysis of nystagmus slow phases provides an excellent means for testing both hypotheses, we examined 19 patients with DN during a fixation task and compared them with healthy subjects. We show that the presentation of DN patients is not uniform; they can be grouped according to their deficits: DN with vertical integrator leakage, DN with vertical and horizontal integrator leakage, and DN without integrator leakage. The 3-D analysis of the slow phases of DN patients revealed that DN is most likely neither caused by damage to central vestibular pathways carrying semicircular canal information nor by a smooth pursuit imbalance. We propose that the observed effects can be explained by partial damage of a brain stem-cerebellar loop that augments the time constant of the neural velocity to position integrators in the brain stem and neurally adjusts the orientation of Listing's plane.

Objective: The diagnosis of central paroxysmal positional nystagmus (CPPN) is challenging, and the mechanisms require further elucidation. This study aimed to determine the characteristics and mechanisms of CPPN. Methods: Seventeen patients with CPPN were subjected to analyses of their clinical findings, MRI lesions, and oculographic data on spontaneous and positional nystagmus. Results: The direction of CPPN was mostly aligned with that of the head motion during the positioning, and 3 types of CPPN were identified: downbeat nystagmus on straight-head hanging, upbeat nystagmus on uprighting, and apogeotropic nystagmus during supine head roll test. The direction of CPPN was aligned with the vector sum of the rotational axes of the semicircular canals that were normally inhibited during the positioning. The intensity of evoked nystagmus was at its peak initially and then decreased exponentially over time. The time constants (TC) of the vertical CPPN ranged from 3 to 8 seconds, which corresponds to the TC of the vertical rotational vestibulo-ocular reflex. Sixteen patients (94.1%) showed more than one type of CPPN. Furthermore, persistent downbeat or apogeotropic positional nystagmus was associated in 11 patients (64.7%). Most patients with CPPN from a circumscribed brain lesion showed an involvement of the cerebellar nodulus or uvula. Conclusion: CPPN may be ascribed to enhanced responses of the vestibular afferents due to lesions involving the nodulus and uvula. CPPN could be differentiated from benign paroxysmal positional nystagmus by positional nystagmus induced in multiple planes, temporal patterns of nystagmus intensity, and associated neurologic findings suggestive of central pathologies.

Downbeat nystagmus (DBN) is rarely caused by lesions in sites other than the cerebellar (para-)floccular lobe. We describe a case of DBN secondary to a hemorrhaged venous cavernoma at the pontomedullary junction. This case provides new insights into the neuro-anatomical substrate of DBN. We propose that DBN arises from lesions in a brainstem-cerebellar feedback loop, which comprises cells of the pontine paramedian tract (PMT).

Even though joint action is highly developed in humans, not much is known about motor control in physical joint-action tasks. Here we investigated a physical handover task: one subject sequentially passed wooden cubes to another without communicating verbally. Temporal parameters such as reaction time decreased on a trial-to-trial basis, showing that the efficiency of the task is optimized on-line by implicit negotiation between the partners. In contrast, the spatial position of the handover was found to be invariant and trial-independent. Thus, our results suggest that physical joint-action is guided by on-line adaptation and a priori assumptions.

A basic version of a 3-D mathematical model for simulation of otolithic control of binocular static eye position was extended by introducting excitatory commissural fibers between the vestibular nuclei, and physiological non-linearities: the force-response relationship of utricular neurons and a quadratic relationship between eye muscle innervation and force. These modifications appeared to be necessary in order to simulate the gain asymmetry of ocular counter-roll to lateral head tilt in patients with unilateral utricular loss. The current model can adequately simulate skew-torsion in patients with unilateral utricular loss, lesions of the vestibular nuclei, and central graviceptive pathway lesions. The direction of simulated skew-torsion corresponds satisfactorily to data from normals and patients with acute vestibular loss. The relatively low values of predicted eye deviations for peripheral vestibular lesions suggest that part of the effects seen in patients is caused by affection of the semicircular canals.

Our objective was to investigate how cooling of the arm and vision influence pointing movements in healthy subjects and patients with cerebellar limb ataxia due to clinically proven multiple sclerosis. An infrared video motion analysis system was used to record the unrestricted, horizontal pointing movements toward a target under three different conditions involving a moving, stationary, or imaginary target; a visual, or acoustic trigger; and vision or memory guidance. All three tasks were performed before and after cooling the arm in ice water. Patients had more hypermetric and slower pointing movements than controls under all tested conditions. Patients also had significantly larger three-dimensional finger sway paths during the postural phase and larger movement angles of the wrist joint. Memory-guided movements were the most hypermetric recorded in both groups. Cooling of the limb had no effect on amplitude or peak velocity of the pointing movement in either group under all tested conditions, but significantly reduced the three-dimensional finger sway path during the postural phase in patients with limb ataxia. Cooling-induced reduction of the finger sway was largest in those patients with the largest finger sway before cooling. In conclusion, the cooling-induced reduction of the proprioceptive afferent inflow, most probably of group I spindle afferents, reduces postural tremor of patients with cerebellar dysfunction.

One of the key computations performed in human brain is multi-sensory cue integration, through which humans are able to estimate the current state of the world to discover relative reliabilities and relations between observed cues. Mammalian cortex consists of highly distributed and interconnected populations of neurons, each providing a specific type of information about the state of the world. Connections between areas seemingly realize functional relationships amongst them and computation occurs by each area trying to be consistent with the areas it is connected to. In this paper using line-attraction dynamics and divisive normalization, we present a computational framework which is able to learn arbitrary non-linear relations between multiple cues using a simple Hebbian Learning principle. After learning, the network dynamics converges to the stable state so to satisfy the relation between connected populations. This network can perform several principle computational tasks such as inference, de-noising and cue-integration. By applying a real world multi-sensory integrating scenario, we demonstrate that the network can encode relative reliabilities of cues in different areas of the state space, over distributed population vectors. This reliability based encoding biases the network’s dynamics in favor of more reliable cues and realizes a near optimal sensory integration mechanism. Additional important features of the network are its scalability to cases with higher order of modalities and its flexibility to learn smooth functions of relations which is necessary for a system to operate in a dynamic environment.

International audience; In the present study, we compared the effects of two processing modes on the updating of the location and orientation of a previously viewed object in space during a guided walk without vision. In Experiment 1, in order to measure the error for initial perception of object's orientation, 12 subjects rotated a miniature model until it matched the memorized orientation of its counterpart object in space. In Experiment 2, they attempted either to keep track of the object continuously (in the object-centered [OC] task) or to estimate the object's perspective only at the terminal vantage point given the trajectory they walked (in the trajectory-centered [TC] task). Subjects indicated the location of the object by facing it, and then rotated the model in order to indicate its orientation from the new vantage point. Results showed that, with respect to the TC mode, the OC mode induced a slow-down of the subjects' self-paced locomotion velocity for both linear and angular movements, and a decrease of the latencies as well as smaller absolute errors for the orientation-of-the-object response. Mean signed errors on object's orientation were equivalent for both processing modes, suggesting that the latter induced different allocations of processing resources on a common representation of space updated by "path integration."

Human head and eye rotate in coordination to rapidly project images of targets from the visual space. Once a target is fixated, the head/eye complex maintains the stability of the image, even while the head continues to move. The dynamics of the head is constrained by Donders' Law whereas the final position of the eye satisfies Listing's constraint with respect to the final position of the head. The vestibuloocular reflex rotates the eye opposite to the forward movement of the head in order to compensate and maintain image stability. Using a dynamic model of the head and eye, the eye movement is modeled as a tracking control problem, where the tracking signal depends on the head movement trajectory. The torques required for the head and eye movements are computed by minimizing a suitable cost function. The paper computes the optimal torque and compares simulation results with experimentally measured data on human head/eye movement.

The rostral interstitial nucleus of the medial longitudinal fasciculus (riMLF) and the interstitial nucleus of Cajal (iC) are involved in the generation of vertical and torsional saccades and gaze holding.Midbrain lesions involving either riMLF or iC produce a tonic torsional ocular deviation to the contralesional side. Lesions of the iC are associated with an ipsilesional torsional nystagmus (TN). For the first time, we describe a patient with a unilateral midbrain lesion showing a contralesionally beating torsional nystagmus. This nystagmus was probably caused by a lesion involving the riMLF.NEUROLOGY 1996;47: 482-486

The control of head direction in humans walking along planned circular trajectories was investigated in this study. Five healthy volunteers were asked to walk at a constant speed along circular trajectories in the light and while blindfolded. Head and walking directions were analysed on a real-time basis. Head direction systematically anticipated changes in the direction of locomotion (by about 200 ms). The anticipation interval depended on the curvature of the circle. In the light, head orientation was deviated with respect to the walking direction, toward the inner concavity of the performed trajectory. The results suggest that head direction is controlled on a step-by-step basis in a predictive fashion. A 'go where you look' strategy seems to underlie steering along circular trajectories.

Although opioid-induced nausea and vomiting (OINV) is common and debilitating, its mechanism is still unclear. Recently, we suggested that opioids affect semicircular canal function and that this leads to a mismatch between canal input and other sensory information during head motion, which triggers OINV. Here, we assess if visual input is relevant for this mismatch. In a randomized-controlled crossover study 14 healthy men (26.9±3.4 years, mean±SD) were tested twice, once blindfolded and once with eyes open, with at least one-day washout. The opioid remifentanil was administered intravenously (0.15 μg/kg/min) for 60 minutes. After a thirty-minutes resting period, subjects' head and trunk were passively moved. Nausea was rated before remifentanil start (T0), before the movement intervention (T30) and after 60 minutes (T60) of administration. At rest (T0, T30), median nausea ratings were zero whether subjects were blindfolded or not. Movement triggered nausea independently of visual input (nausea rating 1.5/3.0 (median/interquartile range) in the blindfolded, 2.5/6 in the eyes-open condition, χ2(1) = 1.3, p = 0.25). As movement exacerbates OINV independently of visual input, a clash between visual and semicircular canal information is not the relevant trigger for OINV. To prevent OINV, emphasis should be put on head-rest, eye-closure is less important.

The capacity to understand goals and intentions emerges early and universally in humans and is a basic precondition for the interpretation and prediction of others' actions, be it other humans, animals, or even robots. It is unclear, however, how this goal attribution system is acquired, in particular with regard to the role of prior experience with the actor and visual characteristics that are necessary. In four preferential looking time experiments we examined how familiarity, appearance, and movement of different agents influence the capability of marmosets to perceive the behavior of these agents as goal directed. To this end we compared the monkeys' reactions to the same goal-directed actions performed by four different agents: a human actor, a conspecific, a monkey-like small robot, and a black box. The results showed that monkeys attributed goals to the human actor, the conspecific, and the robot, but not the box. Thus, the monkeys extended their capacity for goal attribution not only to familiar agents, but also to agents not previously encountered, provided that they had some conspecific-like features. Our results suggest that in non-human primates, the system for goal attribution does not require previous experience with a specific agent or agent-category, as long as it exhibits certain visual characteristics like face, body or legs. Furthermore, the results suggest that the capacity to attribute goals emerged very early during evolution and, at least in marmoset monkeys, does not necessarily require pre-learned associations in order to fulfill its function when dealing with unfamiliar agents.

The ability to infer intentions and predict actions enables coordinating of one's own actions with those of another human and allows smooth and intuitive interaction. The aim to achieve equally effective human-robot interactions is a crucial aspect of current robotic studies. Thus, we assume that studying human-human interaction provides valuable insights allowing to implement mutual intention recognition and action prediction in robotic systems. A common scenario of interaction, be it in everyday life or in an industrial setting, is that two or more agents share the same workspace and perform tasks without interference. If humans are involved, the robots should act sufficiently predictable to enable the human to attribute goals and predict motion trajectories. In the present work, we first analyzed how well a human recognizes the goal of another person entering the room, and whether this ability is deteriorated by concealing gaze direction of the other person. In a second setup, the same experiment was repeated by replacing the approaching person with a wheeled robot. On average, the distance at which subjects predicted the goal of the approaching agent was approx. 4 m and depended on subject and goal position, but not on the type of agent. However, goal attribution showed a considerable proportion of errors for the robot (19%), much less for a human with hidden gaze direction (6%), and almost none for a human with visible gaze (1%). Thus, our subjects apparently decided on the goal of the approaching agent without taking into account the reliability of directional cues, thus resulting in more errors. In a human-robot setting, such wrong predictions about robotic behavior may easily lead to dangerous situations. For smooth and safe interaction, it is therefore important to ameliorate the predictability of robotic actions.

International audience; Our objective is to improve legibility of robot navigation behavior in the presence of moving humans. We examine a human-aware global navigation planner in a path crossing situation and assess the legibility of the resulting navigation behavior. We observe planning based on fixed social costs and static search spaces to perform badly in situations where robot and human move towards the same point. To find an improved cost model, we experimentally examine how humans deal with path crossing. Based on the results we provide a new way of calculating social costs with context dependent costs without increasing the search space. Our evaluation shows that a simulated robot using our new cost model moves more similar to humans. This shows how comparison of human and robot behavior can help with assessing and improving legibility.

Recent findings in neuroscience suggest an overlap between brain regions involved in the execution of movement and perception of another’s movement. This so-called “action-perception coupling” is supposed to serve our ability to automatically infer the goals and intentions of others by internal simulation of their actions. A consequence of this coupling is motor interference (MI), the effect of movement observation on the trajectory of one’s own movement. Previous studies emphasized that various features of the observed agent determine the degree of MI, but could not clarify how human-like an agent has to be for its movements to elicit MI and, more importantly, what ‘human-like’ means in the context of MI. Thus, we investigated in several experiments how different aspects of appearance and motility of the observed agent influence motor interference (MI). Participants performed arm movements in horizontal and vertical directions while observing videos of a human, a humanoid robot, or an industrial robot arm with either artificial (industrial) or human-like joint configurations. Our results show that, given a human-like joint configuration, MI was elicited by observing arm movements of both humanoid and industrial robots. However, if the joint configuration of the robot did not resemble that of the human arm, MI could longer be demonstrated. Our findings present evidence for the importance of human-like joint configuration rather than other human-like features for perception-action coupling when observing inanimate agents.

When shifting gaze to foveate a new target, humans mostly choose a unique set of eye and head movements from an infinite number of possible combinations. This stereotypy suggests that a general principle governs the movement choice. Here, we show that minimizing the impact of uncertainty, i.e., noise affecting motor performance, can account for the choice of combined eye–head movements. This optimization criterion predicts all major features of natural eye–head movements—including the part where gaze is already on target and the eye counter-rotates—such as movement durations, relative eye–head contributions, velocity profiles, and the dependency of gaze shifts on initial eye position. As a critical test of this principle, we show that it also correctly predicts changes in eye and head movement imposed by an experimental increase in the head moment of inertia. This suggests that minimizing the impact of noise is a simple and powerful principle that explains the choice of a unique set of movement profiles and segment coordination in goal-directed action.

OBJECTIVE: Animal experiments have demonstrated that aminopyridines increase Purkinje cell excitability, and in clinical studies, 4-aminopyridine (4-AP) and 3,4-diaminopyridine (3,4-DAP) improved downbeat nystagmus. In this double-blind, prospective, crossover study, the effects of equivalent doses of 4-AP and 3,4-DAP on the slow-phase velocity (SPV) of downbeat nystagmus were compared. METHODS: Eight patients with downbeat nystagmus due to different etiologies (cerebellar degeneration [n = 1], bilateral vestibulopathy [n = 1], bilateral vestibulopathy and cerebellar degeneration [n = 1], Arnold-Chiari I malformation and cerebellar ataxia [n = 1], cryptogenic cerebellar ataxia [n = 4]) were included. They were randomly assigned to receiving a single capsule of 10 mg of 3,4-DAP or 4-AP followed by 6 days with no medication. One week later, the treatment was switched, that is, 1 single capsule (10 mg) of the other agent. Recordings with 3-dimensional video-oculography were performed before and 45 and 90 minutes after drug administration. RESULTS: Both medications had a significant effect throughout time (pre vs post 45 vs post 90) (F() = 8.876; P < 0.01). Following the administration of 3,4-DAP, mean slow velocity decreased from -5.68°/s (pre) to -3.29°/s (post 45) to -2.96°/s (post 90) (pre vs post 45/post 90 P < 0.01). In 4-AP, the mean SPV decreased from -6.04°/s (pre) to -1.58°/s (post 45) to -1.21°/s (post 90) (pre vs post 45/post 90 P < 0.00001). Both after 45 and after 90, the mean SPVs were significantly lower for 4-AP than for 3,4-DAP (P < 0.05). None of the patients reported serious side effects. CONCLUSION: Based on these results, 10-mg doses of 4-AP lead to a more pronounced decrease of the SPV of downbeat nystagmus than do equivalent doses of 3,4-DAP.

While moving through the world, humans as well as animals can make use of motion cues during both active and passive whole-body motion to track their own position in space. However, the functional neuroanatomy of self-motion processing remains poorly understood. In the present study we aimed to characterize brain networks reflecting whole-body self-motion experience. We used retrieval of previously experienced events, which is known to involve cortical representations of the modalities used to perceive these events. Recollection of self-motion experience may thus engage motor and sensory brain areas, reflecting the active or passive nature of the experienced movement, but may engage also common brain areas processing self-motion. We further compared the retrieval networks of self- and observed motion: even though actual action observation has been shown to recruit brain networks similar to those active during mental simulation, it is unclear to which extent recollection networks of these experiences overlap. Brain activation patterns were recorded using fMRI during mental simulation of recent episodes of (1) experiencing linear whole-body motion (active locomotion and passive transport) and (2) observing another person performing the same tasks. Following the experiential phase, participants recalled the episodes during a MR session. We found that primary sensorimotor brain areas dominate the composition of the recollection network of active walking, while recalling passive transport recruits higher level association areas. Common to both self-motion conditions was activation in the medial temporal lobe. Recollection of self-experienced and observed movement overlapped in motor planning areas. Our results provide evidence that the medial temporal lobe is specifically relevant for retrieval of self-motion information and that motor coding during action observation is reflected in recollection networks.

We introduce a novel approach for evaluation of neuronal tuning functions, which can be expressed by the conditional probability of observing a spike given any combination of independent variables. This probability can be estimated out of experimentally available data. By maximizing the mutual information between the probability distribution of the spike occurrence and that of the variables, the dependence of the spike on the input variables is maximized as well. We used this method to analyze the dependence of neuronal activity in cortical area MSTd on signals related to movement of the eye and retinal image movement.

1. Passive linear self-motion estimation along the X and Y axes was investigated in human subjects. 2. A target was viewed from a distance of 0.8 or 2.4 m from the starting point. Subjects were then blindfolded and transported toward the target on a distance of 3.2 m and back to the start. Acceleration was constant: +/- 0.2 or +/- 1 m/s2. The subjects pushed a button on both outward and return paths, when they passed the previously seen target. 3. The results showed anticipation of the target on the outward path, and not on the return. This was identical for both axes and both accelerations. 4. The data are in accord with a model of double integration of the otolith signal, suggesting that linear path integration is a basic sensory mechanism.
