The rat incisors obtained from rats perfused with high-calcium solution containing 30 to 50 mM CaCl2 were processed for rapid freeze/freeze-substitution and embedded in epoxy resin. GBHA staining, a histochemical staining for calcium, or unhydrously prepared sections revealed a large number of granular Ca-GBHA reactions in the enamel organ, most of which being located along the lateral plasma membranes of the ameloblasts. In the ameloblast layer, the reaction was negative in the presecretory stage, became intense in concert with the onset of enamel matrix formation, and remained so by the end of the transitional stage where the reaction gradually diminished. In the maturation stage, similar GBHA reactions emerged only in association with the smooth-ended ameloblasts and were absent in the ruffle-ended ameloblasts. Some granular reactions of much smaller diameter were occasionally noted in other cells of the enamel organ at both the secretory and maturation stages. Electron microscopy confirmed large electron-dense granules, each showing distinct peaks for Ca and P by microanalysis, associated with the cytoplasmic aspect of lateral plasma membranes of secretory, transitional, and smooth-ended ameloblasts. The present study thus revealed a strong calcium-binding property of the cytoplasmic aspect of lateral plasma membranes of secretory ameloblasts and its fluctuation in the maturation ameloblasts under the given experimental conditions. A necessity for an extensive analysis of membrane constituents of the enamel organ was proposed for better understanding of cellular roles in calcium regulation during amelogenesis.

Vinblastine is known to affect secretory and transport functions of ameloblasts. The effects of vinblastine on distribution patterns of membrane-associated calcium and Ca2+,Mg(2+)-ATPase in maturation ameloblasts were investigated cytochemically. The potassium pyroantimonate (PPA) method was used for localizing calcium and a modified Wachstein-Meisel medium was used to localize Ca2+,Mg(2+)-ATPase. Ultrastructural changes induced by vinblastine included dislocated organelles and reduction or elimination of the ruffled border of the ameloblasts. Membrane-associated calcium pyroantimonate deposits were markedly reduced. The intensity of Ca2+,Mg(2+)-ATPase reaction product was also markedly reduced by vinblastine. Concomitant reduction of membrane-associated calcium and Ca2+,Mg(2+)-ATPase lends support to a role for maturation ameloblasts in control of a cyclic pattern of influx of calcium to mineralizing enamel.

The enamel organs of rat incisors were separated from the enamel surface and processed for rapid freezing and freeze-substitution. A histochemical stain for calcium (GBHA) of thick Epon sections revealed intense calcium reactions in the secretory ameloblasts, exclusively in the tubulovesicular structures extending throughout their distal cytoplasm. Electron microscopy revealed a thin layer of amorphous material with clusters of electron-dense granules along the distal surface of secretory ameloblasts. In young secretory ameloblasts without typical Tomes' processes, a considerable number of mitochondria were located in the distal cytoplasm and contained numerous electron-dense granules. Similar dense granules as well as fine ribbon-like electron-dense figures, all containing significant amounts of calcium, were observed in some of the tubulovesicular structures at the distal end of these cells. A putative exocytotic figure of such dense granules was also observed. The electron-dense granules were rare in more differentiated ameloblasts with elongated Tomes' processes, which occasionally displayed ribbon-like figures in some of the tubulovesicular structures in the process region. No significant calcium peak was detected in the extracellular amorphous material, secretory granules, or along the lateral plasma membranes. These observations may imply high calcium concentrations in mitochondria and tubulovesicular structures in the distal cytoplasm of secretory ameloblasts relative to that of the cytosol and support the possible contribution of these organelles in secretory ameloblasts to cellular calcium regulation at least in the early stage of amelogenesis.

Colchicine is known to affect secretory, transport, and degradative functions of ameloblasts. The effects of colchicine on membrane-associated calcium and Ca2+,Mg2(+)-ATPase in secretory and maturation ameloblasts were investigated cytochemically. The pyroantimonate (PPA) method was used for localizing calcium and a modified Wachstein-Meisel medium was used to localize Ca2+,Mg2(+)-ATPase. Sections representing secretory and early maturation stages were examined by transmission electron microscopy. Morphological changes induced by colchicine included dislocated organelles and other well-established reactions to such anti-microtubule drugs. Calcium pyroantimonate (Ca-PA) deposits in most ameloblast types were markedly reduced, with the greater reduction occurring in those cells more severely altered morphologically. However, the cell membranes of both control and experimental smooth-ended maturation ameloblasts were essentially devoid of Ca-PA. The normal distribution and intensity of Ca2+,Mg2(+)-ATPase was not affected by colchicine. Because the observed reduction of membrane-associated calcium is apparently not mediated by Ca2+,Mg2(+)-ATPase in this case, other aspects of the calcium regulating system of ameloblasts are apparently targeted by colchicine.

The distribution and movement of calcium through the enamel organ and into the forming enamel was examined by means of quantitative 45Ca radioautography in rat incisors. Dislocation of radiocalcium in the specimen was minimized during histologic and radioautographic processing by using rapid freeze/freeze-substitution and dry emulsion coating methods. At 30 sec. after the 45Ca injection, distinct peaks of radioactivity occurred in the connective tissue immediately adjacent to the enamel organ and the infranuclear compartment of secretory ameloblasts. An intense labeling also occurred in the superficial layers of the forming enamel extending 15 microns below the surface. The grain density in the distal cytoplasm of secretory ameloblasts increased at later time periods, whereas all other regions of the enamel organ showed a considerable decrease in radioactivity. The radioactivity in the infranuclear compartment of ameloblasts with numerous mitochondria remained relatively high at 2 min. but was abolished by 10 min. after the injection. The grain density in the enamel matrix became much stronger but the labeled regions only extended to 20, 30, and 40 microns below the surface at 2, 10, and 60 min. after the injection, respectively. The application of wet emulsion over similarly prepared sections caused a severe dislocation of radiocalcium in the specimens. These data confirmed the rapid penetration of systemically administered radiocalcium into the surface layers of forming enamel and its slow diffusion to the deeper layers. The time-related changes in relative grain densities at various regions of the ameloblasts support the coexistence of a relatively slow transcellular pathway for calcium through the secretory ameloblast layer.

Ultrastructural localization of calcium in preodontoblasts and preameloblasts was investigated using the potassium pyroantimonate technique, and it was confirmed that there were clear differences in calcium distribution pattern between preodontoblasts and preameloblasts. In preodontoblasts, pyroantimonate reaction products were mainly observed in the Golgi region, lateral intercellular spaces, and secretory granules, especially in the distal portion of cell body; however, few were found in mitochondria and on the plasma membrane. In preameloblasts, on the other hand, the precipitates were located in mitochondria, nuclei, and on the inner face of the plasma membrane; however, few reaction products were observed in the intercellular spaces, lysosomelike granules, secretory granules, and stippled materials. Granular reaction product approximately 20-40 nm in diameter adhered preferentially to the growing end of needlelike crystals in the initial enamel matrix.

Energy dispersive X-ray microanalysis was applied to freeze-dried blocks of enamel organ tissue to determine levels of calcium in various cellular regions. The tissue blocks were dissected free from adjacent forming enamel following injection of cobalt or fluoride ions, both of which temporarily inhibit enamel mineralization. In all control and experimental specimens there was an increasing gradient of calcium from the stratum intermedium cells to the distal ends of the ameloblasts. Calcium levels were significantly reduced near the distal ends of the ameloblasts following cobalt or fluoride injection as compared with controls. It is suggested that evidence of an intercellular buildup of calcium near the distal ends of the ameloblast supports a controlling function of these cells. The changes in calcium levels are correlated with alterations in mineralization known to occur in the adjacent enamel of the model systems employed.

Migration of mitochondria and modulation of Ca-ATPase activity in secretory ameloblasts were investigated ultrastructurally and ultracytochemically by using lower incisors taken from normally fed, 30-hr-fasted, and calcium (Ca)-loaded rats. In normally fed rats, almost all mitochondria were localized in a narrow infranuclear compartment between the nucleus and proximal cell webs of secretory ameloblasts. In 30-hr-fasted rats, a prominent migration of many mitochondria into the supranuclear region of the cells was noted. Mitochondria returned to the infranuclear compartment and seldom appeared in the supranuclear region when fasted rats were Ca-loaded by transcardiac perfusion with physiological Ca solution. Normally, the mitochondria of secretory ameloblast exhibited moderate Ca-ATPase activity along their inner membranes. This mitochondrial Ca-ATPase was decreased by a 30-hr fast and became prominent again after Ca loading. Plasma-membrane Ca-ATPase was demonstrated in the entire cell surface of secretory ameloblasts. An especially abundant reaction was found along the invaginated cell surface of the Tomes process. This Ca-ATPase also became very weak and was almost abolished from the Tomes process after fasting, but Ca loading caused reappearance of an intense Ca-ATPase activity on the entire cell surface, including along Tomes's processes. These results suggest that 1) mitochondrial localization in secretory ameloblasts is influenced by the Ca concentration of the extracellular milieu, and 2) the level of mitochondrial and cell-membrane ATPase activity is responsive to the concentration of extracellular calcium.

X-ray micro-analysis and electron energy loss analysis were used to confirm cytochemical localization by potassium pyro-antimonate of calcium in secretory ameloblasts. Neither intercellular calcium concentration between ameloblasts nor over-all calcium levels of the enamel organ were lowered during a period of inhibition of enamel mineralization by injected fluoride. Calcium concentrations in mitochondria and secretory granules were reduced.

Rapidly frozen growing rat incisors were freeze fractured and freeze dried in preparation for energy dispersive X-ray emission microanalysis in a scanning electron microscope. Ca levels were found to be elevated in the distal cell body of odontoblasts, whereas Ca was uniformly low over all parts of the cell body of secretory ameloblasts. The results suggest fundamental differences in the mechanisms by which these two cell types process Ca, and that Ca possibly diffuses through the secretory ameloblast layer on its way to the enamel.

Scanning electron microscopy was used to distinguish the topographical characteristics of two maturation ameloblast types in freeze-dried blocks of enamel organ tissue. This distinction was based primarily upon the configuration of the distal ends of the ameloblasts and the presence or absence of wide intercellular spaces. Energy dispersive x-ray spectrometry was applied to compare calcium levels in various regions of tissue identified as constituting either ruffle-ended or smooth ended ameloblasts. Greater levels of calcium were found in the distal ends of the ruffle-ended cells than in their proximal ends. In addition, greater calcium levels were found in the distal ends of the ruffle-ended cells than the distal ends of the smooth-ended cells. The higher calcium levels in ruffle-ended cells correlates with the view that these cells are actively involved in control of movement of calcium to the enamel front.

Fluoride in high concentrations is known to have an adverse effect on the formation of enamel. The effect of a single injection of two concentrations of sodium fluoride on inner enamel secretory ameloblasts was investigated morphologically by electron microscopy and functionally by assessing the location and relative amount of available calcium, using the potassium pyroantimonate method. The results showed that acute doses of fluoride interfere with the normal function of secretory ameloblasts. The increase in the population of lysosome-like structures observed after fluoride administration is suggestive of defects in the synthetic pathway. Concomitant with the effect of fluoride on secretory ameloblasts is an inhibition of enamel formation, resulting in incomplete enamel rods and leaving large remnants of Tomes' processes buried in the enamel. The distribution of the calcium pyroantimonate deposits found tends to support the concept of calcium traveling between the cells to the enamel. Acute doses of fluoride also reduce the amount of calcium available for complexing with pyroantimonate in the intercellular region.

The influence of vanadate, a potent inhibitor of Ca2+-ATPase and Na+-K+-ATPase, on 45Ca uptake in maturing enamel of the rat incisor was investigated by a vascular perfusion method combined with 45Ca autoradiography. The morphological integrity of the maturation-stage enamel organ was well-retained during vascular perfusion under all the experimental conditions. Distinct patterns of 45Ca labeling, comparable with those found in previous in vivo 45Ca autoradiographic studies, appeared in the maturing enamel after vascular perfusion with a standard perfusate. One mmol/L vanadate added to the standard perfusate caused a drastic decrease in 45Ca uptake in the maturing enamel, corresponding to the ruffle-ended ameloblasts, leaving narrow peaks of moderate intensity corresponding to the bands of the overlying smooth-ended ameloblasts. The in vitro labeling of exposed enamel surfaces with 45Ca revealed blackening of autoradiographic emulsion in wide bands separated by unlabeled or slightly labeled narrow ones resembling the distribution of smooth-ended ameloblasts in both control and vanadate-treated incisors. Our observations indicate that the ruffle-ended ameloblasts of the rat incisor serve as an efficient diffusion barrier to calcium ions and regulate transcellular calcium transport to the maturing enamel, at least in part, by a vanadate-sensitive mechanism.

In order to detect the precise location and pathways of calcium in the ameloblastic layer with EM and EDX, frozen ultrathin-sections, potassium pyroantimonate (PPA) method and 45Ca-autoradiography (A.R) were employed. A population of dense granules containing Ca and P was mainly observed within the nuclei and mitochondria of the frozen-sectioned ameloblasts, whereas PPA-reaction was seen not only within them but in ER, Golgi vesicles and intercellular spaces. No granules were found at the enamel matrix by both methods, but peaks for Ca and P were distinct. Silver grains of 45Ca-A.R were located in nuclei, mitochondria. Golgi complex and intercellular spaces. These results indicate that calcium can transport across the ameloblastic layer through two main pathways, extracellular and transcellular.

We investigated the ultrastructural distribution of calcium in several kinds of hard tissue forming cells (secretory and maturation ameloblasts, odontoblasts osteoblasts, chondrocytes, and osteodentine forming cells) of mammals, amphibians, and fish by use of the potassium pyroantimonate technique. The calcium distribution pattern is compared among these cells, and its biological significance is discussed. Except for mammalian odontoblasts, all types of the hard tissue forming cells exhibited fundamentally the same distribution pattern of calcium; the antimonate reaction product was mainly localized on the inner face of the plasmalemma and inside mitochondria. On the other hand, in mammalian odontoblasts, the reaction product was found within secretory granules and in the intercellular spaces. Thus, the calcium distribution pattern in odontoblasts of lower vertebrates differed from that of mammalian odontoblasts and was similar to that of the osteoblasts or chondrocytes of the vertebrates examined. The differences in calcium distribution pattern among these hard tissue forming cells were not related to their origin, ectodermal or mesodermal (ectomesenchymal). We suggest on the basis of previous studies cited in this paper and of the present data that they are closely associated with the phylogeny and physiological system of Ca-ATPase.

The distribution of calcium in relation to secretory ameloblasts of the rat incisor was studied. An experimental model system in which enamel mineralization was temporarily inhibited by injecting sodium fluoride and cobalt chloride was used. Potassium pyroantimonate (PPA) cytochemistry, electron energy loss spectroscopy (EELS), and energy dispersive X-ray spectrometry (EDS) were used to clarify the role of the ameloblast in controlling calcium distribution during normal and experimentally altered enamel mineralization. Secretory ameloblasts chemically-preserved in glutaraldehyde either with or without PPA were analyzed for calcium; those preserved with PPA showed higher concentrations of calcium than did those preserved with glutaraldehyde only. Freeze-dried control and experimental tissues showed an increasing gradient of calcium from stratum intermedium cells to the distal ends of the ameloblasts. Calcium levels were reduced near the distal ends of the cells following fluoride and cobalt injections, while magnesium levels were increased markedly in the same region. This multi-method approach showed correlated calcium localization in specific regions of this cell in relation to changes in function. The study thus provides additional evidence for active involvement of the ameloblasts in enamel mineralization.

45Ca transfer through the stratum intermedium and the secreting ameloblasts towards enamel has been studied by quantitative electron microscopical autoradiography in tooth-germs of newborn cats following intravenous injection of the isotope. Two transfer pathways were demonstrated. The relatively more important direct path passed through the stratum intermedium and ameloblast intercellular spaces and reached the enamel directly. The second pathway consisted in an intracellular transfer through the ameloblasts. 45Ca penetrated the cell through its basal pole. The mitochondria were the most highly labeled organelles at the different experimental time intervals studied. A maximum of radioactivity was respectively noticed at 30 min and 1 h in the endoplasmic reticulum and the Golgi apparatus. A total absence of silver grains was noted over the secretory ameloblastic bodies. At 6 h, the highest labeling was observed over enamel and especailly over the inner enamel along the enameldentin junction.
