The present detailed study aimed to establish for the first time the ovarian and uterine hemodynamic change in dairy cows after the end of the puerperal period to investigate a possible association between Doppler indices and volume of blood flow. Twenty cows weighing 500-600 kg (mean 400 ± 50 kg) and 4-5 years of age (mean 3.2 ± 0.5 years) categorized into two main groups (true anestrum, with corpus luteum, n = 10) and (normal cyclic, n = 10), The period of examination started from day 39 till day 70 after calving with day after day routinely examination by b-mode and Doppler performed on both ovaries with ovarian arteries (OA) and uterus with uterine arteries (MUA) in the ipsilateral (ipsi) and contralateral (contra) side to ovulation, in addition to a thickness in horns and body was measured. Estradiol and progesterone were also measured. Results showed that both Doppler indices in the OA and MUA ipsi and contra had a positive (P ≤ 0.001) correlation with contra Doppler indices, but revealed a negative (P ≤ 0.05) correlation with ipsi and contra Doppler velocities, blood flow rate and volume in anestrum cows. Both ovarian and uterine ipsi indices showed an increase (P ≤ 0.05) in the anestrum cows, and both PSV and EDV of both arteries ipsilateral showed (P ≤ 0.05) a decrease in the anestrum cows, the ipsi and contra ovarian and uterine colored % were lower in anestrum group than the normal group. Estradiol (P ≤ 0.05) decreased in anestrum cows than the normal, while progesterone increased in the anestrum group. The objective of this study was to assess the uterine blood flow following estradiol benzoate administration in Holstein-Friesian dairy cows by trans-rectal color Doppler ultrasonography. Six healthy lactating Holstein-Friesian cows were examined daily for 10 days starting at 4 weeks postpartum. All the cows, which were clinically healthy based on vaginal mucus scoring and endometrial cytology, were examined by trans-rectal Doppler ultrasonography to measure pulsatility index (PI), resistance index (RI), time average maximum velocity (TAMAX), blood flow volume (BFV) and diameter in the uterine arteries ipsilateral and contralateral to the previously pregnant uterine horn. On the third day of the experiment, the six cows were administered 10 mg intramuscular injection of estradiol-17β (E 2 ).Blood samples were collected at the time of daily examination for the assessment ofE 2 concentrations.The PI and RI values decreased while TAMAX, BFV and diameter of uterine arteries increased in response toE 2 administration (P < 0.05).There was a high correlation between both the ipsilateral and contralateral uterine arteries for all variables that were studied(r = 0.860, P < 0.0001, r = 0.922, P < 0.0001, r = 0.651, P < 0.0001, r = 0.879, P < 0.0001, r = 0.861, P < 0.0001 for the PI, RI, TAMAX, BFV and uterine arteries diameter, respectively).In conclusion, the greater blood concentrations ofE 2 may be responsible for the greater TAMAX, BFV, increased diameters and decreased PI and RI of the uterine arteries during the puerperium in dairy cows., (Copyright © 2018 Elsevier B.V. Previous studies have characterized ovarian steroid synthesis which directly affects uterine environment and blood flow. Clearance of steroids occurs primarily in hepatic tissues, however, it was discovered that there is an abundant activity of the phase II steroid metabolizing enzyme UDP-glucuronosyltransferase (UGT) in uterine biopsies. No minimally invasive techniques for collecting endometrial perfusion, which is affected by steroids and indicative of reproductive health, have been developed for livestock. The objective of the present study was to characterize UGT activity and endometrial blood perfusion during a normal estrous cycle of cattle. It was hypothesized that there would be increased steroid metabolism during the luteal phase of the estrous cycle and in the uterine horn ipsilateral to the corpus luteum (CL). During the first synchronized estrous cycle, progesterone and UGT activity increased on Day 6 compared with 0 and 3, with the first day of estrus being considered Day 0 of the study. Endometrial perfusion was greater ipsilateral to the CL compared with contralateral on Day 12, and was less ipsilateral to the CL compared with contralateral on Day 18. Similar to perfusion results, nitric oxide metabolites (nitrites) were greatest in the endometrium ipsilateral as compared with contralateral to the CL. Moreover, there was a positive correlation (r = 0.28; P = .04) between endometrial perfusion and nitrite concentration. It is concluded that activity of UGT within the endometrium is affected by the contralateral or ipsilateral location of the CL, and collection of endometrial perfusion data using a laser Doppler probe could be a viable measurement technique as indicated by associated nitrite concentrations in the present study., (Copyright © 2018 Elsevier B.V. The objective of this study was to determine the effect of rumen-protected arginine on median caudal artery blood flow and LH dynamics in cows fed toxic endophyte-infected tall fescue seed. Four ruminally cannulated nonlactating beef cows (539 ± 30 kg) were used in a 2 × 2 factorial arrangement of treatments utilizing a 4 × 4 Latin square design with 4 periods of 31 d each. Each cow was assigned to individual pens and fed orchardgrass hay (10.3% CP and 85% NDF; OM basis) during a 10-d adaptation period, followed by a 21-d collection period in which each cow was assigned 1 of 4 treatments: 1) rumen-protected Arg (180 mg/kg of BW) and 1.0 kg/d of toxic endophyte-infected fescue seed (AE+), 2) rumen-protected Arg (180 mg/kg of BW) and 1.0 kg/d of endophyte-free fescue seed (AE-), 3) 1.0 kg/d of toxic endophyte-infected fescue seed (E+) alone, or 4) 1.0 kg/d of endophyte-free fescue seed (E-) alone. In each period, Doppler ultrasound measurements for blood flow parameters were quantified on d 1, 5, 10, 15, and 20. On d 20 of each period, blood samples were collected every 10 min for 6 h and then once every hour for 12 h for LH response following exogenous GnRH. There was an Arg × fescue seed type interaction ( = 0.05) for median caudal artery blood flow due to an increase in blood flow in cows fed rumen-protected Arg with endophyte-free fescue seed. In addition, mean blood flow velocity in the artery was greater ( = 0.01) with the inclusion of rumen-protected Arg in the diet. Median caudal artery area ( = 0.03) and diameter ( = 0.01) were decreased in cows consuming E+ compared to those consuming E- with no effect ( ≥ 0.38) by Arg inclusion. Circulating nitric oxide (NO) concentrations tended to be influenced ( = 0.09) by the interaction of Arg × fescue seed type with E+ alone decreasing NO concentrations. Circulating NO concentrations were unaffected by rumen-protected Arg ( = 0.48). Mean serum LH concentration exhibited ( = 0.02) an Arg × fescue seed type interaction. Cows consuming E+ had decreased ( < 0.05) LH concentrations compared to all other treatments. However, cows consuming AE+ had ( ≥ 0.67) LH concentrations similar to those of cows consuming AE- and E-. Thus, supplementing rumen-protected Arg to cows consuming toxic endophyte-infected fescue seed has the potential to increase reproductive performance and peripheral blood flow.

The aim of this study was to investigate the compatibility of the visual evaluation result of the blood flow characteristics and the blood flow measurements of the CL and the predictability of the responses given by corpora lutea with varying levels of blood flow to an induction of luteolysis by a PGF 2α injection and to determine the possibility of increase in serum estrogen and progesterone concentrations in parallel with increased luteal blood flow (LBF). The cows, bearing a CL (n = 60; postpartum 35 days), were injected with PGF 2α and were monitored for signs of estrous following the first injection. The cows, which did not show estrous signs, were examined for the presence of a CL on Day 14, whereas those that showed signs of estrous were examined on Day 10 following the onset of estrous. The level of LBF was visually graded as + (low; GI), ++ (medium; GII), +++ (high; GIII), and ++++ (very high; GIV). Immediately after the examination of LBFs, a second intramuscular injection of PGF 2α was injected. In the cows, which were determined to be in estrous, the diameter of the Graafian follicles was measured by B-mode ultrasonography. Subsequently, these animals were artificially inseminated. The animals, which did not show estrous after the second injection, were examined as previously described and monitored for signs of estrous. A strong correlation (r = 0.654; P < 0.001) was determined to exist between the results of the visual examination of the images and the results obtained for the LBF area with the use of the Pixel Flux software. GIII (0.83 ± 0.15 cm 2 ) and GIV (1.03 ± 0.48 cm 2 ) were found to differ from GI (0.47 ± 0.23 cm 2 ) and GII (0.51 ± 0.12 cm 2 ) for the size of the LBF (P < 0.001). Serum progesterone levels in groups (GI, GII, GIII, and GIV) were determined to be 4.44 ± 2.42 ng/mL, 6.03 ± 2.37 ng/mL, 7.01 ± 2.94 ng/mL, and 7.17 ± 1.69 ng/mL, respectively. The comparative evaluation of the study groups showed that the groups did not statistically differ for the period between PGF 2α injection and the onset of estrous, mean Graafian follicle size and estrogen levels. The massive increase in size of the fetus and uterus in the last trimester is accompanied by an increasing demand for nutrients and oxygen, and it is assumed that this demand is met by increasing uterine and fetal perfusion. The goals of this study were to measure the perfusion of the uterine arteries and the placentomes in the last month of gestation and to investigate the effect of epidural anesthesia and isoxsuprine on perfusion. During the last month of gestation, eight Braunvieh cows underwent nine color Doppler sonographic examinations of the uterine arteries to determine diameter (DM), pulse rate (PR), resistance index, time-averaged maximum blood flow velocity (TAMV), and blood flow volume (BFV), and power-mode Doppler sonography was used to determine perfusion of placentomes. The PR increased (P < 0.001), and the BFV and TAMV of the ipsilateral uterine artery decreased between 4.5 and 0.5 weeks prepartum (BFV, 236.8 ± 65.80 and 208 ± 41.52 cm(3)/s, P < 0.01; TAMV, 140.0 ± 26.53 cm/s and 125.2 ± 18.46 cm/s, P < 0.05). After sonographic examination, the cows received epidural administration of local anesthetic (100-mg lidocaine) in the sacrococcygeal space or isoxsuprine (200 mg/cow, iv), and the sonographic measurements were repeated 30 minutes later. After epidural anesthesia, the TAMV and BFV of the contralateral uterine artery increased by 5.4% (P < 0.05) and 7.9% (P < 0.01). In the placentomes of the gravid uterine horn, the relative placentome perfusion and the color pixel grading (Cp) increased by 10.1% (P < 0.05) and 11.5% (P < 0.01) after epidural anesthesia. After isoxsuprine, the DM, PR, and BFV increased by 4.7%, 49.3%, and 16.9% in the ipsilateral uterine artery and by 10.8%, 48.7%, and 22.8%, respectively in the contralateral uterine artery. The TAMV of the ipsilateral uterine artery increased by 7.1% (P < 0.01), and the resistance index decreased in both uterine arteries (ipsilateral 24.2%, contralateral 14.9%, both P < 0.00001). Isoxsuprine increased the relative placentome perfusion and the Cp of the placentomes by 18.1% and 18.3% in the gravid horn and by 10.2% and 24.2% in the nongravid horn. Blood flow variables changed little in the last month of gestation. Transrectal color Doppler ultrasonography is a useful technique to get new information about physiologic and pathophysiologic alterations of the uterus and ovaries in female cattle. During all reproductive stages characteristic changes in uterine blood flow are observed. Cows with puerperal disturbances show delayed decrease in uterine blood flow in the first few weeks postparturition compared with healthy cows. Measurement of follicular blood flow is used to identify normally developing follicles and predict superovulatory response. Determination of luteal blood is more reliable than B-mode sonography to distinguish between functional and nonfunctional corpora lutea. The maternal portion of the bovine placenta receives blood mainly from the uterine arteries (AUT) and the fetal portion from the umbilical arteries (AUM). Placental perfusion is crucial for fetal development and undergoes adaptive changes during pregnancy according to the fetal requirements. One goal of this study was to investigate changes in Doppler sonographic measurements of blood vessels that supply blood to the placenta in cows during the last 4 weeks of pregnancy. Another goal was to examine how these measurements are affected by three drugs commonly used in cows at the time of parturition. Nine cows underwent Doppler sonographic examination of the AUT ipsilateral and contralateral to the pregnant horn and one AUM three times per week during the last 4 weeks of gestation. This was followed by the randomized administration of one of the three following experimental drugs per day: isoxsuprine (200 mg/cow, iv), xylazine (2 mg/100 kg, iv), and lidocaine for epidural anesthesia (100 mg/cow). Doppler sonographic examination was repeated 30 minutes after medication. Maternal pulse rate increased during the study period (P < 0.001), and the diameter of the contralateral AUT was smaller in the last week before birth than in the two preceding weeks. The resistance index (RI) of the ipsilateral AUT was smaller in the last week than in the first 2 weeks of the study period. Uterine blood flow volume increased after isoxsuprine by 5% and after epidural anesthesia by 6% (both P ≤ 0.05) and decreased after xylazine by 10% (P < 0.001). Isoxsuprine was the only drug that elevated the blood flow volume in the AUM (P ≤ 0.05). Xylazine increased the RI of both AUT (both P < 0.001) and significantly reduced maternal and fetal pulse frequencies, whereas isoxsuprine significantly reduced the RI of both AUT and the AUM and increased maternal and fetal pulse frequencies. The results reported that Doppler sonographic measurements of uterine and AUM change little in the last month of pregnancy in the cow. Eight cows underwent Doppler sonographic examinations of the milk vein and musculophrenic vein from 9 days prepartum to 300 days postpartum. A 7.5-MHz linear transducer was used to determine the inner diameter of the veins and blood flow velocities and volumes on the left side in standing, non-sedated animals. Cows were weighed and milk yield measured at all examination times. The milk vein appeared as a vessel with an inner diameter of 2.01-2.30 cm immediately beneath the skin. The maximum blood flow velocity ranged from 23.84 to 35.76 cm/s before parturition, increased markedly on the day of calving (day 0), peaked at 61.14 cm/s on day 1 and slowly decreased to 23.84 cm/s by day 300. The profiles of the minimum and mean maximum flow velocities and flow volume were similar. The musculophrenic vein had an inner diameter of 0.76-1.07 cm and its distance from the surface of the skin was 1.15-1.31 cm. The profiles of the blood flow variables were irregular and did not resemble those of the milk vein. The maximum blood flow velocity ranged from 87.35 to 114.41 cm/s, the minimum velocity from 5.47 to 7.60 cm/s, the mean maximum velocity from 48.55 to 78.74 cm/s and the blood flow volume from 1.02 to 2.44 l/min. The milk vein had a larger diameter (2.16 versus 0.90 cm; P<0.01), smaller maximum, minimum and mean maximum blood flow velocities (P<0.01) and a larger blood flow volume than the musculophrenic vein (P<0.01)., (Copyright © 2012 Elsevier Ltd. Background: This study investigated the effect of milk yield on blood flow variables in the milk vein and musculophrenic vein in dairy cows., Methods: Five healthy dry cows, five cows with a daily milk yield of 10 kg and five others with a daily milk yield of 20 kg underwent B-mode and colour Doppler sonographic examination. The diameter of the veins, blood flow velocities and blood flow volumes were measured on both sides in standing, non-sedated cows using a 7.5 MHz linear transducer., Results: Lactating cows had significantly higher blood flow velocities in the milk vein than dry cows; the maximum blood flow velocity of dry cows and those with a daily milk yield of 10 and 20 kg were 14.04, 38.77 and 39.49 cm/s, respectively, the minimum velocities were 0.63, 3.02 and 2.64 cm/s, respectively, and the mean maximum velocities were 8.21, 26.67 und 28.22 cm/s, respectively. Cows producing 20 kg of milk a day had a blood flow volume of 3.09 l/min, which was significantly higher than 0.79 l/min recorded in dry cows. Lactating cows had significantly higher mean maximum blood flow velocities in the musculophrenic vein than dry cows. Blood flow variables of both veins did not differ significantly between the left and right side., Conclusion: This study showed that milk yield has a profound effect on blood flow variables in the milk vein and to a lesser extent the musculophrenic vein. This must be taken into consideration in future Doppler sonographic studies of these veins and possibly other vessels. Furthermore, measurements on one side are representative of both sides.

The goal of this study was to determine the effects of the length of the preovulatory phase on genital blood flow and mRNA expression of endometrial hormone receptors in cattle (Bos Taurus). Ovulation was synchronized in 50 Holstein-Friesian cows using a modified Ovsynch (ovulation synchronization) protocol, in which the second gonadotropin-releasing hormone (GnRH) administration was given 40h (G40, n=17) or 60h (G60, n=16) after the prostaglandin F(2alpha) (PGF(2alpha)) administration. The third group (S, n=17) did not receive a second GnRH administration. Transrectal color Doppler examinations were carried out 24h before (Day -1) and on Day 7 after ovulation (Day 0). Follicular size (FS) and luteal size (CLS) were quantified by measuring the areas of these structures on cross-sectional B-mode ultrasound images. Follicular blood flow (FB) and luteal blood flow (CLB) were quantified by determining the colored areas of these structures. Uterine blood flow was measured using the time-averaged maximum velocities (TAMVs) and the pulsatility indices (PIs) of both uterine arteries. Endometrial mRNA transcript abundance of estrogen receptors alpha and beta as well as oxytocin and progesterone receptor were determined on Days -1 and 7 in G40 and G60 cows. In all cows, plasma progesterone (P(4)) values were measured on Day 7. On Day -1, FS and FB values were lower (P < or = 0.05) in G40 cows compared with those in S cows but did not differ (P>0.05) between G60 cows on the one hand and G40 cows and S cows, respectively, on the other hand. On Day 7, CLS and P(4) did not differ (P>0.05) between cows of Groups G40, G60, and S; CLB was lower (P< or =0.05) in G40 cows than in G60 and S cows, but no difference (P>0.05) occurred between G60 and S cows. The uterine TAMV and PI values did not differ among the three groups (P>0.05). Gene expression of hormone receptors did not differ (P>0.05) between Groups G40 and G60 on Days -1 and 7. Results of this study indicate that a shortened preovulatory follicle phase primarily affects ovarian but not the measured uterine events in cows undergoing synchronization of ovulation.

The goal of the present study was to examine the musculophrenic vein of 29 healthy Swiss Braunvieh cows using colour-Doppler sonography to determine vessel morphology and diameter, and blood flow velocity. The hair over the reticular region was clipped, and the left musculophrenic vein was examined before and 10min after sedation using 0.03mg/kg xylazine. The musculophrenic vein appeared as a vessel with a diameter of 0.5-1.1cm located in the diaphragmatic musculature. The spectral display was a broad band structure with a wave-like shape. The Doppler measurement point was 1.5-2.7cm from the body surface. The diameter of the vein and the blood flow velocity did not differ significantly before and after sedation. Before sedation, the mean diameter (+/-SD) of the musculophrenic vein was 0.7 (0.2) cm, the maximum blood flow velocity 90.2 (38.6) cm/s, the mean blood flow velocity 60.4 (22.3) cm/s and the minimum blood flow velocity 41.4 (24.2) cm/s. There were significant correlations (r=0.45-0.90) between blood flow velocity before and after sedation and between minimum, maximum and mean blood flow velocities.

Two experiments were conducted to compare variation between 2 tracing methods in measuring cross-sectional lumen area of the caudal artery in 5 beef heifers on 3 different dates (Exp. 1) and to compare tracing methods in detecting changes in artery lumen area after 5 heifers were switched from a diet containing nonendophyte-infected tall fescue [Schedonorus arundinaceus (Schreb.) Dumort] seed to one containing endophyte-infected tall fescue seed (Exp. 2). Lumen area determined by tracing the Doppler flow signal was 25% less than that determined by tracing the intima of the connective tissue, but there was no difference (P = 0.90) in the variation of measures between the 2 methods in Exp. 1. Declines in lumen area were detected at the same level of significance (P < 0.01) for both tracing methods after cattle in Exp. 2 were switched from noninfected to infected tall fescue diets. Variation in lumen areas was different between noninfected and infected diets with tracing the Doppler flow signal (P < 0.05) or the intima of the connective tissue (P < 0.01). Results indicated that lumen area of the caudal artery can be measured with similar precision by tracing the intima of the connective tissue in the artery wall or the outer boundary of the Doppler flow signal.

Transrectal color Doppler sonography was used to compare changes in uterine blood flow between cyclic and early pregnant lactating dairy cows. Examinations were carried out in 53 multiparous lactating cows on days 3, 6, 9, 11, 13, 15, 18 and 21 (day 0=estrus). Fourteen cows were examined during the estrous cycle and thirty-nine cows after insemination with frozen/thawed sperm. Uterine blood flow was reflected by the time-averaged maximum velocity (TAMV) and the pulsatility index (PI) in the uterine artery ipsilateral to the corpus luteum. Twenty-one cows that were not pregnant on day 25 were excluded from the study. There was high inter-individual variability in PI- (CV: 22-62%) and TAMV-values (CV: 22-42%) at all examinations. In cyclic cows, TAMV values increased between days 13 and 18 and PI values decreased between days 15 and 21 (P<0.05). In pregnant cows (n=18), TAMV values increased from days 9 to 11 and then decreased to minimal values by day 18 (P<0.05). The PI values decreased between days 3 and 11 and then increased to maximum levels on day 18 (P<0.05). On day 21, both variables reached (P<0.05) values that did not differ (P>0.05) from those on day 11. The changes in TAMV were correlated with estrogen and progesterone concentrations (r=0.69 and -0.70, respectively; P<0.05) in cycling cows, but not in pregnant cows (P>0.05). The PI values did not correlate with steroid hormone levels (P>0.05). Differences in uterine blood flow between cycling and early pregnant cows were observed only on day 18 (P<0.05). The results show that in pregnant cows changes in uterine blood supply can be detected already in the second week after insemination; these changes do not occur in the second week of the cycle.

Objective: The objective of the study was to report normal ultrasonographic appearance and intra-cardiac dimensions in two dairy breeds and to calculate cardiac output (CO) using echocardiography., Background: Intra-cardiac dimensions, time indices and CO estimation have not previously been reported in adult cattle., Animals, Materials and Methods: Echocardiograms were obtained from healthy adult dairy cows (10 Jersey (J) and 12 Holstein Friesians (HF)) in the body weight range of 400 to 700 kg. Standard echocardiographic images were obtained from the left and right hemithoraces. Velocity time integrals were obtained in order to calculate CO using pulsed wave Doppler of aortic flow in the J cows. Measurements obtained included pulmonary artery and aortic diameters, left and right ventricular diameters (and calculated fractional shortening and left ventricular ejection fraction), left atrial size and time indices assessing valve function., Results: HF cows had significantly (p<0.05) larger pulmonary artery and aortic diameters, larger left atrial diameters and left ventricular internal diameters during diastole, but these were not different when corrected for body weight. Left and right ventricular dimensions, adjusted for body weight, were significantly larger (p=0.02 and p=0.035 respectively) in J cows when compared to HF cows. No differences were noted in the time indices between the two groups. No significant differences were noted in intra-operator variability and the only significant difference in inter-operator variability was in measurement of the pulmonary artery (p=0.03; ICC=0.63)., Conclusions: It is possible to obtain repeatable, reliable echocardiograms in order that meaningful intra-cardiac dimensions can be obtained in adult dairy cattle.

The development of Doppler processing extended the scope of sonographic imaging from an anatomical to a physiological basis. This technique became established as a clinical tool in human gynaecology. For example, it has been discussed that the implantation of an embryo is influenced by the uterine blood flow. In cows, this uterine blood flow was investigated, using surgically implanted Doppler ultrasonic or electromagnetic blood flow probes prior to the introduction of colour Doppler sonography in bovine medicine. Therefore, the aims of our studies were to use transrectal Doppler sonography for the non-invasive measurement of uterine and ovarian blood flow in cows and to determine changes in genital perfusion during the oestrous cycle, pregnancy and puerperium, respectively. The results of our studies show that transrectal flow imaging can be used to obtain blood flow velocity waveforms from the uterine arteries at any time during the oestrous cycle, pregnancy and puerperium. During all these phases, characteristic changes in the uterine blood flow could be observed. This uterine blood flow was low during diestrus and high during proestrus and oestrus. During pregnancy, an exponential rise in uterine blood supply could be detected. There was a positive relationship between the uterine blood flow volume (BFV) at the end of gestation and the birth weight of calves. During puerperium, the uterine BFV declined tremendously, especially during the first week after birth. In cows, with pathological disturbances of the pueperium a delayed decrease in the uterine BFV was observed. Characteristic alterations occurred also in the luteal blood flow during the oestrous cycle, which were highly related to those of the progesterone levels. Furthermore, it has been detected by the colour Doppler technique that there is no decrease, but an increase of the luteal blood flow at the beginning of luteolysis in cows. Another group has found that there are close relationships between the LH-surge and the follicular blood flow before ovulation. In conclusion, these studies show that transrectal colour Doppler sonography is a useful technique for the investigation of the genital blood flow and provides new information about physiological changes of the genital organs during, all reproductive phases. The influence of the genital blood flow on fertility in cows needs to be examined further in future studies.

Dairy calf weaning is associated with ketone concentrations that exceed the levels occurring in adults, and weaning represents a potential energy loss that may be mitigated by ionophore supplementation. To assess the effects of weaning and ionophore supplementation on net nutrient flux across portal-drained viscera (PDV) tissues in dairy calves, concentrations of glucose, acetoacetate (ACAC), beta-hydroxybutyrate (BHBA), nonesterified fatty acids, volatile fatty acids, lactate, pyruvate, insulin, and glucagon and PDV flux rates were determined in Jersey bull calves (n = 19) at 35, 56, 84, and 112 d of age. Calves were randomly assigned at birth to either a commercial pelleted starter without (CON) or with lasalocid (TRT; 83 mg/kg of dry matter). Calves were fed only milk replacer from d 3 to 34 (d 3 to 20 = 454 g/d; d 21 to 34 = 568 g/d). After blood sampling on d 35, calves received replacer (d 35 to 41 = 454 g/d; d 42 to 48 = 227 g/d) and had free access to the CON or TRT starter, and from d 49 to 112 they received CON or TRT ad libitum. Catheters were implanted in the portal vein and in the mesenteric vein and artery between d 21 and 28. Blood flow was measured by continuous infusion of p-aminohippurate into the mesenteric vein. Six serial samples were taken at 30-min intervals from the arterial and portal vein catheters simultaneously. Portal blood flow increased with age but did not differ between CON and TRT calves. Glucose was released preweaning and was extracted postweaning by PDV, but was not affected by ionophore. The portal flux of nonesterified fatty acids was not different from zero during any of the 4 sample ages. Fluxes of ACAC and BHBA in CON and TRT calves went from no measurable flux preweaning to a postweaning PDV release that peaked at d 84, but the d-84 release of ACAC and BHBA was lower in TRT calves. The portal flux of volatile fatty acids increased with age, and PDV release of both butyrate and propionate was lower at d 84 in TRT than in CON calves. However, TRT calves had a greater PDV release of lactate on d 84, partially compensating for the lower release of propionate. Glucagon was greater in CON than in TRT calves at d 84 and could be a response to the elevated ketogenesis observed in CON calves during this period. Changes in the metabolic profile and nutrient flux of transition calves were demonstrated in response to weaning and ionophore supplementation. Inclusion of an ionophore appeared to moderate alimentary output at a postweaning period (d 84) at which ketone concentrations have the potential to exceed the whole animal capacity for utilization.

Objective: To determine blood flow patterns in the common carotid artery and external jugular vein in cows before and after sedation achieved by administration of xylazine hydrochloride., Animals: 30 clinically normal Swiss Braunvieh cows., Procedure: A 5.0-MHz sector transducer was used to examine the common carotid artery and external jugular vein before and after cows were sedated by administration of xylazine. Several variables were calculated, including diameter of the blood vessels, blood flow velocity, and flow-time volume., Results: The common carotid artery before sedation had a maximum systolic velocity of 89 +/- 8.5 cm/s, maximum diastolic velocity of 36 +/- 6.0 cm/s, mean velocity of 35 +/- 5.4 cm/s, and flow-time volume of 28.2 +/- 3.48 cm3/s. In all cows, sedation achieved by administration of xylazine resulted in a significant decrease in velocity of arterial blood flow and flow-time volume. The external jugular vein before sedation had a maximum velocity of 65 +/- 8.3 cm/s, maximum velocity of retrograde venous blood flow of 20 +/- 6.6 cm/s, and flow-time volume of 29.7 +/- 5.42 cm3/s. These values decreased significantly after cows were sedated by administration of xylazine., Conclusions and Clinical Relevance: Doppler ultrasonography is particularly suitable for evaluation of blood flow patterns in the common carotid artery and external jugular vein of healthy cows. The results reported here provide a basis for use in examination of cows with cardiac and blood vessel disease.

Transrectal Doppler ultrasound was used for the noninvasive investigation of uterine blood flow in cows. Both the left and right Aa. uterinae were scanned to obtain blood flow velocity waveforms over 2 consecutive estrous cycles. Blood flow was reflected by the resistance index (RI) and the time-averaged maximum velocity (TAMV). Intra-observer reproducibility of Doppler measurements was evaluated. The intra-class correlation coefficient (Intra-CC) was 0.97 for the RI and 0.95 for TAMV. While RI values did not differ between the left and right A. uterina (P > 0.05), differences in TAMV occurred between both vessels in 2 cows. These differences were not related to the ovary bearing the dominant follicle or to the corpus luteum (P < 0.001). As in all cows, changes of RI and TAMV values between the left and right artery during the estrous cycle were correlated (correlation coefficient r > 0.72; P < 0.0001); the mean values of both sides were used for subsequent analyses. Variance component estimates for the effect of cow on RI and TAMV were 8 and 13% and for the influence of day of estrous cycle they were 70 and 47%, respectively (P <0.0001). Between estrous cycles no significant differences could be measured within cows (P > 0.05). The highest RI and lowest TAMV values occurred on Day 0 (= day of ovulation) and Day 1, while the lowest RI and highest TAMV values were measured between Days -3 and -1 of the estrous cycle, respectively. There was a positive correlation between TAMV and estrogen concentrations and a negative correlation between RI and plasma estrogen levels. Plasma progesterone levels and TAMV were negatively correlated, but no correlation could be measured (P > 0.05) between RI values and plasma progesterone concentrations. While there were no differences in plasma concentrations of estrogens and progesterone between estrous cycles within cows, the levels of these hormones differed between cows. The results show that transrectal Doppler sonography is a useful, noninvasive method for examining uterine blood flows in cows. If there is an influence of uterine perfusion on fertility in cows its role needs further investigation.

Blood flow across the lactating bovine mammary gland was measured by two techniques. The use of transit time flow probes appeared to give flows which correlated well with dye dilution in only one of five cows, although the relative changes in flow were similar between the techniques in four of the cows. Further studies were made on the effect of posture on mammary blood flow using both techniques. The crossover of venous blood from one side of the mammary gland was also studied using the dye dilution technique, and revealed large differences between animals and also with posture. These observations suggest that particular care should be taken when sampling blood from the milk vein of cows, if a representative sample is required. Changes in blood flow with posture may be indicative of a repartitioning of flow within the body, and the physiology of such a mechanism would be of interest in itself. The control of this mechanism may be useful in modifying blood flow to the mammary gland and thus milk yield, since blood flow is related to the level of milk production.

The objective was to compare blood flow to the hindquarters of steers measured by transit time ultrasound with blood flow determined by indicator (p-aminohippurate) dilution. Five Hereford steers had ultrasonic flow probes on the abdominal aorta and catheters in the abdominal aorta and inferior vena cava inserted through both sets of circumflex iliac vessels. Indicator was infused continuously into the abdominal aorta through both arterial catheters simultaneously, then through each of the arterial catheters in succession. Samples of blood from the inferior vena cava and jugular vein were taken during infusion for measurement of p-aminohippurate. Blood flow determined by the ultrasonic flow probe was averaged over each blood sampling interval. Compared with the ultrasonic flow probe there was no difference in mean blood flow measured by p-aminohippurate, regardless of method of infusion. Correlation of individual values between ultrasound and p-aminohippurate was .87 when p-aminohippurate was infused into both arterial catheters, .44 when p-aminohippurate was infused into the left arterial catheter, and .78 when p-aminohippurate was infused into the right arterial catheter. The respective ranges for ultrasonic measurements and p-aminohippurate were 3.62 to 10.99 L/min and 2.25 to 30.43 L/min. Although means by the two methods do not differ, there is a greater range and incidence of occasional high values with p-aminohippurate dilution.

A useful approach of the study of nutrient absorption and metabolism is in vivo measurement of blood flow across portal-drained viscera and liver, and flux of bloodborne metabolites, successful application of the approach requires correct placement of chronic catheters in appropriate blood vessels. Additionally, catheters must stay patent long enough to allow the animal to recover from surgery and to complete an experimental protocol. This paper describes surgical techniques to install chronic catheters in mesenteric veins, the hepatic portal vein, and an hepatic vein of cattle. Techniques for access to arterial blood are described also. Materials, equipment, and supplies required for surgery, blood sampling, and blood flow determination are described. Commercial sources of supplies are suggested. Blood flow is measured by downstream dilution of para-aminohippurate, which is infused into a mesenteric vein. Examples of blood flow data for three types of cattle are provided.

Lactating cattle were used to validate a transit time ultrasonic blood flow metering system for measuring mammary gland arterial blood flow. Blood flow probes were surgically placed around the right external pudic artery. An electromagnetic flow probe was implanted in tandem with the ultrasonic probe in two cows for comparative measurements. The absolute accuracy of the implanted flow probes was assessed in vivo by mechanical means on anesthetized cows after 2 to 3 wk of implantation. The zero offset of the ultrasonic probes ranged from -12 to 8 ml/min. When the ultrasonic probe was properly implanted, the slopes of the calibration curves were linear and ranged from .92 to .95, tracking absolute flow to within 8%. The transit time instrument's performance was examined under a variety of physiological conditions. These included milking and hormone injections. The transit time ultrasonic flow meter accurately measured physiological changes in mammary arterial blood flow in chronically prepared conscious cattle. Blood flow increased 29% during milking. Epinephrine decreased mammary blood flow by 90 to 95%. Oxytocin doses increased mammary blood flow by 15 to 24%.

With the advancement of gestation, blood flow increases preferentially to the caruncular bed of the gravid uterus in association with a decreasing sensitivity of the uterus to the vasoconstrictive effects of circulating catecholamines. This study directly compared the sensitivity of the caruncular artery (CA) of the isolated bovine placentome to phenylephrine (PE), a specific alpha 1-adrenergic receptor (AR) agonist, with that to norepinephrine (NE) and epinephrine (E), both of which are alpha 1-, alpha 2-, and beta-AR agonists, at two stages of gestation (140 to 170 d, mid-pregnant; 210 to 270 d, late pregnant). The CA of each placentome was perfused with oxygenated Krebs Ringer solution into which PE, NE or E were administered; increases in intra-arterial pressure were recorded. Further, NE content and numbers of alpha 1- and alpha 2-AR in the CA, intercaruncular arteries (ICA) and uterine arteries (UA) were quantitated. The CA from mid-pregnant cows exhibited greater (P less than .05) contractile responses to NE and E than did the CA from late pregnant cows, whereas responsiveness to PE remained constant. No difference in NE content, alpha 1-AR or alpha 2-AR numbers were observed in the UA, ICA or CA between mid-pregnant or late pregnant cows. Alpha 1-AR numbers were similar in CA, ICA and UA. However, CA contained threefold greater alpha 2-AR numbers than either the ICA or UA (50.2 +/- 6.1 vs 14.6 +/- 1.6 and 14.8 +/- 2.4 fmol/mg protein, respectively; P less than .01).(ABSTRACT TRUNCATED AT 250 WORDS)

The effect of xylazine (Rompun) upon hepatic glucose production and blood flow rate was measured in two cows. Doses of 0-16 or 0-18 mg/kg bodyweight increased blood glucose concentrations by 200 per cent and hepatic glucose production by 400 per cent. Maximum blood glucose concentrations were reached approximately 40 minutes after dosing and did not start to fall until 185 minutes. Concentrations were near normal 24 hours after dosing. The increase in hepatic glucose production was greatest 20 minutes after dosing and production had returned to control rates 150 minutes after dosing. Visceral glucose utilisation was also increased. Blood flow rates in the hepatic and portal veins were reduced to 50 to 60 per cent of their predosing values. It is concluded that the prolonged hyperglycaemia which persists beyond 150 minutes is produced either by continued glucose production from sites other than liver and viscera or by reduced utilisation of the blood glucose by peripheral tissue.

Blood flow to each uterine horn of cows during the oestrous cycle and early pregnancy was determined daily by use of electromagnetic blood flow probes placed around both middle uterine arteries. The pattern of blood flow to uteri of pregnant and non-pregnant cows was similar until Day 14 after mating or oestrus. Between Days 14 and 18 of pregnancy blood flow to the uterine horn containing the conceptus increased (P less than 0.01) 2- to 3-fold, whereas blood flow to the non-gravid uterine horn in these cows remained constant. No corresponding increase in blood flow to the uterine horn ipsilateral to the ovary bearing the CL was observed in non-pregnant cows during this 4-day period. By Day 19 of pregnancy, blood flow to the gravid uterine horn had returned to a level similar to that observed on Day 13. Blood flow to both uterine horns of pregnant cows remained constant from Days 19 to 25 and then increased to the gravid horn (P less than 0.01) markedly until Day 30 whereas blood flow to the non-gravid horn remained low. Uterine blood flow during the oestrous cycle of non-pregnant cows was positively correlated (P less than 0.01) with systemic concentrations of oestradiol and the ratio of oestradiol (pg/ml) to progesterone (ng/ml). There was no association between oestradiol concentrations and blood flow to the gravid uterine horn. These data indicate local control of uterine blood flow by the bovine conceptus which may function to create optimal conditions for the continuation of pregnancy.
