The adsorption of lignin to cellulase is the major obstacle in the sugar-platform conversion of lignocellulosic bioresources. In this study, the adsorption behavior of untreated and pretreated lignin samples from corn stover to cellulase was investigated, in particular the effects of lignin adsorption on the composition and spatial conformation of free enzymes were explored. The results showed that pretreatments decreased the hydrophobic groups contents of lignin, i.e., aromatic ring, ether and carbonyl, as well as the content of ionizable group, i.e., carboxyl, which reduced its hydrophobicity and negative charge density, thus weakening the adsorption ability of lignin to cellulase. The lignin samples mainly adsorbed the CBHII component of cellulase to inhibit the synergistic effect of free enzymes. Lignin adsorption altered the spatial position of tryptophan residues in free enzymes, exposing them to the protein surface. In addition, the secondary structure of free enzymes was altered, with a decrease in the alpha-helix content and an increase in the random coil content, thus loosening the spatial conformation of free enzymes. The change degree in the spatial structure of free enzymes correlated with the adsorption capacity of the lignin, i.e., lignin with low adsorption capacity caused the least damage to free enzyme, with NaOH pretreated lignin being the best. It appears that appropriate pretreatment and chemical modification of enzymes to resist lignin adsorption is a promising long-term pathway to overcome the lignin inhibition during sugar-platform conversion of lignocellulosic bioresources. Effective enzymatic hydrolysis of lignocellulosic materials to produce fermentable sugars requires close cooperation between cellulase components. However, lignin remaining in the pre-treated substrate plays an inhibitory role in the synergistic action of enzymes through irreversible adsorption. In this study, the effect of cellulolytic enzyme lignin (CEL) isolated from pre-treated furfural residues and dissolved lignins (DLs) precipitated from black liquor on cellulase and cellobiase adsorption behavior was investigated. Green liquor (GL) combined with H2O2 (GL-H2O2) or ethanol (GL-ethanol) pre-treatments markedly decreased the adsorption capacity of CELs. However, the DLs had the strongest binding ability, which was approximately two to three times higher than that of CELs. According to SDS–PAGE analyses, only CBHII and EGII in cellulase and enzymes of 245 kDa and 55 kDa molecular weight in cellobiase were easily bound onto CELs and DLs. Solid-state 13C NMR spectra and FT-IR analysis indicated that pre-treatments reduced the hydrophobic structure of aromatic rings, β-O-4, and carbonyl, and increased the number of hydrophilic carboxyl groups in CELs. For this reason, the hydrophobicity of CELs was decreased, thus reducing their adsorption capacity. Compared with GL-H2O2 pre-treatment, GL-ethanol pre-treatment had a stronger effect on decreasing enzyme adsorption of lignin due to its high hydrophilicity. Heavy damage to aromatic structures, low molecular weight, and the large number of aliphatic portions in DLs caused their strong affinity for enzymes, although their BET surface area and hydrophobicity were low. The introduction of specific choline chloride (ChCl)- and betaine (Bet)-based deep eutectic solvents (DESs) into the conventional acetic acid–sodium acetate solution led to a significant increase in cellulase activity and stability. In particular, a higher enhancement in β-glucosidase activity was obtained, which plays a key role in improving cellulase function. The DESs prolonged the half-life of cellulase. The addition of 10 % (v/v) ChCl/1,3-propanediol (1:2) and 5 % (v/v) Bet/glycerol (1:2) resulted in the most significant improvement, with the half-life of cellulase reaching three times the initial half-life value. The above both DESs allowed the cellulase to maintain an excellent level of activity over time, even after 72 h. The DESs tightened the structure of the enzyme by changing its secondary structure, which had a stabilizing effect on the overall conformation. The density functional theory calculations of DESs revealed that the DES with a more stable structure more effectively protected the active conformation. The molecular dynamics simulation on β-glucosidase indicated that the hydrogen bond donor end of the DES primarily formed hydrogen bonds with the enzyme, which induced a spatially compact conformation by increasing the α-helix content while reducing the β-sheet content. It also restricted the movement of amino acids, particularly stabilizing the active site, and increased the hydrophilic region, thereby enhancing enzyme activity and stability. In addition, the DES formed a protective layer that prevented the unproductive adsorption of lignin on the enzymes during the hydrolysis of lignocellulosic feedstocks. Herein, a green facile approach to improve the flexibility of unbleached bamboo kraft pulp (UBKP) via an immobilized enzyme technology is proposed. Polydopamine (PDA) acts as versatile modification and coating materials of cellulose nanocrystals (CNC) for assembling versatile bio-carriers (PDA@CNC). Cellulase biomacromolecules are efficiently immobilized on PDA@CNC to form cellulase@PDA@CNC nanocomposites. The relative enzyme activity, temperature/pH tolerance, and storage stability of cellulase were significantly improved after immobilization. The degree of polymerization treated UBKP decreased by 5.42 % (25 U/g pulp) compared to the control sample. The flexibility of treated fibers was 6.61 × 1014/(N·m2), which was 96.93 % higher (25 U/g) compared to the control and 3.88 times higher than that of the blank fibers. Cellulase@PDA@CNC performs excellent accessibility to fiber structure and induces high degree of fibrillation and hydrolysis of UBKP fibers, which contributes high softness of obtained tissue handsheets. The bio-carrier PDA@CNC within paper framework may further enhance tissue tensile strength. This study proposes a practical and environmentally friendly immobilization approach of cellulase@PDA@CNC for improving the hydrolysis efficiency and flexibility of UBKP fibers, which provides the possibility to maintain the strength of tissue paper while improving its softness, thus broadening the high-value application of immobilized enzyme technology in tissue production. Abstract In this work, cellulase, low-concentration cold alkali and cellulase combined with cold alkali were used to pretreat unbleached bagasse pulp from which cellulose nanofibrils (CNFs), about 30 nm in diameter, were successfully prepared through ultrafine grinding and high-pressure homogenization. X-ray diffraction analysis showed that cellulase pretreatment increased the crystallinity of CNFs. After low-concentration cold alkali pretreatment, the crystallinity of CNFs significantly reduced and the crystal structure of the cellulose changed from type I to type II. Thermogravimetric analysis showed that CNFs prepared by cellulase combined with cold alkali treatment produced more regenerated cellulose and had lower thermal stability. The use of cellulase and low-concentration cold alkali pretreatments combined with ultrafine grinding and high-pressure homogenization is an environment-friendly method for preparing CNFs. Tobacco based cellulose nanofiber (TCNF) is a novel nanocellulose that has recently been used to replace undesirable wood pulp fibers in the preparation of reconstructed tobacco sheets (RTS). However, given the strict requirements for controlling toxic chemical content in tobacco products, there is a global interest in developing a green, efficient, and toxic-chemical free approach to isolate TCNF from tobacco stem as a bioresource. In this study, we propose a creative and environmentally friendly method to efficiently and safely isolate TCNF from tobacco stem pulp, which involves integrated biological pretreatment followed by a facile mechanical defibrillation process. Feruloyl esterase is used to pretreat the stem pulp by disrupting the ether and ester bonds between lignin and polysaccharide carbohydrates within the fiber wall, which effectively facilitates cellulase hydrolysis and swelling of the stem pulp fiber, as well as the following mechanical shearing treatment for TCNF isolation. The results demonstrate that TCNF obtained by the comprehensive feruloyl esterase/cellulase/mechanical process exhibit uniform and well-dispersed nanofiber morphology, higher crystallinity, and stronger mechanical properties than those of the control. The addition of 0.5 % TCNF can replace wood pulp by 18 wt% ~ 25 wt% in the production of RTS samples while maintaining their reasonable strength properties. The lignin-derived inhibition on cellulase is the most critical bottleneck that must be addressed when converting lignocellulosic biomass through the sugar-platform process. Herein, a novel modified cellulase with high activity, great stability and excellent lignin-resistant ability was synthesized by chemical modification using hydrophilic and negatively charged polyethylene glycol (PEG) derivatives including HO-PEG-COOH and HOOC-PEG-COOH. Both modifications obviously increased the cellulase activity, with 37% (HO-PEG-COOH modification) and 53% (HOOC-PEG-COOH modification) increase, respectively. In addition, the stabilities of both modified cellulases were significantly enhanced, seeing from their much higher activities at all selected temperatures and pH values. The modification resulted in a movement of tryptophan residues to the exterior of enzyme, allowing the substrate chains to be more easily recruited and thus initiating the hydrolysis. The compactness of secondary structure for enzyme was improved, especially, the α-helix content was increased which requires strong hydrogen bonding to generate it, thus enhancing the stability. High hydrophilicity, negative charge and compactness of enzyme conferred modified cellulase good ability to resist lignin adsorption and destruction, thus improving the enzymatic hydrolysis of corn stover.
