Serotonin (5-HT) plays pivotal roles in many physiological processes including reproduction of crustaceans, which are mediated 5-HT receptors. The distributions of 5-HT and its receptor have never been explored in Portunus pelagicus. To validate the targets which indirectly indicate the roles of 5-HT in this crab, we have investigated the distribution of 5-HT in the central nervous system (CNS) and ovary using immunohistochemistry and tissue expression of its receptor by RT-PCR. In the brain, 5-HT immunoreactivity (-ir) was detected in clusters 6, 7, 8, 11, 14, 15 and the fibers. In the ventral nerve cord (VNC), 5-HT-ir was detected in pairs of neurons and the fibers connected to the neurons. In the ovary, 5-HT-ir was intense in the oocyte step 1 (Oc1) and Oc2, and its intensity was slightly decreased in Oc3 and Oc4. The 5-HT receptor was molecularly characterized to be type 7, and it was strongly expressed in the eyestalk, brain, VNC, mature ovary and muscle. Due to the presence of 5-HT receptor we suggest that 5-HT acts primarily at the CNS and ovary, thus implicating its role in reproduction especially in the development of oocytes though its exact function in this crab needed to be explored further., (Copyright © 2019 Elsevier GmbH. The mud crab, Scylla olivacea, is a high value economic marine animal in Thailand. However, collection of these crabs from natural habitat for local consumption and export has caused rapid population decline. Hence, aquaculture of this species is required and to this measure understanding of endocrine control of their reproduction must be understood. Egg laying hormone (ELH) is a neuropeptide synthesized by the bag cells (neurons) in the abdominal ganglia of Aplysia gastropods. It plays a critical role in controlling egg production and laying in gastropods, and its possible homolog (ELH-like peptide) was reported in the neural and ovarian tissues of prawns and recently in female reproductive tract of the blue swimming crab, Portunus pelagicus. In this study, we have studied the histology of the male reproductive tract in Scylla olivacea which are comprised of anterior testis, posterior testis, early proximal spermatic duct (ePSD), proximal spermatic duct (PSD), middle spermatic duct (MSD) and distal spermatic duct (DSD), by immunohistochemistry, detected an abalone ELH- immunoreactivity (aELH-ir) in epithelium of ducts in posterior testis and epithelium of all parts of spermatic duct. Furthermore, we could detect aELH-ir in neurons of cluster 9, 11, olfactory neuropil (ON) in the brain and in the small neurons located between the third and the fourth thoracic neuropils (T3-T4) and between the fourth and the fifth thoracic neuropils (T4-T5) of thoracic ganglia. Prostaglandins have important physiological roles in marine invertebrates, including larval development and reproduction. The prostaglandin E concentration fluctuates during the ovarian development of crustaceans. The biosynthetic pathway of prostaglandin, however, has not been well studied in portunid crabs, including in the mud crab, Scylla olivacea. In this study, the aim was to investigate the presence of prostaglandin E synthase (PGES), enzyme that catalyzes the terminal conversion in the prostaglandin E2 (PGE 2 ) biosynthesis, and its gene expression in the central nervous system (CNS) and ovary during ovarian maturation of S. olivacea. cDNA sequence encoding PGES was cloned from the S. olivacea ovary. The PGES transcript of S. olivacea (Scyol-PGES) consists of 1258 nucleotides, which encodes for 420 amino acid PGES protein precursor. Investigation of gene expression by RT-PCR indicated that Scyol-PGES was detected in all organs studied. Based on in situ hybridization, Scyol-PGES was detected in the I to III stages for oocyte development of Stage 3 of ovarian development, and in the CNS, including the various neuronal clusters of the brain. In the ventral nerve cord, the Scyol-PGES gene was expressed in the neurons within the subesophageal, thoracic and abdominal ganglia. The Scyol-PGES gene expression as indicated by relative abundance of mRNA in the Stage 4 of ovarian development was greater than that at Stages 1 to 3 of ovarian development. In the present study, the presence and distribution of leptin receptor (LEP-R) in central nervous system, digestive organs, gonads of giant freshwater prawn, Macrobrachium rosenbergii, were investigated with Western blot and immunohistochemistry. By Western blot a LEP-R with a molecular weight (MW) of 100 kDa was detected in the brain, thoracic ganglia, abdominal ganglia, hepatopancreas, all parts of the gastrointestinal tract, ovaries, and testes. In hepatopancreas and foregut, another intense positive band was detected at molecular weight of 30 kDa, which could be an isotype of LEP-R. By immunohistochemistry, LEP-R-ir was detected in the neurons, and neuropils in the brain, thoracic ganglia, and abdominal ganglia. In the gastrointestinal tract, there was intense LEP-R-ir in the apical part of the epithelial cells of the foregut, midgut, and hindgut. In addition, LEP-R-ir was found in the Restzellen(R)cells and Fibrillenzellen(F) cells in the hepatopancreas. In the ovary, LEP-R-ir was detected in early stage of oocytes and mature oocytes. Intense LEP-R-ir was observed in spermatogonia and spermatocytes of the small and orange claw male prawns. In addition, LEP-R was seen in the high epithelium of spermatic ducts from all male morphotypes. In summary, the detection of the LEP-R-ir suggests the existence of a LEP-R in several organs of M. rosenbergii. Our previous studies have demonstrated that lamprey gonadotropin-releasing hormone-III (lGnRH-III)-like peptide occurs in the central nervous system (CNS) of decapod crustaceans (Macrobrachium rosenbergii, Penaeus monodon, Portunus pelagicus), and that lGnRH-III is the most potent in stimulating ovarian maturation compared with other GnRH isoforms. In this study, we examined the localization of lGnRH-III-like peptide in the CNS and male reproductive organs of the blue swimming crab by using anti-lGnRH-III as a probe. In the brain, lGnRH-III immunoreactivity (-ir) was detected in neurons of clusters 6, 10, 11, 14/15, 16, and 17 and in many neuropils. In the subesophageal ganglion, lGnRH-III-ir was present in neurons of the dorso-lateral and ventro-medial clusters. In the thoracic ganglia, lGnRH-III-ir was observed in the large-sized neurons between the thoracic neuropils and in the ventromedial cluster of the abdominal ganglia. In the testis, lGnRH-III-ir was detected in nurse cells, hemocytes, spermatids 2, and the outer and inner zones of the acrosomes of spermatozoa. Bioassay showed that lGnRH-III significantly increased the testis-somatic index, the percentage of late stages of seminiferous tubules (stages VII-IX), the diameter of the seminiferous tubules, and the number of BrdU-labeled early germ cells compared with the control groups. Thus, lGnRH-III-like peptide exists in the male crab and possibly enhances germ cell proliferation and maturation in the testes, leading to increased sperm production.

In crustaceans serotonin (5-HT) and dopamine (DA) are neurotransmitters that play roles in the modulation of numerous physiological functions, including reproduction. However, in the mud crab, Scylla olivacea, the distributions of 5-HT and DA in the CNS have not yet been investigated. The aim of our study was to map the distributions of these two neurotransmitters in the central nervous system (CNS) of the female of this crab during the late stage of ovarian development. We found 5-HT immunoreactivity (-ir) and DA-ir in many parts of the CNS, including the eyestalk, brain, and thoracic ganglia. In the eyestalk, 5-HT-ir was localized in the medulla terminalis (MT), hemi-ellipsoid body (HB), and protocerebral tract (PT), whereas DA-ir was present in neuronal cluster 1, the LG neuropils, and PT. In the brain, 5-HT-ir and DA-ir were detected in cells and fibers of neuronal clusters 6, 7, 8, 9, 10, 11, 14, and 15. In the ventral nerve cord, 5-HT-ir was present in neurons of the abdominal ganglia, whereas DA was only present in fibers. These spatial distributions of 5-HT and DA suggest that they may be involved in the neuromodulation of important physiological functions, including ovarian maturation, as shown in other non-crab decapods., (Copyright © 2014 Elsevier GmbH. Octopamine (OA) is a major neurotransmitter that has not been studied in the Pacific white shrimp, Litopenaeus vannamei. Therefore, we investigated changes in OA levels, its distribution in regions of the central nervous system (CNS) and ovary during the ovarian maturation cycle, as well as its possible role in regulating ovarian maturation. OA exhibited the highest concentration in the brain and thoracic ganglia at ovarian stage II, and then declined to the lowest concentration at ovarian stages III and IV. In the cerebral ganglia, OA-immunoreactivity (OA-ir) was present in neurons of clusters 6, 17, the anterior and posterior medial protocerebral, olfactory, antenna II, and tegumentary neuropils. In the circumesophageal, subesophageal, thoracic ganglia and abdominal ganglia, OA-ir was detected in several neuropils, neurons and fibers. The high level of intensity in OA immunostaining was observed in early developmental stage of oocyte by comparison with low level of OA-ir in late stages of oocyte development. Functionally, OA-injected female shrimps at doses of 2.5×10(-7) and 2.5×10(-6)mol/shrimp, showed significantly decreased gonado-somatic indices, oocyte diameters, and hemolymph vitellogenin levels, compared with control groups. This study showed changes of OA in the CNS and ovary reaching the highest level in early ovarian stages and declining in late stages, and it decreased hemolymph vitellogenin levels, suggesting significant involvement of OA in female reproduction in this species., (Copyright © 2014 Elsevier Inc. The mud crab, Scylla olivacea, is one of the most economically valuable marine species in Southeast Asian countries. However, commercial cultivation is disadvantaged by reduced reproductive capacity in captivity. Therefore, an understanding of the general and detailed anatomy of central nervous system (CNS) is required before investigating the distribution and functions of neurotransmitters, neurohormones, and other biomolecules, involved with reproduction. We found that the anatomical structure of the brain is similar to other crabs. However, the ventral nerve cord (VNC) is unlike other caridian and dendrobrachiate decapods, as the subesophageal (SEG), thoracic and abdominal ganglia are fused, due to the reduction of abdominal segments and the tail. Neurons in clusters within the CNS varied in sizes, and we found that there were five distinct size classes (i.e., very small globuli, small, medium, large, and giant). Clusters in the brain and SEG contained mainly very small globuli and small-sized neurons, whereas, the VNC contained small-, medium-, large-, and giant-sized neurons. We present a detailed histological description of the central nervous system (CNS: brain, subesophageal ganglion, thoracic ganglia, abdominal ganglia) of the blue crab, Portunus pelagicus. Because the presence of gonadotropin-releasing hormone (GnRH) in crustaceans has been disputed, we examine the presence and localization of a GnRH-like peptide in the CNS of the blue crab by using antibodies against lamprey GnRH (lGnRH)-III, octopus GnRH (octGnRH) and tunicate GnRH (tGnRH)-I. These antibodies showed no cross-reactivity with red-pigment-concentrating hormone, adipokinetic hormone, or corazonin. In the brain, strong lGnRH-III immunoreactivity (-ir) was detected in small (7-17 μm diameter) neurons of clusters 8, 9 and 10, in medium-sized (21-36 μm diameter) neurons of clusters 6, 7 and 11 and in the anterior and posterior median protocerebral neuropils, olfactory neuropil, median and lateral antenna I neuropils, tegumentary neuropil and antenna II neuropil. In the subesophageal ganglion, lGnRH-III-ir was detected in medium-sized neurons and in the subesophageal neuropil. In the thoracic and abdominal ganglia, lGnRH-III-ir was detected in medium-sized and small neurons and in the neuropils. OctGnRH-ir was observed in neurons of the same clusters with moderate staining, particularly in the deutocerebrum, whereas tGnRH-I-ir was only detected in medium-sized neurons of cluster 11 in the brain. Thus, anti-lGnRH-III shows greater immunoreactivity in the crab CNS than anti-octGnRH and anti-tGnRH-I. Moreover, our functional bioassay demonstrates that only lGnRH-III has significant stimulatory effects on ovarian growth and maturation. We therefore conclude that, although the true identity of the crab GnRH eludes us, crabs possess a putative GnRH hormone similar to lGnRH-III. The identification and characterization of this molecule is part of our ongoing research.

The crustacean X-organ-sinus gland (XO-SG) complex controls molt-inhibiting hormone (MIH) production, although extra expression sites for MIH have been postulated. Therefore, to explore the expression of MIH and distinguish between the crustacean hyperglycemic hormone (CHH) superfamily, and MIH immunoreactive sites (ir) in the central nervous system (CNS), we cloned a CHH gene sequence for the crab Portunus pelagicus (Ppel-CHH), and compared it with crab CHH-type I and II peptides. Employing multiple sequence alignments and phylogenic analysis, the mature Ppel-CHH peptide exhibited residues common to both CHH-type I and II peptides, and a high degree of identity to the type-I group, but little homology between Ppel-CHH and Ppel-MIH (a type II peptide). This sequence identification then allowed for the use of MIH antisera to further confirm the identity and existence of a MIH-ir 9kDa protein in all neural organs tested by Western blotting, and through immunohistochemistry, MIH-ir in the XO, optic nerve, neuronal cluster 17 of the supraesophageal ganglion, the ventral nerve cord, and cell cluster 22 of the thoracic ganglion. The distributions of neuropeptides in paraffin-embedded tissue sections (PETS) of the eyestalk, brain, and thoracic ganglia of the shrimp Penaeus monodon were visualized by imaging mass spectrometry (IMS). Peptide signals were obtained from PETS without affecting morphological features. Twenty-nine neuropeptides comprising members of FMRFamide, SIFamides, crustacean hyperglycaemic hormone, orcokinin-related peptides, tachykinin-related peptides, and allatostatin A were detected and visualized. Among these findings we first identified tachykinin-related peptide as a novel neuropeptide in this shrimp species. We found that these neuropeptides were distributed at specific areas in the three neural organs. In addition, 28 peptide sequences derived from 4 types of constitutive proteins, including actin, histones, arginine kinase, and cyclophilin A were also detected. All peptide sequences were verified by liquid chromatography-tandem mass spectrometry. The use of IMS on acetic acid-treated PETS enabled us to identify peptides and obtain their specific localizations in correlation with the undisturbed histological structure of the tissue samples., (Copyright © 2011 Elsevier Inc. We investigated changes in serotonin (5-HT) and dopamine (DA) levels and in their distribution patterns in the central nervous system (CNS) and ovary during the ovarian maturation cycle in the Pacific white shrimp, Litopenaeus vannamei. The concentrations of these two neurotransmitters were determined by using high performance liquid chromatography with electrochemical detection. The 5-HT concentration exhibited a gradual increase in the brain and thoracic ganglia during early ovarian stages I, II, and III, reaching a maximum at the mature ovarian stage IV, whereas DA showed its highest concentration at ovarian stage II in the brain and thoracic ganglia and then declined to its lowest concentration at ovarian stage IV. In the ovaries, 5-HT was lowest at ovarian stage I and gradually increased to a peak at ovarian stage IV. Conversely, the concentration of DA was highest at ovarian stages I and II and lowest at ovarian stage IV. In the brain, 5-HT immunoreactivity (-ir) from stage IV and DA-ir from stage II were distributed extensively in neurons of clusters 6, 11, and 17, in fibers, and in the anterior and posterior medial protocerebral, olfactory, antenna II, and tegumentary neuropils. In the circumesophageal, subesophageal, thoracic, and abdominal ganglia, both 5-HT-ir and DA-ir were detected in neuropils and surrounding neurons and fibers. 5-HT-ir and DA-ir were more intense in the thoracic ganglia than in other parts of the CNS. In the ovary, 5-HT-ir exhibited high intensity in late oocytes, whereas DA-ir was more intense in early oocytes. Thus, opposing changes occur in the levels of these two neurotransmitters and in their specific localizations in the CNS and ovary during ovarian maturation, indicating their important involvement in female reproduction.

Immunohistochemistry was used to identify the distribution of both APGWamide-like and RFamide-like peptides in the central nervous system (CNS) and ovary of the mature female giant freshwater prawn, Macrobrachium rosenbergii. APGWamide-like immunoreactivity (ALP-ir) was found only within the sinus gland (SG) of the eyestalk, in small- and medium-sized neurons of cluster 4, as well as their varicosed axons. RFamide-like immunoreactivity (RF-ir) was detected in neurons of all neuronal clusters of the eyestalk and CNS, except clusters 1 and 5 of the eyestalk, and dorsal clusters of the subesophageal, thoracic, and abdominal ganglia. The RF-ir was also found in all neuropils of the CNS and SG, except the lamina ganglionaris. These immunohistochemical locations of the APGWamide-like and RF-like peptides in the eyestalk indicate that these neuropeptides could modulate the release of the neurohormones in the sinus gland. The presence of RFamide-like peptides in the thoracic and abdominal ganglia suggests that it may act as a neurotransmitter which controls muscular contractions. In the ovary, RF-ir was found predominantly in late previtellogenic and early vitellogenic oocytes, and to a lesser degree in late vitellogenic oocytes. These RFs may be involved with oocyte development, but may also act with other neurohormones and/or neurotransmitters within the oocyte in an autocrine or paracrine manner.

We used antibodies against octopus gonadotropin-releasing hormone (octGnRH) and tunicate GnRH (tGnRH-I) in order to investigate the existence and distribution of GnRH-like peptides in the central nervous system (CNS) and in the ovary during various stages of the ovarian cycle of the white shrimp, Litopenaeus vannamei. OctGnRH-immunoreactive and tGnRH-I-immunoreactive neurons and fibers were present in several regions of the supraesophageal ganglion (brain), subesophageal ganglion (SEG), thoracic ganglia, and abdominal ganglia. In the brain, both octGnRH immunoreactivity (ir) and tGnRH-I-ir were detected in neurons of clusters 6, 11, 17, and associated fibers, and the anterior medial protocerebral, posterior medial protocerebral, olfactory, and tegumentary neuropils. In the SEG and thoracic ganglia, octGnRH-immunoreactive and tGnRH-I-immunoreactive neurons and fibers were present in dorsolateral and ventromedial cell clusters and in surrounding fibers. Only immunoreactive fibers were detected in the abdominal ganglia. In the ovary, both octGnRH and tGnRH-I were detected at medium intensity in the cytoplasm of early step oocytes (Oc2) and, at high intensity, in Oc3. Furthermore, octGnRH-ir and tGnRH-I-ir were intense in follicular cells surrounding Oc2 and Oc3. The presence of GnRH-ir in the CNS and ovary indicates that GnRH-like peptides occur in the white shrimp, and that GnRHs are involved in the reproductive process, especially ovarian maturation and the differentiation of oocytes, as reported in other species.

Dopamine (DA), octopamine (OA) and serotonin (5-HT) are the key neurotransmitters that control gonadal development in decapod crustaceans. 5-HT stimulates, while DA and OA delay gonadal development in Macrobrachium rosenbergii. In the present study, we have further investigated the distribution patterns of DA and OA in the central nervous system (CNS) and ovary during various stages of the ovarian maturation cycle of this giant freshwater prawn. DA- and OA-immunoreactive neurons and fibers were distributed extensively in several regions of the brain, subesophageal ganglion (SEG), thoracic ganglia and abdominal ganglia. In the brain, the two neurotransmitters were present in neurons of clusters 6, 7, 11, 17, and nearby neuropil regions. In the SEG, thoracic ganglia and abdominal ganglia, immunoreactive neurons and fibers were found along the midline and in several neuronal clusters around each neuropil region. Staining for DA and OA was more intense in the thoracic ganglia than in other parts of the CNS. In the ovary, DA- and OA-immunoreactivities were present at high intensity in early oocytes. The presence of DA- and OA-immunoreactivities in neural ganglia as well as ovary suggests that DA and OA may also be involved in the reproductive process, particularly ovarian development and differentiation of oocytes in this species.

Serotonin or 5-hydroxytryptamine (5-HT) and dopamine (DA) are the two key neurotransmitters that control gonadal development in decapod crustaceans. This study investigated changes in the levels of 5-HT and DA in the CNS and ovary during different phases of the ovarian cycle of the freshwater prawn, Macrobrachium rosenbergii. The levels of 5-HT and DA were quantified by using High Performance Liquid Chromatography with electrochemical detection (HPLC-ECD). Moreover, changes of vitellogenin (Vg) concentrations in the hemolymph after treatment with 5-HT and DA (at doses of 2.5 x 10(-6) and 2.5 x 10(-7)mol per prawn) were also examined. 5-HT exhibited a gradual increase in concentration in the brain and thoracic ganglia from ovarian stage I (0.12+/-0.01 nmol/mg, 0.22+/-0.01 nmol/mg, respectively) to reach a maximum (0.66+/-0.03 nmol/mg, 1.48+/-0.03 nmol/mg, respectively) at ovarian stage IV. In contrast, DA in the brain and thoracic ganglia showed the highest concentrations at ovarian stage II (0.20+/-0.01 nmol/mg, 1.27+/-0.06 nmol/mg, respectively) and then decreased to the lowest concentrations (0.06+/-0.01 nmol/mg, 0.28+/-0.04 nmol/mg, respectively) at ovarian stage IV. The ovarian concentration of 5-HT was 0.53+/-0.11 nmol/mg at ovarian stage I and gradually increased to 1.63+/-0.16 nmol/mg at ovarian stage IV. In contrast, the concentration of DA was highest at ovarian stage I (29.05+/-1.31 nmol/mg), and lowest at the ovarian stage IV (11.43+/-0.74 nmol/mg). Injecting 5-HT into prawns significantly increased Vg concentration in the hemolymph at ovarian stage IV compared to control groups, and injecting DA into prawns had the opposite effect. The inverse relationship between 5-HT and DA levels in neural ganglia and ovaries, and their opposing effects on hemolymph Vg levels suggest that these two transmitters play opposite regulatory roles in controlling ovarian maturation and oocyte development in this species.

In the present study, we demonstrated the existence of GnRH-like peptides in the central nervous system (CNS) and ovary of the giant freshwater prawn, Macrobrachium rosenbergii using immunocytochemistry. The immunoreactivity (ir) of lamprey (l) GnRH-III was detected in the soma of medium-sized neurons located in neuronal cluster number 11 in the middle part of supraesophageal ganglion (deutocerebrum), whereas ir-octopus (oct) GnRH was observed in the soma of both medium-sized and large-sized neurons in thoracic ganglia, as well as in the fibers innervating the other medium-sized and large-sized neuronal cell bodies in the thoracic ganglia. In addition, ir-lGnRH-I was observed in the cytoplasm of late previtellogenic oocyte and early vitellogenic oocyte. These data suggest that M. rosenbergii contain at least three isoforms of GnRH: two GnRH isoforms closely related to lGnRH-III and octGnRH in the CNS, whereas another isoform, closely related to lGnRH-I, was localized in the ovary. This finding provides supporting data that ir-GnRH-like peptide(s) may exist in this decapod crustacean.

The distribution and presence of gonadotropin-releasing hormone (GnRH) in the central nervous system (CNS) of Penaeus monodon were examined by immunocytochemistry, high performance liquid chromatography (HPLC), and radioimmunoassay (RIA). We demonstrated the existence of octopus (oct)GnRH-liked immunoreactivity (ir-octGnRH) and lamprey (l)GnRH-III-liked immunoreactivity (ir-lGnRH-III) in cell bodies of medium-sized neurons of the anterior part (protocerebrum) of the supraesophageal ganglion (brain). In addition, only the ir-octGnRH was detected in the nerve fibers located in the brain and segmental ganglia (subesophageal, thoracic, and abdominal ganglia). Moreover, some branches of these fibers also innervated the neurons in the middle (deutrocerebrum), posterior (tritocerebrum) brain and segmental ganglia. There was no ir-lGnRH-I and ir-salmon (s)GnRH detected in the shrimp CNS. The results from HPLC and RIA showed ir-GnRH in the CNS using anti-lGnRH-III, but not with anti-mammalian (m)GnRH. The data from immunocytochemistry, HPLC and RIA suggest that ir-GnRH in shrimp may be more similar to octGnRH and lGnRH-III than the other forms. These findings support the hypothesis that GnRH-liked factor(s) may be an ancient peptide that also exists in this decapod crustacean.

Gamma-aminobutyric acid (GABA) is a neurotransmitter that is widely spread in vertebrate and invertebrate nervous systems and modulates essential physiological roles. Previous studies have reported the distribution of several neurotransmitters throughout the central nervous system (CNS) of decapod crustaceans. However, the existence and distribution of GABA in the mud crab's, Scylla olivacea , CNS has still not been reported. In this study, we investigated the distribution of GABA using immunohistochemistry. The result revealed that GABA immunoreactivity (-ir) was observed in neurons and fibres throughout the CNS, including the eyestalk, brain, and ventral nerve cord of S. olivacea. Therefore, the existence and extensive distribution pattern of GABA in the CNS of the male mud crab suggest its possible roles in feeding, locomotion, and also reproduction. We previously analyzed the central nervous system (CNS) transcriptome and found three isotypes of long neuropeptide F (MrNPF-I, −II, −III) and four isoforms of short NPF (sMrNPF) in the giant freshwater prawn, Macrobrachium rosenbergii. We now validate the complete sequences of the MrNPF-I and −II precursor proteins, which show high similarity (91-95 %) to NPFs of the penaeus shrimp (PsNPF). MrNPF-I and -II precursors share 71 % amino acid identity, whereas the mature 32-amino-acid MrNPF-I and 69-amino-acid MrNPF-II are identical, except for a 37-amino-acid insert within the middle part of the latter. Both mature MrNPFs are almost identical to PsNPF-I and −II except for four amino acids at the mid-region of the peptides. Reverse transcription plus the polymerase chain reaction revealed that transripts of MrNPF-I and -II were expressed in various parts of CNS including the eyestalk, brain and thoracic and abdominal ganglia, with the highest expression occurring in the brain and thoracic ganglia and with MrNPF-I showing five- to seven-fold higher expression than MrNPF-II. These peptides were also expressed in the midgut hindgut, and hepatopancreas, with MrNPF-I expression in the former two organs being at the same level as that in the brain and thoracic ganglia and about 4-fold higher than NPF-II. The expression of NPFs was also detected in the testes and spermatic duct but appeared much weaker in the latter. Other tissues that also expressed a considerable amount of NPF-I included the hematopoeitic tissue, heart and muscle. By immunohistochemistry, we detected MrNPFs in neurons of clusters 2, 3 and 4 and neuropils ME, MT and SG of the optic ganglia, neurons in cluster 6 and neuropils AMPN, PMPN, PT, PB and CB of the medial protocerebrum, neurons in clusters 9 and 11 and neurophils ON and OGTN of the deutocerebrum and neurons in clusters 14, 15 and 16 and neuropils TN and AnN of the tritocerebrum. Because of their high degree of conservation and strong and wide-spread expression in tissues other than CNS, we believe that, in addition to being a neuromodulator in controlling feeding, MrNPFs also play critical roles in tissue homeostasis. Macrobrachium rosenbergii is the most economically important of the cultured freshwater crustacean species, yet there is currently a deficiency in genomic and transcriptomic information for research requirements. In this study, we present an in silico analysis of neuropeptide genes within the female M. rosenbergii eyestalk, central nervous system, and ovary. We could confidently predict 37 preproneuropeptide transcripts, including those that encode bursicons, crustacean cardioactive peptide, crustacean hyperglycemic hormones, eclosion hormone, pigment-dispersing hormones, diuretic hormones, neuropeptide F, neuroparsins, SIFamide, and sulfakinin. These transcripts are most prominent within the eyestalk and central nervous system. Transcript tissue distribution as determined by reverse transcription-polymerase chain reaction revealed the presence of selected neuropeptide genes of interest mainly in the nervous tissues while others were additionally present in the non-nervous tissues. Liquid chromatography-mass spectrometry analysis of eyestalk peptides confirmed the presence of the crustacean hyperglycemic hormone precursor. This data set provides a strong foundation for further studies into the functional roles of neuropeptides in M. rosenbergii, and will be especially helpful for developing methods to improve crustacean aquaculture. The giant freshwater prawn, Macrobrachium rosenbergii , is an important aquaculture species in many Asian-Pacific countries due to its high economic value. A better understanding of hormones that regulate reproductive processes, including development and maturation of gametes, will facilitate more efficient breeding and larval production in prawn aquaculture. In this study, we identified four gene transcripts from the M. rosenbergii that encode crustacean female sex hormones (CFSHs), which were designated as MroCFSH1a, MroCFSH1b, MroCFSH2a and MroCFSH2b , based on primary sequence and phylogeny. We also show that CFSH may have evolved from an ancestral protein similar to the interleukin 17. All MroCFSH genes are present in the central nervous system (CNS) as well as in other tissues, including the ovaries and testes. The expression level of all MroCFSH during ovarian development is significantly higher in the CNS compared to the ovary. In the brain, MroCFSH expression is localized to small-sized neurons of neuronal clusters 9, 10, and 11, and in the medium- and large-sized neurons of neuronal clusters 14 and 15; regions known to regulate feeding and reproductive behaviors. In the ovary, MroCFSHs expression are found in the oogonia, previtellogenic and early vitellogenic oocytes. Egg-laying hormone (ELH) is a neuropeptide secreted from the neural ganglia of mollusks that can stimulate the egg-laying and associated behaviors. In the present study, using a polyclonal antibody against an abalone, H asinina , ELH (aELH) we could detect the aELH-like immunoreactivity (aELH-l-ir) in the central nervous system (CNS) and ovary of the blue swimming crab Portunus pelagicus . In protocerebrum, aELH-l-ir was detected in the small- and medium- sized neurons of cluster 6, anterior/posterior medial protocerebral neuropils, protocerebral bridge and central body. In deutocerebrum, the aELH-l-ir was observed in small-sized neurons of cluster 9, fibers between small-sized neurons of cluster 10, medium-sized neurons of cluster 11, olfactory neuropil and medial/lateral antenna I neuropils. In tritocerebrum, the medium- sized neurons of cluster 16, the tritocerebral commissure, and tegumental neuropil showed strong aELH-l-ir. In the ventral nerve cord, the aELH-l-ir was detected in the small- and medium-sized neurons of dorsolateral clusters of subesophageal ganglion. There was no aELH-l- ir in neurons of thoracic ganglion, but aELH-l-ir exhibited a dot-like structure in the subcapsular space around the thoracic neuropils. In abdominal ganglion, the aELH-l-ir was detected in some medium-sized neurons and abdominal neuropils. In the ovary, the aELH-l-ir was detected in the cytoplasm of oogonia, and all steps of developing oocytes. The effect of aELH on ovarian maturation in the crabs was examined by bioassay. When mature female crabs were injected with a dose of 500 ng/g body weight per crab, the ovarian maturation was accelerated as indicated by the rising of gonado-somatic index and levels of vitellin in the ovarian tissue and hemolymph, which was confirmed by histological studies. The aELH also enhanced the proliferation of follicular cells as indicated by the increased number of BrdU-labeled nuclei. In addition, the aELH also significantly induced the 17β-estradiol and progesterone secretion in the cultured ovarian explant. In conclusion, this study demonstrated the existence of an aELH-liked-peptide in the CNS and ovary of the P . pelagicus , and it can induce the ovarian 17β-estradiol and progesterone production resulting in early ovarian maturation in this crab. Recently, the neuronal classification of the ivory shell Spotted Babylon, Babylonia areolata , was readily demonstrated. Regarding its importance as marine economic molluscan species, the attempt to understand the neuroendocrine regulation is necessary. This study firstly demonstrated the neurosecretory cells as well as the existence and distribution of the egg-laying hormone (ELH)-like peptide in the central nervous system (CNS) and ovary of the B. areolata . The neurosecretory cell was characterized by the cytoplasmic purple dot-like structure as stained by the Gomori’s paraldehyde fuchsin. Using the anti-abalone (a) ELH, we detected the aELH-like-peptide in neurons (Nr) and neurosecretory cells (Ns) of all ganglia including the cerebral, pleural, parietal, pedal and buccal ganglia. The aELH-like peptide was also present in the neuropil of each. It was noted that not all Ns presented the aELH-like peptide. In the ovary, the aELH-like peptide was slightly detected in early developing oocytes and strongly detected in late developing oocytes and follicular cells. This study firstly reported the evidence of ELH-like peptide in the CNS and ovary of the B. areolata. Leptin, a highly conserved adipocyte-derived hormone, plays important roles in a variety of physiological processes, including the control of fat storage and metabolic status which are linked to food intake, energy homeostasis, and reproduction in all vertebrates. In the present study, we hypothesize that leptin is also present in various organs of the fresh water prawns, Macrobrachium rosenbergii . The existence and distribution of a leptin-like peptide in prawn tissues were verified by using Western blotting (WB) and immunohistochemical detection (ID) using primary antibody against human leptin. With WB, a leptin-like peptide, having a molecular weight of 15 kDa, was detected in the brain, thoracic ganglia, abdominal ganglia, parts of the gastro-intestinal tract, hepatopancreas, adipocytes and gonads. By ID, leptin immunoreactivity (leptin-ir) was detected in the brain, thoracic ganglia and intersegmental commissural nerve fibers of abdominal ganglia. In the gastrointestinal tract, there was intense leptin-ir in the apical part of the epithelial cells of the cardiac and pyloric parts of the stomach. In the midgut and hindgut, the leptin-ir was detected in epithelial cells and basal cells located near the basal lamina of the epithelium. In addition, there was leptin-ir in the Restzellen cells in the hepatopancreas which produce digestive enzymes. In the ovary, the strong intensity of a leptin-ir was detected in the cytoplasm of middle to late stage oocytes, whereas no positive staining was detected in follicular cells. An intense leptin-ir was detected in spermatocytes and sustentacular cells in the seminiferous tubules in the testes of small and orange claw males. Taken together, the detection of the leptin-ir in several organs implicates the existence of a leptin-like peptide in various organs of the freshwater prawn; and like in vertebrates this peptide may be an important hormonal factor in controlling feeding and reproductive process. Crustacean hyperglycemic hormone (CHH) has gained wide attention for its diverse physiological functions, including in metabolism and reproduction. In this study, cDNA sequence encoding CHH was cloned from the brain and ventral nerve cord (VNC) of the mud crab, Scylla olivacea . CHH of S. olivacea ( Scyol-CHH ) consists of 642 nucleotides, which encode a 108-amino acid CHH prepropeptide. Based on in situ hybridization, Scyol-CHH was expressed in the neurons within the eyestalk, including at neuronal clusters 1–4. Scyol-CHH was also detected in the neurons within the neuronal clusters 6 and 8–11 of the brain. In the VNC, its expression was found in the neurons within the subesophageal ganglion as well as the abdominal ganglion. We found that administration of serotonin (5-HT) significantly increased the expression of Scyol-CHH in the brain and VNC at 3 and 6 h post-injections, respectively, as compared with the controls. This suggests that 5-HT may play an important role in the regulation of CHH expression in the nervous system. We demonstrated for the first time the existence and distribution of kisspeptin-like peptides (Kiss-l) and their receptors in the central nervous system (CNS) of the giant freshwater prawn, Macrobrachium rosenbergii , and that these peptides could induce the oogonial proliferation, ovarian maturation, and spawning. Using immunohistochemistry, kisspeptin1-like-signal (Kiss1-l signal) was detected in the neurons of clusters 6, 11 and 14/15 and also in olfactory neuropil (ON). Kisspeptin2-like-signal (Kiss2-l signal) was observed in clusters 6, 9, 10, 16 and 17 as well as in the anterior and posterior median protocerebral neuropils (AMPN/PMPN), ON, central body (CB), protocerebral bridge (PB), and olfactory globular tract (OGT). In the subesophageal (SEG) and thoracic ganglia (TG), both Kiss1-l and Kiss2-l signals were detected in some neurons of dorsolateral cluster (DLC), ventromedial cluster (VMC) and also in fibers of associated neuropils. In abdominal ganglia (AG), Kiss1-l signal was detected in some neuronal clusters, whereas Kiss2-l signal was detected only in fibers of the AG. Functional bioassays demonstrated that both Kiss1- and Kiss2-treated prawns showed significantly shortened ovarian cycle (16 ± 1.63 and 22.86 ± 1.07 days) compared with the control (36 ± 2 days). Moreover, both Kisses significantly increased the numbers of oogonial proliferation and spawned eggs, while the percentages of fertilized eggs in all treated groups were not significantly different from the control. These findings suggest that Kisses are highly conserved peptides that may also exist in the prawns and control their oogonial proliferation and ovarian maturation. Abstract: Egg-laying hormone (ELH) is a peptide hormone that is involved in spawning and egg-laying behaviors in many mollusks. The present study demonstrated the presence of an ELH-like hormone in the ovary of the freshwater prawn, Macrobrachium rosenbergii, using a polyclonal antibody against abalone ELH (aELH). The immunoreactivity against aELH (aELH-ir) was detected in follicular cell type 2 (Fc2) of stages 0 to III ovaries, and in the cytoplasm of mature oocytes of stage IV ovaries. In contrast, there was no aELH-ir detected in the central nervous system (CNS). The effect of aELH on reproductive processes in this species was determined by in vivo bioassays, using intramuscular (i.m.) injections. The gonadosomatic index (GSI) values of control prawns at 15days post-treatment was 0.5±0.1% (X̅±S.D.), but 5.8±0.6% when treated with 50ng/g BW of aELH, and 5.4±1.1% with a 500ng/g BW dose. The time for ovarian maturation and spawning of the control group was 40.0±3.4days, but only 22.0±2.4days following injections with 50ng/g BW of aELH, and 21.2±2.8days with a 500ng/g BW dose. However, there were no differences in the number of eggs spawned and fertilization rates after aELH treatments. These findings support the hypothesis that an ELH-like hormone is present in M. rosenbergii, and could have a significant role in female spawning. [Copyright &y& Elsevier]

The authors recently reported the presence and distribution of oxytocin/vasopressin-like peptide in Portunus pelagicus as well as demonstrated its function to inhibit ovarian steroid release (Saetan et al., 2018). Here, the full-length receptor of this peptide, namely oxytocin/vasopressin-like peptide receptor (Ppel OT/VP-like peptide receptor) is reported. The coding region of the Ppel OT/VP-like peptide receptor contained 1497 bp which translationally corresponded to 499 amino acids. Sequence analysis revealed its seven transmembrane characteristics, with -two N-linked glycosylation residues located before the first transmembrane domain (TM I). The phylogenetic tree revealed that the Ppel OT/VP-like peptide receptor was placed in the group of invertebrate OT/VP-like receptors, and was clearly distinguishable from the V1R, V2R and OTR of vertebrates. Also, this receptor gene transcript was detected in several organs of the blue swimming crab with highest abundance found in brain tissue. In situ hybridization exhibited its distribution in all neuronal clusters of the eyestalk, brain, ventral nerve cord (VNC), as well as in the ovary. Comparative gene expressions between this receptor and its corresponding peptide in immature and mature female crabs revealed no significant difference of the Ppel OT/VP-like peptide receptor gene expression in the central nervous system (CNS) and ovary. In contrast, the Ppel OT/VP-like peptide gene was shown to significantly express higher in the VNC of immature crabs and in the ovary of mature crabs. Changes in expression of this peptide gene, but not its receptor, might result in ovarian steroid release inhibition.
