The gram-negative bacterium Pseudomonas aeruginosa is an opportunistic human pathogen. Metabolomics was used to elucidate the influence of P. aeruginosa infection on the host metabolism. The used infection models, based on Caenorhabditis elegans and HeLa cells showed a substantial influence on the host central carbon metabolism. Das gram-negative Bakterium Pseudomonas aeruginosa ist ein opportunistischer, humaner Krankheitserreger. Metabolomeuntersuchungen wurden genutzt um den Einfluss einer P. aeruginosa-Infektion auf den Wirtsmetabolismus zu untersuchen. Die verwendeten Infektionsmodelle, basierend auf Caenorhabditis elegans und HeLa-Zellen, zeigten einen substantiellen Einfluss der Infektion auf den zentralen Kohlenstoffwechsel der Wirtszellen.

Non-targeted metabolomics is a promising tool for the investigation and evaluation of diseases. In this work the development of a metabolomic platform that integrates the chromatographic resolution of UPLC with the high mass accuracy of FT-ICR-MS for the analysis of human plasma samples was proposed. The work flow leads to the experimental application; the study of the non-targeted metabolome of fatty liver individuals, whose pre-diabetic state was characterized by their insulin sensitivity. Ungerichtete Metabolomik ist viel versprechendes Werkzeug für die Untersuchung und Evaluierung von Krankheiten. In dieser Arbeit wurde die Entwicklung einer Metabolomik Plattform präsentiert, welche chromatographische Auflösung mittels UPLC und hohe Massengenauigkeit mittels FT-ICR-MS für die Analyse von humanem Blutplasma verbindet. Der Arbeitsablauf wird schließlich auf Plasmata von Fette-Leber-Patienten angewandt, welche der Insulinresistenz nach ISI-Matsuda folgend klassifiziert waren.

Based on non-targeted Metabolomics analysis using UPLC-MS and ICR/FT-MS, an overview of metabolite permutations and biomarker candidates in C. pneumoniae infection of human cells could be defined. Effects of the pathogen on the lipid homeostasis are investigated in C. trachomatis infections. A modulated glycerophospholipid content is evidenced, including the identification of bacterial synthesized plasmalogens. The developed analytical and data analysis strategy is presented, as well as the biological results. Ein Überblick über die Metabolitveränderungen und Biomarkerkandidaten in C. pneumoniae Infektionen konnten in der UPLC-MS und ICR/FT-MS nutzenden nicht-zielgerichteten metabolischen Untersuchung erarbeitet werden. Effekte des Pathogens auf den Lipid Haushalt wurden in C. trachomatis Infektionen untersucht. Es wurde gezeigt, dass Glycerophospholipide in der Infektion stark verändert sind. Plasmalogene bakteriellen Ursprungs konnten identifiziert werden. Das entwickelte analytische Konzept, die Strategie der Datenauswertung und die biologischen Ergebnisse werden dargestellt.

Capillary electrophoretic methods were developed for the determination of saturated and unsaturated heterocyclic and aliphatic amines in metalworking and atmospheric aerosol. The target substances are derivatives of (i) benzotriazoles and benzothiazoles, (ii) hexahydrotriazines and oxasolidines, (iii) aminoalcohols and (iv) volatile and biogenic alkylamines. The overall analytical process includes the sampling of the vapour and condensed phase parallel, an easy extraction and separation of the analytes form mixtures. To adopt capillary electrophoresis to analytic of small bases in aerosol (i) mobility scale transformation was used for improving the precision (ii) the methods were MS compatible for improving the selectivity and (iii) a novel at-line preconcentration was developed to increase the sensitivity of the methods. Die vorliegende Arbeit beschreibt die Entwicklung von Methoden zur Analyse von gesättigten und ungesättigten heterozyklischen und aliphatischen Aminen in Kühlschmierstoffen und atmosphärischen Aerosolen durch Kapillarelektrophorese (CE). Die Zielsubstanzen waren hierbei Derivate von (i) Benzotriazolen und Benzothiazolen, (ii) Hexahydrotriazinen und Oxazolidinen, (iii) Aminoalkoholen und (iv) flüchtigen und biogenen Alkylaminen. Der analytische Prozeß beinhaltet die parallele Probennahme aus der Gas- und der kondensierten Phase, eine einfache Aufreinigung sowie die Trennung der Analyten aus Mischungen. Zur Anpassung an die Analytik niedermolekularer Basen in Aerosolen wurden (i) die Mobilitätstransformation zur Erzielung einer höheren Methodenpräzision verwendet, (ii) mit der Massenspektrometrie kompatible Methoden entwickelt und (iii) eine neuartige Methode der at-line Probenaufkonzentrierung zur Erzielung höherer Sensitivitäten eingesetzt.

In dieser Arbeit sollte die räumliche Verteilung von Alkaloiden als Sekundärmetabolite in der Tabakpflanze "Nicotiana tabacum" untersucht werden. Diese Pflanze diente im Rahmen der Untersuchungen als Modellsystem zur Aufklärung grundlegender Fragen bezüglich der Regulation der Alkaloidsynthese und deren Verteilung. Die Experimente sollten zum Verständnis der Wechselwirkung zwischen variierendem Resourcenangebot und der Bildung von Sekundärmetabolite sowie deren Verteilung auf Zellebene beitragen. Neben der damit verbundenen Untersuchung der Verknüpfung von Primär- und Sekundärstoffwechsel, sollte der Pflanze noch ein zusätzlicher Stressfaktor in Form von mechanischer Verwundung zugefügt werden. Unter Berücksichtigung dieser Bedingungen wurden die Experimente mit Hilfe der Einzelzellanalytik durchgeführt. Als instrumentelle analytische Methode wurde dabei die Kapillarelektrophorese/Massenspektrometrie verwendet. Die Elektronenmikroskopie diente zur strukturellen Veranschaulichung der Untersuchungsobjekte. In this study, the spatial distribution of alkaloids as secondary metabolites of the tobacco plant “Nicotiana tabacum” was investigated. In line with the investigation, the plant exemplified model system, clarifying basic questions concerning the regulation of alkaloid synthesis and the distribution of alkaloids. The experiments should contribute to the understanding of the interaction between the variation of resource supply and the formation of secondary metabolites, as well as their distribution on the cellular level. In the context of the investigation of the relationship between primary and secondary metabolism, the plant received another stress factor namely mechanical wounding. In view of these requirements the experiments were carried out with single cell analysis. Capillary electrophoresis in connection with UV- detection or mass spectrometry was utilized as instrumental analytical method. Electron microscopy enabled structural visualisation of the research probes.

N-Acyl-Homoserinlactone (AHL, HSL) spielen eine zentrale Rolle als Semiochemikalien bei der zelldichteabhängigen Kommunikation von Bakterien (Quorum Sensing, QS). Ungeachtet der weitreichenden Bedeutung des QS im medizinischen, landwirtschaftlich-ökonomischen und ökologischen Bereich erschien die Bandbreite der verfügbaren Nachweismethoden für HSL eingeschränkt. In der vorliegenden Arbeit werden unabhängige Strategien zur Analyse von HSL durch massenspektrometriegekoppelte Kapillartrenntechniken (micellare elektrokinetische Chromatographie, miniaturisierte Flüssigkeitschromatographie, Kapillarzonenelektrophorese, Gaschromatographie) in Verbindung mit spezifischen Aufkonzentrierungs- und Extraktionstechniken sowie durch ultrahochauflösende Fourier-Transform-Ionencyclotron-Resonanz-Massenspektrometrie vorgestellt. Durch Realprobenmessungen wird die Eignung der entwickelten Methoden für unterschiedliche Fragestellungen demonstriert. N-acylhomoserine lactones play a central role as semiochemicals in cell-density dependent communication phenomena in bacteria (quorum sensing, QS). Despite the high relevance of QS processes in medical, agricultural, economic and ecological contexts, the bandwidth of available methods for HSL detection and quantification was limited. In this work, independent strategies for the analysis of HSL by mass spectrometry-coupled capillary separation techniques (micellar electrokinetic chromatography, miniaturized liquid chromatography, capillary zone electrophoresis, gas chromatography) in connection with specific preconcentration and extraction methods and by ultrahigh resolution Fourier transform ion cyclotron resonance mass spectrometry are presented. The applicability of the methods for specific questions is demonstrated by real sample measurements.
