Electrochemically deposited sensors and DNA biosensors have been widely used in the food industry, and various other fields due to their high sensitivity, fast response, easy operation, no pollution, and low price. A rare earth electrochemically deposited gadolinium ferricyanide thin film modified glassy carbon electrode, and the investigation of electrocatalytic oxidation of guanine by GaHCF/GCE is reported. We deposited electrochemically a rare earth terbium ferricyanide thin film modified glassy carbon electrode. OH was investigated through various characteristics such as capacitance–voltage, Differential Pulse Voltammetric and many more by the comparison of catalytic oxidation signal of guanine with ·OH in Fenton reaction.

Integrating the merits of long lifespan and excellent energy as well as power densities, potassium-ion hybrid capacitors (PIHCs) exhibit great prospects for future energy storage devices. To boost comprehensive performance of PIHCs, heteroatom-doping and morphology-tuning as two comprehensive strategies have been devoted to designing uniquely structural carbon-based materials with favorable advantages. An ideal strategy for simultaneous atomic doping and structural regulation is expected to be developed. Herein, we propose a novel “Killing Two Birds with One Stone” strategy to prepare a tri-elements doped hollow carbon sphere (TED-HCS) as PIHCs anodes, that is, a single template of spherical CoP particles is rationally adopted, which not only provides both a P source for heteroatom-doping but also acts as a self-sacrificial template for hollow-structure engineering. The multifunctional TED-HCS presents a high capacity of 473.0 mAh g−1 and excellent rate performance of 212.5 mAh g−1 at 5.0 A g−1. Remarkably, the as-assembled PIHCs show outstanding energy/power density (40.4 Wh kg−1/10500 W kg−1) and remain high-capacity retention of 89.15% even cycling 12,000 times. The “Killing Two Birds with One Stone” strategy offers new insight into the search for the preparation of carbon-based materials with multi-elements doping and specific morphology structure.

The Sanchakou Cu deposit is located in the eastern section of the Dananhu magmatic arc in the Eastern Tianshan orogenic belt, northwest China. Sanchakou is hosted by quartz diorite and granodiorite intrusions. Chalcopyrite and bornite are the dominant ore minerals and occur as disseminations, patches, veins, and veinlets. Secondary ion mass spectrometry (SIMS) U-Pb dating of zircons shows that the ore-bearing intrusions were emplaced at ca. 435–432 Ma, recording the early subduction of the Paleo-Tianshan oceanic plate. The enrichment in large ion lithophile elements (LILEs), depletion in high field strength elements (HFSEs), and moderate Mg# values, together with mantle-like bulk Sr-Nd and zircon Hf-O isotope signatures (δ18O = 4.0–5.3‰), suggest that they were generated from partial melting of metasomatized mantle materials by subducted slab fluids. In situ S and whole-rock Pb isotope results imply that the Sanchakou diorite magmas provided ore-forming components (S and metals), with additional minor metals (e.g., Cu and Pb) sourced from crustal components beneath the Dananhu arc. The redox state of diorite magmas ranges from initial high fO2 (>FMQ + 2, where FMQ is the fayalite-magnetite-quartz buffer) to relatively low fO2 (7 wt %), normal S (8–393 ppm), and systematically low Cl (38–1,100 ppm) contents. A constant and favorable elevated magma oxidation state appears critical for generating an economic porphyry Cu deposit. Additionally, Cl melt concentrations may be a key factor that controlled metal fertility of the deposits in the Eastern Tianshan, although the mineralization potential may also relate to depth of emplacement of the hydrothermal system. The anomalous presence of stellerite with chalcopyrite in late-stage veins indicates that original porphyry-style mineralization at Sanchakou underwent deformation-related modification after its formation.

Homocysteine (Hcy) is a sulfur-containing amino acid derived from the essential amino acid methionine (Met). Circulating levels of Hcy in animals can be increased by feeding on Met-enriched diets, which is generally considered harmful. Spiders are one of the largest groups of obligate carnivores and feed on animals high in protein and Met. We analyzed the Hcy metabolism pathways in 18 species of three taxa (Mammalia, Insecta, and Arachnida) and found that the betaine-dependent remethylation pathway (BRP) was present in all carnivorous arachnid species and mammals but absent in insects and red spider mites. We then studied the Hcy metabolism pathway in Pardosa pseudoannulata. In P. pseudoannulata, Hcy is metabolized through the transsulfuration pathway, BRP, and S-methylmethionine-dependent remethylation pathway. Because of a prior duplication event of the betaine homocysteine S-methyltransferase (BHMT) gene in the BRP, BHMTa and BHMTb are present in tandem in the genome of P. pseudoannulata. The high expression levels of BHMTa and its high abundance in detoxification tissues indicate that it plays an important role in the BRP; the ability of BHMTa and BHMTb to remethylate Hcy using betaine as substrate was similar. Compared with other Hcy metabolic enzyme genes, BHMT responded quickly to the application of Hcy or betaine. Macrophage infiltration is a hallmark pathological change observed in early stage myocardial ischemia/reperfusion (MI/R) injury and one of the main causes of myocardial damage. Here, we investigated the effects of p-Coumaric acid (p-CA) on macrophage polarization following MI/R injury and its mechanisms. In vitro, p-CA decreases the expression of LPS/IFN-γ-induced M1 macrophage markers (TNF-α, IL-6, iNOS and CCL2) and increases IL-4-induced M2 macrophage markers (IL-10, CD206, Arg1 and Mrc) in mouse bone marrow-derived macrophages (BMDMs). Additionally, p-CA elevated indoleamine 2, 3-dioxygenase (IDO) protein expression levels, M2 macrophage polarization and M2 macrophage markers through IL-4. In contrast, repression of IDO attenuated p-CA functions regulating BMDMs through IL-4. In vivo, IDO expression was downregulated in mouse hearts subjected to MI/R injury. Treatment of p-CA increased IDO expression in the hearts of MI/R mice. Functionally, p-CA decreases M1 macrophage markers, the number of M1 macrophages and inflammation around heart tissue following MI/R injury. Importantly, p-CA reduces cardiomyocyte apoptosis caused by MI/R. Altogether, our study identified that p-CA modulates macrophage polarization by promoting IDO expression and that p-CA attenuates macrophage-mediated inflammation following MI/R by promoting M2 macrophage polarization through IDO.

In the present study, via using the mixed ligand synthesis approach, two coordination polymers (CPs) based on Cu(II) and Co(II) ions as nodes, namely, [Cu3(L1)2(1,2,4-BTC)2(H2O)4]n (1) and [Co3(L2)(1,2,4-BTC)2(H2O)4]n (2) (L1 is 3,6-bis(imidazol-1-yl)pyridazine, L2 is 3,6-bis(benzimidazol-1-yl)pyridazine and 1,2,4-H3BTC is benzene-1,2,4-tricarboxylic acid), were synthesized by reaction of the 1,2,4-H3BTC ligand and relevant metal salts with the aid of various nitrogen-donor co-ligands. Their application values on the cerebral ischemia were evaluated and the specific mechanism was studied simultaneously. At first, the enzyme linked immunosorbent assay (ELISA) was carried out to assess the inflammatory cytokines content released into cerebrospinal fluid. Besides, the neuro-apoptosis induced by cerebral ischemia was tested through the Annexin V-FITC Apoptosis Detection Kit.

Aspergillus flavus is saprophytic soil fungus that contaminates seed crops with the carcinogenic secondary metabolite aflatoxin, posing a significant threat to humans and animals. Ferrous sulfate is a common iron supplement that is used to the treatment of iron-deficiency anemia. Here, we identified an unexpected inhibitory role of ferrous sulfate on A. flavus. With specific fluorescent dyes, we detected several conidial ferroptosis hallmarks in conidia under the treatment of 1 mM Fe2+, including nonapoptosis necrosis, iron-dependent, lipid peroxide accumulation, and ROS burst. However, unlike traditional ferroptosis in mammals, Fe2+ triggered conidial ferroptosis in A. flavus was regulated by NADPH oxidase (NOXs) activation instead of Fenton reaction. Transcriptomic and some other bioinformatics analyses showed that NoxA in A. flavus might be a potential target of Fe2+, and thus led to the occurrence of conidial ferroptosis. Furthermore, noxA deletion mutant was constructed, and both ROS generation and conidial ferroptosis in ΔnoxA was reduced when exposed to Fe2+. Taken together, our study revealed an exogenous Fe2+-triggered conidial ferroptosis pathway mediated by NoxA of A. flavus, which greatly contributes to the development of an alternative strategy to control this pathogen.

In this letter, we achieved significantly enhanced avalanche ruggedness and surge current capability in GaN junction barrier Schottky (JBS) diode for highly reliable power operation. Based on the selective Mg-ion implantation technology, the GaN JBS diode obtains superior electrostatic performances, including 830-V breakdown voltage, 150-mΩ specific on -state resistance, and 0.5-V turning on voltage. Meanwhile, zero reverse recovery behaviors are observed even under extreme switching conditions of 600 V/10 A. During the reliability evaluation in the inductive load circuits, crucial avalanche capability with avalanche breakdown voltage over 965 V, avalanche energy up to 57.8 mJ, and more than 10 000 repetitive avalanche breakdown events are demonstrated. Together with the surge current tolerance up to 65 A and surge energy of 6.0 J, a large safe-operation-area under both forward and reverse inductive spikes is realized for the GaN-based rectifier.

A convenient and efficient strategy for the synthesis of dibenzoxazepinamines and dibenzothiazepinamines has been developed. This three-component approach started from 2-nitrobenzaldehydes, 2-aminophenols, and methoxyammonium chlorides under metal-free conditions. The protocol has the advantages of readily available starting materials, simple and facile conditions, gram-scale synthesis, and broad substrate scope, providing an efficient and practical strategy for the preparation of potential drug-active dibenzoxazepinamines and dibenzothiazepinamines in one pot.

As a novel type of theranostic radioactive agents, 177Lu-labeled nanomaterials conjugated to macromolecules have been described. The study aimed to fabricate PAMAM-G4-(177Lu-dendrimer)-bombesin-folate in the dendrimeric cavity, assess the radiopharmaceutical ability for specifically targeted radiotherapy and simultaneously detects gastrin-releasing peptide receptors (GRPR) and folate receptors (FRs) overexpressed in lung carcinoma cells, respectively. In an aqueous-basic media, p-SCN-benzyl-DOTA was conjugated to the dendrimer. This dendrimer was formed by activating the carboxylic acid groups of DOTA-folic acid and bombesin with HATU and conjugating them to develop the dendrimer. As part of this process, the conjugate was combined with 1% HAuCl4, added NaBH4 and filtered by ultrafiltration. Infrared, UV-Vis, TEM analysis, dynamic light scattering (DLS), and fluorescence spectroscopy were employed to observe the composition of the fabricated sample. Radio-labeled 177LuCl3 was used to label the conjugate, which was then evaluated using the radio-HPLC method. Findings demonstrated dendrimeric functionalization with remarkable radiochemical composition purity up to >96%. Because of fluorescence studies, it was determined that the occurrence of AuNMs in the dendrimeric cavities gives beneficial photo-physical characteristics to the radiopharmaceutical for bio-imaging. HEL-299 lung cancer cells exhibited a selective absorption of the drug (%). It might be helpful as nuclear and optical imaging agents for lung cancers that overexpress FRs and GRPR and as a specific target for radiation therapy if combined with folate-bombesin.

Hundreds of C60 derivatives stand out as electrontransporting materials(ETMs), for example, in perovskite solar cells(PSCs), due to their properties on electron extraction or defect passivation. However, it still lacks of guidelines to update C60-based ETMs with excellent photoelectric properties. In this work, crystallographic data of eight C60-based ETMs, including pristine C60 and the well-known PCBM as well as six newly synthesized fullerenes, are analyzed to establish the connections between derivatized structures and photoelectric properties for the typical carbon cluster of C60. In terms of packing centroid-centroid distance between neighboring carbon cages, the crystallographic data are useful for probing photoelectric properties, such as electrochemical properties, electron mobility and photovoltaic performances, and therefore facilitate to design novel C60-based ETMs for PSCs with high performances.

Suppressing the dissolution and accelerating conversion kinetics of intermediate polysulfides are central to the practical usage of lithium−sulfur (Li–S) battery in a wide temperature range. Herein, an atomic engineering strategy is innovatively proposed to constructing a sulfur host consisting of chemical-anchoring zinc atomic cluster within double layer N-doped carbon matrix (ZnAC CM@CL) for addressing concerns. The as-prepared ZnAC CM@CL@S can deliver capacity as high as 1451 mAh g−1 at 0.2 C with electrolyte/sulfur ratio (E/S ratio) of 5.5 μL mg−1 and maintain 749 mAh g−1 after 1200 cycles (fading rate of 0.021% per cycle) at 5 C. Remarkably, the reversible capacity holds 627 mAh g−1 at 0.2 C at the −25 ℃. By a further combination analysis of electron holography (EH) and geometry phase analysis (GPA), the outstanding performance is revealed to be mainly traced to synergistic effect of the polarity N-doped multi-layer carbon matrix and internal periodical charge field induced by atomic strain, greatly enhancing the capability towards on the immobilization and conversion of intermediate polysulfides. More importantly, such an interplay also renders the strongly coupled cathode achieve a high area specific capacity of 8.71 mAh cm−2 even the area sulfur loading obtain to 7.34 mg cm−2. This atomic-level engineering strategy might shed light on the novel insights on the design of high performance for sulfur hosts.

In recent years, synergistic chemo-photothermal therapy has revealed promising potential in treatments against various kinds of cancer. However, the development of superb photothermal agents with high drug loading capacity is still highly required. In this work, a hollow mesoporous polyaniline nanoparticle (HPANI NP) has been developed for encapsulating chemotherapeutic drug doxorubicin (DOX) with an remarkable drug loading content as high as 37.5%. Additional PEG modification endowed the drug-loaded HPANI NPs with improved water-dispersibility and bioavailability. Such PEG-HPANI-DOX NPs exhibited strong NIR absorbance and robust photothermal conversion capacity, exhibiting highly efficient synergistic cancer treatment. More interestingly, the responsively released DOX molecules could emit strong red fluorescence, which could be employed to monitor the cellular endocytosis and drug release profile of PEG-HPANI-DOX NPs. Finally, the as-fabricated NPs showed good biocompatibility and low toxicity, serving as a promising nanoagent for highly efficient drug delivery and cancer combination therapy.

Biogenic amines (BAs) are potential amine hazards that are detected in fermented foods and alcoholic beverages. Excessive intake of BAs may lead to allergic symptoms such as difficulty in breathing, nausea, and vomiting. Degradation of BAs by multicopper oxidase (MCO) is a promising method as it has little effect on the fermentation process, food nutrition, and flavor. However, the application of MCO in food industry was restricted due to its poor catalytic properties and low productivity. In this work, food-grade expression of the Bacillus amyloliquefaciens MCO (MCOB) and its three mutants were successfully constructed in Lactococcus lactis NZ3900. The expression level of MCOB in L. lactis NZ3900 was dramatically enhanced by optimizing the cultivation conditions, and the highest expression level reached 4488.1 U/L. This was the highest expression level of food-graded MCO reported so far, to our knowledge. Interestingly, the optimal reaction pH of MCOB expressed in L. lactis NZ3900 switched to 4.5, it would be more suitable for degrading BAs in food as the pH value of most fermented foods was found to be 4.5. Moreover, MCOB expressed in L. lactis NZ3900 was quite stable (with more than 80% residual activity) in the pH range of 4.0–5.5, the catalytic rate constant (kcat) and specific activity of MCOBLS were all dramatically increased compared with that of MCOB expressed in Escherichia coli. Using histamine as the substrate, the degradation of BAs within 24 h by MCOB expressed in L. lactis NZ3900 was 69.7% higher than that expressed in E. coli. The results demonstrated the potential applications of MCOB in food industry for reduction of biogenic amines.

Ten types of Tf2O/TTBP-mediated amide transformation reactions were investigated. The results showed that compared with pyridine derivatives 2,6-di-tert-butyl-4-methylpyridine (DTBMP) and 2-fluoropyridine (2-F-Pyr.), TTBP can serve as an alternative amide activation system for the direct transformation of both secondary and tertiary amides. For most surveyed examples, higher or comparable yields were generally obtained. In addition, Tf2O/TTBP combination was used to promote the condensation reactions of 2-(tert-butyldimethylsilyloxy)furan (TBSOF) with both tertiary and secondary amides, the one-pot reductive Bischler-Napieralski-type reaction of tertiary lactams, and Movassaghi and Hill's modern version of the Bischler-Napieralski reaction. The value of the Tf2O/TTBP-based methodology was further demonstrated by the concise and high-yielding syntheses of several natural products.

Schizonepeta tenuifolia Briq. (ST) has been used as an aromatic exterior-releasing medicine in clinical practice for thousands of years in China. Previous researches have revealed both volatile oil (STVO) and aqueous extract (STAE) from ST showed significant pharmacological activities, such as anti-virus, anti-inflammation, anti-oxidation, and immunoregulation. However, the influence between each other was still unknown. The purpose of this study was to compare the pharmacokinetic profiles of three main flavonoids (luteoloside, apigetrin, and hesperidin) in STAE to illustrate the influence of STVO. A liquid chromatography-tandem mass spectrometry (HPLC-MS) method was established to quantitatively analyze the three absorbed ingredients in the plasma of healthy rats. Biological samples were analyzed on an Agilent Eclipse Plus C18 column (3.0 mm × 150 mm, 3.5 μm) with gradient mobile phase (containing 0.2% formic acid and acetonitrile) at a flow rate of 0.8 mL/min. All the analytes and quercitrin (IS) were investigated with an electrospray ionization source (ESI) using multiple-reaction monitoring (MRM) in negative ionization mode. In addition, this quantitative method showed good linearities (r ≥ 0.9995) and the lower limits of quantification were 0.590–1.19 ng/mL. The intra- and inter-day precisions ranged 3.47–10.45% and 4.29–11.28% for the three analytes. The mean extraction recoveries were in the range of 77.41–109.79% and the average matrix effects were within 83.41–112.67%. The validated method has been fully applied to compare the pharmacokinetic parameters of the three flavonoid glycosides in rat plasma after oral administration of STAE and STAE+STVO. In comparison of luteoloside, apigetrin, and hesperidin in STAE group, it was found that different degree of increasing existed for the time to reach the maximum concentration (T max), elimination half-life time (T 1/2), the area under the concentration curves (AUC0→t and AUC0→∞) and the maximum concentrations (C max) in STAE+STVO group. As can be seen from above results, STVO could improve the absorption and bioavailability of the three analytes. These findings would provide some active and strong basis of safe clinical application for ST and further exploitation for STVO from the perspective of drug–drug interaction.

Prior to Ginkgo leaf harvest, we implemented four exogenous applications of salicylic acid (SA: 50, 80, and 100 mg·L−1) or methyl jasmonate (MJ: 20, 40, and 80 mg·L−1) as leaf spray to investigate their effect on flavonoids content and related enzyme activity. Compared with the control, a significant increase of 12% and 26% in flavonoid content was observed 7 days after the first application of 100 mg·L−1 SA and 20 mg·L−1 MJ, respectively; both were determined to be the best treatments. Operationally, administering either elicitor (SA or MJ) 1 week prior to leaf harvest is deemed to be the most practical and economic option. Additionally, enzymes activity involved in flavonoid metabolism (phenylalanine ammonia-lyase, cinnamate 4-hydroxylase, and 4-coumarate:coenzyme A ligase) and antioxidants (peroxidase, superoxide dismutase, and catalase) significantly increased in most SA and MJ treatments. The results strongly support Ginkgo leaf response to SA- or MJ-induced stress through increased nonenzymatic antioxidant compounds (flavonoids) and activity of antioxidant enzymes.

Dysregulation of deubiquitination has been reported to contribute to carcinogenesis. However, the function and mechanism of deubiquitinating enzyme 26S proteasome non‐ATPase regulatory subunit 14 (PSMD14) in the progression of ovarian cancer (OV), the deadliest gynecological cancer, still remains to be characterized. The present study demonstrated that PSMD14 was overexpressed in OV tissues and its higher levels correlated with a higher International Federation of Gynecology and Obstetrics (FIGO) stage in OV patients. A high level of PSMD14 expression was related to poor survival in OV patients. Knockdown and overexpression experiments elucidated that PSMD14 stimulated OV cell proliferation, invasion, and migration in vitro. Repression of PSMD14 suppressed OV tumor growth in vivo. PSMD14 inhibitor O‐phenanthroline (OPA) effectively attenuated malignant behaviors of OV cells in vitro and OV tumor growth in vivo. Mechanistically, we uncovered that PSMD14 was involved in post‐translational regulation of pyruvate kinase M2 isoform (PKM2). PSMD14 decreased K63‐linked ubiquitination on PKM2, downregulated the ratio of PKM2 tetramers to dimers and monomers, and subsequently diminished pyruvate kinase activity and induced nuclear translocation of PKM2, contributing to aerobic glycolysis in OV cells. Collectively, our findings highlight the potential roles of PSMD14 as a biomarker and therapeutic candidate for OV., PSMD14 decreases K63‐linked ubiquitination on PKM2 and downregulates the ratio of PKM2 tetramers to dimers and monomers. Subsequently, this diminishes pyruvate kinase activity and induces nuclear translocation of PKM2 as well as transcription of downstream genes, contributing to aerobic glycolysis in ovarian cancer cells and progression of ovarian cancer.

Main-group (s- and p-block) metals are generally regarded as catalytically inactive due to the delocalized s/p-band. Herein, we successfully synthesized a p-block antimony single-atom catalyst (Sb SAC) with the Sb-N4 configuration for efficient catalysis of the oxygen reduction reaction (ORR). The obtained Sb SAC exhibits superior ORR activity with a half-wave potential of 0.86 V and excellent stability, which outperforms most transition-metal (TM, d-block) based SACs and commercial Pt/C. In addition, it presents an excellent power density of 184.6 mW cm-2 and a high specific capacity (803.5 mAh g-1 ) in Zn-air battery. Both experiment and theoretical calculation manifest that the active catalytic sites are positively charged Sb-N4 single-metal sites, which have closed d shells. Density of states (DOS) results unveil the p orbital of the atomically dispersed Sb cation in Sb SAC can easily interact with O2 -p orbital to form hybrid states, facilitating the charge transfer and generating appropriate adsorption strength for oxygen intermediates, lowering the energy barrier and modulating the rate-determining step. This work sheds light on the atomic-level preparing p-block Sb metal catalyst for highly active ORR, and further provides valuable guidelines for the rational design of other main-group-metal SACs.

Rational composite design is highly important for the development of high-performance composite polymer electrolytes (CPEs) for solid-state lithium (Li) metal batteries. In this work, Li closo-borohydride, Li2B12H12, is introduced to poly(vinylidene fluoride)-Li-bis-(trifluoromethanesulfonyl) imide (PVDF-LiTFSI) with a bound N-methyl pyrrolidone plasticizer to form a novel CPE. This CPE shows superb Li+ conduction properties, as evidenced by its conductivity of 1.43 × 10−4 S cm−1 and Li+ transference number of 0.34 at 25°C. Density functional theory calculations reveal that Li2B12H12, which features electron-deficient multicenter bonds, can facilitate the dissociation of LiTFSI and enhance the immobilization of TFSI to improve the Li+ conduction properties of the CPE. Moreover, the fabricated CPE exhibits excellent electrochemical, thermal, and mechanical stability. The addition of Li2B12H12 can help form a protective layer at the anode/electrolyte interface, thereby preventing unwanted reactions. The above benefits of the fabricated CPE contribute to the high compatibility of the electrode. Symmetric Li cells can be stably cycled at 0.2 mA cm−2 for over 1200 h, and Li∥LiFePO4 cells can deliver a reversible specific capacity of 140 mA h g−1 after 200 cycles at 1 C at 25°C with a capacity retention of 98%.

Objective: To develop a rapid ultra-performance liquid chromatographic (UHPLC)-UV method for vancomycin determination in human serum for therapeutic drug monitoring (TDM). Methods: Human serum samples were precipitated with 10% perchloric acid, and the supernatant after centrifugation was analyzed on an ACQUITY UHPLC BEH C18 column (2.1 × 50mm, 1.7 μm) via gradient elution with a flow rate at 0.3 mL/min. The mobile phase consisted of acetonitrile and 0.005M KH2PO4 buffer (containing 0.1% triethylamine, pH 3.4). The detection wavelength was set at 210 nm, and the column temperature was set at 40°C. The total runtime was 6.0 min per analysis. Results: After comprehensive validation, the method was applied to determine the concentration of vancomycin in human serum. The chromatographic peaks of vancomycin and internal standard were not interfered by endogenous matrices. The Retention Time (RT) of vancomycin was 1.91 min, while the internal standard was 1.58 min. The good linearity range of vancomycin concentration was 2.5-120 μg/mL (R2>0.999). The lower Limit of Quantitation (LLOQ) was 2.5 μg/mL. The precision at three Quality Control (QC) levels (including LLOQ) was restricted within 85-115%. The extraction recovery rate of QC samples (4.0, 20.0, and 60.0 μg/mL) were 101.16%, 97.70%, and 94.90%, respectively. Inter- and intra-day precision was less than 8% (RSD). Stability tests under different storage conditions were satisfactory. In patients, the concentration of vancomycin ranged from 7.30 to 89.12 μg/mL determined by the fully validated method. Conclusion: The rapid sample pre-treatment procedures and short analysis time made this UHPLC-UV method suitable for therapeutic drug monitoring (TDM) of vancomycin.

Two novel 3-D Ln(III)–Cu(I) heterometallic coordination polymers, [Ln2Cu2(L3)4(C2O4)2]·H2O (Ln = Sm, Eu; HL3 = 5-aminonicotinic acid), were hydrothermally produced by reacting lanthanide oxide and copper halide with 5-aminonicotinic acid and orotic acid. In the two isostructural complexes, {Ln6}-wheel clusters link each other via edge-sharing mode, constructing honeycomb-like layers which stack in –ABAB– mode. On the whole, the architectures can be described as sandwich framework constructed by {Ln6}-wheel layers pillared by interlamellar [CuOL32] triangle pillars. The electronic structure was theoretically calculated by the density functional theory, confirming the direct transition character. The steady and transient photoluminescent spectra were measured, from which the intensity parameters, radiative lifetime and quantum efficiency were calculated within the framework of the Judd–Ofelt theory. [Eu2Cu2(L3)4(C2O4)2]·H2O can be regarded as a potential candidate for LED applications.

Objective. To investigate the effects of Danggui Buxue Tang (DBT) on rats with pulmonary fibrosis (PF) and the underlying mechanism. Methods. Sixty specific pathogen-free (SPF) male Sprague-Dawley (SD) rats were randomly divided into 4 groups: control, PF, prednisone treatment, and DBT treatment. Intratracheal instillation of bleomycin (BLM) was performed to establish a PF rat model. DBT was administered to PF rats concurrently for 2 weeks. Lung samples were then collected for HE and Masson staining after pulmonary function testing, and semiquantitative analysis for the degree of alveolitis and fibrosis was performed using the Szapiel and Ashcroft score systems. Myeloperoxidase (MPO) activity, hydroxyproline (HYP), hyaluronic acid (HA), and inflammatory cytokine content were measured. Western blotting was performed to detect fibrotic marker and TLR4/NLRP3 signaling pathway changes. Results. Oral administration of DBT attenuated weight loss, survival rate, and pulmonary index. Lung histopathologic lesions were also reduced. DBT inhibited PF by decreasing the secretion of inflammatory cytokines and collagen deposition. Specifically, DBT reduced tumor necrosis factor-alpha (TNF-α), interleukin 1 beta (IL-1β), IL-6, HYP, alpha-smooth muscle actin (α-SMA), collagen I, and collagen III levels. Corollary experiments identified a potential mechanism involving suppression of TLR4/MyD88/NF-κB signaling pathway activation and the NLRP3/ASC/caspase-1 axis, the downstream regulatory pathway. Conclusion. DBT exhibited a potent effect on BLM-induced PF rats by inhibiting the TLR4/NLRP3 signaling pathway. Thus, DBT alleviates pulmonary inflammation to inhibit fibrotic pathology and should be considered as a candidate for the clinical treatment of PF.
