Redox tailoring enzymes play key roles in generating structural complexity and diversity in type II polyketides. In chartreusin biosynthesis, the early 13 C-labeling experiments and bioinformatic analysis suggest the unusual aglycone is originated from a tetracyclic anthracyclic polyketide. Here, we demonstrated that the carbon skeleton rearrangement from a linear anthracyclic polyketide to an angular pentacyclic biosynthetic intermediate requires two redox enzymes. The flavin-dependent monooxygenase ChaZ catalyses a Baeyer-Villiger oxidation on resomycin C to form a seven-membered lactone. Subsequently, a ketoreductase ChaE rearranges the carbon skeleton and affords the α-pyrone containing pentacyclic intermediate in an NADPH-dependent manner via tandem reactions including the reduction of the lactone carbonyl group, Aldol-type reaction, followed by a spontaneous γ-lactone ring formation, oxidation and aromatization. Our work reveals an unprecedented function of a ketoreductase that contributes to generate structural complexity of aromatic polyketide.

In this study, through employing the mixed‐ligand synthesis method, a novel coordination polymer (CP) containing Cu(II) has been synthesized in success with the reaction of 4‐bmbpd and Cu(NO3)2·3H2O in the presence of auxiliary polycarboxylate ligand 1,3,5‐H3BTC under hydrothermal conditions, and the chemical formula of this compound is [Cu(4‐bmbpd)(1,3,5‐HBTC)]·2H2O (1) (1,3,5‐H3BTC is 1,3,5‐benzenetricarboxylic acid and 4‐bmbpd is N,N′‐bis(4‐methylenepyridin‐4‐yl)‐1,4‐benzenedicarboxamide). The assessment of its prevention and predictive nursing activity on the hypothermia during laparoscopic cholecystectomy operation was conducted and the corresponding mechanism was studied simultaneously. The molecular docking simulation helps for identifying that the carboxylate group and the amide group form multiple H‐bonds with respect to the active sites within the docking pocket of the target protein.

As classical MRPs, the toxic effects of furosine, pyralline, and 5‐hydroxymethylfurfural (5‐HMF) in liver tissue are evaluated and the related mechanism is investigated here, and the protective effects of lactoferrin on liver injury caused by Maillard reaction products (MRPs) with furan ring are proved in vitro and in vivo. First, we detect the concentrations of furosine, pyralline, and 5‐HMF in several foods using ultrahigh‐performance liquid chromatography (UHPLC). Then, the effects of the three MRPs on liver cells (HL‐7702) viability, as well as liver tissue, are performed and evaluated. Furthermore, the regulations of three MRPs on necroptosis‐related pathway in liver cells are investigated. Additionally, the effects of lactoferrin in alleviating liver injury, as well as regulating necroptosis pathway, were evaluated. Results elucidate that lactoferrin protects liver injury caused by MRPs with furan ring structure through activating RIPK1/RIPK3/p‐MLKL necroptosis pathway and downstream inflammatory reaction., Lactoferrin protects liver injury caused by MRPs with furan ring structure through activating RIPK1/RIPK3/p‐MLKL necroptosis pathway and downstream inflammatory reaction.

Because the conventional methods for furosine analysis are time-consuming, a modified method is presented to improve analysis efficiency. Microwave-assisted HCl hydrolysis was performed at 140, 150, and 160°C for 10 to 200 min, with 6, 8, or 9 M HCl. The hydrolysate purification process was carried out using only paper and membrane filtration. An ultra-performance liquid chromatography (UPLC) system was used to achieve rapid analysis of furosine. The results showed that microwave-assisted HCl hydrolysis at 8 M and 160°C led to a stable furosine yield and took only 40 min. The UPLC analysis was completed in 8 min. The modified method was validated and obtained limit of detection at 3 µg/L, limit of quantitation of 10 µg/L, linearity range of 0.2 to 5.0 mg/L, 80.5 to 94.2% recoveries from spiked samples, and coefficients of variation of 2.2 to 6.8%. The modified method is rapid and reliable for the determination of furosine in milk.

The effective chemotherapy of cancer is usually hindered by the unsatisfied cell internalization of the drug delivery systems (DDS) as well as drug resistance of cancer cells. In order to solve these dilemmas in one design, red blood cell membrane (RBM)-coated silica nanoparticles (RS) were fabricated to codeliver doxorubicin (Dox) and indocyanine green (ICG) to effectively treat the model lung cancer using photothermal-assisted chemotherapy. Our results demonstrated that the RS/I-D was the nanoparticle at around 100 nm with superior stability and biocompatibility. Especially, the photothermal effects of ICG were well preserved and could be applied to accelerate the drug release from the DDS. More importantly, the RBM modification can mediate enhanced cell internalization of drugs as compared to their free forms, which finally resulted in enhanced anticancer efficacy in Dox-resistant A549 cells (A549/Dox) both in vitro and in vivo with enhanced cell apoptosis and cell arrest.

Copper-based materials are efficient electrocatalysts for the conversion of CO2 to C2+ products, and most these materials are reconstructed in situ to regenerate active species. It is a challenge to precisely design precatalysts to obtain active sites for the CO2 reduction reaction (CO2 RR). Herein, we develop a strategy based on local sulfur doping of a Cu-based metal-organic framework precatalyst, in which the stable Cu-S motif is dispersed in the framework of HKUST-1 (S-HKUST-1). The precatalyst exhibits a high ethylene selectivity in an H-type cell with a maximum faradaic efficiency (FE) of 60.0 %, and delivers a current density of 400 mA cm-2 with an ethylene FE up to 57.2 % in a flow cell. Operando X-ray absorption results demonstrate that Cuδ+ species stabilized by the Cu-S motif exist in S-HKUST-1 during CO2 RR. Density functional theory calculations indicate the partially oxidized Cuδ+ at the Cu/Cux Sy interface is favorable for coupling of the *CO intermediate due to the modest distance between coupling sites and optimized adsorption energy.

The mangrove-derived endophytic fungus Peniophora incarnata Z4 produced seven new xanthone derivatives, including four new tetrahydroxanthones (1-4), one new chromone (5), one new xanthone (6), and one new xanthone dimer (7), together with one known compound, globosuxanthone B (8). Their structures were determined by an extensive analysis of 1D and 2D NMR, HRESIMS, ECD, and single-crystal X-ray diffraction data. In cytotoxic activity assays, compound 2 showed cytotoxicity against three carcinoma cell lines with IC50 values less than 10 μM.

Cancer therapeutic strategies based on angiogenesis attract great attention from fundamental and clinical research. Blocking oxygen and nutrition supply to tumor cells could inhibit the growth of tumors based on occlusion of blood vessels in the tumor. Herein, we report a dual-responsive peptide-based nanoparticle, mimicking the laminin fibrillogenesis specifically and highly efficiently in tumor vessels, resulting in the blockage of tumor vessels and the growth inhibition of tumors. The laminin mimic peptide (LMMP) is designed with a fibrillation sequence, a pH-responsive sequence, and a targeting sequence. The LMMP in nanoformulations is delivered to blood vessels in the tumors, where the microenvironment (pH and microthrombus) enable LMMP to process laminin fibrillogenesis, constructing fibrous networks. The laminin-like fibrous networks capture red blood cells etc., forming occlusion specifically in the tumor blood vessels to inhibit the growth of the tumor.

Heterostructure engineering of electrode materials, which is expected to accelerate the ion/electron transport rates driven by a built-in internal electric field at the heterointerface, offers unprecedented promise in improving their cycling stability and rate performance. Herein, carbon nanotubes with Co9 S8 /ZnS heterostructures embedded in a N-doped carbon framework (Co9 S8 /ZnS@NC) have been rationally designed via an in-situ vapor chemical transformation strategy with the aid of thiophene, which not only acted as carbon source for the growth of carbon nanotubes but also as sulfur source for the sulfurization of metal Zn and Co. Density functional theory (DFT) calculation shows an about 3.24 eV electrostatic potential difference between ZnS and Co9 S8 , which results in a strong electrostatic field across the interface that makes electrons transfer from Co9 S8 to the ZnS side. As expected, a stable cycling performance with reversible capacity of 411.2 mAh g-1 at 1000 mA g-1 after 300 cycles, excellent rate capability (324 mAh g-1 at 2000 A g-1 ) and a high percentage of pseudocapacitance contribution (87.5% at 2.2 mv/s) for lithium-ion batteries (LIBs) are achieved. This work provides a possible strategy for designing multicomponent heterostructural materials for application in energy storage and conversion fields.

Sugars are abundant natural sources existing in biological systems, and bioactive saccharides have attracted much more attention in the field of biochemistry and biomaterials. For better understanding of the sugar-based biomaterials and biological sciences, aggregation-induced emission luminogens (AIE-gens) have been widely employed for detection, tracing, and imaging. This review covers the applications of AIE molecules on sugar-based biomaterials by three parts, polysaccharide, oligosaccharide, and monosaccharide, mainly focusing on saccharide detection, stimuli response materials preparation, bioimaging, and study of the AIE mechanism. These excellent works suggest the promising future of the sugar-based AIE bioconjugates, considering that the naturally designed and elaborately functionalized saccharides play discriminate roles in biological processes and AIE-tagged species may work as an indicator in each case. However, there are a lot of sugar-based biological species that have not been touched, such as mucopolysaccharides and glycoproteins on the cell surface and in the cell plasma. Based on these features, we enthusiastically look forward to more glorious developments in this bright research area.

In this work, an improved 4H-SiC insulated gate bipolar transistor (IGBT), or CTH-IGBT, with a trench p-polySi/p-SiC heterojunction on the backside of the device is proposed to reduce the turn-off energy loss (Eoff) and turn-off time (Toff). The electrical properties of the proposed and contrast structures are all simulated using the ATLAS simulation software to research the working mechanism of this improved structure. For the static performance, the specific ON-resistance (Ron,sp) and the figure of merit (FOM = VBR2/Ron,sp) are not influenced much as compared to the traditional structure at the same breakdown voltage (VBR) of 12 kV. However, with a prominent electron current path formed by the heterojunction region of CTH-IGBT, a very available conduction path to discharge the electrons during turn-off process is proved in this article. The simulation results demonstrate that compared with the traditional structure, the turn-off energy loss of the CTH-IGBT is reduced by 76.4%, while the turn-off time is reduced by 85.0%.

We developed a sensitive and selective isotope dilution ultra-high-performance liquid chromatography-tandem mass spectrometry (UPLC-MS/MS) method for the determination of sulbactam residue in raw bovine milk. Sulbactam and internal standard, sulbactam-d5, were extracted from raw bovine milk via liquid-liquid extraction and enriched with strong anion exchange solid-phase extraction cartridges and finally analyzed by using UPLC-MS/MS with multiple reaction monitoring mode. The method was validated according to European regulations. The calibration curve showed good linearity, with a correlation coefficient of 0.9998. Decision limit and detection capability of sulbactam were determined by matrix calibration curve and were 0.0445 and 0.0517 μg/L, respectively. The recoveries of sulbactam in fortified raw bovine milk ranged from 72.1 to 91.5%, with the intra- and interday relative standard deviations ranging from 3.0 to 18.9%. Furthermore, the developed method was applied to analyzing real raw bovine milk samples collected from dairy farms in Beijing, China. Sulbactam was not determined in all samples. The proposed method could ultimately serve as a methodological foundation for the determination of sulbactam in different types of raw milk and dairy products.

Resources in urine-containing wastewater are useful if recovered as nutrients. In this study, a three-chamber resource recovery microbial fuel cell (RRMFC) is proposed to treat synthetic urine-containing wastewater with various organic pollutants and recover N, P, and S nutrients. In the treatment, urea hydrolysis was increased by microbial and electrical processes. Ions migration driven by the self-generated electric field was used to recover nutrients from the wastewater. Over one cycle (∼3 days), 99% of urea, 97% of COD, 99% of histidine, 91% of creatinine, 99% of sodium acetate, 98% of SO42−, and 99% of PO43− were removed from the wastewater, and at the same time, 42% of total nitrogen, 37% of PO43−, 59% of SO42−, and 33% of total salts were recovered in the middle chamber. This technology is very attractive for sustainable resource recovery from urine-containing wastewater.

Epithelial ovarian cancer has the highest mortality rate of all malignant ovarian cancer types. Great progress has been made in the treatment of ovarian cancer in recent years. However, drug resistance has led to a low level of 5-year survival rate of epithelial ovarian cancer, and the molecular mechanism of which remains unknown. The aim of the present study was to identify the role of redox status in the cisplatin (CDDP) resistance of ovarian cancer. CDDP-resistant SK-OV3 (SK-OV3/cddp) cells were prepared and their reactive oxygen species and glutathione levels were investigated. The effects of hydrogen peroxide on the CDDP sensitivity of the SK-OV3/cddp cells and their expression levels of the redox-associated protein growth arrest and DNA damage 45a (GADD45α) were also investigated. In addition, the impact of GADD45α overexpression on cell viability was evaluated in vitro and in vivo, and the levels of Ser-139 phosphorylated H2A histone family member X (γ-H2AX), which is associated with DNA damage, were detected. The results suggested that redox status affected the drug resistance of the ovarian cancer cells by increasing the expression of GADD45α. The overexpression of GADD45α reversed the CDDP resistance of the SK-OV3/cddp cells and increased the level of γ-H2AX. In conclusion, GADD45α alleviated the CDDP resistance of SK-OV3/cddp cells via the induction of redox-mediated DNA damage.

Pharmacological intervention played an important role in the management of tic disorder. Large-scale prescription data for pediatric tic disorder patients in the real-world setting were scarce. The demographic and prescription data of tic disorder were extracted from the electronic medical records database of Beijing Children's Hospital from 2018 to 2020. The intervention choice for outpatient pediatric tic disorder patients was analyzed. A total of 20 417 patients were included, 28.1% (n = 5028) of them did not receive any pharmacological treatment. Over 70% were prescribed with anti-tic medication. For children less than 6 years of age, clonidine adhesive patches (CAPs) and traditional Chinese medicine (TCM) were the common choice. With the age growing, the use of antipsychotics was on the rise; 22% (n = 3389) were prescribed for at least two anti-tic medication, and the most common medication combination group was tiapride and TCM (33.7%), followed by CAP and TCM (22.1%). The clinical intervention choice for tic disorder is highly individualized. The pharmacological choice was influenced by severity, duration of symptom, age, the acceptance of parents and other factors.

A laboratory-scale study was conducted to investigate the effect of bioaugmentation (BA) and biostimulation (BS) on the remediation of oily sludge with high total petroleum hydrocarbon (TPH) content (269,000 mg/kg d.w. sludge). TPH concentration significantly decreased by 30.4% (P 0.05) in the BS treatment after 13-week incubation, and 17.0 and 9.1% of TPH was removed in the BA and control treatments (amended with sterile water only), respectively. Aliphatic and other fractions (i.e., saturated n-alkanes and cyclic saturated alkanes) were reduced in the BS treatment, whereas no decrease in aromatic hydrocarbons occurred in any treatment. Gas chromatography-mass spectrometry analysis of aliphatic fractions showed that low-chain-length alkanes (C8-C20) were the most biodegradable fractions. The BS treatment supported fungal proliferation, with Sordariomycetes and Eurotiomycetes as the dominant classes. BS increased fungal diversity and decreased fungal abundance, and changed bacterial community structure. The findings show the potential of using BS to treat oily sludge with high TPH content. The persulfate/Fe2+ system has been proposed for the chemical oxidation for soil remediation, however, the homogeneous iron catalyst was hard to reuse which limited the further application. Cobalt oxyhydroxide (CoOOH) existed as a mineral in nature, which was environmentally friendly. Thus, in this study, CoOOH was selected as an efficient heterogeneous catalyst for peroxymonosulfate (PMS) activation to remediate oil contaminated soil by chemical oxidized reaction. 88.3% of oil at the initial concentration of 78–99 mg/kg can be removed within 24 h under the conditions of 1.0 g/L CoOOH and 0.1 M PMS at room temperature. The residual oil content was approximately 11.5 mg/kg which was lower than the standard of petroleum hydrocarbons for residential land (30 mg/kg), published by the Canadian Council of Ministers of the Environment (CCME). Specifically, the PMS/CoOOH system had a relatively high apparent reaction rate constant (0.3078 h−1), which was approximately twice that of the PS/Fe2+ system (0.1601 h−1). Furthermore, multiple radicals and reactive oxygen species (ROS), such as SO4 −, O2 − and 1O2, were involved in the oil removal oxidation reaction. Moreover, 73% total organic carbon (TOC) had been removed after the reaction. The findings of this study suggested that the oil-contaminated soil and CoOOH could both be recycled after remediation using the PMS/CoOOH system. In summary, the results indicated that CoOOH is a promising heterogeneous catalyst, and the PMS/CoOOH system could be considered as a feasible alternative to the PS/Fe2+ system for the remediation of oil-contaminated soil.

Foods supplemented with lactoferrin are an increasing trend owing to its multiple functions. Therefore, a reliable analytical method for the nutritional assessment and quality control of lactoferrin containing foods must be established. A method for the determination of native lactoferrin in milk using a HiTrapTM Heparin HP column coupled with HPLC was developed and validated. The conditions for lactoferrin enrichment in milk using the HiTrapTM Heparin HP column were optimized. A linear range of 2 to 100 mg L-1 was obtained for lactoferrin standards, with a R2 value of 0.9989. The detection limit (LOD) and quantification limit (LOQ) of the lactoferrin standards were 2.00 and 6.67 mg L-1, respectively. Considering the enrichment factor involved in the pre-treatment procedures, LOD and LOQ for detection of milk sample were 0.57 and 1.90 mg L-1, respectively. In addition, milk samples spiked with lactoferrin at three concentration levels (2, 5, and 10 mg/L) showed overall mean recovery of 88.3%, 90.2% and 95.1%, respectively. The relative standard deviations of intra-day and inter-day precision were 1.3% ~ 4.8%, and 2.1% ~ 5.7%, respectively, indicating that the present method showed good performances. Furthermore, the successful quantification of lactoferrin in raw milk and processed milk demonstrated the usefulness and effectiveness of the proposed method.

We assessed the presence of tetracyclines, quinolones, lincomycin, and streptomycin in ultra-high temperature (UHT) milk samples (n = 148) and pasteurized milk samples (n = 50) from China using an enzyme linked immunosorbent assay-based visualization microarray chip technique. The detection rates of tetracyclines, quinolones, lincomycin, and streptomycin were 4.7%, 3.3%, 2.7%, and 15.5%, respectively, in UHT milk and 16.0%, 4.0%, 2.0%, and 14.0%, respectively, in pasteurized milk. The maximum levels of tetracyclines, quinolones, lincomycin, and streptomycin in the sample were 9.06 μg kg−1, 4.06 μg kg−1, 7.66 μg kg−1, and 8.92 μg kg−1, respectively, which are lower than the maximum residue levels established in China, the Codex Alimentarius Commission, and the European Union, indicating the milk production/marketing system might be adequately controlling the antibiotic residue risk. However, considering the limited sample numbers, and relative high detection rates in spring and summer which might pose a health risk on population, control measures still need to be strictly enforced to keep these residues at safe levels.

To understand genes involved in neutral lipid accumulation upon nitrogen deprivation (ND) in a novel isolate of Nannochloropsis sp. PJ12, we performed comparative transcriptomic and lipidomic analyses of cells under ND and NR (nitrogen replete) conditions. Transcriptomic profiling indicated that, while enzymes involved in TCA cycle in PJ12 under ND condition were upregulated compared to that under NR condition, those involved in Calvin cycle and glycolysis under ND condition were downregulated. Furthermore, we showed that enzymes involved in fatty acid synthesis and glycerolipid synthesis were downregulated but not β-oxidation. Lipidomic profiling indicated that, while the level of neutral lipids in ND cells was increased compared to that of NR cells, level of photosynthetic membrane-lipids DGDG and PG was decreased. Taken together, our analysis indicated that TAG accumulation is attributed to the modification of membrane lipids derived primarily from “prokaryotic” pathway and secondarily from “eukaryotic” pathway based on the 16:X or 18:X fatty acid at the sn2 position of the glycerol backbone. We propose that two-phase (NR-ND) growth is ideal for biomass and biofuel production because ND reduces cell growth rate due to the loss of photosynthetic membrane and decreased quantum yield.

Five isomorphic lanthanide metal-organic frameworks constructed from 5-(3-carboxy-phenyl)-pyridine-2-carboxylic acid (H2cppca) and oxalate (H2ox), {[Ln2(cppca)2(ox)(H2O)2]·2H2O}n [Ln = Eu (1), Gd (2), Nd (3), Pr (4),Tb (5)] have been successfully synthesized and characterized by single-crystal X ray diffraction, elemental analysis, IR spectroscopy, thermal analysis, and powder X-ray diffraction. In compounds 1–5, inorganic helical Ln-O chain and cppca2- anions are interconnected to form a one-dimension (1D) helical metal-organic chain, then 1D metal-organic chains are linked into a three-dimension achiral network with 1D channel of 6.31 × 10.13 A2, showing an unprecedented (4,5)-connected topology net with Schlafli symbol {44.62}{44.64.82}. Fluorescent experiment indicated that compounds 1 and 5 show sensitive luminescence quenching response to aniline, and the corresponding quenching constants (Ksv) are 2.81 × 104 M−1 and 3.14 × 104 M−1, respectively. The detection limits of aniline are 3.06 μM for 1 and 2.29 μM for 5. Moreover, compounds 1 and 5 are very stable and their reusability is relatively high and the fluorescence can be easily recovered after washing.

Considering the distinctions of cancer cell microenvironments, smart systems with stimulus-triggered features have become increasingly popular nowadays. These smart systems are regarded as efficient approaches for targeted cancer cell imaging and selective tumoricidal therapy. On account of the highly reductive micro-environment in tumor tissues, a redox-responsive fluorescent AIE bioprobe based on a disulfide bond is designed and synthesized. This AIE bioprobe possesses excellent water dispersibility and nano-size stability under physiological conditions. Under the reductive microenvironment of tumor cells, the water dispersibility of the AIE probe becomes worse owing to the cleavage of carboxyl groups. The dispersibility alteration further induces the aggregation of AIE nanoparticles and enhances intracellular retention, allowing for targeted imaging reinforcement and selective suppression of cancer cells.

A novel Cu(I)-cluster based coordination compound formulated as {(Cu2I2)2[Cu6(dmpymt)6]}n (Hdmpymt = 4,6-dimethylpyrimidine-2-thione) was synthesized under solvo-thermal condition and characterized by single-crystal X ray diffraction, elemental analysis, IR spectroscopy, thermal analysis, and powder X-ray diffraction. Compound 1 is an infinite 3D structure composed of Cu2I2 and Cu6S6 binary Cu(I) clusters with 66 topology, and displays high thermal stability and good solvent stability. Fluorescence experiments illustrate that compound 1 displays apparently red luminescence emission and selectively turn-off fluorescence response for CrO42− and Cr2O72− in water, showing potential luminescent probes for detecting Cr(VI) ions.

Myo-inositol and its metabolic derivatives such as pinitol, galactinol, and raffinose affect growth and development and are also involved in stress adaptation. Previous studies have identified myo-inositol transporters (INTs) as transporters of Na+ from root to shoot in the halophyte ice plant (Mesembryanthemum crystallinum). We found that the supply of myo-inositol could alleviate the dehydration effects of salt-stressed ice plant seedlings by decreasing the Na/K ratio in roots and increasing the Na/K ratio in shoots. Analyses of the uptake of exogenous myo-inositol revealed that ice plant seedlings contained intrinsic high-affinity transporters and inducible low-affinity uptake systems. The presence of Na+ facilitated both high- and low-affinity myo-inositol uptake. Six INT genes were identified from the ice plant transcriptome and named McINT1a, 1b, 2, 4a, 4b, and 4c, according to the classification of the Arabidopsis INT family. In seedlings treated with myo-inositol, salt, or myo-inositol plus salt, the expression patterns of all McINT members differed in shoot and root, which indicates organ-specific regulation of McINTs by salt and myo-inositol. The expression of McINT2, 4a, 4b, and 4c was induced by salt stress in shoot and root, but that of McINT1a and 1b was salt-induced only in shoot. The expression of pinitol biosynthesis gene IMT1 was induced by salt and myo-inositol, and their combination had a synergistic effect on the accumulation of pinitol. Supply of myo-inositol to salt-treated seedlings alleviated the detrimental effects by maintaining a low root Na/K ratio and providing precursors for the synthesis of compatible solute to maintain the osmotic balance.

Sparganii rhizoma (SL) has potential therapeutic effects on gastric cancer (GC), but its main active ingredients and possible anticancer mechanism are still unclear. In this study, we used HPLC-Q-TOF–MS/MS to comprehensively analyse the chemical components of the aqueous extract of SL. On this basis, a network pharmacology method incorporating target prediction, gene function annotation, and molecular docking was performed to analyse the identified compounds, thereby determining the main active ingredients and hub genes of SL in the treatment of GC. Finally, the mRNA and protein expression levels of the hub genes of GC patients were further analysed by the Oncomine, GEPIA, and HPA databases. A total of 41 compounds were identified from the aqueous extract of SL. Through network analysis, we identified seven main active ingredients and ten hub genes: acacetin, sanleng acid, ferulic acid, methyl 3,6-dihydroxy-2-[(2-hydroxyphenyl) ethynyl]benzoate, caffeic acid, adenine nucleoside, azelaic acid and PIK3R1, PIK3CA, SRC, MAPK1, AKT1, HSP90AA1, HRAS, STAT3, FYN, and RHOA. The results indicated that SL might play a role in GC treatment by controlling the PI3K-Akt and other signalling pathways to regulate biological processes such as proliferation, apoptosis, migration, and angiogenesis in tumour cells. In conclusion, this study used HPLC-Q-TOF–MS/MS combined with a network pharmacology approach to provide an essential reference for identifying the chemical components of SL and its mechanism of action in the treatment of GC.

Aim: To explore the mechanism of gastric carcinogenesis by mining potential hub genes and to search for promising small-molecular compounds for gastric cancer (GC). Materials & methods: The microarray datasets were downloaded from Gene Expression Omnibus database and the genes and compounds were analyzed by bioinformatics-related tools and software. Results: Six hub genes ( MKI67, PLK1, COL1A1, TPX2, COL1A2 and SPP1) related to the prognosis of GC were confirmed to be upregulated in GC and their high expression was correlated with poor overall survival rate in GC patients. In addition, eight candidate compounds with potential anti-GC activity were identified, among which resveratrol was closely correlated with six hub genes. Conclusion: Six hub genes identified in the present study may contribute to a more comprehensive understanding of the mechanism of gastric carcinogenesis and the predicted potential of resveratrol may provide valuable clues for the future development of targeted anti-GC inhibitors.

Background Flap Endonuclease 1(FEN1) has been considered as a new tumor marker in recent years and Jianpi Yangwei Decoction (JPYW) is a basic Traditional Chinese Medicine (TCM) for the treatment of gastric cancer. This study aimed to explore the role of FEN1-mediated DNA damage repair in the drug resistance of gastric cancer and the effect of JPYW on it by employing BGC823/5-Fu drug-resistant cell model. Methods The DNA repair efficiency of BGC823 and BGC823/5-Fu was compared intracellularly and extracellularly using an extrachromosomal assay system and the reconstituted base excision repair assay. By comparing gene and protein expression and identifying cell survival rates after knockdown or high expression of FEN1, the correlation between FEN1 high expression and 5-Fluorouracil (5-Fu) drug resistance was revealed. The effect of JPYW on DNA damage repair and FEN1 expression was observed by the degree of γ-H2AX phosphorylation in the cells, DNA repair efficiency and enzyme activity, et al. Results BGC823/5-Fu had a higher DNA repair efficiency than BGC823(P 32p labeled DNA substrates were decreased. Conclusion FEN1 was highly expressed in drug-resistance gastric cancer cells BGC823/5-Fu, which leading to BGC823 resistant to (5-Fu) by acting on DNA damage repair. JPYW inhibited DNA damage repair and reversed 5-Fu drug resistance by reducing FEN1 expression and inhibiting FEN1 functional activity.

Cesium (Cs) contained triple-cation and mixed halide perovskite (CsFAMA) is broadly employed as light absorption layers for efficient and stable perovskite solar cells (PSCs) fabrication with high reproducibility. On the other hand, thermal annealing is a universal post-treatment method for perovskite films preparation. Moreover, thermal management highly depends on perovskite materials. However, no specialized study has been reported on CsFAMA perovskite to date. Herein, we have systematically investigated the influence of thermal annealing and annealing time on CsFAMA films and their solar cells. We demonstrated that heating time of 45 or 60 min at 100 °C is desirable. More interestingly, we found that the unannealed CsFAMA films exhibit ultrahigh photoluminescence (PL) intensities, much stronger than that of annealed films. Note that PL intensities gradually weaken as a function of annealing time. In particular, the PL intensities of fresh films (after antisolvent dripping) are at least 200 times higher than that of 60 min annealed films. To our knowledge, it is the first time to report this PL behavior. We speculate that it is due to quantum confinement effect of perovskite crystal nuclei and "cage effect" of DMSO intermediates in the fresh films. To this point, the unannealed CsFAMA films may have great potential in PL emission applications.

Two new 2-carboxymethyl-3-hexyl-maleic anhydride derivatives, arthrianhydride A (1) and B (2), along with three known compounds 3-5, were isolated from the fermentation broth of a grasshopper-associated fungus Arthrinium sp. NF2410. The structures of new compounds 1 and 2 were determined based on the analysis of the HR-ESI-MS and NMR spectroscopic data. Furthermore, compounds 1 and 2 were evaluated on inhibitory activity against the enzyme SHP2 and both of them showed moderate inhibitory activity against SHP2.

A novel pillared-layer coordination polymer containing -Pb-O-Pb-Cu-Cl- inorganic sheets, [PbCuCl(3.5-pydc)]n (1) (3.5-H2pydc = pyridine-3,5-dicarboxylic acid), was obtained through the hydrothermal method. In compound 1, two symmetry-related Pb centers are bridged into a Pb2O2 dimer by two symmetrically carboxyl oxygen atoms from 3,5-pydc2− anions, each Pb2O2 dimer links four equivalent units into a 2D puckered Pb-O-Pb layer. Two symmetry-related Cu ions are bridged into a Cu2Cl2 dimer by two chlorine ions. Thus the -Pb-O-Pb- layer and Cu2Cl2 dimers are joined into a novel 2D puckered -Pb-O-Pb-Cu-Cl- inorganic layer. The 2D -Pb-O-Pb-Cu-Cl- inorganic layers are further pillared to form a 3D network through 3,5-pydc2− anions. To the best of our knowledge, the pillared-layer framework based on -Pb-O-Pb-Cu-Cl- inorganic layers has never been reported previously. Compound 1 exhibits weak blue light emission with the maximum at 383 nm upon excitation at 260 nm.

The oleaginous alga Botryococcus braunii (Chlorophyta) is known to accumulate hydrocarbons up to 80% of cell dry weight. Races A, B, and L of B. braunii species accumulate alkadiene/triene, botryococcene, and lycopadiene, respectively. Transcriptome analyses in race A and race B have identified various transcripts encoding enzymes involved in the biosynthesis of alkadiene/triene and botryococcene. However, the transcriptome of B. braunii race L has not been analyzed. In this study, we report the de novo assembly of the transcriptome of B. braunii AC768 race L. AC768 culture in 2×BB medium reaches a maximum density (cell dry weight) of 5.5 g L−1, of which, 6% is lycopadiene. Transcriptional profiles of AC768 show that ESTs involved in energy metabolisms are most abundantly expressed, suggesting an energy requirement for lipid accumulation. Transcriptomic analysis indicates the presence of a putative lycopaoctaene synthase that synthesizes the precursor of lycopadiene in AC768. Comparative analysis between races reveals that botryococcene synthase and lycopaoctaene synthase are specific to races B and L of B. braunii, respectively. Taken together, our results indicate that hydrocarbon chemicals accumulated in different races of B. braunii are determined by their transcriptional profiles.

This paper presents the development and application of a sensitive and specific method for the analysis of multi-mycotoxins in raw milk using ultrahigh performance liquid chromatography quadrupole orbitrap mass spectrometry (UHPLC/Q-Orbitrap). Mycotoxins (a total of 14) included in this study were aflatoxin B1, aflatoxin B2, aflatoxin G1, aflatoxin G2, aflatoxin M1, aflatoxin M2, ochratoxin A, ochratoxin B, zearalenone, zearalanone, α-zeralanol, β-zeralanol, α-zeralenol, and β-zeralenol. Mycotoxins in milk were extracted and purified with a multi-mycotoxins AOZ immunoaffinity column in a single run, which made the matrix effect be negligible. The deliberate addition of small amount of acetonitrile was found to be beneficial for the extraction of mycotoxins from the complex milk matrix, especially for zearalenone and its derivatives. The final reconstituted extracts were analyzed using ultra high performance liquid chromatography coupled to quadrupole orbitrap mass spectrometry (UHPLC Q-Orbitrap mass spectrometry). Antimony has emerged as a promising candidate for electrode materials attributed to tempting specific capacity and low cost. Nevertheless, the notorious volume fluctuation and unstable solid-electrolyte interphase (SEI) film give rise to structural degradation and rapid capacity decay. Herein, a remarkable dual-modification approach is investigated on Sb-based anode by Fe doping and double carbon coatings via self-assembling of novel Fe/Sb–hexamine bimetal–organic frameworks strategy followed by thermal treatment and chemical vapor deposition simultaneously. In the as-prepared composite, Fe-doped Sb nanoparticles decorated with in-situ formed amorphous carbon coating are dispersed in interconnected carbon matrices with robust contact (Fe-Sb@NSC). Fe-doping would effectively enhance the electric conductivity, regulate the dispersion of Sb particles, and act as buffer layer to hinder the aggregation of Sb during repeated discharge/charge process. Meanwhile, double carbon layers not only reduce electrolytic corrosion and enhance electric conductivity, but also provide robust shelter layer to restrict the volume variation and coarsening of Sb-based materials. Therefore, the dual-modified sample shows enhanced lithium storage performance with high reversible capacity, outstanding long-term stability and stable structure. Moreover, the kinetics analysis of Li+ storage indicates that the increased pseudocapacitive contribution and decreased charge transfer barriers after cycling may account for the promoted performance.

Emergent and long-term hemorrhage control is requisite and beneficial for reducing global mortality and postoperative complications (e.g., second bleeding and adverse tissue adhesion). Despite recent advance in injectable hydrogels for hemostasis, achieving rapid gelation, strong tissue-adhesive property and stable mechanical strength under fluid physiological environment is still challenging. Herein, we developed a novel chitosan hydrogel (CCS@gel) via dynamic Schiff base reaction and mussel-inspired catechol chemistry. The hydrogel possessed high gelation rate (10 s), strong wet adhesiveness, excellent self-healing performance and biocompatibility. More importantly, the CCS@gel exhibited saline-induced contractile performance and mechanical enhancement, promoting its mechanical property in moist internal conditions. In vivo studies demonstrated its superior hemostatic efficacy for diverse anticoagulated visceral and carotid bleeding scenarios, compared to commercialized fibrin glue. The hydrogel-treated rats survived for 8 weeks with minimal inflammation and postoperative adhesion. These results revealed that the promising CCS@gel would be a facile, efficient and safe sealant for clinical hemorrhage control.

Different from chemical (small molecular inhibitor) and biological (monoclonal antibody) drugs, herein, based on angiogenesis-related neuropilin-1 (NRP-1), we develop a biomimetic superstructure drug, i.e. an antibody-like peptidic network (ALPN) to achieve the high-efficient treatment of choroidal neovascularization (CNV). The ALPN in nanoparticulated formulation (ALPN-NPS) can bind NRP-1 through targeting unit and form fibrous peptidic networks trapping NRP-1 on the surface of endothelial cells (ECs), leading to anti-angiogenesis. The ALPN shows high-efficacy against angiogenesis in CNV rat model ascribed to the superstructure-enhanced binding and blockage of NRP-1. The very low dose of ALPN (0.263 μg/Kg) exhibits similar anti-angiogenesis effect comparing with monoclonal antibody bevacizumab (23.5 μg/Kg), which shows potential advantages over traditional monoclonal antibodies.

The effective chemotherapy of cancer is usually hindered by the unsatisfied cell internalization of the drug delivery systems (DDS) as well as drug resistance of cancer cells. In order to solve these dilemmas in one design, red blood cell membrane (RBM)-coated silica nanoparticles (RS) were fabricated to codeliver doxorubicin (Dox) and indocyanine green (ICG) to effectively treat the model lung cancer using photothermal-assisted chemotherapy. Our results demonstrated that the RS/I-D was the nanoparticle at around 100 nm with superior stability and biocompatibility. Especially, the photothermal effects of ICG were well preserved and could be applied to accelerate the drug release from the DDS. More importantly, the RBM modification can mediate enhanced cell internalization of drugs as compared to their free forms, which finally resulted in enhanced anticancer efficacy in Dox-resistant A549 cells (A549/Dox) both in vitro and in vivo with enhanced cell apoptosis and cell arrest.

Poly(butyl acrylate) (PBA) side chain equipped isoindigo-bithiophene (II2T) conjugated polymers have been designed and synthesized for stretchable electronic applications. The PBA segment possesses low glass transition temperature and high softness, offering a great opportunity to improve the mechanical property of semiconducting polymer thin films that typically contain lots of rigid conjugated rings. Polymers with 0, 5, 10, 20 and 100% of PBA side chains, named PII2T, PII2T-PBA5, PII2T-PBA10, PII2T-PBA20, and PII2T-PBA100, were explored, and their polymer properties, surface morphology, electrical characteristics, and strain-dependent performance were investigated systematically. The series polymers showed a charge carrier mobility of 0.06–0.8 cm2 V–1 s–1 with an on/off current ratio over 105 dependent on different amounts of PBA chains as probed using a top-contact transistor device. Ion-pair amphiphiles (IPAs, also known as catanionic surfactants) are lipid-like double-chained molecules potentially used for fabricating liposome-like vesicular drug and gene carriers. Frequently ethanol and cholesterol are added to modulate the properties of their bilayer membranes. Effects of ethanol and cholesterol on the fundamental properties of IPA bilayers such as thermotropic phase behavior, however, is not known. In this work, the bilayer phase transition behavior of two IPAs (decyltrimethylammonium-tetradecyl sulfate, DeTMA-TS, and dodecyltrimethylammonium-dodecyl sulfate, DTMA-DS) in tris buffer with various amounts of ethanol was studied by using differential scanning calorimetry (DSC). Effect of cholesterol (CHOL) addition on bilayer phase transition of IPAs with 20 vol% ethanol was thereafter systematically investigated. The experimental results showed that the main phase transition temperature (Tm) was monotonously decreased with the increase of ethanol concentration up to 30 vol%. The degree of Tm depression by ethanol is essentially the same for the two IPAs regardless of different symmetry in the hydrocarbon chains. Further addition of CHOL, however, caused a slight decrease in Tm on the one hand and a significant decrease in the enthalpy of phase transition on the other hand. When the added CHOL exceeded a specific amount, the phase transition disappeared. More hasty disappearance of phase transition was found for IPA with asymmetric structure than the symmetric one. Possible mechanisms of ethanol effect based on binding in the headgroup region of the bilayers and CHOL effect based on opposite (condensing and disordering) interactions with IPA molecules in bilayers, respectively, were proposed.

LiMnPO 4 has been considered as one of the most promising high voltage cathode materials for next-generation lithium ion batteries. However, LiMnPO 4 suffers from intrinsic drawbacks of extremely low electronic conductivity and ionic diffusivity between LiMnPO 4 /MnPO 4 . In this paper, mesoporous LiMnPO 4 nanoparticles are synthesized successfully via a facile glycine-assisted solvothermal rout. The as-prepared mesoporous LiMnPO 4 /C nanoparticles present well-defined abundant mesoporous structure (diameter of 3 ∼ 10 nm), uniform carbon layer (thickness of 3 ∼ 4 nm), high specific surface area (90.1 m 2 /g). As a result, the mesoporous LiMnPO 4 /C nanoparticles achieve excellent electrochemical performance as cathode materials for lithium ion batteries. It demonstrates a high discharge capacity of 167.7, 161.6, 156.4, 148.4 and 128.7 mAh/g at 0.1, 0.5, 1, 2 and 5C, and maintains a discharge capacity of 130.0 mAh/g after 100 cycles at 1C. The good electrochemical performance is attributed to its special interpenetrating mesoporous structure in LiMnPO 4 nanoparticles, which significantly enhances the ionic and electronic transport and additional capacitive behavior to compensate the sluggish kinetics.
