Objective: Serum phosphorous is a significant risk factor for increased carotid intima-media thickness. Increased thickness of the carotid intima is a known cause of cardiovascular disease. Coronary heart disease is a significant cause of mortality and morbidity in postmenopausal women. This study aimed to compare the relationship between serum phosphorous concentration and carotid intima-media thickness in healthy asymptomatic postmenopausal women. Methods: A retrospective review of the medical records from a health checkup center in Gangnam Severance hospital between March 2007 and September 2017 was conducted. We examined asymptomatic postmenopausal female patients with age range between 56 and 66 (N = 361) who underwent measurement of carotid intima-media thickness by B-mode ultrasonography. The physiological variables analyzed included mean blood pressure, body mass index, renal function (serum creatinine and estimated glomerular filtration rate), cholesterol levels (total cholesterol, triglyceride, and high- and low-density lipoprotein), serum phosphorous, calcium, electrolytes, diabetic status, hypertension, and albumin. Thyroid hormone (TH) signaling plays an important role in the regulation of long-wavelength vision in vertebrates. In the retina, thyroid hormone receptor β ( thrb ) is required for expression of long-wavelength-sensitive opsin ( lws ) in red cone photoreceptors, while in retinal pigment epithelium (RPE), TH regulates expression of a cytochrome P450 enzyme, cyp27c1 , that converts vitamin A 1 into vitamin A 2 to produce a red-shifted chromophore. To better understand how TH controls these processes, we analyzed the phenotype of zebrafish with mutations in the three known TH nuclear receptor transcription factors ( thraa , thrab , and thrb ). We found that no single TH nuclear receptor is required for TH-mediated induction of cyp27c1 but that deletion of all three ( thraa −/− ;thrab −/− ;thrb −/− ) completely abrogates its induction and the resulting conversion of A 1 - to A 2 -based retinoids. In the retina, loss of thrb resulted in an absence of red cones at both larval and adult stages without disruption of the underlying cone mosaic. RNA-sequencing analysis revealed significant down-regulation of only five genes in adult thrb −/− retina, of which three ( lws1 , lws2 , and miR-726 ) occur in a single syntenic cluster. In the thrb −/− retina, retinal progenitors destined to become red cones were transfated into ultraviolet (UV) cones and horizontal cells. Taken together, our findings demonstrate cooperative regulation of cyp27c1 by TH receptors and a requirement for thrb in red cone fate determination. Thus, TH signaling coordinately regulates both spectral sensitivity and sensory plasticity.

The exercise pressor reflex (EPR) is activated by muscle contractions to increase heart rate and blood pressure during exercise. While this reflex is beneficial in healthy individuals, the reflex activity is exaggerated in patients with cardiovascular disease, which is associated with increased mortality. Group III and IV afferents mediate the EPR and have been shown to express both tetrodotoxin-sensitive (TTX-S, NaV1.6, and NaV1.7) and -resistant (TTX-R, NaV1.8, and NaV1.9) voltage-gated sodium (NaV) channels, but NaV1.9 current has not yet been demonstrated. Using a F−-containing internal solution, we found a NaVcurrent in muscle afferent neurons that activates at around −70 mV with slow activation and inactivation kinetics, as expected from NaV1.9 current. However, this current ran down with time, which resulted, at least in part, from increased steady-state inactivation since it was slowed by both holding potential hyperpolarization and a depolarized shift of the gating properties. We further show that, following NaV1.9 current rundown (internal F−), application of the NaV1.8 channel blocker A803467 inhibited significantly more TTX-R current than we had previously observed (internal Cl−), which suggests that NaV1.9 current did not rundown with that internal solution. Using immunohistochemistry, we found that the majority of group IV somata and axons were NaV1.9 positive. The majority of small diameter myelinated afferent somata (putative group III) were also NaV1.9 positive, but myelinated muscle afferent axons were rarely labeled. The presence of NaV1.9 channels in muscle afferents supports a role for these channels in activation and maintenance of the EPR.NEW & NOTEWORTHY Small diameter muscle afferents signal pain and muscle activity levels. The muscle activity signals drive the cardiovascular system to increase muscle blood flow, but these signals can become exaggerated in cardiovascular disease to exacerbate cardiac damage. The voltage-dependent sodium channel NaV1.9 plays a unique role in controlling afferent excitability. We show that NaV1.9 channels are expressed in muscle afferents, which supports these channels as a target for drug development to control hyperactivity of these neurons.

Meat and food products adulterated with pork fat concern Islamic consumers whose dietary laws prohibit consumption of pork meat. The objectives of this study are to investigate the existence of thermal stable-soluble protein (TSSP) in pork fat tissues and to develop and characterize monoclonal antibodies (MAbs) highly specific to the TSSP in pork fat tissues. The TSSP extracted was used as an immunogen. After cell fusion and cloning, seven monoclonal antibodies (MAbs; PF 2B8-3, 2B8-9, 2B8-20, 2B8-28, 2B8-31, 2B8-32, and 2B8-33) were developed and confirmed to be specific to pork fat protein without cross-reaction to other animal meats and fats in an indirect ELISA and Western blot. The ELISA assay based on MAb PF 2B8-31 can sensitively detect 1% pork fat protein mixed in other animal meats. These results support that the developed MAbs especially PF 2B8-31 can be sufficiently used as a bio-receptor in the development of immunoassay for the easy and rapid detection of pork fat adulteration. In addition, the ELISA based on the MAbs developed could be a useful tool to rapidly detect low levels of pork fat fraudfully adulterated in meat and food products.

EUSO-Balloon is a pathfinder mission for the Extreme Universe Space Observatory onboard the Japanese Experiment Module (JEM-EUSO). It was launched on the moonless night of the 25th of August 2014 from Timmins, Canada. The flight ended successfully after maintaining the target altitude of 38 km for five hours. One part of the mission was a 2.5 hour underflight using a helicopter equipped with three UV light sources (LED, xenon flasher and laser) to perform an inflight calibration and examine the detectors capability to measure tracks moving at the speed of light. We describe the helicopter laser system and details of the underflight as well as how the laser tracks were recorded and found in the data. These are the first recorded laser tracks measured from a fluorescence detector looking down on the atmosphere. The current investigation was performed to evaluate the chemical analytical techniques for PAHs (organic, carcinogenic and mutagenic compounds) in food. It also determines the content of eight polycyclic aromatic hydrocarbons (PAHs) in frequently consumed fruit, vegetables, meats and their products. The methodology included saponification or ultrasonication, liquid-liquid extraction with solvents like n-hexane, clean-up using a silica solid phase extraction cartridge and a gas chromatography-mass spectrometry technique. A good linearity (R2 > 0.99) was achieved for the PAHs at different concentrations. Recovery results for PAHs extended from 88.75 to ∼100.00%. The limit of detection was 0.08–0.25 µg/kg and the limit of quantification was 0.24–0.75 µg/kg. The mean concentration (n = 3) of benzo[a]pyrene was not identified in fruits, was 0.05 µg/kg in vegetables, and 0.64 µg/kg in meat products. Overall concentration of the eight PAHs was 0.67 µg/kg in fruits, 0.82 µg/kg in vegetables, and 3.37 µg/kg in meat products.

Ochratoxin A (OTA), a mycotoxin classified as a potentially carcinogenic to humans, is slightly soluble in water. In this study, the transfer rate of OTA in herbal medicines to decoctions was investigated. Herbal medicine decoctions were prepared by boiling and autoclaving herbal medicines contaminated with OTA after soaking and not soaking in water. OTA amounts in decoctions and residues were analyzed using HPLC. Transfer of OTA (12.72–61.33%) from herbal medicines to decoctions was observed. Soaking and autoclaving steps enhanced the transfer rate of OTA to decoctions. The results meant that there is a potential risk for consumption of herbal medicine decoctions. Therefore, regular monitoring for mycotoxins, including OTA, in herbal medicines and proper management to ensure the safety of herbal medicines is necessary to protect consumers.

UNLABELLED The objective of this study was to develop a 1-step simultaneous lateral flow strip test for the rapid and simple detection of deoxynivalenol (DON) and zearalenone (ZEA) in grains. Two monoclonal antibodies (MAbs) against DON and ZEA were respectively conjugated with gold nanoparticles and used to develop a lateral flow strip test for a single toxin and multiple toxins. First, individual lateral flow strips for a single toxin were optimized, and their conditions were used to develop a simultaneous lateral flow strip for multiple toxins. Limits of detection of both lateral flow strip tests for DON and ZEA were the same (DON: 50 ng/mL, ZEA: 1 ng/mL). Both methods showed cross-reactivity for α-zearalenol and β-zearalenol, but no cross-reaction to other mycotoxins. The results can be completed obtained within 15 min. The cut-off values of the simultaneous lateral flow strip for the spiked rice and corn were 500 and 10 ng/g for DON and ZEA, respectively. The results demonstrated that the developed simultaneous lateral flow strip test offers a rapid, easy-to-use, and portable analytical system and can be used as a convenient qualitative tool for the on-site detection of DON and ZEA in food and agricultural commodities. PRACTICAL APPLICATION Simultaneous lateral strip test is useful for a rapid detection of DON and ZEA at a time in food and grain samples.

This research is focused on coagulation and sand filtration process as a pretreatment of RO seawater desalination. RO systems require sufficient and reliable pretreatment process to produce superior quality of RO feedwater that can mitigate RO membrane fouling. This experiment was conducted to investigate the effectiveness of coagulation and filtration process under various experimental conditions including different coagulant dose, flocculation mixing intensity and time, turbidity, and filtration rate. The experimental results showed that the optimum pretreatment conditions resulting in lower SDI value suitable for RO feedwater were coagulation pH 6.5, raw water turbidity greater than 4 NTU, and media bed depth greater than 550 mm. However, flocculation mixing intensity, coagulant dose, and filtration rate relatively affected little on the filtration efficiency.

To analyze the hazards associated with cucumber and hot pepper cultivation areas, a total of 72 samples were obtained and tested to detect the presence of biological (sanitary indicative, pathogenic bacteria and fungi) and chemical hazards (heavy metals and pesticide residues). The levels of sanitary indicative bacteria (aerobic plate counts and coliforms) and fungi were ND-7.2 and ND-4.8 log CFU/(g, mL, hand, or 100 cm 2 ) in cucumber cultivation areas, and ND- 6.8 and 0.4-5.3 log CFU/(g, mL, hand, or 100 cm 2 ) in hot pepper cultivation areas. More specifically, the soil of hot pepper cultivation areas was contaminated with coliforms at a maximum level of 5.6 log CFU/g. Staphylococcus aureus was detected only in glove samples at a level of 1.4 log CFU/100 cm 2 and Bacillus cereus was detected in the majority of samples at a level of ND-4.8 log CFU/(g, mL, hand, or 100 cm 2 ). Listeria monocytogenes, Escherichia coli O157:H7, and Salmonella spp. were not detected. Heavy metal (Zn, Cu, Ni, Pb, and Hg) chemical hazards were detected at levels lower than the regulation limit. Residual insecticides were not detected in cucumbers; however, hexaconazole was detected at a level of 0.016 mg/kg (maximum residue limit: 0.3 mg/kg) in hot peppers.

A rapid immunochromatographic (ICG) strip based on a conjugate of colloidal gold and monoclonal antibody (mAb) was developed for the rapid, sensitive, and on-site detection of sulfamethazine in meat and egg samples. The detection limit of the ICG strip is 2 ng/mL, and the assay can be completed in 10 min. A cross-reactivity test indicated that the ICG strip was highly specific to sulfamethazine with no cross-reaction with sulfonamide compounds and other antibiotics. The results of the recovery test from meat and egg samples spiked with sulfamethazine were in good agreement with those obtained by the indirect competitive enzyme-linked immunosorbent assay. These results demonstrated that the ICG strip can be used as a rapid and qualitative tool for on-site screening of sulfamethazine in meat and egg samples. Practical Application The present immunochromatographic strip could be used as a rapid and on-site screening tool for sulfamethazine in livestock products.

This study was carried out to find the optimal conditions for red cabbage seed sprouts in terms of their physicochemical and sensory qualities by electron-beam irradiation, cultivation and storage using the response surface methodology (RSM). Moisture content (R 2 =0.9638) was affected by irradiation dose and cultivation time. Total phenolics content (R 2 =0.9117) was mainly affected by irradiation dose, but carotenoid content (R 2 =0.8338) was affected in the order of irradiation dose, cultivation time and storage time. Sensory properties were also affected by irradiation dose, and thus scores decreased as irradiation dose increased. The optimum conditions estimated by superimposing total phenolics content and overall acceptance were 2.2-3.8 kGy of the irradiation dose, 3.0-4.0 days of cultivation and 2.0-3.0 days of storage.

Reference materials for quantitative determination of Cd, Cr, Hg and Pb in polycarbonate were developed. Reference materials with two concentration level of elements were prepared by adding appropriate amounts of chemicals to a blank polycarbonate base material. It was shown that ten bottles with triplicate analysis are enough to demonstrate the homogeneity of these candidate reference materials. The statistical results also showed no significant trends in both short-term stability test for four weeks and long-term stability test for twelve months. The certification of the four elements was carried out by isotope-dilution-inductively coupled plasma mass spectrometry (ID-ICP-MS) with microwave-assisted digestion. Certification of candidate reference materials in a single laboratory was confirmed with interlaboratory comparison participated by a certain number of well-recognized testing laboratories in Korea. The certified values and expanded uncertainties (k = 2) for the candidate reference material with low level and the one with high level were (51.7 ± 2.1) mg kg−1 Cd, (103.8 ± 2.9) mg kg−1 Cd, (98.8 ± 4.5) mg kg−1 Cr, (1004 ± 49.8) mg kg−1 Cr, (107.4 ± 4.6) mg kg−1 Hg, (1133 ± 50.7) mg kg−1 Hg, (94.8 ± 3.7) mg kg−1 Pb and (988.4 ± 53.6) mg kg−1 Pb, respectively. The reference materials developed in this study demonstrated their suitability for the quality assurance in Cd, Cr, Hg and Pb analysis for the implementation of RoHS Directive.

The effect of different light conditions on the specificity and reliability of thermoluminescence (TL) analysis to detect irradiated samples (potatoes and garlic) during 2 years of storage was investigated. The silicate minerals separated from the nonirradiated samples provided a low-intensity TL glow curve with a maximum peak after 300 °C demonstrating the absence of an irradiation history. The TL glow curve from the irradiated samples provided easy discrimination with TL glow curves of high intensity and maximum peak in temperatures range of 160–185 °C. The results were also confirmed by calculating the TL ratio (TL1/TL2) through normalization (re-irradiation) step. The key parameters of the TL analysis including the TL intensity, TL glow curve shape, and the TL ratio exhibited a time-dependent change. The bleaching effect of different light conditions was also significant, which was most prominent in the case of natural light with a prominent decrease in the TL intensity and ratio. The maximum TL glow peak also showed a shift towards the higher temperature. However, all irradiated samples, irrespective of the storage conditions and time, were easily discernable from the nonirradiated ones considering the shape and intensity of the TL glow curve.

Green tea leaves were fermented for 15 and 30 days with Monascus pilosus which is known to produce functional statins (TMs), and the content of various biochemical constituents such as total polyphenol (TP), total flavonoid (TF), theaflavin, and thearubigin were analyzed and compared with that of non- fermented green tea (GT) and Pu-erh Chinese post-fermented tea (PU). In addition to the electron donating ability (EDA), ferric iron reducing power (FIRP), xanthine oxidase (XO) inhibitory activity, superoxide dismutase (SOD)-like activity, iron chelating activity (ICA) and hydrogen peroxide contents were also measured and compared with that of GT and PU. Content of TP and TF in the water and ethanol extracts in TMs were lower than those in GT and PU. Theaflavin and thearubigin contents of water and ethanol extracts in TMs were higher than those of GT. And, these components were increased depending on the period of fermentation. While, EDA and FIRP of TMs were lower than those of GT, XO inhibitory activity of TMs was higher than non- fermented tea. While, ICA of TMs was slightly higher than GT and PU, the content of hydrogen peroxide in TMs was markedly lower than GT. This results suggested that the green tea fermented by M. pilosus was valuable for oxidative stress-induced diseases by decreasing hydrogen peroxide, and forming theaflavins and thearubigins with functionality of genus Monascus.

The epidermal recovering-effects of the Cnidium officinale extract after inducing contact-dermatitis by 1-chloro-2,4-dinitrochlorobenzene (DNCB) showed by measuring thickness of skin, identifying IgE amounts in plasma, and activities of anti-oxidant enzymes using histochemical and biochemical methods. By three-time applications of the extract, morphological changes of dermatitis in impaired region were recovered dramatically and shape of skin surface and thickness of epidermis were restored to be normal. Also, lipid content was recovered to the level of normal state. We suggested that extract treatment lowered a hypersensitive reaction by decreasing of IgE level in blood and restored activities of superoxide dismutase and catalase. In conclusion, we proposed that C. officinale extract might be used as natural resources for treating effectively allergic contact-dermatitis.

Response surface methodology (RSM) was used for the microwave-assisted extraction (MAE) of the effective components of grape seed, such as its antioxidative and nitrite-scavenging abilities. Microwave power (2,450 MHz, 0-160W), ethanol concentration (0-100%), and MAE time (1-5 min) were used as independent variables (Xi) for the central composite design to yield 16 different MAE conditions. The optimum MAE conditions were predicted for the dependent variables of the extracts, such as the total phenolic content () antioxidative ability (), and nitrite-scavenging ability (), depending on different microwave powers, ethanol concentrations, and MAE times. The determination coefficients () of the regression equations for the dependent variables ranged from 0.8024 to 0.9498. The maximal values of each dependent variable were predicted at different MAE conditions, as follows: 3.19% total phenolic content at 142.32W, 44.30% ethanol, and 4.36 min, and 1.22 antioxidative ability at 84.44W, 56.60% ethanol, and 3.28 min. More than 99.5% nitrite-scavenging ability was predicted at pH 1.2-3.0, 30.80-106.58W, 49.32-55.18% ethanol, and 3.72-4.58min, respectively. The results indicated that the total phenolic content and anti oxidative ability showed a higher correlation with each other in that they were more influenced by microwave power than by the other variables, while the nitrite-scavenging ability was largely influenced by the ethanol concentration.

Here, we elucidated the roles of mast cells in a myocardial infarction (MI) rat model. Parkinson's disease (PD) involves an irreversible degeneration of the nigrostriatal pathway. As most cases of PD are sporadic, environmental risk factors may underlie neurodegeneration in dopaminergic neurons. One such factor is 6-hydroxydopamine (6-OHDA), which is widely used as a parkinsonian mimetic. Studies have shown that 6-OHDA generates reactive oxygen species and induces cell stress, the unfolded protein response, and apoptosis. Present findings show that 6-OHDA, but not hydrogen peroxide, MPP+, or rotenone, leads to the rapid formation of high-molecular-weight species of protein disulfide isomerase-associated protein 3 (ERp57) in a dose- and time-dependent fashion. Moreover, ERp57 conjugates are blocked by N-acetylcysteine and glutathione, suggesting that they represent oxidized forms of protein. Surprisingly, conjugates are complexed with DNA, because treatment with DNase reduces their appearance. Subcellular fractionation indicates that both nuclear and mitochondrial DNA are associated with the protein. Finally, toxin-treated ERp57 rapidly forms juxtanuclear aggresome-like structures in dopaminergic cells, suggesting that ERp57 plays an early adaptive response in toxin-mediated stress. Understanding the signaling mechanisms associated with parkinsonian mimetics, as well as their temporal induction, may aid in designing better interventions in models of PD.

Transferrin is a molecule carrying iron to store and maintain for iron homeostasis of living organisms. In this study, we have purified transferrin, as an iron-binding protein, from the last larval haemolymph of Papilio xuthus by KBr density gradient ultracentrifugation and gel filtration (superose 6 HR) using fast protein liquid chromatography (FPLC) and transferrin containing iron was identified by Ferene S staining. The purified haemolymph transferrin was shown to have molecular mass of 78 and 80 kDa and amino acid composition of transferrin was rich in aspartic acid, valine, leucine and glutamic acid. With immuno-diffusion assay, we confirmed the existence of the transferrin in the haemolymph and fat body by detection of visible and clear positive reaction. From the quantitative comparison by rocket immuno-electrophoresis process, the amount of transferrin were increased in the haemolymph of 3 days after pupation and the whole 5 days after pupation. Here, with biochemical and immunohistochemical analysis, we speculate the relationship of transferrin between the physical characteristics and distribution during metamorphosis of P. xuthus.

Response surface methodology (RSM) was applied in steaming and roasting processes of Polygonatum odoratum roots in order to monitor hypoglycemic components and sensory property. In steaming and roasting processes based on the central composite design with variations in steaming time , roasting temperature and roasting time , coefficients of determinations were 0.8691 (p, hypoglycemic component (total), and overall acceptability, respectively. The maximum value of soluble solid was in 65.24 min of steaming time, of roasting temperature and 37.58 min of roasting time. The maximum value of hypoglycemic component (total) was in 107.76 min, and 14.70 min. Meantime, the maximum value of overall acceptability was 6.89 in 126.04 min, and 43.93 min. The predicted values in optimum conditions for hypoglycemic components and sensory property were in good agreement with experimental values.

Extracts of 33 samples of seaweed, shrimp, fish and shellfish, including two certified reference materials, were investigated for their contents of arsenic compounds (arsenic speciation).An anion exchange high performance liquid chromatography procedure was optimized to separate six arsenic compounds present in the seafood samples with dynamic reaction gas cell by inductively coupled plasma mass spectrometry. The concentration of each species in the sample were: arsenobetaines - 0.019-1.04 mg/kg, arsenocholine - 0.033-69.0 mg/kg, arseniousacid - ND-1.25 mg/kg, dimethylarsinate - ND-3.75 mg/kg, monomethylarsonate - ND-8.33 mg/kg, arsenic acid - ND-0.55 mg/kg. Additionally, unknown arsenic species were present in most of samples. The intake of inorganic arsenic via ingestion of the seafood samples that were analyzed did not represent a toxicological problem to humans. The limits of detection (LOD) were in the range of 0.5-2.5 µg/kg .

Nitrous oxide (N2O), one of the greenhouse effect gases, has not been known that how much N2O is produced from municipal wastewater treatment and what its management should be. In this study, for controlling nitrous oxide emission and removing nitrogen from municipal wastewater, we experimented the three phase fluidized bed process equipped with draft tube along with immobilized Alcaligenes faecalis, a typical heterotrophic nitrifer and a predominant genus. Also we evaluated the optimum treatment condition of the three phase fluidized bed process for emitting nitrous oxide. The results of this study showed that the three phase fluidized bed process was more effective than the activated sludge process for controlling nitrous oxide emission and removing nitrogen. Increasing amount of A. faecalis in reactor should be encouraged for controlling nitrous oxide emission and removing nitrogen. In addition, the activated sludge process using immobilized A. faecalis as a carrier had more nitrogen removal efficiency than conventional activated sludge process. The accumulation of NO2-N, NO3-N resulted in high N2O emission. Therefore, we suggested that it is necessary to reduce NO2-N and NO3-N for both reducing N2O emission and improving nitrogen removal.

Objectives: Although the direct cause for periodontitis is oral bacterial infection, its progression depends upon genetic and environmental factors. Smoking, one of the environmental factors, is a risk factor for the development and severity of periodontitis. Therefore, individual susceptibility to periodontitis may be influenced by the polymorphisms of genes coding for enzymes metabolizing tobacco-derived substances. The object of this study is to investigate roles of genetic polymorphisms of these metabolizing enzymes in the risk for periodontitis. Material and Methods: We investigated three important enzymes: cytochrome P450 (CYP) 1A1, CYP2E1 and glutathione S-transferase (GST) Ml, involved in the metabolic activation and detoxification of tobacco-derived substances. The prevalence of the polymorphisms of these genes was examined in 115 patients with periodontitis as well as in 126 control subjects. Results: Significantly increased risk for periodontitis was observed for subjects with the polymorphic CYP1A1 m2 allele (odds ratio (OR) = 2.3, 95% confidence interval (CI) = 1.2-4.4). A significant risk increase for periodontitis associated with the GSTM1 allele was observed (OR = 2.1, 95% CI = 1.3-3.6). However, no association was observed between the CYP2E1 Pst1 polymorphism and risk for periodontitis (OR = 1.3, 95% CI = 0.6-2.5). Conclusion: These results suggest that the GSTM1 and CYP1A1 polymorphisms may play an important role in risk for periodontitis.

We synthesized an estrogen analog, ZYC-5, lacking activity at the classical estrogen receptor and examined its neuroprotective potential against necrosis induced by N -methyl- d -aspartate (NMDA) and apoptosis/necrosis induced by the NMDA receptor antagonist (+)-3-(2-carboxypiperazine-4- yl )-propyl-1-phosphonic acid (CPP). ZYC-5 protected cortical neurons in a dose-dependent manner, and the neuroprotection was more robust than with 17β-estradiol. The effect of ZYC-5 was not mediated by the classical estrogen receptor, because it was unaffected by the antagonists 4-hydroxytamoxifen and ICI 182,780. The ZYC-5 protection against excitotoxicity was not directly mediated through the NMDA receptor, because there was no effect of ZYC-5 on NMDA current or the intracellular calcium increase induced by NMDA. Results obtained with the free-radical-sensitive dye, dihydroethidium, suggested that the neuroprotection of ZYC-5 was partly related to its radical scavenging properties. Although some of estrogen's neuroprotective effects may depend upon the estrogen receptor, our results suggest the possibility of neuroprotection without hormonal side effects.

L-DOPA may cause side-effects during the treatment of Parkinson’s disease. We investigated the role of glutathione peroxidase (GSHPx) in cellular defense against L-DOPA cytotoxicity. A line of PC12 cells overexpressing GSHPx with plasmid pRc/CMV-GSHPx was established and stable transfectants overexpressing GSHPx were used for this study. GSHPx activity was found to be 1.5-fold higher in GSHPx-transfectants than in mock-controlled transfectants. Transfectants over expressing GSHPx were also significantly more resistant to exposure to either L-DOPA or t-butyl hydroperoxide than mock-transfected cells. Results suggested that L-DOPA may cause neuronal cell death by an oxidative pathway and GSHPx may play an important role in cellular defense against oxidative stress.

5-Aminolevulinic acid and its derivatives, which are known to affect the early diagnosis and treatment of cancer, have been synthesized. Simple methods for the synthesis of 5-aminolevulinic acid (ALA), a precursor of porphyrins, have been developed in our laboratory for use in studies on the biosynthesis of porphyrins.

Rationale: Increased arginine-vasopressin (AVP) secretion is a key physiological response to hyperosmotic stress and may be part of the mechanism by which high-salt diets induce or exacerbate hypertension. Objective: Using deoxycorticosterone acetate-salt hypertension model rats, we sought to test the hypothesis that changes in GABA A receptor–mediated inhibition in AVP-secreting magnocellular neurons contribute to the generation of Na + -dependent hypertension. Methods and Results: In vitro gramicidin-perforated recordings in the paraventricular and supraoptic nuclei revealed that the GABAergic inhibition in AVP-secreting neurons was converted into excitation in this model, because of the depolarization of GABA equilibrium potential. Meanwhile, in vivo extracellular recordings in the supraoptic nuclei showed that the GABAergic baroreflexive inhibition of magnocellular neurons was transformed to excitation, so that baroreceptor activation may increase AVP release. The depolarizing GABA equilibrium potential shift in AVP-secreting neurons occurred progressively over weeks of deoxycorticosterone acetate-salt treatment along with gradual increases in plasma AVP and blood pressure. Furthermore, the shift was associated with changes in chloride transporter expression and partially reversed by bumetanide (Na + -K + -2Cl – cotransporter inhibitor). Intracerebroventricular bumetanide administration during deoxycorticosterone acetate-salt treatment hindered the development of hypertension and rise in plasma AVP level. Muscimol (GABA A agonist) microinjection into the supraoptic nuclei in hypertensive rats increased blood pressure, which was prevented by previous intravenous V1a AVP antagonist injection. Conclusions: We conclude that the inhibitory-to-excitatory switch of GABA A receptor–mediated transmission in AVP neurons contributes to the generation of Na + -dependent hypertension by increasing AVP release. We speculate that normalizing the GABA equilibrium potential may have some utility in treating Na + -dependent hypertension.

An organic-assisted alkaline extraction method was developed for the determination of hexavalent chromium (Cr(VI)) in plastics. The solubilization of polymer as a pre-step of the alkaline extraction provided good extraction efficiency of Cr(VI) from the sample. The optimization of the experimental conditions affecting the extraction and UV-vis spectrophotometric analysis was accomplished by evaluating the recovery rate of Cr(VI) through the analysis of Cr(VI) in in-house polymer reference materials (RMs). With the proposed method, most of the Cr(VI) in polymers was released within a short extraction time of 30 min and the Cr(III)-DPCO complex can be kept stable for 24 h. The heating for the extraction of the Cr(VI) was not necessary. The optimal pH of the final solution was fixed at 2.0. The proposed extraction method was applied successfully for the determination of Cr(III) and Cr(VI) in spiked samples. The practical applicability of this new method was evaluated through the analysis of Cr(VI) in in-house polymer RMs. The good linearity was demonstrated at desired concentrations of the range 0-3.3 mg L(-1). The detection limits were quite low, varying from 0.0061 to 0.0285 mg L(-1). The recovery of Cr(VI) was between 97 and 106%, and the relative standard deviation (R.S.D.) was below 6%.

The protective effects of freeze-dried privet (Ligustrum obtusifolium) fruits (PFs) were observed in streptozotocin (STZ)-induced diabetic rats on a high fat diet by measuring levels of blood glucose, serum insulin, fructosamine, and hepatic reactive oxygen species generating and scavenging enzyme activities. A PF-supplemented diet was prepared by mixing an AIN-76 diet with powdered PF (final concentration, 1% or 2%). It was fed to STZ-induced diabetic rats on a high fat diet for 6 weeks. Diabetic animals receiving the PF-supplemented diet showed a significant increase in body weight, feed efficiency ratio, liver, kidney, and heart weight, and serum glucose, insulin, and fructosamine levels compared with high fat diet-fed diabetic animals. The treatment with PF showed improved hepatic glutathione S-transferase, superoxide dismutase, and xanthine oxidase activities as well as glutathione and lipid peroxide levels in the diabetic animals. Intracellular swelling and vacuole formation in diabetic pancreatic beta- and delta-cells were ameliorated by the PF-supplemented diet. Furthermore, necrosis of tubular epithelial cells and dilatation of luminal space in diabetic kidneys exhibited near-noninjured condition. This is the first time an antihyperglycemic effect of L. obtusifolium fruit in STZ-induced diabetic rats has been identified.

Based on the biomimetic approaches the present work describes a straightforward technique to mimic not only the architecture (the morphology) but also the chemistry (the composition) of the lowest level of the hierarchical organization of bone. This technique uses an electrospinning (ES) process with polyvinyl alcohol (PVA) and hydroxyapatite (HAp) nanoparticles. To determine morphology, crystalline structures and thermal properties of the resulting electrospun fibers with the pure PVA and PVA/HAp nanocomposite (NC) before electrospinning various techniques were employed, including transmission electron microscopy (TEM), high-resolution TEM (HR-TEM), scanning electron microscopy (SEM), x-ray diffraction (XRD), differential scanning calorimetry (DSC) and thermogravimetric analysis (TGA). In addition, FT-IR spectroscopy was carried out to analyze the complex structural changes upon undergoing electrospinning as well as interactions between HAp and PVA. The morphological and crystallographic investigations revealed that the rod-like HAp nanoparticles exhibit a nanoporous morphology and are embedded within the electrospun fibers. A large number of HAp nanorods are preferentially oriented parallel to the longitudinal direction of the electrospun PVA fibers, which closely resemble the naturally mineralized hard tissues of bones. Due to abundant OH groups present in PVA and HAp nanorods, they strongly interact via hydrogen bonding within the electrospun PVA/HAp NC fibers, which results in improved thermal properties. The unique physiochemical features of the electrospun PVA/HAp NC nanofibers prepared by the ES process will open up a wide variety of future applications related to hard tissue replacement and regeneration (bone and dentin), not limited to coating implants.

The antihyperglycemic effect of a water extract (WE) and an alkali extract (AE) of the Fomitopsis pinicola fruit body was studied in streptozotocin (STZ)-induced diabetic rats. The STZ-induced diabetes mellitus (DM) control group lost a significant amount of body weight, whereas the normal control group (NC) gained weight; however, the DM-AE group gained a significant amount of weight, with weight gain approaching normal. Feed intake by the DM-AE group was also similar to the NC group. The liver and kidney weights per body weight increased with the STZ treatment; however, the weights were lower in the F. pinicola-treated groups and nearly normalized in the DM-AE group. The weights of the heart, lungs, and spleen were not influenced by the STZ treatment. Blood glucose levels of F. pinicola-treated DM groups were significantly lower than that of the DM group. In particular, STZ-induced hyperglycemia was remarkably inhibited by the AE-supplemented diet. Serum insulin levels were decreased with STZ injection; however, the decreased levels were almost restored to the NC level with F. pinicola supplementation. The increased serum fructosamine levels associated with hyperglycemia were decreased with the F. pinicola treatment. Cells of the pericentral regions were found to have significant swelling, and some necrotic cells were observed in the pancreas of DM animals; however, pancreatic tissue damage by STZ in the F. pinicola-supplemented diet groups was ameliorated. In this study, the AE from F. pinicola showed the highest antidiabetic effect among the treatments. These results indicate that constituents of F. pinicola may regulate hyperglycemia via either increased insulin secretion during recovery or the prevention of STZ-induced pancreatic damage. This is the first report of antihyperglycemic effects of F. pinicola in STZ-induced DM rats.

Neurons in the suprachiasmatic nucleus (SCN) are responsible for the generation of circadian oscillations, and understanding how these neurons communicate to form a functional circuit is a critical issue. The neurotransmitter GABA and its receptors are widely expressed in the SCN where they mediate cell-to-cell communication. Previous studies have raised the possibility that GABA can function as an excitatory transmitter in adult SCN neurons during the day, but this work is controversial. In the present study, we first tested the hypothesis that GABA can evoke excitatory responses during certain phases of the daily cycle by broadly sampling how SCN neurons respond to GABA using extracellular single-unit recording and gramicidin-perforated-patch recording techniques. We found that, although GABA inhibits most SCN neurons, some level of GABA-mediated excitation was present in both dorsal and ventral regions of the SCN, regardless of the time of day. These GABA-evoked excitatory responses were most common during the night in the dorsal SCN region. The Na+-K+-2Cl−cotransporter (NKCC) inhibitor, bumetanide, prevented these excitatory responses. In individual neurons, the application of bumetanide was sufficient to change GABA-evoked excitation to inhibition. Calcium-imaging experiments also indicated that GABA-elicited calcium transients in SCN cells are highly dependent on the NKCC isoform 1 (NKCC1). Finally, Western blot analysis indicated that NKCC1 expression in the dorsal SCN is higher in the night. Together, this work indicates that GABA can play an excitatory role in communication between adult SCN neurons and that this excitation is critically dependent on NKCC1.

Ceramics were introduced in total hip arthroplasty to address the problems of friction and wear that were reported with metal on polyethylene articulations. This remarkable sliding characteristics and wettable material could limit wear debries generation and provide longer lifetime of the artificial hip. Extensive clinical study shows that alumina to alumina is the best used in young and active patients, while the classic metal on polyethylene couple remains the gold standard in the remaining population. Compared with metal, ceramic is weak in tension and brittle and fracture is well recognized problem.

Streptococcus mutans (S. mutans) is most commonly founded Mutans Streptococci group in human oral cavity. This study aimed to compare the adhesion ability of Xylitol-sensitive (Xs) and Xylitol-resistant (Xr) on three filling materials which are currently used by dentists. Hydroxyapatite (HA) disk (control group, n=36), sealant (n=36), resin (n=36), glass-ionomer (n=36) disks were made by mold (10.2 mm O, 3 mm hight). Xs and Xr of S. mutans KCTC3065 and S. mutans UA159 were formed and cultured in TYE media. After culture, 500 ㎕ of 0.01% crystal violet was added to the plate, dyeing was performing, and mixed 1 ㎖ solution of ethanol:acetone (4:1 w/w) for reaction termination. The absorbance of the extracted solution was measured by using ELISA reader and the extracted solution on disk was observed with SEM. The adhesion ability was calculated by ELISA reader. Adhesion ability of Xs and Xr from S. mutans of resin was higher than those of HA disk and the adhesion ability of sealant was lower compared to those of HA disk. Sealant had the lowest adhesion ability of dental caries-inducing bacteria among three filling materials when viewed in terms of relevance of the material and decay-inducing bacteria in oral cavity.

In this study, a newly-synthesized metalloporphyrin, Gd-chlorin (PB Chlorin), was investigated by using a simple tissue phantom to test its efficacy as an MRI contrast, agent. This study demonstrated the potential activity of Gd-chlorin as not only a MRI contrast agent, but also as a PDT photosensitizer by using a simple tissue phantom and conducting a very brief MRI experiment.

Flavonoids are naturally occurring polyphenolic compounds that have many biological properties, including antioxidative, anti-inflammatory and neuroprotective effects. Here, we report that amentoflavone significantly reduced cell death induced by staurosporine, etoposide and sodium nitroprusside in neuroblastoma SH-SY5Y cells. In post-natal day 7 rats, hypoxic-ischemic (H-I) brain damage induced by unilateral carotid ligation and hypoxia resulted in distinct features of neuronal cell death including apoptosis and necrosis. In this model, a systemic administration of amentoflavone (30 mg/kg) markedly reduced the H-I-induced brain tissue loss with a wide therapeutic time window up to 6 h after the onset of hypoxia. Amentoflavone blocked the activation of caspase 3, characteristic of apoptosis, and the proteolytic cleavage of its substrates following H-I injury. Amentoflavone also reduced the excitotoxic/necrotic cell death after H-I injury in vivo and after oxygen/glucose deprivation in mouse mixed cultures in vitro. Treatment of mouse microglial cells with amentoflavone resulted in a significant decrease in the lipopolysaccharide-induced production of nitric oxide and induction of inducible nitric oxide synthase and cyclo-oxygenase-2. Furthermore, amentoflavone decreased the inflammatory activation of microglia after H-I injury when assessed by the microglial-specific marker OX-42. These data demonstrate for the first time that amentoflavone strongly protects the neonatal brain from H-I injury by blocking multiple cellular events leading to brain damage.

Copper toxicity contributes to neuronal death in Wilson's disease and has been speculatively linked to the pathogenesis of Alzheimer's and prion diseases. We examined copper-induced neuronal death with the goal of developing neuroprotective strategies. Copper catalyzed an increase in hydroxyl radical generation in solution, and the addition of 20 microM copper for 22 hours to murine neocortical cell cultures induced a decrease in ATP levels and neuronal death without glial death. This selective neuronal death was associated with activation of caspase-3 and was reduced by free radical scavengers and Z-Val-Ala-Asp fluoromethylketone, consistent with free radical-mediated injury leading to apoptosis. Pyruvate dehydrogenase is especially vulnerable to inhibition by oxygen free radicals, and the upstream metabolites, pyruvate, phosphoenolpyruvate, and 2-phosphoglycerate were elevated in cortical cells after toxic exposure to copper. One approach to protecting pyruvate dehydrogenase from oxidative attack might be to enhance binding to cofactors. Addition of thiamine, dihydrolipoic acid, or pyruvate reduced copper-induced neuronal death. To test efficacy in vivo, we added 1% thiamine to the drinking water of Long Evans Cinnamon rats, an animal model of Wilson's disease. This thiamine therapy markedly extended life span from 6.0 +/- 1.6 months to greater than 16 months.
