Direct absorption of sunlight with carbon nanofluids is a facile way to efficiently harvest solar-thermal energy, but poor dispersion stability of nanofluids under elevated temperatures severely limits their applications. Herein, we report a general strategy to prepare stably-dispersed medium-temperature solar-thermal nanofluids by employing crumpled particles of ethanol-wetted graphene oxide sheets as the self-dispersible photothermal converters. The crumpled particles simultaneously possess intensively-deformed surface structure, a low density and a small particle size. Such features weaken the inter-particle van der Waals attraction and gravitational sedimentation of the crumpled particles, thereby enabling their long-term stable dispersion within commercial thermal storage oils. Uniform dispersion of nanofluids was maintained after continuous heating for 2 weeks under a heating temperature up to 200 °C. The homogenously dispersed nanofluids also achieved high solar absorptance with low particle loadings, and retained the large heat capacity and suitable viscosity. These combined advantageous thermophysical properties enabled consistent high-performance medium-temperature direct absorption-based nanofluidic solar-thermal energy harvesting under large-flux solar illumination.

The alkyl substituent(s) in the scaffold of the alkylphosphanyl PNP ligands of the form Ph2PN(cyclopentyl)PPhR, Ph2PN(cyclopentyl)PR2, and RPhPN(cyclopentyl)PPhR have been observed to have a marked impact on the catalytic performance of Cr(III) catalysts in ethylene oligomerization reactions. Precatalyst 6, bearing diisopropylphosphanyl substituent, afforded the highest catalytic activity of 4490 kg(product)·g(Cr)−1·h−1 and 51.0% C8 selectivity with good product purity under suitable conditions. Precatalyst 7, having diethylphosphanyl substituent in the PNP scaffold, exhibited 72.0% high C8 selectivity with 1717 kg(product)·g(Cr)−1·h−1 improved catalytic activity. Precatalysts based on monoalkylphosphanyl PNP (2–4) and symmetrical alkylphosphanyl PNP (9–10) exhibited poor catalytic performance toward selective ethylene oligomerization. Single crystal analysis suggested that the alkyl substituents in the scaffold of the alkylphosphanyl PNP ligands may effectively induce the complex structure and consequently the catalytic performance. Density functional theory (DFT) calculations revealed that in comparison to the trimerization pathway, the rate-determining states in the tetramerization pathways of precatalysts 6–7 may face low energy barriers and therefore may offer high C8 selectivity.

CuO–CeO2 (Cu–Ce) catalyst with a CuO/CeO2 mass ratio of 1 prepared by a sol–gel method is used in the CO catalytic oxidation reaction in the actual industrial sulfur-containing atmosphere. At a reaction temperature of 200 °C, the catalyst exhibits quite different stability under sulfur-containing and sulfur-free conditions. When 30 ppm SO2 was added to the feed gas, the Cu–Ce catalyst had an initial CO conversion rate of 100%, gradually decreasing after 26 h, and this catalyst completely deactivated at about 50 h. However, the CO conversion rate of the catalyst under sulfur-free conditions could be nearly maintained at 100% within the measured time range (60 h). The results of IR, Raman, and XPS characterizations proved that the accumulation of cerium sulfate on the Cu–Ce catalyst would cover the active sites of the catalyst, eventually leading to the complete deactivation of the catalyst, which provides favorable evidence for the actual industrial anti-sulfur application.

CeO2/g-C3N4 heterojunction photocatalyst had been successfully fabricated through a one-step in-situ pyrolysis formation of 3D hollow CeO2 mesoporous nanospheres and 2D g-C3N4 nanosheets together with simultaneous removal of carbon sphere templates after heat treatment. The sample shows high catalytic performances for photocatalytic hydrogen generation and photocatalytic oxidation of Bisphenol A (BPA) under visible light irradiation, and the catalytic degradation route of BPA was suggested by the degradation products determined by GC–MS. The enhancing catalytic activity was attributed to the effective interfacial charge migration and separation. Finally, it was proposed that the CeO2/g-C3N4 heterojunction photocatalyst could follow a more appropriate Z-scheme charge transfer mechanism, which was confirmed by the analysis of experiment and theoretical calculation results.

Animal derived biomaterials have attracted much attentions in treating large size bone defect due to their excellent biocompatibility and potent bioactivities offered by the biomacromolecules and growth factors contained in these materials. Dermis-derived matrix (ADM) has been used as skin grafts and wound dressings for decades, however its application in bone tissue engineering has been largely limited as ADM possesses a dense structure which does not support bone tissue ingrowth. Recently, we have successfully fabricated porous scaffold structure using an ADM with the aid of micronization technique. When integrated with inorganic components such as calcium phosphate, ADM could be transformed to bone graft substitutes with desirable osteogenic properties. While purified and chemically cross-linked collagen has lost its natural structure, our ADM successfully preserved natural tropocollagen structure, as well as other bioactive components. A composite scaffold was fabricated by incorporating dicalcium phosphate (DCP) microparticles into ADM microfibers and freeze-dried to form a highly porous structure. Unlike conventional ADM materials, this scaffold possesses high porosity with interconnected pores. More importantly, our evaluation data demonstrated that it performed much more effective in treating critical bone defects in comparison with best commercial product on the market. In a head-to-head comparison with a commercial bone graft material Bongold®, the ADM/DCP scaffold showed superior osteogenic capacity by filling the defect with well-organized new bone tissue in a rabbit radius segmental defect model. Put together, our results exhibited a novel bone graft substitute was developed by circumventing processing barriers associated with natural ADM, which offers another novel bone graft substitute for bone regeneration.

Mesoporous graphene and other carbon derivative nanospheres have attracted intensive research attention owing to their many unique characteristics and fascinating properties. However, facile synthesis of these mesoporous nanospheres with a small particle size and controllable morphology is still a grand challenge. Herein, we report a Kirigami-inspired approach to prepare mesoporous spherical graphene oxide (MSGO) particles through spray drying of holey GO sheets. By increasing pore size and porosity of the holey GO sheets, the spray-dried particles underwent the transition from rough crumpled structure to smooth spherical shape. The prepared MSGO particles have tunable diameters smaller than 200 nm and a high specific surface area of 503 m2/g, which enable consistent high-efficiency physical adsorption removal of organic dyes such as Rhodamine B from polluted water with a high adsorption rate of 335.6 mg/(g min). Moreover, the reported synthetic route provides the freedom to easily incorporate other functional nanoparticles such as TiO2, CuO or post-dope MSGO with nitrogen elements for construction of multifunctional composite particles with tailorable structures, which may enable a broad range of promising applications.

Ambient air pollution is a leading cause of non-communicable disease in the world. PM2.5 has the potential to change the miRNAs profiles, which in turn causes cardiovascular effects. Hypoxia-inducible factor (HIF)-1 plays a critical role in the development of atherosclerosis. Yet, the possible role of miR-939-5p/HIF-1α in PM2.5-induced endothelial injury remains elusive. Therefore, the study aims to investigate the effects of miR-939-5p and HIF-1α on PM2.5-triggered endothelial injury. The results from immunofluorescence, qRT-PCR, LSCM, and western blot assays demonstrated that PM2.5 increased the levels of HIF-1α, inflammation and apoptosis in human aortic endothelial cells (HAECs). Yet, the inflammatory response and mitochondrial-mediated apoptosis pathway were effectively inhibited in HIF-1α knockdown HAECs lines. The expression of miR-939-5p was significantly down-regulated in HAECs after exposed to PM2.5. The luciferase reporter, qRT-PCR and western blot results demonstrated that miR-939-5p could directly targeted HIF-1α. And the miR-939-5p overexpression restricted PM2.5-triggered decreases in cell viability and increases in lactic dehydrogenase (LDH) activity, reactive oxygen species (ROS), mitochondrial membrane potential (MMP) and inflammation. In addition, miR-939-5p overexpression remarkably suppressed PM2.5-triggered BcL-2/Bax ratio reduction and Cytochrome C, Cleaved Caspase-9 and Cleaved Caspase-3 expression increase, revealed that miR-939-5p hampered PM2.5-induced endothelial apoptosis through mitochondrial-mediated apoptosis pathway. Our results demonstrated that PM2.5 increased the expression of HIF-1α followed by a pro-inflammatory and apoptotic response in HAECs. The protective effect of miR-939-5p on PM2.5-triggered endothelial cell injury by negatively regulating HIF-1α. miR-939-5p might present a new therapeutic target for PM2.5 induced endothelial injury.

After adding either organic or inorganic ligands, sulfidated nano-zero-valent iron (SnZVI) was used for aerobic degradation of phenol, and the effect of the ligand species on oxidation performance was investigated. We found that SnZVI hardly degraded phenol in the absence of ligand addition. Ligands initiated and promoted the degradation of pollutants by SnZVI. The data herein show that a characteristic inorganic ligand, tripolyphosphate (TPP), is more effective in enhancing oxidation than a characteristic organic ligand oxalate. In addition to the scavenging of reactive oxidants by the organic ligand, more ferrous ion (Fe(II)) dissolution from SnZVI in the TPP system is another cause for the superior enhancement by the inorganic ligand. In the oxalate system, as the sulfur content of SnZVI increased, the oxidation efficiency increased because FeS shell promoted the transfer of electrons to produce more reactive oxygen species (ROS). In TPP system, the effect of sulfur content on oxidation performance is more complex. The SnZVI with low sulfur content showed poor oxidation performance compared with that of nZVI. Further experiments proved that sulfidation might weaken the complexation of TPP with surface bound Fe, which would slow down the ionic Fe(II) dissolution rate. Therefore, sulfidation has the dual effects of enhancing electron transfer and inhibiting the complexation of inorganic ligands. In addition, the mechanisms of ROS generation in different ligand systems were investigated herein. Results showed that the critical ROS in both the oxalate and TPP systems are hydroxyl radicals, and that they are produced via one-electron activation of O2.

Currently, the development of high-performance and stable non-noble metal catalysts for low-temperature CO2 methanation is quite challenging for practical industrial applications in terms of highly efficient and renewable energy storage and conversion. In this work, highly dispersed Ni nanoparticles over a perovskite-type AlCeO3 solid-solution support as catalysts for exceptional low-temperature CO2 methanation were synthesized via an innovative single-source Ni–Al–Ce layered double hydroxide (LDH) precursor route. It was demonstrated that the as-fabricated Ni-based catalyst with a Ce/(Ce + Al) molar ratio of 0.2 displayed superior low-temperature catalytic activity for CO2 methanation compared to the Ce-free Ni catalyst and Ni/CeO2 and Ni/Al2O3 catalysts obtained via the impregnation method for comparison, with a high CO2 conversion of 83.2% at only 200 °C and a high CO2 turnover frequency value of 18.2 h−1 achieved at a low reaction temperature of 175 °C. This remarkable low-temperature activity of the catalyst for CO2 methanation outperforms all other supported Ni catalysts reported thus far. It was verified that the favorable Ce3+ sites in the perovskite-type AlCeO3 solid solution contribute to the enhanced medium-strength surface basicity, which is beneficial for CO2 adsorption and the formation of formate intermediate species, thus greatly accelerating the transformation of formate intermediates to the target methane product. Furthermore, for the Ni/AlCe-0.2 catalyst, no deactivation could be observed, indicating the good stability and reusability of the Ce-containing Ni-based catalysts. The present findings provide a new stable and high-performance non-noble metal-based catalyst by integrating Ni nanoparticles with perovskite-type AlCeO3 solid-solution support for exceptional low-temperature CO2 methanation.

DNA cages are ideally suited to make nanostructures which serve as containers for drug delivery. Using fewer strands to assemble DNA cages is of importance to the design of DNA molecules with multiple component strands. In this study, we propose a rational assembling procedure to design and analyze DNA bipyramids with minimum strands. The results show that the odd-half twist has a major impact on assembling strands required to construct DNA cages and this method could offer a search of DNA bipyramids with minimum component strands faster. This research provides new insights into design and synthesis for DNA bipyramid-like cages from mathematical perspective.

An efficient metal-free hydroalkoxylation reaction of ynesulfonamides with esters under mild conditions is described. Under the catalysis of TMSOTf, various ynesulfonamides are transformed into the corresponding alkoxy-substituted enamides in high yields with complete regioselectivity and high to excellent stereoselectivity.

Ethylene glycol (EG) nanofluids have been intensively explored as one of the most promising solid-liquid phase change materials for subzero cold thermal energy storage (CTES). However, the prepared nanofluids usually suffer from a large supercooling degree, a long freezing period, reduced storage capacity and poor dispersion stability. Herein, we overcome these issues by developing stable EG nanofluids that are uniformly dispersed with low concentrations of monolayer ethanol-wetted graphene oxide nanosheets. The homogeneously dispersed monolayer sheet not only improves the thermal conductivity of the nanofluids (12.1%) but also provides the heterogeneous nucleation sites to trigger the crystal formation, thereby shortening the freezing time and reducing the supercooling degree. Compared with the base fluid, the nanofluids have reduced the supercooling degree by 87.2%, shortened the freezing time by 78.2% and maintained 98.5% of the latent heat. Moreover, the EG nanofluids have retained their initial stable homogeneous dispersion after repeated freezing/melting for 50 cycles, which ensures consistent CTES behavior during long-period operations. The facile preparation process, low loading requirement and consistent superior thermophysical properties would make the EG nanofluids loaded with monolayer graphene oxide sheets promising coolants for high-performance phase change-based CTES.

Oxidative stress and inflammation are necessary pathogenic factors contributing to the aetiology of diabetic retinopathy (DR). Triptolide (TPL) is derived from the traditional Chinese herb lei gong teng with anti-inflammatory, immunosuppressive and antitumor activities. This article was developed to examine the effect of TPL on DR. ARPE-19 cells were pre-treated with TPL and then stimulated by high glucose (HG). We found that TPL treatment enhanced cell viability, decreased apoptosis and ROS production in HG-treated RPE cells. MiR-29b was low-expressed in HG-treated cells, but TPL raised its expression. In addition, the protective activity of TPL towards ARPE-19 cells was attenuated when miR-29b was reduced. By utilising bioinformatics evaluation, PTEN was predicted as a downstream target of miR-29b. Also, TPL obstructed PI3K/AKT signalling pathways in HG-treated ARPE-19 Cells. Taken together, TPL secured ARPE-19 cells from HG-induced oxidative damage via regulating miR-29b/PTEN axis.

To enable the use of anthocyanins in food with high oil content, bilberry anthocyanins were acylated with cinnamic acids to address their poor lipid solubility. Structural analyses based on Fourier-transform infrared spectroscopy (FTIR), X-ray diffraction (XRD), X-ray photoelectron spectroscopy (XPS), and 1H nuclear magnetic resonance analyses indicated that cinnamic acids were efficiently grafted onto 6-OH of glucoside and galactoside and 5-OH of arabinose through an esterification reaction. The higher the dose of the acylating agent, the higher the acylation degree (AD) and the lower the total anthocyanidin content (TAC) of bilberry anthocyanins. An-Ci4 presented the highest AD value (6.61%), and An-Ci3 exhibited the lowest TAC value (50.16 mg/g). After acylating with lipophilic cinnamic acids, the lipid solubility of acylated bilberry anthocyanins significantly improved. The color of the native bilberry anthocyanin solution dissolved in ethyl acetate and dioxane was transparent. By contrast, the acylated anthocyanin solution dissolved in these solvents was unmistakably red. In terms of the antioxidant activity, acylated bilberry anthocyanins demonstrated inferior performance in 2,2-diphenyl-1-picrylhydrazyl (DPPH) clearance but a better inhibition ratio in β-carotene bleaching assay compared with native bilberry anthocyanins. As AD value increased, the DPPH clearance of acylated anthocyanins decreased and their inhibition ratio increased in β-carotene bleaching assay.

Description, EDS1 acts as a transcriptional coactivator that interacts with NPR1 and Mediator to promote plant defense., The phytohormone salicylic acid (SA) plays a pivotal role in plant defense against biotrophic and hemibiotrophic pathogens. NPR1 and EDS1 function as two central hubs in plant local and systemic immunity. However, it is unclear how NPR1 orchestrates gene regulation and whether EDS1 directly participates in transcriptional reprogramming. Here, we show that NPR1 and EDS1 synergistically activate pathogenesis-related (PR) genes and plant defenses by forming a protein complex and recruiting Mediator. We discover that EDS1 functions as an autonomous transcriptional coactivator with intrinsic transactivation domains and physically interacts with the CDK8 subunit of Mediator. Upon SA induction, EDS1 is directly recruited by NPR1 onto the PR1 promoter via physical NPR1-EDS1 interactions, thereby potentiating PR1 activation. We further demonstrate that EDS1 stabilizes NPR1 protein and NPR1 transcriptionally up-regulates EDS1. Our results reveal an elegant interplay of key coactivators with Mediator and elucidate important molecular mechanisms for activating transcription during immune responses.

Pomelo seeds (PS) are important by-product of pomelo fruits (Citrus grandis Osbeck). The value-added utilization of PS remains highly challenged. This study aimed to investigate the utilization potential of PS as natural antioxidant, antibacterial, herbicidal agents, and their functional components. The ethanolic extract (EE) of PS and its four fractions as PEE (petroleum ether extract), AcOEtE (ethyl acetate extract), BTE (butanol extract), and WE (water extract), were prepared and biologically evaluated. BTE exhibited the best antioxidant activity among all these extracts, in both ABTS (2,2-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) diammonium salt) and FRAP (ferric reducing antioxidant power) assays. AcOEtE was superior to other extracts in herbicidal assay against both Festuca elata Keng (IC50 of 0.48 mg mL-1 ) and Amaranthus retroflexus L. (IC50 of 0.94 mg mL-1 ). Meanwhile, both AcOEtE and BTE demonstrated inhibitory effects against Bacillus subtilis, Escherichia coli, and Xanthomonas citri subsp. citri, with MIC ranging 2.5-5.0 mg mL-1 . Furthermore, the primary chemical components involving naringin, deacetylnomilin, limonin, nomilin, and obacunone, were quantified in all these extracts. PCA (principal component analysis) suggested that naringin might highly contribute to the antioxidant activity of PS, and the herbicidal activity should be ascribed to limonoids. This study successfully identified AcOEtE and BTE as naturally occurring antioxidant, antibacterial, and herbicidal agents, showing application potential in food and cosmetics industries, and organic farming agriculture.

Delayed chest closure (DSC) is widely performed during the treatment of congenital heart diseases. However, the high prevalence of surgical site infection (SSI) in patients undergoing DSC affects prognosis negatively. Herein, we designed a suturable poly (vinyl alcohol)/keratin film loaded with silver nanoparticles (AgNPs) as an alternative material for DSC, which was named PVA/Keratin/AgNPs. The PVA/Keratin/AgNPs films exhibited significantly enhanced mechanical strength after crosslinking by sodium trimetaphosphate (STMP). These films were non-toxic, and cells proliferated with good morphology after 1 week of culture. In addition, PVA/Keratin/AgNPs films provided superior antibacterial ability, as evidenced by the eradication and lower growth rate of Staphylococcus aureus and Escherichia coli. Finally, the PVA/Keratin/AgNPs films were demonstrated to successfully cover the chest cavity temporarily and protect the chest cavity from bacterial infection. These results indicated that the PVA/Keratin/AgNPs films have great prospects to be further exploited for clinical applications in DSC.

Two novel rhodamine-based polystyrene solid-phase fluorescent sensors PS-AC-I and PS-AC-II with different coordination atoms (O or S) are synthesized and shown to be able to detect Hg(II) ions. They are characterized by Fourier-transform infrared spectroscopy and by scanning electron microscopy analysis. Their fluorescent properties, including response time, pH effects, fluorescence titrations, metal ion competition and recycling, are investigated and compared. Sensor PS-AC-II displayed higher selectivity and sensitivity to Hg(II), with a lower detection limit of 0.032 µM, which was 15 times better than PS-AC-I. A detection mechanism involving the Hg(II) chelation-induced ring-opening of the rhodamine spirolactam is proposed with the aid of theoretical calculations.

Two novel naphthalimide-piperazine-pyridine-based polystyrene solid-phase fluorescent sensors PS-NA and PS-ND with different lengths of the linker were synthesized and shown to be able to detect Hg(ii) ions. Their structures were characterized by Fourier transform infrared (FTIR) spectroscopy and scanning electron microscopy (SEM) analysis. Fluorescence properties, including response time, pH effects, fluorescence titration, metal ion selectivity and regeneration, were investigated and compared. Sensor PS-NA displayed a higher fluorescence response to Hg(ii) than PS-ND, with a lower detection limit of 1.01 μM. The detection mechanism involving the Hg(ii) chelation-induced photo-induced electron transfer (PET) was proposed with the aid of density functional theory (DFT) calculations. Sensors PS-NA and PS-ND with seven other similar sensors from our previous studies were collected together for thorough structure-fluorescence relationship (SFR) studies. Sensor PS-NA being recyclable and environmentally friendly was successfully employed in the fluorescence detection of Hg(ii) in real water samples, indicating its good potential in practical application.

The interaction between water and graphene oxide (GO), including water adsorption and desorption, has been widely used to demonstrate moisture-electric energy transformation (MEET). By taking advantage of water desorption stimulated by infrared (IR) light from a graphene oxide film with a gradient of oxygen-containing groups (O-group), a self-powered IR detection approach that can convert IR light into electric signals is demonstrated. The temperature sensitivity reaches 4.2 mK. With further optimization of the materials and structures, the performance of such self-powered IR detection can be further improved. The mechanism of the detection involving an IR/thermal effect on the interaction between vapor molecules and solid materials also has potential applications in thermal detection, thermal energy conversion, and chemical sensing.

Polyphosphate (copper polyphosphate: CuPPA, zinc polyphosphate: ZnPPA, cobalt polyphosphate: CoPPA and nickel polyphosphate: NiPPA) was used as surface modifiers, smoke suppressants and flame retardants to modify the magnesium hydroxide (MH). Core–shell flame retardants composed of transition metals (Cu, Zn, Co, Ni) and P were prepared. The morphology, chemical composition and structure of core–shell flame retardants were characterized by scanning electron microscopy, Fourier-transform infrared spectrum and X-ray diffraction. Compared with MH, the core–shell flame retardant endows EVA resin with better flame retardant, smoke suppression and mechanical properties. Under the same loading of flame retardant, the peak heat release rate, total heat release and CO production of EVA + Cu decreased by 41.45%, 7.51% and 54.84%, respectively, and the mass residue increased by 12.42%. These improvements can be attributed to the catalytic carbonation of transition metal compounds and better protection of char layer formation. The synergistic effect between polyphosphate and MH was discussed. Due to the extensive synergy between flame retardant metal ion and polyphosphate group, it had a potential application prospect in the preparation of various flame retardant polymers.

Two novel rhodamine-based polystyrene solid-phase fluorescence sensors PS-PA-I and PS-PA-II with different lengths of polyamines were synthesized for Hg(II) determination. The detection mechanism involving the Hg(II) chelation-induced spirocycle open of rhodamine was proposed with the aid of theoretical calculation. The stronger N—Hg bond and the longer polyamine chain in PS-PA-II led to a better selectivity, much higher and more quickly fluorescence response to Hg(II).

Ferroptosis is a recently emerging non-apoptotic mode of cell death involving the production of iron-dependent reactive oxygen species (ROS). Here we described a mitochondria-targeted iridium (III) complex IrFN that exhibited potent antiproliferative activity against a variety of cancer cells, especially the A2780 human ovarian cancer cells, through the ferroptosis pathways. Mechanistic studies by label-free quantitative proteomics profiling indicated that heme oxygenase 1 (HMOX1)-mediated ferroptosis process was activated by IrFN. The study on iron-dependent cell death, ROS accumulation, lipid peroxidation, and over released iron further confirmed the ferroptosis processes. mRNA transcription quantification, in vitro over-expression of HMOX1, and RNAi-mediated knock-down experiments suggested that IrFN activated the over-expression of HMOX1. Our report revealed the first case of anticancer iridium complex leading to ferroptosis, highlighting ferroptosis as a promising approach in future design of metallodrugs.

Epidemiological evidence showed that the particulate matter exposure is associated with atherosclerotic plaque progression, which may be related to foam cell formation, but the mechanism is still unknown. The study was aimed to investigate the toxic effects and possible mechanism of PM2.5 on the formation of macrophage foam cells induced by oxidized low density lipoprotein (ox-LDL). Results showed that PM2.5 induced cytotoxicity by decreasing the cell viability and increasing the LDH level in macrophage foam cells. PM2.5 aggravated the lipid accumulation in ox-LDL-stimulated macrophage RAW264.7 within markedly increasing level of intracellular lipid by Oil red O staining. The level of ROS increased obivously after co-exposure to PM2.5 and ox-LDL than single exposure group. In addition, serious mitochondrial damage such as the mitochondrial swelling, cristae rupturing and disappearance were observed in macrophage foam cells. The loss of the mitochondrial membrane potential (MMP) further exacerbated the mitochondrial damage in PM2.5-induced macrophage foam cells. The apoptotic rate increased more severely via up-regulated protein level of Bax, Cyt C, Caspase-9, Caspase-3, and down-regulated that of Bcl-2, indicating that PM2.5 activated the mitochondrial-mediated apoptosis pathway. In summary, our results demonstrated that PM2.5 aggravated the lipid accumulation, mitochondrial damage and apoptosis in macrophage foam cells, suggesting that PM2.5 was a risk factor of atherosclerosis progression.

In recent years, an increasing number of defucosylated therapeutic antibodies have been applied in clinical practices due to their better efficacy compared to fucosylated counterparts. The establishment of stable and clonal manufacturing cell lines is the basis of therapeutic antibodies production. Bioprocess development of a new cell line is necessary for its future applications in the biopharmaceutical industry. We engineered a stable cell line expressing defucosylated anti-HER2 antibody based on an established α-1,6-fucosyltransferase (FUT8) gene knockout CHO-S cell line. The optimization of medium and feed was evaluated in a small-scale culture system. Then the optimal medium and feed were scaled up in a bioreactor system. After fed-batch culture over 13 days, we evaluated the cell growth, antibody yield, glycan compositions and bioactivities. The production of anti-HER2 antibody from the FUT8 gene knockout CHO-S cells in the bioreactor increased by 37% compared to the shake flask system. The N-glycan profile of the produced antibody was consistent between the bioreactor and shake flask system. The antibody-dependent cellular cytotoxicity activity of the defucosylated antibody increased 14-fold compared to the wild-type antibody, which was the same as our previous results. The results of our bioprocess development demonstrated that the engineered cell line could be developed to a biopharmaceutical industrial cell line.

Biomineralization is a phenomenon in which organisms form crystals. Studies have shown that many fungi have the ability to biomineralize, it can exhibit calcium oxalate crystals on their hyphae and fruiting body. Schizophyllum commune is a common saprophytic fungus distributed all over the world, but there is little research on its biomineralization. In this paper, S. commune fruiting body from three different provinces of China were collected, and isolation for hyphal cultured to obtain several samples. Utilizing light microscope, FE-SEM, and EDAX, the existence of crystals on the fruiting body and mycelium of each strain was found, and their morphological characteristics and ion content were analyzed. It was ultimately established that biomineralization occurs on S. commune.

Background The less volatile fraction of perfumes is susceptible to contamination by phthalate esters (PAEs), which are endocrine-disrupting chemicals encountered during the production, bottling, and transportation processes. There is a need to establish an innovative and rapid method for the determination of trace PAEs in commercial perfumes. Objective Therefore, self-built spray-inlet microwave plasma torch (MPT) ionization coupled with quadrupole time-of-flight (QTOF) tandem MS (MS/MS) was developed for the direct analysis of PAEs in perfumes without sample pretreatment. Methods Perfumes were sprayed directly onto the flame of the MPT by means of a sampling pump at a rate of 10 µL/min and the ions produced by the MPT were introduced into the QTOF-MS equipment with the MPT operating at 2450 MHz and 40 W at a flow rate of 800 ml/min of argon. Results For the established method, excellent linearity was obtained with a correlation coefficient of 0.9799 for di-n-pentyl phthalate and of greater than 0.99 for the other substances tested. The LOD and LOQ obtained were in the ranges of 1.452–18.96 and 4.839–63.19 ng/g, respectively. The spiked recoveries of PAEs ranged from 100.1 to 105.2% with satisfactory intraday RSDs and interday RSDs ranging from 1.51 to 4.34% and 3.45 to 5.65%, respectively. PAEs in commercial perfumes were successfully detected by spray-inlet MPT-MS/MS with low concentrations ranging from 0.036 to 1.352 µg/g. Conclusions The method is a promising tool for the on-site analysis of PAEs in cosmetic solutions requiring rapid qualitative and quantitative analysis. Highlights The method was successfully applied to the analysis of the commercial perfume samples within 30 s, and displayed the merits of simplicity, sensitivity, environmental friendliness and ease of operation.

The effects of different surfactants (rhamnolipid, trehalolipid and citrate) on phosphorus (P) release and acidogenic fermentation of waste activated sludge (WAS) containing different aluminium phosphate forms (AlPO4, Al(PO3)3) were investigated. Results showed that rhamnolipid was the most effective surfactant to release P from aluminum phosphates (AlPs)-rich sludge. Al(PO3)3 was easier to release P than AlPO4 in WAS due to their different crystal structures. Different surfactants promoted the production of different types of protein. The addition of rhamnolipid was conducive to produce propionate from WAS, while trehalolipid and citrate increased the production of n-butyrate and acetate, respectively. Citrobacter played an important role in producing phosphatase continuously for P release with rhamnolipid addition. Predictive functional profiling indicates that rhamnolipid greatly facilitated adenosine triphosphate (ATP)-binding cassette transporter and quorum sensing. These important discoveries help to enrich P recovery paths from sludge produced with Al-based coagulants in wastewater treatment plants.

Zero valent iron (ZVI) applications to remediation of shallow groundwaters can be affected by dissolved oxygen (DO) and organic ligands. To explore the intersection between these complicating factors, this study thoroughly characterized the reactions of nitrobenzene (NB) with ZVI in the presence DO and the model ligand ethylene diamine tetraacetic acid (EDTA). The results showed that NB is degraded by both ZVI reduction and ZVI-induced advanced oxidation under oxygen-limited conditions. The contribution of ·OH to the degradation of NB increased with time so that nearly 39% of NB was oxidized by ·OH at 15 min (pH = 3), but reduction was still the main pathway of NB transformation throughout. NB reduction products, such as aniline (AN), were also oxidized by ·OH. The lower the pH, the greater the contribution of advanced oxidation, but DO was the limiting factor for ·OH generation. Only 4.7% NB was fully degraded by ring opening and/or mineralization because the production of •OH was limited by low DO. After the transformation of NB and AN, other benzene ring and nitrogen-containing intermediates were identified (e.g., p-nitrophenol, p-aminophenol, hydroquinone, and p-benzoquinone). The removal of total organic carbon and total organic nitrogen was minimal. The results suggested that the relative doses of ZVI, DO, and iron-complexing ligands can be balanced for the optimal (rapid and deep) removal of organic contaminants.

Disulfide-bond A oxidoreductase-like protein (DsbA-L) is a molecular chaperone involved in the multimerization of adiponectin. Recent studies have found that DsbA-L is related to metabolic diseases including gestational diabetes mellitus (GDM), and can be regulated by peroxisome proliferator-activated receptor γ (PPARγ) agonists; the specific mechanism, however, is uncertain. Furthermore, the relationship between DsbA-L and the novel adipokine chemerin is also unclear. This article aims to investigate the role of DsbA-L in the improvement of insulin resistance by PPARγ agonists in trophoblast cells cultured by the high-glucose simulation of GDM placenta. Immunohistochemistry and western blot were used to detect differences between GDM patients and normal pregnant women in DsbA-L expression in the adipose tissue. The western blot technique was performed to verify the relationship between PPARγ agonists and DsbA-L, and to explore changes in key molecules of the insulin signaling pathway, as well as the effect of chemerin on DsbA-L. Results showed that DsbA-L was significantly downregulated in the adipose tissue of GDM patients. Both PPARγ agonists and chemerin could upregulate the level of DsbA-L. Silencing DsbA-L affected the function of rosiglitazone to promote the phosphatidylinositol 3-kinase (PI3K)-protein kinase B (PKB)/AKT pathway. Therefore, it is plausible to speculate that DsbA-L is essential in the environment of PPARγ agonists for raising insulin sensitivity. Overall, we further clarified the mechanism by which PPARγ agonists improve insulin resistance.

A novel metal-free [3+2] annulation of ynamides with anthranils provides a facile, flexible, environmentally friendly, and atom-economical route to 2-aminoindoles. This synthetic process proceeds with efficiency, excellent regioselectivity, and wide functional group tolerance under mild conditions. Moreover, the obtained 2-aminoindole products represent a multifunctional platform for the construction of various 2-aminoindolyl frameworks.

Objective. To explore the possible mechanism of electroacupuncture to improve insulin sensitivity in type 2 diabetes rats. Methods. Fourteen Zucker Diabetic Fatty (ZDF) rats were randomly divided into two groups: a model group and an electroacupuncture group, with 7 rats in each group. Seven Zucker Lean (ZL) rats served as a control group. All rats were fed with Purina #5008 for 4 weeks, and the electroacupuncture group received 4-week electroacupuncture intervention, while the control group and model group received no intervention. We measured fasting blood glucose (FBG) on the fourth weekend. After 4 weeks of intervention, the expression levels of insulin receptor substrate-1 (IRS-1) tyrosine phosphorylation, IRS-1 serine/threonine phosphorylation, and GLUT4 in quadriceps femoris muscles were detected by western Blot. Results. Compared with the model group, the electroacupuncture group had a lower level of fasting blood glucose, serum insulin level, and insulin resistance index ( P < 0.05 ). The electroacupuncture group had lower IRS-1 serine/threonine phosphorylation than the model group, with the difference showing statistical significance ( P < 0.05 ). Furthermore, the mean score (MS) of the control group showed the lowest phosphorylation expression, followed by the electroacupuncture group, while the model group had the highest level of phosphorylated protein expression. The level of IRS-1 tyrosine phosphorylation at Tyr895 sites was compared, and the result showed that there was no significant difference between the electroacupuncture group and the control group ( P > 0.05 ), and the electroacupuncture group had higher phosphorylation expression than the model group ( P < 0.05 ). Compared with the control group and the model group, the expression level of GLUT4 protein in the electroacupuncture group was significantly increased ( P < 0.05 ). Conclusion. Electroacupuncture has the effect to improve the insulin sensitivity of type 2 diabetic ZDF rats by reducing fasting blood glucose, insulin level, and insulin resistance index, effectively up regulating the expression of GLUT4 protein in quadriceps femoris muscle. The mechanism is related to the regulation of skeletal muscle IRS-1 serine/threonine and tyrosine phosphorylation levels.

S100B is a calcium-binding protein that governs calcium-mediated responses in a variety of cells&mdash, especially neuronal and glial cells. It is also extensively investigated as a potential biomarker for several disease conditions, especially neurodegenerative ones. In order to establish S100B as a viable pharmaceutical target, it is critical to understand its mechanistic role in signaling pathways and its interacting partners. In this report, we provide evidence to support a calcium-regulated interaction between S100B and the neuronal calcium sensor protein, neurocalcin delta both in vitro and in living cells. Membrane overlay assays were used to test the interaction between purified proteins in vitro and bimolecular fluorescence complementation assays, for interactions in living cells. Added calcium is essential for interaction in vitro, however, in living cells, calcium elevation causes translocation of the NCALD-S100B complex to the membrane-rich, perinuclear trans-Golgi network in COS7 cells, suggesting that the response is independent of specialized structures/molecules found in neuronal/glial cells. Similar results are also observed with hippocalcin, a closely related paralog, however, the interaction appears less robust in vitro. The N-terminal region of NCALD and HPCA appear to be critical for interaction with S100B based on in vitro experiments. The possible physiological significance of this interaction is discussed.

Transient protein expression in a heterologous system has been very useful in many research fields. As a plant expression system, tobacco has some unique advantages including big leaves, simple infiltration and transformation, high activity in expressing transgenes, and easy sampling for microscopy. Because of these advantages, tobacco system has been extensively used for many purposes, such as large-scale expression and purification of proteins of interest, protein colocalization, protein degradation, protein-protein interaction assays including co-immunoprecipitation (CoIP), fluorescence resonance energy transfer (FRET), and bimolecular fluorescence complementation (BiFC), transcription regulation, plant-pathogen interactions, and functional verification of small RNAs. A large number of publications have used this system and generated critical results to support their conclusions. The results obtained from tobacco system are highly reproducible and mostly consistent with those generated from traditional techniques, indicating its reliability. Here we describe a protocol for studying protein-protein interactions in tobacco system, which could be applied to multiple experimental purposes as the procedure of tobacco leaf infiltration is basically shared among them.

Co-immunoprecipitation (CoIP) assay has been used as a powerful and routine technique to detect in vivo protein-protein interactions. Not only can it probe stable interactions, but also it is applicable for semiquantitative and inducible protein associations. Here we describe the protocol for detecting blue light-dependent protein interactions, particularly for blue light receptor cryptochrome-mediated complex formation. In addition, we present some notes which may be helpful for common Co-IP study as well.

This study evaluates the applicability of nano-sized calcium peroxide (CaO2) as a source of H2O2 to remediate 2,4-dichlorophenol (2,4-DCP) contaminated groundwater via the advanced oxidation process (AOP). First, the effect and mechanism of 2,4-DCP degradation by CaO2 at different Fe concentrations were studied (Fenton reaction). We found that at high Fe concentrations, 2,4-DCP almost completely degrades via primarily the oxidation of ·OH within 5 h. At low Fe concentrations, the degradation rate of 2,4-DCP decreased rapidly. The main mechanism was the combined action of ·OH and O2·−. Without Fe, the 2,4-DCP degradation reached 13.6% in 213 h, primarily via the heterogeneous reaction on the surface of CaO2. Besides, 2,4-DCP degradation was significantly affected by solution pH. When the solution pH was>10, the degradation was almost completely inhibited. Thus, we adopted a two-dimensional water tank experiment to study the remediation efficiency CaO2 on the water sample. We noticed that the degradation took place mainly in regions of pH < 10 (i.e., CaO2 distribution area), both upstream and downstream of the tank. After 28 days of treatment, the average 2,4-DCP degradation level was ≈36.5%. Given the inadequacy of the results, we recommend that groundwater remediation using nano CaO2: (1) a buffer solution should be added to retard the rapid increase in pH, and (2) the nano CaO2 should be injected copiously in batches to reduce CaO2 deposition.

Poor dispersion stability of carbon nanofluids is one of the key issues limiting their solar-thermal applications especially under medium-to-high temperatures. Herein, this work reported a facile way to prepare stably dispersed graphene quantum dot-ethylene glycol (GQD-EG) medium-temperature solar-thermal nanofluids. The hydroxyl-terminated GQDs were synthesized by a scalable hydrothermal approach. The obtained GQDs have a small particle size, narrow particle size distribution and are self-dispersible within EG fluids. The GQD-EG nanofluids maintained their uniform dispersion after continuous heating at 180 °C for 7 days. The hydrogen bonding between the hydroxyl group on the surface of GQDs and the EG molecules helped achieve homogenous dispersion of GQDs in the nanofluids, and the small particle size and low density of GQDs helped mitigate the sedimentation tendency. The dispersed GQD-EG nanofluids have demonstrated broadband absorption of sunlight, high specific heat capacity and low viscosity, which are all desired for high-performance direct absorption-based solar-thermal energy. The prepared GQD-EG nanofluids have exhibited consistent volumetric harvesting of solar-thermal energy under concentrated solar illumination with a heating temperature up to 170 °C.

Salicylic acid (SA) plays pivotal roles in plant defense against biotrophic and hemibiotrophic pathogens. Tremendous progress has been made in the field of the SA biosynthesis pathways and SA-mediated plant defense signaling networks in the past three decades. As one of the SA receptors, NONEXPRESSOR OF PATHOGENESIS-RELATED GENES1 (NPR1) functions as a master regulator of SA-mediated plant defense. The function of NPR1 is tightly regulated by transcriptional and post-translational regulation. This review discusses recent advances in SA and NPR1 biology, including SA perception, SA signaling, the function of SA and NPR1 in plant immunity, and the transcriptional and post-translational regulation of NPR1.

Background The efficacy and safety of β-lactam/β-lactamase inhibitors (BL/BLIs) versus carbapenems for febrile neutropenia empiric therapy are controversial. Methods PubMed, Embase, Cochrane library databases, Web of Science and Google scholar were searched up to 1 April 2020. Studies were included if they compared BL/BLIs versus carbapenem for febrile neutropenia patients undergoing chemotherapy for either solid tumours or haematological malignancies among adults and children. We pooled the treatment success rate, mortality and adverse events. Results Nine RCTs were included. There was no differences between carbapenems and BL/BLIs were observed in terms of treatment success without modification (RR 1.04, 95% CI 0.93–1.15), no differences were observed in the subgroups of BL/BLIs, adults and children. No significant differences were found in all-cause mortality (RR 1.15, 95% CI 0.64–2.06). Our study shows that gastrointestinal events are the most common adverse effects, nausea/vomiting were significantly more common with carbapenems (RR 2.83, 95% CI 1.35–5.92, P = 0.006), however, diarrhea were more common with BL/BLIs (RR 0.47, 95% CI 0.27–0.80, P = 0.006). Conclusions The efficacy and safety of BL/BLIs with carbapenems were comparable in empiric treatment of febrile neutropenia.
