The liver consists of parenchymal hepatocytes and non-parenchymal cells. Non-parenchymal cells, Kupffer cells, hepatic stellate cells and cholangiocytes have crucial roles in liver homeostasis and liver pathology. To establish baseline data, this study investigated immunohistochemically the distribution of non-parenchymal cells in perivenular areas (PV), periportal areas (PP) and Glisson's sheath (GS) of adult rat liver. Liver tissues were collected from the left lateral lobe of rats. CD163-positive macrophages were seen along the sinusoid of PV and PP areas, indicating Kupffer cells. Double immunofluorescence showed, Kupffer cells partly co-expressed CD68 and MHC class II antigens in the liver. The numbers of Kupffer cells were significantly high in PP areas as compared with PV or GS areas. CD68-positive exudative macrophages were highly localized in PP and GS areas and a comparatively low PV area. MHC class II-positive dendritic cells (activated macrophages) were localized mainly in GS. Granzyme B-positive NK cells were mainly localized in the Glisson's sheath. CD3-positive T cells and CD20-positive B cells were distributed along the sinusoids of the PP and PV areas of hepatic lobules. Vimentin and glial fibrillary acidic protein (GFAP)-positive hepatic stellate cells were localized along sinusoids in the hepatic lobules of the liver. Cholangiocytes reacting to cytokeratin 19 were seen on interlobular bile ducts in Glisson's sheath of the liver. This study shows that heterogeneous macrophage populations, liver-resident lymphocytes and hepatic stellate cells localized in PP and PV areas or GS areas of the liver with cells specific patterns.

Blood parameters are an important index to assess the body homeostasis in mammals. Here, the effect of pesticide (imidacloprid) contaminated feed exposure on the haematological parameters of adult rabbits (n=15) was studied. Pesticides (Imidacloprid, Bildor® 0.5ml (100mg)/L water spray on vegetables and green grass) exposed feed fed to rabbits with wash (washed feed rabbit) or without a wash (not washed feed rabbit) in the once-daily morning for every alternative day up to 15 days. Control rabbits have received a standard diet (fresh vegetables and green grass). The blood cell analysis showed that the total erythrocyte count, packed cell volume, and haemoglobin values were not changed significantly in pesticide exposed rabbit. For differential leukocyte count, the percentage of neutrophil and eosinophil was significantly decreased in the pesticide-exposed rabbit as compared to the control rabbit. The lymphocyte percentage was increased significantly in pesticide-exposed rabbits compared to control rabbits. Interestingly, the number of lymphocytes was significantly increased in not washed feed rabbits compared to wash feed rabbits. Formaldehyde (FA), a ubiquitous environmental pollutant, has long been suspected to possess reproductive toxicity. Here, we investigated the histopathological alteration of male gonads following exposure to FA-contaminated feed (40% aqueous solution of FA; 2.5 ml formalin/kg feed) in pigeons for 15 days. The mean body weights were not changed significantly in FA-contaminated feed exposed pigeons compared with control pigeons. The hemoglobin concentration was significantly decreased and serum enzyme aspartate transaminase (AST) was significantly increased in FA-exposed pigeons in comparison with control pigeons. Histologically, the structural components of the testes are the seminiferous tubules and interstitial tissues, which are surrounded by a connective tissue capsule. In control pigeons, the size and shape of seminiferous tubules were normal with a regular arrangement of spermatogenic cells. In FA-exposed pigeons, the testicular capsule was thickened and degeneration of spermatogenic cells in the seminiferous tubules was observed. The number of spermatogenic cells was significantly decreased in the seminiferous tubules of FA-exposed pigeons in comparison with control pigeons, indicating that the low exposure of FA affects the spermatogenic cells populations in male birds. Short-statured and lodging resistant rice plants with long to medium slender grain are the expected criteria for aromatic rice. However, most of the aromatic rice varieties in Bangladesh do not meet the expected criteria. Therefore, this study was undertaken to detect short-statured rice plants with aromatic and long to medium slender grain where twelve advanced rice lines (derived from the local rice germplasm) with a local check Kataribhog were evaluated. All the genotypes demonstrated significant variation for different parameters such as plant height at harvest, yield per hectare, grain length, grain size, grain shape, and aroma. At harvest, the shortest plant height was obtained from SAU ADL11 (107.09cm), and it was statistically similar to SAU ADL4 (111.68cm) and SAU ADL5 (111.89cm). However, the plant height of check variety Kataribhog was 167.17cm. The highest grain yield per hectare was found in SAU ADL5 (4.79 tha-1), where it was 2.71 tha-1 in Kataribhog. Besides, the grain yield of shorter genotypes SAU ADL4 and SAU ADL11 was 3.47 t ha-1 and 3.84 t ha-1 , respectively. The SAU ADL1 provided the highest kernel length (7.31mm), and the lowest kernel length (4.87 mm) was recorded in the check. Kernel length of short-statured genotypes ranged from 7.01 mm to 6.57 mm. Kernel size of all evaluated genotypes was long to medium; whereas, it was short for Kataribhog. Kernel shape of these short-statured genotypes was slender to medium. In case of aroma, SAU ADL3, SAU ADL5, SAU ADL7, SAU ADL9, SAU ADL10, and SAU ADL11 were moderately aromatic, and other genotypes were non-aromatic. However, Kataribhog was strongly aromatic. Finally, SAU ADL5 and SAU ADL11 were evaluated as important germplasms in respect of different characters such as shortstatured plant, long to medium slender grain, and aroma. Effects of pesticide exposure on public health are monitored in developed countries, which is rare in the least developed countries like Bangladesh. Here, a surveillance study was conducted to investigate the farmer's knowledge and perception toward pesticide use and the impact of occupational exposure as well as an environmental hazard in six agro-based districts (Gazipur, Khulna, Manikganj, Naogaon, Mymensingh and Tangail) in Bangladesh. A questionnaire was designed for a face to face interview of the farmers (at least 100 farmers in each district), and the data was collected. The farmer during cultivations used pesticides as single, double or several times in a single crop cycle. Among different types of pesticides, insecticides were highly used (38%), whereas Imidacloprid, and Lamda cyhalothrin insecticide was top used by the farmers. Weedicides or herbicide, fungicides and rodenticides were used 11%, 48%, and 0.5%, respectively. The knowledge and perception of the farmers towards pesticide usage are minimum, whereas the environmental pollution aspect is deficient. The knowledge of using personal protective equipment is the least. Eighty five percent of farmers use pesticides by spraying in the field. Thirty nine percent of farmers suffered from sneezing, burning sensation on face, conjunctivitis, dizziness and headache during pesticide application, and 3% suffered from skin rash of long time exposed with pesticide. The exposure of pesticides is widespread in the professional life of a farmer in Bangladesh. So, it is essential to ensure different training and awareness building program for the farmers to cope the adverse situation.

Left ventricular (LV) heart failure (HF) is a significant and increasing cause of death worldwide. HF is characterized by myocardial remodeling and excessive fibrosis. Transcriptional co-activator Yes-associated protein (Yap), the downstream effector of HIPPO signaling pathway, is an essential factor in cardiomyocyte survival, however, its status in human LV HF is not entirely elucidated. Here, we report that Yap is elevated in LV tissue of patients with HF, and is associated with down-regulation of its upstream inhibitor HIPPO component large tumor suppressor 1 (LATS1) activation as well as upregulation of the fibrosis marker connective tissue growth factor (CTGF). Applying the established profibrotic combined stress of TGFβ and hypoxia to human ventricular cardiac fibroblasts in vitro increased Yap protein levels, down-regulated LATS1 activation, increased cell proliferation and collagen I production, and decreased ribosomal protein S6 and S6 kinase phosphorylation, a hallmark of mTOR activation, without any significant effect on mTOR and raptor protein expression or phosphorylation of mTOR or 4E-binding protein 1 (4EBP1), a downstream effector of mTOR pathway. As previously reported in various cell types, TGFβ/hypoxia also enhanced cardiac fibroblast Akt and ERK1/2 phosphorylation, which was similar to our observation in LV tissues from HF patients. Further, depletion of Yap reduced TGFβ/hypoxia-induced cardiac fibroblast proliferation and Akt phosphorylation at Ser 473 and Thr308, without any significant effect on TGFβ/hypoxia-induced ERK1/2 activation or reduction in S6 and S6 kinase activities. Taken together, these data demonstrate that Yap is a mediator that promotes human cardiac fibroblast proliferation and suggest its possible contribution to remodeling of the LV, opening the door to further studies to decipher the cell-specific roles of Yap signaling in human HF.

An experiment was conducted at Regional Agricultural Research Station, Jashore, Bangladesh during rabi season of (2019-2020) to evaluate the effect of foliar application of urea on growth and yield of short durative lentil variety (BARI Masur-9). The experiment was conducted in split plot design with three replications where time of urea spraying, P1= at branching stage, P2= at pod initiation stage were distributed in main plots and doses of urea spraying T1= application of 100% recommended fertilizers as basal except urea, T2= application of 100% recommended fertilizers as basal, T3= application of 75% of urea with 100% of other fertilizers as basal and rest of the 25% of urea by spraying, T4= application of 25% of urea with 100% of other fertilizers as basal and rest of the 75% of urea by spraying, T5= application of 50% of urea with 100% of other fertilizers as basal and rest of the 50% of urea by spraying were distributed in sub plots. The highest plant height, number of pods per plant were obtained from application of 75% of urea with 100% of other fertilizers as basal and rest of the 25% of urea spraying at pod initiation stage. The highest seed yield was also found from application of 75% of urea with 100% of other fertilizers as basal and rest of the 25% of urea spraying at pod initiation stage followed by application of 100% recommended fertilizers as basal and application of 50% of urea with 100% of other fertilizers as basal and rest of the 50% of urea by spraying at branching stage. Plant height, pods per plant and days to maturity showed positive correlation with seed yield. On the other hand days to flower and plants per m2 has no linear relationship with seed yield. The highest marginal benefit cost ratio (3.29) was recorded from application of 75% of urea with 100% of other fertilizers as basal and rest of the 25% of urea by spraying at pod initiation stage. So based on findings to increase the yield potential of BARI Masur-9 foliar application of urea may be a tool and hence application of 75% of urea with 100% of other fertilizers as basal and rest of the 25% of urea by spraying at pod initiation stage may be consider as the best treatment.

Objective: Doxycycline hyclate is a broad spectrum antibiotic with activity against a wide range of gram-positive and gram-negative bacteria and it is widely used as a pharmacological agent. A simple, selective, linear, precise and accurate ultraviolet detection (UV) method has been developed and applied for the determination of doxycycline hyclate in different pharmaceutical samples. Methods: Acid-base analysis and titrimetric method were utilized to determine the value of pH and moisture content of purchased pharmaceutical samples. A mixture of methanol and hydrochloric acid (0.01N Methanolic HCl) was used to determine the biochemical properties of doxycycline hyclate. UV detector set at 349 nm was used to monitor the effluent. The purified water was used as solvent. Results: In 1% aqueous solution of doxycycline, three samples (4th, 5th and 7th) showed lower pH values of 1.97, 1.98, and 1.99 respectively. Furthermore, the same samples indicated the additional moisture contents of 2.81%, 2.85% and 2.83% respectively while considering the acceptance level (1.4% to 2.8%). The method proved to be linear (R2=0.993), precise (RSD=0.79% for inter-day precision), accurate (Recovery=100.59%) and selective regarding possible impurities and excipients of the samples. The doxycycline content obtained in the sample analysis was within the range of 84.05% to 85.80%. Conclusion: The optimized and validated method may be successfully employed to perform routine quality control analyses. Investigation of the pH, moisture content and potency of doxycycline hyclate in different samples give a general view of local pharmacies trade and ensure that the method applied here was validated for this kind of analysis.

Bacteria and Eukarya share essentially the same family of protein-serine/threonine kinases, also known as the Hanks-type kinases. However, when it comes to protein-tyrosine phosphorylation, bacteria seem to have gone their own way. Bacterial protein-tyrosine kinases (BY-kinases) are bacterial enzymes that are unique in exploiting the ATP/GTP-binding Walker motif to catalyze phosphorylation of protein tyrosine residues. Characterized for the first time only a decade ago, BY-kinases have now come to the fore. Important regulatory roles have been linked with these enzymes, via their involvement in exopolysaccharide production, virulence, DNA metabolism, stress response and other key functions of the bacterial cell. BY-kinases act through autophosphorylation (mainly in exopolysaccharide production) and phosphorylation of other proteins, which have in most cases been shown to be activated by tyrosine phosphorylation. Protein-tyrosine phosphorylation in bacteria is particular with respect to very low occupancy of phosphorylation sites in vivo; this has represented a major challenge for detection techniques. Only the recent breakthroughs in gel-free high resolution mass spectrometry allowed the systematic detection of phosphorylated tyrosines by phosphoprotomics studies in bacteria. Other pioneering studies conducted in recent years, such as the first structures of BY-kinases and biochemical and phyiological studies of new BY-kinase substrates significantly furthered our understanding of these enzymes and highlighted their importance in bacterial physiology. Having no orthologues in Eukarya, BY-kinases are receiving a growing attention from the biomedical field, since they represent a particularly promising target for anti-bacterial drug design.

Despite of considerable progress in our understanding of the role of this modification in bacterial physiology, the dynamics of protein phosphorylation during bacterial growth has rarely been systematically addressed. In addition, little is known about in vivo substrates of bacterial Ser/Thr/Tyr kinases and phosphatases. An excellent candidate to study these questions is the Gram-positive bacterium Bacillus subtilis, one of the most intensively investigated bacterial model organism with both research and industrial applications. Here we employed gel-free phosphoproteomics combined with SILAC labeling and high resolution mass spectrometry to study the proteome and phosphoproteome dynamics during the batch growth of B. subtilis. We measured the dynamics of 1666 proteins and 64 phosphorylation sites in five distinct phases of growth. Enzymes of the central carbon metabolism and components of the translation machinery appear to be highly phosphorylated in the stationary phase, coinciding with stronger expression of Ser/Thr kinases. We further used the SILAC workflow to identify novel putative substrates of the Ser/Thr kinase PrkC and the phosphatase PrpC during stationary phase. The overall number of putative substrates was low, pointing to a high kinase and phosphatase specificity. One of the phosphorylation sites affected by both, PrkC and PrpC, was the Ser281 on the oxidoreductase YkwC. We showed that PrkC phosphorylates and PrpC dephosphorylates YkwC in vitro and that phosphorylation at Ser281 abolishes the oxidoreductase activity of YkwC in vitro and in vivo. Our results present the most detailed phosphoproteomic analysis of B. subtilis growth to date and provide the first global in vivo screen of PrkC and PrpC substrates.

The present experiment was conducted to prepare soymilk from whole soya seed by using various techniques. Twelve (12) soymilk samples were prepared by three different methods with four concentration level. Each sample was analyzed for proximate composition and sensory qualities were evaluated by a taste panel. No significant difference was found between dehulled soy seed blending milk and whole soy seed powder milk. But 125g concentration obtained highest score from the judges. From the results of chemical analysis, significant difference was found in terms of moisture, total solids, solids- not- fat, protein, fat, carbohydrate, ash content and acidity percentage. Dehulled soy seed blending milk contained more moisture, protein, fat and ash content. On the other hand, a sharp increase of nutrient concentration was observed with the increase of soymilk concentration. But soymilk of 150 and 175g concentration become thicker after boiling which had fewer acceptances by the judges. Considering the physical and chemical properties of all samples, it is recommended that 125g concentrate dehulled soy seed blending milk may be prepared in industrial scale.
